This work aimed to develop the submerged cultivation conditions for improved exopolysaccharides (EPS) production by Armillaria luteo-virens Sacc. The effects of culture temperature, aeration rate, inoculum level, initial pH, and additives on EPS formation and mycelial growth are investigated. The aeration rate, initial pH, and inoculum level significantly affected EPS production under the submerged cultivation. The developed conditions were as follows: cultivation temperature 23 °C, initial pH 5.0, aeration rate 0.5 vvm, 0.5% Tween 80, inoculum level 5% (v/v), and shaking speed 120 r/min. Under the developed conditions, the highest EPS production was 13.01 g/L at 5 days culture time. EPS production was examined in a 5 L bioreactor, and an unstructured kinetic model for EPS formation was well developed. The verified investigations in the large-scale cultivation system showed that the developed models are able to predict the submerged cultivation process of EPS formation. Current results revealed that the submerged cultivation conditions can be utilized to control EPS production, and the unstructured models developed are suitable for explaining EPS production by A. luteo-virens Sacc QH in a large-scale cultivation bioreactor.  相似文献

For the cultivation of mammalian cells on microcarriers a minimum inoculum concentration is required to initiate cell attachment and subsequent cell growth. A critical cell number model has been proposed to elucidate the mechanism of the inoculum requirement. In this model it was hypothesized that after inoculation a critical number of cells per microcarrier is required for normal growth to occur; failure to acquire enough cells will impede cell growth. This critical cell number model was expressed mathematically and used to simulate cell distribution and growth on microcarriers under different cultivation conditions. By comparing the simulated growth kinetics with the experimental results, the actual critical cell number per microcarrier was identified. The critical number could be reduced by employing an improved medium for the cultivation.  相似文献

微载体是动物细胞高密度培养的有效手段。首先在硅化的方瓶中对Cytodex 1、Cy-todex 3、Biosilon、Bellco Glass Microcarrier、CT-1、CT-3、MC-1、CT-28种国产和进口微载体进行了比较和筛选。确定以Biosilon作为Vero细胞高密度培养的首选微载体。用500mlWheaton搅拌瓶探索影响Vero细胞高密度培养的条件,表明50~60mg/ml的微载体浓度、1~2×106/ml的细胞接种密度、适当的通气(95%O_2+5%CO2)对该细胞的高密度培养具有重要意义。在200ml培养体积的Wheaton搅拌瓶中,微载体浓度为50~60mg/ml,细胞接种密度为9.24×105/ml,搅拌速度为65~85r/min,经25d培养,Vero细胞密度可达2.34×107/ml,表明50~60mg/ml的微载体浓度对培养细胞没有毒性。接着在1.5L CelliGen生物反应器中进行培养,细胞接种密度为4.98×105/ml,培养体积为1.2L,日灌流量从0.20L逐渐加大到3.65L,经22d连接灌流培养,最终细胞密度可达2.05×107/ml。  相似文献

The noncoagulative type of pellet formation can be induced in submerged cultivation of the filamentous fungus Rhizopus nigricans. The size and constitution of the hyphal agglomerates obtained varied with changes in inoculum size and agitation speed for given media composition and cultivation conditions. The physiological state of mycelium, used for a further process of biotransformation, was estimated by following the growth kinetics, pH value and substrate utilization during submerged cultivation. Namely, differences in pellet morphology and physiology affect the ability of R. nigricans to hydroxylate progesterone at the 11α position. A repeated batch procedure revealed the best maintenance of biotransformation capacity for pellets, obtained from the growth phase of cultivation at high agitation speed and with low inoculum size.  相似文献

以马铃薯葡萄糖综合培养基(PDP)为基础培养基,采用正交试验法优化杏鲍菇菌丝体多糖发酵条件,对接种量、摇床转速和培养时间等因素对多糖含量的影响进行了研究。采用水提醇沉法提取多糖,苯酚—浓硫酸法进行多糖含量测定。结果表明,最佳培养条件:接种量为每瓶1块直径为1cm的菌块,转速为140r/min,培养时间为8d。此时杏鲍菇菌丝体多糖含量最高,为75.1mg/g。  相似文献

This study aimed to evaluate the influence of magnetic fields (MF) on inoculum cultivation and carotenoid production by Phaffia rhodozyma. The application of MF in the inoculum culture was evaluated (0 m T – control and 30 m T). Cellular concentration increased by 12.8 % after 24 h-culture with MF application compared to the control assay, and this was the best alternative for the preparation of inoculum. Different intervals of MF application were evaluated over 168 h. The highest volumetric carotenoids concentration was achieved by applying MF throughout cultivation, with values of 1146.39 ± 26.18 μg L−1 and carotenoid productivity of 11.94 ± 1.11 μg L−1 h−1 in 96 h. As a result, carotenoid production increased by 59.4 % and carotenoid productivity by 99.3 %. This study is one of the first to consider MF application in carotenoid production using P. rhodozyma as a viable and low-cost alternative for carotenoid production in a shorter cultivation time.  相似文献

Three strains of Aspergillus oryzae were used for fungal protein production from starch processing wastewater (SPW) as part of a comprehensive SPW utilization and treatment program. The mycelial morphologies of the three strains varied from fluffy, clumpy mycelia to compact pellets with various strains and cultivation conditions of growth pH, inoculum quantity and superficial air velocity in an air lift bioreactor process. An inoculum technique was developed to ensure a desirable morphology with an improved fungal protein yield. Experimental relationship between the morphology and the yield revealed that the formation of small compact pellets under designed cultivation conditions favoured higher fungal protein yields, easier product separation and better process operation.  相似文献

Effect of composition of the medium used for the inoculum cultivation, of the age and amount of the inoculum was investigated using a 3-L glass fermentor with a working volume of 1 L. The highest productivity of the culture was obtained when using a 20% (V/V) 1-d inoculum grown in the MRS medium. Yields of lactic acid were 88-97%, while the L(+)-isomer represented about 80% of the total product.  相似文献

通过晚稻田大面积放养固氮蓝藻试验,我们初步提出了土池大量培养和茬口田大面积生产藻种等一系列的培养和生产藻种的技术措施;生产了30吨的鲜藻种接种到晚稻田中。经过加强管理,先锋大队990亩晚稻田全部养藻化。接种的藻生长旺盛,平均产量达到1000斤/亩以上。抽样测产估计,晚稻田中鲜藻量达到500吨。晚稻田养藻使水稻增产约10%,个别达到20%以上。因此,晚稻田放养固氮蓝藻为晚稻开辟了新的有希望的肥源。    相似文献

The initial moisture content, cultivation time, inoculum size and concentration of basal medium were optimized in solid state fermentation (SSF) for the production of xylanase by an Aspergillus niger mutant using statistical experimental designs. The cultivation time and concentration of basal medium were the most important factors affecting xylanase activity. An inoculum size of 5 x 10(5) spores/g, initial moisture content of 65%, cultivation time of 5 days and 10 times concentration of basal medium containing 50 times concentration of corn steep liquor were optimum for xylanase production in SSF. Under the optimized conditions, the activity and productivity of xylanase obtained after 5 days of fermentation were 5,071 IU/g of rice straw and 14,790 IU l(-1) h(-1), respectively. The xylanase activity predicted by a polynomial model was 5,484 IU/g of rice straw.  相似文献

Summary A knowledge based system has been shown to be a powerful tool for diagnosing microbial activities during a fermentation process. Knowledge about lactic acid fermentation was collected by an experimental study ofLactobacillus casei. The effects of the inoculum properties and sterilization time on the cultivation were expressed in a form of a fuzzy rule-based knowledge network. The system was able to detect abnormal inoculum or sterilization conditions which caused malfunctions in the cultivations.  相似文献

Existence of autocrine growth factors (aGFs) may influence the serum requirement for growth of hybridoma cells and thus significantly influence process economics. For the murine hybridoma cell line S3H5/2bA2, critical inoculum density (cID) and serum requirement for growth were inversely related for cultivation in both T flasks and spinner flasks. In spinner flasks, an inoculum density of 106 cells/ml was necessary for the cells to grow in RPMI 1640 medium without serum supplement, and an inoculum density of 103 cell/ml was necessary in RPMI 1640 medium with 10% serum. In T flasks, where the local cell density is higher than in spinner flasks, an inoculum density of 106 cells/ml was necessary for the cells to grow in RPMI 1640 medium without serum supplement, and an inoculum density of 1 cell/ml was also necessary in RPMI 1640 medium with 10% serum. Further, immobilized cells at high local cell density could grow under conditions where cells in T flasks at corresponding overall cell density could not grow. The cells at high inoculum density were less sensitive to shear induced by mechanical agitation than the cells at low inoculum density. Taken together these observations support the existence of secreted aGF(s) by the hybridoma cell line used. Since the specific MAb production rate was independent of cultivation method and inoculum density, the existence of autocrine growth factors would suggest that the use of immobilized cells should improve the economics of MAb production.  相似文献

探索Bacillus pumilusWHK4以羽毛粉为底物产酶的最佳条件和最佳培养基组成。以羽毛粉发酵培养基为基础,首先采用单因子试验考察底物浓度、初始pH、接种量、外加碳源、外加氮源对WHK4产酶活力的影响。在单因子试验的基础上采用正交试验设计对底物浓度、温度、初始pH、接种量、外加(NH4)2SO4、外加麦芽糖进行优化。结果显示:Bacillus pumilusWHK4最佳的产酶条件为初始pH7.38,菌龄16 h,接种量5%,37℃。最佳的培养基组成为:1 L基础发酵培养基,40.0 g羽毛粉,10.0 g(NH4)2SO4和10.0 g麦芽糖。在优化的条件下Bacillus pumilusWHK4 24 h产蛋白酶活力为每毫升90 U。对培养条件和培养基的优化为Bacillus pumilusWHK4产蛋白酶的分离纯化奠定了基础。  相似文献

Industrially acceptable production of ergot alkaloids in surface culture under sterile conditions is described. After elucidation of the biochemical and morphological differentiation in the production microorganisms it is possible to elaborate a relatively simple cultivation method with good yields. Surface cultivation is especially advantageous for the production of spores in large quantities which are used as inoculum in industrial fermentations or as infection material (field production of ergot alkaloids, mycoinsecticides).  相似文献

Identical masses of submerged Trichoderma viride mycelia of various ages were used as inoculum for a second submerged cultivation lasting for 24 h. It was found that the growth yield of secondary culture was dependent on the age of inoculum. The growth yields increased when the age of primary culture was less than 3 d, and decreased down to zero when older mycelia were inoculated. The mycelia were living even after 1 month of submerged cultivation, as they formed conidia after inoculating onto solid medium. In order to elucidate underlying biochemical processes, developmental changes of specific activities of organellar marker enzymes were measured in the mitochondrial/vacuolar and microsomal fractions of mycelia. These activities changed during the growth of mycelia in a biphasic manner and their time courses were remarkably similar. Only the H+-ATPase activity decreased monophasically with the age of mycelia. Membrane-bound proteases of both membrane fractions changed differently upon ageing. These results could not be explained as a consequence of nutrient starvation and indicate that the prolonged submerged cultivation triggers coordinated series of biochemical events which leads to the loss of growth competence.  相似文献

High-density cultivation of Perilla frutescens cells for anthocyanin production was carried out in both batch and fed-batch modes in a 500-ml shake flask. In fed-batch cultures, a high cell density of 27.7 g dry cells l−1 and a total anthocyanin production of 3.87 g l−1 by intermittent feeding of all medium components except hormones were obtained. In batch cultures, both initial sucrose concentration and inoculum size showed a conspicuous effect on the kinetics of cell growth, sugar consumption, and secondary metabolite (anthocyanins) production by suspended P. frutescens cells. At an inoculum size of 50 g wet cells l−1, the maximum cell density of 38.3 g dry cells l−1 was obtained after 11 days of cultivation at an initial sucrose concentration of 60 g l−1, the highest pigment production of>5.8 g l−1 was attained after 10 days of cultivation at an initial sucrose concentration of 45 g l−1. These amounts of cell mass and anthocyanin pigments were 3.3 and 24 times higher than those at an initial sucrose concentration of 15 g l−1 and inoculum size of 15 g wet cells l−1, respectively.  相似文献

Inoculum size (1.5-6.0g dry weight/l) significantly affected cell growth and accumulation of intracellular and extracellular taxol in Taxus chinensis. A shorter cultivation time and a higher biomass productivity were achieved using inoculum size of 6.0g DW/l. Both the intracellular content and total production of taxol were increased almost 30% with an increase of inoculum size from 1.5 to 3.0g DW/l, while an even higher inoculum size decreased taxol formation. The extracellular taxol concentration was relatively higher (0.091mg/l) at low inoculum sizes of 1.5 and 2.0g DW/l; and in various cases it was less than 25% of the total amount of taxol produced.  相似文献

The optimal parameters for the cultivation in 10-l fermenters of a mutant strain Humicola lutea 120-5 were established:temperature 30°C, inoculum size 6%, inoculum age 24 h, aeration rate 0,6 vol/vol · min, medium agitation 620 rpm and cultivation time 72 h. A maximal proteolytic activity of 2000 µg tyrosine liberated from 2%casein ml?1 culture filtrate min?1 at pH 3.0 and 40°C was obtained under the fixed conditions. α-Amylase biosynthesis during the cultivation of H. lutea 120-5 was observed but it was insignificant to the 72nd h. It is demonstrated that starch can be used as alternative to glucose carbon source. It is proved that the mutant strain H.lutea 120-5 produced two acid proteinases.  相似文献

The effect of specific inductors at different stages of Streptomyces recifensis var. lyticus 2435 cultivation was studied. It is shown that introduction of killed bacterial cells into inoculum not only influences the level of accumulation of lytic enzymes by the strain S. recifensis var. lyticus 2435 but also determines the qualitative composition of the synthesized complex. The yield of bacteriological glycosidases grows with introduction of the Micrococcus lysodeikticus cells into the inoculum or of dissolvable chitin into the enzymic medium. The possibility to preserve the initial level of the lytic activity in the producer during re-inoculations by introducing the Bacillus subtilis cells into the cultivation medium has been studied.  相似文献
