For the most part, research in the area of cytogerontology, i.e., investigation of the mechanisms of aging in the experiments on cultured cells, is carried out using the "Hayflick's model". More than forty years have passed since the appearance of that model, and during this period of time, very much data were obtained on its basis. These data contributed significantly to our knowledge of the behavior of both animal and human cultured cells. Specifically, we already know of the mechanisms underlying the aging in vitro. On the other hand, in my opinion, little has changed in our knowledge of the aging of the whole organism. In all likelihood, this can be explained by that the Hayflick's model is, like many others used in the experimental gerontology, correlative, i.e. based on a number of detected correlations. In the case of Hayflick's model, these are correlations between the mitotic potential of cells (cell population doubling potential) and some "gerontological" parameters and indices: species life-span, donor age, evidence of progeroid syndromes, etc., as well as various changes of normal (diploid) cells during long-term cultivation and during aging of the organism. It is, however, well known that very frequently a good correlation has nothing to do with the essence (gist) of the phenomenon. For example, we do know that the amount of gray hair correlates quite well with the age of an individual but is in no way related to the mechanisms of his/her aging and probability of death. In this case, the absence of cause-effect relationships is evident, which are, at the same time, indispensable for the development of gist models. These models, as distinct from the correlative ones, are based on a certain concept of aging. In the case of Hayflick's model, such a concept is absent: we cannot explain, using the "Hayflick's limit", why our organism ages. This conclusion was convincingly confirmed by the discovery of telomere mechanism which determines the aging of cells in vitro. That discovery initiated the appearance of theories attempting to explain the process of aging in vivo also on its basis. However, it has become clear that the mechanisms of aging of the entire organism, located, apparently, in its postmitotic cells, such as neurons or cardiomyocytes, cannot be explained in the framework of this approach. Hence, we believe that it is essential to develop "gist" models of aging using cultured cells. The mechanisms of cell aging in such models should be similar to the mechanisms of cell aging in the entire organism. Our "stationary phase aging" model could be one of such models, which is based on the assumption of the leading role of cell proliferation restriction in the processes of aging. We assume that the accumulation of "senile" damage is caused by the restriction of cell proliferation either due to the formation of differentiated cell populations during development (in vivo) or to the existence of saturation density phenomenon (in vitro). Cell proliferation changes themselves do not induce aging, they only lead to the accumulation of macromolecular defects, which, in turn, lead to the deterioration of tissues, organs, and, eventually, of the entire organism, increasing the probability of its death. Within the framework of our model, we define cell aging as the accumulation in a cell population of various types of damage identical to the damage arising in senescing multicellular organism. And, finally, it is essential to determine how the cell is dying and what the death of the cell is. These definitions will help to draw real parallels between the "genuine" aging of cells (i.e., increasing probability of their death with "age") and the aging of multicellular organisms.  相似文献

"太岁"是一种存在于地球上分类地位尚不清楚的生物体。对实验室保藏的7种不同的"太岁"样品进行了生物学组分的定性定量分析,旨在科学地了解"太岁"的组成成分,同时为"太岁"有效成分的开发利用和分类鉴定提供理论依据。对实验室保藏的7种"太岁"样品做生物组分及含量分析,具体包括:苯酚硫酸法测粗多糖含量、凯氏定氮法测粗蛋白含量、索氏提取法测粗脂肪含量、钒钼酸显色法测核酸含量、灼烧重量法测粗灰分含量、恒温干燥法测水分含量、原子吸收法测微量元素含量。实验结果表明,7种"太岁"样品含水量均较高,除LNUT006含水量73.58%外,其余"太岁"样品均达到90%以上;"太岁"样品中粗多糖含量在8.97~46.57 mg/g之间,其中LNUT006多糖含量明显高于其他"太岁"样品;粗蛋白含量在20.35~184.18 mg/g之间,其中LNUT006蛋白含量明显高于其他"太岁"样品;"太岁"样品中净核酸含量在2.053~35.128 6μg/g之间;灰分含量在42.93~246.4 mg/g之间;"太岁"样品钙、铁含量较高,分别为2 907.09μg/g和3 929.09μg/g,铅、镉含量最低。首次对未知生物"太岁"样品的组成成分进行初步系统的研究,为明确其组分和对其进一步的开发利用均有很重要的意义。  相似文献

Origin of complex neuroendocrine systems in evolution seems to be based on the signalling functions of molecules produced by separate groups of cells and glands, and each of these molecules "is coding" some distinct conditions of internal milieu and environment for the cell and the whole organism. The definition "physiological age" of animals has been revised. A new definition has been formulated on the basis of V. N. Beklemishev's (1962) and Ju. S. Balashov's (1962) theses as follows: physiological age of animals (= biological) is an extent of general irreversible morphophysiological change of organism during its whole life and determined by the accumulation of irreversible changes as a result of its normal vital activity. This definition is equivalent to that of "biological ages" of man. The contents of this definition is enlarged and the possibility of its use for different animals including their males is unified. On the basis of the general concept this definition is specified in regard to ixodid ticks: physiological age of hungry ixodid tick is an extent of general irreversible morpholophysiological change of its organism during the whole life and determined by the state of reserve nutrient and excretory substances. According to the amount of reserve and excretory substances in the organism of ixodid tick we distinguish 4 main physiological ages: new-born, young, mature and old which reflect general biological regularity of age development of animals.  相似文献

The H(2)-oxidizing lithoautotrophic bacterium Ralstonia eutropha H16 is a metabolically versatile organism capable of subsisting, in the absence of organic growth substrates, on H(2) and CO(2) as its sole sources of energy and carbon. R. eutropha H16 first attracted biotechnological interest nearly 50 years ago with the realization that the organism's ability to produce and store large amounts of poly[R-(-)-3-hydroxybutyrate] and other polyesters could be harnessed to make biodegradable plastics. Here we report the complete genome sequence of the two chromosomes of R. eutropha H16. Together, chromosome 1 (4,052,032 base pairs (bp)) and chromosome 2 (2,912,490 bp) encode 6,116 putative genes. Analysis of the genome sequence offers the genetic basis for exploiting the biotechnological potential of this organism and provides insights into its remarkable metabolic versatility.  相似文献

An attempt is made to consider disease and aging following from the concepts of the essence of life. The proposed definition of life represents a modified Engels' (1878) definition. Proceeding from the analysis of possible mechanisms of different disturbances in the life process leading to a decreased probability of the organism existence it is concluded that disease develops either as a result of hereditary changes in the genome or due-to disorders in its realization under certain unfavorable conditions. The method for studying the effects of weak magnetic fields and "magnetic vacuum" on the psychophysiological state of a human organism is proposed. This method includes the system of the exposure of a human organism to uniform constant and alternating magnetic fields and the system of computerized psychological tests. The influence of the weakening of constant magnetic field on the psychophysiological state of human organisms was studied. The short-term color memory and reaction rates of 30 subjects have been examined in the local geomagnetic field and in a magnetic field which was reduced in 10 and more times. Statistically significant differences in the color memory test was found in the magnetic field 4 +/- 1 microT in comparison with the results in the geomagnetic field. In the magnetic field 0 +/- 1 microT, slight impairment of color memory was found. Preliminary results in the test of reaction rates showed the tendency to slowing down the reaction rates in the weakened magnetic fields.  相似文献

Putative gene predictions of the Gram positive actinobacteria Micrococcus luteus (NCTC 2665, "Fleming strain") was used to construct a genome scale reconstruction of the metabolic network for this organism. The metabolic network comprises 586 reactions and 551 metabolites, and accounts for 21% of the genes in the genome. The reconstruction was based on the annotated genome and available biochemical information. M. luteus has one of the smallest genomes of actinobacteria with a circular chromosome of 2,501,097 base pairs and a GC content of 73%. The metabolic pathways required for biomass production in silico were determined based on earlier models of actinobacteria. The in silico network is used for metabolic comparison of M. luteus with other actinomycetes, and hence provides useful information for possible future biotechnological exploitation of this organism, e.g., for production of biofuels.  相似文献

Photomicrographic data were collected to measure the kinetics of elongation of "Lineola longa," a large gram-negative rod ranging from 5 to 10 microns long, during the exponential phase of growth. Its large size makes this organism especially well suited for light microscopic observations. Because this organism is aerobic, it was necessary to ensure a saturating supply of oxygen during growth. Oxygen was supplied by using Chlorella species, in a Wheaton microculture slide, as an oxygen donor separated from the bacteria by a thin layer of agar. In another set of experiments, water-saturated air replaced Chlorella species, with similar results. Statistical analysis of the data showed that "L. longa" elongates in an exponential manner. Minicell-like structures, small spherical cells lacking DNA, were occasionally seen to be produced by aberrant septation. These minicells were seen most often at the end of the exponential-growth phase. A model of cell growth is proposed to account for these observations.  相似文献

A fascinating property of germ cells is their ability to maintain totipotency throughout development. At fertilization, this totipotency is unleashed and the egg generates all the cell types needed to make a brand new organism. Occasionally, germ cells differentiate precociously in the embryo or in the gonads and form teratomas, tumors containing many differentiated somatic cell types. Until recently, the genetic basis for teratoma formation was not known. The unexpected discovery of a teratoma in a C. elegans double mutant points to translational control as a key mechanism to maintain totipotency in developing germ cells.  相似文献

"Candidatus Accumulibacter phosphatis" is considered a polyphosphate-accumulating organism (PAO) though it has not been isolated yet. To reveal the denitrification ability of this organism, we first concentrated this organism by flow cytometric sorting following fluorescence in situ hybridization (FISH) using specific probes for this organism. The purity of the target cells was about 97% of total cell count in the sorted sample. The PCR amplification of the nitrite reductase genes (nirK and nirS) from unsorted and sorted cells was performed. Although nirK and nirS were amplified from unsorted cells, only nirS was detected from sorted cells, indicating that "Ca. Accumulibacter phosphatis" has nirS. Furthermore, nirS fragments were cloned from unsorted (Ba clone library) and sorted (Bd clone library) cells and classified by restriction fragment length polymorphism analysis. The most dominant clone in clone library Ba, which represented 62% of the total number of clones, was not found in clone library Bd. In contrast, the most dominant clone in clone library Bd, which represented 59% of the total number of clones, represented only 2% of the total number of clones in clone library Ba, indicating that this clone could be that of "Ca. Accumulibacter phosphatis." The sequence of this nirS clone exhibited less than 90% similarity to the sequences of known denitrifying bacteria in the database. The recovery of the nirS genes makes it likely that "Ca. Accumulibacter phosphatis" behaves as a denitrifying PAO capable of utilizing nitrite instead of oxygen as an electron acceptor for phosphorus uptake.  相似文献

The population genetics and structure of P. falciparum determine the rate at which malaria evolves in response to interventions such as drugs and vaccines. This has been the source of considerable recent controversy, but here we demonstrate the organism to be essentially sexual, in an area of moderately high transmission in the Lower Shire Valley, Malawi. Seven thousand mosquitoes were collected and dissected, and genetic data were obtained on 190 oocysts from 56 infected midguts. The oocysts were genotyped at three microsatellite loci and the MSP1 locus. Selfing rate was estimated as 50% and there was significant genotypic linkage disequilibrium (LD) in the pooled oocysts. A more appropriate analysis searching for genotypic LD in outcrossed oocysts and/or haplotypic LD in the selfed oocysts found no evidence for LD, indicating that the population was effectively sexual. Inbreeding estimates at MSP1 were higher than at the microsatellites, possibly indicative of immune action against MSP1, but the effect was confounded by the probable presence of null mutations. Mating appeared to occur at random in mosquitoes and evidence regarding whether malaria clones in the same host were related (presumably through simultaneous inoculation in the same mosquito bite) was ambiguous. This is the most detailed genetic analysis yet of P. falciparum sexual stages, and shows P. falciparum to be a sexual organism whose genomes are in linkage equilibrium, which acts to slow the emergence of drug resistance and vaccine insensitivity, extending the likely useful therapeutic lifespan of drugs and vaccines.  相似文献

Cell reaction of the interstitial tissue in the aseptic inflammation areas was studied to determine the nonspecific reactivity of the organism in patients with infiltrative tuberculosis of the lungs. Rebuck's "skin window" method modified by Shurenkova was used. Results of investigations conducted in 15 patients and 10 healthy individuals indicated the absence in them of any significant difference in the character of the inflammatory exudate. Consequently, infiltrative tuberculosis of the lungs coursed against the background of unchanged nonspecific reactivity of the organism.  相似文献

In this article is presented the result of the experiments on mice-hybrids F1(CBA x C57B1/6), which indicates the presence of the reaction of "ischemia/reperfusion" for stem cells of two "critical" cell renewal systems of organism (bone marrow and intestinal epithelium) during the irradiation under the conditions of hypoxic radioprotector application. The additional injection of the source of nitric oxide radicals-sodum nitroprusside (SNT) to the mice right after the irradiation under the conditions of hypoxic protection by serotonin, resulted the substantial increase of the survival rate of hematopoietic stem cells (registered by the methods of endogenous and exogenous colony forming in spleen) and stem cells of intestinal epithelium (registered by the method of intestinal "microcolonies"). The similar radioprotective effect was also registered during the test of survival rate of mice under tests of "bone marrow" and "intestinal" forms of radiation lethality that is evidence of the importance of the realization of phenomenon "ischemia/reperfusion" in the reaction of whole organism on the acute radiation injury. As SNP weakens the manifestation apoptosis and necrosis through competition with active forms of oxygen (AFO) during the period of "reperfusion" on basis of the found out phenomenon experimental model for studying mechanisms of stem cells damage in vivo induced by AFO and for the search of the modifiers weakening or strengthening such damage can be developed.  相似文献

Interaction of ATP and its derivative "ATP-LONG", the preparations raising general resistanse of an organism to physical stress, with the model phospholipid monolayer membrane of distearoylphosphatidylcholine has been investigated. It is shown, that these substances interact with a monolayer under partial embedding of a purine base between the phospholipid molecules. The antitumor and toxic effects of methylnitrosourea (MNU) are determined through its metabolic pathways. In organism MNU is subject to hydrolytic decomposition and denitrosation. It has been shown in vivo studies that MNU abdominal injections of therapeutic doses caused the inhibition of ribonucleotide reductase in mouse spleen, and therefore the DNA synthesis depress. The effect may apparently contribute to antitumor property of MNU. It has been estimated that destruction of M2 subunit of the enzyme is occurred. The relation between the loss of ribonucleotide reductase activity and the inhibition of protein synthesis was discussed. Besides, the cancerogenic and mutagenic properties of MNU were discussed as a result of imbalance of DNA precursor pools. Changes in contents of Fe(3+)-transferrin, ceruloplasmin, methemoglobin in blood and spleen of animals after MNU injections have been found. The changes were reversible after single MNU injection and became irreversible after multiple injections.  相似文献

Evidence has accumulated that cancer develops from a population of quiescent tissue committed/pluripotent stem cells (TCSC/PSC) or cells developmentally closely related to them that are distributed in various organs. To support this notion, stem cells (SC) are long lived cells and thus may become the subject of accumulating mutations that are crucial for initiation/progression of cancer. More important, they may maintain these mutations and pass them to the daughter stem cells. Therefore, mutations that occur in normal SC, accumulate during the life of an organism at the clonal level in the stem cell compartment committed to a given tissue/organ. As a consequence, this may lead to the malignant transformation of SC and tumor initiation. Furthermore, many biological features of normal and cancer SC such as the physiological trafficking of normal and metastasis of cancer stem cells involve similar molecular mechanisms, and we discuss these similarities here. Therefore, looking both at the origin and behavioral aspects we can envision cancer SC being normal SC "Jedi" that went over to the "dark side".  相似文献
