Littorina tenebrosa is a small fragile-shelled periwinkle which lives on permanently submerged algae in coastal lagoons and non-tidal brackish pools. This periwinkle is a member of the rough periwinkle group which also comprises Littorina saxatilis, L. arcana, L. compressa and L. neglecta and is most closely related to L. saxatilis although its exact systematic status is in some doubt. Based on its unique ecology many believe L. tenebrosa to be a valid species. However, shell morphometric and allozyme analyses on Scottish and Swedish populations of L. tenebrosa and L. saxatilis have indicated that the two periwinkles are virtually identical. Preliminary results on five allozyme loci (AAT-1, GPI, PGM-1, PGM-2 and PNP) in samples of L. tenebrosa and L. saxatilis from Golam Head, Lettermullen, and other locations on the west coast of Ireland show L. tenebrosa to be genetically differentiated from L. saxatilis At Golam Head, where opportunities for gene flow occur between the two taxa, L. tenebrosa is as genetically differentiated from local L. saxatilis as it is from L. saxatilis from more geographically distant locations.  相似文献

Twenty-three enzymes and five shell parameters were screened in 11 subpopulations of Littorina saxatilis Olivi (= L. rudis Maton) occupying different habitats over a 1 km stretch of coastline. Shell morphology varied considerably and consistently with respect to degree of exposure, and since there is evidence that such morphology is at least partly under genetic control, it is likely that natural selection selects particular genotypes at particular locations. There was significant allozyme heterogeneity between neighbouring subpopulations, sometimes only metres apart, but little of the allozyme variability could be related directly to environmental pressures. Thus, with the exception of the Odh locus, the considerable morphological differentiation between snails from exposed and sheltered sites was not reflected in differentiation of those genes coding for electrophoretically assayed enzymes. At the Odh locus, virtually all the genetic differentiation between subpopulations was attributable to differentiation between habitat types. Two loci, Sod-1 and Aat-1, showed highly significant genetic disequilibrium, and possible reasons for this are explored. The population structure as assessed electrophoretically accords well with the stepping-stone model which permits greater differentiation of neighbouring populations than the island model, and which seems realistic in the ovoviviparously reproducing L. saxatilis, where the greater part of gene flow is likely to occur through the occasional migration of adults between contiguous populations.  相似文献

Studies of allozyme variation may reveal unexpected patterns of genetic variation which challenge earlier conclusions of species delimitations based on morphological data. However, allozyme variation alone may not be sufficient to resolve this kind of problem. For example, populations of the marine intertidal snail Littorina fabalis (=Littorina mariae) from wave exposed parts end from protected parts of the same shores are distinguished by different alleles of arginine kinase (Ark) while indifferent, or very nearly so, in another 29 loci. Intermediate populations have large deficiencies of exposed/sheltered heterozygote classes of Ark and we have earlier suggested habitat-related selection in this locus as the explanation. In this study we estimated growth rate of individual snails of different Ark-genotypes in three different habitats (exposed, sheltered and intermediate). In all habitats the snails homozygous for alleles of ‘exposed’ type grew faster and matured at a larger size than did snails homozygous for alleles of ‘sheltered’ types. This relationship was indirectly confirmed in three additional sites of intermediate exposure where exposed AA-genotypes dominated among large (>8 mm) snails while the sheltered genotypes dominated among small (<5 mm) snails of truly sympatric samples. We furthermore found small differences in allele frequencies of two other loci (Pgi and Pgm-2) and in shell colour frequencies, comparing sympatric snails of exposed and sheltered Ark-homozygotes. Although we found no signs of habitat-related selection among snails of different Ark-genotype, or selection against heterozygotes, we cannot reject selection in Ark, as our experiments only covered one island, one season and grown-up snails. The coupling between allozyme and phenotypic characters in strictly sympatric samples of snails suggests the presence of two gene pools. Perhaps the large and small forms of L. fabalis represent very closely related cryptic taxa. However, introgression between them seems a possible explanation for the striking similarities in the vast majority of morphological and allozyme characters.  相似文献

Two South African populations of Littorina saxatilis were examined by starch-gel electrophoresis at 16 enzyme loci and compared with 13 populations of North Atlantic saxatilis from both American and European coasts, and with six British populations of the closely related species Littorina arcana. The South African animals showed a severely reduced heterozygosity (– 0.052) compared with Atlantic populations of saxatilis ( = 0.181), and the mean genetic distance between the two areas was high ( = 0.203) compared with distances within the North Atlantic saxatilis populations (D = 0.034). In fact, the saxatilis from South Africa were genetically more distant from the North Atlantic samples of L. saxatilis than were the arcana from British shores. The reduced genetic heterozygosity and genetic divergence of the South African populations is attributed to founder effects following a postulated recent introduction by man.  相似文献

The population genetic structure of marine species lacking free-swimming larvae is expected to be strongly affected by random genetic drift among populations, resulting in genetic isolation by geographical distance. At the same time, ecological separation over microhabitats followed by direct selection on those parts of the genome that affect adaptation might also be strong. Here, we address the question of how the relative importance of stochastic vs. selective structuring forces varies at different geographical scales. We use microsatellite DNA and allozyme data from samples of the marine rocky shore snail Littorina saxatilis over distance scales ranging from metres to 1000 km, and we show that genetic drift is the most important structuring evolutionary force at distances > 1 km. On smaller geographical scales (< 1 km), divergent selection between contrasting habitats affects population genetic structure by impeding gene flow over microhabitat borders (microsatellite structure), or by directly favouring specific alleles of selected loci (allozyme structure). Six populations of Littorina rudis and ihree of L. arcana were screened for elcctrophorctically dctci table variation at 21 enzyme loci. These species reproduce by ovoviviparity and oviparily respectively, and both are members of the L. saxatilis species complex. The mean coefficient of genetic identity between the two species was high (0.957) and no diagnostic loci were observed. Allele frequency differences are maintained in sympatric populations of the two species. Intraspecific values of/ ranged from 0.944 to 0.995 and both species showed marked inter-population genetic heterogeneity, a feature consistent with their limited powers of dispersal. Heterozygosity is high in all populations of both species. Populations of I. rudis have higher mean heterozygosities (= 0.153) then L. arcane ( H = 0.132).  相似文献

Earlier studies of Swedish populations of the marine snail Littorina fabalis show that snails from different microhabitats (with a greater and lesser exposure to wave action) have almost diagnostic differences in one allozyme locus (arginine kinase, Ark ), and differ in adult size. Snails with 'sheltered' and 'exposed' Ark genotypes occur in sympatry in intermediary exposed sites and here adult sizes remain distinct. Approaching the microgeographic differentiation we studied the parts of two populations where the frequency of Ark changes dramatically over zones 50–120 m wide. The aim was to test if the transitional zones are best described as areas of mixing of two genetically separate populations, or if hybridization between the exposed and sheltered groups occurs. Heterozygotes were in deficiency along both clines but were still roughly twice as common as expected from a pure mixing of 'sheltered' and 'exposed' groups suggesting hybridization. Hybridization was also supported by the observation that snails homozygous for sheltered and exposed alleles mated at random with each other in both populations. On the macrogeographic scale, we found populations from exposed and sheltered sites in France and Wales being fixed for the same exposed and sheltered Ark alleles as found in Sweden. However, variation in three other highly polymorphic loci indicated geographic affinity rather than habitat similarity being the main factor of genetic coherence. These observations support a hypothesis of gene flow between exposed and sheltered populations of L. fabalis . Two Spanish populations were remarkably different with unique alleles at high frequencies in three of four strongly polymorphic loci.  相似文献

Allozyme variation in natural populations of basidiomycetes fungus Pleurotus ostreatus (88 individuals) from three regions of central Russia was studied. The species was shown to have 92.86% of polymorphic allozyme loci and expected heterozygosity He = 0.49. The mean number of alleles per locus was 3.5. The genetic differences among populations were supported by F-statistics (FST = 0.750). The low level of inbreeding (FIS = 0.018) suggests that the P. ostreatus populations are panmictic, and the main reproduction mode involves basidiospores dispersing at long distances. Using cluster analysis, geographically isolated populations and intersterile groups were differentiated within the complex P. ostreatus species.  相似文献

12 rocky shore populations of Littorina saxatilis from three islands of Chupa Inlet (Kandalaksha Bay, White Sea) were examined for infection with trematodes. Morphometric characters (6 indexes of the shell and aperture shape) of molluscs were investigated for all these populations. Exposed and sheltered sites were considered at every island and high and low littoral samples were fulfilled at every site. Seven species of trematodes, Podocotyle atomon, Cryptocotyle lingua, Renicola sp., Himasthla sp., Microphallus piriformes, M. pygmaeus, M. pseudopygmaeus, were found. Uneven distribution of trematodes was confirmed by log-linear analysis. Sheltered populations of L. saxatilis have the greater infection prevalence than exposed ones. This is due to the heavy infection with M. piriformes and M. pygmaeus. The prevalences by these trematodes are up to 52.97% and 27.16% respectively in sheltered populations of the host. The prevalence of M. piriformes tend to be higher at the upper shore level of sheltered sites. In a contrast, the prevalence of M. pygmaeus is significantly higher at the low part of such sites. Factor analysis shows a significant association of the indices of L. saxatilis shell shape with three factors. The first one is associated with the "elongation" of a shell and reveals L. saxatilis from the exposed rocky shore to be more elongated than the molluscs from sheltered sites. The second one is connected with the "aperture shape" index. There is an association of this factor with the shore level position of samples. The third factor reflects the affect of trematodes on the shell shape. The molluscs infected with M. piriformes show "elongated" shell shape and relatively smaller aperture. Shall peculiarities of the hosts infected with M. piriformes and M. pygmaeus are somewhat different. The results of the factor analysis is justified by the series of analysis of variances on the values of shell indices (MANOVA) according to the factors "exposure", "shore level" and "infection".  相似文献

The genetic structure of Pogonus chalceus from 11 Atlantic and seven Mediterranean Western European populations was analysed. Results from seven allozyme loci were compared to five microsatellites to test the hypothesis that some enzymatic loci undergo selection. Both allozyme and microsatellite results showed that Mediterranean beetle populations are genetically distinct from Atlantic populations. The analysis of the genetic structure showed that FST values derived from all microsatellite loci were much smaller than those obtained from allozymes. The enzymatic locus Idh-1 exhibited a high value compared to the other loci, suggesting that it is non-neutral. The same Idh-1 locus was implicated in differentiation between temporary and stable populations, as followed also from a highly significant relationship between the allele two of this Idh-1 locus and dispersal power population estimates. The 'parallel evolution' model may account for the diversification of locally adapted Pogonus chalceus populations between different microhabitats.  相似文献

The genetic subdivision of the endemic minnow, Zacco pachycephalus, in 12 rivers of Taiwan was studied by allozyme electrophoresis. Among 26 loci surveyed, six were significantly differentiated among sites. Allozyme genotype frequencies within samples accord with Hardy-Weinberg expectations. A highly divergent genetic structure among sites (FST = 0.497) with low genetic variability within samples (H = 0.006-0.062, mean = 0.03) suggests that local populations originated from a small number of founders. UPGMA and Fitch trees derived from Nei's genetic distance and hierarchical analysis of FST values reveal that samples of Z. pachycephalus can be divided into northern, middle, and southern subgroups and that most genetic variation (87%) is distributed among rather than within groups. It is proposed that there were two isolated refugia during the most recent glacial period, and that populations of the southern group (Group I) originated from a southern refugium in Taiwan below the boundary of the Kaoping River, and other groups (Group II), including northern and middle subgroups, originated from a refugium located on the extinct land-bridge between Taiwan and mainland China. The postglacial division between northern and middle subgroups may be due to hydrological differences between these two regions.  相似文献

Primers for amplification of four novel, unlinked nuclear DNA loci, the first reported for the rough periwinkles of the genus Littorina, are described. Patterns of restriction site polymorphism for these loci are detailed within the rough periwinkles. RFLPs are not found to be diagnostic for any of the currently accepted species within this group, nor for any of the contentious subspecies, or forms, whose taxonomic status is uncertain. However, there are important differences in allele frequencies between these taxa and certain of these mirror differences detected in a previous study of the mitochondrial DNA. These allele frequency data are used to construct a phylogeny in which groupings of the three recognised species are obvious when either Nei's genetic distances or Reynold's distances are clustered. Contentious forms (L. neglecta, L. saxatilis 'b' and L. tenebrosa) do not cluster as distinct taxa, although populations of L. neglecta have important allele frequency differences from L. saxatilis. These four loci have confirmed the consensus view of Littorina phylogeny and provided important information on population structure-however four loci is insufficient for reaching definitive conclusions. Since analysis of nuclear DNA polymorphisms such as these is invaluable for analysis of phylogeny, population structure and phylogeography, identification of additional loci is considered imperative.  相似文献

Different classes of molecular markers occasionally yield discordant views of population structure within a species. Here, we examine the distribution of molecular variance from 14 polymorphic loci comprising four classes of molecular markers within approximately 400 blue marlin individuals (Makaira nigricans). Samples were collected from the Atlantic and Pacific Oceans over 5 years. Data from five hypervariable tetranucleotide microsatellite loci and restriction fragment length polymorphism (RFLP) analysis of whole molecule mitochondrial DNA (mtDNA) were reported and compared with previous analyses of allozyme and single-copy nuclear DNA (scnDNA) loci. Temporal variance in allele frequencies was nonsignificant in nearly all cases. Mitochondrial and microsatellite loci revealed striking phylogeographic partitioning among Atlantic and Pacific Ocean samples. A large cluster of alleles was present almost exclusively in Atlantic individuals at one microsatellite locus and for mtDNA, suggesting that, if gene flow occurs, it is likely to be unidirectional from Pacific to Atlantic oceans. Mitochondrial DNA inter-ocean divergence (FST) was almost four times greater than microsatellite or combined nuclear divergences including allozyme and scnDNA markers. Estimates of Neu varied by five orders of magnitude among marker classes. Using mathematical and computer simulation approaches, we show that substantially different distributions of FST are expected from marker classes that differ in mode of inheritance and rate of mutation, without influence of natural selection or sex-biased dispersal. Furthermore, divergent FST values can be reconciled by quantifying the balance between genetic drift, mutation and migration. These results illustrate the usefulness of a mitochondrial analysis of population history, and relative precision of nuclear estimates of gene flow based on a mean of several loci.  相似文献

Four populations of Anopheles sacharovi Favre occurring in different ecological subregions at altitudes between 353-1,126 m in the Sanliurfa Province of southeast Turkey were compared using morphometric and allozyme analyses. Four allozyme loci were assessed for genetic differentiation among samples from four localities. The similarity phenogram obtained from the allozyme data showed that populations at Birecik and Sandi branched as a separate group from the Pamuklu and Gedik populations. The Gedik population at the highest altitude (1,126 m) was clustered as a separate branch when linear measurements of 63 morphological characteristics were examined. The UPGMA phenogram also showed that Pamuklu and Sandi formed a cluster while Birecik and Gedik formed separate groups.  相似文献

The genetic structure of five natural populations of common wild rice Oryza rufipogon Griff. from China, was investigated with 21 microsatellite loci and compared to estimates of genetic diversity and genetic differentiation detected by 22 allozyme loci. Microsatellite loci, as expected, have much higher levels of genetic diversity (mean values of A = 3.1, P = 73.3%, Ho = 0.358 and He = 0.345) than allozyme loci (mean values of A = 1.2, P = 12.7%, Ho = 0.020 and He = 0.030). Genetic differentiation detected by microsatellite loci ( FST = 0.468, mean I = 0.472) was higher than that for allozyme loci ( FST =0.388, mean I = 0.976). However, microsatellite markers showed less deviation from Hardy-Weinberg expectation (Wright's inbreeding coefficient FIS = -0.069) than do allozymes ( FIS = 0.337). These results suggest that microsatellite markers are powerful high-resolution tools for the accurate assessment of important parameters in population biology and conservation genetics of O. rufipogon, and offer advantages over allozyme markers.  相似文献

We introduce an approach for the analysis of Mendelian polymorphisms in nuclear DNA (nDNA), using restriction fragment patterns from anonymous single-copy regions amplified by the polymerase chain reaction, and apply this method to the elucidation of population structure and gene flow in the endangered green turtle, Chelonia mydas. Seven anonymous clones isolated from a total cell DNA library were sequenced to generate primers for the amplification of nDNA fragments. Nine individuals were screened for restriction site polymorphisms at these seven loci, using 40 endonucleases. Two loci were monomorphic, while the remainder exhibited a total of nine polymorphic restriction sites and three size variants (reflecting 600-base pair (bp) and 20-bp deletions and a 20-bp insertion). A total of 256 turtle specimens from 15 nesting populations worldwide were then scored for these polymorphisms. Genotypic proportions within populations were in accord with Hardy-Weinberg expectations. Strong linkage disequilibrium observed among polymorphic sites within loci enabled multisite haplotype assignments. Estimates of the standardized variance in haplotype frequency among global collections (FST = 0.17), within the Atlantic-Mediterranean (FST = 0.13), and within the Indian-Pacific (FST = 0.13), revealed a moderate degree of population substructure. Although a previous study concluded that nesting populations appear to be highly structured with respect to female (mitochondrial DNA) lineages, estimates of Nm based on nDNA data from this study indicate moderate rates of male-mediated gene flow. A positive relationship between genetic similarity and geographic proximity suggests historical connections and/or contemporary gene flow between particular rookery populations, likely via matings on overlapping feeding grounds, migration corridors or nonnatal rookeries.  相似文献

Phylogeographic studies provide critical insight into the evolutionary histories of model organisms; yet, to date, range-wide data are lacking for the rough periwinkle Littorina saxatilis, a classic example of marine sympatric speciation. Here, we use mitochondrial DNA (mtDNA) sequence data to demonstrate that L. saxatilis is not monophyletic for this marker, but is composed of two distinct mtDNA lineages (I and II) that are shared with sister species Littorina arcana and Littorina compressa. Bayesian coalescent dating and phylogeographic patterns indicate that both L. saxatilis lineages originated in the eastern North Atlantic, around the British Isles, at approximately 0.64 Ma. Both lineages are now distributed broadly across the eastern, central and western North Atlantic, and show strong phylogeographic structure among regions. The Iberian Peninsula is genetically distinct, suggesting prolonged isolation from northeastern North Atlantic populations. Western North Atlantic populations of L. saxatilis lineages I and II predate the last glacial maximum and have been isolated from eastern North Atlantic populations since that time. This identification of two distinct, broadly distributed mtDNA lineages further complicates observed patterns of repeated incipient ecological speciation in L. saxatilis, because the sympatric origins of distinct ecotype pairs on eastern North Atlantic shores may be confounded by admixture of divergent lineages.  相似文献

The genetic diversity and population differentiation of Parashorea chinensis, a rare and endangered dipterocarp, and listed in the Plant Red Book under Grade I, were examined using allozyme analysis of horizontal sliceable starch gel electrophoresis. Considering its narrow distribution, which was limited in the South & Southeast of Yunnan and Southwest of Guangxi, allozyme data for 16 loci of 11 enzymes in 9 natural population (as a comparison, two cultivated populations in Xishuangbanna Tropical Botanical Garden were analyzed) indicated extremely low level of genetic variability in these eleven populations with the proportion of polymorphic loci(P) ranging from 6.25% to 12.5%(mean 6.82%).  相似文献

The null assumption of molecular variation is that most of it is neutral to natural selection. This is in contrast to variation in morphological traits that we generally assume is maintained by selection, and therefore often by selection coupled to environmental heterogeneity in time and space. Examples of molecular variation that vary over habitat-shifts, particularly in allozymes, show that the relative impact of non-neutral variation as compared to neutral variation might be substantial in some systems. To assess the importance of habitat-generated variation in relation to variation generated by random processes in nuclear DNA markers at small spatial scales, we compared the effects of island isolation and habitat heterogeneity on genetic substructuring in a rocky shore snail ( Littorina saxatilis ). This species has a restricted migration among islands owing to the lack of free-floating larvae. Earlier studies show that allozymes vary extensively as a consequence of isolation by water barriers among islands, but also as a consequence of divergent selection among different microhabitats within islands. In the DNA markers we observed genetic differentiation owing to island isolation at three of nine loci. In addition, variation at three loci correlated with habitat type, but the correlation for two of the loci was weak. Overall, isolation contributed slightly more to the genetic variation among populations than did habitat-related factors but the difference was small. It is concluded that both island isolation, which interrupts gene flow, and a heterogeneous habitat cause genetic substructuring at the DNA level in L. saxatilis in the studied area, and thus in this species we need to be somewhat concerned about habitat heterogeneity also at DNA loci.  © 2004 The Linnean Society of London, Biological Journal of the Linnean Society , 2004, 82 , 377–384.  相似文献
