不同饵料对史氏鲟仔鱼消化酶活性的影响   总被引:12,自引:0,他引:12  
用活饵料(水丝蚓)、商品饵料、添加消化酶制剂的商品饵料、活饵料与商品饵料交替使用(对照组)来投喂10~40日龄(DPH,Day_past_hatch)的史氏鲟仔鱼。其中对照组仔鱼体内蛋白酶活性和商品饵料组及添加消化酶组的差异极显著(P<0.01),与活饵料组差异不显著(P>0.05),活饵料组仔鱼蛋白酶活性与商品饵料组差异极显著(P<0.01),与添加消化酶组差异显著(P<0.05),商品饵料组和添加消化酶组之间仔鱼的蛋白酶活性差异不显著(P>0.05);各饵料组仔鱼的淀粉酶活性差异均不显著(P>0.05);活饵料组和对照组仔鱼的脂肪酶活力与商品饵料及添加消化酶组仔鱼的脂肪酶活性差异极显著(P<0.01),在这两组之间,仔鱼的脂肪酶活性差异均不显著(P>0.05)。  相似文献

研究了不同温度下虎斑颈槽蛇胃、胰腺、小肠的蛋白酶、淀粉酶、脂肪酶的活性。结果表明,随着温度的升高,各种酶的活性在一定范围之内均表现为升高。在不同组织中同一种酶活性存在差异。3种组织中,蛋白酶的最适温度均为40℃;胃和胰腺淀粉酶的最适温度为35℃,小肠淀粉酶为30℃;胰腺脂肪酶的最适温度为30℃,小肠脂肪酶为35℃。  相似文献

兰州鲇与鲇消化系统的形态学及组织学比较研究   总被引:1,自引:0,他引:1  
为探究黄河濒危鱼类兰州鲇(Silurus lanzhouensis)消化系统的形态学和组织学结构特点,以鲇(Silurus asotus)为对照,对兰州鲇消化系统形态学和组织学进行了深入研究。结果表明:(1)兰州鲇与鲇的消化道和消化腺形态相似,具有肉食性鱼类的特征。兰州鲇消化道较短,有发达的“U”型胃,胃内皱褶明显,无幽门盲囊,肠道短且粗,可分为前肠、中肠和后肠三部分,前肠粗大,后肠较细。两种鲇属鱼类都有独立致密的肝脏和胰脏。(2)兰州鲇的比肠长显著大于鲇(P < 0.05),比胃重、比肝胰脏重显著低于鲇(P < 0.05),但二者的比肠重无显著性差异(P>0.05)。(3)兰州鲇胃的皱襞幅度小于鲇,且环肌层比兰州鲇薄。兰州鲇与鲇前肠的肠黏膜均形成了大量皱襞,肠黏膜、褶皱粗大,但鲇的褶皱分支较细密。兰州鲇与鲇的后肠与前肠相比,肠腔变小,褶皱数量明显减少,高度降低。黏膜层分布有杯状细胞和柱状细胞。兰州鲇与鲇的肝脏肝小叶间缺少结缔组织,分界不明显,而兰州鲇肝细胞的密度大于鲇。综上所述,兰州鲇与鲇的消化系统相似,均符合肉食性鱼类消化系统特征,结合消化生理等研究结果,表明兰州鲇的消化能力弱于鲇,这可能是在自然情况下兰州鲇的分布区域及适应性不及鲇的原因之一。  相似文献

本研究的目的在于认识兰州鲇(Silurus lanzhouensis)染色体组特征,为兰州鲇的细胞遗传学和人工育种研究提供基础参考资料。以兰州鲇培养肾细胞为材料,采用空气干燥法制备染色体玻片标本,分析了兰州鲇的染色体组型。兰州鲇的染色体数2n=58,核型公式为2n=20m+18sm+16st+4t,染色体总臂数(NF)为96。通过比较分析认为,兰州鲇在鲇属鱼类中属于较为原始的类群。  相似文献

温度对南疆沙蜥和密点麻蜥消化酶活性的影响   总被引:1,自引:0,他引:1  
为探究不同温度对南疆沙蜥Phrynocephalus forsythii和密点麻蜥Eremias multiocellata消化酶活性的影响,本研究测定了不同温度下南疆沙蜥和密点麻蜥的肝脏、胃及十二指肠的消化酶——淀粉酶(碘-淀粉比色法)、脂肪酶(比浊法)及胰蛋白酶(紫外线吸收法)的活性。结果显示,南疆沙蜥和密点麻蜥消化酶活性在4~45℃随着温度升高呈现先上升、后下降的趋势;40℃驯化的南疆沙蜥、35℃驯化的密点麻蜥肝脏、胃及十二指肠的淀粉酶、脂肪酶及胰蛋白酶活性最高。南疆沙蜥和密点麻蜥消化酶活性具有温度依赖性,这与机体在不同温度下的生理机能是密切相关的。同时,2种蜥蜴的消化酶活性最大时的驯化温度明显不同,这与它们偏好的环境温度密切相关。  相似文献

兰州鲇与鲇、黄河鲤肌肉品质比较研究   总被引:2,自引:0,他引:2  
为深入探究兰州鲇(Silurus lanzhouensis)肌肉品质特性, 对黄河中兰州鲇、鲇(Silurus asotus)和黄河鲤(Cyrinus carpio)肌肉品质指标进行了测定。结果表明: 兰州鲇肌肉的pH、滴水损失、熟肉率、胶原蛋白、肌原纤维耐折力、黏附性、内聚性、胶黏性、咀嚼性、弹性和回复性等指标与鲇差异不显著(P>0.05), 失水率、肌纤维直径和硬度在二者间差异显著(P<0.05); 兰州鲇肌肉的pH、肌纤维直径、硬度、黏附性、回复性、胶黏性和咀嚼性与黄河鲤差异不显著(P>0.05), 肌肉滴水损失、熟肉率、失水率、胶原蛋白、肌原纤维耐折力、内聚性和弹性在二者间差异显著(P<0.05)。相关性分析表明, 兰州鲇肌肉咀嚼性与硬度呈极正相关, 其关系式为: y=0.3651x+25.339(R=0.97); 回复性与内聚性呈极正相关, 其关系式为: y=0.6279x-0.0929(R=0.91); 胶黏性与硬度呈极正相关, 其关系式为: y=2.4104x-41.155(R=0.97)。对兰州鲇7个质构指标进行了主成分分析, 提取出3个主成分, 累计方差贡献率为96.08%, 其中硬度是影响兰州鲇质构特性的主要因素。黄河中兰州鲇和鲇作为鲇属鱼类中形态十分相似的两个物种, 在肌肉品质特性上既有很大的相似性, 又有一定的不同, 这可能与这两种鱼种质特性的异同有关。  相似文献

采用酶学分析方法,测定了温度和pH对橄榄蚶蛋白酶、淀粉酶、纤维素酶活力的影响。结果表明:温度和pH显著影响橄榄蚶蛋白酶、淀粉酶和纤维素酶的活力;蛋白酶、淀粉酶和纤维素酶的最适温度分别为60℃、40℃和30℃~40℃,最适pH分别为3.6、5.2和6.0。橄榄蚶3种消化酶活性大小为淀粉酶>蛋白酶>纤维素酶。  相似文献

采用毒性试验方法,研究了安全浓度(1.288 mg/L)条件下亚砷酸钠(NaAsO2)对兰州鲇(Silurus lanzhouensis)脑、鳃、肝脏、肌肉4种组织中6-磷酸葡萄糖脱氢酶(G-6-PDH)和乳酸脱氢酶(LDH)活性,以及RNA和蛋白质含量的影响。结果表明,染毒21d时,As(Ⅲ)可显著降低4种组织中G-6-PDH和LDH活性、RNA和蛋白质含量(P0.05)。撤毒后21d,除脑和肝组织中蛋白质含量未恢复到对照组水平(P0.05),肝脏中G-6-PDH活性超过了对照组水平(P0.05)外,其余各组织中G-6-PDH和LDH活性、RNA和蛋白质含量均可恢复到对照组水平(P0.05)。以上结果表明,As(Ⅲ)对兰州鲇组织中代谢酶活性具有明显的抑制作用,可致组织细胞RNA损伤和可溶性蛋白质减少,但这种影响是可逆的,撤毒后一定时间内可恢复到正常水平。    相似文献

针对目前兰州鲇(Silurus lanzhouensis)种质资源救护保存和良种选育等研究工作中面临的亲子鉴定及系谱管理等问题,研究应用微卫星荧光标记多重PCR与自动测序分型技术,建立了2组四重PCR和2组三重PCR体系,并成功应用于3个家系亲子鉴定中。利用Cervus v.3.0软件对110尾兰州鲇进行遗传多样性分析,结果显示:研究筛选的14个微卫星标记的平均观测杂合度(Ho)为0.750,平均期望杂合度(He)为0.667,平均多态信息含量(PIC)为0.624,具有丰富的遗传多样性。对已知系谱信息的3个兰州鲇家系的90尾子代和20尾候选亲本进行亲子鉴定分析,结果表明,双亲基因型未知累积排除概率(CE-1P)、单亲基因型已知累积排除概率(CE-2P)和双亲基因型已知累积排除概率(CE-PP)分别为0.99753092、0.99983971和0.99999964。4组多重PCR累积模拟鉴定率为100%,累积实际鉴定率为83%。采用50尾个体进行双盲验证,利用MEGA7.0对3个家系50尾个体进行聚类分析,结果表明同一家系94%的个体聚类分析...  相似文献

在模拟人体胃肠环境中研究不同发酵时期的冠突散囊菌黑茶发酵液对淀粉酶、脂肪酶和蛋白酶活性的影响。结果表明冠突散囊菌黑茶发酵液能显著提高α-淀粉酶、蛋白酶活力,并有效抑制脂肪酶活力。冠突散囊菌发酵液利于淀粉、蛋白质消化吸收,抑制脂肪分解吸收,为解释茯砖茶的保健功能提供了理论依据。  相似文献

The proteolytic activity of 34 commercial lipase preparations (CLP) was determined using a labeled casein substrate. Only three CLP were free from proteolytic activity. Porcine pancreatic lipases exhibited levels of proteolytic activity comparable to or greater than that of a reference porcine trypsin. Bacterial lipases contained up to 10% of the proteolytic activity of commercial trypsin. Proteolytic activities in lipases from fungal species were present at low levels (<1% of the activity in trypsin). Among preparations of fungal origin, lipases from Aspergillus niger and Mucor javanicus were highest in proteolytic activity; Aspergillus oryzae and Pseudomonas cepacia lipases were lowest. Proteins in CLP were separated by non-denaturing PAGE; between 4 and 17 protein bands in the range &#104 6.5- &#83 200 kDa were observed. With the exception of a single pair of Rhizomucor miehei lipases, the distribution of apparent molecular weights (AMW) was unique to each preparation. Bands of caseinolytic activity in commercial lipases were visualized by applying a zymographic technique. CLP contained between 0 (P. cepacia lipases) and 6 (porcine pancreas lipase and Rhizopus oryzae lipase) discrete proteolytic bands. Common themes of proteolytic AMW emerged, including 21-23 kDa and 30-35 kDa bands.  相似文献

采用常规方法测定了鲇(Silurusasotus)的消化道指数,其比肠长、比胃重、比肠重、比肝胰脏重和比内脏重分别为0726±0087、0027±0015、0013±0008、0022±0004和0081±0041。体重与体长的回归方程Y=00124X2.8451(r=09940,P<001);鲇的胃、前肠、中肠和后肠中消化液的pH值分别为48~62、66~70、69~75、72~77,且消化道各部分具有较高的蛋白酶和淀粉酶活性,蛋白酶的活性为前肠>胰脏>胃>中肠>后肠;而淀粉酶的活性则为胰脏>胃>前肠>中肠>后肠;肌肉中粗蛋白的含量1881%,粗脂肪含量152%;必需氨基酸含量占氨基酸总量的4451%,鲜味氨基酸含量占氨基酸总量的4598%,综合肌肉的常规营养成分和氨基酸分析的结果,鲇是一种营养价值较高的优质鱼类。  相似文献

Nine healthy female subjects were studied when exposed to the natural light-dark cycle, but living for 17 “days” on a 27h day (9h sleep, 18h wake). Since the circadian endogenous oscillator cannot entrain to this imposed period, forced desynchronization between the sleep/activity cycle and the endogenous circadian temperature rhythm took place. This enabled the effects of activity on core temperature to be assessed at different endogenous circadian phases and at different stages of the sleep/activity cycle. Rectal temperature was measured at 6-minute intervals, and the activity of the nondominant wrist was summed at 1-minute intervals. Each waking span was divided into overlapping 3h sections, and each section was submitted to linear regression analysis between the rectal temperatures and the total activity in the previous 30 minutes. From this analysis were obtained the gradient (of the change in rectal temperature produced by a unit change in activity) and the intercept (the rectal temperature predicted when activity was zero). The gradients were subjected to a two-factor analysis of variance (ANOVA) (circadian phase/ time awake). There was no significant effect of time awake, but circadian phase was highly significant statistically. Post hoc tests (Newman-Keuls) indicated that gradients around the temperature peak were significantly less than those around its trough. The intercepts formed a sinusoid that, for the group, showed a mesor (±SE) of 36.97 (±0.12) and amplitude (95% confidence interval) of 0.22°C (0.12°C, 0.32°C). We conclude that this is a further method for removing masking effects from circadian temperature rhythm data in order to assess its endogenous component, a method that can be used when subjects are able to live normally. We suggest also that the decreased effect of activity on temperature when the endogenous circadian rhythm and activity are at their peak will reduce the possibility of hyperthermia.  相似文献

About 34 wild fungal species associated with edible oil mill wastes were isolated by the serial dilution technique. Methods for rapid screening of fungal species against production of extracellular enzymes such as amylase, protease, cellulase, and lipase are reported. Among all the species, Aspergillus versicolor exhibited high amlylolytic and gelatinolytic activity, whereas Penicillium citrinum showed only high amylolytic activity. Maximum cellulolytic activity was recorded for Absidia corymbifera, As. niger, Cunninghamella echinulata, Curvularia lunata, Fusarium solani, Mucor racemosus, Paecilomyces variotii, and Syncephalastrum racemosum. The fungal species Ab. corymbifera, As. fumigatus, As. japonicus, As. nidulans, As. terreus, Cun. verticillata, Cur. pallescens, F. oxysporum, Geotrichum candidum, M. racemosus, Pe. citrinum, Pe. frequentans, Rhizopus stolonifer, and Trichoderma viride exhibited maximum lipase activity. This study confirms the isolated fungi present on a wide range of substrates in the ambient environment, and these results could provide basic data for further investigations on fungal extracellular enzymes.First two authors equally contributed to this work  相似文献

本研究在不同p H和温度条件下以离体方式对野生棘胸蛙(Quasipaa spinosa)不同消化器官的蛋白酶活力进行了分析。结果表明,食道、胃、胰的蛋白酶活力受到酶体系p H的显著影响(P0.05),且随p H升高呈现典型的单峰型活力曲线。食道和胃的蛋白酶活力在p H为1.5时达到峰值,胰的酶活力在p H为9.6时达到峰值,而肠道酶活力在p H为7.4时达到峰值。各消化器官蛋白酶活力具有明显的温度依赖性(P0.05),酶活力随温度升高也均呈典型的单峰型活力曲线,不同消化器官的最大酶活力温度分别为,食道50℃、胃50℃、胰45℃、前肠45℃、后肠45℃、直肠45℃。在最大酶活力的p H和30℃条件下,各消化器官蛋白酶活力由高到低依次为胰、食道、胃、直肠、前肠、后肠。由此可见,蛋白酶在棘胸蛙消化系统的分布具有明确的规律性,且不同来源的蛋白酶需要在特定p H和温度下才能表现出最大的反应活性。  相似文献
