降钙素对骨质疏松大鼠骨密度形态计量学与骨代谢的影响   总被引:2,自引:1,他引:2  
目的探讨降钙素(密盖息)对骨质疏松大鼠骨密度、骨形态计量学影响以及与血钙、磷、维生素D代谢和生长因子的关系。方法用摘除大鼠双侧卵巢的方式制备骨质疏松模型(OVX),实验动物分为4个组:模型对照组、密盖息治疗组,盐酸雷洛昔芬治疗组,假手术组。应用HOLOGIC第4代双能X线4500W骨密度仪测定大鼠腰椎、股骨上段骨密度值(BMD);以骨形态计量学测股骨骨小梁面积、矿化沉积率;用ELISA法测定血清IGF-1水平和血清25OHVitD浓度以及血淋巴细胞维生素D受体(VDR)含量。结果密盖息治疗组、盐酸雷洛昔芬治疗组均较OVX组腰椎、股骨上段骨密度增高,组间比较差异有显著性(P<0.01)。密盖息治疗组较盐酸雷洛昔芬治疗组股骨上段骨密度增高,两组之间差异有显著性(P<0.01)。密盖息治疗组骨小梁面积明显增加、矿化沉积率增高。密盖息治疗组、盐酸雷洛昔芬治疗组血清IGF-1浓度值、血清25-OHVitD浓度值升高,与OVX组比较差异有显著性(P<0.01)。各组血淋巴细胞VDR含量无明显变化,与OVX组比较差异无显著性(P>0.05)。结论密盖息能够预防腰椎、股骨上段骨密度丢失,使骨小梁面积明显增加、矿化沉积率增高并且血清IGF-1及血清25-OHVitD浓度值升高,但对VDR含量无明显作用。  相似文献

斯钙素(stanniocalcin,STC)是一种最早在硬骨鱼中发现的糖蛋白类激素. 哺乳动物斯钙素在体内钙磷代谢、肌肉骨骼系统的发育等方面均起到重要作用,并且在心血管疾病、肿瘤发展以及神经系统疾病中也扮演重要角色. 近年,斯钙素在骨骼发育中的作用逐渐引起科学界的关注. 骨组织中STC由成软骨细胞、成骨细胞分泌,并以自分泌/旁分泌的形式作用于局部组织细胞中,主要影响软骨形成和骨重建过程.本文以斯钙素为主题,综述了其生化分子特性、其在骨组织中的表达分布特点,以及该分子在成熟骨组织骨重建过程中的作用机制. 本文将为深入了解斯钙素在骨组织代谢中的作用提供帮助.  相似文献

在砂培条件下 ,研究施加钙、砷对蜈蚣草生长和砷、磷和钙的吸收及转运的影响。添加砷对蜈蚣草的生物量 (根、叶柄和羽叶的干物重 )虽未达到显著影响 (p<0 .0 5) ,但添加 0 .1 mmol/L砷时 ,表现出刺激生长效应。提高介质中钙浓度明显抑制蜈蚣草根系生长 ,钙浓度过高还会显著限制地上部生长。供应 0 .0 3mmol/L钙时 ,蜈蚣草羽片砷浓度为 42 1 8mg/kg,明显高于 2 .5和 5 mmol/L钙处理下相应的砷浓度。砷的转运系数 (羽片 /根 )随着介质中砷浓度的升高而增大 ,随着介质中钙浓度的升高而减少。这说明一定范围内提高介质中砷浓度促进砷向地上部运输 ,而钙却明显抑制砷向地上部转运。钙和砷浓度过高时 ,植株均会出现中毒症状。钙中毒表现为叶脉变褐和叶肉坏死 ;而砷中毒现象表现在叶尖和叶缘变褐。介质中砷限制蜈蚣草根部对磷的吸收 ,但对地上部磷浓度无显著影响。介质中添加砷 ,植物体内钙浓度升高 ,可能起缓解砷毒的作用。钙、砷对蜈蚣草羽片砷累积量和总累积量均有极显著的交互作用 ,钙是负交互效应 ,砷是正交互效应。添加 2 .5和 5.0 mmol/L钙时 ,相对于 0 .0 3 mmol/L钙处理分别减少地上部砷累积量 2 0 .8%和73.1 %。这表明在应用蜈蚣草进行植物修复时 ,介质中出现过高浓度的钙是不利于提高土壤修复效率  相似文献

目的:研究骨碎补总黄酮对切除卵巢的雌性大鼠血清骨钙素和骨形态蛋白-2(bone morphogenetic protein-2,BMP-2)在骨组织中表达的影响。方法:取3月龄雌性大鼠30只,随机分成假手术组、给药组和空白对照组。假手术组仅去除卵巢周围脂肪,给药组在去除卵巢后连续15周灌胃给服强骨胶囊,其余给等量自来水。15周后处死所有大鼠,取股骨中段组织,行BMP-2免疫组化染色,镜下观察染色标本摄像并用Image图形分析软件分析各标本阳性表达率。另取大鼠血清以放免法测定其血清骨钙素含量。结果:免疫组化染色后,图片分析显示给药组和假手术组bmp-2阳性率明显高于对照组(p<0.05);给药组和假手术组血清骨钙素的表达明显高于对照组(p<0.05)。结论:骨碎补总黄酮对卵巢切除大鼠的BMP-2和血清骨钙素表达有促进作用。  相似文献

将不同剂量的重组人骨形态发生蛋白-7(rhBMP-7)与脱钙骨基质(DBM)分别复合后,植入小鼠股部内侧肌间隙,三周后取材,通过组织学检查、碱性磷酸酶(ALP)及钙含量的测定比较各组的骨诱导活性。结果显示,三组复合物均有骨组织生成,中、高剂量组可见骨小梁、板层骨和原始骨髓腔,血管和骨髓丰富;低剂量组的成骨量明显少于中、高剂量组,且新骨的成熟度低于其他两组,DBM少部分吸收;单独植入rhBMP-7组有编织骨形成;而DBM组可见成骨细胞的聚集。rhBMP-7/DBM复合组在ALP和Ca含量水平上与同等剂量的两对照组相比均有显著性差异(P<0.01),而rhBMP-7三种剂量之间均有显著性差异(P<0.01)。这充分说明DBM作为rhBMP-7的合适载体,具有缓释作用,且二者复合可起到双重骨诱导活性;而且rhBMP-7的骨诱导活性具有一定的剂量依赖性。  相似文献

比较不同频率的正弦交变电磁场对SD青年大鼠骨密度及骨形态计量指标的影响,筛选可有效提升大鼠骨密度的频率参数。将32只8周龄SD雌性大鼠随机分为4组:对照组、15 Hz组、30 Hz组、45 Hz组;除对照组外,实验组大鼠每天都给予相应频率的1.8 m T正弦交变电磁场干预,干预时间为90 min。磁场干预8周后,双能X射线骨密度仪检测大鼠全身骨密度、右侧股骨骨密度和椎骨骨密度,ELISA分析血清中骨形成与骨吸收生化指标的含量,右侧胫骨进行荧光间距测量与骨形态计量分析。相比于对照组,15 Hz组、45 Hz组大鼠的全身骨密度、股骨骨密度、椎骨骨密度均明显升高(P0.05),血清中骨钙素与骨保护素含量也显著提升(P0.05);实验组大鼠的胫骨双荧光间距与骨组织静态参数均高于对照组(P0.05)。结果表明,15 Hz、45 Hz正弦交变电磁场可有效提升青年大鼠的骨密度,从而可预防骨质疏松的发生。  相似文献

骨唾液酸蛋白(Bonesialoprotein,BSP)是细胞外基质中一种高度磷酸化和糖基化分泌性蛋白,它是多种癌症(乳腺癌,前列腺癌和肺癌等)进程中的重要参与者。乳腺癌细胞转染实验和裸鼠移植模型证明过表达BSP可以促进乳腺癌细胞骨转移。BSP抗体和反义核酸均可有效抑制乳腺癌细胞骨转移的发生,故BSP有可能成为一种新的诊断和治疗乳腺癌骨转移的靶蛋白。  相似文献

目的研究饲料中不同的钙磷含量和钙磷比对生长期实验大鼠体重及骨骼发育的影响。方法采用完全随机化设计方案,取1月龄(100±10)g清洁级Wistar雄性大鼠70只,随机分成7组,每组10只,饲喂7种不同钙磷含量的饲料。自由采食,饮用去离子水,试验期42 d。各组于实验开始和结束时称重。试验结束后,处死动物并分离一侧胫骨和股骨用于实验指标的检测。结果当保持饲料中钙或者磷中的一种含量不变的情况下,调整另一种的含量即改变钙磷比例时,1.2∶1的正常钙磷比例组生长期实验大鼠增重及骨骼发育要明显好于0.4∶1的低钙磷比例组和4∶1的高钙磷比例组(P〈0.05),并且当保持饲料中钙磷比例不变时,即使在低钙低磷和高钙高磷进食水平下,正常钙磷比例组大鼠增重及骨骼发育都较好,但低钙磷比例组和高钙磷比例组大鼠增重及骨骼发育则由于饲料中钙磷含量的变化波动较大(P〈0.05)。结论饲料中钙和磷需按比例添加,如果两者绝对含量相差过大就会影响到动物的增重及骨骼发育,本实验证明饲料钙磷比为1.2∶1时生长期实验大鼠增重及骨骼发育较好。  相似文献

The effects of estradiol-17β (E2) on bone resorption and formation as well as its effects on scale resorption were investigated in rainbow trout in order to elucidate the role of the hormone in calcium mobilization from calcified tissues, and to clarify the importance of scale and bone as calcium reserves during sexual maturation. Furthermore, the effects of nutritional status on calcified tissues and E2-induced calcium mobilization were studied. In fed as well as fasted rainbow trout, E2 treatment increased scale osteoclastic activity measured as tartrate-resistant acid phosphatase activity, and reduced scale calcium content, suggesting that E2 increases scale resorption in both the fed and fasted fish. Using histomorphometry, E2 treatment was found to decrease pharyngeal bone resorption in fed, but not in fasted rainbow trout. The E2 effect on rainbow trout bone is consistent with its physiological role in mammals and birds where E2 has been reported to decrease bone resorption. It appears therefore that rainbow trout protect their skeleton and instead use scales as a source of calcium during E2-induced calcium mobilization. The formation of pharyngeal bone was decreased by fasting, and the importance of the nutritional status for the activity of the bone cells in rainbow trout is therefore emphasized. Fluorochrome labelling in Roman period skeletons from Dakhleh Oasis, Egypt   总被引:1,自引:0,他引:1  
A histological aging study of femoral midshafts in a late Roman period sample from the Dakhleh Oasis, Egypt, showed discrete fluorochrome labelling. The fluorochrome is yellow-green in colour and fluoresces at a wavelength of 525 nm. The labelling occurs at the mineralization fronts and is so distinct that several histomorphometric measurements were made. The percent labelled surface values ranged from 6.03% to 59.34%, and the mean distance between labels ranged from 13.57 to 20.63 microns. Teeth from several individuals were also labelled within the enamel matrix. Comparisons were made between the patterns of fluorescent labelling from this population and from patients treated with interval and continuous dosages of tetracycline. The results collectively indicate that the fluorochrome is most likely tetracycline that made its way into the bone (in vivo) via stored grain contaminated by Streptomycetes. The labelling was differential, much like that of the patient on interval doses of tetracycline, so it is argued that the tetracycline was ingested occasionally, probably on a seasonal basis. The lack of bone infection in the adults (n = 29) suggests that the tetracycline may have provided some antibiotic protection in this population.  相似文献

Bone sialoprotein is an extracellular noncollagenous acidic protein that plays a role in bone mineralization and remodeling. Its expression is restricted to mineralized tissues and is subjected to variety of posttranslational modifications including phosphorylation and glycosylation. We have expressed the full-length and half domains of bovine bone sialoprotein in a prokaryotic system and identified the phosphorylation sites of casein kinase II. The N-terminal automated solid-phase sequencing defined four phosphorylated peptides: residues 28-38 (LEDS(P)EENGVFK), 51-86 (FYPELKRFAVQSSS(P)DS(P)S(P)EENGNGDS(P)S(P)EEEEEEEETS(P)), 151-165 (EDES(P)DEEEEEEEEEE), and 295-305 (GRGYDS(P)YDGQD). Nine phosphoserines were identified within the four peptides. Seven of them were in the N-terminus (S31, S64, S66, S67, S75, S76, and S86) and two were in the C-terminus (S154 and S300) of the protein.  相似文献

The effect of extracellular calcium on human osteoblast-like cells (HOS) has been demonstrated. An experimental setup was used for applying defined rates of change in the extracellular calcium concentration. The intracellular calcium concentration was monitored using the fluorescence dye fura-2. HOS cells showed qualitatively different responses of the intracellular calcium concentration to changes of the extracellular calcium concentration depending on its changing rate. A small rate caused only a small and slow increase of the intracellular calcium concentration, whereas faster changes are able to cause a rapid transient increase followed by a sustained elevation of the internal calcium level. Surprisingly, both an increasing as well as a decreasing external calcium concentration is able to cause cellular responses. These signals could be reduced by the IP3-inhibitor neomycin. We propose that the G-protein dependent signalling pathway of HOS cells can not only sense the extracellular calcium concentration but also its time derivative. 2型糖尿病模型GK大鼠骨形态学和生物力学特点观察   总被引:1,自引:0,他引:1  
目的观察2型糖尿病模型GK大鼠的骨代谢特点以及骨密度和骨生物力学特性的变化。方法采用雄性6月龄GK大鼠10只,以月龄、性别匹配的健康Wistar大鼠作为正常对照。颈静脉取血检测与骨代谢有关的生化指标。DXA法测定股骨和腰椎骨密度,并行股骨三点弯曲实验和腰椎压缩实验。甲基丙烯酸甲酯包埋胫骨干骺端以制备不脱钙骨切片。应用多媒体病理图像分析软件进行骨组织形态计量学分析。结果GK大鼠体重明显低于健康对照Wistar大鼠(P〈0.01)。与对照组相比,GK大鼠血清骨钙素水平明显降低[(4.97±0.49,6.75±0.71)μg/mL,P〈0.01],而抗酒石酸酸性磷酸酶活性明显升高[(17.92±5.23,8.31±2.69)U/L,P〈0.01],但血钙和血磷无明显变化(P〉0.05);股骨和腰椎骨密度显著降低[(0.16±0.01,0.22±0.02;0.12±0.01,0.16±0.02)g/cm2,P〈0.01];骨强度和腰椎的弹性模量明显降低(P〈0.01)。骨形态学分析显示GK大鼠股骨长度和第五腰椎高度分别降低10.3%和9.5%(P〈0.01),股骨和腰椎横截面积无明显变化(P〉0.05)。骨组织形态计量学分析显示,GK大鼠骨小梁体积、骨小梁厚度、类骨质表面和厚度明显降低[(15.72±1.18,19.13±1.13)%,(61.91±4.54,74.43±3.63)μm,(18.18±1.25,19.63±1.07)%,(3.68±0.48,4.34±0.35)μm,P〈0.01或0.05],动态参数如矿化表面、矿化沉积率和骨形成率也明显降低[(17.77±1.54,19.56±2.07)%,(1.07±0.22,1.35±0.16;0.20±0.03,0.26±0.04)μm/day,P〈0.05或0.01],而矿化延迟时间显著延长(2.66±0.56,2.12±0.35,P〈0.05)。结论非肥胖的GK大鼠表现有骨量减少和骨折危险性增加;2型糖尿病本身可干扰成骨细胞功能和活性而导致骨重建失衡。  相似文献

The determination of calcium content in human bone tissue is very useful in metabolic diseases of bone, such as renal osteodystrophy, osteoporosis, hyperparathyroidism, and osteomalacia of diverse etiology. The PIXE technique allows calcium to be directly determined in bioptic tissue sections properly, sampled for histological optical and/or electron microscopy examination. Bone semithin sections (3 μm thick, 4×4 mm2 dimensions), cut by ultramicrotome and deposited onto polyvinyl acetate films, underwent PIXE analysis using the CISE set-up. Histomorphometric (after standard staining), evaluation of calcified bone volume (CBV) in absolute value allows calcium density to be determined. A total of nine bone biopsies were analyzed (three sections each) obtaining values ranging between 352 and 482, with an average value of 421.5±15.3 (M±SE) μg/μL, in good agreement with literature data (obtained by AAS technique on dissected bone samples). The aim of this paper is to emphasize the usefulness, of combined PIXE and histomorphometric techniques for the study of calcium content in bone tissue in both healthy and diseased bones.  相似文献
