As evidenced by ion-selective electrode potentiometry, the hexameric R-phycoerythrin (RPE) molecule binds 20–4000 cadmium ions (Cd2+) depending on Cd2+ concentration in the solution. Cadmium ions bound to RPE serve as the nuclei of cadmium sulfide crystallization in the presence of sulfide ions. According to spectrometric, electron-microscopic, and capillary electrophoresis data, the particles are heteroaggregates of 3.2 × 6 nm in size. The fact that the particle size fits the size of the central tunnel of the RPE molecule and the similarity between the electrophoretic patterns of free RPE and the RPE-CdS complex indicate that the tunnel space, limiting the crystal growth, is the most probable site of nanoparticle formation. The adaptation of Klebsiella aerogenes to high levels of cadmium was studied in continuous culture under conditions of glucose limitation. When up to 6 × 10−4 M cadmium was added to a culture in steady state, growth ceased instantaneously but resumed within 5 h (dilution rate, 0.1 h−1). When again in steady state, these adapted cells exhibited a far greater tolerance to cadmium than did unadapted cells (not previously exposed to cadmium) when tested on solid media containing different concentrations of cadmium. This relative insensitivity of adapted cells to cadmium was subsequently lost in continuous culture within 5 days after omitting cadmium from the influent medium. Thus, the phenomenon was an inducible physiological process. Adapted cells contained substantial amounts of cadmium (up to 2.4% of the bacterial dry weight). The cadmium content of the cells was dependent on growth conditions and was found to be proportional to the inorganic sulfide content of the cells in all cases. This suggested that formation of CdS is probably the most important mechanism of detoxification in this organism. The presence of large numbers of electron-dense granules on the cell surface (absent in cultures without added cadmium) provided additional support for this conclusion.  相似文献

Summary Vanadium bromoperoxidase is a naturally occurring vanadium-containing enzyme isolated from marine algae. V-BrPO catalyzes the oxidation of halides by hydrogen peroxide which can result in the halogenation of organic substrates. Bromoperoxidase activity is measured by the halogenation of monochlorodimedone (2-chloro-5,5-dimethyl-1,3-dimedone, MCD). In the absence of an organic substrate, V-BrPO catalyzes the halide-assisted disproportionation of hydrogen peroxide yielding dioxygen. The dioxygen formed is in the singlet excited state (1O2). V-BrPO is quite stable to thermal denaturation and denaturation by certain organic solvents which makes V-BrPO an excellent candidate for industrial applications. The stability of V-BrPO in the presence of strong oxidants and in the presence of phosphate is reported. Incubation of V-BrPO in phosphate buffer (1–100 mM at pH 6; 2–10 mM at pH 5) inactivates the enzyme. The inactivity can be fully restored by the addition of vanadate if excess phosphate is removed. The inactivation of V-BrPO by phosphate can be prevented by the presence of H2O2 (4–40 mM). We are currently investigating the mechanism of V-BrPO inactivation by phosphate. V-BrPO was not inactivated by HOCl (1 mM) nor H2O2. In addition V-BrPO was not inactivated under turnover conditions of 1 mM H2O2 with 0.1–1 M Cl at pH 5 nor 2 mM H2O2 with 0.1 M Br.  相似文献

Nitrogen fixation (C2H2 reduction) in a sediment-water system was studied under anaerobic incubation conditions. Sodium sulfide at low concentrations stimulated activity, with a twofold increase in C2H4 production occurring in the presence of 8 μmol of S2− per ml of stream water. Sodium sulfide at concentrations of 16 μmol of S2− per ml or greater inhibited nitrogen fixation, with 64 μmol of S2− per ml being completely inhibitory. Sulfide at levels of 16 μmol/ml or above inhibited CO2 production, and the degree of inhibition increased with increasing concentration of sulfide. Titanium (III) citrate (used to modify Eh levels) stimulated both nitrogen fixation and CO2 production, but could not duplicate, at any concentration tested, the twofold increase in nitrogen fixation caused by 8 μmol of S2− per ml. Sulfide additions caused pH changes in the sediment, and when the sediment was adjusted and maintained at pH 7.0 all concentrations of sulfide inhibited nitrogen fixation activity. From considerations of the redox equilibria of H2, H2S, and other sulfur species at various pH values, it appeared that H2S was the toxic entity and that HS was less toxic. The observed stimulation of activity was apparently due to a pH change coupled with the concurrent production of HS from H2S.  相似文献

Hydroponically grown tomato plants (Lycopersicon esculentum P. Mill. cv Golden Boy) exposed to 100 micromolar cadmium sulfate produced metal-(γEC)nG peptide complexes containing acid-labile sulfur. The properties of the complexes resemble those of the cadmium-(γEC)nG peptide complexes from Schizosaccharomyces pombe and Candida glabrata known to contain a cadmium sulfide crystallite core. The crystallite is stabilized by a sheath of peptides of general structure (γGlu-Cys)n-Gly. The cadmium-peptide complexes of tomato contained predominantly peptides of n3, n4, and n5. spectroscopic analyses indicated that the tomato cadmium-sulfide-peptide complex contained CdS crystallite core particles smaller than 2.0 nanometers in diameter.  相似文献

Proton translocation by washed cells of the sulfate-reducing bacterium Desulfovibrio desulfuricans strain Essex 6 was studied by means of pH and sulfide electrodes. Reversible extrusion of protons could be induced either by addition of electron acceptors to cells incubated under hydrogen, or by addition of hydrogen to cells incubated in the presence of an appropriate electron acceptor. Proton translocation was increased in the presence of ionophores that dissipate the membrane potential (thiocyanate, methyl triphenylphosphonium cation, but not valinomycin) and was sensitive to the uncoupler carbonylcyanide m-chlorophenylhydrazone (CCCP). Upon micromolar additions of H2, usually sulfide was formed in stoichiometric amounts, and extrapolated H+/H2 ratios were 1.8±0.5 with sulfate, 2.3±0.3 with sulfite and 0.5±0.1 with thiosulfate. In several experiments hydrogen pulses caused increased proton extrusion not associated with sulfide production. This was a hint that sulfite might be reduced via intermediates. In the absence of H2S formation, extrapolated H+/H2 ratios were 3.1±0.8 with sulfate, 3.4±1.1 with sulfite, 4.4±0.8 with thiosulfate and 6.3±1.2 with oxygen. Micromolar pulses of electron acceptors to cells incubated under H2 caused less proton translocation than H2 pulses in presence of excess of electron acceptor; extrapolated H+/H2 ratios were 1.3±0.4 with sulfite, 3.3±0.9 with nitrite and 4.2±0.5 with oxygen. No proton translocation was observed after micromolar pulses of sulfate, thiosulfate or nitrate to cells incubated under hydrogen in the presence of thiocyanate. Inhibition experiments with CO and CuCl2 revealed that the hydrogenase activity was localized in the intracellular space, and that no periplasmic hydrogenase was present. A lithotrophic freshwater Beggiatoa strain was enriched in O2-H2S gradient tubes to investigate its ability to oxidize sulfide with NO3 as an alternative electron acceptor. The gradient tubes contained different NO3 concentrations, and the chemotactic response of the Beggiatoa mats was observed. The effects of the Beggiatoa sp. on vertical gradients of O2, H2S, pH, and NO3 were determined with microsensors. The more NO3 that was added to the agar, the deeper the Beggiatoa filaments glided into anoxic agar layers, suggesting that the Beggiatoa sp. used NO3 to oxidize sulfide at depths below the depth that O2 penetrated. In the presence of NO3 Beggiatoa formed thick mats (>8 mm), compared to the thin mats (ca. 0.4 mm) that were formed when no NO3 was added. These thick mats spatially separated O2 and sulfide but not NO3 and sulfide, and therefore NO3 must have served as the electron acceptor for sulfide oxidation. This interpretation is consistent with a fourfold-lower O2 flux and a twofold-higher sulfide flux into the NO3-exposed mats compared to the fluxes for controls without NO3. Additionally, a pronounced pH maximum was observed within the Beggiatoa mat; such a pH maximum is known to occur when sulfide is oxidized to S0 with NO3 as the electron acceptor.  相似文献

An advanced electrochemical sensor for the detection of enrofloxacin (ENR) based on the use of a modified electrode containing cadmium sulfide (CdS) nanoparticles (NPs) is reported. The CdS NPs were synthesized and characterized and then coated onto the electrode to fabricate a modified electrode that exhibited a lower limit of detection of 9.5 × 10?8 mol·L?1. This detection limit compares with a traditional electrode that exhibited a concentration detection range of 1.0 × 10?2 to 1.0 × 10?7 mol·L?1. This modified electrode demonstrated good selectivity, reproducibility, response time (<40 s), lifetime (up to 12 wk), and pH range (3.3‐7.2) for the determination of ENR in real samples (eg, pig urine).  相似文献

Pure cadmium oxalate trihydrate (COT) and barium added cadmium oxalate (BCO) single crystals were grown by controlled diffusion of Cd2+ and Ba2+ ions in silica gel at ambient temperature. A single test tube technique coupled with gel aging conferred maximum size crystals by controlling the nucleation rate. It was found that the pH and age of the gel greatly influenced the crystal quality, their size and transparency. Grown crystals CdC2O4 · 3H2O and Ba0.5Cd0.5(C2O4)2 · 5H2O were characterized by X-ray diffraction, Fourier transform infrared spectroscopy and thermal analysis. Effect of barium dopant on the growth and morphology of cadmium oxalate was studied. Pure cadmium oxalate crystallized in triclinic system and the barium-doped cadmium oxalate crystallized in hexagonal system with massive changes in their unit cell parameters. The infrared spectrum revealed the presence of oxalate ligands and water of hydration in both the pure and barium-doped crystals. Thermal analysis showed that the grown crystals were dehydrated thermally even from lower temperatures and the doped crystals were found more stable.  相似文献

Heavy metal resistance by bacteria is a topic of much importance to the bioremediation of contaminated soils and sediments. We report here the isolation of a highly cadmium-resistant Klebsiella planticola strain, Cd-1, from reducing salt marsh sediments. The strain grows in up to 15 mM CdCl2 under a wide range of NaCl concentrations and at acidic or neutral pH. In growth medium amended with thiosulfate, it precipitated significant amounts of cadmium sulfide (CdS), as confirmed by x-absorption spectroscopy. In comparison with various other strains tested, Cd-1 is superior for precipitating CdS in cultures containing thiosulfate. Thus, our results suggest that Cd-1 is a good candidate for the accelerated bioremediation of systems contaminated by high levels of cadmium.  相似文献

Biological CO2 sequestration through acetogenesis with H2 as electron donor is a promising technology to reduce greenhouse gas emissions. Today, a major issue is the presence of impurities such as hydrogen sulfide (H2S) in CO2 containing gases, as they are known to inhibit acetogenesis in CO2-based fermentations. However, exact values of toxicity and inhibition are not well-defined. To tackle this uncertainty, a series of toxicity experiments were conducted, with a mixed homoacetogenic culture, total dissolved sulfide concentrations ([TDS]) varied between 0 and 5 mM and pH between 5 and 7. The extent of inhibition was evaluated based on acetate production rates and microbial growth. Maximum acetate production rates of 0.12, 0.09 and 0.04 mM h-1 were achieved in the controls without sulfide at pH 7, pH 6 and pH 5. The half-maximal inhibitory concentration (IC50qAc) was 0.86, 1.16 and 1.36 mM [TDS] for pH 7, pH 6 and pH 5. At [TDS] above 3.33 mM, acetate production and microbial growth were completely inhibited at all pHs. 16S rRNA gene amplicon sequencing revealed major community composition transitions that could be attributed to both pH and [TDS]. Based on the observed toxicity levels, treatment approaches for incoming industrial CO2 streams can be determined.  相似文献

We report microbially facilitated synthesis of cadmium sulfide (CdS) nanostructured particles (NP) using anaerobic, metal-reducing Thermoanaerobacter sp. The extracellular CdS crystallites were <10 nm in size with yields of ~3 g/L of growth medium/month with demonstrated reproducibility and scalability up to 24 L. During synthesis, Thermoanaerobacter cultures reduced thiosulfate and sulfite salts to H2S, which reacted with Cd2+ cations to produce thermodynamically favored NP in a single step at 65 °C with catalytic nucleation on the cell surfaces. Photoluminescence (PL) analysis of dry CdS NP revealed an exciton-dominated PL peak at 440 nm, having a narrow full width at half maximum of 10 nm. A PL spectrum of CdS NP produced by dissimilatory sulfur reducing bacteria was dominated by features associated with radiative exciton relaxation at the surface. High reproducibility of CdS NP PL features important for scale-up conditions was confirmed from test tubes to 24 L batches at a small fraction of the manufacturing cost associated with conventional inorganic NP production processes.  相似文献

Consistent with data in animal systems, experimental evidence highlights sulfide as a signaling molecule of equal importance to NO and H2O2 in plant systems. In mammals, two cytosolic enzymes, cystathionine β-synthase (CBS) and cystathionine γ-lyase (CSE), have been shown to be responsible for the endogenous production of sulfide. L-cysteine desulfhydrase 1 (DES1) has been recently established as the only enzyme that is involved in the generation of hydrogen sulfide in plant cytosol. Although plants have an available source of sulfide within chloroplasts, the basic stromal pH prevents sulfide release into the cytosol. Therefore, DES1 is essential for the production of sulfide for signaling purposes.  相似文献

Chemoautotrophic symbioses, in which endosymbiotic bacteria are the major source of organic carbon for the host, are found in marine habitats where sulfide and oxygen coexist. The purpose of this study was to determine the influence of pH, alternate sulfur sources, and electron acceptors on carbon fixation and to investigate which form(s) of inorganic carbon is taken up and fixed by the gamma-proteobacterial endosymbionts of the protobranch bivalve Solemya velum. Symbiont-enriched suspensions were generated by homogenization of S. velum gills, followed by velocity centrifugation to pellet the symbiont cells. Carbon fixation was measured by incubating the cells with 14C-labeled dissolved inorganic carbon. When oxygen was present, both sulfide and thiosulfate stimulated carbon fixation; however, elevated levels of either sulfide (>0.5 mM) or oxygen (1 mM) were inhibitory. In the absence of oxygen, nitrate did not enhance carbon fixation rates when sulfide was present. Symbionts fixed carbon most rapidly between pH 7.5 and 8.5. Under optimal pH, sulfide, and oxygen conditions, symbiont carbon fixation rates correlated with the concentrations of extracellular CO2 and not with HCO3 concentrations. The half-saturation constant for carbon fixation with respect to extracellular dissolved CO2 was 28 ± 3 μM, and the average maximal velocity was 50.8 ± 7.1 μmol min−1 g of protein−1. The reliance of S. velum symbionts on extracellular CO2 is consistent with their intracellular lifestyle, since HCO3 utilization would require protein-mediated transport across the bacteriocyte membrane, perisymbiont vacuole membrane, and symbiont outer and inner membranes. The use of CO2 may be a general trait shared with many symbioses with an intracellular chemoautotrophic partner.  相似文献

Production of hydrogen and organic compounds by an electrosynthetic microbiome using electrodes and carbon dioxide as sole electron donor and carbon source, respectively, was examined after exposure to acidic pH (∼5). Hydrogen production by biocathodes poised at −600 mV vs. SHE increased>100-fold and acetate production ceased at acidic pH, but ∼5–15 mM (catholyte volume)/day acetate and>1,000 mM/day hydrogen were attained at pH ∼6.5 following repeated exposure to acidic pH. Cyclic voltammetry revealed a 250 mV decrease in hydrogen overpotential and a maximum current density of 12.2 mA/cm2 at −765 mV (0.065 mA/cm2 sterile control at −800 mV) by the Acetobacterium-dominated community. Supplying −800 mV to the microbiome after repeated exposure to acidic pH resulted in up to 2.6 kg/m3/day hydrogen (≈2.6 gallons gasoline equivalent), 0.7 kg/m3/day formate, and 3.1 kg/m3/day acetate ( = 4.7 kg CO2 captured).  相似文献

In this study, CdS quantum dots (QDs) capped with mercaptosuccinic acid (MSA) were prepared in one step. The size, shape, component and spectral properties of MSA‐capped CdS QDs were characterized by transmission electron microscopy (TEM), photoluminescence (PL) and infrared (IR) spectrometry. The results showed that the prepared QDs with an average diameter of 6 nm have favorable fluorescence, which is greatly influenced by the pH of the environment. The interaction of some heavy metal ions including Ag+, Hg2+, Cu2+, Ni2+ and Co2+ with MSA‐capped CdS QDs was investigated in different buffering pH media. Based on the fluorescence quenching of the QDs in the presence of each of the metal ions, the feasibility of their determinations was examined according to the Stern–Volmer equation. The investigations showed that Hg(II) ions can be determined in the presence of many co‐existing metal ions at a buffering pH of 5. Desulfotomaculum carboxydivorans, recently isolated from a full-scale anaerobic wastewater treatment facility, is a sulfate reducer capable of hydrogenogenic growth on carbon monoxide (CO). In the presence of sulfate, the hydrogen formed is used for sulfate reduction. The organism grows rapidly at 200 kPa CO, pH 7.0, and 55°C, with a generation time of 100 min, producing nearly equimolar amounts of H2 and CO2 from CO and H2O. The high specific CO conversion rates, exceeding 0.8 mol CO (g protein)−1 h−1, makes this bacterium an interesting candidate for a biological alternative of the currently employed chemical catalytic water–gas shift reaction to purify synthesis gas (contains mainly H2, CO, and CO2). Furthermore, as D. carboxydivorans is capable of hydrogenotrophic sulfate reduction at partial CO pressures exceeding 100 kPa, it is also a good candidate for biodesulfurization processes using synthesis gas as electron donor at elevated temperatures, e.g., in biological flue gas desulfurization. Although high maximal specific sulfate reduction rates (32 mmol (g protein)−1 h−1) can be obtained, its sulfide tolerance is rather low and pH dependent, i.e., maximally 9 and 5 mM sulfide at pH 7.2 and pH 6.5, respectively.  相似文献

Summary The continuously operated suspended growth anaerobic contact system was utilized to estimate the effect of sulfate reduction on the thermophilic (55°C) methane fermentation process. Results indicated that reduction in methanogenesis in the presence of sulfate was due to two separate, but related, processes;i.e. competitive and sulfide inhibition. Although prevention of competitive inhibition would be difficult under normal fermenter operation, sulfide inhibition could be minimized by environmental selection of sulfide tolerant microbial populations through biomass recycle and pH control. Stable fermenter operation was achieved at soluble sulfide concentrations as high as 330 mg/l soluble sulfide. Using batch fermenters, a maximum thermophilic sulfate reduction rate of 3.7 mg SO4 2––S/g volatile solids (VS)-day was estimated. In the Mammoth Springs (Yellowstone National Park) waters with near neutral pH and soluble sulfide (H2S, HS, S2–) of over 1–2 mg/liter (30–60M) are characterized by substrate covers of phototrophic bacteria (Chloroflexus and aChlorobium-like unicell) above 50C and by a blue-green alga (Spirulina labyrinthiformis) below this temperature.Synechococcus. Mastigocladus, and other blue-green algae typical of most hot springs of western North America are excluded, apparently by sulfide. The sulfide-adaptedSpirulina photosynthesized at maximum rates at 45C and at approximately 300 to 700Ein/m2/sec of visible radiation. Sulfide (0.6–1.2 mM) severely poisoned photosynthesis of nonadapted populations, but those continuously exposed to over 30M tolerated at least 1 mM without inhibition. A normal14C-HCO3 photoincorporation rate was sustained with 0.6–1 mM sulfide in the presence of DCMU (7M) or NH2OH (0.2 mM), although both of these photosystem II inhibitors prevented photoincorporation without sulfide. Other sulfur-containing compounds (S2O3 2– SO3 2–, S2O4 2– thioglycolic acid cysteine) were unable to relieve DCMU inhibition. The lowering of the photoincorporation rate by preferentially irradiating photosystem I was also relieved by sulfide. The most tenable explanation of these results is that sulfide is used as a photo-reductant of CO2, at least when photosystem II is inhibited. It is suggested that in some blue-green algae photosystem II is poisoned by a low sulfide concentration, thus making these algae sulfidedependent if they are to continue photosynthesizing in a sulfide environment. Presumably a sulfidecytochrome reductase enzyme system must be synthesized for sulfide to be used as a photo-reductant.  相似文献

The isotope exchange between35S-labeled sulfur compounds of sulfate (SO4 2–), elemental sulfur (S0), polysulfide (Sn 2–), hydrogen sulfide (HS: H2S + HS + S2–), iron sulfide (FeS), and pyrite (FeS2) was studied at pH 7.6 and 20 °C in anoxic, sterile seawater. Isotope exchange was observed between S0, S2 2– HS, and FeS, but not between35S labeled SO4 2– or FeS2 and the other sulfur compounds. Polysulfide mediated the isotope exchange between S0 and bisulfide (HS). The isotope exchange between S0 and Sn 2–) reached 50% of equilibrium within < 2 min while exchange between S2 2– and HS approached equilibrium within 0.5-1 h. In all the experiments HS, revealed a fraction exchange from 0.79 to 1.00. Isotope exchange between S2– and FeS took place only via S2 2– and/or HS. The isotope exchange between iron sulfide and the other sulfur compounds was not complete within 24 h as shown by a fraction exchange of 0.07–0.83. This lack of equilibrium (fraction exchange < 1) was due to the isotope exchange between dissolved compounds and surfaces of sulfur particles. The isotopic exchange reactions limit the usefulness of radiotracers in process studies of the inorganic sulfur species. Exchange reactions will also affect the stable isotope distribution among the sulfur species. The kinetics of the isotopic exchange reactions, however, depend on both pH and temperature.  相似文献
