Metabolic disturbances inNicotiana glauca, a symptomless carrier of potato witches’ broom were studied. The dry weight content of leaves of diseased plants was slightly decreased, that of the stems increased. The ash content in the dry weight of the whole diseased plant was decreased by 11%. Contrary to tomato plants which had shown considerable disproportions in the distribution of metabolites among the individual organs investigated (ULRYCHOVÁ, LIMBERK 1964), the content of the total nitrogen and of the individual nitrogen fractions of both leaves and stems of diseasedNicotiana glauca were decreased. The content of glutamine was increased in both organs in spite of the decrease of ammonia nitrogen by nearly 30% in both cases. The most striking disturbance found in the diseased plants ofNicotiana glauca was the 67% increase of a low-molecular phosphorus fraction (P extractable with 0-2N HCIO4) in the leaves. This result agrees with that found in diseased tomato plants and may be primarily associated with the virus infection. The content of all other phosphorus fractions of both leaves and stems was decreased.  相似文献

Enzyme-linked immunosorbent assay was used to measure the concentration of potato leafroll virus (PLRV) antigen in different parts of field-grown secondarily infected plants of three potato genotypes known to differ in resistance to infection. The antigen concentration in leaves of cv. Maris Piper (susceptible) was 10–30 times greater than that in cv. Pentland Crown or G 7445(1), a breeder's line (both resistant). Differences between genotypes in antigen concentration were smaller in petioles and tubers (5–10-fold) and in above-ground stems (about 4-fold), and were least in below-ground stems, stolons and roots (about 2-fold). PLRV antigen, detected by fluorescent antibody staining of tissue sections, was confined to phloem companion cells. In Pentland Crown, the decrease in PLRV antigen concentration in leaf mid-veins and petioles, relative to that in Maris Piper, was proportional to the decrease in number of PLRV-containing companion cells; this decrease was greater in the external phloem than in the internal phloem. The spread of PLRV infection within the phloem system seems to be impaired in the resistant genotypes. Green peach aphids (Myzuspersicae) acquired < 2800 pg PLRV/aphid when fed for 4 days on infected field-grown Maris Piper plants and < 58% of such aphids transmitted the virus to Physalis floridana test plants. In contrast, aphids fed on infected Pentland Crown plants acquired <120 pg PLRV/aphid and <3% transmitted the virus to P. floridana. The ease with which M. persicae acquired and transmitted PLRV from field-grown Maris Piper plants decreased greatly after the end of June without a proportionate drop in PLRV concentration. Spread of PLRV in potato crops should be substantially decreased by growing cultivars in which the virus multiplies to only a limited extent.  相似文献

Multiple components of the resistance of potatoes to potato leafroll virus   总被引:2,自引:0,他引:2  
In glasshouse experiments the ranking of potato genotypes for resistance to infection with potato leafroll virus (PLRV) using three concentrations of aphid-borne inoculum was the same as their field resistance ratings. In field-grown plants this resistance to infection increased in all genotypes as the plants aged but its rate of increase differed between genotypes. In tests on field-grown plants infected by aphid- or graft-inoculation, the proportion of virus-free progeny tubers increased the later the date of inoculation but was greater in resistant than in susceptible genotypes. This trend was most pronounced in the resistant clone G7445(1), in which the virus failed to move from the foliage to the tubers of some plants infected in glasshouse tests. The spread of PLRV will thus be minimised in crops of resistant compared with susceptible genotypes for three reasons: plants have greater resistance to infection, systemic spread of virus from their foliage to tubers is less likely and, as shown previously, the low concentration of virus particles in leaf tissue makes infected plants less potent sources of inoculum for aphids.  相似文献

Temperature both before and after aphid inoculation with potato leafroll virus (PLRV) greatly influenced the susceptibility of potato plants to infection and virus accumulation, as evaluated with ELISA using cultivars with different ratings for the resistance to PLRV. Pre-incubation at 15 compared to 27 °C increased the susceptibility of plants to infection and a subsequent PLRV accumulation. The virus was detected by ELISA in a greater proportion of plants and reached a higher concentration, when the plants were kept at 27 than at 15 °C after inoculation. The mean ELISA values obtained with PLRV-infected plants in the 15/27 combination of the pre-/post-inoculation temperatures over the period 1—6 wks after inoculation were significantly higher than those in the 27/27, 15/15 and 27/15 treatments, and the values obtained in the 27/27 treatment were significantly higher than those in the 15/15 and 27/15 ones. A hypersensitive-like intolerance reaction to PLRV occurred in the resistant cv. Irga only in the plants kept at 27 °C after inoculation.  相似文献

Methods are described for preparing potato mesophyll protoplasts that are suitable for infection with inocula of virus nucleoprotein or RNA. The protoplasts could be infected with four sap-transmissible viruses (tobacco mosaic, tobacco rattle, tobacco ringspot and tomato black ring viruses) and with potato leafroll virus, which is not saptransmissible. No differences were observed in ability to infect protoplasts with potato leafroll virus strains differing either in virulence in intact plants or in aphid transmissibility.  相似文献

Potato cultivars were tested for susceptibility to two British isolates of beet western yellows virus originally obtained from sugar beet and oil seed rape. Tre potato cultivars Erika and Krasava are unable to reproduce two strains of TMV systemically except when in graft symbiosis under normal greenhouse conditions with tomato which has been infected with the virus. The escape resistance of potato to TMV infection due to slight hypersensitivity was not changed either by long-lasting non-infectious symbiosis with TMV-sensitive tomato or in a constant environment (32±2 °C, 9 700 lx) which was adequate to change the hypersensitive reaction of some other TMV-hosts. On the contrary, enhanced temperature provoked a still more severe local and necrotic response in the potato varieties. This led to systemic necrosis and the quick death of potato components systemically infected in graft symbiosis with tomato under normal greenhouse conditions when transferred into an environment of enhanced temperature.  相似文献

Race T1 of Phytophthora infestans was first isolated in Central Europe. Tomato varieties carrying the dominant gene Ph were easily infected by T1 but showed a different degree of field resistance. In September 1967 more than 80% of all isolations from the experimental plots at Greifswald were determined as the aggressive race T1. This race was also isolated in a large screening test over several provinces of the GDR from potato and tomato fields where no plants carrying dominant resistance genes were present in the neighbourhood. The fungus was able to pass from potato leaves or tubers to detached tomato leaves or intact tomato plants (cv. Fanal) without any reduction in sporulation. The field-resistant tomato variety Ru?ový Ker and the highly field-resistant variety Atom were crossed. In F2 and subsequent generations, young plants were selected following inoculation of 14 mm leaf-discs. Weak sporulations on the leaf-discs were almost invariably correlated with a reduced level of infection of fruit in the field. Data from crosses of Atom with more or less susceptible varieties suggested the presence of incompletely dominant genes. It is concluded that at least two genes in the variety Atom control field resistance and the gene-symbols Phf and Phf-2 are proposed for them.  相似文献

By growing the excised apical meristems of sprouts from the potato varieties King Edward and Arran Victory, infected respectively with potato paracrinkle virus and potato virus S , virus-free plants were obtained. Although the method failed to produce virus-free plants from varieties infected with potato virus X , this virus also seems not to be present in apical meristems, for no virus could be demonstrated in callus tissue that developed from excised meristems less than 200 μ across. The concentration of tobacco mosaic virus in tomato roots and tobacco stems is also much less near the growing point than in older cells, but there is no evidence that the meristematic region is virus-free.  相似文献

Summary In a soil pot culture experiment conducted under greenhouse conditions, two varieties of cowpea (Vigna unguiculata (L) Walp) were found to be differentially susceptible to phosphorus-induced zinc deficiency. Although phosphate application, in the absence of applied zinc, caused growth disorder in both the varieties, the symptoms were more virulent, appeared early and were induced with both 25 and 75 ppm phosphorus in variety HFC-42-1, as against FOS-1, in which case zinc deficiency symptoms appeared under P75Zn0 treatment only. Contrary to the expectation the more susceptible variety was found to have higher zinc concentration in its shoots and thus the differential susceptibility of these varieties of cowpea could not be related to their zinc absorption characteristics. High phosphate application caused efflux of zinc during 16–32 day growth in both the varieties. However, higher sensitivity to phosphorus-induced zinc disorder in variety HFC-42-1, appeared to be due to nearly 2 times higher P concentration in this variety as against FOS-1. The results indicate that variety HFC-42-1 has a greater need for zinc because of its inherent capacity to accumulate more phosphorus. The relationship between inoculum density and mortality or infection was studied for various pepper varieties (Capsicum annuum L.) inoculated with zoospores of two P. capsici isolates. The inoculum concentrations required for 50% mortality (LD 50) varied greatly between pepper varieties and P. capsici isolates: with one isolate, LD 50 was 40 zoospores/ml for a susceptible variety and reached 4,380 to 97,300 zoospores/ml for resistant varieties. For another isolate, LD 50 for the, same varieties ranged from 26 to 800 zoospores/ml. Comparisons between LD 50 and inoculum doses required for 50 % Infection (ID 50) also revealed differences between varieties but not between isolates. After multiple infection correction, regression slopes of infections/inoculum concentration were low for resistant varieties (0.28 to 0.50) but higher for susceptible varieties (0.72 to 0.94), indicating strong competition between spores for infection of resistant plants, but not for infection of susceptible plants. This analysis provided many criteria which can be used to differentiate susceptible from resistant varieties and to evaluate with precision the resistance level of the different resistance genitors used in our breeding program.  相似文献

The susceptibility of stems of six potato cultivars to Erwinia carotovora subsp. atroseptica was assessed in two years (1981 and 1982) either by direct inoculation in the field or by inoculation of detached stems in the laboratory. These six and a further 22 cultivars were also assessed in three years (1982-84) by inoculating stems of glasshouse-grown plants. Different methods of inoculation and types of inocula were tested. In the field, wooden toothpicks rubbed in bacterial slime were more successful in establishing infection than when dipped in a bacterial suspension, but injection of bacterial suspension with a hypodermic needle was reliable in establishing infection over a range of concentrations. Detached stems were more readily infected and gave more consistent results compared with inoculation in the field. The range of reaction of the six cultivars was similar in both detached stem and glasshouse tests. The early cultivars Pentland Javelin and Ulster Sceptre were most susceptible and of the maincrop cultivars, Maris Piper was intermediate and Desiree and King Edward least susceptible whereas Pentland Crown showed greater resistance in the glasshouse than in the field. Glasshouse tests using hypodermic inoculation indicated a range of susceptibilities; the early cultivars Manna, Maris Bard and Estima were most susceptible and the maincrop Pentland cultivars Crown, Dell, Hawk, Ivory and Squire least susceptible.  相似文献

The relationship between age-related resistance of peper plants to Phytophthora capsici and contents of carbohydrates, amino acids, phenolics and mineral nutrients in pepper stems was studied using two pepper cultivars, Hanbyul (susceptible) and Kingkun (resistant). With increasing age of pepper plants, the two cultivars, which differ in their susceptibility to Phytophthora blight, became gradually resistant to the disease. The cultivar Kingkun distinctly showed the age-related resistance to Phytophthora blight at the second branch stage. The weight of dry matter in healthy stems of pepper plants at the second branch stage was twice that at the six leaf stage. The resistant cultivar Kingkun contained lower levels of fructose, glucose and sucrose in stems than the susceptible cultivar Hanbyul at the different developmental stages. No consistent differences between the developmental stages of the plants were recognized with regard to their glucose content. However, the contents of fructose and sucrose in the cultivar Hanbyul greatly increased at the second branch stage. The levels of inositol reduced in both pepper cultivars during plant development. In view of the fact that there were only slight changes in the amount of total amino acids, it seems unlikely that there is a relationship between the amino acid metabolism and the retardation of Phytophthora infection during plant development. The amounts of total phenolic compounds in pepper stems were relatively low at the later growth stages of the plants and also in the resistant cultivar Kingkun. The contents of macroelemental nutrients such as nitrogen, phosphorus, potassium, calcium and magnesium were drastically reduced in pepper stems at the later plant growth stage. No significant differences between the cultivars or the plant growth stages were found in the silicon and microelemental nutrients such as sodium, iron, zinc and manganese. These results suggest that the expression of age-related resistance of pepper plants may be due to the morphological and nutritional changes in tissues of pepper stems during ageing, i.e. the pronounced increase in weight of dry matter, the significant decrease in amounts of mineral nutrients such as nitrogen, phosphorus, potassium, calcium and magnesium, and the tow contents of fructose, glucose and sucrose in the stem tissues.  相似文献

Plants of two potato clones which, in preliminary greenhouse assessments, showed resistance to multiplication and accumulation of potato leafroll virus (PLRV) were graft or aphid inoculated with the virus and grown in the greenhouse; plants of a moderately susceptible cultivar were used for comparison in all experiments. A high concentration of aphid‐borne inoculum was used to ensure strong infection pressure. Clone M62759 appeared to be highly resistant to PLRV infection, whereas clone PS1706 was more susceptible. Both clones expressed a high level of resistance to virus multiplication, when primary or secondary infection was assayed by enzyme‐linked immunosorbent assay. Moreover, PLRV was detected in only few or none of the progeny plants of clone M62759, which thus strongly inhibited virus transport to tubers. The study on PLRV translocation from aphid‐inoculated shoots to uninoculated shoots sprouted from the same tubers showed that no specific mechanisms are likely to impair PLRV movement through the tubers of the resistant genotypes. These results indicate that three valuable components of the resistance to PLRV are probably closely linked in the genotype, a combination that seems to occur rather rarely in potato clones. Nevertheless, selecting potato genotypes for the complex resistance to PLRV may prove to be a worthwhile part of breeding programmes, provided that the genetic mechanisms governing particular types of resistance are better recognized.  相似文献

The wheat variety Red Standard was more susceptible to infection by Erysiphe graminis at 14–20° C. than at c. 7° C. Conidia were produced per unit area of pustule more than ten times as rapidly at 14° C. as at 7° C.
The increased susceptibility of wheat to mildew after applying nitrogenous fertilizer (N) was associated with changes in its growth rate. Both growth rate and susceptibility increased to a maximum and then declined; the curves for the two were parallel, with a lag of some days between effect on growth rate and effect on susceptibility. Plants that had passed through the susceptible phase and became resistant to mildew, again became susceptible when supplied with more N. Nitrogen-deficient plants continuously resisted infection.
The higher the average growth rate during an experiment the greater was the total amount of infection. Increasing the average growth rate was soon followed by a sharp increase in the amount of infection. When plants of two size groups received the same amount of N the initially smaller plants became more heavily infected than the larger plants.  相似文献

‘Early-dying’ disease was examined in potato plants (King Edward) with varying numbers of main stems and lateral stems. Infection with Verticillium affected neither the number of main stems produced nor the stem number/ lateral number/yield inter-relationships, but did slightly reduce lateral development. The host growth pattern, however, markedly influenced the severity of disease: infected plants with a single main stem or a few, compared with those having many main stems, showed delayed symptom expression and also produced many laterals which further reduced disease severity and increased longevity. Thus yield reductions as a result of infection are likely to be smaller in single-stemmed plants showing considerable lateral development than in those plants with many main stems and few or no laterals.  相似文献

The pathophysiology of tomato plants infected with potato witches’ broom was studied. Changes in dry weight, content and composition of ash and the amount of different phosphorus and nitrogen fractions were investigated. All analyses were carried out separately in leaves, stems and preliminarily also in flowers. Considerable disproportions found in the distribution of metabolites among the individual organs investigated correspond to a great extent to the morphological picture of the disease. The leaves of diseased plants are actually starving and their growth and development are greatly inhibited. On the other hand, the stems are overflowing with metabolites, in particular with nitrogenous ones. They are thickened and along their entire length rootlets and sprouts appear forming the characteristic brooms. The authors assume that the disturbances in flower formation in diseased plants are due to the lack of essential metabolites. Diseased plants contain an increased amount of alkaline metals and alkaline earths.  相似文献
