The role of a recently identified K+ATP channel in preventing H2O2 formation was examined in isolated pea stem mitochondria. The succinate-dependent H2O2 formation was progressively inhibited, when mitochondria were resuspended in media containing increasing concentration of KCl (from 0.05 to 0.15  M ). This inhibition was linked to a partial dissipation of the transmembrane electrical potential (ΔΨ) induced by KCl. Conversely, the malate plus glutamate-dependent H2O2 formation was not influenced. The succinate-sustained H2O2 generation was also unaffected by nigericin (a H+/K+ exchanger), but completely prevented by valinomycin (a K+ ionophore). In addition, cyclosporin A (a K+ATP channel opener) inhibited this H2O2 formation, while ATP (an inhibitor of the channel opening) slightly increased it. The inhibitory effect of ATP was strongly stimulated in the presence of atractylate (an inhibitor of the adenine nucleotide translocase), thus suggesting that the receptor for ATP on the K+ channel faces the intermembrane space. Finally, the succinate-dependent H2O2 formation was partially prevented by phenylarsine oxide (a thiol oxidant).  相似文献

Polyunsaturated fatty acids induce stomatal opening (Y. Lee, H. Lee, R. C. Crain, A. Lee and S. J. Korn. 1994. Cell Signal. 6: 181–186), but it is not known whether they function as second messengers in guard cells exposed to signals that open stomata. To test the hypothesis that phospholipase A2 (PLA2), which produces fatty acids and lysophospholipids, is involved in light signal transduction in guard cells, we treated epidermal peels of Commelina communis L. with PLA2 inhibitors and followed the changes in stomatal apertures in response to light. Stomatal opening by white, blue, or red light was inhibited by 2–3 different PLA2 inhibitors in concentration ranges that have been reported to inhibit PLA2 activity. However, the PLA2 inhibitors could not block stomatal opening induced by a polyunsaturated fatty acid. These results suggest that PLA2 functions as a signal transducer for both blue and red light in guard cells.  相似文献

Light-induced stomatal opening in C3 and C4 plants is mediated by two signalling pathways. One pathway is specific for blue light and involves phototropins, while the second pathway depends on photosyntheticaly active radiation (PAR). Here, the role of Nt MPK4 in light-induced stomatal opening was studied, as silencing of this MAP kinase stimulates stomatal opening. Stomata of Nt MPK4-silenced plants do not close in elevated atmospheric CO2, and show a reduced response to PAR. However, stomatal closure can still be induced by abscisic acid. Measurements using multi-barrelled intracellular micro-electrodes showed that CO2 activates plasma membrane anion channels in wild-type Nicotiana tabacum guard cells, but not in Nt MPK4-silenced cells. Anion channels were also activated in wild-type guard cells after switching off PAR. In approximately half of these cells, activation of anion channels was accompanied by an increase in the cytosolic free Ca2+ concentration. The activity of anion channels was higher in cells showing a parallel increase in cytosolic Ca2+ than in those with steady Ca2+ levels. Both the darkness-induced anion channel activation and Ca2+ signals were repressed in Nt MPK4-silenced guard cells. These data show that CO2 and darkness can activate anion channels in a Ca2+-independent manner, but the anion channel activity is enhanced by parallel increases in the cytosolic Ca2+ concentration. Nt MPK4 plays an essential role in CO2- and darkness-induced activation of guard-cell anion channels, through Ca2+-independent as well as Ca2+-dependent signalling pathways.  相似文献

Abstract: We examined the mechanism underlying the ATP-induced increase in the cytosolic Ca2+ concentration ([Ca]in) in acutely isolated chick ciliary ganglion neurons, using fura-2 microfluorometry. The ATP-induced increase in [Ca]in was dependent on external Ca2+, was blocked in a dose-dependent manner by reactive blue 2, and was substantially inhibited by both L- and N-type Ca2+ channel blockers. ATP was effective in increasing [Ca]in in the presence of a desensitizing concentration of nicotine (100 µ M ), and simultaneous addition of maximal doses of ATP and nicotine caused an additive increase in [Ca]in, suggesting that ATP acts on a site distinct from nicotinic acetylcholine receptors. ATP also increased the cytosolic Na+ concentration as determined by sodium-binding benzofuran isophthalate microfluorometry. These results suggest that ATP increases Na+ influx through P2 purinoceptor-associated channels resulting in membrane depolarization, which in turn increases Ca2+ influx through voltage-dependent Ca2+ channels. However, ATP still caused a small increase in [Ca]in under Na+-free conditions, and this [Ca]in increase was little affected by Ca2+ channel blockers. ATP also increased Mn2+ influx under Na+-free conditions, as indicated by quenching of fura-2 fluorescence. These results suggest that nonselective cationic channels activated by ATP are permeable not only to Ca2+ but also to Mn2+, in addition to monovalent cations.  相似文献

The oxygen requirement for stomatal opening in maize plants ( Zea mays L. hybrid INRA 508) was studied at different CO2 concentrations and light intensities. In the absence of CO2, stomatal opening always required O2, but this requirement decreased with increasing light intensity. In darkness, the lowest O2 partial pressure needed to obtain a weak stomatal movement was about 50 Pa. This value was lowered to ca 10 Pa in light (320 μmol m−2 s−1).
On the other hand. in the absence of O2, CO2enabled stomatal opening to occur in the light, presumably due to the evolved photosynthetic O2. Thus, CO2, which generally reduced stomatal aperture, could induce stomatal movement in anoxia and light. The effect of CO2 on stomatal opening was closely dependent on O2 concentration and light intensity. Stomatal aperture appeared CO2-independent at an O2 partial pressure which was dependent on light intensity and was about 25 Pa at 320 umol m−2 s−1.
The presence of a plasmalemma oxidase, in addition to mitochondrial oxidase, might explain the differences in the O2 requirement at various light intensities. The possible involvement of such a system in relation to the effect of CO2 is discussed.  相似文献

Numerous studies conducted on both whole plants and isolated epidermes have documented stomatal sensitivity to CO2. In general, CO2 concentrations below ambient stimulate stomatal opening, or an inhibition of stomatal closure, while CO2 concentrations above ambient have the opposite effect. The rise in atmospheric CO2 concentrations which has occurred since the industrial revolution, and which is predicted to continue, will therefore alter rates of transpirational water loss and CO2 uptake in terrestrial plants. An understanding of the cellular basis for guard cell CO2 sensing could allow us to better predict, and perhaps ultimately to manipulate, such vegetation responses to climate change. However, the mechanisms by which guard cells sense and respond to the CO2 signal remain unknown. It has been hypothesized that cytosolic pH and malate levels, cytosolic Ca2+ levels, chloroplastic zeaxanthin levels, or plasma-membrane anion channel regulation by apoplastic malate are involved in guard cell perception and response to CO2. In this review, these hypotheses are discussed, and the evidence for guard cell acclimation to prevailing CO2 concentrations is also considered.  相似文献

Abstract: Cultured astroglia express both adenosine and ATP purinergic receptors that are coupled to increases in intracellular calcium concentration ([Ca2+]i). Currently, there is little evidence that such purinergic receptors exist on astrocytes in vivo. To address this issue, calcium-sensitive fluorescent dyes were used in conjunction with confocal microscopy and immunocytochemistry to examine the responsiveness of astrocytes in acutely isolated hippocampal slices to purinergic neuroligands. Both ATP and adenosine induced dynamic increases in astrocytic [Ca2+]i that were blocked by the adenosine receptor antagonist 8-( p -sulfophenyl)theophylline. The responses to adenosine were not blocked by tetrodotoxin, 8-cyclopentyltheophylline, 8-(3-chlorostyryl)caffeine, dipyridamole, or removal of extracellular calcium. The P2Y-selective agonist 2-methylthioadenosine triphosphate was unable to induce increases in astrocytic [Ca2+]i, whereas the P2 agonist adenosine 5'- O -(2-thiodiphosphate) induced astrocytic responses in a low percentage of astrocytes. These results indicate that the majority of hippocampal astrocytes in situ contain P1 purinergic receptors coupled to increases in [Ca2+]i, whereas a small minority appear to contain P2 purinergic receptors. Furthermore, individual hippocampal astrocytes responded to adenosine, glutamate, and depolarization with increases in [Ca2+]i. The existence of both purinergic and glutamatergic receptors on individual astrocytes in situ suggests that astrocytes in vivo are able to integrate information derived from glutamate and adenosine receptor stimulation.  相似文献

External ATP stimulated stomatal opening in epidemal strips of Commelina communis L. in darkness following brief irradiation with white or red light. ATP had no effect in darkness after irradiation with far red light or in continuous light. Mg2+ stimulated stomatal response to ATP, which was shown to be taken-up by the tissue. This implies that a Mg-stimulated ATPase, that interacts with Pfr, may be involved in stomatal opening.  相似文献

Abstract: Inorganic phosphate (Pi) plays a vital role in intracellular energy metabolism. Its many effects include stimulation of glucose use, enhancement of high-energy phosphate concentrations, and modulation of cytosolic free [Ca2+]. Cultured fetal rat cortical neurons constitutively import Pi, and cytosolic levels positively correlate with [ATP], [NADPH], and energy charge. In the present study, we demonstrate that the concentration of intracellular Pi is an important determinant of acute neuronal survival after an excitotoxic or oxidative insult to cultured fetal rat cortical neurons. Extracellular Pi dose-dependently enhanced survival of cortical neurons after exposure to NMDA at early (≤6 h) time points after termination of the insult. Pi similarly increased neuronal survival after exposure to kainic acid or H2O2. Pi-exposed neurons had higher basal intracellular [Pi], [ATP], and [GSH], and slightly lower cytosolic free [Ca2+], compared with Pi-deprived neurons. Pi-exposed neurons maintained increased [ATP] after exposure to NMDA and displayed reduced formation of reactive oxygen species after exposure to kainic acid or H2O2, compared with Pi-deprived neurons. These findings demonstrate that changes in extracellular and intracellular Pi can affect neuronal survival after excitotoxic or oxidative insults.  相似文献

Abstract. The specific effects of hypoxia and various inhibitors on stomatal opening in the light and closing in the dark were characterized in isolated epidermis from Commelina communis L. Reducing the guard cell metabolism with hypoxia and the uncoupler carbonyl cyanide-m-chloro-phenyl-hydrazone, CCCP, respectively, inhibited both stomatal opening and closing. Stomatal closing was very efficiently blocked by CCCP and this effect could be readily reversed by washing out the inhibitor. The authors were unable to inhibit stomatal opening with ATPase-inhibitors, without also affecting closing. Orthovanadate, up to 2 mol m−3, affected neither opening nor closing. Dicyclohexylcarbodiimide, DCCD, and diethylstilbestrol, DES, inhibited opening as well as closing to about 50%. The K+ -channel blocker tetraethylammonium chloride, TEA-Cl, inhibited both stomatal opening and closing, as did phenyl acetic acid, PAA, a compound considered to interfere with blue light induced stomatal opening. The results are discussed in the view that the uncontrolled K+ leakage from the guard cells is low, that K+ efflux during stomatal closing, as well as K+ influx during opening, occurs through specific K+-channels and that ATP and/or a membrane potential seems to be needed to keep these channels open.  相似文献

Abstract: The regulatory role of A2A adenosine receptors in P2 purinoceptor-mediated calcium signaling was investigated in rat pheochromocytoma (PC12) cells. When PC12 cells were treated with 2- p -(2-carboxyethyl)-phenethylamino-5'- N -ethylcarboxamidoadenosine (CGS-21680), a specific agonist of the A2A adenosine receptor, the extracellular ATP-evoked rise in cytosolic free Ca2+ concentration ([Ca2+]i) was inhibited by 20%. Both intracellular calcium release and inositol 1,4,5-trisphosphate production evoked by ATP were not affected by CGS-21680 treatment. However, ATP-evoked Ca2+ influx was inhibited following CGS-21680 stimulation. The CGS-21680-mediated inhibition occurred independently of nifedipine-induced inhibition of the [Ca2+]i rise. The CGS-21680-induced inhibition was completely blocked by reactive blue 2. The CGS-21680 effect was mimicked by forskolin and dibutyryl-cyclic AMP and blocked by Rp -adenosine 3',5'-cyclic monophosphothioate, a protein kinase A inhibitor, or by staurosporine, a general kinase inhibitor. The data suggest that in PC12 cells activation of A2A adenosine receptors leads to inhibition of P2 purinoceptor-mediated Ca2+ influx through ATP-gated cation channels and involves protein kinase A.  相似文献

Abstract. Exposure to low concentration of SO2 increases stomatal conductance in field bean by c 20%. Microscopy of epidermal strips showed that this response was associated with extensive destruction of epidermal cells adjacent to the stomata. Guard cell survival was not affected signficantly. Collapsed epidermal cells were also observed in intact leaf samples using scanning electron miscroscopy, thus confirming that the damage seen in epidermal strips was not caused by the stripping procedure. These observations are discussed in relation to the hypothesis that the increased stomatal opening induced by low SO2 concentrations results from preferential injury to the adjacent cells.  相似文献

Variation in stomatal development and physiology of mature leaves from Alnus glutinosa plants grown under reference (current ambient, 360 μmol mol−1 CO2) and double ambient (720 μmol mol−1 CO2) carbon dioxide (CO2) mole fractions is assessed in terms of relative plant growth, stomatal characters (i.e. stomatal index and density) and leaf photosynthetic characters. This is the first study to consider the effects of elevated CO2 concentration on the distribution of stomata and epidermal cells across the whole leaf and to try to ascertain the cause of intraleaf variation. In general, a doubling of the atmospheric CO2 concentration enhanced plant growth and significantly increased stomatal index. However, there was no significant change in relative stomatal density. Under elevated CO2 concentration there was a significant decrease in stomatal conductance and an increase in assimilation rate. However, no significant differences were found for the maximum rate of carboxylation ( V cmax) and the light saturated rate of electron transport ( J max) between the control and elevated CO2 treatment.  相似文献

Abstract: D1-and D2-dopamine receptors exert important physiological actions on striatal neurons, but the intracellular second messenger pathways activated by these receptors are still incompletely understood. Using primary cultures of rat striatal cells, we have examined the effects of activating D1 or D2 receptors on arachidonic acid (AA) release and cyclic AMP accumulation. In striatal neurons labeled by incubation with [3H]AA, D2-receptor stimulation enhanced release of [3H]AA produced by application of the Ca2+ ionophore A23187 or of the purinergic agonist ATP. By contrast, D1-receptor stimulation inhibited [3H]AA release. This inhibitory effect of D1 receptors was accompanied by stimulation of adenylyl cyclase activity, measured as accumulation of cyclic AMP, and was mimicked by application of the adenylyl cyclase activator forskolin. The results indicate the existence of a novel signaling pathway for D2 and D1 receptors in striatum, potentiation and inhibition, respectively, of Ca2+-evoked AA release.  相似文献

Night-time stomatal opening in C3 plants may result in significant water loss when no carbon gain is possible. The objective of this study was to determine if endogenous patterns of night-time stomatal opening, as reflected in leaf conductance, in Vicia faba are affected by photosynthetic conditions the previous day. Reducing photosynthesis with low light or low CO2 resulted in reduced night-time stomatal opening the following night, irrespective of the effects on daytime stomatal conductance. Likewise, increasing photosynthesis with enriched CO2 levels resulted in increased night-time stomatal opening the following night. Reduced night-time stomatal opening was not the result of an inability to regulate stomatal aperture as leaves with reduced night-time stomatal opening were capable of greater night-time opening when exposed to low CO2. After acclimating plants to long or short days, it was found that night-time leaf conductance was greater in plants acclimated to short days, and associated with greater leaf starch and nitrate accumulation, both of which may affect night-time guard cell osmotic potential. Direct measurement of guard cell contents during endogenous night-time stomatal opening will help identify the mechanism of the effect of daytime photosynthesis on subsequent night-time stomatal regulation.  相似文献

The stomatal response to CO2 is linked to changes in guard cell zeaxanthin*   总被引:4,自引:2,他引:2  
The mechanisms mediating CO2 sensing and light–CO2 interactions in guard cells are unknown. In growth chamber-grown Vicia faba leaves kept under constant light (500 μ mol m–2 s–1) and temperature, guard cell zeaxanthin content tracked ambient [CO2] and stomatal apertures. Increases in [CO2] from 400 to 1200 cm3 m–3 decreased zeaxanthin content from 180 to 80 mmol mol–1 Chl and decreased stomatal apertures by 7·0 μ m. Changes in zeaxanthin and aperture were reversed when [CO2] was lowered. Guard cell zeaxanthin content was linearly correlated with stomatal apertures. In the dark, the CO2-induced changes in stomatal aperture were much smaller, and guard cell zeaxanthin content did not change with chamber [CO2]. Guard cell zeaxanthin also tracked [CO2] and stomatal aperture in illuminated stomata from epidermal peels. Dithiothreitol (DTT), an inhibitor of zeaxanthin formation, eliminated CO2-induced zeaxanthin changes in guard cells from illuminated epidermal peels and reduced the stomatal CO2 response to the level observed in the dark. These data suggest that CO2-dependent changes in the zeaxanthin content of guard cells could modulate CO2-dependent changes of stomatal apertures in the light while a zeaxanthin-independent CO2 sensing mechanism would modulate the CO2 response in the dark.  相似文献

Recent studies have suggested that Ca2+/calmodulin (CaM) or CaM-like proteins may be involved in blue light (BL)-dependent proton pumping in guard cells. As the increase in cytosolic concentration of Ca2+ is required for the activation of CaM and CaM-like proteins, the origin of the Ca2+ was investigated by measuring BL-dependent proton pumping with various treatments using guard cell protoplasts (GCPs) from Vicia faba . BL-dependent proton pumping was affected neither by Ca2+ channel blockers nor by changes of Ca2+ concentration in the medium used for the GCPs. Addition of Ca2+ ionophores and an agonist to GCPs did not induce proton pumping. However, BL-dependent proton pumping was inhibited by 10 m M caffeine, which releases Ca2+ from the intracellular stores, and by 10 μ M 2,5-di-( tert -butyl)-1,4-benzohydroquinone (BHQ) and 10 μ M cyclopiazonic acid (CPA), inhibitors of Ca2+-ATPase in the sarcoplasmic and endoplasmic reticulum (ER). By contrast, the inhibitions were not observed by 10 μ M thapsigargin, an inhibitor of animal ER-type Ca2+-ATPase. The inhibitions by caffeine and BHQ were reversible. Light-dependent stomatal opening in the epidermis of Vicia was inhibited by caffeine, BHQ, and CPA. From these results, we conclude that the Ca2+ thought to be required for BL-dependent proton pumping may originate from intracellular Ca2+ stores, most likely from ER in guard cells, and that this origin of Ca2+ may generate a stimulus-specific Ca2+ signal for stomatal opening.  相似文献

The responses of individual stomata to CO2 concentrations ranging from 0 to 900 μmol mol−1 air were analysed in Ipomoea pes-caprae L. Sweet (Convolvulaceae). The stomata were directly observed using a measurement system that permitted continuous observation of stomatal movement under controlled light and CO2 conditions. A CO2 concentration of 350 μmol mol−1 or higher induced stomatal closure, whereas concentrations below 350 μmol mol−1 did not. The time lag before stomatal closure decreased with increasing CO2 concentration, as did the steady-state aperture of the stomata after a change in CO2 concentration. However, the rate of stomatal closure increased with increasing CO2 concentration. Therefore, not only the stomatal closure rate but also the time from the CO2 concentration change to the beginning of stomatal closure changed with increasing CO2 concentration. These results suggest that atmospheric CO2 may be the stimulus for the closure of guard cells. No significant differences were observed between adaxial and abaxial stomata in terms of their responses to CO2. However, heterogeneous responses were detected between neighbouring stomata on each leaf surface.  相似文献

Abstract: The role of the A2A adenosine receptor in regulating voltage-sensitive calcium channels (VSCCs) was investigated in PC12 cells. Ca2+ influx induced by membrane depolarization with 70 m M K+ could be inhibited with CGS21680, an A2A receptor-specific agonist. Both L- and N-type VSCCs were inhibited by CGS21680 treatment. Effects of adenosine receptor agonists and antagonists indicate that the typical A2A receptor mediates inhibition of VSCCs. Cholera toxin (CTX) treatment for 24 h completely eliminated the CGS21680 potency. Similar inhibitory effects on VSCCs were obtained by membrane-permeable activators of protein kinase A (PKA). These effects were blocked by Rp -adenosine-3',5'-cyclic monophosphothioate, a PKA inhibitor. The data suggest that activation of the A2A receptor leads to inhibition of VSCCs via a CTX-sensitive G protein and PKA. ATP pretreatment caused a reduction in subsequent rise in cytosolic free Ca2+ concentration induced by 70 m M K+, presumably by inactivation of VSCCs. Simultaneous treatment with ATP and CGS21680 produced significantly greater inhibition of VSCCs than treatment with CGS21680 or ATP alone. Furthermore, the CGS21680-induced inhibition of VSCCs was not affected by the presence of reactive blue 2. CGS21680 still significantly inhibited ATP-evoked Ca2+ influx without VSCC activity after cobalt or 70 m M K+ pretreatment. These data suggest that the A2A receptor-sensitive VSCCs differ from those activated by ATP treatment. Although A2A receptors induce inhibition of VSCCs as well as ATP-induced Ca2+ influx, the two inhibitory effects are clearly distinct from each other.  相似文献

Carbon dioxide and abscisic acid (ABA) are two major signals triggering stomatal closure. Their putative interaction in stomatal regulation was investigated in well-watered air-grown or double CO2-grown Arabidopsis thaliana plants, using gas exchange and epidermal strip experiments. With plants grown in normal air, a doubling of the CO2 concentration resulted in a rapid and transient drop in leaf conductance followed by recovery to the pre-treatment level after about two photoperiods. Despite the fact that plants placed in air or in double CO2 for 2 d exhibited similar levels of leaf conductance, their stomatal responses to an osmotic stress (0·16–0·24 MPa) were different. The decrease in leaf conductance in response to the osmotic stress was strongly enhanced at elevated CO2. Similarly, the drop in leaf conductance triggered by 1 μ M ABA applied at the root level was stronger at double CO2. Identical experiments were performed with plants fully grown at double CO2. Levels of leaf conductance and carbon assimilation rate measured at double CO2 were similar for air-grown and elevated CO2-grown plants. An enhanced response to ABA was still observed at high CO2 in pre-conditioned plants. It is concluded that: (i) in the absence of stress, elevated CO2 slightly affects leaf conductance in A. thaliana ; (ii) there is a strong interaction in stomatal responses to CO2 and ABA which is not modified by growth at elevated CO2.  相似文献
