Some medicinal plants need to be cultivated commercially in order to meet the ever-increasing demand for medicinal plants for the indigenous systems of medicine as well as for the pharmaceutical industry; in this regard, it seems significant to test the important medicinal plants for their salt-tolerance capacity, with a view to exploiting the saline lands for medicinal plant cultivation. Phyllanthus amarus plants were grown in the presence of NaCl in order to study the effect of NaCl (80 mM NaCl) in the induction of oxidative stress in terms of lipid peroxidation (TBARS content), H2O2 content, osmolyte concentration, proline(PRO)-metabolizing enzymes, and antioxidant enzyme activities. Groundwater was used for irrigation of control plants. Plants were uprooted randomly on 90 days after sowing (DAS). NaCl-stressed plants showed increased TBARS, H2O2, glycine betaine (GB), and PRO contents, whereas NaCl uptake decreased proline oxidase (PROX) activity and increased gamma-glutamyl kinase (gamma-GK) activity when compared to control. The antioxidant enzymes superoxide dismutase (SOD), peroxidase (POX) and catalase (CAT) were increased under salinity.  相似文献

Triadimefon (TDM), a systemic fungicide with non-traditional plant-growth regulator properties, was administered to Catharanthus roseus (L.) G. Don. plants in order to determine its effects on oxidative injury in terms of H2O2 content, lipid peroxidation (LPO), electrolyte leakage (EL), protein and amino acid contents, as well as proline metabolism. The LPO, estimated as thiobarbituric acid-reactive substances (TBARS), decreased under TDM treatment. It was found that H2O2 and EL were reduced under TDM treatment when compared to control. TDM treatment caused a significant increase in the protein and amino acid contents. Glycine betaine (GB) and proline (PRO) significantly accumulated in C. roseus under stress arisen from fungicide applications. Proline oxidase (PROX) activities reduce the PRO content and gamma-glutamyl kinase (gamma-GK) accelerates the synthesis of PRO. Under TDM treatment, the activity of PROX decreased and the gamma-GK activity increased. From our results, it is suggested that fungicide triadimefon causes activation of metabolic processes in the medicinal plant Catharanthus roseus. These findings are of great significance for the cultivation of this medicinal plant, as it was previously reported that TDM causes an enhancement of antioxidant metabolism and ajmalicine production in C. roseus.  相似文献

The activities of antioxidant enzymes viz. glutathione reductase, GR; superoxide dismutase, SOD; peroxidase, POD; catalase, CAT and glutathione-S-transferase, GST and alkaloid accumulation were investigated in leaf pairs (apical, middle, basal) and in roots of Catharanthus roseus seedlings under the conditions of different nitrogen sources (20 mM KNO(3) and 2 mM NH(4)Cl) and salinity, in the absence (non-saline control) and in the presence of 100 mM NaCl in the nutrient solution. Salinity caused a reduction in plant biomass. The biomass production of ammonium-fed plants was lower than that of nitrate-fed plants. The antioxidant enzymes exhibited higher activity in saline-treated plants. Changes in antioxidant enzyme activity caused by different nitrogen sources differed in all leaf pairs, as well as in roots of C. roseus. Ammonium-fed plants showed higher CAT, GR and GST activity in leaf pairs as well as in roots, while POD and SOD activity were higher in nitrate-fed plants. Higher peroxidase activity concomitant with the increased accumulation of alkaloid was found in all leaf pairs, as well as in roots of C. roseus of NO(3)(-) fed plants as compared to NH(4)(+) fed plants.  相似文献

Nitric oxide (NO), a small diffusible, ubiquitous bioactive molecule, acts as prooxidant as well as antioxidant, and also regulates remarkable spectrum of plant cellular mechanisms. The present work was undertaken to investigate the role of nitric oxide donor sodium nitroprusside (SNP) and/or calcium chloride (CaCl(2)) in the tolerance of excised mustard leaves to salt stress. After 24h, salt stressed leaves treated with SNP and/or CaCl(2), showed an improvement in the activities of carbonic anhydrase (CA) and nitrate reductase (NR), and leaf chlorophyll (Chl) content, leaf relative water content (LRWC) and leaf ion concentration as compared with the leaves treated with NaCl only. Salinity stress caused a significant increase in H(2)O(2) content and membrane damage which is witnessed by enhanced levels of thiobarbituric acid reactive substances (TBARS) and electrolyte leakage. By contrast, such increases were blocked by the application of 0.2mM SNP and 10mM CaCl(2) to salt stressed leaves. Application of SNP and/or CaCl(2) alleviated NaCl stress by enhancing the activities of antioxidative enzymes viz. superoxide dismutase (SOD), catalase (CAT), peroxidase (POX), ascorbate peroxidase (APX) and glutathione reductase (GR) and by enhancing proline (Pro) and glycinebetaine (GB) accumulation with a concomitant decrease in H(2)O(2) content, TBARS and electrolyte leakage, which is manifested in the tolerance of plants to salinity stress. Moreover, application of SNP with CaCl(2) was more effective to reduce the detrimental effects of NaCl stress on excised mustard leaves. In addition to this, ameliorating effect of SNP was not effective in presence of NO scavenger cPTIO [2-(4-carboxyphenyl)-4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxide]. To put all these in a nut shell, the results advocate that SNP in association with CaCl(2) plays a role in enhancing the tolerance of plants to salt stress by improving antioxidative defence system, osmolyte accumulation and ionic homeostasis.  相似文献

To understand protective roles of nitric oxide against salt stress, the effects of exogenous sodium nitroprusside on activities of lipoxygenase, peroxidase, phenylalanine ammonialyase, catalase, superoxide dismutase enzymes, proline accumulation, and distribution of sodium in soybean plants under salt were determined. Application of sodium nitroprusside + bacterium enhanced plant growth-promotion characteristics, activities of different enzymes, and proline accumulation in the presence of sodium nitroprusside under salt stress. Treatment with NaCl at 200 mM and sodium nitroprusside (0.1 mM) reduced Na+ levels but increased K+ levels in leaves in comparison with the NaCl-treated plants. Correspondingly, the plants treated with exogenous sodium nitroprusside and NaCl maintained a lower ratio of [Na+]/[K+] in NaCl-stressed plants.  相似文献

Growth, osmotic adjustment, antioxidant enzyme defense and the principle medicinal component bacoside A were studied in the in vitro raised shoot cultures of Bacopa monnieri, a known medicinal plant, under different concentrations of NaCl [0.0 (control), 50, 100, 150 or 200 mM]. A sharp increase in Na+ content was observed at 50 mM NaCl level and it was about 6.4-fold higher when compared with control. While Na+ content increased in the shoots with increasing levels of NaCl in the medium, both K+ and Ca2+ concentrations decreased. Significant reduction was observed in shoot number per culture; shoot length, fresh weight (FW), dry weight (DW) and tissue water content (TWC) when shoots were exposed to increasing NaCl concentrations (50–200 mM) as compared with the control. Decrease in TWC was not significant at higher NaCl level (150 and 200 mM). At 200 mM NaCl, growth of shoots was adversely affected and microshoots died under prolonged stress. Minimum damage to the membrane as assessed by malondialdehyde (MDA) content was noticed in the controls in contrast to sharp increase of it in NaCl-stressed shoots. Higher amounts of free proline, glycinebetaine and total soluble sugars (TSS) accumulated in NaCl-stressed shoots indicating that it is a glycinebetaine accumulator. About 2.11-fold higher H2O2 content was observed at 50 mM NaCl as compared with control and it reached up to 7.1-folds more at 200 mM NaCl. Antioxidant enzyme activities (superoxide dismutase, catalase, ascorbate peroxidase and guaiacol peroxidase) also increased with a rise in NaCl level. Increase in bacoside A, a triterpene saponin content was observed only up to 100 mM NaCl level. Higher salt concentrations inhibited the accumulation of bacoside A. It appears from the data that accumulation of osmolytes, ions and elevated activities of antioxidant enzymes play an important role in osmotic adjustment in shoot cultures of Bacopa under salt stress.  相似文献

The present study was carried out to examine the effects of seed soaking in 1 mM ascorbic acid (AA) or 1 mM proline on the growth, content of photosynthetic pigments and proline, relative water content, electrolyte leakage, antioxidant enzymes and leaf anatomy of Hordeum vulgare L. Giza 124 seedlings grown in greenhouse under 100 or 200 mM NaCl. The plants exposed to the NaCl stress exhibited a significant reduction in growth, relative water content, leaf photosynthetic pigments, soluble sugars, as well as alterations in leaf anatomy. However, the treatment with AA or proline ameliorated the stress generated by NaCl and improved the above mentioned parameters. NaCl increased electrolyte leakage, proline content, and activities of antioxidant enzymes (SOD, CAT, and POX). The antioxidant enzymes and leaf anatomy exhibited considerable changes in response to AA or proline application in the absence or presence of NaCl.  相似文献

Thermopsis turcica is distributed naturally in saline soils. Interestingly, how T. turcica can live in harsh salt conditions is unknown. To study its defense responses under salinity, T. turcica was grown in a medium containing 100 and 200 mM NaCl for 7 and 14 days. Physiological parameters, ion contents, reactive oxygen species accumulation, activities of antioxidant enzymes/isozymes, NADPH oxidase enzyme/isozyme, lipid peroxidation (TBARS) and osmolyte contents were investigated. Stress caused a rapid decline in relative growth rate, relative water content and chlorophyll fluorescence (F v/F m) under both NaCl treatments. These traits were more suppressed at 200 mM NaCl. The decline in osmotic potential (Ψ Π) with salinity increased the gradient for water flux into the cell and assisted in turgor maintenance. The increased membrane permeability under stress caused the entrance of excess Na+ and K+ into the cell. Stress decreased superoxide dismutase, catalase and peroxidase after 14 days of growth in 200 mM NaCl, whereas glutathione reductase (GR) increased throughout the experiment. While ascorbate peroxidase (APX) increased by 44 % at 7 days, it decreased after 14 days exposure to 200 mM NaCl. 200 mM NaCl caused the highest increase in TBARS at 14 days, indicating a decrease in OH· scavenging activity. Increasing concentrations of salinity caused an increase in glycine betaine (GB) and choline (Cho), though an increase in proline was only observed at 200 mM NaCl for 14 days. Briefly, H2O2 was more efficiently eliminated in 100 mM-treated plants by the ascorbate–glutathione cycle in which APX acts a strong catalyst together with GR. Salt stress-induced changes in antioxidant enzymes, lipid peroxidation, proline and glycine betaine contents, and proline-metabolizing enzymes were examined in the leaves of two mulberry cultivars (Local and Sujanpuri). With increasing salinity up to 150 mM NaCl, superoxide dismutase, catalase, ascor-bate peroxidase, guaiacol peroxidase, glutathione reductase, and monodehydroascorbate reductase activities were increased in both cultivars as compared to control, but more pronounced increase was observed in cv. Local. Salt stress enhanced the rate of lipid peroxidation (as indicated by increasing MDA content) in both cultivars. Under NaCl stress, cv. Local showed less change in the MDA content than cv. Sujanpuri. Salt stress resulted in a significant accumulation of free proline in mulberry leaves, and more accumulation was detected in cv. Local than cv. Sujanpuri. The leaves of cv. Local showed 9-fold accumulation of glycine betaine in comparision with cv. Sujanpuri after 20 days at 150 mM NaCl. A decrease in proline oxidase activity and an increase in γ-glutamyl kinase activity were observed with increasing NaClconcentration. The relative water content and electrolyte leakage also decreased after increasing the NaCl concentration, but a decrease was more pronounced in cv. Sujanpuri than in cv. Local. The results indicate that oxidative stress may play an important role in salt-stressed mulberry plants and cv. Local have more efficient antioxidant characteristics, which could provide for a better protection against oxidative stress.  相似文献

Leaves of 4-week-old (juvenile) and 9-week-old (adult) plants of the halophyte Mesembryanthemum crystallinum L. (the common ice plant), cultured under controlled conditions in the phytotron, were treated with paraquat (0.1 μM), which produces superoxide radical, and (or) paraquat combined with introduction of NaCl (100 mM) or proline (5 mM) into nutrient medium. After a 20-h dark period (23°C), plants were transferred into light (4 h at 54.1 W/m2 of photosynthetically active radiation) for stimulation of O°2 formation in plastids. Activities of antioxidant enzymes, the contents of MDA, H2O2, chlorophyll, and free proline were measured in leaves. Plant responses in two age groups, which differed in the type of photosynthesis (juvenile plants had C3 type of photosynthesis, whereas adult plants were at the transition stage to Crassulacean Acid Metabolism (CAM) photosynthesis), differed in the levels of constitutive proline and proline, induced by NaCl and paraquat, as well as in activities of superoxide dismutase (SOD) and catalase. Changes in SOD activity and proline accumulation in response to paraquat treatment combined with NaCl revealed opposite dependence to accumulation of proline: the more proline accumulated in leaves, the lower activity of the enzyme. In response to paraquat treatment, the content of chlorophylls a and b most drastically declined in juvenile plants. Negative effect of salinity on the content of chlorophylls was lower than that of paraquat and was almost the same in plants of both age groups. Protective effect of exogenous proline was most profound in the case of paraquat treatment. Exogenous proline decreased the rate of lipid peroxidation, the content of superoxide radical and, consequently, SOD activity (almost fivefold), and increased the content of chlorophylls (a and b) in leaves of adult plants. The obtained data suggest that stress-induced accumulation of proline in the common ice plant has both osmoprotectory and antioxidant functions.  相似文献

A sandy culture experiment was conducted to investigate the effects of exogenous CaCl2 on the indole alkaloid accumulation in Catharanthus roseus under salt stress. One-month seedlings of C. roseus were treated with the different concentrations of NaCl (0, 50, and 100 mmol l? 1) and 7.5 mmol l? 1 CaCl2. The plant samples were analyzed after 7 days of the treatments. The NaCl-stressed plants showed decrease of fresh and dry weight and increase of malondialdehyde (MDA) content compared to control. Tryptophan decarboxylase (TDC) activity increased significantly under 50 mmol l? 1 NaCl without CaCl2 addition, 50 mmol l? 1 NaCl with 7.5 mmol l? 1 CaCl2, and CaCl2 treatment without NaCl addition. There was a significant increase in peroxidase activity under NaCl stress compared to control. The vindoline, catharanthine, vincristine, and vinblastine contents increased under salt stress (especially with 50 mmol l? 1 NaCl treatment with or without CaCl2). Addition of CaCl2 to NaCl-stressed plants increased biomass, TDC activity, vindoline, and catharanthine contents and lowered MDA and vincirstine contents compared to the plants without CaCl2. The plants treated with CaCl2 alone showed higher TDC activity, vindoline, catharanthine, and vinblastine content when compared to control. The results showed that exogenous CaCl2 could promote the indole alkaloid metabolism under salt stress.  相似文献

研究了外源油菜素内酯(BL)对NaCl胁迫下棉花幼苗的钠累积、叶片生理特征及叶片差异基因表达水平的影响.结果表明: NaCl胁迫下2个棉花品种各部位钠含量升高,叶片丙二醛(MDA)、脯氨酸含量上升,叶绿素含量下降,叶片基因表达水平受到影响.外源施用油菜素内酯可降低NaCl胁迫下棉花幼苗根、茎、叶的钠含量,降低叶片中MDA含量,提高脯氨酸含量,并且NaCl胁迫下苏棉12号棉花品种更易受油菜素内酯调控.对苏棉12号的叶片进行数字表达谱分析结果表明,NaCl胁迫下苏棉12号叶片中的差异基因表达水平受到油菜素内酯调控,BL+NaCl处理棉花叶片的基因表达模式与CK(正常生长棉株)更为一致.说明外源油菜素内酯可减轻NaCl胁迫下棉花叶片受抑制程度,从而使NaCl胁迫下棉花叶片生理功能增强(叶绿素含量升高),最终使生物量增加.  相似文献

Glycinebetaine is one of the most competitive compounds which play an important role in salt stress in plants. In this study, the enhanced salt tolerance in soybean (Glycine max L.) by exogenous application of glycinebetaine was evaluated. To improve salt tolerance at the seedling stage, GB was applied in four different concentrations (0, 5, 25 and 50 mM) as a pre-sowing seed treatment. Salinity stress in the form of a final concentration of 150 mM sodium chloride (NaCl) over a 15 day period drastically affected the plants as indicated by increased proline, MDA and Na+ content of soybean plants. In contrast, supplementation with 50 mM GB improved growth of soybean plants under NaCl as evidenced by a decrease in proline, MDA and Na+ content of soybean plants. Further analysis showed that treatments with GB, resulted in increasing of CAT and SOD activity of soybean seedlings in salt stress. We propose that the role of GB in increasing tolerance to salinity stress in soybean may result from either its antioxidant capacity by direct scavenging of H2O2 or its role in activating CAT activity which is mandatory in scavenging H2O2.  相似文献

Antioxidant enzymes are related to the resistance to various abiotic stresses including salinity. Barley is relatively tolerant to saline stress among crop plants, but little information is available on barley antioxidant enzymes under salinity stress. We investigated temporal and spatial responses of activities and isoform profiles of superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX), non-specific peroxidase (POX), and glutathione reductase (GR) to saline stress in barley seedlings treated with 200 mM NaCl for 0, 1, 2, 5 days, respectively. In the control plant, hydrogen peroxide content was about 2-fold higher in the root than in the shoot. Under saline stress, hydrogen peroxide content was decreased drastically by 70% at 2 d after NaCl treatment (DAT) in the root. In the leaf, however, the content was remained unchanged by 2 DAT and increased about 14 % at 5 DAT. In general, the activities of antioxidant enzymes were increased in the root and shoot under saline stress. But the increase was more significant and consistent in the root. The activities of SOD, CAT, APX, POX, and GR were increased significantly in the root within 1 DAT, and various elevated levels were maintained by 5 DAT. Among the antioxidant enzymes, CAT activity was increased the most drastically. The significant increase in the activities of SOD, CAT, APX, POX, and GR in the NaCl-stressed barley root was highly correlated with the increased expression of the constitutive isoforms as well as the induced ones. We investigated the effects of exogenous application of jasmonic acid (JA) and nitric oxide (NO) on growth, antioxidant metabolism, physio-biochemical attributes and metabolite accumulation, in tomato (Solanum lycopersicum L.) plants exposed to salt stress. Treating the plants with NaCl (200 mM) resulted in considerable growth inhibition in terms of biomass, relative water content, and chlorophyll content, all of which were significantly improved upon application of JA and NO under both normal and NaCl-stress treatments. Salt treatment particularly 200 mM NaCl caused an apparent increase in electrolyte leakage, lipid peroxidation, and hydrogen peroxide production, which were reduced by exogenous application of JA and NO. Salt treatment triggered the induction of antioxidant system by enhancing the activities of antioxidant enzymes like superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX), and glutathione reductase (GR). Application of JA and NO separately as well as in combination caused a significant improvement in activities of SOD, CAT, APX, and GR activities. JA and NO either applied individually or in combination boosted the flavonoid, proline and glycine betaine synthesis under NaCl treatments. In conclusion, the exogenous application of JA and NO protected tomato plants from NaCl-induced damage by up-regulating the antioxidant metabolism, osmolyte synthesis, and metabolite accumulation.  相似文献

The effect of long-term (30 days) NaCl treatments (100 mM and 200 mM) on the activity of some antioxidant enzymes, level of antioxidant metabolites, water relations and chloroplast ultrastructure, was studied in potato (Solanum tuberosum cv. Désirée) leaves. Salt stress negatively affected relative water content, leaf stomatal conductance and transpiration rate. In treated plants, proline was enhanced, but there was a significant decrease in ascorbate and proteins. Total superoxide dismutase activity was increased. The isozyme patterns detected in native gels from salt-irrigated plants were not changed although all the isoforms appeared more heavily stained due to higher activity. In contrast, at both levels of NaCl, catalase activity decreased and ascorbate peroxidase activity showed no significant change in comparison with an untreated control. At ultrastructural level, only thylakoid swelling and a decrease in the amount of grana stacking was observed in treated plants. The overall behaviour of the antioxidant enzymes suggests an increase of cellular H2O2 that would contribute to the oxidative stress of potato plants, but which may be alleviated somewhat by the enhanced levels of proline.  相似文献

Salt stress causes oxidative damage and cell death in plants. Plants accumulate proline and glycinebetaine (betaine) to mitigate detrimental effects of salt stress. The aim of this study was to investigate the protective effects of proline and betaine on cell death in NaCl-unadapted tobacco (Nicotiana tabacum) Bright Yellow-2 suspension-cultured cells subjected to salt stress. Salt stress increased reactive oxygen species (ROS) accumulation, lipid peroxidation, nuclear deformation and degradation, chromatin condensation, apoptosis-like cell death and ATP contents. Neither proline nor betaine affected apoptosis-like cell death and G(1) phase population, and increased ATP contents in the 200mM NaCl-stressed cells. However, both of them effectively decreased ROS accumulation and lipid peroxidation, and suppressed nuclear deformation and chromatin condensation induced by severe salt stress. Evans Blue staining experiment showed that both proline and betaine significantly suppressed increment of membrane permeability induced by 200mM NaCl. Furthermore, among the ROS scavenging antioxidant defense genes studied here, mRNA levels of salicylic acid-binding (SAbind) catalase (CAT) and lignin-forming peroxidase (POX) were found to be increased by proline and betaine under salt stress. It is concluded that both proline and betaine provide a protection against NaCl-induced cell death via decreasing level of ROS accumulation and lipid peroxidation as well as improvement of membrane integrity.  相似文献

The effect of silicon on the growth, boron concentrations, malondialdehyde (MDA) content, lipoxygenase (LOX) activity, proline (PRO) and H2O2 accumulation, and the activities of major antioxidant enzymes [superoxide dismutase (SOD), catalase (CAT), and ascorbate peroxidase (APX)] and non-enzymatic antioxidants (AA) of wheat grown in soil originally with toxic B concentrations were investigated. Applied of 5.0 and 10.0 mM Si to the B toxic soil significantly increased Si concentration of the wheat and counteracted the deleterious effects of B on shoot growth. The contents of PRO, H2O2, MDA, and LOX activity of wheat grown in B toxic soil were significantly reduced by Si treatments. Compared with control plants, the activities of SOD, CAT, APX and content of AA were decreased by applied Si. Based on the present work, it can be concluded that Si alleviates B toxicity of wheat by preventing oxidative membrane damage and also translocation of B from root to shoot and/or soil to plant.  相似文献

Changes in tocopherol, chlorophyll and TBARS levels and the activities of antioxidant enzymes i.e., GSH-Px, GST, and SOD in chloroplasts of tomato plants subjected to moderate (50 mM) and severe (150 mM) NaCl stress were determined. Increase in tocopherol content around the second day under both stresses did not correlate with the chlorophyll degradation while such correlation was observed from the fifth day of severe stress. The activities of GSH-Px and GST as well as TBARS content showed NaCl-induced enhancement which was dose- and time-dependent. However, chloroplastic SOD was rather not involved in the response of tomato plants to NaCl stress. The obtained results suggest that under the moderate stress similarly as in the early phase of severe stress tocopherol functions as a typical antioxidant, while in the late phase of the latter it may be involved in senescence signaling pathway and enables the recovery and recycling of the compounds significant for a plant organism.  相似文献
