Two populations of Epistylis wuhanensis n. sp., a new freshwater peritrich ciliate, were isolated from different freshwater ponds located in Hubei, China. Their morphological characteristics were investigated using live observation, protargol impregnation, and scanning electron microscopy (SEM). Specimens from the two populations showed identical arrangement of the infraciliature and identical small subunit ribosomal RNA (SSU rRNA) gene and ITS1‐5.8S‐ITS2 sequences. The zooids present bell‐shaped and 90–175 × 27–54 μm in vivo. Macronucleus is variable in shape and located in the middle of cell. Pellicle is usually smooth with 139–154 and 97–105 striations above and below the trochal band, respectively. SSU rRNA gene and ITS1‐5.8S‐ITS2 sequences of E. wuhanensis n. sp. did not match any available sequences in GenBank. Phylogenetically, E. wuhanensis n. sp. clusters with the other Epistylis within the family Epistylididae, but is distinct from the major clades of Epistylis. Above all, the morphological characteristics and molecular analyses support that the present Epistylis is a new species. Expanded phylogenetic analyses of sessilids based on both SSU rRNA gene sequences and ITS1‐5.8S‐ITS2 sequences reveal that the genus Epistylis consists of Epistylis morphospecies and taxonomic revision of the genus is needed.  相似文献

Recent molecular analyses of Dictyosphaerium strains revealed a polyphyletic origin of this morphotype within the Chlorellaceae. The type species Dictyosphaerium ehrenbergianum Nägeli formed an independent lineage within the Parachlorella clade, assigning the genus to this clade. Our study focused on three different Dictyosphaerium species to resolve the phylogenetic position of remaining species. We used combined analyses of morphology; molecular data based on SSU and internally transcribed spacer region (ITS) rRNA sequences; and the comparison of the secondary structure of the SSU, ITS‐1, and ITS‐2 for species and generic delineation. The phylogenetic analyses revealed two lineages without generic assignment and two distinct clades of Dictyosphaerium‐like strains within the Parachlorella clade. One clade comprises the lineages with the epitype strain of D. ehrenbergianum Nägeli and two additional lineages that are described as new species (Dictyosphaerium libertatis sp. nov. and Dictyosphaerium lacustre sp. nov.). An emendation of the genus Dictyosphaerium is proposed. The second clade comprises the species Dictyosphaerium sphagnale Hindák and Dictyosphaerium pulchellum H. C. Wood. On the basis of phylogenetic analyses, complementary base changes, and morphology, we describe Mucidosphaerium gen. nov with the four species Mucidosphaerium sphagnale comb. nov., Mucidosphaerium pulchellum comb. nov., Mucidosphaerium palustre sp. nov., and Mucidosphaerium planctonicum sp. nov.  相似文献

A new filamentous cyanobacterial species of the genus Brasilonema was isolated from the island of Oahu, Hawaii. The taxon is distinguished from the seven other species in the genus by attenuation of trichomes, and is here described as Brasilonema angustatum sp. nov. It possesses the cytoplasmic kerotimization and reddish‐brown coloration of several species in the genus. Phylogenetic analysis based on 16S rRNA gene sequence shows B. angustatum within a highly supported clade containing all sequenced Brasilonema species. We compared the secondary structure of the 16S–23S ITS regions for B. octagenarum and B. angustatum. The structurally conservative D1–D1′ and V3 helices show similar motifs between the two taxa, but differ structurally and in sequence, providing additional justification for erection of the new species. The Box‐B helix has identical secondary structure. The existence of tapering in a Brasilonema species is unique in this genus, and requires modification of the current concept of the genus Brasilonema, which was described as being unattenuated. Our phylogenetic evidence supports the hypothesis that tapering has developed repeatedly in separate cyanobacterial lineages and lacks the taxonomic significance once assumed by early workers.  相似文献

Microstomum lineare is a common species of fresh and brackish waters found worldwide. Three genes (18S, CO1 and ITS) were sequenced from specimens of M. lineare collected from four countries, and the levels of cryptic diversity and genetic structuring were assessed. Results showed M. lineare has very wide haplotype distributions suggesting higher than expected dispersal capabilities. In addition, three new species were described on the basis of molecular taxonomy: Microstomum artoisi sp. nov., Microstomum tchaikovskyi sp. nov. and Microstomum zicklerorum sp. nov.  相似文献

Erotini is a small tribe of net‐winged beetles known from the northern temperate zone. We investigated relationships and limits of tribes and genera using a molecular phylogeny inferred from rrnL, cox1 and nad5 mtDNA fragments. Lopheros Leconte, 1881 and Pseudaplatopterus Kleine, 1940 were inferred in current molecular analyses as terminal lineages within Erotini. Therefore, we consider Lopherotini Kazantsev, 2012 and Pseudaplatopterina Kazantsev, 2012 as junior synonyms of Erotini Leconte, 1881. The Platycis genus group comprises 19 species representing several genus‐group taxa previously placed in Platycis s. l. Konoplatycis Nakane, 1969 was recovered as a deeply rooted lineage of Erotini and we inferred another independent lineage for which we propose a new genus Sinoplatycis gen. nov. Additionally, we studied the diversity of Platycis s. l. in Asia, found their highest diversity in the temperate forest habitats of Eastern Asia and inferred the paraphyletic character of the subgenus Erotides s. str. Five new species are described from China: Sinoplatycis cardinalis sp. nov. , S. zhani sp. nov. (type species of Sinoplatycis gen. nov. ), Erotides brunnescens sp. nov. , E. pusillus sp. nov. and E. slipinskii sp. nov. The molecular phylogeny shows deep splits of Japanese species with respect to the continental Eurasian fauna and the role of shallow seas and mountain systems in the diversification of the Platycis genus group.  相似文献

Knowledge of the tropical terrestrial cyanobacterial flora from the African continent is still limited. Of 31 strains isolated from soil and subaerial samples collected in Lagos State, Nigeria, three were found to be in the Oculatellaceae, including two species in a new genus. Subsequently, isolates from microbial mats in White Sands National Park in New Mexico, United States, and from a rock near the ocean in Puerto Rico, United States, were found to belong to the new genus as well. Cyanobacterial isolates were characterized microscopically, sequenced for the 16S rRNA gene and associated ITS region, and phylogenetically analyzed. Egbenema gen. nov., with three new species, as well as two new species of Albertania were differentiated from all other Oculatellaceae. Both genera belong to a supported clade within the Oculatellaceae that includes Trichotorquatus and Komarkovaea. The two new species of Albertania, A. egbensis and A. latericola, were from the same sample, but were evolutionarily separate based on 16S rRNA gene phylogenies, percent identity below the 98.7% threshold, and ITS rRNA percent dissimilarity >7.0%. Egbenema aeruginosum gen. et sp. nov. was phylogenetically separated from Trichotorquatus and Albertania but was in a clade with other strains belonging to Egbenema. The two Egbenema strains from the United States are here named Egbenema epilithicum sp. nov. and Egbenema gypsiphilum sp. nov. Our results support the hypothesis that further species discoveries of novel cyanobacteria will likely be made in soils and subaerial habitats, as these habitats continue to be studied, both in tropical and temperate biomes.  相似文献

The complete life cycle and developmental stages of the fluke, Metorchis ussuriensis sp. nov. (Trematoda, Platyhelminthes), are herein described. The results of the present experiments showed that, for flukes from the Primorsky Region in the Russian southern Far East, the first intermediate hosts are the snails Parafossarulus spiridonovi and Boreoelona ussuriensis, and the second intermediate hosts are freshwater fish, tadpoles, and snails. The definitive host in this experiment was Anas platyrhynchos dom. Morphometric parameters of M. ussuriensis sp. nov. demonstrate similarities with Metorchis taiwanensis, but the two species differ in the sizes of their bodies, sizes of suckers of adult worms, and sizes of cercariae, as well as respective positions of the finfold in cercariae. Phylogenetic reconstructions and genetic distances using the cox1 gene sequences support the conclusion that M. ussuriensis sp. nov. is well distinguished from all other species of the genus Metorchis, while sequences of internal transcribed spacers (ITS1 and ITS2) failed to separate Mussuriensis sp. nov., Metorchis bilis, and Metorchis xanthosomus. In addition, we sequenced 1,402 bp of the 28SrRNA gene of Mussuriensis sp. nov. being the first 28S sequences in the genus Metorchis. Comparison to other trematodes suggests that 28SrRNA could proof suitable for the differentiation of trematode species.  相似文献

Two new species of the methanol assimilating ascosporic yeast genus Komagataella are described. Komagataella populi sp. nov. (NRRL YB-455, CBS 12362, type strain, MycoBank accession number = 564110) was isolated from an exudate on a cottonwood tree (Populus deltoides), Peoria, Illinois, USA, and Komagataella ulmi sp. nov. (NRRL YB-407, CBS 12361, type strain, MycoBank accession number = 564111) was isolated from the exudate on an elm tree (Ulmus americana), also growing in Peoria, Illinois. The species were resolved from divergence in gene sequences for domains D1/D2 LSU rRNA, ITS1-5.8S-ITS2, mitochondrial small subunit rRNA, RNA polymerase subunit 1 and translation elongation factor-1α. Species of Komagataella assimilate few carbon compounds and are unlikely to be resolved from differences in standard growth and fermentation tests. For this reason, separation of species is dependent on gene sequence analysis.  相似文献

The six synonyms currently accepted under Saccharomycodes ludwigii were investigated for by phenotypic properties, however, the sequence diversity of the rRNA and protein coding genes have not yet been determined. Nine strains including the type strains of synonyms of S. ludwigii deposited in the CBS yeast collection, Westerdijk Fungal Biodiversity Institute, Utrecht, The Netherlands, were analyzed using a multi-locus sequence analysis (MLSA) approach that included sequences of 18S ribosomal RNA (rRNA), the D1/D2 domains of the 26S rRNA, the ITS region (including the 5.8S rRNA) and fragments of genes encoding the largest subunit of the RNA polymerase II (RPB1 and RPB2) and translation elongation factor 1-α (TEF1). Our results showed that the nine strains have identical D1/D2, 18S and RPB2 sequences and similar ITS, RPB1 and TEF1 sequences, which indicated that they are conspecific. In addition, a novel species of Saccharomycodes, S. pseudoludwigii sp. nov. (type CGMCC 2.4526 T) that was isolated from fruit and tree bark in China, is proposed. The MycoBank number of this new species is MB 811,650.

Several new genera originally classified as the genus Phormidium, a polyphyletic and taxonomically complex genus within the Oscillatoriales, were recently described. The simple morphology of Phormidium does not reflect its genetic diversity and the delimitation of a natural group is not possible with traditional classification systems based on morphology alone. Therefore, this study used morphological, ecological, and molecular approaches to evaluate four populations morphologically similar to Ammassolinea, Kamptonema, and Ancylothrix (simple, curved, and gradually attenuated at the ends trichome), found in subtropical and tropical Brazilian regions. 16S rRNA gene sequences grouped all the strains in a highly supported clade with other two European strains isolated from thermal springs surrounding areas. The 16S‐23S ITS secondary structure corroborated the phylogenetic analysis with all the strains having similar structures. Consequently, a genetically well‐defined and cryptic new genus, Koinonema gen. nov., is proposed containing the aquatic, mesophilic, and morphologically homogeneous new species, Koinonema pervagatum sp. nov.  相似文献

Based upon COI‐5P, LSU rDNA, and rbcL sequence data and morphological characteristics, six new members of the noncalcified crustose genus of red algae Ethelia are described in a new family, Etheliaceae (Gigartinales), sister to the recently described Ptilocladiopsidaceae. The novel species are described from subtropical to tropical Atlantic and Indo‐Pacific Ocean basins; E. mucronata sp. nov. and E. denizotii sp. nov. from southern and northern Western Australia respectively, E. wilcei sp. nov. from the Cocos (Keeling) Islands of Australia, E. suluensis sp. nov. from the Philippines, E. umbricola sp. nov. from Bermuda and E. kraftii sp. nov. from Lord Howe Island, Australia. The generitype, Ethelia biradiata, originally reported from the Seychelles, Indian Ocean, is added to the Western Australian flora.  相似文献

One new genus and five new species of dragonflies (Odonata) are described and figured from the Yixian Formation of northeastern China, viz. two gomphaeschnids Sinojoria magna nov. sp. and S. cancellosa nov. sp., plus three progobiaeshnids Mongoliaeshna hadrens nov. sp., M. exiguusens nov. sp., and Decoraeshna preciosus nov. gen., nov. sp. These new discoveries confirm the apparently sudden great diversification in China of the clade Aeshnoptera during the Middle-Upper Jurassic, together with the Upper Mesozoic to modern lineages of dragonflies. At the same time, some ‘ancient’ groups of Odonata became extinct.  相似文献

Two populations of Rivularia‐like cyanobacteria were isolated from ecologically distinct and biogeographically distant sites. One population was from an unpolluted stream in the Kola Peninsula of Russia, whereas the other was from a wet wall in the Grand Staircase‐Escalante National Monument, a desert park‐land in Utah. Though both were virtually indistinguishable from Rivularia in field and cultured material, they were both phylogenetically distant from Rivularia and the Rivulariaceae based on both 16S rRNA and rbcLX phylogenies. We here name the new cryptic genus Cyanomargarita gen. nov., with type species C. melechinii sp. nov., and additional species C. calcarea sp. nov. We also name a new family for these taxa, the Cyanomargaritaceae.  相似文献

Five yeast morphotypes were isolated from biogas reactors at North Wyke Research, Okehampton, UK. Out of the five morphotypes, four were identified as known species. In contrast, the fifth morphotype strain, Bio10T, was found to differ from Bullera dendrophila and Kwoniella mangroviensis, its closest phylogenetic neighbours, by 2.6–2.9% with respect to the nucleotide sequence of the D1/D2 domain of the 26S rRNA gene and by 5.6–6.2% with respect to the internal transcribed spacer 1 (ITS1)-5.8S rRNA gene-ITS2 region. Bio10T also differs from these two species by a number of phenotypic characteristics. Thus, based on the phenotypic differences and phylogenetic analysis, strain Bio10T is assigned the status of a new species of Cryptococcus, for which the name Cryptococcus shivajii sp. nov. is proposed. The type strain is Bio10T (NCYC 3541T = CBS 11374T).  相似文献

Soil cyanobacteria are crucial components of biological soil crusts and carry out many functions in dryland ecosystems. Despite this importance, their taxonomy and population genetics remain poorly known. We isolated 42 strains of simple filamentous cyanobacteria previously identified as Pseudophormidium hollerbachianum from 26 desert locations in the North and South America and characterized these strains using a total evidence approach, that is, using both morphological and molecular data to arrive at taxonomic decisions. Based on a phylogenetic analysis of 16S rRNA gene sequences, we propose and characterize Myxacorys gen. nov. with two new species Myxacorys chilensis, the generitype, and Mcalifornica. We also found distinct 16S‐23S ITS sequence variability within species in our dataset. Especially interesting was the presence of two distinct lineages of M. californica obtained from locations in close spatial proximity (within a few meters to kilometers from each other) suggesting niche differentiation. The detection of such unrecognized lineage‐level variability in soil cyanobacteria has important implications for biocrust restoration practices and conservation efforts.  相似文献
