为探究林下参内生真菌球毛壳菌(Chaetomium globosum)FS-01菌株对人参病原菌的抑菌作用,该研究在实验室条件下,测定了FS-01菌株菌丝、发酵液和孢子悬浮液对人参黑斑病菌(Alternaria panax)、人参菌核病菌(Sclerotinia schinseng)、人参灰霉病菌(Botrytis cinerea)、人参立枯病菌(Rhizoctonia solani)、人参根腐病菌(Fusarium solani)5种人参病原菌的抑制作用。结果表明:内生真菌球毛壳菌FS-01对5种病原菌均有抑制作用,其中,对人参黑斑病菌的抑制作用最高,为30.80%,其次是人参立枯病菌、人参菌核病菌、人参根腐病菌和人参灰霉病菌; 发酵液抑菌实验结果表明,在加入内生真菌球毛壳菌FS-01菌株发酵液的PDA培养基上,对人参灰霉病菌的抑制作用最高,为82.09%,其次是人参菌核病菌、人参黑斑病菌、人参立枯病菌和人参根腐病菌; 孢子抑菌实验结果表明,在加入内生真菌球毛壳菌FS-01菌株孢子悬浮液的PDA培养基上,对人参黑斑病菌的抑制作用最高,为83.72%,其次是人参灰霉病菌、人参立枯病菌、人参菌核病菌和人参根腐病菌。综上结果认为,内生真菌球毛壳菌FS-01菌株对人参病原菌均有很高的抑菌作用,可作为人参病原菌的生防菌株资源。  相似文献

Pairs of fungi were incubated on wheat straw in microcosms for 10 weeks. Release of Na+, K+ and NH4 +-N was similar from all combinations, but Ca2+, Mg2+ and PO4 3--P release depended on the species. In Agrocybe gibberosa/Chaetomium globosum and Sphaerobolus stellatus/Chaetomium globosum combinations, there was evidence of interactions which suppressed the predicted rate of phosphate release, and in all the mixed species combinations there were interactions which increased the rate of fungal respiration above that of the more combative fungus in pure culture. ei]{gnR}{fnMerckx}  相似文献

The effect of cellulosic substrates on the production of extra-cellular cellulases and their cellulolytic activity inChaetomium globosum has been studied in shake flask cultures. Production of endoglucanase, exoglucanase and filter paper cellulase was highest with pure cellulose whereas -glucosidase was maximally induced by wheat straw. A suitable pretreatment for wheat straw was peracetic acid followed by NaOH and that of bagasse with NaOH for saccharification.

La paille de froment est traitée de manière adéquate pour la saccharification par l'acide peracétique suivi de NaOH, la bagasse, par NaOH.

The potential of an antibiotic-producing isolate of Chaetomium globosum (CgA-1) to suppress Diaporthe phaseolorum f. sp. meridionalis (Dpm) in soybean stubble was studied in field microplots of no-tillage, minimum-tillage, and shallow plowing. Mature soybean stems colonized in vitro with Dpm were spread on the soil surface and C. globosum ascospore suspension, without nutrient supply, was sprayed over the entire plot prior to any tillage operation. Perithecial formation and survival of Dpm in soybean stems, concomitantly with colonization by C. globosum, were monitored for a 180-day period (mid-autumn through winter and mid-spring), which is the normal interval between soybean harvest and sowing. The proportion of soybean stem segments occupied by Dpm and number of perithecia formed decreased linearly with time and showed a strong negative correlation with increase in the occupation by C. globosum. At the end of the study, which coincided with the soybean sowing season, the soybean stubble was free from viable Dpm and was colonized by C. globosum. The effectiveness of C. globosum in eliminating the pathogen from surface-borne residue or harrowed-in residue was similar but much slower than in the shallow-plowed microplots. C. globosum successfully competed with major interfering fungi such as, Trichoderma, Nigrospora, and Fusarium in colonizing the soybean stems above and under the soil surface. The data provide strong evidence for use of the antibiotic-producing isolate of C. globosum to control soybean stem canker disease.  相似文献

The genus Chaetomium is a rich source of novel and bioactive secondary metabolites of great importance. To date, a variety of more than 200 secondary metabolites belonging to diverse structural types have been discovered. Fungal enzymes are used in food, beverages, confectionaries, textiles, and leather industries to simplify the processing of raw materials. They are often more stable than enzymes derived from other sources. Ten isolates of Chaetomium globosum recovered and designated as TUCg1 to TUCg10 were identified by morphological and molecular biology means and submitted to the GenBank. These isolates were screened for extracellular enzymes such as amylase, cellulase, laccase, lipase, pectinases, protease and chitinase on solid media. All Chaetomium globosum isolates screened for potential enzymes showed amylolytic, cellulolytic, and proteolytic activities; six isolates were chitinolytic and laccase producers; and five and three isolates showed pectinolytic and lipolytic activities, respectively. The produced array of enzymes differed among isolates. Molecular techniques such as internal transcribed spacer (ITS) region sequencing and specific genes random primers polymerase chain reaction (SGRP-PCR) have shown high DNA polymorphism of Chaetomium globosum. In conclusion, SGRP-PCR is a rapid and valuable tool for assessment and characterization of genetic diversity of Chaetomium globosum, which suggests the use of this technique for identification of different fungal isolates.  相似文献

Purpose of Review

This study aimed to isolate and characterize filamentous fungi onychomycosis agents in a military population assisted at a hospital outpatient clinic.

Recent Findings

In onychomycosis, the fungi colonize the subungual region causing thickening, discoloration, or cracking of the nail bed. Samples were collected from patients with clinical sights of onychomycosis.

Summary

Among 80 samples collected, 50 (62.5%) had positive culture. Isolated dermatophytes (86%) were Trichophyton rubrum (21; 42%), T. mentagrophytes var. interdigitale (19; 38%), and Microsporum gypseum (3; 6%) and non-dermatophyte molds were Fusarium spp. (1; 2%), Scytalidium spp. (1; 2%), and Chaetomium globosum (5; 10%). Minimal inhibitory concentrations (mg/L) of terbinafine, itraconazole, and fluconazole necessary to inhibit 50/90% of the isolates were respectively 0.015/0.06, 0.06/0.12, and 32/32. Etiological agents of onychomycosis in a military hospital are similar as reported in studies for the general population. High prevalence of non-dermatophytic agents was observed, especially for Chaetomium globosum.

Chaetomium globosum, the most common species within this genus, produces chaetoglobosins A and C when cultured on building material. Relatively low levels of these compounds have been shown to be lethal to various tissue culture cell lines. This study had two major objectives: (1) to determine the frequency at which Chaetomium species are isolated in water-damaged buildings and (2) to examine the production of chaetoglobosins A and C in isolates of C. globosum obtained from different buildings. Out of 794 water-damaged buildings, Chaetomium species were isolated in 49% of these structures. C. globosum ATCC 16021 was grown on four different media: oatmeal agar (OA), potato dextrose agar (PDA), corn meal agar (CMA), and malt extract agar (MEA). After 4 weeks, fungal growth was evaluated based on colony diameter and the quantity of spores produced on agar plates. In addition, production of chaetoglobosin A and C was monitored using high performance liquid chromatography. Colony diameter, spore production, and mycotoxin production by C. globosum were the highest on OA. Out of 30 C. globosum isolates cultured on OA for 4 weeks, 16 produced detectable amounts of chaetoglobosin A and every isolate produced chaetoglobosin C.  相似文献

Objective

We aimed to identify new high-yield dextranase strains and study the catalytic potential of dextranase from the strain in industrial applications.

Results

Dextranase-producing strains were screened from soil samples, and a potential strain was identified as Chaetomium globosum according to its phenotype, biochemical characteristics, and rDNA analysis. Crude dextranase was purified to reach 10.97-fold specific activity and 18.7% recovery. The molecular weight of the enzyme was 53 kDa with an optimum temperature and pH of 60 °C and 5.5, respectively. Enzyme activity was stable at pH 4.0–7.0 and displayed sufficient thermal stability at temperatures?<?50 °C. Mn2+ (10 mM) enhanced dextranase activity by 134.44%. The enzyme was identified as an endodextranase. It displayed very high hydrolytic affinity toward high-molecular weight dextran T2000, reaching 97.9% hydrolysis within 15 min at 2 U/mL.

Conclusion

Collectively, these results suggest that Chaetomium globosum shows higher production and specificity of dextranase than that from other reported strains. These findings may offer new insights into the potential of dextranase in the sugar, medical, and food industries.

为探究球毛壳ND35微生物菌剂对楸树幼苗生长及土壤肥力的作用机制,本研究楸树幼苗为研究对象,采用室内盆栽试验,设计0(CK),10(T1),15(T2),20(T3)4种微生物菌剂施用量,测定幼苗生长情况、土壤微生物组成结构、土壤酶和土壤养分等特征。研究结果如下:(1)球毛壳ND35微生物菌剂可显著促进楸树幼苗的生长,株高、地径、地上及地下生物量显著提高(P<0.05),T2处理下促生效果最好。(2)施用球毛壳ND35微生物菌剂可显著提高土壤中有机质、硝态氮、铵态氮含量及脲酶、磷酸酶、蔗糖酶活性(P<0.05)。(3)球毛壳ND35微生物菌剂可显著影响土壤细菌群落组成,提高细菌群落的丰富度和多样性,使土壤中β-变形菌纲(Betaproteobacteria)、γ-变形菌纲(Gammaproteobacteria)的相对丰度显著下降,α-变形菌纲(Alphaproteobacteria)、δ-变形菌纲(Deltaproteobacteria)的相对丰度呈显著提高,可使土壤中鞘氨醇单胞菌属(Sphingomonas)的相对丰度显著提高21.88%-103.56%(P<0.05),芽孢杆菌属(Bacillus)的相对丰度提高66.28%-65.97%(P<0.05),酸杆菌属(Acidibacter)的相对丰度提高12.76%-38.06%。(4)冗余分析(RDA)结果表明,土壤硝态氮、铵态氮、有机质是影响土壤细菌群落分布和多样性的重要环境因子,土壤细菌群落结构的改变会显著影响土壤脲酶、蔗糖酶、碱性磷酸酶的活性。因此,施用球毛壳ND35微生物菌剂可通过影响植物根际土壤的化学性质及生物性质,促进楸树幼苗的生长。这一研究结果为楸树繁育提供了新的指导方向,亦为将其用于困难立地及退化生态系统植被恢复提供基础理论指导。  相似文献

Chaetomium globosum has been well-known potential antagonist of several seed and soilborne fungus. Eight isolates of C. globosum were obtained from different sources and were identified by morphological characters. C. globosum isolates examined for the presence of extra cellular proteins, cellulases and antifungal metabolites in culture filtrate by sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE), thin-layer chromatography and high-performance liquid chromatography. Variation in the mycelial protein of C. globosum isolates was noted in the SDS-PAGE analysis. Different C. globosum isolates that showed more number of bands in protein profile was further screened for the production of cellulases in culture filtrate. Cellulase activity of C. globosum isolates revealed that maximum activity was observed in the isolate Cg-6 after 11?days of incubation, while Cg-2 had least activity. C. globosum isolates were tested for antibiotic production, among which three isolates viz. Cg-6, Cg-7 and Cg-5 were found to produce the antibiotic Chaetoglobosin A in the culture filtrate. The antibiotic Chaetoglobosin A appeared blue colour under UV spectrum with a wavelength of 250?nm.  相似文献

Pseudolaric acid B (1) is a natural product with potent antifungal activity. We discovered that pseudolaric acid B did not kill but only suppress the growth of the filamentous fungus Chaetomium globosum. It was proposed that pseudolaric acid B was converted to metabolites with decreased antifungal activities. In this study, a scaled-up biotransformation of pseudolaric acid B by C. globosum produced five metabolites, including three new compounds, pseudolaric acid I (2), pseudolaric acid B 18-oyl-alanine (4) and pseudolaric acid B 18-oyl-serine (6), together with two known compounds, pseudolaric acid F (3) and pseudolaric acid B 18-oyl-glycine (5). The structures were characterized by NMR and MS spectroscopy. The major biotransformation reaction was conjugation with amino acids. None of the metabolites showed inhibitory effects on the growth of Candida albicans. The results suggested that biotransformation might be a detoxification process for fungi to resist antifungal drugs.  相似文献

Chaetomium globosum grows vigorously on media containing, assource of nitrogen, ammonium pyruvate, lactate, succinate ortartrate, or urea. Most common amino-acids, except lysine, permitgrowth to varying degrees. The carbon of those compounds whichsustain growth is also utilized by C. globosum, but usuallyonly when glucose is also available. Sporulation is vigorouson media containing ammonium pyruvate or lactate, -alanine oraspartic acid; less so on other amino-acids, and very weak onammonium tartrate. Vigour of fruiting was apparently connectedwith the presence of organic phosphates in the media, and itis suggested that the favourable influence of pyruvate, lactate,and -alanine is due to their ready transformation into 3-phosphoglycericacid; this transformation has been demonstrated for -alanineby the use of the 14C-labelled compound.  相似文献

A novel polyhydroxylated C29-sterol, 25ξ-methyl-22-homo-5α-cholest-7,22-diene-3β,6β,9α-triol, designated globosterol (1), together with one known tetrahydroxylated ergosterol (22E, 24R)-ergosta-7,22-diene-3β,5α,6β,9α-tetraol (2) has been isolated from the cultures of an endophytic fungus, Chaetomium globosum ZY-22 originated from the plant Ginkgo biloba. The structures and relative configurations of 1 and 2 were established on the basis of extensive spectroscopic analyses including 1D and 2D NMR (1H-1H COSY, HSQC, HMBC, and NOESY) experiments and comparison with the literature. Globosterol (1) possesses an unprecedented 25-methyl Δ22-C10-side chain and Δ7-3β,6β,9α-hydroxy-steroid nucleus, which represents the first example for C29-steroids of the group.  相似文献

From the culture of an endophyte Chaetomium globosum found in Ginkgo biloba, twelve metabolites, including three diketopiperazines, two cytochalasins, two sterols, two simple phenolic compounds, one complex aromatic compound, a nonprotein amino acid and a linear triterpene were isolated. Of them, eight compounds are first reported for the genus Chaetomium. Gliotoxin (8) has good antifungal activity against plant pathogenic fungi.

Submerged cultures of the ascomycete Chaetomium globosum oxidised the exogenous sesquiterpene (+)-valencene to nootkatone via the stereoselective generation of -nootkatol. Inhibition experiments suggested that the first introduction of oxygen, the rate-limiting step of the bioconversion, may have been catalysed by a cytochrome-P450-monooxygenase. However, nootkatone was not the final metabolite: further flavour-active and inactive, non-volatile oxidation products were identified. (+)-Valencene and the flavour-active mono-oxyfunctionalised transformation products, -nootkatol, nootkatone, and valencene-11,12-epoxide accumulated preferably inside the fungal cells. Di- and poly-oxygenated products, such as nootkatone-11,12-epoxide, were found solely in the culture medium, indicating an active transport of these metabolites into the extracellular compartment during (+)-valencene detoxification. These metabolic properties may have contributed to the high tolerance of the fungus towards the exogenous hydrocarbon.  相似文献

Summary Kinnow-mandarin waste (peel, pulp and seeds) was assessed for single-cell protein (SCP) production byChaetomium globosum andSporotrichum pulverulentum in shake-flask culture. The maximum protein enrichment (32% and 34%) of the substrate was achieved after 5 and 7 days of incubation by the two organisms, respectively. Of various nitrogen sources, NaNO3 and NH4Cl gave maximum protein enrichment of the substrate byC. globosum andS. pulverulentum, respectively.

Résumé On a examiné la possibilité de produire des protéines uni-cellulaires (POU) à partir de résidus de la mandarine-kinnow (pelures, pulpe, et pépins) par la culture deChaetomium globosum et deSporotrichum pulverulentum en flacons agités. L'enrichissement maximum en protéines du substrat, soit 32 et 34% est obtenu après 5 et 7 jours d'incubation respectivement par les deux organismes. Parmi les diverses sources d'azote, NaNO3 et NH4Cl ont permis respectivement l'enrichissement maximum en protéines du substrat parC. globosum etS. pulverulentum.

Abstract

Crude glycerol is becoming a financial and environmental liability due to its surplus production from biodiesel industry, and its utilization as a fermentation feedstock for value-added chemicals production has been widely studied. In present work, the capacity of an endophytic fungus, Chaetomium globosum CGMCC 6882, using glycerol and crude glycerol for polysaccharide production was investigated. Results showed that the polysaccharide titers from glucose and glycerol were 1.85 and 3.8?g/L, respectively. Moreover, spore morphology of C. globosum CGMCC 6882 was favorable for polysaccharide production. Meanwhile, impurities in crude glycerol have no effect on polysaccharide production by C. globosum CGMCC 6882. Finally, characteristic results of polysaccharides produced from glucose, glycerol, and crude glycerol have suggested that metabolic flux might be a determinant factor on polysaccharide structure. Taken together, this research provided an innovative approach of utilizing crude glycerol produced from the biodiesel production process.  相似文献

A wide range of field and storage fungi were isolated from black pepper, white pepper and Brazil nut kernels from Amazonia. A total of 42 species were isolated from both peppers. Aspergillus flavus and A. niger were isolated more frequently from black than from white pepper. Other potential mycotoxigenic species isolated included: A. ochraceus, A. tamarii, A. versicolor, Emericella nidulans and Chaetomium globosum, Penicillium brevicompactum, P. citrinum, P. islandicum and P. glabrum. Species isolated from pepper for the first time were Acrogenospora sphaerocephala, Cylindrocarpon lichenicola, Lacellinopsis sacchari, Microascus cinereus, Petriella setifera and Sporormiella minima. Seventeen species were isolated from Brazil nut kernels. A. flavus was the dominant species followed by A. niger. P. citrinum and P. glabrum were the only penicillia isolated. Species isolated for the first time included Acremonium curvulum, Cunninghamella elegans, Exophiala sp., Fusarium oxysporum, Pseudoallescheria boydii, Rhizopusoryzae, Scopulariopsis sp., Thielavia terricola and Trichoderma citrinoviride. Considerably more metabolites were detected from black than white pepper in qualitative analyses. Chaetocin, penitrem A, and xanthocillin were identified only from black pepper, and tenuazonic acid was identified from both black and white pepper. Aflatoxin G2, chaetoglobosin C, and spinulosin were identified from poor quality brazil nuts. Aflatoxin B1 and B2 were also only detected in poor quality brazil nuts at concentrations of 27.1 g kg–1 and 2.1 g kg–1 respectively (total 29.2 g kg–1).This revised version was published online in October 2005 with corrections to the Cover Date.  相似文献
