The linkage of the different types of cytoskeletal proteins to cell adhesion structures at the cytoplasmic membrane and the connection of these contact sites to corresponding sites of adjacent cells is a prerequisite for integrity and stability of cells and tissues. The structurally most prominent types of such cell-cell adhesion complexes are the desmosomes (maculae adhaerentes), which are found in all epithelia and certain non-epithelial tissues. As an element of the cytoskeleton, intermediate filaments are connected to the adhesive desmosomal transmembrane proteins by the cytoplasmic desmosomal plaque proteins. At least three different types of proteins are found in the desmosomal plaque, one of which is represented by the plakophilins, a recently described sub-family of sequence-related armadillo-repeat proteins. Consisting of three isoforms, plakophilins (plakophilin 1 to 3, PKP 1 to 3) are located in all desmosomes in a differentiation-dependent manner. While PKP 2 and PKP 3 are part of almost all desmosome-bearing cell types (PKP 2 except for differentiated cells of stratified epithelia and PKP 3 for hepatocytes and cardiomyocytes), PKP 1 is restricted to desmosomes of cells of stratified and complex epithelia. Besides the architectural function that plakophilins seem to fulfill in the desmosomes, at least PKP 1 and 2 are also localized in the nucleus independently of any differentiation-related processes and with an up to now enigmatic function in this compartment. In the following article we want to summarize the current knowledge concerning structure, function and regulation of the plakophilins that has been achieved during the last decade.  相似文献

Summary The cell junctions of the cyst envelope in the testes of Bombyx mori were examined by electron microscopy utilizing a thin-sectioning technique following conventional fixation, tannic acid fixation and lanthanum tracer study, and also using a freeze-fracture technique. There are three kinds of junctions; septate junctions, gap junctions and tight junctions. Septate junctions are of the pleated type. Gap junctions are characterized by four electron-dense lines and three electronlucent lines in the reduced intercellular spaces seen by thin-sectioning. They are of the E type, having clusters of intramembraneous particles on the E-fracture face. The most striking finding is the frequent presence of tight junctions on the fracture planes, while focally fused outer leaflets of the junctional unit membranes are rarely detected on thin-sectioned preparations. Tight junctions are characterized by branching zigzag ridges on the P-fracture face and complementary grooves on the E-fracture face. It is proposed that tight junctions are new morphological evidence of blood-germ cell barrier in an insect. Vegetation changes during the Holocene in the North Iberá, Corrientes, Argentina. Wetlands are very important sites for palynological studies, since they represent one of the most suitable environments for fossil pollen preservation. The aim of this work was to determine, by palynological analysis of lacustrine sediments, the vegetal communities and the predominant environment during the Holocene in NW of Iberá. Two lagoons were studied: San Sebastián and San Juan Poriahú. Sediment samples were obtained with witness using a "Levingstone square-rod sampler", processed with Faegri e Iversen techniques and dated with C14. The palynological graphs were divided in zones using the Tilia program. The palynological analysis allowed visualizing diverse changes in the vegetation: from 6 140 +/- 50 to 5 170 +/- 100 a. C., the NW of Iberá was characterized by marsh-herbaceous vegetation and arboreal vegetation typical of dry vegetation. From 5 170 +/- 100 to 3 460 +/- 60 a. C., a decrease in the species frequency, typical of wet environments, is produced, and the clogging of the waterbody, from 3460 +/- 60 a. C. onwards, while continuing the dominance of herbaceous vegetation typical of these environments, the arboreal pollen, indicates the beginning of a hygrophilous forest development.  相似文献

Oscillations of the “magnetosphere-solar wind” system are studied analytically in the framework of a plane-stratified model of the medium. The properties of oscillations are determined by three phenomena: Kelvin-Helmholtz instability on the tangential discontinuity (magnetopause) separating the magnetosphere and the solar wind, the presence of a waveguide for fast magnetosonic waves in the magnetosphere, and the Alfvén resonance—a sharp increase in the amplitude of oscillations having the properties of Alfvén waves—in the inner magnetosphere. The oscillations of the system form a discrete spectrum of eigenmodes. Analytical expressions are obtained for the frequency and growth rate of instability of each mode, as well as for the functions describing the spatial structure of these modes. All these characteristics of the eigenmodes are shown to depend on the velocity of the solar wind as a parameter. The dependences of the main mode characteristics (such as the instability thresholds, the points of the maximum and minimum growth rate, and the spatial distributions of the oscillation energy) on this parameter are determined for each eigenmode.  相似文献

Cultural and environmental factors interact in determining the genetic structure of human populations. Bio-demographic investigations of ethnic minorities are able to disentangle the influences that these two components have on the evolution of the genetic structure of a population. The ethnic minority of the Arb?reshe of the province of Cosenza (Calabria, southern Italy) is analyzed in this paper and its bio-demographic structure in the early 1800s is compared with that of neighboring Italian populations. The data derive from surnames recorded in the birth registers of the 19 Arbdreshe municipalities of the province of Cosenza and in 5 non-Arb?reshe municipalities of the same province. Isonymy and repeated pairs of surnames are used to analyze the bio-demographic structure of these populations, while analysis of isonymic relationships is used to investigate the variability between populations. Higher values of marital isonymy and subdivision into subpopulations characterize the Arb?reshe populations with respect to their non-Arb?reshe neighbors. However, the high range of variability of these parameters suggests a strong influence of geographic location on the marriage pattern of each community. At the same time, cultural differences linked to group identity had a strong impact in limiting marriage exchanges between the different ethnic groups living in the province of Cosenza in the early 1800s. In fact, the analysis of isonymic relationships demonstrates that geographic location shaped kinship patterns among the Arbereshe communities, but it also shows that the non-Arb?reshe neighbors formed a clearly separate reproductive cluster.  相似文献

Some of the recent commitment by the government may be the result of enlightened thinking, but some of it has resulted from the aftermath of at least two major healthcare incidents. The acquisition of hypsodont molars is often regarded as a key innovation in the history of ruminant ungulates. Hypsodont ruminants diversified rapidly during the later Neogene, circa 15-2 Myr ago, and came to dominate the ruminant fossil record in terms of species diversity. Here we show that hypsodont clades had higher speciation and diversification rates than other clades. Hypsodont species had, on average, shorter stratigraphic durations, smaller range size and lower occupancy than non-hypsodont species. Within hypsodont clades, some species were very common and acquired large geographical ranges, whereas others were quite rare and geographically limited. We argue that hypsodont clades diversified in an adaptive radiation-like fashion, with species often splitting cladogenetically while still in the expansive phase of their occupancy history.  相似文献

Surnames analysis is useful for populations in which only written documents remain, as is the case for historical populations. In Córdoba province, Argentina, census records contain nominal data of inhabitants, including information on sex, age, ethnosocial category, civil status, occupation, place of birth, and residence, that can be analyzed using surnames. Relationship indicators within and among ecclesiastic units in Córdoba were estimated by isonymy for the adult white population registered in the 1813 census. The Rii, Rij, and R(ST) coefficients and the surname abundance indicator (a) were calculated. Lasker's distances among categories of population units were used to cluster the 16 provincial population categories. Gradients for kinship within population and for surname diversity were in agreement with the principal areas and waves of original settlement in the province. The main population clusters reflect those areas, whereas minor clusters coincide with the network of roads existing in the territory by 1813. The structure of the white population in Córdoba province was determined by the geographic location of the original waves of settlement, and it followed a pattern of relationships conditioned by the routes connecting population units in the Colonial period.  相似文献

Cathepsins D, K, and L were immunolocalized in tissue undergoing endochondral ossification in the human. Cathepsins D, K, and L were localized in osteoclasts and chondroclasts attached to bone matrix and cartilage matrix, respectively. Cathepsins D and L were immunostained in chondrocytes. Immunolocalization of cathepsin D was limited to hypertrophic chondrocytes adjacent to the osteochondral junction. In contrast, cathepsin L was immunolocalized in both proliferating and hypertrophic chondrocytes. In the bone marrow space, cathepsins D, K, and L were localized in multinucleated cells. Cathepsin D was diffusely detected in mononuclear bone marrow cells which were negative for cathepsins K and L. The present findings indicated that cathepsins K, D, and L were associated with the process of endochondral ossification in the human, and suggested that these cathepsins share roles in bone and cartilage turnover in the human.  相似文献

Archaeobotanical investigations at Qara Qorum (Karakorum), Mongolia, reveal information about diet and land use from the 13th to the 15th century A.D. People grew Panicum miliaceum, Hordeum vulgare, Triticum aestivum and Setaria italica in nearby irrigated fields but additionally imported all other known cereals, including Oryza sativa, in small amounts as well as oil and fibre plants and pulses. The most common oil and fibre plant was Cannabis sativa. At least ten species of vegetables and spices such as Carum carvi, Coriandrum sativum, Apium graveolens, Beta vulgaris, Lycium chinense and Piper nigrum were either gathered from the wild, grown locally or imported. Apart from some wild gathered species like Pinus sibirica and Fragaria vesca, most of the fruits and nuts as for instance Vitis vinifera, Ficus carica, Ziziphus jujuba, Prunus dulcis, P. insititia, P. avium and P. persica, Cucumis melo and Juglans regia must also have been imported from quite long distances. First pollen results from lake Ugii Nuur, 50 km north of Qara Qorum indicate a much earlier beginning of agriculture than in the high and late Medieval.  相似文献

A block to polyspermy is required for successful fertilisation and embryo survival in mammals. A higher incidence of polyspermy is observed during in vitro fertilisation (IVF) compared with the in vivo situation in several species. Two groups of mechanisms have traditionally been proposed as contributing to the block to polyspermy in mammals: oviduct‐based mechanisms, avoiding a massive arrival of spermatozoa in the proximity of the oocyte, and egg‐based mechanisms, including changes in the membrane and zona pellucida (ZP) in reaction to the fertilising sperm. Additionally, a mechanism has been described recently which involves modifications of the ZP in the oviduct before the oocyte interacts with spermatozoa, termed “pre‐fertilisation zona pellucida hardening”. This mechanism is mediated by the oviductal‐specific glycoprotein (OVGP1) secreted by the oviductal epithelial cells around the time of ovulation, and is reinforced by heparin‐like glycosaminoglycans (S‐GAGs) present in oviductal fluid. Identification of the molecules contributing to the ZP modifications in the oviduct will improve our knowledge of the mechanisms of sperm‐egg interaction and could help to increase the success of IVF systems in domestic animals and humans.  相似文献

The data on the composition and abundance of nekton species and their interannual variations within the upper epipelagic layer (0–50 m) in the Aleutian and Commander basins in the western Bering Sea that are considered in the article were collected during complex surveys that were conducted by the Pacific Research Fisheries Center in September and October of 2002–2013. The core species structures of the nekton communities in these two areas were similar: the chum salmon Oncorhynchus keta and the boreopacific gonate squid Boreoteuthis borealis were the most abundant species. Simpson’s dominance index varied synchronously in both areas, with higher values in the Aleutian Basin, until 2009. In the subsequent years, the values of the index for the studied areas became equal and varied asynchronously. An analysis of abundance showed that two types of species structure most often prevailed (with the dominance of either chum salmon or squid); in some years the species structure differed from that in other years. Species of the low-boreal and low-boreal-subtropical complexes were more abundant in the Commander Basin, reaching particularly high proportions in 2006, 2008, and 2012. Certain species showed similarity in the year-to-year dynamics of their abundance; however, these coincidences were frequently accidental. After the climate regime shift in 2006–2007, the total biomass of nekton in the Aleutian Basin decreased from 3241 to 1736 kg/km2 (46%); in the Komandorskiye Basin it decreased from 2459 to 1976 kg/km2 (20%).  相似文献

Summary The guinea-pig intestine was found to harbor nerve fibers containing immunoreactive cholecystokinin (CCK), gastrin-releasing peptide (GRP), neurotensin or - endorphin. Such fibers occurred in the myenteric and submucous ganglia and in the smooth muscle. GRP- and CCK-fibers, in addition, were found in the mucosa. Following colchicine treatment, neuronal perikarya in the myenteric ganglia displayed CCK-, GRP-, or -endorphin immunoreactivity. CCK-immunoreactive perikarya were located also in the submucous ganglia. Neurotensin-immunoreactive cell bodies could not be detected. The presence of immunoreactive neuronal perikarya in intramural ganglia indicates that CCK-, GRP- and -endorphin-containing fibers are intrinsic to the gut wall. GRP, neurotensin, and -endorphin were identified in extracts of smooth muscle by immunochemical and Chromatographic analysis.CCK-8, GRP and neurotensin contracted the isolated taenia coli. Tetrodotoxin reduced the response to CCK-8 but not that to GRP and neurotensin, suggesting that the two latter peptides act directly on smooth muscle receptors. The effect of CCK-8 is partly mediated by cholinergic nerves, since not only tetrodotoxin but also atropine greatly reduced the CCK-8-induced contractile response. The substance P (SP) antagonist, (d-Pro2, d-Trp7,9)-SP1–11 had no effect on the CCK-8-induced contraction of the taenia. CCK-8 enhanced the SP-mediated (atropine-resistant) contractile response to electrical stimulation but not that mediated by acetylcholine. -Endorphin had no effect on the tension of the muscle but reduced the response to electrical stimulation (cholinergic as well as SP-mediated) through a naloxone-sensitive mechanism.While CCK-8 and -endorphin seem to play neuromodulatory roles in the taenia coli, the significance of GRP and neurotensin remains enigmatic.  相似文献

The relationship between myonuclear number, cellular size, succinate dehydrogenase activity, and myosin type was examined in single fiber segments (n=54; 9±3 mm long) mechanically dissected from soleus and plantaris muscles of adult rats. One end of each fiber segment was stained for DNA before quantitative photometric analysis of succinate dehydrogenase activity; the other end was double immunolabelled with fast and slow myosin heavy chain monoclonal antibodies. Mean±S.D. cytoplasmic volume/myonucleus ratio was higher in fast and slow plantaris fibers (112±69 vs. 34±21x103 m3) than fast and slow soleus fibers (40±20 vs. 30±14x103 m3), respectively. Slow fibers always had small volumes/myonucleus, regardless of fiber diameter, succinate dehydrogenase activity, or muscle of origin. In contrast, smaller diameter (<70 m) fast soleus and plantaris fibers with high succinate dehydrogenase activity appeared to have low volumes/myonucleus while larger diameter (>70 m) fast fibers with low succinate dehydrogenase activity always had large volume/myonucleus. Slow soleus fibers had significantly greater numbers of myonuclei/mm than did either fast soleus or fast plantaris fibers (116±51 vs. 55±22 and 44±23), respectively. These data suggest that the myonuclear domain is more limited in slow than fast fibers and in the fibers with a high, compared to a low, oxidative metabolic capability.  相似文献
