采集生长于恶劣环境和中生环境的普通木麻黄(Casuarina)小枝,超速离心提取粗质膜制剂后,用两相系统法纯化得到质膜微囊,研究不同生境下木麻黄的质膜ATPase活性,并测定木麻黄小枝的游离脯氨酸含量。实验结果表明:同一生境中的木麻黄ATPase活性相对一致,而同一树种木麻黄不同生境下质膜微囊H~+-ATPase、Ca~(2+)-ATPase、K~+-ATPase活性有显著差异,表现出以渗透胁迫为主的恶劣环境下的木麻黄质膜微囊ATPase活性和木麻黄细胞内游离脯氨酸明显高于中生环境下生长的木麻黄。说明普通木麻黄在干旱和盐胁迫下能调整生理生化过程来提高其质膜ATPase活性和增加细胞内脯氨酸含量提高渗透调节能力以保证其在恶劣环境的正常生长。 相似文献

以小麦为实验材料,研究了盐胁迫对根质膜H^ —ATPase、Ca^2 —ATPase活性及H^ —ATPase蛋白表达的影响。结果显示:50、100、150mmol/L的NaCl处理72h后,小麦根质膜H^ —ATPase、Ca^2 —ATPase活性均降低。100mmol/L NaCl对质膜ATPase活性的抑制程度随处理时间的延长而增强,在处理24h后,H^ —ATPase和Ca^2 —ATPase的活性分别降为对照的72%和75%,而处理72h后,酶活性分别减小到对照的50%和48%。50、100、150mmol/L的NaCl直接作用于提取的质膜微囊,H^ —ATPase的活性分别降低约5%、8%和16%。Western blotting分析结果显示100mmol/L NaCl处理72h后,质膜H^ —ATPase的含量与对照比有所减少。本研究表明:盐胁迫抑制小麦根质膜H^ —ATPase、Ca^2 —ATPase的活性,酶含量的减少可能是盐胁迫导致质膜H^ —ATPase活性降低的原因。 相似文献

以耐冷性不同的两个水稻品种为材料,比较研究了幼苗根系质膜、液泡膜ATP酶对低温(8℃)及高pH(8.0)胁迫的反应。结果表明:水稻根细胞质膜和液泡膜上均存在Ca^2+-ATP酶,但活性远低于H^+-ATP酶。耐冷品种武育粳3号经低温(8℃)处理2d,根系质膜和液泡膜H^2+-ATP酶、Ca^2+-ATP酶海性均明显升高,至冷处理12d,H^+-ATP酶、Ca^2+-ATP酶活性有所下降,但仍与对照 相似文献

以盐生植物盐芥和中生植物拟南芥幼苗为材料,研究了盐胁迫对它们叶片和根质膜、液泡膜H+-ATPase、Ca2+-ATPases和K+-ATPase活性以及H+-ATPase、Na+/H+ 逆向转运蛋白表达的影响.结果显示:在NaCl胁迫下,盐芥叶片和根质膜的H+-ATPase活性分别比对照显著升高41%~212%和35%~53%,液泡膜的H+-ATPase分别显著升高281%~373%和4%~38%,而拟南芥却比相应对照都显著降低;相同盐浓度胁迫下,盐芥叶片的H+-ATPase活性比根部高4~8倍,盐芥根也远高于拟南芥.在NaCl胁迫下,盐芥叶片和根的液泡膜H+-ATPase蛋白质β亚基含量变化与其酶活性变化趋势一致,质膜Na+/H+ 逆向转运蛋白的表达量与Na+含量变化趋势一致.盐胁迫下盐芥根中Ca2+-ATPases和K+-ATPase活性的增加与根中Ca2+和K+含量呈显著正相关.研究发现,在盐胁迫条件下,盐芥能有效增强H+-ATPase蛋白和Na+/H+逆向转运蛋白表达,显著提高其根系与叶片质膜和液泡膜的H+-ATPase、Ca2+-ATPase和K+-ATPase活性,维持细胞质中较高的Ca2+和K+水平,从而缓解盐胁迫的伤害,增强耐盐性. 相似文献

以三叶期的水稻幼苗为材料,研究了干旱(-0.6MPa PEG模拟)、盐(0.15ml/L NaCl)和低温(6℃)胁迫下,不同水稻品种脯氨酸积累的变化。结果表明,干旱、盐和低温胁迫下稻苗均可积累脯氨酸,且随着胁迫时间的处长而加剧。在同一胁迫条件下,耐性强的品种脯氨酸积累较少,而敏感品种脯氨酸积累则较多。脯氨酸的积累不宜作为稻苗抗逆性的筛选指标。 相似文献

In this study an experiment was carried out to study the process of stress adaptation in Groenlandia densa (opposite-leaved pondweed) grown under cadmium stress (0–20 mg L?1 Cd). The results showed that Cd concentrations in plants increased with increasing Cd supply levels and reached a maximum of 0.43 mg kg?1 DW at 0.5 mg L?1 Cd concentrations. The level of photosynthetic pigments and soluble proteins decreased only upon exposure to high Cd concentrations. At the same time, the level of malondialdehyde (MDA) increased with increasing Cd concentration. These results suggested an alleviation of stress that was presumably the result of by antioxidants such as superoxide dismutase (SOD), catalase (CAT), glutathione reductase (GR) and glutathione S-transferase (GST) as well as ascorbate peroxidase (APX), which increased linearly with increasing Cd levels. Cellular antioxidants levels showed a decline suggesting a defensive mechanism to protect against oxidative stress caused by Cd. In addition, the proline content in G. densa increased with increasing cadmium levels. These findings suggest that G. densa is equipped with an efficient antioxidant mechanism against Cd-induced oxidative stress which protects the plant's photosynthetic machinery from damage.Our present work concluded that G. densa has a high level of Cd tolerance and accumulation. We also found that moderate Cd treatment (0.05–5 mg L?1 Cd) alleviated oxidative stress in plants, while the addition of higher amounts of Cd (10–20 mg L?1) could cause an increasing generation of ROS, which was effectively scavenged by the antioxidative system. 相似文献

Summary An increase in the specific activity of kinase and transferase is observed when late growth phase is compared with early exponential phase. The increase is more pronounced in the presence of saturating inducer concentrations than in uninduced cultures. Epimerase does not increase in late growth phase in the presence of D-fucose, provided the other enzymes of the galactose pathway are present. It is believed that increases observed are due to an increased rate of synthesis of the galactose enzymes in late growth phase, and that the lack of coordination found for epimerase is due to an inactivation of the latter enzyme, which occurs in the presence of D-fucose and needs an intact galactose pathway. 相似文献

The levels of some enzymatic activities involved in protoheme synthesis have been measured in subcellular fractions obtained at different stages of the growth of the yeast Saccharomyces cerevisiae grown anaerobically and aerobically with glucose (50 or 6 g/ liter), and ethanol (20 g/liter) as the carbon source. The degree of repression of the respiratory system is estimated by the respiratory capacity of whole cells, by the activities of succinate-cytochrome c reductase and cytochrome c oxidase of the mitochondrial particles, and by the cytochrome spectra. The results show that (i) the more porphyrins (cytochromes) that are synthesized by the cells, the lower is the specific activity of δ-aminolevulinic acid (ALA) synthetase and the higher is the specific activity of ALA dehydratase, the activity ratio ALA synthetase/ALA dehydratase decreasing at least 10-fold compared to the repressed cells; (ii) the amount of intracellular ALA found under all conditions tested (from 0.05 to 1.5 mm in the cell sap) correlates well with the measured ALA synthetase activity; its presence argues against a rate-limiting function for ALA synthetase and rather favors such a role for the ALA dehydratase in the formation of heme in yeast; (iii) the rate of porphyrin synthesis measured in vitro is higher in the case of cells with high cytochrome contents; and (iv) the specific activities of succinyl CoA synthetase and protoheme ferrolyase are always present in nonlimiting amounts. Some experiments are described showing that the values of the activities which are calculated from these in situ and in vivo experiments compare well with the values measured in vitro in the acellular extracts. The results concerning the enzymatic activities, together with (i) the excretion of coproporphyrin(ogen) and the accumulation of protoporphyrin + Zn-protoporphyrin in anaerobiosis, (ii) the presence of protoporpho(di)methene (P503) in anaerobic and repressed cells, and (iii) the presence of intracellular ALA under all growth conditions, are discussed in terms of possible control(s) of heme synthesis in yeast. 相似文献

Summary Free proline accumulation was measured in leaves of intact sorghum (Sorghum bicolor L. cv. Pioneer 846) and soybean (Glycine max. L. cv. Calland) grown in growth chambers and subjected to ‘normal’ drought stress. Stomatal diffusive resistance and leaf
water potential were used to determine the degree of stress at the time of proline analysis.
Free proline did not accumulate markedly in either species until each was severely stressed, indicating that proline is not
a sensitive indicator of drought stress. Free proline accumulated under less stress in soybean than in sorghum. Since soybean
is less drought resistant than sorghum, proline accumulation may be an indicator of drought resistance or susceptibility.

Basal and Na+-K+ stimulated ATPase (ATP phosphohydrolase, E.C. 3.6.1.3) are both present in isolated preparations of purified cell wall and plasma membrane from cotyledon tissue of Phaseolus vulgaris. A comparison of the enzymes in the two fractions has revealed that the specific activities of basal and cation-sensitive ATPase are markedly higher in isolated cell wall than in the plasma membrane fraction. In addition, enrichments of both enzymes calculated on a protein basis relative to corresponding homogenates were considerably higher for cell wall than for plasma membrane. Thus, while part of the ATP-hydrolyzing activity of the wall may be attributable to the enzymatic properties of imbedded plasma membrane, there must also be additional non-membranous ATPase in the protein complement of the wall itself. 相似文献

Changes in photosynthetic performance, osmolyte accumulation and the activities of superoxide dismutase (SOD), ascorbate peroxidase (APX), catalase (CAT) and polyphenol oxidase (PPO) were investigated in one-year-old olive cultivars (Chemlali, Meski and Picholine) subjected to contrasting water availability regimes under arid climatic conditions in Tunisia. Shoot elongation rates (SER) and photosynthetic performance were markedly reduced by the water deficit regime (WD) in all cultivars except for Chemlali, which proved to be superior to the other two cultivars with respect to drought tolerance. Higher photosynthetic performance (net photosynthesis (Pn), stomatal conductance (gs) and transpiration rates (E)) in the Chemlali and Meski cvs. compared to Picholine olive allowed them to maintain better plant water status and shoot elongation rates. Under WD conditions, Chemlali showed a greater capability for proline accumulation. Leaves grown under WD conditions showed signs of oxidative stress such as reduced chlorophyll and carotenoid concentrations. Nevertheless, different cultivars developed certain antioxidative defense mechanisms, including elevated SOD, APX and CAT activities. In contrast, PPO activity decreased under WD circumstances. Comparatively, Chemlali olive displayed better antioxidative enzyme activity, and thus better protection against oxidative stress. These results show that the ability of olive trees to up-regulate the enzymatic antioxidative system might be an important attribute linked to drought tolerance. These findings demonstrate that the association of higher Pn, proline accumulation and antioxidative defenses could be effective in a water-limited environment and may be useful selection criteria in breeding programs with the objective of improving drought tolerance and growth of olive trees, at least under the described environmental conditions. 相似文献

The activity of the plasma membrane ATPase in growing yeast is increased by low pH and by glucose, conditions which result in a higher demand for proton pumping. The amount of enzyme is not significatively modified under these conditions. The amount of ATPase is only slightly increased by introducing extra copies of its gene in autonomous plasmids. In addition, the expression of the ATPase gene in a multi-copy plasmid causes a reduction of the copy number of the plasmid and slows growth. Therefore, overexpression of the ATPase is detrimental for the cell, justifying a regulatory mechanism based on increasing the catalytic activity and not the amount of enzyme. 相似文献

Changes in activities of plasma membrane enzymes during liver regeneration may be related to the maintenance of hepatic function or to the regulation of cell proliferation. Plasma membranes were isolated from rat livers at various times after partial hepatectomy, and the specific activities of alkaline phosphatase, (Na+ + K+)-ATPase, leucine aminopeptidase, 5'-nucleotidase, and adenylate cyclase (basal and with glucagon or epinephrine) were measured. Alkaline phosphatase and (Na+ + K+)-ATPase activity increased 3.6-fold and 2-fold respectively, during the first 48 h after partial hepatectomy. The time of onset and duration of change suggest that these increases in activity are involved in the maintenance of bile secretion. Decreases in leucine aminopeptidase activity at 48--108 h and in 5'-nucleotidase activity at 12--24 h were observed, which may be involved in the restoration of protein and accumulation of RNA. The basal activity of adenylate cyclase increased after partial hepatectomy. The response of adenylate cyclase to epinephrine showed a transitory increase between 36 and 108 h after surgery, while the response to glucagon was decreased by approximately 50% at all time points through 324 h after surgery. These changes in the hormone responsiveness of adenylate cyclase are similar to those previously observed in fetal and preneoplastic liver. 相似文献

A variety of the biochemical properties of the electrogenic plasma membrane ATPase of Neurospora crassa are described. The enzyme catalyzes the hydrolysis of ATP, resulting in the formation of ADP and inorganic phosphate. Optimal activity is observed between pH 6 and 6.5. ATP hydrolysis approaches a maximum rate at an Mg-ATP concentration of 10–20 mm with a half-maximal velocity around 2 mm Mg-ATP. The enzyme requires a divalent cation for activity in the following order of preference at 10 mm: Mg2+, Co2+ > Mn2+ > Zn2+ > Fe2+, Ca2+, Cu2+. The enzyme is quite specific for ATP compared to the other nucleotides tested. Treatment of the plasma membranes with sodium deoxycholate inactivates the ATPase and the inactivation can be prevented by the addition of certain acidic phospholipids with the deoxycholate. Other classes of lipids cannot prevent the deoxycholate inhibition. The organic mercurials parachloromercuribenzoate and parachloromercuriphenylsulfonate are potent inhibitors of the ATPase, but N-ethylmaleimide at a similar concentration is not inhibitory. The organic mercurial inhibition is not reversed by mercaptoethanol. Under appropriate conditions, the inhibitory effect of p-chloromercuribenzoate is suppressed in the presence of ATP. Treatment of the plasma membranes with trypsin leads to a marked inhibition of the ATPase activity and this inhibition can be prevented by Mg-ATP. Neither the organic mercurial reactive site(s) nor the trypsin-sensitive site(s) are accessible from the outer surface of the plasma membranes.
