Azetidine-2-carboxylic acid (AZC), which occurs naturally inLiliaceous plants, is reported to be a proline (pro) analoguePlant cell walls contain ‘extensin’, which is richin hydroxyproline (hyp). Peptidyl hyp arises through hydroxylationof peptidyl pro followed by glycosylation (arabinose attachment)of hyp Because AZC replaces peptidyl prolyl residues, it maybe a useful tool for evaluating the significance of hyp-o-arabinoselinkages in cell elongation. Therefore, we determined the effectof AZC on [14C]pro uptake, incorporation and conversion to wall-bound[14C]hyp in relation to elongation of lily pollen tubes whosewalls consist, in part, of hyp-containing glycopeptides TheAZC suppressed pollen germination 9–42 per cent (1–10mM) and subsequent tube elongation 40–54 per cent (0·1–1mM without affecting respiration In contrast, similar hyp concentrationswere without effect on tube elongation Whereas uptake of [14C]prowas 16·5–6·2 per cent of the control at0·1–1 mM AZC, [14C]leucine uptake was 85–25per cent of the control. Light microscope radioautography revealedfewer silver grains over tubes elongated in 0·1–1mM AZC than in its absence. Incorporation of [14C]pro into tnchloroaceticacid (TCA)-precipitable cytoplasm was reduced by only 10 percent at 0·01–1 mM but 43 per cent at 10 mM AZCGel filtration of cytoplasm from pollen germinated without AZCbut with [14C]pro resulted in labelled void volume (V) and threeretarded peaks (RI–III) Incorporation into V and RI wasinhibited at both 0·01 and 1 mM AZC These AZC concentrationsreduced conversion of [14C]pro to wall-bound hyp by 20 percent However, total incorporation of [14C]pro into salt-water-purifiedwall fractions was suppressed 47–53 per cent (0·1–1mM AZC). Young plants of uniculm barley were grown singly in pots ina growth room at 23/21 °C, and an irradiance of 655 µEm–2s–1 during each 12 h photoperiod. At the fifth leaf stage,they were subjected to 80 h of continuous darkness during whichthe rates of CO2 efflux of vegetative shoot meristems, and maturefully expanded leaves, were separately monitored. Respiratoryefflux from the meristematic tissue was initially high, 12–15mg CO2 g–1 h–1, equivalent to a daily loss in weightof 20–25 per cent. It remained high, or even rose slightly,during what would have been the normal dark period, but thenfell sharply. Even so, it was still three times that of themature tissue at the end of the experimental period. The rateof CO2 efflux of the mature tissue began low, and fell evenfurther during the first 12 h of darkness. It then levelledoff at a rate of 2·0–2·5 mg CO2 g–1h–1, equivalent to a daily loss in weight of about 3 percent. It is suggested that the rate of ‘mature tissue’respiration, established after 12–24 h of darkness, mightbe a useful selection criterion to employ in attempts to increasethe total dry matter yield of the grass crop by breeding. Determination of Nitrate Reductase Activity in Barley Leaves and Roots   总被引:4,自引:0,他引:4  
The inactivation of nitrate reductase in the leaves and rootsof barley (Hordeum vulgare L. cv. Mazurka) during and afterextracting was investigated. At 0 °C in the absence of casein,25 per cent of ‘total’. i.e. maximal in vitro, nitratereductase activity was lost during the 2 min extraction process,followed by a slower loss of activity while the extract wasstored in ice. Activity was maintained by adding a minimum of1 per cent casein to the extraction medium containing 0·1M phosphate (pH 7·5), 1 mM EDTA and 1 mM dithiothreitol.Nitrate reductase was stable for several hours in these extracts,but declined in a first order manner in the absence of dithiothreitol.Casein also prevented the initial loss while making root extracts,but had less effect during storage. Using casein and thiols, nitrate reductase activity in light,(as product of maximal in vitro rates and wt g–1) in leaveswas 98 per cent of the total activity in 31-day-old plants grownwith full nutrient in water culture and 60-day-old field-grownplants receiving no fertilizer. Field-grown plants, however,exhibited only 17 per cent of the activity of culture-grownplants. Nitrate reductase in leaves of barley plants grown in waterculture had a diurnal rhythm. During the first 3 h of the lightperiod, activity increased to 1·3 x the ‘dark’value. This was followed by a temporary decrease and then byanother increase to a maximum of 1·7 x the ‘dark’value, occurring about 8 h after illumination. Lolium temulentum plants were grown at 20 °C, under an 8-hdaylength, in a controlled-environment chamber, and the kineticsof leaf expansion were observed by measuring the movement ofan optical grid attached to the fourth leaf. The leaf emerged23–24 d after sowing and was fully expanded 9–10d later. Extension rate was maximal between the second and fifthdays after emergence and declined markedly thereafter. Duringthe rapid growth phase the rate of elongation exhibited a distinctdiurnal rhythm, fluctuating between 1.9 to 2.3 mm h–1in the light period, and 1.3 to 1.7 mm h–1 in the dark.A circadian oscillation with a period of about 27 h was observedin leaves elongating in continuous darkness. When plants weretransferred to 5 °C soon after emergence of the fourth leafthere was an immediate reduction in rate of growth to about22 per cent of the rate at 20 °C: the Q10 for the mean elongationrate in the range 20–5 °C was 3.7. When plants weretransferred from 20 to 2 °C at fourth leaf emergence, meanextension rate declined to less than 5 per cent, correspondingto a Q10 in the range 5–2 °C of more than 300. Furthermore,growth at 2 °C was confined almost entirely to the darkphase of the photoperiod cycle. The responsive tissue was shownto be a small area of expanding leafless than 1.5 cm above theshoot apex and the possible mechanisms underlying low temperatureeffects in this region are discussed. Potassium transport has been studied in the marine euryhalinealga, Enteromorpha intestimlis cultured in seawater and in low-salinitymedium (Artificial Cape Banks Spring Water, ACBSW; 25·5mol m–3 Cl, 20·4 mol m–3 Na+, 0·5mol m–3 K+). K+ fluxes were measured using 42K+ and 86Rb+although 86Rb+ does not act as an efficient K+ analogue in thisplant. 42K+ experiments on seawater plants typically exhibiteda single protoplasmic exchange phase whereas 86Rb+ exhibitedtwo exchange phases. Compartmental analysis of 86Rb+ effluxexperiments on seawater-grown Enteromorpha plants were usedto deduce the intracellular partition of K+ between the cytoplasm(279±38 mMolal) and vacuole (405±68 mMolal). Theplasmalemma K+ flux in plants in seawater was greater in thelight than in the dark (563±108 nmol m–2 s–1versus 389±66·7 nmol m–2 s–1). Inlow-salinity plants, separate cytoplasmic and vacuolar exchangephases were apparent. Analysis of 42K+ efflux experiments onlow-salinity plants yielded a cytoplasmic K+ of 222±38mMolal and a vacuolar K+ of 82±11 mMolal. The plasmalemmaand tonoplast flux was 23±4·5 nmol m–2 s–1. The Nernst equation showed that, although K+ was close to electrochemicalequilibrium, active accumulation of K+ across the plasmalemmaoccurred in plants in seawater and ACBSW both in the light anddark. K+ was also actively transported inwards across the tonoplastin low-salinity plants. The electrochemical potential for K+across the plasmalemma ranged from 2·41±0·60kJ mol–1 in plants grown in seawater in the light to 5·79±0·87kJ mol–1 for plants in ACBSW in the light. Although K+is close to electrochemical equilibrium, the flux of K+ in plantsin both seawater and ACBSW media is high, hence the power consumptionof K+ transport is high. The permeability of K+ (PK+) was significantlyhigher in the light than in the dark in plants in seawater (about7·0 versus 2·5 nm s–1) but in plants inlow-salinity (ACBSW) medium the permeability was independentof light (about 12 nm s–1). Wheat plants were grown in a controlled environment with daytemperatures of 18 ?C and with 500 µ Einsteins m–28–1 of photosynthetically active radiation for 16 h. Beforeanthesis and 2 to 3 weeks after, rates of net photosynthesiswere measured for leaves in 2 or 21% O2 containing 350 vpm CO2at 13, 18, 23, and 28 ?C and with 500 µEinsteins m–2s–1 of photosynthetically active radiation. Also, underthe same conditions of light intensity and temperature, therates of efflux of CO2 into CO2-free air were measured and,for mature flag leaves 3 to 4 weeks after anthesis, gross andnet photosynthesis from air containing 320 vpm 14CO2 of specificactivity 39?7 nCi µmol–1. When the O2 concentration was decreased from 21 to 2% (v/v)the rate of net photosynthesis increased by 32 per cent at thelowest temperature and 54 per cent at the highest temperature.Efflux of CO2 into CO2-free air ranged from 38 per cent of netphotosynthesis at 13 ?C to 86 per cent at 28 ?C. Gross photosynthesis,measured by the 14C assimilated during 40 s, was greater thannet photosynthesis by some 10 per cent at 13 ?C and 17 per centat 28 ?C. The respiratory losses and the pattern of carbon supply froma leaf of unicuim barley were examined during a complete diurnalperiod using a steady state 14C-labelling technique. After a delay of c. 1 h a portion of the 14C exported from acontinuously assimilating leaf was lost in respiration in thelight. This respiratory loss amounted to c. 20% of the total14C fixed. A further 28% of the total 14C fixed was respiredduring the dark period. In the light, carbon was fixed at a rate of c. 8·9 mgC dm–2 h–1 and exported from the leaf at c. 5·3mg C dm–2 h–1. Dark export averaged c. 31% of theday-time rate. Carbohydrate was stored in the leaf during the day and was almostcompletely remobilized during the dark. Sucrose, the major reservecarbohydrate, was exported first whilst the starch level remainedconstant. After some 9 h of darkness, sucrose declined to alow level and starch remobilization began.  相似文献

The rates of 14CO2 fixation and translocation of 14C labelledassimilates were measured in field experiments at two timesof the day in two sugar-cane clones known to have differentrates of sucrose uptake in vitro but the same weight of leafper unit weight of cane. The rate of 14CO2 fixation and the velocity and rate of translocationwere significantly greater at both times in the clone with thehigher rate of sucrose uptake in vitro. The velocities of translocationwere 2.18 and 2.36 cm/min–1 for the clone with high sucroseuptake and 1.46 cm min–1 at both times in the clone withlow uptake. It is suggested that among sugar-cane clones the ability oftheir canes to store sugar may play a part in determining theirrates of photosynthesis and translocation.  相似文献

Duckweed, Lemna minor L., grown on inorganic nutrient solutionscontaining either NH4+ or NO3 as nitrogen source wasallowed to assimilate [1-14C]- or [2-14C]glycollate during a20 min period in darkness or in light. The incorporation ofradioactivity into water-soluble metabolites, the insolublefraction, and into the CO2 released was measured. In additionthe extractable activity of phosphoenolpyruvate carboxylasewas determined. During the metabolism of [2-14C]glycollate in darkness, as wellas in the light, NH4+ grown plants evolved more 14CO2 than NO3grown plants. Formate was labelled only from [2-14C]glycollateand in NH4+ grown plants it was significantly less labelledin light than in darkness. In NO3 grown plants formateshowed similar radioactivity after dark and light labelling.The radioactivity in glycine was little influenced by the nitrogensource. Amounts of radioactivity in serine implied that thefurther metabolism of serine was reduced in darkness comparedwith its metabolism in the light under both nitrogen regimes.In illuminated NH4+ plants, serine was labelled through a pathwaystarting from phosphoglycerate. After [1-14C]glycollate feedingNH4+ grown plants contained markedly more radioactive aspartateand malate than NO3 plants indicating a stimulated phosphoenolpyruvatecarboxylation in plants grown on NH4+. The rate of carbon transport from an old tomato leaf (54 days),grown at 80 W m–2, was measured under light flux densitiesbetween 7 and 90 W m–2. Under low light, the rate of carbontransport over a 6 h period was about 1 mg C dm–2 h–1,well in excess of the concurrent photosynthetic rate. The lossfrom these leaves of 14C-leaf assimilate which was fixed beforethe experimental period amounted to 62 per cent of the totalinitial uptake and was higher than that from leaves with higherconcurrent photosynthetic rates. The higher loss of 14C fromleaves with low photosynthetic rates was due to a greater contributionof 14C from the starch and residue fractions. The rate of transportappeared to be determined by the concentration of the labilesucrose, not the total sucrose concentration. In comparisonwith young fully-expanded tomato leaves (Ho, 1976) the sizeof the labile sucrose pool appeared to decrease with age. Thephotosynthesistranslocation coefficient was low (k1k2=0•21)for an old tomato leaf. Based on these results a scheme of carbonpartitioning in relation to translocation is proposed. The present study deals with structure and function of fourareas of Himalayan chir pine forest. Tree layer was monospecificon all sites with varied density and basal cover in the rangeof 540–1630 individuals per ha and 25·0–47·2m2ha–1, respectively. Shrubs having low density were sparselydistributed. All allometric equations relating to biomass ofdifferent components, to circumference at breast height (cbh)were significant, with the exception of that for cone biomass.Total vegetation biomass (115–236 t ha–1) was distributedas 113–283 t ha–1 in trees. 0·56–0·82t ha–1 in shrubs and 1·63–2·57 t ha–1in herbs. Total forest floor biomass including herbaceous litterranged between 9·6 and 13·6 t ha–1. Of thetotal annual litter fall (4·26–7·38 t ha–1),60·3–75·1 per cent was distributed in leaflitter and 24·9–39·7 per cent in wood litter.Turnover rate of tree litter varied from 0·45 to 0·53,whereas rates for shrubs and herbs were assumed to 1. Net primaryproduction of total vegetation ranged between 9·91 and21·2 t ha–1 year–1, of which the contributionof trees, shrubs and herbs was 76·5– 88·1per cent 0·6–1·8 per cent and 11·3–21·5per cent, respectively. A compartment model of dry matter onthe basis of mean data across sites was developed to show drymatter storage and flow of dry matter within the ecosystem. Carbon dioxide efflux from 5- to 20-day-old pea fruits was measuredfor plants grown in controlled environment at 15 °C and600 µmol s–1 m–2 photon flux density in a16 h photoperiod. The rate of CO2 output per fruit increasedquickly from 0.005 to 0.018 mg CO2 min–1 during fruitelongation and subsequently more slowly to 0.030 mg CO2 min–1as the fruits inflated. On a d. wt basis the rate was highest,0.175 mg CO2 g–1 min–1, in the youngest fruits anddeclined curvilinearly with increasing fruit weight to 0.02mg CO2 g–1 min–1. Separation of maintenance andgrowth components was achieved by starvation methods and bymultiple regression analysis. From the latter method estimatesof the maintenance coefficient declined hyperbolically from150±8.7 mg carbohydrate g–1 d. wt day–1 inthe very young fruits (0.05 g) to 10.4±0.36 mg carbohydrateg–1 d. wt day–1 in older fruits (2.0 g). On a nitrogenbasis maintenance costs decreased from 2240 to 310 mg carbohydrateg–1 nitrogen day–1 while nitrogen concentrationfell from 6.7 to 3 per cent d. wt. A simple linear relationshipbetween maintenance cost per unit d. wt and nitrogen concentrationwas not observed. A growth coefficient of 50±6.7 mg carbohydrate g–1growth (equivalent to a conversion efficiency, YG, of 0.95)was estimated for all fruits examined. The overall efficiency, Y, increased from a mean of 0.70 to0.85 during fruit elongation and subsequently declined to 0.80.For a given fruit weight, efficiency increased asymptoticallywith relative growth rate; both asymptote and slope of the relationshipincreased as the fruits grew. Phototropic responses were studied in light-grown mustard hypocotyls.Phototropism was induced by adding 0.27 µmol m–2s–1 unilateral blue light to a background of low pressuresodium (SOX) lamp light. Curvatures of some 6° from thevertical were reached by 60 min, the curvature rate between20 min and 60 min being 0.14° min–1. From the axialgrowth rate and tissue geometry the local growth rates of illuminatedand shaded sides of the hypocotyl were calculated to be 1.5and 4.5 µmin–1 respectively. Turgor pressures ofexpanding cells in control plants and in the shaded and illuminatedsides of the blue light illuminated hypocotyls were measuredto be 0.40-0.55 MPa with a pressure probe. No changes in turgorpressure were observed on initiation of curvature. The decayof pressure in the cells of non-transpiring plants followingexcision indicated that the yield stress threshold of the tissuemay be as low as 0.1 MPa. Seeds of 19 native British herbaceous species (14 grasses andfive forbs) were exposed to white light at three photon fluencerates: high (19–2 mol m–2 d–1), medium (9·6mol m–1 d–1) and low (2·3 mol m–2 d–2) These photon doses have been found by previous workers to inhibitgermination in several species. High and low photon doses wereapplied only as continuous light, but the medium dose was appliedas both continuous light and as a 12 h light/12 h dark photoperiod.All four treatments, plus a dark control, were carried out at15 °C; the high and low doses were also applied with a dailyalternation of 10/20 °C. The majority of species (15) fell into one of two groups. Inseven species germination was relatively high and consistentacross all treatments, including darkness; in the other eightspp germination was inhibited only in darkness. Mostly thesedata confirmed published results for the same species In contrast in Agrostis capillaris and A. stolonifera germinationwas high only at alternating temperatures, irrespective of photondose, but was also slightly promoted by a constant temperaturecombined with light/dark alternations. The influence of salinity in the growing media on ribulose-1,5-bisphosphate (RuBP) carboxylase and on CO2 fixation by intactsugar beet (Beta vulgaris) leaves was investigated. RuBP carboxylase activity was mostly stimulated in young leavesafter exposure of plants for 1 week to 180 mM NaCl in the nutrientsolution. This stimulation was more effective at the higherNaHCO2 concentrations in the reaction medium. Salinity also enhanced CO2 fixation in intact leaves mostlyat rate-limiting light intensities. A 60 per cent stimulationin CO2 fixation rate was obtained by salinity under 450 µEm–2 s–1. At quantum flux densities of 150 µEm–2 s–1 (400–700 nm) this stimulation was280 per cent. Under high light intensities no stimulation bysalinity was found. In contrast, water stress achieved by directleaf desiccation or by polyethylene glycol inhibited enzymeactivity up to fourfold at –1.2 MPa. Intact chloroplasts from young pea leaves were able to incorporate[3H]thymidine into DNA at relatively high rates (50 pmol mg–1chlorophyll h–1 or more), using light as the sole energysource. The intact plastids were also able to synthesize DNAin darkness, but only if ATP and MgCl2 (MgATP) were both present.The rates of MgATP-driven assimilation in darkness were equalto or greater than light-driven activity. Neither light nordithiothreitol pretreatments enhanced thymidine incorporationin darkness, suggesting that enzymes of chloroplast DNA (ctDNA)biosynthesis are not regulated via a thioredoxin-type system.Although exogenous nucleosides (other than [3H]thymidine) werenot an absolute requirement, dramatically elevated rates ofincorporation (over 300 pmol mg–1 chlorophyll h–1)were seen when adenosine, cytidine, guanosine and thymidinewere supplied in combination (500 mmol m–3 each). RadiolabelledDNA synthesized by the isolated chloroplasts was prepared usinga new heat extraction method. After digestion by restrictionendonucleases, ctDNA synthesized in organello was found to givetypical autoradiography patterns for chloroplast DNA. ExonucleaseIII studies suggested that 5% to 15% of the newly synthesizedDNA might be in a closed circular form. MgATP-driven synthesisin darkness was highly age-dependent. Chloroplasts from young(6 to 8-d-old) plants, or alternatively the youngest leavesof more mature plants, were 4–10 times more active thanthose from older tissues. Although these data do not establishconclusively that replication-type synthesis was occurring inthe isolated chloroplasts, they are consistent with this suggestion. The short-term dependence of NO3 uptake upon photosynthesisand sugar supply to the roots of soybean plants was investigatedin a series of experiments where CO2 availability, light intensityor conduction of phloem sap to the roots were severely limited.Removal of CO2 from the atmosphere or girdling of the stem equallyprevented the stimulation of NO3 uptake when plants weretransferred from darkness to the light. The effect of thesetwo treatments can be reversed by CO2 re-supply or by additionof 10 mM glucose in the nutrient solution, respectively. Glucosewas also more effective in stimulating NO3 uptake byintact plants in darkness than in light. Collectively, theseobservations are interpreted as evidence that the diurnal changesin NO3 uptake are due to decreased phloem transport ofphotosynthates in darkness. Accordingly, the magnitude of thesechanges was much dependent on starch accumulation in the leavesat the end of the photo-period. Shading the plants lowered thisaccumulation, and resulted in an amplification of the diurnalchanges in NO3 uptake. These results are discussed inconnection with the hypothesis that the carbon-dependent plasticityof the night/day ratio of NO3 uptake is an importantfeature of the co-ordination of the acquisition of N and C bythe plant. The effects of blue light (B) pretreatments on internode extensiongrowth and their possible interaction with phytochrome mediatedresponses were examined in Sinapis alba seedlings grown for11 d under 280 µmol m–2 s–1 of continuousblue-deficient light from low pressure sodium lamps (SOX). SupplementaryB (16 µmol m–2 s–1) caused no detectable inhibitionof the first internode growth rate under continuous SOX, butgrowth rate was inhibited after transfer to darkness. This effect,and the growth promotion caused by far-red bend-of-day' lightpulses were additive. The addition of B at 16 µmol m–2s–1 during 11 d, or only during the first 9 or 10 d orthe latest 0.75, 1 or 2 d of the SOX pretreatment caused approximatelythe same extent of inhibition after the transition to darkness.A single hour of supplementary B before darkness caused morethan 50% of the maximum inhibition. However, 24 h of lower fluencerates of B (4 or 7 µmol m–2 s–1) were ineffective.Covering the internode during the supplementary B period didnot prevent the response to B after the transition to darkness.Far-red light given simultaneously with B (instead of the SOXbackground) reduced the inhibitory effect of B. Above a given threshold fluence rate, B perceived mainly inthe leaves inhibits extension growth in subsequent darkness,provided that high phytochrome photo-equilibria are presentduring the irradiation with B. The effects of chloramphenicol and kinetin on uptake and incorporationof 35S-methionine and some 14C-amino acids have been investigatedin leaf-disks of Nicotiana rustica in light and dark. Chloramphenicolin a concentration of 1 mg per ml inhibits the uptake of aminoacids from 30 to 60 per cent compared with the water control.The incorporation of amino acids into bulk protein is stronglyinhibited in light (40 to 70 per cent), but only to a smalldegree in dark (10 to 20 per cent), as revealed also by 14CO2-photosynthesisof the disks and following treatment with chloramphenicol indark. The stimulating effect of kinetin on uptake and incorporationof amino acids is dependent upon its concentration (10–5to 10–6 M ; but 10–4 M solution inhibits stronglyboth uptake and incorporation). The stimulation seems to influencemore incorporation than uptake processes. Possible interactionsof chloramphenicol and kinetin in the protein metabolism oftobacco leaves have been discussed. Phaseolus vulgaris cv. Glamis plants grown at 7 and 28 W m–228 W m–2 in controlled environment cabinets showed copiousnodulation and high levels of acetylene reducing activity. Earlydifferences in nodulation were apparent before differences inphotosynthesis and were attributed to an effect of far-red lighton nodule development. Total plant nitrogen content was greater at 28 W m–2 thanat 7 W –2 but nitrogen content as a percentage of d. wtwas greater at the lower irradiance level. Total acetylene reducing activity (nmol. min–1 root–1)was greater at 28 W m–2 than at 7 W –2, but therewas no irradiance effect on specific activity (nmol. min–1g d. wt of pink nodules–1 or nmol. min–1 pink nodule–1). Transfer of 40-day-old plants from 7 W m–2 to 28 W m–2resulted in increased nodulesize(due toincreased size of infectedcells), accompanied by increased total, but not specific, acetylenereducing activity. Transfer of plants from 28 W m–2 to 7 W m–2 resultedin a fall of total acetylene reducing activity within 24 h,and senescence of large nodules. Specific acetylene reducingactivity was unaffected The results are interpreted as an effect of light on the productionof nitrogen fixing tissue, rather than on nitrogenase activity.  相似文献
