A new tropone,3,8-dimethyl-5-isopropyl-2,3- dihydro(1H)azulen-6-one, named orobanone, has been isolated for the first time from a plant (Orobanche rapum-genistae, Orobanchaceae). This tropone is synthesized by the parasite and exists, with other related compounds, in various species of broom-rapes.  相似文献

Three new sesquiterpenes were isolated from Severinia buxifolia, and identified as α-santalen-11-one, dihydro-α-santalen-12-one, and 12,13-epoxy-α-santalene, respectively. α-Photosantalol A, Δ13,14-iso-α-santalol, α-santalene and (E)-5-(2,3-dimethyl-3-nortricyclyl)pent-3-en-2-one were also isolated and characterized.  相似文献

Chemical studies on the constituents of Eranthis cilicica led to isolation of ten chromone derivatives, two of which were previously known. Comprehensive spectroscopic analysis, including extensive 1D and 2D NMR data, and the results of enzymatic hydrolysis allowed the chemical structures of the compounds to be assigned as 8,11-dihydro-5-hydroxy-2,9-dihydroxymethyl-4H-pyrano[2,3-g][1]benzoxepin-4-one, 5,7-dihydroxy-8-[(2E)-4-hydroxy-3-methylbut-2-enyl]-2-methyl-4H-1-benzopyran-4-one, 5,7-dihydroxy-2-hydroxymethyl-8-[(2E)-4-hydroxy-3-methylbut-2-enyl]-4H-1-benzopyran-4-one, 7-[(β-d-glucopyranosyl)oxy]-5-hydroxy-8-[(2E)-4-hydroxy-3-methylbut-2-enyl]-2-methyl-4H-1-benzopyran-4-one, 7-[(β-d-glucopyranosyl)oxy]-5-hydroxy-2-hydroxymethyl-8-[(2E)-4-hydroxy-3-methylbut-2-enyl]-4H-1-benzopyran-4-one, 9-[(O-β-d-glucopyranosyl-(1→6)-β-d-glucopyranosyl)oxy]methyl-8,11-dihydro-5,9-dihydroxy-2-methyl-4H-pyrano[2,3-g][1]benzoxepin-4-one, 8,11-dihydro-5,9-dihydroxy-9-hydroxymethyl-2-methyl-4H-pyrano[2,3-g][1]benzoxepin-4-one, and 7-[(O-β-d-glucopyranosyl-(1→6)-β-d-glucopyranosyl)oxy]methyl-4-hydroxy-5H-furo[3,2-g][1]benzopyran-5-one, respectively. From Acnistus breviflorus the new 27-hydroxy-5β,6β-epoxy-1-oxo-(22R)-witha-24-enolide (2,3-dihydrojaborosalactone A) as well as seven known withanolides, withaferin A, 2,3-dihydrowithaferin A, 6α-chloro-5β-hydroxywithaferin A, 5,6-deoxywithaferin A, jaborosalactone A, jaborosalactone D and jaborosalactone E were isolated and characterized by means of spectroscopic (1H NMR, 13 C NMR and mass spectral) methods. Depending on the extraction solvent (methanol or ethanol), a known artifact (3β-methoxy-2,3-dihydrowithaferin A) and the new 5α-methoxy-4,5-dihydrojaborosalactone B and 5α-ethoxy-4,5-dihydrojaborosalactone B were also isolated and characterized.  相似文献

A new class of pyrrolo[2,3-d]pyrimidin-4-one corticotropin-releasing factor 1 (CRF1) receptor antagonists has been designed and synthesized. In general, reported CRF1 receptor antagonists possess a sp2-nitrogen atom as hydrogen bonding acceptor (HBA) on their core scaffolds. We proposed to use a carbonyl group of pyrrolo[2,3-d]pyrimidin-4-one derivatives as a replacement for the sp2-nitrogen atom as HBA in classical CRF1 receptor antagonists. As a result, several pyrrolo[2,3-d]pyrimidin-4-one derivatives showed CRF1 receptor binding affinity with IC50 values in the submicromolar range. Ex vivo 125I-sauvagine binding studies showed that 2-(dipropylamino)-3,7-dimethyl-5-(2,4,6-trimethylphenyl)-3,7-dihydro-4H-pyrrolo[2,3-d]pyrimidin-4-one (16b) (30 mg/kg, po) was able to penetrate into the brain and inhibit radioligand binding to CRF1 receptors (frontal cortex, olfactory bulb, and pituitary) in mice. We identified pyrrolo[2,3-d]pyrimidin-4-one derivatives as the first CRF1 antagonists with a carbonyl-based HBA.  相似文献

Two novel lactones have been isolated from the stem barks of Garcinia conrauana and G. mannii. The major component of the bark of G. conrauana was identified as 3-(3,? 3″-dimethylallyl)-conrauanalactone [4-hydroxy-3-(3″, 3″-dimethylallyl)-6-pentadecylpyran- 2-one] by comparison of spectral data of the isolated compound and two methylethers with that obtained for the previously isolated conrauanalactone. A minor component of the bark of G. mannii was tentatively identified as 3-α-hydroxy-5-(heptadec-8′-enyl)-tetrahydro- furan-2-one on the basis of spectral data from the isolated compound and its monoacetate. The distributions of biflavonoids and related compounds and benzophenones in the stem bark, heartwood, seeds and leaves of the two species are reported.  相似文献

The synthesis and pharmacological evaluation of new 3-(imidazol-4(5)-ylmethylene)-2,3-dihydrobenzo[b]furan-2-ones 8-10 and 3-(3,5-dimethylpyrrol-2-ylmethylene)-2,3-dihydrobenzo[b]furan-2-one 11, analogues of SU-5416, as potential inhibitors of angiogenesis, are reported. Compounds 8 and 11 were prepared by a Knoevenagel reaction starting from 2-hydroxyphenylacetic acid 2 and 4-formylimidazole 5 or 2-formyl-3,5-dimethylpyrrole 7, followed by acid-catalysed cyclodehydration. For compounds 9 and 10, an alternative method was used; it consisted in carrying out the Knoevenagel reaction with the 2,3-dihydrobenzo[b]furan-2-ones 3 and 4. The antiangiogenic activity of these compounds was evaluated in the three-dimensional in vitro rat aortic rings test at 1 μM. At this concentration, compound 11 induced a decrease of angiogenesis comparable to that observed with SU-5416; the vascular density index at 1 μM of 11 and SU-5416 were 30±10 and 22±4% of control, respectively.  相似文献

The isolation of two polysubstituted arylbenzofurans from the heartwood of Dalbergia parviflora is described. Their structures were elucidated mainly by spectroscopic techniques (UV, IR, 1H and 13C NMR). They were named parvifuran (5-hydroxy-6-methoxy-3-methyl-2-phenylbenzofuran) and isoparvifuran (5-hydroxy-6-methoxy-2-methyl-3-phenylbenzofuran) and are the first compounds of this class isolated from a Dalbergia species.  相似文献

Reaction of hydrazonoyl halides with 6-(benzylidenamino)-2-thioxo-2,3-dihydro-1H-pyrimidin-4-one and 2,3-diaminoquinazolin-4-one site-selectively afforded 3-substituted-7-(benzylidenamino)-1-phenyl-[1,2,4]triazolo[4,3-a]-pyrimidin-5(1H)-ones, [1,2,4,5]tetrazino[6,1-b]quinazolin-6(4H)-one, and 3-methyl-2-(4-substituted-phenylhydrazo)-[1,2,4]triazino[3,2-b]quinazolin-10-ones in good yields. The structures of the newly synthesized compounds were elucidated by chemical evidence and their IR, 1H, 13C NMR, and MS spectra. Furthermore, some of the products were screened against different strains of bacteria and fungi.  相似文献

Using well-established techniques, samples were obtained of the volatile essential oils of the two types of curry leaf, Murraya koenigii and Pandanus latifolius. Both contained mainly terpenes, and M. koenigii produced less than 4% of other components with eight monoterpene hydrocarbons (ca 16%) and 17 sesquiterpene hydrocarbons (ca 80%) being obtained. The most important constituents of M. koenigii are β-caryophyllene, β-gurjunene, β-elemene, β-phellandrene and β-thujene. The volatile essential oil of P. latifolius also contained mainly sesquiterpene hydrocarbons (6–42%) but the only monoterpene was linalool (ca 6%). Nearly 2000 times the total quantity of aroma volatiles was produced by M. koenigii compared with P. latifolius, and this partly explains the observed stronger flavour potency of the former.  相似文献

In this study, some 4-(1,5-diarylimidazol-2-yl)thioacetyl-1-phenyl-2,3-dimethyl-3-pyrazoline-5-one derivatives were prepared by reacting 4-(2-chloroacetyl)-1-phenyl-2,3-dimethyl-3-pyrazoline-5-one and 2-mercapto-1, 5-diarylimidazole derivatives. The antinociceptive and anticancer activities of the compounds obtained were investigated. It was observed that some of the compounds, 2a, 2d, 2g, and 2j, showed remarkable antinociceptive activity, and one of the compounds, 2i, showed weak anticancer activity.  相似文献

2-Deoxy-2,3-dehydro-N-acetylneuraminic acid and its methyl ester are competitive inhibitors of Arthrobacter sialophilus neuraminidase with Ki = 1.4 × 10?6M and 4.8 × 10?5M, respectively. The Km for the substrate, N-acetylneuraminlactose, is 1.0 × 10?3M. These data, taken together with the conformation of these compounds, indicate that these compounds are transition-state analogs of the enzyme. These results also suggest that the substrate upon binding to neuraminidase is distorted to a conformation approaching that of a half-chair.  相似文献

Samples of the aroma volatiles of globe artichoke and Jerusalem artichoke were obtained by well established methods and were analysed by routine GC and GC/MS. Eight sesquiterpene hydrocarbons afforded the major group of components (over 42%) in globe artichoke samples, with β-selinene (ca 32%) as the main constituent. Previously reported caryophyllene could not be detected. α-Cedrene was found to have globe artichoke aroma characteristics on odour evaluation of separated components at an odour port at the exit of the GC column. Jerusalem artichoke samples contained one major component (β-bisabolene, ca 51%) and a range of saturated long-chain hydrocarbons (ca 22%). The sesquiterpene presumably contributes appreciably to the characteristic flavour of Jerusalem artichoke. Both types of artichoke gave a low concentration of total volatiles.  相似文献

A new series of pyrazolo[3,4-d]pyrimidine-6-one derivatives (2a–2j) were prepared by using the Biginelli multicomponent cyclocondensation of 3-methyl-1-phenyl-1H-pyrazol-5(4H)-one (1a), different aromatic aldehydes, and urea with a catalytic amount of HCl at reflux temperature. These compounds were characterized by IR, 1H NMR, 13C NMR, and Mass spectral data. In vitro antiamoebic activity was performed against HM1:IMSS strain of Entamoeba histolytica. The results showed that the compounds 2b, 2i, and 2j with IC50 values of 0.37 µM, 0.04 µM, and 0.06 µM, respectively, exhibited better antiamoebic activity than the standard drug metronidazole (IC50?=?1.33 µM). The toxicological studies of these compounds on human breast cancer MCF-7 cell line showed that the compounds 2b, 2i, and 2j exhibited >80% viability at the concentration range of 1.56–50 µM.  相似文献
