为了研究核盘菌的分类和生理分化问题,对供试的28个核盘菌株进行了可溶性蛋白质,芳香基酯酶和酸性磷酸酯酶同工酶电泳谱带以及紫外光吸收峰图形的试验研究,其结果,可分为五种类型。其中属于核盘菌属有四个种,即核盘菌Sclerotinia sclerotiorum(Lib.)de Bary(=Whetzelinia sclerotiorum(Lib.)Korf & Dumont),三叶草核盘菌S.trifoliorum Erikss.,细辛核盘菌(S.asari Wu et C.R.Wang和人参菌核病菌Sclerotinia sp。人参菌核病的另一分离菌和油菜菌核病的一个分离菌其谱带与灰葡萄孢(Botrytis cinerea)谱带相似,应归属于与葡萄孢属(Botrytis)相应的核盘菌。在核盘菌S.sclerotiorum种群中,南方菌系与北方菌系的可溶性蛋白质稍有差异,芳香基酯酶和酸性磷酸酯酶活性强度也稍有不同,可能存在生理分化现象。  相似文献

观察和测定了核盘菌(Sclerotinia sclerotiorum (Lib.) de Bary)弱毒株Ep-1PN单孢分离物的生长、菌落扩展和菌落形态等培养性状.结果发现来自7个子囊盘的1574个单孢分离物中有1560个分离物的培养性状与正常菌株的没有显著差异,与亲本相似的分离物只有6个,其它8个分离物介于弱毒株Ep-1PN与正常菌株之间.弱毒株Ep-IPN培养性状的有性遗传不遵循孟德尔核遗传规律.  相似文献

利用公共的真菌基因组数据库资源, 对核盘菌(Sclerotinia sclerotiorum)和灰葡萄孢(Botrytis cinerea)基因组中SSRs的结构类型、分布、丰度及最长序列等进行了系统分析, 并与已经研究过的禾谷镰孢菌(Fusarium graminearum), 稻瘟病菌(Magnaporthe grisea)和黑粉菌(Ustilago maydis)等几种植物病原真菌基因组中的SSRs进行了比较。结果表明: 核盘菌和灰葡萄孢基因组中的SSRs非常丰富, 分别为6 539和8 627个, 并且在结构类型和分布规律上具有一定的相似性; 与其他几种病原真菌相比, 核盘菌和灰葡萄孢基因组中长重复的四、五、六核苷酸基序更为丰富, 从而使得这两种真菌具有更高的变异性。同时, 我们发现真菌基因组中SSRs的丰度与基因组的大小及GC含量没有必然的关系。文章对核盘菌和灰葡萄孢基因组中SSRs的丰度、出现频率及最长基序的分析为快速、便捷地设计多态性丰富的SSRs引物提供了有益的信息。  相似文献

本文报道了采自人参(Panax ginseng C. A.Mey (保留名nom. Coiiscry))的核盘菌属一新种——人参核盘菌(Sclerotinla.Ginseng Wang C. R.,C. F. Chen et J. Chen)。该种在形态学以及可溶性蛋白、果胶(甲)酯酶和多聚半乳糖醛酸酶谱带等方面,均不同于已知种核盘菌(S. sclerotiorum (Lib.)de Bary),小核盘菌(S. minor Jagger)车轴草核盘菌(S.Trifoliorum Erikss.)和细辛核盘菌(S.Asari Wu et C.R. 南方红豆杉Taxus chinensis var.maimi内生真菌紫杉木霉Trichoderma taxi菌株ZJUF0986发酵液对15种常见植物病原真菌具有很强的抑菌活性;用石油醚、乙酸乙酯和正丁醇对发酵液进行活性物质的提取分离,并通过硅胶柱层析和制备HPLC纯化,得到3个活性代谢产物Ⅰ、Ⅱ和Ⅲ,其中主要活性代谢产物Ⅰ具有广谱高效抑制植物病原真菌的特点,特别是对灰葡萄孢Botrytis cinema、立桔丝核菌Rhizoctonia solani、核盘菌Sclerotinia sclerotiorum具有很强的抑菌活性,其IC50为1.1mg/L.  相似文献

菌核病(Sclerotinia stem rot,SSR)是我国油料作物生产中主要病害之一,严重制约长江中下游地区油菜主产区的产业发展。菌核病的病原是子囊菌门的核盘菌,是一种世界性分布的重要植物病原真菌。其寄主范围广泛,引起的菌核病对多种作物的产量和品质造成重要影响。核盘菌作为典型的死体营养型病原真菌,侵染寄主植物时通过直接杀死细胞和破坏组织攫取生长所需的营养物质。核盘菌的致病机理相对复杂,前期研究主要集中在其分泌的水解酶类(角质酶、细胞壁降解酶和蛋白酶等)和草酸在核盘菌侵染寄主植物中的作用。近些年来,越来越多的研究证实了分泌蛋白在核盘菌的致病过程中同样发挥着重要作用。分泌蛋白(效应蛋白)主要通过诱导植物细胞死亡或抑制寄主细胞的免疫反应,促进核盘菌的侵染和定殖。综述了水解酶类、草酸和分泌蛋白等在核盘菌致病机制中的作用,并对核盘菌致病机理研究进行了展望,以期为菌核病的安全防控提供理论参考。  相似文献

Aims:  To characterize the interaction of Sclerotinia sclerotiorum and S. minor with strains of the mycoparasite and commercial biocontrol agent Coniothyrium minitans using novel perfusion chamber gasket co-culture.
Methods and Results:  Sclerotinia were cultured in perfusion chamber gaskets and then flooded with Coniothyrium conidia. After germination, Coniothyrium failed to show any form of directed growth, making contact with Sclerotinia hyphae in a random manner. In turn, some Coniothyrium hyphae coiled round Sclerotinia counterparts and although no intracellular growth was observed, Coniothyrium proliferated, while the hyphae of Sclerotinia became vacuolated and lost the cytoplasm. When co-cultures of Sclerotinia with Coniothyrium were flooded with FITC-lectins, small difference in fluorescence between the fungi was found with FITC-Con A suggesting that cell walls of both the species exposed mannose. In contrast, Coniothyrium fluoresced poorly in comparison with Sclerotinia when FITC-wheat germ agglutinin was used, indicating a marked paucity of N -acetylglucosamine exposure by cell walls of Coniothyrium, hence reduced exposure to chitinolytic enzyme action.
Conclusions, Significance and Impact of the Study:  The approach employed supported direct sequential microscopic observation of Coniothyrium and Sclerotinia as well as the utilization of representative fluorescent moieties to characterize relative carbohydrate cell wall exposure.  相似文献

Electrophoretic karyotypes (EKs) of 83 isolates were variable within agricultural and natural populations of Sclerotinia sclerotiorum, as well as among S. sclerotiorum, Sclerotinia minor, and Sclerotinia trifoliorum. Variation in EKs was not observed within six mitotic or three meiotic lineages of isolates. EKs of 8 to 10 chromosome-sized DNAs were observed. There is confusion as to whether the Sclerotinia on Vicia faba is a normal strain of Sclerotinia trifoliorum as found on clover and related legumes or a distinct variety (var. fabae ). Pathologically the two fungi appear to be the same, and field observations indicate that beans in the rotation may aggravate the incidence of clover rot. A re-investigation of the morphology of the two fungi is made to assess the status of var. fabae.
The two fungi could not be distinguished by apothecial characters or cultural behaviour. Ascospore measurements confirm the existence of S. trifoliorum var. fabae , although 5. trifoliorum proper also occurs on beans. Var. fabae has not been found on any legume besides beans. Isolates from trefoil and sainfoin agree in spore size with S. trifoliorum on clover.
When isolates of S. trifoliorum are co-inoculated on the same Petri plate, they either do not react, or react to form a 'demarcation line'. Although a demarcation line is assumed to imply a genetic difference, non-reaction does not necessarily imply genetic identity. Non-reaction and the formation of a demarcation line also occur when var. fabae strains are co-inoculated together. When an isolate of S. trifoliorum is co-inoculated with an isolate of S. trifoliorum var. fabae an 'incompatibility line' is formed. Use can be made of this fact in identification.
On the basis of co-inoculations 5. trifoliorum and S. trifoliorum var. fabae are both shown to constitute a diverse range of biotypes. Segregation of biotypes in the ascus as a result of heterokaryosis in the ascus-initial is demonstrated. The view is expressed that heterokaryosis is the mechanism underlying the production of new biotypes.  相似文献

The relative saturations of the following fungicides, in dilute McIlvaine buffer containing sucrose, required for 50% inhibition of spore germination of Sclerotinia laxa were determined as under: phenylmercuric chloride, 0.0002; nystatin, 0.028; dichlone, 0.034; copper-8-quinolinolate, 0.039; 2:4-dichloro-6-( o -chloroanilino)-s-triazine, 0.131; captan, 0.132; bis-(5-chloro-2-hydroxyphenyl)methane, 0.234.
