The rate of introduction of exotic marine species has dramatically increased during the 19th and 20th centuries. Exemplifying this trend, the marine gastropod Ocinebrellus inornatus was first detected outside its native range in 1924 on the American Pacific coast, then in 1995 on the French Atlantic coast. To determine the origin of the French populations of this invasive species, we compared a French population with populations collected in Asia—the native range—and with a population collected in the United States. Analyses of mitochondrial DNA and allozyme polymorphism revealed that the French and American populations were closely related and substantially differentiated from the Asian populations. According to our results, the most likely scenario is that the source population of the French Atlantic coast populations was located in the United States. Indeed, taken altogether, the genetic structure of Asian populations, the time lag separating the introduction on the American Pacific coast from the introduction on the French Atlantic coast and the high level of genetic diversity in the two introduced areas (indicating an absence of major founder events) are hardly compatible with a scenario in which French population resulting only from primary introduction events from the native area. Finally, although similar, the French and American populations were not identical. Thus, even if the main source population of the French populations was located in the United States, the genetic structure of French populations may have been modified by cryptic and recurrent introduction events directly from Asia.  相似文献

Chaetognaths are one of the most numerous organisms in the zooplankton community off the coast of North Carolina. During two years of offshore sampling in the late winter to early spring, sixteen chaetognath species were identified, four of which had not previously been reported in the waters of the United States South Atlantic Bight. Offshore samples were dominated by Sagitta enflata Grassi, 1881, one of the larger species, which contributed > 61% of total chaetognath abundance while dominant coastal species were S. tenuis Conant, 1896 and S. hispida Conant, 1895. Abundances, body sizes and spatial distributions were determined for the most abundant chaetognath species along with the overall abundance of three common co-occurring larval fish species (spot, Atlantic croaker and Atlantic menhaden). In addition, laboratory feeding experiments were conducted using S. tenuis and S. hispida to estimate the potential impact of chaetognath predation on representative North Carolina larval fish which spawn offshore and subsequently migrate into local estuaries. Feeding rates (no. prey items day− 1) varied with prey type and the condition (starved/fed) of the chaetognath. Weight specific daily rations (SDR) were found to vary inversely with chaetognath size, decreasing exponentially with an increase in chaetognath length. The observed abundance and distribution data indicate that wintertime chaetognath populations in offshore waters of Onslow Bay, North Carolina have the potential to interact with recently spawned larval fish and may simultaneously act as competitors, predators, or prey. Furthermore, calculations using published values of chaetognath abundance, zooplankton standing crops, and our SDR estimates indicate that chaetognaths in a representative North Carolina estuary would require a minimum of 5.96 cal m− 3 day− 1 to sustain their biomass. Allowing for the reported spatial and temporal variability in zooplankton abundance in these systems, chaetognaths should consume 4.4%-20.9% of the estimated total zooplankton production day− 1. This further emphasizes the role of chaetognaths not only as predators, but also as competitors with larval fish for zooplankton food stocks in southeastern United Sates estuaries.  相似文献

We investigated the host specificity of two cryptic microsporidian species (Anostracospora rigaudi and Enterocytospora artemiae) infecting invasive (Artemia franciscana) and native (Artemia parthenogenetica) hosts in sympatry. Anostracospora rigaudi was on average four times more prevalent in the native host, whereas E. artemiae was three times more prevalent in the invasive host. Infection with An. rigaudi strongly reduced female reproduction in both host species, whereas infection with E. artemiae had weaker effects on female reproduction. We contrasted microsporidian prevalence in native A. franciscana populations (New World) and in both invaded and non-invaded Artemia populations (Old World). At a community level, microsporidian prevalence was twice as high in native compared with invasive hosts, due to the contrasting host-specificity of An. rigaudi and E. artemiae. At a higher biogeographical level, microsporidian prevalence in A. franciscana did not differ between the invaded populations and the native populations used for the introduction. Although E. artemiae was the only species found both in New and Old World populations, no evidence of its co-introduction with the invasive host was found in our experimental and phylogeographic tests. These results suggest that the success of A. franciscana invasion is probably due to a lower susceptibility to virulent microsporidian parasites rather than to decreased microsporidian prevalence compared with A. parthenogenetica or to lower microsporidian virulence in introduced areas.  相似文献

Pathogens have been implicated as potential factors in the recent decline of some North American bumble bee (Bombus) species, but little information has been reported about the natural enemy complex of bumble bees in the United States. We targeted bumble bee populations in a state-wide survey in Illinois and several sites in California and Oregon where declines have been reported to determine presence and prevalence of natural enemies. Based on our observations, most parasites and pathogens appear to be widespread generalists among bumble bee species, but susceptibility to some natural enemies appeared to vary.  相似文献

Little is known about the variation in demographics, flower number, fruit set, and summer dormancy in Allium species native to the Intermountain West region of the United States. The purpose of this study was to investigate demographic patterns, summer dormancy traits, and variation in reproductive characteristics among habitats. Three populations of each of three species, Allium acuminatum, A. brandegei, and A. passeyi, were studied along an altitudinal gradient in northern Utah, USA, from 2004 to 2006. Demographic data were collected from individuals in permanent plots established in each population. Individual flowering plants were periodically monitored and used to calculate reproductive characteristics. Leaf area measurements throughout the 2005 growing season and soil temperature sensors were used to assess the relationship between habitat and dormancy characteristics. Populations of the widely distributed species, A. acuminatum and A. brandegei, were dominated by individuals in seedling and flowering stages while the restricted A. passeyi had populations containing a majority of individuals in the post-seedling, but non-flowering stage-class. There was a strong directional trend in A. acuminatum reproductive traits, with flower number and fruit set decreasing with increasing elevation. The mid-elevation A. brandegei populations were reproductively more successful than the high and low-elevation populations. The high-elevation A. passeyi population had more flowers per plant and higher fruit set in comparison to the lower elevation populations. The onset of summer dormancy as indicated by the timing of leaf senescence, was associated with high soil temperatures specific to each species. In general, the timing of leaf senescence was later in higher elevation populations in all species. A smaller proportion of non-flowering individuals in A. acuminatum and A. brandegei may indicate a high seedling mortality rate or a shorter juvenile phase in comparison to A. passeyi. The A. passeyi populations exhibit demographic characteristics of rare taxa; low seed production and low seedling recruitment. The high number of non-flowering plants in A. passeyi likely includes many reproductively mature individuals which are influenced by the unique habitat and environment of this species. Temperature is likely the key environmental cue inducing summer dormancy in all three species.  相似文献

Saprolegniosis, the disease caused by Saprolegnia sp., results in considerable economic losses in aquaculture. Current control methods are inadequate, as they are either largely ineffective or present environmental and fish health concerns. Vaccination of fish presents an attractive alternative to these control methods. Therefore we set out to identify suitable antigens that could help generate a fish vaccine against Saprolegnia parasitica. Unexpectedly, antibodies against S. parasitica were found in serum from healthy rainbow trout, Oncorhynchus mykiss. The antibodies detected a single band in secreted proteins that were run on a one-dimensional SDS-polyacrylamide gel, which corresponded to two protein spots on a two-dimensional gel. The proteins were analysed by liquid chromatography tandem mass spectrometry. Mascot and bioinformatic analysis resulted in the identification of a single secreted protein, SpSsp1, of 481 amino acid residues, containing a subtilisin domain. Expression analysis demonstrated that SpSsp1 is highly expressed in all tested mycelial stages of S. parasitica. Investigation of other non-infected trout from several fish farms in the United Kingdom showed similar activity in their sera towards SpSsp1. Several fish that had no visible saprolegniosis showed an antibody response towards SpSsp1 suggesting that SpSsp1 might be a useful candidate for future vaccination trial experiments.  相似文献

The emerging invasive fungal pathogen Aspergillus fumigatus causes very serious infections among immunocompromised patient populations. While the genome of this pathogen has been sequenced, a major barrier to better understanding the complex biology of this eukaryotic organism is a lack of tools for efficient genetic manipulation. To improve upon this, we have generated a new gene deletion system for A. fumigatus using yeast recombinational cloning and Agrobacterium tumefaciens mediated transformation (ATMT) employing a recyclable marker system. This system reduced the time for generating a gene deletion strain in our hands by two-thirds (12 weeks to 3 weeks) using minimal human labor, and we demonstrate that it can be used to efficiently generate multiple gene deletions within a single strain.  相似文献

Individual females from 19 populations of Heterodera glycines from China, Japan, and the United States were analyzed for esterase allozyme polymorphism. Eight esterase electrophoretic phenotypes were resolved. Four putative loci, est-1, est-2, est-3, and est-4, were identified, having one, one, two, and one allele, respectively. The four loci expressed six genotypes in the four Chinese populations. Loci est-2, est-3, and est-4 were identified in five Japanese populations and expressed five genotypes, whereas only loci est-2 and est-3 were identified in 10 populations from the United States and expressed four genotypes. Putative alleles at each locus were defined as characters for data analysis. Phylogenetic analysis using parsimony (PAUP) was utilized to determine relationships among the 19 populations. More loci and alleles in populations from China and phylogenetic similarities among populations from Japan and the United States are consistent with a founder effect resulting from dissemination of progenitor H. glycines from China to Japan and subsequent introductions of founder populations from Japan to the United States.  相似文献

Cryptosporidium species and Giardia intestinalis are the most common enteric protozoan pathogens affecting humans worldwide. In recent years, nitazoxanide has been licensed in the United States for the treatment of cryptosporidiosis in non-immunodeficient children and adults, becoming the first drug approved for treating this disease. There is a need for a highly effective treatment for cryptosporidiosis in immunodeficient patients, but the quest for such a drug has proven to be elusive. While not effective against Cryptosporidium, nitroimidazoles such as metronidazole or tinidazole are effective treatments for giardiasis and can be administered as a single dose. Albendazole and nitazoxanide are effective against giardiasis but require multiple doses. Nitazoxanide is the first new drug developed for treating giardiasis in more than 20 years. New potentially promising drug targets in Cryptosporidium and Giardia have been identified, but there appears to be little activity toward clinical development of new drugs.  相似文献

Ludwigia hexapetala and L. grandiflora are recent, aggressive invaders of freshwater wetlands in California. To assess the relative role of sexual versus clonal reproduction in invasive spread, we used AFLP markers to genotype 794 ramets of L. hexapetala sampled from 27 populations in three watersheds of California, and 150 ramets of L. grandiflora from five populations in a fourth watershed. We then used two analytical methods, similarity thresholds and character incompatibilities, to distinguish genotypic variation within genets (clones) from variation between genets. Our analyses revealed extremely limited genotypic and genet variation in invasive L. hexapetala and L. grandiflora within California. Within L. hexapetala, 95% of the ramets analyzed represented a single genet. The genet was the only one detected in 20 populations. The remaining seven populations contained two to nine genets. Within L. grandiflora, all ramets were of only one genotype. Thus, invasive spread within and between populations, and across watersheds, appears to be almost exclusively clonal and brought about by the dispersal of vegetative propagules. The extremely low seedling recruitment indicates that management should target vegetative dispersal and growth.  相似文献

The Asian shore crab, Hemigrapsus sanguineus, is one of the most abundant invasive crabs along the east coast of the United States. Larval stages are generally planktonic, but the megalopa stage settles to the substratum near the time of metamorphosis. Reducing the time to metamorphosis may result in higher recruitment and survival. Previous work has shown that a water-soluble cue produced by adult H. sanguineus can induce metamorphosis of conspecific megalopae. Here we report the results of experiments in which megalopae were exposed to cues produced by different life stages of H. sanguineus. We also provide data from experiments that investigated the temporal stability, detection threshold, and chemical classification of the cue. Our results indicate that an active cue is produced by juveniles as well as adults. The cue is proteinaceous and begins to degrade within 2 days of production. The threshold for detection of the cue by megalopae lies between 0.1 and 0.01 µg of protein per ml.  相似文献

Background and Aims

Black cherry (Prunus serotina) is a North American tree that is rapidly invading European forests. This species was introduced first as an ornamental plant then it was massively planted by foresters in many countries but its origins and the process of invasion remain poorly documented. Based on a genetic survey of both native and invasive ranges, the invasion history of black cherry was investigated by identifying putative source populations and then assessing the importance of multiple introductions on the maintenance of gene diversity.

Methods

Genetic variability and structure of 23 populations from the invasive range and 22 populations from the native range were analysed using eight nuclear microsatellite loci and five chloroplast DNA regions.

Key Results

Chloroplast DNA diversity suggests there were multiple introductions from a single geographic region (the north-eastern United States). A low reduction of genetic diversity was observed in the invasive range for both nuclear and plastid genomes. High propagule pressure including both the size and number of introductions shaped the genetic structure in Europe and boosted genetic diversity. Populations from Denmark, The Netherlands, Belgium and Germany showed high genetic diversity and low differentiation among populations, supporting the hypothesis that numerous introduction events, including multiple individuals and exchanges between sites, have taken place during two centuries of plantation.

Conclusions

This study postulates that the invasive black cherry has originated from east of the Appalachian Mountains (mainly the Allegheny plateau) and its invasiveness in north-western Europe is mainly due to multiple introductions containing high numbers of individuals.  相似文献

One to two hundred nematodes from each of seven Xiphinema americanum-group populations were measured to determine the range of stylet and body lengths for juveniles and adults. First-stage juveniles were identified by the position of the replacement odontostyle (i.e., the tip of the replacement odontostyle overlapped the base of the odontophore). Nematodes were identified as second stage if the functional odontostyle was the same length as the replacement odontostyle of the first stage. Subsequent stages were similarly identified by establishing the range of corresponding replacement and functional odontostyle lengths. In all populations examined, this procedure created natural divisions that clearly grouped nematodes by stylet and body length. Presumably these groups identified all juvenile and adult stages. Populations of X. americanum, X. rivesi, and X. californicum from the United States had three juvenile stages, but a population of X. pachtaicum from Bulgaria had four juvenile stages.  相似文献

Nosema ceranae is a recently described pathogen of Apis mellifera and Apis cerana. Relatively little is known about the distribution or prevalence of N. ceranae in the United States. To determine the prevalence and potential impact of this new pathogen on honey bee colonies in Virginia, over 300 hives were sampled across the state. The samples were analyzed microscopically for Nosema spores and for the presence of the pathogen using real-time PCR. Our studies indicate that N. ceranae is the dominant species in Virginia with an estimated 69.3% of hives infected. Nosema apis infections were only observed at very low levels (2.7%), and occurred only as co-infections with N. ceranae. Traditional diagnoses based on spore counts alone do not provide an accurate indication of colony infections. We found that 51.1% of colonies that did not have spores present in the sample were infected with N. ceranae when analyzed by real-time PCR. In hives that tested positive for N. ceranae, average CT values were used to diagnose a hive as having a low, moderate, or a heavy infection intensity. Most infected colonies had low-level infections (73%), but 11% of colonies had high levels of infection and 16% had moderate level infections. The prevalence and mean levels of infection were similar in different regions of the state.  相似文献

Seven field populations of Xiphinema americanum sensu lato from California''s major agronomic areas were tested for their ability to transmit two nepoviruses, including the prune brownline, peach yellow bud, and grapevine yellow vein strains of tomato ringspot virus and the bud blight strain of tobacco ringspot virus. Two field populations transmitted all isolates, one population transmitted all tomato ringspot virus isolates but failed to transmit bud blight strain of tobacco ringspot virus, and the remaining four populations failed to transmit any virus. Only one population, which transmitted all isolates, bad been associated with field spread of a nepovirus. As two California populations of Xiphinema americanum sensu lato were shown to have the ability to vector two different nepoviruses, a nematode taxonomy based on a parsimony of virus-vector relationship is not practical for these populations. Because two California populations of X. americanum were able to vector tobacco ringspot virus, commonly vectored by X. americanum in the eastern United States, these western populations cannot be differentiated from eastern populations by vector capability tests using tobacco ringspot virus.  相似文献

Though in the past Xiphinema americanum has been the most commonly reported dagger nematode in the eastern United States, our studies revealed the presence in Pennsvlvania of a previously unrecognized and unreported species related to X. americanum, Morphometric data and photomicrographs establish the identity of this form as X. rivesi and show expected variations in populations of this species from various locations. Similar data and illustrations are given for X. americanum populations from Pennsylvania and other areas, showing variations and relationships. Xiphinema rivesi is widely distributed in the fruit producing area of south-central Pennsylvania and is also reported herein from raspberry in Vermont and apple in Maryland and New York. This species is frequently found in fruit growing areas of Pennsylvania associated with tomato ringspot virus-induced diseases and is also found associated with corn, bluegrass sod, and alfalfa.  相似文献

The known range of the eastern oyster (Crassostrea virginica) parasite, Perkinsus marinus, expanded into the northeastern United States in the early 1990s. We used both in vitro and in vivo data to test the hypothesis that the northward expansion was associated with a low-temperature adapted strain of the parasite. In vitro proliferation of nine P. marinus isolates from three geographic sites, Massachusetts and New Jersey in the new range, and South Carolina in the historic southern range, was measured at seven temperatures (5 to 35 °C) using a tetrazolium blue dye assay. We wanted to determine if there were between- and within-geographic location differences in the P. marinus proliferation rate, and if so, whether they were associated with temperature. We found no evidence of low-temperature adaptation based on the fact that net proliferation rates for isolates from all three geographic locations were similar at temperatures from 5 to 20 °C. On the other hand, at temperatures of 25 to 35 °C, the South Carolina isolates exhibited higher proliferation rates than the northern isolates suggesting possible high-temperature adaptation of parasite strains that are routinely exposed to higher temperatures. Although there was significant within-location variation among isolates, the data tended to group together by geographic location supporting the hypothesis that there is an important regional component to the proliferation rate of P. marinus isolates. A survey of published data showed that the temperature at which in vivo proliferation was first observed in oysters at sites from the Gulf of Mexico to Massachusetts was typically between 20 and 23 °C with no evidence of a geographic cline. These results lend support to the hypothesis that the recent warming trend in the northeastern US is the most likely explanation for the P. marinus range extension.  相似文献

Morphometrics of 23 United States populations of Xiphinema americanum sensu lato, sharing the characteristics of an offset lip region and conoid tail, were examined and analyzed statistically by canonical discriminant analysis (CDA). Specimens were collected from Arkansas, Georgia, Tennessee, Mississippi, Florida, Oklahoma, California, and North Dakota. Eleven measurements and body ratios obtained from female specimens were used in the analysis. Xiphinema americanum, X. bricolensis, X. californicum, X. citricolum, X, intermedium, X. tarjanense, and X. thornei, and one undescribed species were identified among the 23 populations. Three groups -- X. americanum-group, X. californicum-group, and X. intermedium-group (X. intermedium and X. tarjanense) -- were formed and four populations belonging to four different species were separated consistently from these groups in CDA scatterplots of the 23 populations. Composition of the groups was somewhat related to the geographical origins of the populations in the groups. A population from California had morphometrics intermediate between X. americanum and X. californicum. Separation between the X. americanum-group and X. californicum-group in the CDA scatterplots was not as distinct as that between them and the X. intermedium-group or between any of the three groups and the four single outlying populations.  相似文献
