Mechanisms of biocontrol of soil-borne plant pathogens by Rhizobacteria   总被引:3,自引:0,他引:3
Bacterial antagonism, responsible for biological control, may operate by antiobiosis, competition or parasitism. Parasitism relies on lytic enzymes for the degradation of cell walls of pathogenic fungi. Serratia marcescens was found to be an efficient biocontrol agent of Sclerotium rolfsii and Rhizoctonia solani under greenhouse conditions. Populations of 105 or 106 colony forming units g-1 soil were the most effective. Drench and drip application of S. marcescens suspension were more effective in controlling S. rolfsii than spraying, mixing in soil or seed coating. The highest population density of the bacteria in the rhizosphere was found on the proximal portion of the root, decreasing significantly until the tips, where it increased again. The isolated Serratia, found to possess chitinolytic activity, was able to release N-acetyl D-glucosamine from cell walls of S. rolfsii. The gene coding for chitinase was cloned into Escherichia coli and the enzyme was uniquely excreted from the bacterium into its growth medium. When S. rolfsii was sprayed by partially purified chitinase produced by the cloned gene, rapid and extensive bursting of the hyphal tips was observed. This chitinase preparation was effective in reducing disease incidence caused by S. rolfsii in beans and R. solani in cotton, under greenhouse conditions. A similar effect was obtained when a viable E. coli cell, containing the plasmid with the chitinase gene (pLCHIA), was applied. It appears that genetic engineering of the lytic enzymes, such as chitinase which play an important role in plant disease control, may improve the efficacy of biocontrol agents.  相似文献

Novel strains of rhizobacteria, Pseudomonas fluorescens Pf 9A‐14, Pseudomonas sp. Psp. 8D‐45 and Bacillus subtilis Bs 8B‐1, showed broad‐spectrum antagonistic activity and provided suppression of Pythium damping‐off and root rot of cucumber. Their biocontrol potential was further investigated for suppression of additional seedling diseases of cucumber (Phytophthora capsici) and radish (Rhizoctonia solani). Bacterial strains were also characterised for production of antibiotics, metabolites, volatiles, phytohormones and lytic enzymes. Seed and pre‐plant applications of all three antagonistic bacteria as cell suspension and talc or irradiated peat formulations to the infested potting mix provided overall high level of suppression of Phytophthora damping‐off and root rot of cucumber (66–85% healthy seedlings) and relatively low level of suppression of Rhizoctonia damping‐off of radish (18–38% healthy seedlings). Bacterial treatments also resulted in higher plant fresh masses. Seed coating with irradiated peat formulation of a mixture of three bacteria resulted in superior control of Phytophthora damping‐off and root rot of cucumber and much higher plant fresh masses. The presence of genes for biosynthesis of phenazine‐1‐carboxylic acid, 2,4‐diacetylphloroglucinol, pyrrolnitrin and pyoluteorin was confirmed in Pseudomonas strains, and that of fengycin, bacillomycin, bacilysin, surfactin and iturin A in B. subtilis Bs 8B‐1. All three strains produced HCN, salicylic acid, indole‐3‐acetic acid, protease and β‐1,3‐glucanase. Both Pseudomonas strains produced siderophores and only P. fluorescens Pf 9A‐14 showed phosphate solubilisation and chitinase activity. All three strains inhibited pathogen growth by producing volatiles, and gas chromatography–mass spectrometry analysis revealed eight compounds in Pf 9A‐14, 10 in Bs 8B‐1 and 4 in Psp 8D‐45, some with known antifungal activity. The antagonistic and plant‐growth promotion activities of these strains might be due to production of antibiotics, metabolites, lytic enzymes or phytohormones.  相似文献

Five strains of Streptomyces sp. were evaluated in vitro for their ability of inhibiting the mycelial growth of Macrophomina phaseolina, the causal agent of root rot of mung bean (Vigna radiata L.). Among the Streptomyces sp. strains tested, PDK showed the maximum in vitro inhibition of mycelial growth of M. phaseolina and recorded an inhibition zone of 21?mm. The strains CBE, MDU, SA and ANR recorded inhibition zones of 18, 16, 13 and 11?mm, respectively. These Streptomyces sp. strains were tested for their growth-promoting efficiency on mung bean seedlings. Among them, CBE and PDK recorded the maximum increase in shoot length, root length and seedling vigour compared with control, followed by MDU. Three Streptomyces sp. strains (CBE, MDU and PDK) that showed higher levels of inhibition of growth of M. phaseolina in dual culture assay and plant growth-promoting activity were tested for their biocontrol activity against root rot under greenhouse and field conditions. Seed treatment or soil application with powder formulation of Streptomyces sp. strains CBE, MDU and PDK was effective in controlling root rot disease; but, combined application through seed and soil increased the efficacy in both the greenhouse and field trials. Among the treatments, seed treatment plus soil application with powder formulation of Streptomyces sp. strain CBE proved to be most effective, which reduced the root rot incidence from 26.8% (with non-bacterised seeds) to 4.0% in Trial I and from 32.0 to 4.9% in Trial II. The above treatment recorded the highest yield in both the field trials, and the yield increase was 78 and 74% over control in Trial I and Trial II, respectively. Isozyme analysis of the Streptomyces sp.-treated plants indicates that seed treatment plus soil application strongly induce the activities of peroxidase (PO-1 and PO-2) and polyphenol oxidase (PPO-2 and PPO-3) in mung bean. Among the three strains tested, Streptomyces sp. strain MDU- treated plants showed higher levels of activities of PO and PPO. Based on the above findings, it can be concluded that both the direct inhibition of pathogen and induced resistance might be involved in the control of root rot of mung bean by Streptomyces sp.  相似文献

Biological control of fungi causing root rot on sugar beet by native Streptomyces isolates (C and S2) was evaluated in this study. The dry weight and colony forming unit (CFU) of S2 and C increased when 300 mM NaCl was added to medium. The in vitro antagonism assays showed that both isolates had inhibitory effect against Rhizoctonia solani AG-2, Fusarium solani and Phytophthora drechsleri. In dual culture, Streptomyces isolate C inhibited mycelial growth of R. solani, F. solani and P. drechsleri 45%, 53% and 26%, respectively. NaCl treatment of medium increased biocontrol activity of soluble and volatile compounds of isolate C and S2. After salt treatment, growth inhibition of R. solani, F. solani and P. drechsleri by isolate C increased up to 59%, 70% and 79%, respectively. To elucidate the mode of antagonism, protease, chitinase, beta glucanase, cellulase, lipase and α-amylase activity and siderophore and salicylic acid (SA) production were evaluated. Both isolates showed protease, chitinase and α-amylase activity. Also, biosynthesis of siderophore was detectable for both isolates. Production of siderophore and activity of protease and α-amylase increased after adding salt for both isolates. In contrast, chitinase activity decreased significantly. Production of SA, beta glucanase and lipase by isolate S2 and biosynthesis of cellulase by isolate C were observed in presence and absence of NaCl. Soil treatment with Streptomyces isolate C inhibited root rot of sugar beet caused by P. drechsleri, R. solani and F. solani. Results of this study showed that these two Streptomyces isolates had potential to be utilized as biocontrol agent against fungal diseases especially in saline soils.  相似文献

Trichoderma has been used to manage a large number of pathogens, but there is a gap in the mechanisms used by these biocontrol agents regarding the physiological response of cassava plants (Manihot esculenta) when it is subjected to cassava root rot. The aims of this study were to investigate the antagonist activity of ten Trichoderma isolates against Fusarium solani on potato dextrose Agar (PDA), to quantify the chitinase production, to select and test in vivo the best isolate from each experiment and to assess the physiological response of cassava to the production of oxidative enzyme complex production (ascorbate peroxidase, catalase, peroxidase and polyphenol oxidase). All Trichoderma isolates have shown competitive capability against F. solani, and Trichoderma hamatum URM 6656 showed the highest inhibition of pathogen growth (88.91%). All isolates have shown chitinase activity, but Trichoderma aureoviride URM 5158 produced the highest amount of chitinase. T. hamatum URM 6656 and Taureoviride URM 5158 were selected to be applied in vivo. The two Trichoderma strains reduced 64 and 60% of the disease severity in the shoot and 82 and 84% in the root. Cassava plants infected with Trichoderma have shown the highest peroxidase and ascorbate peroxidase production. Our results have indicated that T. aureoviride URM 5158 is an effective biocontrol agent against cassava root rot caused by F. solani, because it presented competitive antagonist capability in vitro, the highest chitinase production, and reduced the cassava root rot severity. The application of T. aureoviride has led to the maximum enzyme activity of reactive oxygen species group in cassava plants.  相似文献

The current study aimed at developing diverse Trichoderma fusants for fungicides, drought, and salt tolerance with enhanced antagonistic activity against Sclerotium rolfsii Sacc. Trichoderma virens NBAII Tvs12 (mycoparasitic) and Trichoderma koningii MTCC796 (multistress tolerant) were used as parental strains for development of interspecific protoplast fusants. A total of 36 stable fusants were used for mycoparasitism, fungicides, and abiotic stresses (drought and salt) tolerance. The results revealed 20 homozygous progenies showing characteristics of either one parental strain and 14 heterozygous mutants depicting traits of both parental strains. A novel concept of inhibition coefficient was established using growth-related key parameters that represent the pathogen biology and the biocontrol-related biophysics of Trichoderma fusants. The results indicated a differential inhibition coefficient of the test pathogen and the highest (92.88%) inhibition coefficient of S. rolfsii was observed by interstable fusant Fu21. It also grew better under fungicides and abiotic stress (drought and salt) conditions. The molecular characterization and heterozygosity analysis evidenced the highest observed heterozygosity (0.5441) and gene flow (0.3872) in stable heterozygous Fu21. Principal coordinates analysis exhibited 62.7% of total variability. The ecofriendly heterozygous Trichoderma fusant (Fu21) might be useful for biocontrol of stem rot disease under adverse conditions or as a part of integrated disease management.  相似文献

Pseudomonas fluorescens BAM-4, Burkholderia cepacia BAM-6 and B. cepacia BAM-12 isolated from the rhizosphere of moong bean (Vigna radiata L.) showed significant growth-inhibitory activity against a range of phytopathogenic fungi. Light and scanning electron microscopic (SEM) studies showed morphological abnormalities such as fragmentation, swelling, perforation and lysis of hyphae of pathogens by Pseudomonas and Burkholderia. Two of the strains (BAM-4 and BAM-6) produced siderophore in CAS agar plates, whereas all three strains produced chitinase. Bacterization of seeds of moong bean with pseudomonads has been reported as a potential method for enhancing plant growth and yield, and for providing protection against Macrophomina phaseolina. Seed bacterization with these plant growth-promoting rhizobacteria (PGPR) showed a significant increase in seed germination, shoot length, shoot fresh and dry weight, root length, root fresh and dry weight, leaf area and rhizosphere colonization. Yield parameters such as pods, number of seeds, and grain yield per plant also enhanced significantly in comparison to control. The disease suppression and plant growth enhancement along with the positive rhizosphere colonization by these strains indicate their possible use as PGPR/biocontrol agents against charcoal rot.  相似文献

Aims: To determine the role of phenazines (PHZ) and lipopeptide surfactants (LPs) produced by Pseudomonas in suppression of stem rot disease of groundnut, caused by the fungal pathogen Sclerotium rolfsii. Methods and Results: In vitro assays showed that PHZ‐producing Pseudomonas chlororaphis strain Phz24 significantly inhibited hyphal growth of S. rolfsii and suppressed stem rot disease of groundnut under field conditions. Biosynthesis and regulatory mutants of Phz24 deficient in PHZ production were less effective in pathogen suppression. Pseudomonas strains SS101, SBW25 and 267, producing viscosin or putisolvin‐like LPs, only marginally inhibited hyphal growth of S. rolfsii and did not suppress stem rot disease. In contrast, Pseudomonas strain SH‐C52, producing the chlorinated LP thanamycin, inhibited hyphal growth of S. rolfsii and significantly reduced stem rot disease of groundnut in nethouse and field experiments, whereas its thanamycin‐deficient mutant was less effective. Conclusions: Phenazines and specific lipopeptides play an important role in suppression of stem rot disease of groundnut by root‐colonizing Pseudomonas strains. Significance and Impact of the Study: Pseudomonas strains Phz24 and SH‐C52 showed significant control of stem rot disease. Treatment of seeds or soil with these strains provides a promising supplementary strategy to control stem rot disease of groundnut.  相似文献

The potential use of Bacillus thuringiensis UM96 as a biocontrol agent for the grey mould phytopathogen Botrytis cinerea was evaluated. In order to dissect the mode of action of this UM96 strain, we also examined the role of lytic activities in the antagonism. First, B. thuringiensis UM96 was characterised based on 16S rRNA and gyrA gene sequencing and phenotypic traits. Petri dish biocontrol assays demonstrated that when strain UM96 was inoculated 24 h previous to B. cinerea, the mycelial growth was inhibited by up to 70%. Test for lytic enzymes activities of cellulase and glucanase was negative. Chitinase was the only positive enzyme activity in two different culture media. PCR detection of the chiB gene was also positive. Chitinolytic supernatants, obtained from rich and minimal media supplemented with colloidal chitin as the sole carbon source, from B. thuringiensis UM96 showed a strong inhibitory effect of B. cinerea that was not observed with heat-treated supernatant. Interestingly, when the supernatant was supplemented with 100 µM allosamidin, a chitinase specific inhibitor, the antagonistic activity was suppressed significantly. A lack of chitinase activity was also observed in allosamidin-treated supernatants. Our pathogenic B. cinerea strain also exhibited susceptibility to pure Streptomyces griseus chitinase. Finally, the chitinolytic strain B. thuringiensis UM96 was able to protect Medicago truncatula plants in vitro from B. cinerea infection and significantly reduced the necrotic zones and root browning of the plants. Together, these results suggest a potential use of B. thuringiensis UM96 for the biological control of B. cinerea and a role for chitinases during the antagonism displayed.  相似文献

Vermicompost-based bioformulations of bacterial and fungal biocontrol agents were examined against sugarbeet root rot caused by Sclerotium rolfsii. The result showed that the Pseudomonas fluorescens strain Pf1 in combination with either Trichoderma asperellum strain TTH1 or Bacillus subtilis strain EPCO-16 performed better in reducing disease next to the chemical difenoconazole. Similarly, enhanced yield was observed in the same combination treatments under both pot and field conditions.  相似文献

The effect of application of different biocontrol agents (Trichoderma spp (eight isolates), Pseudomonas fluorescens (four isolates) and Bacillus subtilis (two isolates)) was tested against head rot fungus, Sclerotinia sclerotiorum and root-knot nematode, Meloidogyne incognita complex diseases. In vitro studies showed that three Trichoderma isolates (Tvc1, Tvc2 and Thc) were effective in inhibition of mycelial growth of the fungus as well as egg hatching ability of the nematode. Application of talc based formulation of biocontrol agents (bioformulations) individually as well as in mixtures with or without chitin was tested against head rot–root-knot disease complex under greenhouse conditions. The combined application of bioformulation mixture (Tvc1, Tvc2 and Thc) along with chitin reduced the incidence of the complex diseases and induced significantly increased activities of phenylalanine ammonia-lyase (PAL), peroxidase (PO), polyphenol oxidase (PPO) and chitinase in cabbage. Activities of PAL and chitinase reached maximum levels within 10 – 20 d, while the activity of PAL continued to be maintained up to 40 d after the application of Tvc1 + Tvc2 + Thc + chitin. Isozyme analyses observed that unique PO (PO1, PO2 PO3 and PO4) and PPO (PPO1, PPO2 and PPO3) enzymes were induced after 10 d in cabbage plants treated with Tvc1 + Tvc2 + Thc + chitin upon challenge inoculation with head rot–root-knot pathogens. Similarly, the bioformulation mixture with chitin was successful at reducing the incidence of head rot–root-knot apart from enhancing the crop yield under field conditions. The mechanism associated with reduced incidence of head rot–root-knot in cabbage may be due to induction of defense proteins (PAL, PO, PPO and chitinase) in the crop.  相似文献

This study investigates the biochemical relationships between carrot roots and Pythium violae, the pathogen responsible for cavity spot (CS) disease. P. violae isolates obtained from CS lesions, cultured in Petri dishes on agar were used for inoculation of uninfected mature carrots. The fungus secreted a wide spectrum of enzymes that degraded the cellulose and pectic substances of the carrot cell walls. Cellulase and polygalacuronase (pg) showed the highest activity during the first day post-inoculation, subsequently declining. Pectin lyase (PnL), pectate lyase (PeL) and pectin methylesterase (PME) gradually increased to their highest levels of activity 14 to 30 days post-inoculation. This pattern of activity enables the penetration of the fungus through the walls of the host cells and the establishment of the hyphae. Several plant pathogen-related substances such as peroxidase, chitinase, glucanase and polyphenol oxidase were produced in the infected tissue. Peroxidase activity rose in the inoculated roots from day 1 post-inoculation. Chitinase, glucanase and polyphenol oxidase activities first appeared 3–4 days post-inoculation. At this time, two bands corresponding to chitinase at about 26 and 33 KDa and one band corresponding to glucanase at about 24 KDa could be resolved by SDS-PAGE.  相似文献

Abstract

The effect of Salicylic Acid (SA) in inducing resistance in groundnut plants against Alternaria alternata was investigated. Foliar application of SA at the concentration of 1 mM significantly reduced the leaf blight disease intensity and increased the pod yield under glasshouse conditions. Changes in the activities of phenylalanine ammonium lyase, chitinase β-1,3 glucanase and in phenolic content on groundnut after application of SA and inoculation with A. alternate were studied. In SA-treated leaves (plants) an increase in phenolic content was observed five days after challenge inoculation with A. alternata in groundnut plants pretreated with SA. There was a marked increase in chitinase and pathogen inoculation in SA-treated leaves. In chitinase, β-1,3 glucanase activities were observed in response to plants with an increase in SA treated leaves. Foliar applications of SA-induced in peroxidase and polyphenol oxidase activities were observed upon challenge inoculation with pathogen.  相似文献

Melanins and Resistance of Fungi to Lysis   总被引:13,自引:4,他引:9
Hyphal walls of Aspergillus phoenicis and Sclerotium rolfsii are composed of large amounts of glucose- and N-acetylhexosamine-containing polysaccharides, and the walls are extensively digested by streptomycete culture filtrates or by a mixture of purified chitinase and β-(1 → 3) glucanase preparations with the release of the monomeric units. A. phoenicis conidial walls also contain polymers of glucose and N-acetylhexosamine, but these walls are resistant to digestion by microorganisms or the enzyme combination active on the hyphae. When the melanin-containing spicules were removed from the spore surface, however, the chitinase and glucanase partially digested the underlying structural components. Microorganisms decomposing hyphal walls of S. rolfsii did not attack the melanin-covered sclerotia produced by this fungus. No microorganism capable of lysing two fungi, Rhizoctonia solani and Cladosporium sp., producing hyphae containing abundant melanin was found. The ecological significance of these findings and possible mechanisms for the protective influence associated with melanins are discussed.  相似文献

Intercrossing red clover plants, selected for reaction to Fusarium root rot (F. avenaceum), produced progenies with lower root rot scores than the parental cultivar Hermes II. Eleven percent of the progenies had less than 2 mm root rot after two cycles of selection, compared to less than 1% in the parental cultivar. Root rot severity in two old local strains of red clover, as well as in two populations of wild red clover, was lower than in cv. Hermes II. This could indicate that a natural selection for root rot resistance can occur in red clover.  相似文献

Abstract

Fluorescent pseudomonads based bioformulation was evaluated for their ability to control Macrophomina root rot disease in mungbean (Vigna mungo). P. fluorescens isolate Pf1 showed the maximum inhibition in mycelial growth of Macrophomina phaseolina under in vitro conditions. Bioformulation of Pf1 with chitin was effective in reducing the root rot incidence in green gram both under glasshouse and field conditions. The rhizosphere colonization of P. fluorescens was observed appreciable with the green gram plants. However, Pf1 amended with chitin colonized effectively. Furthermore, the induction of defence-related enzymes and chemicals in plants by Pf1 amended with or without chitin and neem were tested. Increased accumulation of defence enzymes viz., phenylalanine ammonia lyase (PAL), peroxidase (PO), polyphenol oxidase (PPO), chitinase, β-1,3-glucanse and phenolics were observed in Pf1 bioformulation amended with chitin, pre-treated plants challenge inoculated with M. phaseolina under glasshouse conditions. The present study reveals that in addition to direct antagonism and plant-growth promotion, PGPR strains amended with chitin bioformulation induced defence-related enzymes and pathogenesis related (PR) proteins which collectively enhance the resistance in green gram against the infection of M. phaseolina.  相似文献

Trichoderma harzianum and Trichoderma viride, which were grown on modified media of rice straw waste, were used as suitable natural media for determining the population of the bioagents and stimulating the production of antimicrobial substances, i.e. toxins, enzymes at different periods (1, 2, 3, 4, 5 and 6 weeks). Also, the evaluation of synergistic effect between biocontrol fungi and bacteria may play a key role in the natural process of biocontrol. Synergism can occur when different agents are applied together and cell wall degrading enzymes produced by fungi can increase the efficacy of bacteria. The evaluation of the bioagents formulated on rice straw waste has influenced the percentage of infection of root rot on bean plants under green house and field conditions. The formulation of the Trichoderma spp. to reduce the incidence of the diseases caused by soil-borne fungi in the field has great importance in the biocontrol of the diseases. This work was aimed to determine and improve the efficacy of application and formulation of Trichoderma spp. on the rice straw waste against root-rot disease of bean under green house and field conditions.  相似文献

Antagonistic Trichoderma spp. are used throughout the world for the biological control of soil-borne plant diseases. This approach has stimulated an on-going search for more efficient mycoparasitic strains with a high potential for producing extracellular lytic enzymes. This study compares the production of lytic enzymes by native strains of Trichoderma asperellum and Trichoderma longibrachiatum on substrates of differing complexity. The quantity of protein induced by Agaricus bisporus-based medium was higher than that induced by Phymatotrichopsis omnivora-based medium. In P. omnivora medium, T. asperellum exhibited higher chitinolytic and β-1,3-glucanolytic activities than T. longibrachiatum. The enzyme profile was related to the previously reported ability of these strains to inhibit the growth of several soil-borne plant pathogens. NAGase production was similar among the tested indigenous strains of T. longibrachiatum; T479 and T359 produced more endochitinase, T479 produced more glucanase, and T341 and T359 produced more β-1,3-glucanase. The detected variations in glucanase and β-1,3-glucanase activities suggest that the production of these enzymes is strongly influenced by the substrate. Strains T397 and T359 exhibited xylanase activity, which triggers defence mechanisms in plants. Thus, these strains may utilise an additional mechanism of biocontrol.  相似文献

We isolated 137 fungal strains from females and eggs of Meloidogyne spp. collected from plant roots and infested soil in Jiangsu, China, and examined their in vitro protease production and chitinase activities. We then selected 20 strains, each with a protease production higher than 2 (protease production was measured as the ratio of diameters of the zone of clearing to that of the colony) and a chitinase activity higher than 0.2 U/min mL. For these strains, we examined their egg parasitic rates, mortality rates of stage 2 juvenile (J2), egg hatching rates and evaluated their biocontrol efficacies under greenhouse conditions. The biocontrol efficacies of these 20 potential biocontrol fungal strains ranged from 29 to 58%. The actual biocontrol efficacy against Meloidogyne root-knot was highly correlated with the in vitro protease production (r=0.82), chitinase activity (r=0.80), and mortality of J2 (r=0.76), but poorly correlated with the egg parasitic rate (r=0.51) and the egg hatch rate (r=0.52). Based on these results, we suggest that in vitro protease production or chitinase activities of the fungal strains could be an applicable indicator for assessing biocontrol efficacy against nematodes. This notion was also supported by our field experiments on tomatoes with Paecilomyces lilacinus strains XT18, XCS46 and Pochonia chlamydosporia strain XT124, which reached the biocontrol efficacies of 60.7, 51.5 and 48.1% in 2007; and 67.8, 57.3 and 53.2% in 2008, respectively.  相似文献
