Microrheologic properties (aggregation and deformability) were examined in young (upper fraction) and old (bottom fraction) erythrocytes separated by density gradient centrifugation at 30000 g. A significant difference in microrheologic properties of the young and old erythrocytes was revieled. Erythrocyte aggregation was studied in the plasma and serum. Aggregation of the young and old cells in plasma was higher than in serum, which points to the importance of the presence of fibrinogen in the aggregation medium. The study of aggregation kinetics showed that the absence of fibrinogen in the medium produced an insignificant effect on the rate of aggregate formation, but it dramatically affected the degree of aggregation (12 and 88% difference between aggregate number in the plasma and serum, respectively). In young erythrocytes, the difference of the degree of aggregation and kinetics of aggregate formation in the plasma and serum was not so pronounced (32 and 43%, respectively). Analysis of correlations between aggregation and deformation of the young and old erythrocytes revealed more pronounced interrelations of these parameters in the plasma compared to serum, indicating an important role of fibrinogen for the manifestation of microrheologic properties of erythrocytes. These interrelations were most pronounced during aggregate formation, which supported the bridging theory of aggregation.  相似文献

Acetohydroxyacid synthetase activity of crude extracts ofBacillus subtilis grown in pH 7.0 minimal medium has a pH optimum of 7.5. However, the activity of extracts of cells grown in minimal medium of pH 6.0 shows a pH optimum of 6.5. Acetate or propionate induces formation of the pH 6.5 activity. Hydroxyapatite chromatography of a crude extract of cells grown in pH 7.0 medium shows one major and one minor peak of enzymatic activity. Both peaks have a pH optimum of 7.5–8.0. However, chromatography of an extract of cells grown in the presence of acetate reveals three peaks of activity: one major peak with a pH optimum of 6.5 and two minor peaks both having a pH optimum of 7.5–8.0.  相似文献

The phenomenon of flocculation in Azospirillum brasilense (MTCC-125) was studied under different combinations of carbon and nitrogen sources. Fructose and Potassium nitrate at a pH of 6.4 in the cultural medium favour a higher bio-floc production. The biofloc was studied for root adhesion and its survival efficiency in the rhizoplane and rhizosphere of certain crops such as sorghum and sunflower under dryland condition. It has been demonstrated that the flocculated cultures of Azospirillum were found to have maximum adhesion to the root surface and higher survival rate in the rhizoplane and rhizosphere under different moisture stressed conditions as compared to the log phase cells of Azospirillum.  相似文献

Oscillating nitrogenase activity in long lasting batch cultures ofAzospirillum brasilense ATCC 29145 is independent of the carbon source malate. With fumarate, succinate or pyruvate as sole carbon source nitrogenase activity is also oscillating. Cultivation in a medium with 20-fold the buffer concentration also results in oscillating nitrogenase activity. Nitrogen-fixing cultures ofAzospirillum brasilense ATCC 29145 excrete ammonia into the culture medium varying between 0.02 and 0.04 mM concentrations. This is not sufficient to cause a drop of nitrogenase activity inAzospirillum brasilense after the first maximum. During growth under nitrogen-fixing conditions with malate as carbon source, the cells excrete significant quantities of succinate into the culture medium. Cultures with only 0.05% malate reutilized the excreted succinate as soon as malate disappeared from the medium. Azospirillum brasilense ATCC 29145 is shown to have the capability of encystation. Encysted cells are different from vegetative cells in their resistance to desiccation, by the spherical shape and by immotility. The results indicate that oscillating nitrogenase activity in long lasting cultures reflects the development from vegetative cells to cysts and again to vegetative cells under microaerobic conditions.  相似文献

The gel-filtration behaviour of calf thymus histone fraction F2(b) was studied at three different salt concentrations (0.01m-, 0.10m- and 1.00m-sodium chloride) and two different pH ranges (pH3–4 and pH6.7–7.1). Other histone fractions [F1, F2(a) and F3] were also utilized to assist interpretation of the data. It was found that the Stokes radius of histone fraction F2(b) was not significantly changed when the salt concentration was increased, implying that the aggregation of the individual histone molecules (Edwards & Shooter, 1969) resulted in only relatively minor changes in the hydrodynamic volume. Aggregation would appear to be due to the salting out of hydrophobic regions giving rise, in the aggregate, to a compact core of hydrophobic groups from which protrude the remaining basic parts of the molecule. Repulsion between charged groups on the basic regions of individual histone molecules would give the aggregate approximately spherical symmetry, the diameter of the aggregate approximating to the length of a single histone molecule.  相似文献

Aggregative growth of non-slime-forming strains of Zoogloea ramigera was induced by growing the organisms at a depressed pH. Calcium and magnesium ion was found to reverse aggregative growth of the organisms. Conversely, aggregation was stimulated when the available inorganic cation concentration of the growth medium was lowered by the use of a chelating agent. The aggregative effects of pH depression or cation depletion and the dispersal effects of cation supplementation were observed only during cellular growth. The data suggest that aggregate formation of non-slime-forming strains of Z. ramigera may be related to the calcium or magnesium metabolism of the organisms, or both.  相似文献

We studied the effects of cAMP on phosphohydrolase activity in the microsomal fraction and its 0.5 M KCl extract. In both preparations, cAMP inhibited the activity of acid phosphatases. In the microsomal fraction, a 50% inhibition of acid cation-dependent (Mg2+, Cu2+) phosphatase was induced at pH 6.0 by 10 M cAMP; whereas, for cation-independent phosphatase, this effect was exerted only by 50 M cAMP. In the KCl extract, the sensitivity of both cation-dependent and cation-independent acid phosphatases to cAMP was higher than in the microsomal fraction. Cyclic AMP did not reduce the activity of nucleosidases. Selectivity in the cAMP action on acid phosphatases is assumed to be one of the mechanisms controlling the biochemical activity outside the barley root cells.  相似文献

Escherichia coli O111 is an emerging non-O157:H7 serotype of Shiga toxin-producing E. coli (STEC). We previously reported that outbreak and environmental, but not sporadic-case, strains of STEC O111 share a distinct aggregation phenotype (M. E. Diodati, A. H. Bates, M. B. Cooley, S. Walker, R. E. Mandrell, and M. T. Brandl, Foodborne Pathog Dis 12:235−243, 2015, http://dx.doi.org/10.1089/fpd.2014.1887). We show here the natural occurrence of nonaggregative variants in single STEC O111 strains. These variants do not produce curli fimbriae and lack RpoS function but synthesize cellulose. The deletion of csgBAC or rpoS in an aggregative outbreak strain abolished aggregate formation, which was rescued when curli biogenesis or RpoS function, respectively, was restored. Complementation of a nonaggregative variant with RpoS also conferred curli production and aggregation. These observations were supported by Western blotting with an anti-CsgA antibody. Immunomicroscopy revealed that curli were undetectable on the cells of the nonaggregative variant and the RpoS mutant but were present in large quantities in the intercellular matrix of the assemblages formed by aggregative strains. Sequence analysis of rpoS in the aggregative strain and its variant showed a single substitution of threonine for asparagine at amino acid 124. Our results indicate that the multicellular behavior of STEC O111 is RpoS dependent via positive regulation of curli production. Aggregation may confer a fitness advantage in O111 outbreak strains under stressful conditions in hydrodynamic environments along the food production chain and in the host, while the occurrence of nonaggregative variants may allow the cell population to adapt to conditions benefiting a planktonic lifestyle.  相似文献

Summary A soluble extract from purified cell walls of C. reinhardii has been separated by gel filtration into three fractions which together account for 94% of the cell wall. The major fraction (accounting for 70% of the extract) is a glycoprotein, with a molecular wt. in sodium perchlorate of 298,000, which can be split into 4 electrophoretically distinct species. It contains 35% protein with high levels of hydroxyproline, arabinose and galactose, and is capable of self assembly into crystalline structures identical to those found within the cell wall. The second fraction (25% of the extract) is a similar glycoprotein, but contains 24% protein, a higher proportion of mannose, and is incapable of self assembly. Summary Growth and ethanol production by three strains of the bacterium Zymomonas mobilis were tested, at 40°C, in a medium containing cellulose hydrolysate (hexose fraction) as carbon source. The thermotolerant mutant C107 exhibited the best growth compared to wild type ZM4 and to the osmotolerant mutant SBE15. When cultivated in media supplemented with various nutrients, growth was only observed in presence of yeast extract (10 g/l) which acted both as a vitamin supplier and pH stabilizer. Using calcium pantothenate instead of yeast extract and sodium acetate to control pH resulted in growth inhibition by the high medium osmolality. Batch fermentation with pH control (KOH addition) showed good growth and ethanol production with the mineral medium.  相似文献

To identify clinically relevant parameters of red blood cell (RBC) aggregation, we examined correlations of aggregation parameters with C-reactive protein and fibrinogen in unstable angina (UA), acute myocardial infarction (AMI), and bacterial infection (BI). Aggregation parameters were derived from the distribution of RBC population into aggregate sizes (cells per aggregate) and changing of the distribution by flow-derived shear stress. Increased aggregation was observed in the following order: UA, AMI, and BI. The best correlation was obtained by integration of large aggregate fraction as a function of shear stress. To differentiate plasmatic from cellular factors in RBC aggregation, we determined the aggregation in the presence and absence of plasma and formulated a "plasma factor" (PF) ranging from 0 to 1. In AMI the enhanced aggregation was entirely due to PF (PF = 1), whereas in UA and BI it was due to both plasmatic and cellular factors (0 < or = PF < or = 1). It is proposed that clinically relevant parameters of RBC aggregation should express both RBC aggregate size distribution and aggregate resistance to disaggregation and distinguish between plasmatic and cellular factors.  相似文献

Summary The involvement of Concanavalin A (ConA), -methylmannose and glucuronic acid in the reaggregation of fresh water sponge cells has been tested. Glucuronic acid has been found to be the potent inhibitor and the indication is that this group is involved in the binding of aggregation factor to the cell surface. When cells were washed with EDTA and periodate to inactivate the aggregation factor, a lag phase of 5–6 h was required for the cells to recover the adhesive capacity. Puromycin does not inhibit this recovery process of EDTA washed cells. Con A, -methylmannose and glucuronic acid interfere with this recovery of adhesion and the cells remain nonaggregative, probably due to their interference with the insertion of aggregative molecules on the cell surface.  相似文献

Aggregation of suspended HeLa cells is increased on removal of cell surface sialic acid. Calcium ions promote aggregation whereas magnesium ions have no effect. The calcium effect is abolished by previous treatment of the cells with neuraminidase. Trypsinization of the HeLa cells followed by thorough washing diminishes the rate of mutual cell aggregation. Subsequent incubation with neuraminidase restores the aggregation rate to the original value before trypsin treatment. Cells which had acquired a greater tendency for aggregation after removal of peripheral sialic acid lose this property when subsequently treated with trypsin. Calcium ions have no aggregative effect on trypsinized cells. In contrast to HeLa cells, aggregation of human erythrocytes was not increased after treatment with neuraminidase or on addition of calcium. The results with HeLa cells are interpreted as follows: (a) Trypsin-releasable material confers adhesiveness upon the cells. (b) The adhesive property of this material is counteracted by the presence of cell surface sialic acids. (c) Calcium ions exert their effect by attenuating the adverse effect of sialic acid.  相似文献

A crude mosquito larvae and dialysed extract alters the mitotic rate of several epithelial cell populations in normal young and adult hepatectomized mice. A crude extract also showed a biphasic effect on the proliferation of human mononuclear cells (MNCs), either stimulating or inhibiting them depending on the dose applied. In the present paper, we assayed the effect of the dialysed mosquito larvae extract and two different protein fractions on human MNCs. Analysis of cell viability after culture indicated that the extract did not have toxic effects. Our results show a dual response of the MNCs to the dialysed, as well as to the protein fraction, with the highest molecular weight inhibiting or stimulating proliferation, depending on the dose applied. The protein fraction with the lowest molecular weight (range between 12-80 kDa) showed only an inhibitory effect on cell proliferation.  相似文献

There is broad agreement that platelet aggregation is generally dependent on fibrinogen (Fg) binding to the glycoprotein (GP) IIb-IIIa receptor expressed on the activated platelet surface. We therefore compared rates and extents of aggregation and of fibrinogen receptor expression and specific Fg binding to activated platelets, as a function of ADP concentration. Human citrated platelet-rich plasma (diluted 10-fold) was stirred with adenosine diphosphate (ADP) for 10 s or 2 min to measure rates and extent of aggregation, respectively, determined from the decrease in the total number of particles. The number of fibrinogen receptors and bound Fg were measured from mean fluorescence values obtained with FITC-labeled IgM monoclonal antibody PAC1 and the IgG monoclonal antibody, 9F9, respectively, using flow cytometry as presented in part I (Frojmovic et al., 1994). Because flow cytometric and aggregation measurements were routinely determined at room temperature and 37 degrees C, respectively, we also compared and found temperature-independent initial rates of aggregation. The fraction of platelets with fluorescence values above one critical threshold value, corresponding to maximally "activated" platelets (P*), increased with increasing activator concentration and correlated linearly with the fraction of platelets recruited into aggregates for ADP (r > 0.9). Aggregation was not rate-limited by fibrinogen receptor expression or by Fg binding. It appears that each platelet expresses its maximal Fg receptors at a critical ADP concentration, i.e., occupancy of ADP receptors. This, in turn, leads to rapid Fg occupancy and capture of such "quantally activated" platelets into aggregates.  相似文献

Mosquito larvae crude extract have been found to alter the mitotic rate of several mouse epithelial cell populations such as enterocytes and tongue keratinocytes. Also, the dialysed fraction inhibits hepatocyte proliferation in hepatectomized males. These experiments suggested an inhibitory effect on the G1/S interphase. Consequently, we suggested the presence of some molecule or molecules related to the TGF-beta superfamily. In the present paper, we have assayed the crude extract on human mononuclear cells and the dialysed fraction of the extract on tongue keratinocyte proliferation. Furthermore, different protein fractions obtained using a molecular exclusion chromatographic column were assayed on hepatocyte proliferation of hepatectomized mice. Three groups of proteins have been isolated. Results show a dose-dependent effect of crude extract on mononuclear cell proliferation and the dialysed extract caused an inhibitory effect on tongue keratinocyte proliferation. With regard to the hepatocyte mitotic rate, an inhibitory effect appeared only in animals receiving the fraction with lower molecular weight. These results suggest the presence in mosquito larvae of some peptidic molecule or molecules resembling the activity of members of the TGF-beta superfamily.  相似文献

Preservation of cell aggregation is necessary for thyroid follicular differentiation in vitro and requires stimulation by thyrotropin (TSH). We have tested the hypothesis that TSH preferentially increases thyroid cell-cell adhesion relative to cell-substrate adhesion. Cell-cell adhesion was measured in short-term suspension cultures by the decrease in the fraction of single cells remaining in culture (free cell ratio, FCR). When incubated in medium alone freshly isolated cells showed a progressive fall in FCR but this was accelerated by TSH and the cyclic AMP analog, 8-(4-chlorophenylthio)cyclic AMP. Aggregation was dependent upon extracellular Ca2+ and also promoted by a cell-free membrane extract. In contrast, attachment of cells to plastic dishes treated for tissue culture was not affected by TSH. We conclude that thyroid cells possess a TSH-sensitive cell adhesion system. The preferential increase in cell-cell adhesion may be one mechanism by which TSH stimulates the formation and preservation of follicles in vitro.  相似文献

Background

The current study demonstrated the possibility of statistical design tools combination with computational tools for optimization of fermentation conditions for enhanced fibrinolytic protease production.

Methods

The effects of using different carbon and nitrogen sources for protease production by Streptomyces radiopugnans_VITSD8 were examined by a full factorial design method. The incubation time, temperature, pH of the medium, and RPM were assessed by the predictable one factor at a time (OFAT) method. Optimization was carried out using starch and oat meal as carbon source, nitrogen source as peptic and malt extract using Fractional Factorial Design (FFD). The analysis was further continued for medium volume, temperature, initial medium pH, inoculum concentration, high determination co-efficient as (R’-0.965), and lower determination co-efficient of variation (CV-8.19%), which defines a reliable and accurate experimental value.

Results

Analysis of variance by the fixed slope effect by temperature and starch; temperature and L-aspargine, temperature and oat meal, temperature and peptic extracts, temperature and pH, temperature and duration of incubation were more vital for protease production at an interactive level. Response surface plots revealed that temperature, starch, and peptic extracts affix critical concerning in temperature. Programming estimated a 28% increase in protease production. Incubation temperature and medium volume portrayed extreme impact among all factor. Starch, peptic and temperature play an important regulatory role in protease production. Optimium temperature for protease production was 33°C. The ratio of carbon and nitrogen sources and pH were the major regulatory factors in protease production by Streptomyces radiopugnans_VITSD8. It demonstrated a 4% noteworthy change in condition.

Conclusion

Among all the selected parameters, temperature was the most intuitive factor, demonstrating a notable connection with the type of media and pH, while inoculum fixation had a direct impact on protein production.

Poliovirus and reovirus were found to aggregate into clumps of up to several hundred particles when diluted 10-fold into distilled water from a stock preparation of minimal aggregation in 0.05 M phosphate buffer, pH 7.2, plus 22 to 30% sucrose. Reovirus was also found to aggregate when diluted into phosphate-buffered saline. The aggregation was concentration dependent and did not occur when either virus was diluted into water 100-fold or greater. The aggregation of poliovirus was reversible by further addition of saline and produced a dispersed preparation of virus. Reovirus aggregation was not reversible. Both viruses aggregated when diluted into buffers at pH 5 and 3, and poliovirus aggregated at pH 6, and this aggregation of both viruses was reversible when returned to pH 7. Aggregation did not occur at alkaline pH values. Aggregation at low pH could be caused aggregation of either virus at pH 7. Calcium ions, however, were found to aggregate both viruses at a concentration of 0.01 M.  相似文献
