Summary The effect of yeast metabolism on the dispersion characteristics of a fluidised bed fermentor containing flocs of the yeast Saccharomyces carlsbergensis was investigated. Dispersion in the metabolizing fluidised yeast floc system was compared with the dispersion in an inert yeast floc system and in a glass bead system. Breakdown in plug-flow was found to occur in the metabolically active yeast bed when the flow rate was increased over a relatively narrow operating range (up to a dilution rate of 0.08 h-1). Ultrastructural and physiological responses of Prorocentrum mariae-lebouriae (Parke & Ballantine) Faust are reported for cultures maintained at growth irradiances (Ig) ranging from 20.6 to 0.3 E m?2.d?1 and following downward shifts in light intensity. We tested the hypothesis that Prorocentrum grown under light regimes that elicit different responses in photosynthesis and pigmentation exhibit distinctive cell ultrastructures. Prorocentrum from high-light conditions had high saturation intensities for photosynthesis (Ik) and low levels of Chl a, Chl c and peridinin-cell?1 These cultures were morphologically distinguished by a large starch volume fraction (Vv), small chloroplast Vv and fewer thylakoids lamella?1. Ik values were lower and pigment concentrations higher in low-light treatments, and cells showed reduced starch Vv, large chloroplast Vv, and higher numbers of thylakoids · lamella ?1. Cells grown under extremely low-light conditions appeared stressed as indicated by the absence of starch reserves and the presence of large vacuoles within the cytoplasm. Results for presence of large vacuoles within the cytoplasm. Results for quantiative electron microscopy, photosynthesis-irradiance (P-I) relations and cell pigmentation indicate that photoadaptation in P. mariae-lebouriae involves a strategy that encompasses changes in both the “size” and “number” of photosynthetic units.  相似文献

The heritability (h2) of fitness traits is often low. Although this has been attributed to directional selection having eroded genetic variation in direct proportion to the strength of selection, heritability does not necessarily reflect a trait's additive genetic variance and evolutionary potential (“evolvability”). Recent studies suggest that the low h2 of fitness traits in wild populations is caused not by a paucity of additive genetic variance (VA) but by greater environmental or nonadditive genetic variance (VR). We examined the relationship between h2 and variance‐standardized selection intensities (i or βσ), and between evolvability (IA:VA divided by squared phenotypic trait mean) and mean‐standardized selection gradients (βμ). Using 24 years of data from an island population of Savannah sparrows, we show that, across diverse traits, h2 declines with the strength of selection, whereas IA and IR (VR divided by squared trait mean) are independent of the strength of selection. Within trait types (morphological, reproductive, life‐history), h2, IA, and IR are all independent of the strength of selection. This indicates that certain traits have low heritability because of increased residual variance due to the age at which they are expressed or the multiple factors influencing their expression, rather than their association with fitness.  相似文献

Both intrinsic and observed kinetic investigations for those ethanol fermentations using self-flocculated yeast strains have been hindered by the lack of real online monitoring techniques and proper characterization methods for the flocs. An optical detecting technique, the focused beam reflectance measurement probe developed by Lasentec (Redmond, WA) was inserted into a fermentor to monitor the floc chord length distributions. Using a simulating system composed of the floc-buffer suspensions, the total floc chord length counts per second were directly correlated with the floc biomass concentrations so that the floc biomass concentrations can be in situ detected. Furthermore, a characterization method of the flocs was established by properly weighted treatments of the detected floc chord length distributions. When a real yeast floc ethanol fermentation system was detected during its intrinsic kinetic investigations in which the floc size needed to be controlled at a level of micrometer scale to eliminate inner mass transfer limitations, it was found and validated that CO(2) produced during fermentation exerted significant disturbances. By applying 1/length-weighted treatment, these disturbances were effectively overcome.  相似文献

Flocculating agents are used as auxiliary to recover bacterial cells in downstream processes for polyhydroxyalkanoate production. However little is known about the Curpiavidus necator flocs. In this work a new procedure for floc characterization through digital image analysis is presented and validated using the batch settling test. Average diameter, particle size distribution and morphological characteristics of the microbial aggregates were obtained from the flocculation/sedimentation process of the Cupriavidus necator DSM 545 cells by the use of tannin as flocculating agent. The experimental results demonstrated that the proposed method is adequate to determine the average floc diameter with values around 150 μm in accordance with the value obtained from the batch settling test. Nevertheless a morphological characterization of Cupriavidus necator DSM 545 bioaggregates in terms of size distribution and regularity could only be performed by an image analysis procedure. The procedure allowed us to describe the regularity of bacterial flocs through the quantification of morphological parameters of Euclidean [convexity (Conv) and form factor (FF)] and fractal geometry [surface fractal dimension (D BS)], which are important factors to be considered in the settling efficiency of aggregates.  相似文献

Objective: Obese patients without clinically apparent heart disease may have a high output state and elevated total and central blood volumes. Central circulatory congestion should result in elevated pulmonary diffusing capacity (DLCO) and capillary blood volume (Vc) reflecting pulmonary capillary recruitment; however, the effect on membrane diffusion (Dm) is uncertain. We examined DLCO and its partition into Vc and Dm in 13 severely obese subjects (BMI = 51 ± 14 kg/m2) without manifest cardiopulmonary disease before and after surgically induced weight loss. Research Methods and Procedures: DLCO and its partition into Vc and Dm [referenced to alveolar volume (VA)] as described by Roughton and Forster, total body water by tritiated water, and fat distribution by waist‐to‐hip ratio were performed. Results: Despite normal DLCO (mean 98 ± 16% predicted), Vc/VA was increased (mean 118 ± 30% predicted), and Dm/VA was reduced (mean 77 ± 34% predicted). Nine of 13 subjects were restudied after weight loss (mean 52 ± 43 kg); Vc/VA decreased to 89 ± 18% predicted (p = 0.01), and Dm/VA increased to 139 ± 30% predicted (p < 0.01). Increasing total body water was associated with both increasing Vc (r = 0.74, p = 0.01) and increasing waist‐to‐hip ratio (r = 0.65, p = 0.02), indicating that circulatory congestion increases with increasing central obesity. Discussion: Severely obese subjects without manifest cardiopulmonary disease may have increased Vc indicating central circulatory congestion and reduced Dm suggesting associated alveolar capillary leak, despite normal DLCO. Reversibility with weight loss is in accord with reversibility of the hemodynamic abnormalities of obesity.  相似文献

Analytical and numerical calculations of the decay processes of the electric birefringence in an isomerizing system have been performed. Two modes of isomerization are considered: in the first mode, the direction of the axis (of the optical anisotropy) of a molecule is conserved during isomerization; in the second mode, it is not. In the first mode, if GAis not equal to GB, and that, if kAopA2 is not equal to kBopB2 (when the orienting electric field is weak), in which GA, GB and pA, pb are the optical anisotropy and permanent electric dipole moments, of the molecule in two states A and B of the isomerization, and kAo and kBo are the rate constants for the transitions A → B and B → A, respectively, and if diffusion rates are very much slower than the chemical rates, the relaxation due to the chemical reaction can be detected in the decay of the electric birefringence. In the second mode, if diffusion rates are somewhat slower than the chemical rates, the relaxation due to the chemical reaction appears in the decay, even though there are no differences in optical anisotropy and electric moments in the two states. When the rotary diffusion coefficients are different in the two states, the decay process becomes almost one-component, bringing the chemical rate about ten times higher to diffusion coefficients in both modes.  相似文献

The kinetics of γ-benzyl-L -glutamate NCA's polymerizations in dioxane as solvent are discussed. The partial orders respective to [A], the NCA concentration and [I0], the initial initiator concentration are given; the rate constants are deduced and a mechanism is proposed to justify a rate of polymerization Vp = k[A][I0]2. The dependence of the rate constants on the conformation of the growing chains is demonstrated; the acceleration is attributed to the ordered structures favorable to hydrogen bonding. The kinetics of aging have also been examined and discussed; it is shown that they cannot modify the kinetics of polymerization. The DP n were obtained on the same samples before and after debenzylation; it is proved that at any concentration, DP n ? [A0]/[I0]. The molecular-weight distributions were obtained by chromatography and a polydispersity lower than 1.3 was deduced for each sample. The trimodal distribution, appearing as soon as [A0]/[I0] is larger than 3, is attributed to the existence of the three structures σ, β, and α. The weight fraction of each of the structures was correlated to the kinetics of polymerization.  相似文献

A simple, straightforward procedure, which requires no special tables or generators, is presented for constructing resolvable incomplete block designs for v=pk, v=p2k, …, treatments, for kp, in incomplete blocks of size k. Also, it is shown, how to obtain incomplete block designs for any v in blocks of size k and k+1. The procedure allows construction of balanced incomplete block designs for p = k a prime number. For p = n not a prime number, incomplete block designs can be obtained by the procedure, but are not balanced. However, for ps being the smallest prime factor of n, ps + 1 for v = n2, ps2+ ps + 1 for v = n3, …, arrangements can be obtained for which the occurrence of any treatment pair in the blocks is either zero or one. This is called a zero-one concurrence design. Procedures are described for obtaining additional zero-one concurrence arrangements. It is shown that the efficiency of these designs is maximum. Both intra-block and inter-block analyses are described.  相似文献

Detailed topographic and hydrologic surveys were conducted in five intertidal cove marshes in an outer coastal plain landscape to test the hypothesis that the equilibrium geologic state of intertidal habitats residing in similar landscape situations conforms to a consistent geometric form. The equation V=1571.84A1.70 (R2=96.2%) describes the relationship between hectares of marsh (A) and cubic meter volume at mean high tide (V). An empirical relationship between tide height and volume was found to obey the power series Vp=L2.38 (R2=99.6%), where Vp is volume as a percent of full pool and L is water height as a percent of mean high tide. A dimensionless index describing the relationship between area and volume is consistent for each marsh and approaches 0.10. A channel form parameter describing width to channel depth ratios is of consistent value for four of the five marshes. These provide evidence of deterministic rather than stochastic geologic development. The benefits of applying natural basin shape patterns in the design and engineering of created/restored intertidal marshes are highlighted and a generic basin is modeled (based on the geometrical section of a paraboloid retained by simple integration) as an example of the potential applicability of the study.  相似文献

Summary Physical characteristics, namely floc density function, floc size distribution, and relative floc strength, of a number of flocculent yeast types were measured. A straight-line relationship was found to exist between log values of size and density for the yeasts examined. Each yeast type had coefficients from this relationship which could be used to interpret settling behaviour. A cost-minimizing mathematical model for on-line control of dissolved oxygen using agitation speed and aeration rate was developed. In pilot scale monensin fermentation using Streptomyces cinnamonensis, this algortihm provided stable control of dissolved oxygen at 40%, reducing energy usage 27.8%. The agitation and aeration profiles provided by the algorithm respresent the pathway of least energy cost for control at the desired dissolved oxygen level. Other observed advantages of bivariable control were reduction of foaming, evaporation, and gas holdup. Reduced maintenance of compressors and agitator motors could also be expected due to decreased load. Total mercury concentrations (THg) in lake and stream sediments generally decrease with wet-area coverage (AW) per upslope basin area (AB). This was determined by delineating the wet-area component of 12,653 basins above as many sediment-sampling locations of the Geological Survey of Canada. These locations represent four climate regions (maritime, boreal, arctic, alpine) comprising six stream and six lake study areas. The dependence of sediment THg on AW/AB was examined by dividing the 0 < AW/AB < 1 range into 40 equal segments, and obtaining the mean sediment THg value for each segment. The results were evaluated by way of regression analysis using the following equation: mean sediment THg = a (1 ? AW/AB)b + c AW/AB, with a, b and c as area-specific coefficients. The “a” and “c” coefficients could – in part – be inferred from bedrock type, annual atmospheric Hg deposition, and mean monthly air temperatures, and mean annual precipitation. Both “a” and “c” increased with increasing atmospheric Hg deposition for lake sediments. For stream sediments, only “a” did so. The geogenic influence on the THg variations per study area was addressed through multiple regression analyses, using sediment concentrations of other heavy elements and organic matter as independent variables.  相似文献

To define the polymorphism and extent of the mouse immunoglobulin kappa (Igk) gene complex, we have analyzed restriction-enzyme digested genomic DNA from 33 inbred strains of mice with labeled DNA probes corresponding to 16 V x protein groups (1 of them previously undescribed) and the J k/C K region (V, variable; J, joining; C, constant). These probes detected between 1 and 25 distinct restriction enzyme fragments (REF) that appeared in up to eight polymorphic patterns, thus defining eight mouse Jgk haplotypes. The investigated portion of the V A repertoire was estimated to encompass between 60 and 120 discernable V k gene-containing REFs. In contrast to mouse V H gene families, several V k gene families defined by these probes appeared to overlap. Microalgal bacterial floc (MaB‐floc) reactors have been suggested as a more sustainable secondary wastewater treatment. We investigated whether MaB‐flocs could be used as tertiary treatment. Tertiary influent has a high inorganic/organic carbon ratio, depending on the efficiency of the secondary treatment. In this study, the effect of this inorganic/organic carbon ratio on the MaB‐flocs performance was determined, using three sequencing batch photobioreactors. The MaB‐flocs were fed with synthetic wastewater containing 84, 42, and 0 mg L−1 C‐KHCO3 supplemented with 0, 42, 84 mg L−1 C‐sucrose, respectively, representing inorganic versus organic carbon. Bicarbonate significantly decreased the autotrophic index of the MaB‐flocs and resulted in poorly settling flocs. Moreover, sole bicarbonate addition led to a high pH of 9.5 and significant lower nitrogen removal efficiencies. Sucrose without bicarbonate resulted in good settling MaB‐flocs, high nitrogen removal efficiencies and neutral pH levels. Despite the lower chlorophyll a content of the biomass and the lower in situ oxygen concentration, 92–96% of the soluble COD‐sucrose was removed. This study shows that the inorganic/organic carbon ratio of the wastewater is of major importance and that organic carbon is requisite to guarantee a good performance of the MaB‐flocs for wastewater treatment. Data from natural populations have suggested a disconnection between trait heritability (variance standardized additive genetic variance, VA) and evolvability (mean standardized VA) and emphasized the importance of environmental variation as a determinant of trait heritability but not evolvability. However, these inferences are based on heterogeneous and often small datasets across species from different environments. We surveyed the relationship between evolvability and heritability in >100 traits in farmed cattle, taking advantage of large sample sizes and consistent genetic approaches. Heritability and evolvability estimates were positively correlated (r = 0.37/0.54 on untransformed/log scales) reflecting a substantial impact of VA on both measures. Furthermore, heritabilities and residual variances were uncorrelated. The differences between this and previously described patterns may reflect lower environmental variation experienced in farmed systems, but also low and heterogeneous quality of data from natural populations. Similar to studies on wild populations, heritabilities for life‐history and behavioral traits were lower than for other traits. Traits having extremely low heritabilities and evolvabilities (17% of the studied traits) were almost exclusively life‐history or behavioral traits, suggesting that evolutionary constraints stemming from lack of genetic variability are likely to be most common for classical “fitness” (cf. life‐history) rather than for “nonfitness” (cf. morphological) traits.  相似文献

Automated assignment of NOESY spectra is a prerequisite for automated structure determination of biological macromolecules. With the program KNOWNOE we present a novel, knowledge based approach to this problem. KNOWNOE is devised to work directly with the experimental spectra without interference of an expert. Besides making use of routines already implemented in AUREMOL, it contains as a central part a knowledge driven Bayesian algorithm for solving ambiguities in the NOE assignments. These ambiguities mainly arise from chemical shift degeneration which allows multiple assignments of cross peaks. Using a set of 326 protein NMR structures, statistical tables in the form of atom-pairwise volume probability distributions (VPDs) were derived. VPDs for all assignment possibilities relevant to the assignments of interproton NOEs were calculated. With these data for a given cross peak with N possible assignments A i(i = 1,...,N) the conditional probabilities P(A i, a|V 0) can be calculated that the assignment A idetermines essentially all (a-times) of the cross peak volume V 0. An assignment A kwith a probability P(A k, a|V 0) higher than 0.8 is transiently considered as unambiguously assigned. With a list of unambiguously assigned peaks a set of structures is calculated. These structures are used as input for a next cycle of iteration where a distance threshold D maxis dynamically reduced. The program KNOWNOE was tested on NOESY spectra of a medium size protein, the cold shock protein (TmCsp) from Thermotoga maritima. The results show that a high quality structure of this protein can be obtained by automated assignment of NOESY spectra which is at least as good as the structure obtained from manual data evaluation.  相似文献

Abstract Net nitrate uptake rates were measured and the kinetics calculated in non-nodulated Pisum sativum L. cv. Marma and Lemna gibba L. adapted to constant relative rates of nitrate-N additions (RA), ranging from 0.03 to 0.27 d?1 for Pisum and from 0.05 to 0.40 d?1 for Lemna, Vmax of net nitrate uptake (measured in the range 10 to 100 mmol m?3 nitrate, i.e. ‘system I’) increased with RA in the growth limiting range but decreased when RA exceeded the relative growth rate (RGR), Km was not significantly related to changes in RA. On the basis of previous 13N-flux experiments, it is concluded that the differences in Vmax at growth limiting RA are attributable to differences in influx rates. Linear relationships between Vmax and tissue nitrogen concentrations were obtained in the growth limiting range for both species, and extrapolated intercepts relate well with the previously defined minimal nitrogen concentrations for plant growth (Oscarson, Ingemarsson & Larsson, 1989). Analysis of Vmax for net nitrate uptake on intact plant basis in relation to nitrogen demand during stable, nitrogen limited, growth shows an increased overcapacity at lower RA values in both species, which is largely explained by the increased relative root size at low RA. A balancing nitrate concentration, defined as the steady state concentration needed to sustain the relative rate of increase in plant nitrogen (RN), predicted by RA, was calculated for both species. In the growth limiting range, this value ranges from 3.5 mmol m?3 (RA 0.03 d?1) to 44 mmol m?3 (RA 0.21 d?1) for Pisum and from 0.2 mmol m?3 (RA 0.05 d?1) to 5.4 mmol m?3 (RA 0.03 d?1) for Lemna. It is suggested that this value can be used as a unifying measure of the affinity for nitrate, integrating the performance of the nitrate uptake system with nitrate flux and long term growth and demand for nitrogen.  相似文献

The measurement of departure from randomness in spatial distributions has widespread application in ecological work. Several “indices of non-randomness” are compared with regard to their dependence on sample number, sample size and density. Criteria for the best choice of index for specific situations are discussed. A new coefficient Cx is proposed for use with positively contagious distributions and tests of significance are given. When Cx and another index (S2/m−1) are used for positive and negative contagion respectively, values ranging from −1 through 0 (random) to +1 are obtained, regardless of sample number, sample size or density.  相似文献

A new technique is presented for treating the ground state of an heteropolymer with a random sequence of components. An exact system of equations is found for determining the ground state energy E which is equal to the polymer free energy f in the lowest-order approximation in T/V (V/2 is the large “surface” energy arising at the boundaries between coiled and “helical” sections: V ? T, Uk; U1 and –U2 are the free energies of the components counted from the corresponding coiled state energies). These equations are essentially simplified at certain fixed values of the ratio U1/U2. For integer values of U2/U1 and U1/U2 a solution is obtained with an accuracy exp(–V/Uk). The ground-state energy as a function of U1 and U2 is shown to be highly irregular: its derivatives have jumps at an infinite number of points. These jumps provide a fine structure of the melting curves. A smoothed over the jumps function E′ is found by way of analytic continuation from the integer values of U1/U2 and U2/U1. The accuracy of the approximation fE is estimated and the correctional term of order T/V is determined.  相似文献
