Summary The release of substances from wheat roots was found to be directly related to the growth of the root system. Plants whose root system did not grow released almost no exudates.When exudate concentration in the vicinity of the roots was lowered by frequent replacements of the nutrient solution or by a simultaneous cultivation of exudate-utilizing bacteria, the release of exudates was enhanced. In axenic wheat cultures, the amount of exudates during a 12-day cultivation with 2- or 4-day intervals between medium replacements represented 50% of root dry weight and 12% of whole plant dry weight.Wheat plants cultivated in the presence of the bacteriumPseudomonas putida released up to double the amount of exudates compared with axenic variants.  相似文献

Summary Comparisons have been made of the amount and composition of seed and seedling exudates in barley, wheat, cucumber, and bean. Except in the case of wheat a greater proportion of the total nitrogen content in the seed exudates was formed by protein and peptide nitrogen than by nitrogen of free amino acids. In contrast, the greater part of the total nitrogen in the exudates of seedlings was formed by free amino acid nitrogen, except in the case of barley. Peptides represented 8 to 26 per cent of the protein and peptide fraction in seed exudates, the highest amount being found in bean. On an equal weight basis, the spectrum of amino acids released from seeds and seedlings differed little between barley, wheat and cucumber; greater differences were observed in bean exudates. Seedlings exuded reducing substances to a considerably greater extent than seeds. The spectrum of reducing sugars in seed and root exudates differed greatly, especially in the case of keto sugars. Differences in the organic acid spectra were small, except for bean plants, the seedling exudate of which contained more organic acids with a richer spectrum than seed exudate. Both seeds and seedlings of cucumber exuded a small quantity and a poor spectrum of organic acids.  相似文献

A genomic clone of a pea pectinmethylesterase encoding gene, rcpme1, was isolated; the promoter region was found to include regions of homology to phenylalanine ammonia lyase (PAL) and nodulin gene promoters. Agrobacterium rhizogenes mediated hairy roots were used for rcpme1 expression and functional analysis in pea. Patterns of rcpme1 expression in cultured hairy roots, measured using uidA encoding -glucuronidase (GUS) as a reporter gene, were distinct from patterns which occur in normal pea roots. No reporter gene expression occurred in transgenic Arabidopsis thaliana, whose roots do not produce border cells. Border cell number from transgenic hairy roots expressing rcpme1 anti-sense mRNA under the control of its 2.75 kb 5 flanking sequence was reduced by > 50%. Nodulation genes of Rhizobium leguminosarum were used as a marker to document that roots with reduced production of border cells and other root cap exudates have a corresponding reduction in levels of biologically active signal molecules. Direct measurements were used to confirm that most of the exudate harvested from young, unwounded roots of normal pea plants is derived from the root tip region where rcpme1 is expressed. The potential application of the rcpme1 gene as a molecular marker for root exudate production is discussed.  相似文献

Summary The effect of temperature on the exudation of sugars and amino acids from germinating seeds of maize and cucumber and the effect of the so called cold shock on root exudation of plant seedling was studied. After the first forty-eight hours the exudation of these compounds from germinating seeds generally rises in proportion to the temperature. However, certain relative changes were observed in the composition of the exudates. Exudation of maltose and fructose with arabinose, for example is lower at 28°C than at 19°C.When plants previously growing under favourable temperature conditions were exposed for three days to a lower temperature, there was a several times higher exudation from the roots of maize and a marked rise in exudation from roots of cucumber in comparison with control plants. In the exudates of maize affected by cold shock 3 new oligosaccharides and fructose with saccharose in addition to previously detected substances were found.  相似文献

Maize (Zea mays L. cv. Alize) plants were grown in a calcareous soil in pots divided by 30-m nylon nets into three compartments, the central one for root growth and the outer ones for hyphal growth. Sterle soil was inoculated with either (1) rhizosphere microorganisms other than vesicular-arbuscular mycorrhizal (VAM) fungi, (2) rhizosphere microorganisms together with a VAM fungus [Glomus mosseae (Nicol. and Gerd.) Gerdemann and Trappel], or (3) with a gamma-irradiated inoculum as control. Plants were grown under controlled-climate conditions and harvested after 3 or 6 weeks. VAM plants had higher shootroot ratios than non-VAM plants. After 6 weeks, the concentrations of P, Zn and Cu in roots and shoots had significantly increased with VAM colonization, whereas Mn concentrations had significantly decreased. Root exudates were collected on agar sheets placed on the interface between root and hyphal compartments. Six-week-old VAM and non-VAM plants had similar root exudate compositions of 72–73% reducing sugars, 17–18% phenolics, 7% organic acids and 3% amino acids. In another experiment in which root exudates were collected on agar sheets with or without antibiotics, the amounts of amino acids and carbohydrates recovered were similar in VAM and non-VAM plants. However, threeto sixfold higher amounts of carbohydrates, amino acids and phenolics were recovered when antibiotics were added to the agar sheets. Thus, the high microbial activity in the rhizosphere and on the rhizoplane limits the exudates recovered from roots.  相似文献

Summary Amino acids in exudates of uninoculated pea roots were compared quantitatively and qualitatively with exudates of roots inoculated with Gliocladium catenulatum. This fungus has the potential of causing severe root necrosis. Twenty-one amino acids were found in exudates of healthy roots and apparently some of these were utilized by the fungus. A relatively high concentration of ammonia was detected in exudates of inoculated pea roots, indicating an intense deamination by the fungus. No other imbalance in amino acids was found which could be related to known toxic effects of amino acids on plant tissues.  相似文献

This study was conducted to determine if plants can assimilate indole-3-acetic acid (IAA) from rooting media and if exogenous L-tryptophan (L-TRP) can be assimilated and converted by plants into auxins. The addition of 2-14C-IAA (3.7 kBq plant-1) to wheat (Triticum aestivum L.) seedlings of three varieties grown in nutrient solution resulted in the uptake (avg.=7.6%) of labelled IAA. Most of the label IAA was recovered in the shoot (avg.=7.2%) with little accumulation in the root (avg.=0.43%). A portion of the assimilated IAA-label in the plant was identified by co-chromatography and UV spectral confirmation as IAA-glycine and IAA-aspartic acid conjugates. Little of the assimilated IAA label was found as free IAA in the wheat plants. These same assimilation patterns were observed when 2-14C-IAA was added to wheat plants grown in sterile and nonsterile soil. In contrast, the wheat varieties assimilated considerably less (avg.=1.3%) of the added microbial IAA precursor, 3-14C-L-TRP (3.7 kBq plant-1) and thus much lower amounts of IAA conjugates were detected. Glasshouse soil experiments revealed that 2 out of 3 wheat varieties had increased growth rates and increased yields when L-TRP (10-5 and 10-7 M) was added to the root zone. It is surmised that this positive response is a result of microbial auxin production within the rhizosphere upon the addition of the precursor, L-TRP. The amino acid composition of the root exudates plays a critical role in microbial production of auxins in the rhizosphere. This study showed that wheat roots can assimilate IAA from their rooting media, which will supplement the endogenous IAA levels in the shoot tissue and may positively influence plant growth and subsequent yield.  相似文献

Summary The effects of root herbivory by larvae of the scarabaeid, Phyllopertha horticola, on the growth of Capsella bursa-pastoris were examined. Individuals of Aphis fabae were reared on the leaves to determine what effect, if any, root feeding has on the performance of this insect. The experiment was conducted under two watering regimes (low and high). Low watering and root feeding caused water stress in the plants and this was reflected in a reduction in vegetative biomass and an increase in the proportion of material allocated to reproduction. Supplying plants with ample water in the high treatment enabled the water stress caused by root herbivory to be offset, but not completely overcome. Low watering and root feeding caused an increase in aphid weight and growth rate, while root feeding also increased fecundity and adult longevity. These effects are attributed to an improvement in food quality, measured by total soluble nitrogen, and caused by amino acid mobilization due to the water stress. The implications of these results in agricultural and ecological situations are discussed.  相似文献

The changes in the contents of protein and free amino acids in pea plants inoculated with Rhizobium leguminosarum were studied taking into account the susceptibility of roots to root nodule bacteria. The content of cytoplasmic protein during infection increased in the actively growing root zone (0–5 mm) and decreased in the root zones susceptible to rhizobia (5–20 mm from the root tip). The quantitative composition of free amino acids changed essentially upon inoculation of pea seedlings with R. leguminosarum.  相似文献

The localization of enzymes of assimilatory sulfate reduction was examined in roots of 5-d-old pea (Pisum sativum L.) seedlings. During an 8-h period, roots of intact plants incorporated more label from 35SO 4 2- in the nutrient solution into the amino-acid and protein fractions than shoots. Excised roots and roots of intact plants assimilated comparable amounts of radioactivity from 35SO 4 2- into the amino-acid and protein fractions during a 1-h period, demonstrating that roots of pea seedlings at this stage of development were not completely dependent on the shoots for reduced sulfur compounds. Indeed, these roots contained activities of ATP-sulfurylase (EC 2.7.7.4), adenosine 5-phosphosulfate sulfotransferase, sulfite reductase (EC 1.8.7.1) and O-acetyl-l-serine sulfhydrylase (EC 4.2.99.8) at levels of 50, 30, 120 and 100%, respectively, of that in shoots. Most of the extractable activity of adenosine 5-phosphosulfate sulfotransferase was detected in the first centimeter of the root tip. Using sucrose density gradients for organelle separation from this part of the root showed that almost 40% of the activity of ATP-sulfurylase, adenosine 5-phosphosulfate sulfotransferase and sulfite reductase banded with the marker enzyme for proplastids, whereas only approximately 7% of O-acetyl-l-serine sulfhydrylase activity was detected in these fractions. Because their distributions on the gradients were very similar to that of nitrite reductase, a proplastid enzyme, it is concluded that ATP-sulfurylase, adenosine 5-phosphosulfate sulfotransferase and sulfite reductase are also exclusively or almost exclusively localized in the proplastids of pea roots. Exudation of carboxylates represents one the most efficient strategies used by P-starved white lupin (Lupinus albus L.) to acquire phosphorus from sparingly soluble sources. This exudation occurs through proteoid root clusters, with citrate being the predominant organic acid released. The occasional detection of malate in whole root exudates suggests that this acid would also be released, but from tissues other than root clusters. To investigate the spatial and temporal pattern of exudation, citrate and malate exudation and concentration were measured in whole roots and root sections of white lupin, from seedling emergence to plant senescence due to P starvation. Both organic acids were detected in whole root exudates of P-stressed plants, and they were released at similar rates throughout the experiment. Malate was predominantly exuded from apices of both seedling taproots and proteoid roots, whereas citrate exudation was restricted to proteoid root clusters. Studies directed to address the association between carboxylate exudation and concentration in proteoid root clusters showed a non-linear response for citrate, within the range of 7 to 23 mol g–1 fresh weight. This association was further assessed by altering citrate concentration in the whole root. Adding P to 24-day-old P-starved plants reduced citrate concentration and exudation to the level of the control P-fed plants, demonstrating that citrate exudation and concentration are associated. Malate exudation and concentration did not correlate significantly. Results indicate that citrate release by P-starved white lupin would occur whenever a certain threshold of citrate concentration is attained, and that the sites, the rates and the span of transient exudation depend on the physiological age of the tissue.  相似文献

Summary The influence of decapitation and treatment with IAA and/or kinetin on the pattern of distribution of 14C-labelled sucrose applied to the third leaf of 14-day old dwarf pea seedlings was investigated. Decapitation resulted in a diversion of the labelled metabolites to the lateral buds, and greatly increased the radioactivity present in the root system indicating that in these seedlings the roots and apex actively competed for translocates from the third leaf. Application of IAA to the decapitated internode prevented the growth of the lateral buds for the duration of the experiment and restored the pattern of distribution of labelled metabolites found in the intact plant. Application of kinetin alone resulted in a marked accumulation of labelled materials in the lateral buds, but when kinetin was applied with IAA metabolites were once again diverted from the lateral buds to the treated internode. Neither of these treatments had any influence on the proportion of the translocated materials which accumulated in the root system when compared with intact plants. The results are discussed in relation to current concepts of hormone-directed transport of nutrients in plants.  相似文献

We have found that Rhizobium meliloti strain L5.30 exhibits positive chemotaxis towards some amino acids, sugars, exudates and extracts from roots of legume plants. From the investigated compounds sugars were better chemo-attractants than amino acids, but legume root substances were the best ones. Positive chemotaxis towards legume root compounds was supported by clouds of R. meliloti cells observed at the surface of alfalfa roots. A large deletion, in the nodABC region of the symbiotic megaplasmid (Sym), did not eliminate rhizobial chemotaxis.  相似文献

Excised wheat (Triticum aestivum L.) and field pea (Pisum arvense L.) roots, incubated under anaerobic conditions or in the presence of uncouplers of oxidative phosphorylation [2,4-dinitrophenol (DNP), carbonylcyanide-m-chlorophenylhydrazone, pentachlorophenol] accumulated nitrite as a result of an inhibition of nitrite reduction. In isolated root plastids, nitrite reduction was dependent on a supply of glucose-6-phosphate (G6P) and did not require ATP. The estimated Km value for glucose 6-phosphate was 1.25 mM. Glucose and fructose-1,6-diphosphate were ineffective substrates for nitrate reduction. Anaerobic conditions and treatment with DNP, which would result in a cessation of ATP production by the mitochondria and a stimulation of glycolysis via the Pasteur effect, were shown to decrease the G6P content of excised roots of wheat and pea. A negative correlation was observed between the level of G6P and nitrite accumulation on root tissues. It is proposed that an interruption in the supply of G6P to the root plastid under these conditions would result in an inhibition of nitrite reduction leading to nitrite accumulation.Abbreviation G6P glucose-6-phosphate  相似文献

Background and aims

Production of indole-3-acetic acid (IAA) by Azospirillum brasilense is one of the most important mechanisms underlying the beneficial effects observed in plants after inoculation with this bacterium. This study determined the contribution of the hisC1 gene, which encodes aromatic amino acid aminotransferase-1 (AAT1), to IAA production, and analyzed its expression in the free-living state and in association with the roots of wheat.

Methods

We determined production of IAA and AAT activity in the mutant hisC::gusA-sm R . To study the expression of hisC1, a chromosomal gene fusion was analyzed by following β-glucuronidase (GUS) activity in vitro, in the presence of root exudates, and in association with roots.

Results

IAA production in the hisC mutant was not reduced significantly compared to the activity of the wild-type strain. AAT1 activity was reduced by 50% when tyrosine was used as the amino acid donor, whereas there was a 30% reduction when tryptophan was used, compared to the activity of the wild-type strain. Expression of the fusion protein was up-regulated in both logarithmic and stationary phases by several compounds, including IAA, tryptophan, tyrosine, and phenyl acetic acid. We observed the expression of hisC1 in bacteria associated with wheat roots. Root exudates of wheat and maize were able to stimulate hisC1 expression.

Conclusions

The expression data indicate that hisC1 is under a positive feedback control in the presence of root exudates and on plants, suggesting that AAT1 activity plays a role in Azospirillum–plant interactions.  相似文献

Experimental measurements of phosphorus (P) uptake and the forms of soil P depleted from an Ultisol by 6 upland rice cultivars are reported. In both P-fertilized and-unfertilized soil, the majority of P taken up was solubilized from a 0.1 M NaOH-soluble pool by root-induced changes. The soil pH within 4 mm of the roots was lowered by up to 0.5 units (from 4.6), but this by itself could not account for the P solubilized, and nor could increased phosphatase activity near the roots. The possible role of root-released low molecular weight organic acid anions in P solubilization is discussed. No significant differences in the extent of solubilization by a given root mass could be detected between cultivars. In P-unfertilized soil, but not in P-fertilized soil, there were significant differences between cultivars in internal P efficiency as measured by shoot dry weight per unit total plant P. In unfertilized soil, root growth and P uptake were strongly correlated with the P content of the seeds from which the plants were grown.  相似文献

The exudation, composition, and biological activity of the phenolic compounds (PC) of pea (Pisum sativum L.) roots in the light and darkness were studied. The roots of leguminous plants grown for 5 days in darkness exuded a smaller amount of PC that displayed a weaker stimulation of Rhizobium reproduction. Moreover, the root exudates contained antimicrobial compounds, stilbenes. It is assumed that a lower PC exudation by roots and the specific features of PC composition influencing the biological activity are among the reasons causing a delayed nodulation of legumes grown in darkness.  相似文献

Contact between roots and Fe-containing solid substrate is known to facilitate acquisition of iron by plants, but the actual mechanism of this contact effect is not yet clear. This study was undertaken to evaluate the effect of root contact with ballotini (glass spheres) on exudation of substances capable of reducing or chelating insoluble Fe(III) compounds by the roots of barley (Hordeum vulgare L. cv. Europa) seedlings. Seedlings with roots encountering mechanical impedance (i.e., in contact with ballotini) produced more lateral roots than the seedlings with unimpeded (i.e., freely suspended) roots in the nutrient solution. Nutrient solution bathing roots in contact with ballotini showed higher concentrations of Fe(III)-chelating (83% on day 7) and Fe(III)-reducing (107% on day 12) substances than solutions bathing unimpeded roots. The pH of all solutions rose continuously during the course of the experiment but was always lower (by a nonsignificant degree) in the solutions with roots in contact with ballotini than in those with unimpeded roots. The data indicate that under natural soil conditions the amount of Fe-chelating and Fe-reducing root exudates may be higher than is usually measured from roots of terrestrial plants artificially suspended in nutrient solution.  相似文献
