Methyl jasmonate (JA-Me) inhibited or retarded germination of Amaranthus caudatus seeds in darkness at 24°C, Ethephon, ACC and gibberellins (GA3 or GA4+7) partially or completely reversed this inhibition depending on the concentration of JA-Me applied. Both ethephon and the gibberellins were more effective than ACC. BA at 10–5 M, GA3 at 3×10–4 M or GA4+7 at 3×10–5 M partially or largely reversed the inhibition of Amaranthus caudatus seed germination due to JA-Me. BA or GA3 did not affect ethylene production and ACC oxidase activity in vivo in the presence of JA-Me before radicle protrusion. However, both increased ethylene production after 72 h of incubation, when the reversal of the JA-Me inhibition of seed germination was observed. AVG at 3×10–4 M decreased ethylene production when it was applied simultaneously with BA and JA-Me or GA3 and JA-Me, but it had no effect on seed germination. NBD almost completely reversed the stimulatory effect of BA, GA3 or GA4+7 on the germination of seeds in the presence of JA-Me. Exogenous ethylene reversed the inhibitory effect of NBD. The results indicate that action of endogenous ethylene is involved in the response of JA-Me inhibited seeds to BA or GAs.  相似文献

Experiments with Grand Rapids lettuce seeds (Lactuca sativa L.) maintained in darkness or irradiated with red light have shown that the inhibition of germination induced by low concentrations of ABA (2, 4, 6 μM) could be overcome by gibberellins (GA3 or GA4). The same results were obtained, although to a lesser extent, under the influence of two out of the four cytokinins tested (K and BAP) for seeds maintained in darkness. To suppress the block induced by higher concentrations of ABA (for example 8 μM), it was necessary to apply a cytokinin (K, BAP, Z or 2iP) and a gibberellin (GA4 or GA3) simultaneously, or a cytokinin following a red light treatment. Experiments conducted in darkness in which ABA (8 μM) was applied together with a cytokinin (BAP) and a gibberellin (GA4) showed that the gibberellin and the cytokinin played similar roles towards each other and towards ABA.  相似文献

Growth Regulators and the Phytochrome-Mediated Dormancy of Celery Seeds   总被引:2,自引:0,他引:2  
Seeds of five celery (Apium graveolens L.) cultivars germinated at 15°C in the light or dark but at 22°C only in the light. This light requirement was overcome by treatment with a mixture of the gibberellins GA4 and GA7 (GA4/7) but interactions of cytokinins, daminozide, ethephon, EDTA and N-phenyl-N′-4-pyridylurea (NC5392) with GA4/7 were observed. Varietal differences in response to GA4/7 concentration and the requirement for cytokinins were related to the upper temperature limits for germination of the different cultivars. Seeds of cultivars responding to low concentrations of GA4/7 appeared to contain less natural inhibitor than those requiring either high concentrations of GA4/7 or cytokinin in addition to low GA4/7. The cytokinin requirement for germination was partially removed by leaching the seeds with water. Interaction studies with applied hormones indicated that in seeds incubated in the light inhibition by abscisic acid was partially alleviated by N6-benzyladenine but not by GA4/7 application. The implications of these results are discussed in relation to the involvement of natural plant hormones in the dormancy mechanism of celery seeds.  相似文献

Uptake of gibberellic acid as a function of duration of exposure, external concentration, and seed lot was measured in seeds of yellow rocket (Barbarea vulgaris R. Br.) by means of lettuce hypocotyl bioassays of the acidic, basic, or neutral fractions of seed extracts and by uptake of 14C-GA3. In intact seeds, where mM levels of GA3 promoted only 25% germination, uptake was completed within 24 h of exposure. The maximum uptake was about 0.2% of external amount. Although germination promotion by GA3 differed among seedlots of yellow rocket, relative uptake (percentage of the external GA3) was nearly the same. The relative rate of uptake of GA3 was similar for scarified and intact seeds, but germination was promoted in scarified seeds by much lower levels of GA3 than in intact seeds. Total uptake in scarified seeds was much higher, however (about 10% of the external amount). In seeds imbibed in H2O, practically no endogenous GA-like activity was detected in either the acidic, basic, or neutral fractions. It was also apparent that intact seeds could take up quantities of GA3 that failed to promote germination, but were comparable to quantities that promoted germination in scarified seeds.  相似文献

Both dark and red light germination of lettuce seeds (cv. “Maikönig”) as well as their root and hypocotol elongation were inhibited when the seeds were sown in petri dishes together with a few seeds of Heracleum laciniatum Horn. This inhibition was not significantly counteracted by the presence of gibberellic acid (GA3) or/and 6-benzylaminopurine (BA). However, a large proportion of the lettuce seeds germinated abnormally (only cotyledons emerged) when treated with BA in the presence of Heracleum seeds. GA3 had alone no significant effect on abnormal germination, but it counteracted the effect of BA to some extent. The inhibitory effect of Heracleum seeds gradually disappeared during a moist incubation period of one to seven days in darkness at 25°C. When lettuce seeds were pre-incubated together with Heracleum seeds for one to five days the remaining, non-germinated lettuce seeds had lost their ability for subsequent germination in darkness in distilled water. This induced dark dormancy was to a great extent broken by red light, but not by GA3 or/and BA. H. laciniatum seeds inhibited the germination of Salix pentandra seeds and to some extent also the germination of radish but had no effect on the germination of spruce.  相似文献

The temperature-dependent primary dormancy of cv Florida 683 celery seeds in darkness was broken by GA4/7 (2 × 10-4 M) alone but other growth regulators such as BA, ethephon or daminozide were necessary to break dormancy of cv Lathom Blanching seeds in the presence of GA4/7 at this concentration. Although AgNO3 partially inhibited both the ethephon- and BA- induced germination of cv Lathom Blanching seeds in the presence of GA4/7 in the dark it did not affect the promotive action of daminozide. Ethephon did not overcome the inhibitory action of high concentrations of AgNO3 in the light. The ethylene synthesis inhibitor aminoethoxyvinylglycine (AVG) did not inhibit the germination of cv Lathom Blanching seeds induced by growth regulators in the dark or in the absence of growth regulators in the light. Fusicoccin (FC) did not break celery seed dormancy unless applied in the presence of GA4/7. Germination of cv Lathom Blanching celery seeds treated with GA4/7 at 16°C in the dark was inhibited by the K+ ionophore benzo-18-crown-C-6 (18-C-6) and in the presence of Ca2+ by the Ca2+ ionophore A23187; the 18-C-6 inhibition was reversed by BA.It is concluded that the involvement of gibberellin in celery seed dormancy is not dependent on endogenous ethylene and is directly or indirectly controlled through the action of other hormones on transmembrane ion fluxes.  相似文献

The critically endangered and perennial alpine endemic angiosperms, namely, Aquilegia nivalis, Lagotis cashmeriana and Meconopsis latifolia inhabit such habitats in the Kashmir Himalaya that are characterised by short growing season and heavy snow cover for about 3–4 months during winter season. The seeds of these species under natural conditions experience a long period of pre-chilling during winter prior to their germination in following spring season. Taking cue from such a requirement, present study investigated the effect of chilling and exogenous application of growth hormones, NO3 and NH4 + on total percent germination of otherwise deep-dormant seeds of these species, under alternate light/dark and continuous dark light regimes. Prolonged pre-chilling followed by treatment of seeds with different doses of GA3 had a pronounced stimulatory effect on the total germination percentage in all the three species. In fact, highest germination percentage in A. nivalis was recorded only when pre-chilled seeds were treated with 1.5 mM GA3 under alternate light/dark conditions. Likewise, germination of pre-chilled seeds of L. cashmeriana with ruptured seed coats was improved when treated with various concentrations of GA3. Seed germination in M. latifolia was also favourably influenced by treatment of pre-chilled seeds with GA3 or nitrogen applied either as NO3 or as NH4 + under alternate light/dark conditions. Treatment of seeds with kinetin (6-furfuryl-aminopurine) had no significant influence on germination percentage in any of the three species. Thus, prolonged chilling of seeds followed by their treatment with GA3 under alternate light/dark conditions are the requirements necessary for seed germination in these species.  相似文献

以紫斑牡丹种子为试验材料,考察不同浓度赤霉素(GA_3)处理下种子生根情况,同时对其种皮纹饰进行了扫描电镜(SEM)观察,并对胚乳结构进行了荧光倒置显微镜、透射电镜(TEM)观察,以揭示GA_3对紫斑牡丹种子下胚轴解除休眠过程中种子种皮、胚乳结构变化和生根的影响。结果表明:(1)GA_3处理能够促进紫斑牡丹种子生根,以300 mg/L GA_3处理的种子生根效果最好,与对照相比可提前14.66 d生根,生根率可达62.33%。(2)扫描电镜观察发现,紫斑牡丹种皮表面含有蜡质,种子萌发进程中蜡质明显不断增加,种皮表面网眼逐渐消失,种皮皱缩明显。(3)光学显微镜观察发现,GA_3处理的紫斑牡丹种子胚乳细胞中含有较多淀粉体和蛋白体,并随着种子萌发进程逐渐积累,可为种子萌发提供足够的养分。(4)透射电镜观察发现,GA_3处理的紫斑牡丹种子胚乳细胞内大分子营养物质不断降解,线粒体、内质网、高尔基体等细胞器相继出现,种子下胚轴休眠解除,细胞壁较薄后期消融,利于营养物质传送。研究认为,适宜浓度GA_3处理可有效加快紫斑牡丹种皮结构的改变和胚乳细胞内物质代谢进程,促进种子萌发,且300 mg/L GA_3处理紫斑牡丹种子24 h可以获得最好的生根效果。  相似文献

Impaired germination is common among halophyte seeds exposed to salt stress, partly resulting from the salt-induced reduction of the growth regulator contents in seeds. Thus, the understanding of hormonal regulation during the germination process is a main key: (i) to overcome the mechanisms by which NaCl-salinity inhibit germination; and (ii) to improve the germination of these species when challenged with NaCl. In the present investigation, the effects of ABA, GA3, NO3, and NH+4 on the germination of the oilseed halophyte Crithmum maritimum (Apiaceae) were assessed under NaCl-salinity (up to 200 mM NaCl). Seeds were collected from Tabarka rocky coasts (N-W of Tunisia). The exogenous application of GA3, nitrate (either as NaNO3 or KNO3), and NH4Cl enhanced germination under NaCl salinity. The beneficial impact of KNO3 on germination upon seed exposure to NaCl salinity was rather due to NO3 than to K+, since KCl failed to significantly stimulate germination. Under optimal conditions for germination (0 mM NaCl), ABA inhibited germination over time in a dose dependent manner, but KNO3 completely restored the germination parameters. Under NaCl salinity, the application of fluridone (FLU) an inhibitor of ABA biosynthesis, stimulated substantially seed germination. Taken together, our results point out that NO3 and GA3 mitigate the NaCl-induced reduction of seed germination, and that NO3 counteracts the inhibitory effect of ABA on germination of C. maritimum. Swelling of the hypocotyl base induced by 2,4-D in seedlings of marrow was much reduced if GA3 was also present in the incubation medium. At appropriate concentrations kinetin also counteracted this 2,4-D effect, although at higher concentrations appeared to be ineffective. GA3 was also able to overcome the inhibitory effects of 2,4-D on extension growth in the hypocotyl but kinetin was much less effective in this case. None of the treatments employed was able to alleviate the inhibition of radicle extension induced by 2,4-D.Ethephon induced similar responses in the seedlings to those resulting from 2,4-D treatment, while treatment with a mixture of 2,4-D and CoCl2 removed many of these growth abnormalities. These observations are interpreted as indicating that 2,4-D operates at least partly by stimulating the production of ethylene in the tissues. 2,4-D strongly inhibited the accumulation of endogenous gibberellin during the period of seedling development examined, but enhanced cytokinin levels during the later stages of the same period. The possibility of interactions at the biosynthetic level between gibberellins, cytokinins and ethylene and their involvement in the regulation of seedling development are discussed.  相似文献

Imbibition of celery (Apium graveolens L.) seeds at 32°C for up to 96 h lowered the upper temperature limit for germination. If this high temperature treatment was given in the light, these seeds germinated slightly earlier than those treated in the dark although the final percentage germination was similar for both treatments. The inhibitory effect of the high temperature treatment was completely removed by allowing the seeds to imbibe in a mixture of the gibberellins A4 and A7 (GA4/7) and partially removed by the cytokinin N6-benzylaminopurine (BA). GA4/7 was less effective when added before rather than after the high temperature treatment, whereas the opposite was true of BA. At constant temperatures more GA4/7 was required to promote germination as the temperature was raised but addition of BA reduced the concentration of GA4/7 required. A model is proposed for the control of celery seed germination by light and temperature through the action of endogenous cytokinins and gibberellins.  相似文献

Fourty-two kinds of benzaldehyde O-alkyloximes derived from benzaldehydes were prepared and their biological activities were investigated. Introduction of a fluorine or bromine atom to the benzene ring of the oximes enhanced their phytotoxic activity. The O-alkyloximes with a fluorine atom at the 3 or 4 position of the benzene ring were more active than the other oximes in the GA3-induced α-amylase induction inhibition test. In the transpiration test, 4-bromobenzaldehyde O-carboxylmethyloxime was the most active. The O-alkyloximes exhibited weak abscisic acid-like activity by inhibiting not only the germination, root growth and transpiration of higher plants but also GA3-induced α-amylase induction in embryoless barley seeds.  相似文献

The indigenous forage grasses Lasiurus scindicus and Panicum turgidum are candidate species for the restoration of degraded desert rangelands. The impact of five dormancy regulating chemicals on overcoming salinity-induced germination inhibition was assessed under the best germination conditions in the two species. Seeds were germinated in a series of NaCl concentrations: 0–200 mM NaCl for P. turgidum, and 0–300 mM NaCl for L. scindicus. Lasiurus scindicus seeds were more tolerant to salinity than those of P. turgidum. Twenty percent of P. turgidum seeds germinated in 100 mM NaCl and none in the higher levels, but 47.5% and 8.8% of L. scindicus seeds germinated in 100 and 200 mM NaCl, respectively. The five studied chemicals (fusicoccin, GA3, kinetin, nitrate and thiourea) did not succeed in improving germination of non-saline treated seeds of the two species, compared to the control, except thiourea in P. turgidum. The salinity-induced germination inhibition in P. turgidum was completely alleviated by the application of gibberellic acid (GA3), partially alleviated by the application of fusicoccin, kinetin and thiourea, but not affected by nitrate. In L. scindicus, the germination inhibition was completely alleviated by fusicoccin, GA3, nitrate and thiourea, but partially alleviated by kinetin. For using the two grass species in restoration of degraded rangelands affected by higher salinity, the results suggest using fusicoccin, GA3, nitrate and thiourea with L. scindicus and GA3 with P. turgidum seeds as a preseeding treatment can overcome the problem of reduced germination.  相似文献

The effect of the application of gibberellins to Tagetes minuta L. achenes (seeds) was determined at both 25°C, the optimal germination temperature, and 35°C, at which temperature the achenes are thermoinhibited. Both GA3 and GA4+7 accelerated germination at 25°C. Seed germination at 25°C was inhibited by paclobutrazol, but on subsequent application of GA4+7 rapid germination was induced. Following application of GA3 or GA4+7 to thermoinhibited seeds, a significantly higher final germination percentage was observed than in the distilled water control. However, endogenous gibberellin levels in germinating and thermoinhibited seeds did not differ significantly.  相似文献

Abstract

Light and growth-promoting compounds, such as gibberellic acids (GA3), are among the most important factors that can break physiological seed dormancy. Here, we investigate the effects of GA3 and light on germination of five species of Resedaceae that are known to have different levels of physiological dormancy. Seeds were incubated at 20/30?°C in both 12-hr photoperiod and complete darkness. To study the effect of growth hormone on germination, seeds were soaked for 24?h in different concentrations of GA3 before sowing. The annuals (Reseda aucheri and Oligomeris linifolia) and the perennial Ochradenus arabicus had deep physiological dormancy, and exogenous application of GA3 enhanced their germination in the light, but not in darkness; few or no seeds germinated in the dark in these species. Ochradenus aucheri and O. baccatus had intermediate and non-deep dormancy, respectively, and application of GA3 enhanced their germination in both light and darkness. Germination of the annual species was much slower than that of the shrubby perennials. Overall, these results indicate that conditions under which seed developed, matured and stored on maternal plants as well as incubation conditions should be taken into consideration when assessing germination behavior of the perennial species of Resedaceae.  相似文献

Gibberellin A4 (GA4) can substitute for light in the germination of Grand Rapids lettuce seeds. Seeds imbibed in [3H]GA4 do not convert this to other GAs prior to, or immediately following, visible germination: thus GA4 alone can promote radicle expansion. Abscisic acid inhibited [3H]GA4-induced germination, but did not significantly affect [3H]GA4 uptake or metabolism during germination. 6N-benzyladenine overcame the inhibitory effect of abscisic acid and increased [3H]GA4 uptake, although radicle emergence was delayed somewhat.  相似文献

Effects of the application of gibberellin A3 (GA3) on sprouting of rhizomes and germination of seeds were tested in five species of the genus Dioscorea which are considered to be Tertiary relicts growing disjunctively in the Appalachians, the Balkans, and the Caucasus. Applied GA3 at higher than 1 micro;M markedly inhibited sprouting of rhizomes of all species. However, inhibition of seed germination of these species by GA3 application was not observed. It is hypothesized that before disjunction in the Tertiary period GA-induced dormancy probably prevailed in rhizomes of all members of the genus Dioscorea.  相似文献

晚花杜鹃(late flower Rhododendron)是一类花期较晚的杜鹃品种,具有较高的观赏价值,被广泛应用于庭院种植和园林绿化。随着旅游以及经济发展的需要,开发和利用晚花杜鹃资源显得非常迫切。自然条件下晚花杜鹃萌发率相对较低,因此为提高晚花杜鹃萌发率,探究不同浓度赤霉素(GA_3)处理对五种杜鹃种子萌发的影响,该文以五种晚花杜鹃(小白杜鹃、大白杜鹃、桃叶杜鹃、长蕊杜鹃和九龙山杜鹃)为实验材料,通过不同浓度(0、300、400、500、600、700 mg·L~(-1))的GA_3对五种晚花杜鹃种子进行24 h浸种处理,测定其发芽指数、发芽势、发芽率以及成苗率等指标,分别确定五种杜鹃种子萌发的最适GA_3浓度,并对相同处理下五种杜鹃种子的萌发率和成苗率进行比较。结果表明:(1) GA_3浸种对五种晚花杜鹃种子萌发具有促进作用,在适当GA_3浓度下五种杜鹃种子的发芽指数、发芽势和发芽率均显著高于对照组,萌发时滞、萌发高峰期和持续萌发时间均较对照组浓度相对缩短。(2)大白杜鹃、桃叶杜鹃和九龙山杜鹃种子在GA_3浓度为600 mg·L~(-1)处理时各项萌发指标相对较好;长蕊杜鹃以GA_3浓度为700 mg·L~(-1)浸种处理萌发效果相对较好;小白杜鹃以GA_3浓度为400和700 mg·L~(-1)处理最好。因此,在杜鹃的栽培中,可以采用赤霉素GA_3处理法提高种子发芽率,缩短萌发时间。  相似文献

Suaeda salsa, a leaf succulent shrub in the family Chenopodiaceae, is one of the most important halophytes in China. Suaeda salsa produces dimorphic seeds (soft brown seeds and hard black seeds). Seeds of S. salsa were collected from the coastal salt flats near Huanghua City, China. Experiments were conducted to determine the salinity-alleviating effect of plant growth regulators, nitric oxide, nitrate, nitrite and light on the germination of dimorphic seeds of S. salsa. Brown seeds had a higher germination rate than black seeds in all experiments. Black seeds were more sensitive to salt in the absence of light in comparison to brown seeds. Brown seeds absorbed water more quickly in comparison to black seeds and were found to be more tolerant of salt stress. Our results showed that 1-aminocyclopropane-1-carboxylate (ACC, the immediate precursor of ethylene), nitrite, GA4 and BA improved seed germination in the presence of salt. However, nitrate, GA1, GA3 failed to alleviate salt stress. ABA inhibited seed germination and seedling growth. Possible mechanisms involved in the alleviation of salt stress in S. salsa seeds and the ecological adaptation of the seeds to the environment are discussed.  相似文献
