Manometric studies were conducted on Pterocladia capillacea, Gelidium latifolium and Gelidium spinulosum from the Azores, Portugal to determine optimal values of temperature, light and salinity for growth. Physiological responses were considered in relation to vertical distribution patterns of these species commonly observed throughout the Azores. Optimal parameters for the growth of Pterocladia capillacea, Gelidium latifolium and G. spinulosum were 17 to 25 °C, a photon flux density between 200 and 300 µmol m–2 s–1 and salinities of 25 to 35.  相似文献

Juvenile Corbicula fluminea Müller (1774) were cultured at 15.3 °C in the laboratory on eight combinations of suspended sediment and phytoplankton. Sediment concentrations were 2.6, 25, 50, and 150 mg l–1. Chlorophyll a levels were 15.6 and 62.5 µg l–1. Clam tissue growth was found to be independent of silt concentration but increased at the higher chlorophyll level (p < 0.05). The growth experiment was repeated at 24 °C with chlorophyll a concentrations of 18.9 and 112.6 µg l–1. Growth was again greater at the higher phytoplankton level (p < 0.05). These results demonstrate that Asiatic clam populations are food-limited most of the growing season in the Northern and Western portions of California's eutrophic Sacramento-San Joaquin Delta where chlorophyll a levels average less than the lower of these values. Comparisons of clam growth in the laboratory and estuary support the food limitation hypothesis as at the higher food concentration laboratory tissue growth was 2.3 and 3.8 times greater during the high and low temperature evaluations than in the estuary.  相似文献

Cultivar E23, an F1 hybrid of P. edulis and P. edulis f. flavicarpa is usually propagated by shoot-tip grafting. Various media were tested to evaluate the potential of E23 for in vitro propagation. Adult tissue was difficult to culture and did not respond to media containing low (<10 µM) concentrations of growth regulators. Growth of adult buds on intact stem sections was promoted by 1 week of dark incubation on MS basal medium plus 150 µM 2iP, 200 µM adenine sulphate and 17.1 µM IAA (3 mg l–1), and further developed into shoots on MS medium plus 4.9 µM 2iP (1 mg l–1) and 5.7 µM IAA (1 mg l–1). By contrast, juvenile shoots of E23, and Passiflora species: edulis f. flavicarpa, edulis, alata, caerulea, mollissima, coccinea, herbertiana and suberosa grew rapidly on MS medium plus 10 µM kinetin and 5 µM IAA. Single-nodal cuttings of Solanum tuberosum (four cultivars) and Solanum chacoense were induced to produce in vitro microtubers on Murashige & Skoog (MS) medium supplemented with 8 g l–1 sucrose and various concentrations of kinetin and paclobutrazol. The cultures were kept 10 days in darkness and then transferred to a 14 h daylength with 100 µE m–2 sec–1 light intensity at 21 °C. Kinetin (2.5 mg l–1) had no significant influence on tuber formation. However, its addition together with paclobutrazol (0.001 mg l–1) significantly enhanced tuberization. Paclobutrazol alone stimulated early tuber initiation and inhibited stem growth. Despite some genotype × treatment interactions, all genotypes (from very early to late and wild type) formed the maximum proportion of explants bearing microtubers on the media containing both plant growth regulators.  相似文献

Food limitation was tested in the laboratory by individual growth and reproduction of two cladoceran species, Ceriodaphnia richardi and Daphnia gessneri, from the shallow tropical Brazilian Lake Monte Alegre. The cladocerans were fed cultivated green alga Scenedesmus spinosus in concentrations of 0.20, 0.10, 0.05, and 0.025 mg C l−1. Higher biomass and growth rates occurred in the two highest-food concentrations; the two lowest ones negatively affected clutch size and first reproduction. The threshold food concentration is lower than 0.025 mg C l−1 and the incipient limiting level is a value between 0.10 and 0.20 mg C l−1. The largest species, D. gessneri, was more sensitive to low food concentrations. The effects of low and high temperatures (19 and 27°C) were evaluated by life table experiments with three cladocerans from the lake—Daphnia ambigua, D. gessneri, and Moina micrura—with no food limitation (1 mg C l−1 of S. spinosus). Higher population growth rates for the three species were found at 27°C; better performance in most life table parameters was observed for the former two species at the highest temperature, D. gessneri being the most sensitive to the lowest temperature. There are indications that temperature is an important abiotic factor that constrains populations of cladocerans for a short period in winter in the lake, when temperature decreases to 18–19°C. However, its influence cannot be separated from a biotic factor such as food, whose effect is stronger in the cool season, when concentrations are lower and contribution of inedible algae is relatively higher.  相似文献

Pichia anomala, isolated from dried flower buds of Woodfordia fruticosa, produced a high activity of an intracellular phytase, at 68 U per g dry biomass, when grown at 20 °C for 24 h in a medium containing glucose (40 g l–1) and beef extract (10 g l–1) supplemented with Fe2+ (0.15 mM). Partially purified phytase was optimally active at 60 °C and pH 4 with a half life of 7 days at 60 °C. It retained 85% of its activity at 80 °C for 15 min. The enzyme is suitable for supplementing animal feeds to improve the availability of phosphate from phytate.  相似文献

A thermotolerant methylotrophicBacillus sp. (KISRI TM1A, NCIMB 40040), isolated from the Kuwaiti environment and belonging to the group II spore-forming, bacilli, could not be correlated with any knownBacillus sp. It may, therefore, be a new species. It grew at temperatures from 37° to 58°C from pH 6.5 to 9.0 and on methanol up to 40 g l–1. It grew well in a chemostat. Its biomass yield coefficient was improved by about 30% by optimization of medium and growth conditions, reaching a maximum of 0.44g g–1 at 45°C pH 6.8 to 7.0, dilution rate 0.25 h–1 with methanol at 10 g l–1. Average crude protein and amino acid content were 84% and 60%, respectively, and maximum productivity attained under laboratory conditions was 5.06 g l–1h–1. It was concluded that this strain has good potential for use in single-cell protein production.  相似文献

In an attempt to assess bacterioplankton production and growth yieldunder low temperature conditions and to compare bacterioplankton withphytoplankton production in the ice-covered water column of the shallowNeusiedler See, outdoor measurements under near in situ conditions wereperformed during the winter of 1995/96. During the investigation period,mean chlorophyll (Chl) a concentration was 21.03 ± 14.95 µg Chla l-1. Phytoplankton primary production integrated over thewater column ranged from 1.35 to production integrated over the water columnranged from 1.35 to 4.23 mg C m-2 d-1 (mean± SD = 2.46 ± 1.06 mg C m-2d-1). Bacterial abundance varied from 20 to 40×105 ml-1 for most of the investigationperiod and increased by the end of March concomitantly with the increase intemperature from 1.3 to 6.3 °C within 5 days. Mean bacterial productionwas 15.3 ± 12.8 µg C l-1 d-1(range: 3.0 to 41.7 µg C l-1 d-1) and meanbacterial growth rate 0.23 ± 0.16 d-1 following closelythe pattern in bacterial production. DOC concentration declined linearlyfrom 20.7 mg C l-1 to 16.45 mg C l-1 over the 4months period of ice cover. The contribution of humic substances to thetotal DOC pool declined from 43.6% at the end of November to37.3% at the end of March. Calculated on an area basis, phytoplanktonproduction amounted to only 16% of bacterial production which makesit unlikely that phytoplankton supply substrate for bacterioplankton growthin significant quantities when the lake is ice covered. From the observeddecline in DOC over the investigation period and assuming only negligibleinput of DOC from other sources we calculated an average DOC uptake by thebacterioplankton community of 47.5 µg C l-1d-1 resulting in a bacterial growth efficiency of 15.9%for the ice covered conditions. Based on the growth efficiency we estimatethat pelagic primary production amounts to 2.8% of the bacterialcarbon demand. This might indicate that the bacterioplankton in NeusiedlerSee sustain their high growth rates at low temperatures (<2°C formost of the investigation period) by using probably the DOC originating fromthe previous season. This DOM stems most likely from the decay of the reedPhragmites australis and its epiphytes and, probably of minor importance,from phytoplankton leachates.  相似文献

Two strains ofLeptospirillum-like bacteria, L6 and L8, have been isolated from a mixed inoculum, also containingThiobacillus ferrooxidans andT. thiooxidans, cultured for one year with a colbaltiferous pyrite as energy substrate in a 100 I continuous bioleaching laboratory unit. Several physiological properties of the strains are described. The vibrio-shaped microorganisms grew at pH values lower than 1.3. Their growth rate was maximum between 2.5 and 8.0 g l1 ferrous iron. The optimal growth temperature was 37.5° C. Ferric iron had a stimulative effect on bacterial development up to 8 g l–1, and growth was as rapid at 14 g l–1 ferric iron as at 8 g l–1. The negative influence of cobalt on the final cell concentration was observed at 0.5 g l–1, but the growth rate was not affected up to 2 g l–1. The G + C content of strains L8 is 55.6 mol%.  相似文献

Sorghum [Sorghum bicolor (L.) Moench] plants were grown in growth chambers at 20, 25 and 30°C in a low P Typic Argiudoll (3.65 µg P g–1 soil, pH 8.3) inoculated with Glomus fasciculatum, Glomus intraradices, and Glomus macrocarpum to determine effects of vesicular-arbuscular mycorrhizal fungi (VAMF) species on plant growth and mineral nutrient uptake. Sorghum root colonization by VAMF and plant responses to Glomus species were temperature dependent. G. macrocarpum colonized sorghum roots best and enhanced plant growth and mineral uptake considerably more than the other VAMF species, especially at 30°C. G. fasciculatum enhanced shoot growth at 20 and 25°C, and mineral uptake only at 20°C. G. intraradices depressed shoot growth and mineral uptake at 30°C. G. macrocarpum enhanced shoot P, K, and Zn at all temperatures, and Fe at 25 and 30°C above that which could be accounted for by increased biomass. Sorghum plant growth responses to colonization by VAMF species may need to be evaluated at different temperatures to optimize beneficial effects.  相似文献

The influence of two seston fractions, < 20 µm (nanoplankton) and 20 µm (microplankton), on growth and reproduction of cladoceran species with different sizes, from Lake Monte Alegre, was evaluated through individual growth and life table experiments. Size, shape and other features of the algae in the fractions were described. The procedure of filtering lake water through a 20 µm net for seston fractionation caused mutual contamination. The smallest cladoceran species, Ceriodaphnia cornuta Sars and Moina micrura Kurz, produced larger clutch sizes and exhibited higher intrinsic rates of population growth (r) in the nanoplankton, despite contamination of their food by inedible algae. The largest species, Simocephalus mixtus Sars, produced larger clutch sizes in the microplankton. There were no differences in juvenile biomass growth between treatments for M. micrura and Daphnia gessneri Herbst, but lower value of the exponential growth rate (g) in the microplankton was significant for M. micrura. Fecundity (eggs/total female) of M. micrura was significantly lower in the microplankton, while D. gessneri did not reproduce in this fraction, at the end of growth experiments. Spines, colonies, cenobium, filaments, hard cell wall, and gelatinous sheaths, represented constraints to cladoceran reproductive performance, besides algae size. Microplankton contamination by nanoplanktonic algae, in the experiments, probably minimized the negative effect of inedible algae. Nanoplankton was more suitable for the smallest species and microplankton for the largest one.  相似文献

Three micro algae, Ankistrodesmus convolutus, Scenedesmus incrassatulus and Chlorella vulgaris, at three concentrations, were tested as diets for Moina macrocopa. Their effect on reproduction, fecundity, growth and survival was evaluated. All three algae satisfied the nutritional requirements of M. macrocopa, despite their difference in size. The best concentration, expressed in dry weight, was 5 mg l–1 for all three. Time to first reproduction was 4 days with all diets. Average time between clutches was 33 hours, and maximum number of clutches was 12; the highest average number of offspring per brood was 27. The largest broods were the fourth to sixth, depending on the diet. Ephippium hatching depends on temperature, taking only 24 hours at 30 °C and 48 hours at 27 °C. This species could be an important test organism in aquatic bioassays, and a live food in aquaculture.  相似文献

The effects of reduced sucrose concentrations and low temperature on a collection of coffee microcuttings have been examined. Sucrose concentrations of 0.5 g l-1 and 20 g l-1 and temperatures of 20°C and 27°C were compared in three accessions: the Arabusta (interspecific hybrid) and Coffea arabica L. cv. Caturra amarillo and cv. Mokka de Tahiti. After six months, low sucrose concentrations reduced microcutting growth, rooting and survival rate. At 20°C, microcutting growth was also reduced, but leaf loss and survival rate were promoted. The genotypic differences at six months were minor. After one year without subculture, survival rate was influenced by sucrose concentration and by genotype. All five novel, thermotolerant, alcohol-producing Kluyveromyces marxianus yeast strains, capable of growth at 52°C, grew aerobically on lactose (growth rates of 0.43 to 0.5 h-1), whey permeate (0.38 to 0.43 h-1), cellobiose (0.18 to 0.3 h-1) and xylose (0.18 to 0.26 h-1). They also grew anaerobically at 40 (0.18 to 0.19 h-1) and 45°C (0.09 to 0.14 h-1). All fermented glucose to ethanol and tolerated ethanol at 95 g l-1 at 45°C. A micropropagation system for Annona squamosa L. (Sugar Apple) using hypocotyls of seedlings and nodal cuttings from 3-year-old plants was developed. Shoot proliferation was achieved with Woody Plant Medium supplemented with BA. Silver thiosulphate was added at 0.5 mg l–1 to control leaf abscission. Rooting was obtained when subcultured shoots were preconditioned for 2 weeks in medium with 10 g l–1 activated charcoal before treatment with 43 µm NAA or 39 µm IBA. Rooting was improved when galactose was used instead of sucrose in the rooting medium. Evidence for the influence of food type and heavy metals on shell growth and fertility is presented for a freshwater population of the snail P. jenkinsi. When fed an excess of lettuce or lamb heart (protein source), growth rates were higher for lettuce. Highest growth rates occurred at a diet of lettuce plus lamb heart. Fertility was favoured by a diet of lamb heart. When fed an excess of lettuce, the EC50 growth values were 16 µg Cd l–1, 13 µg Cu l–1, and 103 µg Zn l–1 in lake water; snail fertility was inhibited at 25 µg Cd l–1 and 30 µg Cu l–1. A diet of lake detritus spiked with Cd or Cu resulted in a decrease of approximately 50% in growth rates, when compared with growth on non-spiked detritus. Spiked detritus lost metals into lake water. Food type positively interacted with metal stress, both for growth rate and fertility. The assessment of inhibitory effects of detritus contaminated either in the field or, notably, by spiking, and serving as food source for deposit feeders is hampered by sampling problems in the field and by redistribution processes of pollutants between particles and water in laboratory-scale experiments.  相似文献

Laminaria abyssalis occurs in deep water in tropical latitudes of the Brazilian coast (19° 23 S, 38° 28 W to 22° 54 S, 42° 13 09 W). Its life cycle has been completed in the laboratory in seven months using different conditions of light and temperature. The gametophytic stage required for growth the low photon flux density of 1.2 ± 0.3 µmol m–2 s–1 and 18 °C, while the juvenile and adult sporophytes needed 15 µmol m–2 s–1 and 18 °C. The sporophytes became fertile at 23 °C. Our results showed that light and temperature are the main factors regulating the growth and life history of this species under the culture conditions tested.  相似文献

The impact of ethanol and temperature on the dynamic behaviour of Saccharomyces cerevisiae in ethanol biofuel production was studied using an isothermal fed-batch process at five different temperatures. Fermentation parameters and kinetics were quantified. The best performances were found at 30 and 33°C around 120 g l-1 ethanol produced in 30 h with a slight benefit for growth at 30°C and for ethanol production at 33°C. Glycerol formation, enhanced with increasing temperatures, was coupled with growth for all fermentations; whereas, a decoupling phenomenon occurred at 36 and 39°C pointing out a possible role of glycerol in yeast thermal protection.  相似文献

The red alga Stictosiphonia hookeri is epilithic in shaded habitats of the upper intertidal zone from 30 to 55° S. Thalli of this species from Argentina, Chile, South Africa and Australia, usually without reproductive structures when collected, all developed tetrasporangia in culture. Although good vegetative growth occurred in all nine isolates at 20–25 °C, 12:12 light: dark cycle, 10–30 µmol photons m–2 s–1, none reproduced in these conditions except one isolate from Australia. At 15 °C the four South African (34 °S) isolates developed tetrasporangial stichidia, and three completed a Polysiphonia-type life history. Gametophytes were unisexual or bisexual. At 15 °C one isolate from Chile (36 °S) formed tetrasporangia, but sporelings were not viable. At 10 °C isolates from Argentina and Chile (53 °S and 54 °S) formed tetrasporangia; however, only the Chile isolate completed a Polysiphonia-type life history with unisexual gametophytes. The temperature required to induce sporogenesis correlates with the range of water and air temperatures in the natural habitats of each isolate. In irradiances >50 µmol m–2 s–1 the thalli became yellow- brown within two weeks because of phycobiliprotein loss, but this did not impair growth or reproduction. The Argentina and Chile isolates were resistant to freezing in seawater for at least two days, showing no cell damage. The protein cuticle of the outer cell wall is repeatedly shed in culture. This may serve to minimize the attachment of epiphytes in the field.  相似文献
