Biosynthesis and turnover of carnitine acetyltransferase in rat liver   总被引:2,自引:0,他引:2  
Male Wistar rats were fed on a diet with and without di(2-ethylhexyl)phthalate (DEHP) for 2 weeks. Carnitine acetyltransferase in the liver was increased about 100-fold by administration of DEHP. The results of in vivo experiments showed that the incorporation of L-[4,5-3H]leucine into the enzyme was 12-fold higher and the half-life of the labeled enzyme was elongated by a factor 4.6. The results of in vitro translation experiments with total hepatic RNA in a rabbit reticulocytelysate system and the results concerning the synthesis of the enzyme in isolated hepatocytes indicate that the translatable mRNA for the enzyme was increased upon administration of DEHP and that the enzyme is synthesized as a precursor (Mw = 69,000) larger than the mature enzyme (Mw = 67,500). RNA in the free polysomes directed the synthesis of the enzyme precursor five times more actively than RNA in membrane-bound polysomes.  相似文献

The cytosol proteins of mouse or rat liver were separated on the basis of charge and characterized with respect to molecular size, turnover in vivo, and several chemical properties. The basic proteins, which were synthesized and degraded slower than acidic proteins, were generally smaller, as multimers and subunits, than the acidic proteins. Charge and size, therefore, did not appear to be independent characteristics for soluble liver proteins. The amino acid composition of the smaller, basic, stable proteins was very similar to that of the larger, acidic, labile proteins. The charge differences between these two protein fractions could be attributed to the ratio of acidic to basic amino acid residues rather than to carbohydrate, nucleic acid, or phosphate content. The percentage of hydrophobic amino acid residues in the two protein fractions was the same. The acidic labile proteins were more vulnerable to proteolysis and associated with the lysosomal-mitochondrial fractions somewhat more extensively in vitro than the basic, stable proteins. These studies indicate that the information determining the half-lives of soluble enzymes is in the protein moiety of those enzymes and that the factors that correlate with turnover may not be independent variables for soluble mammalian proteins.  相似文献

In plasma membrane glycoproteins of rat liver L-fucose undergoes a rapid intramolecular turnover in that fucose residues are removed from the glycoproteins (Tauber, R., Park, C.S. & Reutter, W. (1983) Proc. Natl. Acad. Sci. U.S.A. 80, 4026-4029). The present paper demonstrates that the intramolecular turnover of L-fucose is markedly decreased during liver regeneration. Turnover half-lives of L-fucose were measured in regenerating liver by pulse-chase experiments in five plasma membrane glycoproteins (Mr 60,000 (gp60), 80,000 (gp80), 120,000 (gp120), 140,000 (gp140), and 160,000 (gp160). The glycoproteins were isolated from plasma membranes by concanavalin A-Sepharose affinity chromatography and semipreparative NaDodSO4 polyacrylamide gel electrophoresis. L-Fucose turned over in the five glycoproteins with heterogeneous half-lives ranging from 22 h (gp160) to 49 h (gp120). The protein moieties of the glycoproteins were degraded with half-lives ranging from 56 h (gp80) to 107 h (gp140). Relative to the half-life of the protein backbone the half-live of L-fucose was increased in the five membrane glycoproteins by 70% (gp60), 150% (gp80), 182% (gp120), 60% (gp140) and 16% (gp160) during liver regeneration when compared to normal liver. The data show that L-fucose turns over in different membrane glycoproteins with individual rates, and that loss of L-fucose from plasma membrane glycoproteins is reduced in rapidly proliferating liver after partial hepatectomy.  相似文献

125I-labelled angiotensin II (A II) specifically binds to solubilized receptors extracted from rat isolated glomeruli using CHAPS (3-[3-( cholamidopropyl ) dimethylammonio ]-1-propanesulfonate). The yield of solubilization of the binding sites was 3.3%. Equilibrium was reached after 15-20 min and specific binding represented 75% of total binding. Dissociation of the hormone-receptor complex after addition of an excess of A II was very slow in the presence of Ca2+ and Mg2+. [Sar1 Ala8] A II and [Sar1 Ile8] A II were more potent as competitive inhibitors of 125I-labelled A II than A II itself and its heptapeptide. These basic features of 125I-labelled A II binding to the extracted material were similar to those observed previously with untreated glomeruli.  相似文献

The protein degradation rate of total liver homogenate as well as of the liver cytosol was measured according to the method introduced by GARLICK and coworkers (Biochem. J. 156 (1976) 657-663). The half-lives were 2.85 days (cytosol) and 2.45 days (homogenate) in the group consisting of 29 months old rats and 2.35 days and 2.15 days in the control group consisting of 12 months old rats. The amount of cathepsin D determined in some organs by two independent methods (immunological and enzymic) revealed differences in the group of the old animals. We suggest the accumulation of cross reacting material of cathepsin D in rats of the old group. It is perhaps one of the reasons why a slower protein degradation is observed in old animals.  相似文献

Endogenous triglyceride turnover in liver and plasma of the dog   总被引:8,自引:0,他引:8  
Radioactive glycerol and S(f) > 20 lipoproteins labeled with it were used to study turnover of plasma S(f) > 20 and hepatic triglyceride in anesthetized dogs. From specific activity-time curves of these lipids after an injection of labeled material, a tentative and incomplete model for the kinetics of endogenous hepatic and plasma triglyceride was defined and partially validated. Pool sizes and turnover rates of triglyceride in liver and S(f) > 20 lipoproteins of plasma were then calculated in seven dogs. Hepatic triglyceride was composed of two compartments: 60% metabolically inert and 40% metabolically active. Although communication between these hepatic compartments surely occurred during the time course of these studies, it was not sufficient to be detected by our present methods. The metabolically active compartment turned over as a single pool but with two destinations: a quite variable proportion (an average of 61%) was secreted into plasma as S(f) > 20 triglyceride, and an average of 39% was presumably hydrolyzed within the liver. The fractional turnover rate of plasma S(f) > 20 triglyceride was 2-3 times that of hepatic triglyceride. This finding, and the parallel decline of specific activities of plasma S(f) > 20 and liver triglyceride after injection of labeled glycerol, confirm the rate-determining role of hepatic triglyceride. In this respect the dog differs importantly from man. Though turnover rates of plasma S(f) > 20 triglyceride fell in the same range in men and dogs, the relationship of turnover rate to plasma concentration of this lipid differed greatly between them. The model for the dog does resemble that previously reported for man, however, in the lack of major recycling of intact plasma triglyceride between the liver and plasma. Lack of such recycling, however, does not exclude return of plasma triglyceride into a hepatic triglyceride sink. The amount of such unidirectional uptake, if any, could not be determined by these techniques.  相似文献

Using kinetics of approach to steady state labeling, we have found that liver poly(A)+ mRNAs fall into three populations, differing in stability and probably in processing, as reflected in their dissimilar delays in reaching polyribosomes and turnover times. There are mRNA-1 (delay 10 min, half-life 1 hr); mRNA-2 (delay 3 hr, half-life 2 hr); and mRNA-3 (delay 40 min, half-life 2.6 hr). The first two species function on free polyribosomes while the third one is operating on bound polyribosomes. The apparent turnover rates of some mitochondrial enzymes can be modified in diabetes. We studied the effect of streptozotocin-diabetes on the half-life of a protein tightly bound to the inner membrane, ATPase. The half-life (t 1/2), measured by the double-isotope technique, decreased by approx. 20% in diabetes (from approximately equal to 2.56 days in controls to approximately equal to 2.06 days in diabetic rats). These results suggest that diabetes produces an increase in degradation of ATPase by a mechanism which is not yet clear, possibly influenced by alterations induced by diabetes in hepatic lysosomes that are associated with hepatic autophagy.  相似文献
