Cold-stored plants of strawberry cultivars Tamella, Cambridge Favourite and Redgauntlet were more susceptible to pathogenic isolates of Phytophthora cactorum than similar plants which had not been cold-stored. Indigenous nonpathogenic isolates of P. cactorum did not cause crown rot in cold-stored plants, although a small number of symptomless latent infections occurred. The majority of P. cactorum isolates causing crown rot symptoms were taken from infected strawberry crowns, although two isolates from gooseberry plants, but of uncertain origin, were also pathogenic. Outbreaks of crown rot in areas with no previous history of the disease therefore probably result from the importation of non-indigenous inoculum with planting material. Assessments of the timing of infection in relation to cold storage revealed that a high incidence of death in the cold store and chronic wilt symptoms on planting from the store resulted from initiating symptomless infections prior to cold storage. However, infection during the period immediately after cold storage resulted in rapid wilt symptoms of Phytophthora crown rot. When plated in sterile distilled water for 24 h, pieces of tissue from infected plants which had died during cold storage produced large numbers of sporangia and zoospores. This indicates that such plant material could provide a potent source of inoculum for infections in the post storage thawing environment. It is proposed that a combination of heightened host susceptibility resulting from cold storage and the presence of scatted latent infections or infected debris among the plants could result in a sudden, large scale appearance of crown rot, as sometimes is seen with cold-stored plantings of strawberries.  相似文献

Four methods were evaluated in measuring resistance of strawberry cultivars to crown rot caused by Phytophthora cactorum. Meristem propagated plants grown in vitro were inoculated with mycelial discs. Four to five days after inoculation, it was possible to distinguish between cultivars with large differences in susceptibility to the disease. Ten days later, all plants were totally necrotic making it impossible to distinguish between cultivars. When detached leaves were inoculated by inoculating a plug of mycelium into the petiole, disease symptoms developed more slowly in resistant cultivars, but leaf age greatly affected the rate of symptom development. When plug plants (not cold stored) were lightly wounded in the rhizome with a scalpel and inoculated with either zoospores or mycelium, differences in disease development between cultivars were mainly as would be expected from previous information on susceptibility, but both age and size of plants influenced the rate of disease development. Unwounded, inoculated plants did not develop symptoms. When cold‐stored plug plants were either unwounded or lightly wounded with a scalpel in the rhizome and inoculated with zoospores, the relative rates of disease development consistently reflected the susceptibility to crown rot. At the time of final assessment, disease was much more severe in wounded plants, but the relative susceptibility of cultivars was not affected by the wounding.  相似文献

Strawberry tissues infected with Phytophthora cactorum were comminuted and plated in a selective antibiotic agar medium to determine levels of tissue colonisation as indicated by the number of colony forming units (CFU) recovered per gramme of infected tissue. The number of CFU recovered per gramme of tissue increased logarithmically with the amount of necrosis in infected crown, leaf and petiole tissues. Under the conditions of enhanced susceptibility to infection and colonisation caused by cold storage treatments, this relationship between colonisation and necrosis was not significantly altered in the susceptible cv. Tamella. A recovery index was used to determine the effect of infected tissues on the recovery of CFU. This indicated that increasing levels of host colonisation stimulated CFU recovery and may partly explain the large increase in CFU g-1 with larger amounts of necrosis. The amount of tissue colonisation was greater in inoculated plants of the susceptible cv. Tamella than in less susceptible cv. Cambridge Favourite, although the necrotic tissues of the latter contained more CFU g-1, indicating a greater level of tolerance to colonisation. In cv. Tamella small amounts of colonisation were capable of causing wilt symptoms, although no wilted plants contained less than 200 CFU g-1. Conversely, plants containing more than 1000 CFU g-1 always wilted. In the early stages of infection, low levels of colonisation could be detected in strawberry crowns in the absence of symptoms. Dormant strawberry plants of cv. Tamella were readily infected by P. cactorum zoospore inoculations but, unlike actively growing plants, the majority of infections remained latent. These latent infections exhibited little or no symptoms and CFU recoveries from infected tissues were always below 100 CFU g-1.  相似文献

Shoot samples of four peach rootstocks that are important to the peach industry in Greece: KID I, GF305, GF677, and PR204, were inoculated with Phytophthora cactorum and Phytophthora megasperma in the field and in the glasshouse and were then evaluated with regard to susceptibility. The pathogenicity of P. cactorum and P. megasperma to peach rootstocks was confirmed with an excised twig assay. The peach rootstocks showed differential susceptibility to P. cactorum . GF305 was the least susceptible and KID I was the most susceptible, which suggests that the latter rootstock is unsuitable for orchards in which the conditions are favourable for Phytophthora diseases. GF677 and PR204 were moderately susceptible. The plants that were inoculated with P. megasperma in the field and in the glasshouse showed no sign of infection whereas twigs inoculated in vitro with P. megasperma developed necrosis. GF305 was the most resistant, KID I was the most susceptible and PR204 and GF677 were moderately resistant. The present results demonstrate that none of the four peach rootstocks used in this study was completely resistant to P. cactorum , particularly when rootstocks were flooded periodically to enhance disease development. Therefore an integrated approach including host and resistance cultural practices is recommended to manage diseases caused by P. cactorum in peach orchards in Greece.  相似文献

 Bulked segregant analysis (BSA) was used to identify seven random amplified polymorphic DNA (RAPD) markers linked to the Rpf 1 gene. Rpf 1 confers resistance to Phytophthora fragariae var. fragariae, the causal agent of red stele root rot in Fragaria spp. The bulked DNAs represented subsets of a F1 population obtained from the cross Md683×Senga Sengana which consisted of 60 plants and segregated in a 1:1 ratio for resistance or susceptibility to race 2.3.4 isolate NS2 of P.  fragariae. Seven markers were shown to be linked to Rpf 1 and were generated from four primers; five of these markers were in coupling phase and two in repulsion phase with respect to the gene. A linkage map of this resistance gene region was generated using JoinMap 2.0TM. The manner in which Rpf 1 and the linked markers co-segregated indicated that they are inherited in a disomic fashion. These markers could enable gene pyramiding and marker-assisted selection of resistance genes in strawberry breeding programmes. Received: 26 August 1996 / Accepted: 20 December 1996  相似文献

Phytophthora root and crown rot (Phytophthora cryptogea) on gerbera is difficult to manage because most gerbera cultivars are susceptible to P. cryptogea. This study was conducted in order to determine the in vivo (pot experiment) efficacy of some fungicides and biofungicides. In pot experiments, fungicides were applied 7 days after inoculation with P. cryptogea, while biofungicide was applied 7 days before inoculation. In this study, soil drenches of five fungicides were tested. “Ametoctradin+dimethomorph (100 ml/day),” “mandipropamid+difenoconazole (60 ml/day),” “propamocarb+fosetyl‐Al (200 ml/day),” “mancozeb+metalaxyl‐M (250 g/day)” and “azoxystrobin+difenoconazole (100 ml/day)” active substances were used. Similarly, one biofungicide Bacillus amyloliquefaciens syn. MBI 600 (50 g/100 L) was applied by soil drenching. Efficacy of treatments was assessed according to the percentage of the root system which was visibly rotten at the end of the experiment. Root and crown rot severity was rated on a scale of 0 = 0% root system necrotic, 1 = 1%‐25% necrotic, 2 = 26%‐50% necrotic, 3 = 51%‐75% necrotic and 4 = 76%‐100% necrotic from 12 to 21 days. In this experiment, “azoxystrobin 200 g/L + difenoconazole 125 g/L” exhibited the highest efficacy against P. cryptogea with a ratio of 43.75%. The other fungicides and biofungicides ametoctradin 300 g/L + dimethomorph 225 g/L, mandipropamid 250 g/L + difenoconazole 250 g/L, propamocarb 530 g/L + fosety‐Al 310 g/L, mancozeb 64%+metalaxyl‐M 4% and Bacillus amyloliquefaciens syn. MBI 600 11% were ineffective. Importance should be given to management strategies of P. cryptogea of and more experiments should be carried out for a better understanding of the use of registered fungicides and biofungicides.  相似文献

Ditylenchus destructor and Ditylenchus dipsaci are economically important plant‐parasitic nematodes, affecting potato production mostly in temperate climates. Management through crop rotation is not feasible because of their wide host range. These nematodes are listed as quarantine pests in many countries. Limited information exists on the resistance and tolerance of currently cultivated potatoes to D. destructor and D. dipsaci. Two greenhouse experiments were conducted to screen 25 potato varieties for resistance to and tolerance for D. destructor and D. dipsaci infections. Reproduction factor (RF) and relative susceptibility (RS) were used to evaluate resistance, while potato tuber damage and tuber weight reduction was used to evaluate tolerance. Based on the RF, 16 varieties were evaluated as susceptible (S) while 5 varieties were evaluated as resistant (R) to D. destructor; varieties ‘Innovator’, ‘Aveka’ and ‘Spunta’ were resistant to D. dipsaci based on RF. ‘Désirée’ was observed to be highly susceptible to D. destructor and D. dipsaci in both experiments and was used as the standard susceptible control variety for the calculation of RS. A scale of 1–9 was used to classify RS of the potato varieties to D. destructor and D. dipsaci, where 9 indicated the highest level of resistance. All classes of resistance to D. destructor and D. dipsaci were observed in the potato varieties tested in the experiments. Six varieties had significantly lower RS to D. dipsaci than the standard susceptible control variety. Tolerant to highly sensitive potato varieties to both nematodes were also observed. RS and external potato tuber damage were identified as suitable methods for resistance and tolerance determination, respectively. This study provides essential information on the status of resistance and tolerance in potato varieties against D. destructor and D. dipsaci but needs to be confirmed under field conditions.  相似文献

A crown rot disease in wheat caused by the fungusFusarium graminearum Schw. Group 1 is a widespread problem in chronically Zn-deficient Australian soils. A link between crown rot and Zn deficiency was established by Sparrow and Graham (1988). This paper reports a test of a further hypothesis, that wheat genotypes more efficient at extracting zinc from low-zinc soils are more resistant to infection by this pathogen. Three wheat cultivars (Excalibur, Songlen and Durati) of differential Zn efficiency were tested at three zinc levels (0.05, 0.5 and 2.0 mg Zn kg−1 of soil) and three levels ofF. graminearum S. Group 1 inoculum (0.1 g and 0.3 g kg−1 live chaff-inoculum and control having 0.1 g kg−1 dead chaff inoculum). Six weeks after sowing dry matter production of shoots and roots was decreased byFusarium inoculation at 0.05 mg and 0.5 mg kg−1 applied Zn.Fusarium inoculum at 0.1 g was as effective as 0.3 g kg−1 for infection and decreasing dry matter. The infection at the basal part of culm decreased significantly by increasing the rate of Zn application. Excalibur, a Zn-efficient cultivar (tolerant to Zn deficiency) produced significantly more shoot and root dry matter, and showed less disease infection compared with Zn-inefficient cultivars (Durati and Songlen) at low (0.05 mg Zn kg−1 soil) and medium (0.5 mg Zn kg−1 soil) Zn fertilization rates. Higher rate of Zn fertilization (2.0 mg Zn kg−1 soil) reduced the disease level in Durati to the level of Excalibur but the disease level of Songlen was still high, indicating its high Zn requirement and or sensitivity to crown rot. The data on Zn uptake show that Excalibur, being Zn-efficient, was able to scavenge enough Zn from Zn-deficient soil, we suggest that besides sustaining growth Excalibur was able to build and maintain resistance to the pathogen; inefficient cultivars needed extra Zn fertilization to achieve performance comparable to that of Excalibur. The present study indicates that growing Zn-efficient cultivars of wheat along with judicious use of Zn fertilizer in Zn-deficient areas where crown rot is a problem may sustain wheat production by reducing the severity of the disease as well as by increasing the plant vigour through improved Zn nutrition. ei]Section editor: R Rodriques-Kalana  相似文献

1 The fecundity, longevity and establishment of Otiorhynchus sulcatus and Otiorhynchus ovatus from the Pacific North‐west U.S.A. was studied on five selected host plants: Picea abies‘Nidiformis’, Picea glauca‘Conica’, Taxus baccata, Rhododendron catawbiense‘Boursault’ and Fragaria×ananassa‘Totem’. 2 Teneral adults were used to study adult longevity and reproductive success. Leaves of these host plants were used for sustenance for 9 months. Larval establishment was studied by infesting potted host plants with eggs. 3 Fragaria×ananassa‘Totem’ produced the longest survival, shortest preoviposition time, the greatest number of eggs, and the highest fertility for adults of both species. Picea spp. were not good adult hosts for O. sulcatus. Taxus was a good adult host for O. sulcatus, but was a nonhost for adults and larvae of O. ovatus. 4 Adult hosts did not affect preoviposition time or egg viability with O. ovatus adults. With O. sulcatus, preoviposition time was greatly increased and egg viability was < 50% on Picea spp. 5 The best larval host was F.×ananassa‘Totem’ for O. sulcatus and P. glauca‘Conica’ for O. ovatus. Rhododendron was a poor larval host for both species. 6 When all of the studies on these two pests are considered, O. sulcatus appears to have varying host preferences from among its many geographical areas of occurrence whereas O. ovatus has a more universal host selection.  相似文献

The progeny of BC6F2-BC9F(2)-4 has been analyzed for resistance to brown rust (Lr genes) and powdery mildew (Pm genes). This progeny was obtained due to introgression of the alien material from the synthetic hexaploid wheat Triticum timopheevii/Aegilops squarrosa (= Triticum tauschii AAGGDD, 2n = 42) into the common wheat variety Saratovskaya 29. Against the background of natural infection, the lines resistant to both diseases and to either of them were developed. The brown-rust and powdery-mildew resistance is controlled by one/two effective independent genes Lr and Pm.  相似文献

Fungal invasions only recently started to receive more attention in invasion biology. This is largely attributed to little or non‐existent information about these inconspicuous organisms. Most invasion hypotheses focus on factors that increase invasion success; few try to explain why invasions fail. Here we hypothesize that a host–pathogen relationships can limit the invasiveness of an alien plant species in a novel range. To test this, we investigate whether the invasiveness of the Australian genus of Proteaceae, Banksia, in South Africa is determined by the alien and major invasive phytopathogen, Phytophthora cinnamomi. The presence of P. cinnamomi in Banksia root and soil was evaluated using morphological and molecular techniques. Isolates were cultured onto selective media and polymerize chain reactions and internal transcribing spacers were used for identification. Acetone leaf extracts of 11 Banksia spp. were screened for antimicrobial activity against P. cinnamomi, using the minimum inhibitory concentration assay. A total of 3840 Banksia individuals from seven localities were surveyed. Phytophthora  cinnamomi was consistently isolated from Banksia species root and soil samples. Out of the 12 Banksia species that were screened for antimicrobial activity, four introduced species, B. burdettii, B. coccinea, Banksia hookeriana, and B. prionotes and the invasive B. integrifolia and B. ericifolia exhibited relatively high antimicrobial activity against P. cinnamomi (strain 696/12). We show that the phytopathogen in the native range has similar impact in the novel range and in doing so may limit invasion success of Banksia species with low antimicrobial activity.  相似文献

Biology of the aphid Therioaphis maculata was studied on alfalfa (Medicago sativa L.), including four resistant (Mesa-Sirsa, CUF101, Baker and Lahontan) and two susceptible (ARC and Caliverde) alfalfa cultivars, and one of the most cropped Brazilian cultivars, Crioula. Under controlled conditions, antibiosis (i.e., reduced longevity, fecundity and increased mortality of the aphid) was observed mainly on the resistant alfalfa cultivars, except on Lahontan. Crioula seemed to be tolerant to aphids. Present data support geographic limitation usage of cultivars, and we suggest Baker and Mesa-Sirsa as sources of antibiosis, and provide biological information of a tropical T. maculata biotype on alfalfa.  相似文献

【背景】寄生帚梗柱孢霉是花生黑腐病的病原菌,被我国列为重要的进境植物检疫性有害生物。该病菌2009年已入侵我国广东,造成花生植株基部腐烂而死亡,严重威胁花生生产安全。筛选与种植抗病品种是防控该病害的重要措施。【方法】收集广东推广种植的15个主要花生品种,通过人工接种方法,鉴定这些品种对花生黑腐病菌的抗性水平。【结果】15个供试花生品种中,湛红2号、湛油62等2个品种表现为抗病;湛油75、湛油82、粤油390、粤油410、仲恺花44、仲恺花99、汕油诱1号等7个品种表现为中抗;花育33号、汕油523、汕油辐1号、粤油18、湛油53等5个品种表现为感病;仲恺花332表现为高感。【结论与意义】目前广东生产上推广种植的花生品种多数对黑腐病菌表现为抗病或中抗水平,部分品种表现为感病或高感。该结果可为我省花生品种的推广与布局提供依据。  相似文献

Photosynthetic performance, contents of chlorophyll and associated pigments, cellular damage and activities of antioxidative enzymes were investigated in two mustard (Brassica juncea L.) cultivars differing in photosynthetic capacity subjected to cadmium (Cd) stress. Exposure to Cd severely restricted the net photosynthetic rate (P(N)) of RH-30 compared to Varuna. This corresponded to the reductions in the activities of carbonic anhydrase (CA) and ribulose-1,5-bisphosphate carboxylase (Rubisco) in both the cultivars. Decline in chlorophyll (Chl) (a+b) and Chl a content was observed but decrease in Chl b was more conspicuous in Varuna under Cd treatments, which was responsible for higher Chl a:b ratio. Additionally, the relative amount of anthocyanin remained higher in Varuna compared to RH-30 even in the presence of high Cd concentration, while percent pheophytin content increased in RH-30 at low Cd concentration. A higher concentration of Cd (100 mg Cd kg(-1) soil) resulted in elevated hydrogen peroxide (H(2)O(2)) content in both the cultivars. However, Varuna exhibited lower content of H(2)O(2) in comparison to RH-30. This was reflected in the increased cellular damage in RH-30, expressed by greater thiobarbituric acid reactive substances (TBARS) content and electrolyte leakage. The enhanced activities of antioxidative enzymes, ascorbate peroxidase (APX), catalase (CAT) and glutathione reductase (GR) and also lower activity of superoxide dismutase (SOD) in Varuna alleviated Cd stress and protected the photosynthetic activity.  相似文献

Inter-section hybrids were successfully obtained by rescuing the immature embryos produced in the cross between Primula sieboldii of Sect. Cortusoides and P. obconica of Sect. Obconicolisteri. In these hybrid plants two types of triploids with different genome combinations were found in addition to the normal diploid hybrids which each had one genome of the parents. Among the five triploids obtained, four had two genomes of P. sieboldii and one genome of P. obconica, whereas the remaining one had one genome of P. sieboldii and two genomes of P. obconica. The possibilities of diploid female gamete formation in P. sieboldii and diploid pollen formation in P. obconica as the causal factors for these triploid formations were discussed. Bovine diarrhea virus (BVDV) causes a variety of economically important enteric and infertility problems in cattle. For that reason, several countries have eradicated the disease, and some others have schemes in progress to achieve freedom. Although there is a considerable amount of information about the risk of BVDV transmission through contaminated semen used for artificial insemination (AI), there is no evidence to indicate whether the resulting embryos, when used for embryo transfer, can lead to the transmission of BVDV to recipients or offspring. For this experiment, semen from a bull persistently infected with BVDV (105 50% tissue culture infective doses/mL NY strain) was used for insemination (two times at estrus) of BVDV-seronegative, superovulated cows (N = 35). Embryos were collected 7 days after insemination and subsequently were washed according to the International Embryo Transfer Society recommendations or left unwashed. Out of 302 collected oocytes and embryos, 173 (57%) were fertilized and the remaining 129 (43%) had degenerated. Infectious BVDV was detected in 24% (17/71) of unwashed and 10% (8/77) of washed embryos, and in all (N = 11) follicular fluid samples, oviductal epithelial cells, endometrium, and corpora lutea tissues as determined by the virus isolation test. After transfer of 39 washed embryos to 27 BVDV-seronegative recipients, 12 (44%) cows became pregnant and 17 calves free of BVDV and BVDV antibodies, including five sets of twins, were born. After embryo transfer, all pregnant and nonpregnant recipients remained free of BVDV and antibodies. In conclusion, results herein suggest that BVDV can be transmitted by AI resulting in the production of some proportion of contaminated embryos. However, it appears that such embryos, when washed according to International Embryo Transfer Society and the World Organization for Animal Health guidelines do not cause BVDV transmission to recipients or their offspring.  相似文献

Graphical analysis of first-flowering dates of selected garden plants 1946–2002 shows an earlier trend, which appears to be linked to temperature and global warming. Some species, however, seem to buck this trend, which may be linked to photoperiodic response. Combined graphs of species selected at intervals throughout the calendar year reveal a rhythm or wave of several years in Leeds 1946–1962 and a less marked rhythm in Richmond 1961–2002.  相似文献
