This is a staining technique for histopathologic evaluation of tissue reaction in the environs of acid-fast tubercle bacilli (avian and bovine) in sections. Fresh tissue is fixed in 10% neutral formalin and processed in the usual manner for embedding in paraffin. Sections are cut approximately 6 μ. thick, dewaxed, hydrated, and stained with Harris' hematoxylin. They are rinsed in tap water, differentiated in add alcohol, washed in tap water, given a distilled water rinse and stained at 20-30° C in a 1% solution of new fuchsin in 5% phenol. Each slide is then handled individually by placing it directly into a saturated aqueous solution of Li2CO3 and agitated gently for a few seconds. This is followed by differentiation with 5% glacial acetic acid in absolute or 95% ethyl alcohol until the color stops running. Two rinses in absolute or 95% ethyl alcohol follow. The sections are then counterstained in the color add of eosin Y prepared according to the method of Schleicher (Stain Techn., 28, 119-23, 1953) and used as an 0.025% solution in absolute alcohol. Following passage through 2 changes of absolute alcohol, the sections are cleared in xylene, then mounted in Permount or similar synthetic resin. The add-fast barilli are emphasized by their bright retractile red color within a contrasting background of hematoxylin and eosin.  相似文献

A modification of the Golgi technic is described in which the reaction takes place in well fixed formalin material. Thin slices (whole sections of adult monkey, cat and rat cerebrum) 2 to 3 mm. thick, from brains fixed 3 to 4 months in 10% formalin, are chromated for two days in 3 g. of zinc chromate dissolved in 98 ml. of distilled water and 2 ml. of formic acid. Slices are then removed, blotted dry and immersed, suspended by a thread, in 0.75% silver nitrate solution for two days. Solution should be changed after the first day. After silvering, the slices are dehydrated rapidly (total time about one hour) in 95% and absolute alcohol, placed in xylene 10 minutes, in low melting point paraffin 10 minutes and embedded in low melting point paraffin. Only surface infiltration is necessary since sections are cut 90 to 100 u. Sections are collected in 95% alcohol, dehydrated in absolute alcohol, cleared in several changes of xylene and mounted in Fisher's Permount. Results with fetal and new born material were not good.  相似文献

Axoplasm is selectively impregnated by the following steps: (1) fixation in 10% formalin or in 10% formalin with added sucrose, 15%, and concentrated NH4OH, 1%, for 1-7 days; (2) frozen sections; (3) extraction of the sections in 95% ethyl alcohol, absolute alcohol, xylene, and 95% ethyl alcohol and absolute alcohol, 1 hr each; (4) distilled water, 3 changes of 10 min each; (5) 20% AgNO3 (aq.) at 25°C, 30 min; (6) distilled water, 3 changes of 1-2 sec each; (7) 6.9% K2CO3, 1 hr; (8) water, 3 changes of about 1 min each; (9) 0.2%AuCl3, 2 min; (10) distilled water; (11) 5% Na2S2O3, 2 min; (12) washing, clearing and mounting. This procedure is proposed as a simplified stain for axoplasm, with other tissue components remaining unstained. The few reagents necessary suit this method for histochemical investigation of the mechanism of silver staining.  相似文献

Sections containing gelatinous fibers were cut at 15 μ from material both fixed and stored in formalin-acetic-alcohol, 5:5:90 (of 70%). These sections were stained 5 min in a 1% aqueous solution of lignin pink (G. T. Gurr), differentiated quickly in water, soaked 5 min in 95% ethyl alcohol, dehydrated in absolute ethyl alcohol and counter stained 5 min with a 1% solution of chlorazol black E (G. T. Gurr) in methyl cellosolve, followed by dehydration in absolute ethyl alcohol, clearing in xylene and mounting in Canada balsam. The gelatinous layer was sharply defined as a dense black zone whilst the remainder of the cell wall stained light pink. The specificity of the technique was superior to that of safranin and light green, and was not easily obscured by overstaining. The technique is particularly useful for locating small zones of gelatinous fibres, and for photomicrographical work.  相似文献

After fixing in phosphate-buffered 5% glutaraldehyde, pH 6.8, by perfusion, brains were sliced to 3-5 mm pieces which were placed in the fixative for 5-7 days. The pieces were washed through several changes of 2.26% NaH2PO4 for 12 hr, 30 μ frozen sections cut, and mordanted 2 days in an equal-parts mixture of 3.5% CrO3 and 5% Na-tartrate, which had been aged at 20-25 C for 20 days prior to use. After washing in distilled water, the sections were put into a solution containing AgNO3, 20 gm; and KNO3, 15 gm, in distilled water, 80 ml; at 30 C for 1.5-2 hr, then reduced at 40-45 C in three pyrogallol solutions as follows: 1-2 sec in 1% pyrogallol in 55% alcohol; 3-4 sec in a 0.67% solution in 33% alcohol, and 5-7 sec in a 0.5% solution in 25% alcohol. Gold toning is optional; dehydration, clearing and covering, routine. The technic shows particularly the perisomatic fibers, boutons en passant and boutons termineaux. Fibers in nerve tracts may be visible but lightly stained; cell nuclei may be dark, but the cytoplasm remains pale.  相似文献

Whole brains of cat were fixed in two changes of cold acetone (24 hours each) and embedded directly in paraffin. The degeneration time recommended is 5 days. Mounted sections 15-20 μ thick were deparaffined, washed in absolute alcohol and given successive treatments of 6 hours each with 1% ammoniated absolute alcohol and pure pyridine, washing well with distilled water between them and after the pyridine. Impregnation in 2% silver nitrate 12 hours at 30°C., rinsing in absolute alcohol and reducing in a 95% alcoholic solution of pyrogallol and formalin (3% and 5%) was followed by 50% alcohol, thorough washing in distilled water, toning in 1% gold chloride and intensification in 1% oxalic acid. Treatment in 10% sodium thiosulfate solution, washing, dehydrating and covering completed the procedure. Normal fibers, degenerating fibers and terminals were stained specifically.  相似文献

Celloidin blocks of Golgi-Cox impregnated material are cut at 50 μ, the sections collected in 70% alcohol, transferred to a 3:1 mixture of absolute alcohol and chloroform for 2 min, and then stored in xylene or toluene for at least 3 min, or up to 2 wk until processed further. Mounting is done on glass slides which have been coated with fresh egg albumen diluted in 0.2% ammonia water (or a 0.5% solution of dry powdered egg albumen) and then dried at 60°C overnight. For attachment to these coated slides, sections are first soaked for 2-3 min in a freshly prepared mixture of methyl benzoate, 50 ml; benzyl alcohol, 200 ml; chloroform, 150 ml; and then transferred quickly to the slides by means of a brush. After 2-3 min the chloroform evaporates and the celloidin softens. The slides are then immersed in toluene which hardens the celloidin and anchors the sections to the slides. Alcohols of descending concentrations to 40% are followed by alkalinizations, first in: absolute alcohol, 40 ml; strong ammonia water 60 ml, for 2 min, then in: absolute alcohol, 70 ml; strong ammonia water, 30 ml, for 1 hr. Excess alkali is then removed by 70% and 40% alcohol, 2 min each, and a 10 min wash in running tap water. Bleaching in 1% Na2S2O3, for 10 min and washing again in tap water for 10 min completes the process preliminary to staining. The preparations are then stained for 90 min in an aqueous solution of either 0.5% cresylecht violet, neutral red, or Darrow red, buffered at pH 3.6. Dehydration and differentiation in ascending grades of alcohol, clearing with toluene or xylene, and applying a cover glass with a mounting medium having a refractive index of about 1.61 completes the process.  相似文献

Rinse in 70% alcohol, pass to distilled water, then overstain (2 hr.) in aniline blue, 0.5% solution in 2.5% acetic acid (aqueous). Precipitate the anilin blue with 0.5 ml. of 0.1% methyl violet solution (aqueous) dropped on die slide. Leave on 2 min. or less. Wash and differentiate in 70% alcohol. (Parietal cells dark blue). Stain 30 min. in a mixture of hematein, 0.10g.; A1C13 cryst., 0.05g.; and 70% alcohol 50 ml., prepared just before use and not filtered. Rinse in 70% alcohol and differentiate with an alcoholic extract of saffron (2 g. saffron pistils in 100 ml. 90% alcohol at 60°C. for 6 hr.) while observing the progress of differentiation microscopically. Dehydrate by dropping a 0.1 % solution of acetic acid in absolute alcohol on the section for 30 sec., followed by pure absolute alcohol, xylene, and covering in balsam.  相似文献

A method of preparing bone or teeth for sectioning is described which involves the following steps: 48 hr. in 1:10 formalin; 24 hr. in 70% alcohol; decalcification for several days in 10% HNO3; rinsing and transferring to 2% potassium alum for 12 hr.; rinsing and treating with 5% NaHCO3 (or Li2CO3) for 24 hr.; washing for 12-24 hr.; then passing through ascending grades of alcohol to xylene. In the case of developing teeth, a slightly different procedure is recommended: fixation in Heidenhain's Susa till hard tissue is decalcified; 24 hr. in 96% alcohol (with three changes); 24 hr. in absolute alcohol (with one change); clearing in xylene or chloroform, and embedding in paraffin.  相似文献

After recordings had been taken from a microelectrode used for mapping nerve impulses, a current of 100 μa from the positive pole of a direct current generator was run through the electrode for 5 sec while it was still in place. On terminating the experiment, in which the use of several electrodes was possible, 50-75 ml of a 1:1 mixture of 4% potassium ferrocyanide and 4% acetic acid was injected into each common carotid artery, and the brain left in situ for 0.5 hr. It was then removed and the electrode-bearing part fixed 5-6 hr in a 1:1 mixture of 40% formalin and 95% ethyl alcohol at 55 °C. This specimen was washed in running water 5-10 min, the electrodes removed and frozen sections of 40-80 μ cut and placed in 95% alcohol. Sections were stained 5-10 min at 25-30°C in 10% silver nitrate solution in 75-80% alcohol acidified by 3-4 drops of glacial acetic acid per 50 ml, washed 4-5 sec in each of 2 baths of 95% alcohol, and reduced while being agitated constantly in a 2% solution of pyrogallol and 6-7% formalin in 75-80% alcohol. Washing in 95% alcohol, clearing in clove oil or methyl salicylate followed by xylene and mounting in synthetic resin or balsam completed the process. Sites of electrolysis at the tips of electrodes (under magnification) were blue before silver staining and black after staining. Axons stained brown to black on a yellow background.  相似文献

Autopsy and biopsy specimens of human skin were fixed overnight in alcoholic Bouin's solution, embedded in paraffin, cut at 7 μ, deparaffinized, hydrated to 70% alcohol, and treated as follows—stained 2 hours in a mixture consisting of: 0.2% orcein in 70% alcohol and 1% HC1 (conc.), 125 ml; 5% hematoxylin in absolute alcohol, 40 ml; 6% FeCl3 in water, 25 ml; and aqueous I2-KI (1:2:100), 25 ml—rinsed in distilled water until the excess stain was removed—differentiated in 1.2% FeCl3, 5-15 sec—washed in running water, 5 min—differentiation completed in 0.01% HC1 acid-alcohol, 1 min—a dip in 95% alcohol—distilled water, 2 min—0.25% aqueous metanil yellow, 5-10 sec—a 95% alcohol dip—dehydrated in absolute alcohol, xylene, and mounted in a resinous medium. The technic combines the orcein of Pinkus' stain and the hematoxylin mixture of Verhoeff into a single staining solution and gives sharp and reliable results for both coarse and extremely delicate elastic fibers. These stain purple; nuclei, violet; and background, yellow. The stain allows the use of formalin, Bouin's fluid and Zenker-formol fixation. The results have been consistent in other primates as well as in man.  相似文献

Fresh tissue slices fixed in chilled acetone for 1 hour and washed in distilled water for 10-30 minutes were incubated for 30-45 minutes at 37°C. in the freshly prepared incubating mixture: filtrate of a mixture of 8% sodium bicarbonate, 100 ml., and MnCl2·4H2O, 1 g. After washing in distilled water for 1 hour, they were dehydrated and embedded in paraffin. Sections were cut 15-20μU, deparaffinized, rinsed in absolute alcohol and placed in a 0.1% solution of potassium periodate for 48 hours at 37°C. The mounted sections were counterstained (if desired), dehydrated in alcohol, cleared in xylene (not carbol-xylene) and mounted in balsam. Many brown granules were produced on the sites of enzyme activity by this procedure. The results obtained seem to be in good agreement with previous findings by biochemical determinations.  相似文献

The following technic is suggested for staining cell walls in shoot apexes: After the usual preliminary steps through 50% ethyl alcohol, stain in 1 % safranin 0 for 24 hours. Rinse in tap water and place in 2% aqueous tannic acid for 2 minutes. After rinsing in tap water, stain for 2 minutes in 1 part Delafield's hematoxylin to 2 parts distilled water and rinse in tap water. Remove excess hematoxylin with acidified water (1 drop cone. HC1 in 200 ml. water), then place slides in 0.5% lithium carbonate for 5 minutes. Dehydrate through an ethyl alcohol series, then transfer from absolute alcohol to a saturated solution of anilin blue in “methyl cellosolve” for 5-10 minutes. Wash in absolute alcohol, rinse in a solution of 25% methyl salicylate, 33% xylene, 42% absolute ethyl alcohol and clear for 10 minutes in a solution of 2 parts methyl salicylate, 1 part xylene, 1 part absolute ethyl alcohol. Transfer through two changes of xylene and mount in “clarite” or suitable alternate. The resulting preparations will have clearly defined, dark-staining cell walls and will photograph well when “Super Panchro-Press, Type B” film (Eastman Kodak Co.) is used in conjunction with suitable Wratten filters.  相似文献

Maxilon blue RL, a basic monoazo dye developed by Geigy S. A., stains metachromatically acid mucopolysaccharide-containing elements in histological sections. This property is due to a chromatographically pure blue fraction which is the main component of the dye. The following routine has been developed for staining sections of formalin-calcium fixed paraffin-embedded tissues. Dewax in xylene and hydrate the sections through alcohol; stain in 0.05% aqueous Maxilon blue RL, from 30-60 sec; wash in distilled water; dehydrate either in tertiary butanol, 2 to 3 min, after removing excess water from the slide by blotting; or, rinse in 70% ethyl alcohol and dehydrate in 2 changes, 2 min each, of absolute alcohol; clear in xylene; mount in DPX or in Canada balsam. Acid mucopolysaccharides are colored red to violet; other basophilic elements, blue.  相似文献

Celloidin sections from formalin-fixed brain and spinal cord of primates are stored in 70% alcohol after cutting, soaked in 2% pyridine in 50% alcohol for 6-8 hr at 37 C, and transferred to 1% concentrated NH4OH in 50% alcohol 15-18 hr at 20-25 C. After washing and flattening, the sections are transferred to 1% silver protein solution containing 30 ml of 0.2 M H3BO3/100 ml. Impregnation is accomplished in 50 ml screw-top jars, 50 mm in diameter, which are filled to a depth of 35 mm, and have 1 gm of copper foil, 0.002 inch thick added. The foil is folded in loose accordion-fashion, pierced and threaded, cleaned in 5% HNO3, rinsed in distilled water, and suspended in the solution just above the sections by fastening the thread to the jar lid. The sections are impregnated for 24 hr at 37 C, rinsed in distilled water, reduced in a solution of 5% Na2SO3 and 1% hydroquinone for 10 min, washed in distilled water and toned in 0.2% gold chloride for 5 min. After rinsing in distilled water, the sections are transferred to 1% oxalic acid for 45-60 sec, washed in distilled water and placed in 5% Na2S2O3 for 5 min. Sections are then washed, dehydrated to 95% alcohol, cleared in terpineol, followed by 3 changes in xylene, and mounted.  相似文献

Cells derived from cultures of bone marrow or leucocytes were treated with hypotonic citrate solution, squashed in 45% acetic acid frozen with CO2 to allow removal of the cover glass without disturbing the smear, and stained by the following schedule: absolute alcohol, 5 min; coat with 0.2% parlodion and air dry; 70% alcohol, 5 min; distilled water, 5 min; stain 2-5 min in a mixture of 45 ml of a 0.3% solution of basic fuchsin in 5% phenol, 6 ml of glacial acetic acid, and 6 ml of 37% formaldehyde. Differentiate and dehydrate in absolute alcohol, clear in xylene and cover. The stain is durable for several weeks if slides are stored in darkness when not in use. Results resemble those obtained by Feulgen or aceto-orcein methods.  相似文献

A technic is described for producing critically stained preparations of phloem tissue. The preparations promise to be relatively stable. Sections of fixed unembedded or of embedded (paraffin or celloidin) phloem, cambium, and xylem are (1) stained in Foster's tannic acid-ferric chloride combination; (2) treated with 1% NaHCOg in 25% or 50% ethyl alcohol for 30 minutes; (3) stained in a saturated solution of lacmoid (made alkaline by adding a few ml. of 1% NaHCO3 in 25% alcohol) for 12 to 18 hours; (4) dehydrated and cleared in a series composed of 1% solution of NaHCOs in 50% ethyl alcohol, 80%, 95%, and absolute alcohol, equal proportions of absolute alcohol, clove oil, and xylene, and finally pure xylene; and (5) mounted in a neutral resin. Callose and lignified secondary walls are blue or blue-green in color, cellulose walls and stainable protoplasmic contents are generally light brown. The technic has been successful with sections from 5 to 40μ in thickness, and the staining has been satisfactory for both color and black and white photomicrography.  相似文献
