Summary A detailed histochemical investigation was carried out on rind, cortical and medullary hyphae of sclerotia ofSclerotinia minor Jagger. Four developmental stages, including mature sclerotia, were studied. The walls and septa of all hyphae contained chitin and -1,3 glucans, while those of the rind contained in addition, a melanin-like pigment. An extracellular matrix, which accumulated around cortical and medullary hyphae, consisted primarily of -1,3 glucans, although another polysaccharide, which could not be identified by histochemical methods, was also present. Phenolic material was deposited around the extracellular matrix and in the few interhyphal spaces that remained at maturity. Glycogen was present throughout the cytoplasm of hyphae of the cortex and medulla, at all stages of their differentiation. Polyphosphate granules were laid down within small vacuoles and as sclerotia matured, became most common in the cortical region. Protein bodies developed rapidly in cortical and medullary hyphae until at maturity, they were the most obvious interhyphal feature. These bodies were either round or elongated in shape, the elongated ones often lying parallel to the long axis of the hyphae, and in close association with strands of endoplasmic reticulum. Summary In freeze-substituted sclerotia stained with aqueous toluidine blue O, metachromatic material was found throughout the cytoplasm in discrete granules. It was also distributed evenly throughout spherical and elongate protein bodies. This material stained at low pH and was extracted by cold acid, indicating that it was polyphosphate. Retention of metachromatic material was much greater than previously reported in chemically fixed, conventionally processed sclerotia. X-ray microanalysis of dry-cut, unstained sections of freeze-substituted sclerotia confirmed that phosphorus was distributed evenly throughout the protein bodies and was not localised in discrete granules but phosphorus levels in the cytoplasm were very low. It is concluded that polyphosphate is lost during conventional preparation procedures but retained in dry-cut, unstained sections of freeze-substituted material. However, when freeze-substituted sections were stained with toluidine blue O, water soluble polyphosphate was extracted and subsequently precipitated in the cytoplasm as polyphosphate granules. Therefore it is considered that polyphosphate granules are an artefact, and that protein bodies are the major site for storage of phosphorus in this fungus.Abbreviations STEM scanning transmission electron microscope - ER endoplasmic reticulum  相似文献

Sclerotia were formed in agar culture by a fungus with clamp connections isolated from rice husks at Tsukuba, Japan. The sclerotia were brown, globose to ellipsoidal, small, up to 200 μm in diameter, and composed of external rind tissue and internal medulla tissue. Such tiny sclerotia have not been commonly reported among basidiomycetous fungi in the literature. The fungus was identified as Coprinopsis cinerea on the basis of morphological characteristics together with molecular analyses. Three reference strains of C. cinerea formed sclerotia similarly under identical cultural conditions.  相似文献

The morphological characteristics of sclerotia were induced in cultures of the fungus Grifola umbellata by introducing an unidentified companion fungus were studied by light microscope, scanning and transmission electron microscope (SEM and TEM). Light microscope and SEM investigations of developing sclerotia revealed that aerial mycelial hyphae diminished with age, and mature sclerotia had two tissue layers, the rind and medulla. The medulla was comprised of thin and thick-walled hyphae of varying diameter. The thick-walled cells always formed below the hyphal tips. Retraction of the cytoplasm was accompanied by the thickening of cell wall. There were crystalline initials in the newly formed sclerotium. Crystalline initials were always formed in the tip of medullary hyphae and were not of regular shape. A series of changes occurred in the cells in which the crystalline initials would be formed, such as enlargement of size, formation of one or several large vacuoles. Crystalline initials developed via amorphous materials in the cytoplasm deposited in the vacuoles. At last crystalline initials was released by degradation of the cell wall.  相似文献

The ultrastructure of the cyanobacterium Spirulina platensis was studied in relation to temperature, light intensity and nitrate concentration. The organism was able to grow in media supplied with nitrate in concentrations up to 250mm. High nitrate concentrations increased the yield and growth rate at temperatures above 35°C. Occurrence, distribution and abundance of cyanophycin granules, polyglucan granules, cylindrical bodies, carboxysomes and mesosomes varied widely in relation to the factors studied. At low temperatures (up to 17°C) cyanophycin was the abundant organelle, especially at high nitrate concentrations, whereas in the temperature range 17–20°C polyglucan was found in large quantities particularly at low nitrate concentrations. Special attention was paid to the cylindrical bodies, the ultrastructure of which was dependent on temperature. Three types of ultrastructure were distinguished each with several possible shapes.  相似文献

Summary Sclerotia ofSclerotinia minor produced in culture are permeable to the apoplastic tracer sulphorhodamine G (SR) in early stages of their development, but become impermeable as the rind differentiates at the onset of maturation. Reduction in permeability corresponds with deposition of the dark brown pigment in the rind cell walls rather than initiation of the rind as a distinct surface layer. Fluorochrome permeation into cut sclerotia indicates that, while the rind is the primary barrier, the walls and extracellular matrix of the cortex and medulla of mature sclerotia also impede SR movement. Some cells take up fluorochrome into the protoplast. This indicates enhanced proton pumping activity at the cell surface, which suppresses ionisation of the fluorochrome, allowing it to cross the plasma membrane and accumulate in the hyphae. In intact sclerotia such hyphae are very rare and were detected only at one stage of development. However, in cut sclerotia at the two earliest stages of development most of the hyphae near the cut surface accumulated SR and it is possible that this is due to proton pumping activity induced by wounding.Abbreviations ECM extracellular matrix - SR sulphorhodamine G  相似文献

Summary The respiration of sclerotia ofS. rolfsii was investigated using the Warburg constant-volume respirometer to measure oxygen uptake. The effects of age of sclerotia, pH, and temperature were studied. Sclerotia produced on prune agar were ideal for respirometric studies, being uniformly round and of approximately equal size. On a dry weight basis, the respiration rates of sclerotia were considerably less than those of vegetative mycelium. Sclerotia showed a decrease in respiration with increasing age. This was accompanied by morphological changes in the outer hyphal rind of the sclerotium during maturation. The respiration rate of sclerotia was approximately the same at 30° and 40° C, but was significantly lower at 45° C. Respiration of sclerotia was not markedly affected by normally encountered hydrogen-ion concentrations. However, a pH of 8.0 markedly repressed oxygen uptake. Sclerotia produced in rye grain cultures were chemically analyzed. The nitrogen content was 4.7 %, the petroleum-ether-soluble lipid content was 0.7 %, and the crude glycogen content was 14.2 % of the oven dry weight of the sclerotia.Contribution No. 345 from The Department of Botany. The effect of insulin (I), cortisol (F) and prolactin (P) on the ultrastructural morphology of epithelial cells of cultured mammary explants from virgin ovariectomized (OV-X) goats were studied. The epithelial cells showed little structural organization and were devoid of fat droplets and secretory protein granules at zero time of culture. The cytoplasm contained few profiles of smooth and rough endoplasmic reticulum and the Golgi apparatus was rudimentary. After being cultured in Waymouth's medium without added hormones the epithelial cells were indistinguishable from epithelial cells of uncultured explants. The addition of I induced changes mainly in the appearance of nucleoli. The nucleoli were enlarged and fibrillogranular areas with light spaces were observed. The most obvious cytological changes of epithelial cells of explants cultured in the presence of I and F are translocation of the nucleus into the basal cytoplasm, increase of rough endoplasmic reticulum, an increase in the size of the Golgi apparatus, presence of one or two lipid droplets and in some cells vacuoles with protein granules were present. Mitochondria were more abundant. The epithelial cells of explants cultured in the presence of I, F and P were characterized by the polarization of organelles within the cytoplasm and by the formation and release of protein granules and small and large fat droplets. The cell nucleus was in the basal cytoplasm, the Golgi apparatus was supranuclear. The rough endoplasmic reticulum was extensively developed and formed large sacs. Summary Brassica napus cv. Topas microspores, isolated and cultured near the time of the first pollen mitosis and subjected to a heat treatment of 24 h, can be induced to develop into haploid embryos. This is a study of microspore structure during induction and embryo determination. Early during the 32.5 °C incubation period the nucleus moved away from the edge of the cell, and granules, 30 to 60 nm in diameter, appeared in the mitochondria and as a cluster in the cytoplasm. Cells divided symmetrically and at the end of the heat treatment, acquired the features of induced bicellular structures described previously. The features persisted as the cells divided randomly within the exine for 4–7 days following heat induction. Multicellular structures released from the exine underwent periclinal divisions resulting in protoderm differentiation of the globular embryo, thus determining embryo development. The cytoplasm of early heart-stage embryos contains abundant polyribosomes. Non-embryogenic development was indicated by large accumulations of starch and/or lipid and thickened cell walls or an unorganized pattern of cell division following release of the multicellular structures from the exine. Summary During imbibition ofPhoenix dactylifera embryos, all cotyledon cells show the same changes: protein and lipid bodies degrade, smooth endoplasmic reticulum (ER) increases in amount, and dictyosomes appear. At germination, the distal portion of the cotyledon expands to form the haustorium. At this time, epithelial cells have a dense cytoplasm with many extremely small vacuoles. Many ribosomes are present along with ER, dictyosomes, and mitochondria. The parenchyma cells have large vacuoles and a small amount of peripheral cytoplasm. Between 2 and 6 weeks after germination, epithelial cells still retain the dense cytoplasm and many organelles appear: glyoxysomes, large lipid bodies, amyloplasts, large osmiophilic bodies, and abundant rough and smooth ER which appear to merge into the plasmalemma. A thin electron-transparent inner wall layer with many small internal projections is added to the cell walls. Starch grains appear first in the subsurface and internal parenchyma and subsequently in the epithelium. Lipid bodies, glyoxysomes, protein, and osmiophilic bodies occur in the epithelial and subepithelial cell layers but not in the internal parenchyma. At 8 weeks after germination, the cytoplasm becomes electron transparent, vacuolation occurs, lipid bodies and osmiophilic bodies degrade, and the endomembranes disassemble. After 10 weeks, the cells are empty. These data support the hypothesis that the major functions of the haustorium are absorption and storage.  相似文献

THE SURVIVAL OF FUNGAL SCLEROTIA UNDER ADVERSE ENVIRONMENTAL CONDITIONS   总被引:4,自引:0,他引:4  
1. Sclerotia have been found to survive under adverse environmental conditions for long periods. Their viability is retained at low and subzero temperatures, while it is progressively reduced at temperatures above about 15° C. Dry conditions and lack of competition from other organisms favour survival. Depletion of food reserves results in loss of resistance to environmental factors. 2. The majority of sclerotia consist of an outer layer of thickened, close-fitting, often pigmented cells that form a rind over a tissue of closely interwoven hyphae. A cortex and medulla is often discernible. 3. Features of sclerotia that are important in overcoming the harmful effects of desiccation and extremes of temperature include: the presence of a rind and sometimes an additional covering of either fungal or host tissue; the compact nature of the sclerotium; miscellaneous protectants on the surface of and in the hyphae; melanized hyphal walls; high intracellular osmotic concentrations; slow air-drying to lower moisture content. Also the buffering action of the soil and the protection afforded by vegetation and plant debris are important. 4. The active exudation of water, the accumulation and secretion of soluble carbohydrates during sclerotium development and maturation, and the formation of large amounts of mucilage are of significance in resistance to desiccation, extremes of temperature and radiations. 5. The nature and the pigmentation of the rind together with the site of sclerotium development may give protection against harmful radiations. 6. The loss of sclerotium viability caused by toxic chemicals in the soil and atmosphere or by the enzymic activities of other organisms is reduced by the rind and the melanization of hyphal walls. 7. Survival under starvation conditions is achieved by the accumulation of abundant food reserves (lipids and/or carbohydrates) and a low level of metabolic activity. In the presence of a compatible host exogenous sources of energy are sometimes used. 8. Although the sclerotium is highly adapted to survive adverse conditions for long periods, sometimes micro-environmental conditions may be so severe that only a few sclerotia can retain their viability. However, even a few such comparatively large bodies, rich in food reserves, have a considerable inoculum potential.  相似文献

Immature zygotic embryos from Howea forsteriana Becc. were cultured on the Murashige and Skoog medium, supplemented with myo-inositol, thiamine-HCl and activated charcoal, in the absence of growth regulators. The fruits were stored for 4 weeks at +4°C and at –18°C. The excised embryos from the fruits stored at +4°C developed into plantlets, showing a well developed primary root, after 40 days in culture, while those excised from fruits stored at –18°C exhibited no growth.This is the first time that in vitro culture and plantlet regeneration from immature embryos of Howea forsteriana has been obtained.  相似文献

Summary The morphology of the Golgi apparatus of Amoeba proteus can be influenced by substances inducing pinocytotic activity as well as by extreme temperatures. During the ingestion of a solution of 0.5% egg white the number of Golgi bodies decreases from 100% measured in control cells to 82% measured in cells showing induced pinocytosis. Simultaneously the ratio of the surface area of the cisternae at the proximal face to that of the vesicles at the distal face of single dictyosomes remains constant (1.74–1.72).The decrease and increase of the temperature of the culture medium to 4° C and 30° C respectively, causes the disappearance of most of the dictyosomes. After keeping the cells for 3–10 h at these temperatures the number of Golgi bodies was only 5–10% of the controls. A continued treatment with cold or warm culture medium leads to a partial reorganization of dictyosomes. After 15 h the number of Golgi bodies counted per cell returned to 57% at 4° C and 38% at 30° C. The ratio of the surface area of the Golgi cisternae to the surface area of the Golgi vesicles also alters under the influence of extreme temperatures. The values measured after treating the cells for 3 h, 4 h 10 h and 15 h at 4° C and 30° C amounted to 0.75, 0.85, 1.14 1.53 and 0.93, 0.38, 0.88, 1.60, respectively, compared to 1.72 of control amoebae.The different values of the ratio of the surface area of cisternae to that of vesicles indicate that there are strong morphological changes of single dictyosomes.  相似文献

Summary The ileal absorptive cells of suckling rats exhibit high levels of endocytic activity being engaged in nonselective uptake of macromolecules from the intestinal lumen. The apical cytoplasm usually contains an extensive network of small, membrane-limited tubules (apical tubules: AT), in addition to newly formed endocytic vesicles and large endocytic vacuoles. To determine whether the AT are directly involved in the endocytic process by carrying the tracer into the cell, we have analysed movements of the apical cell membrane of the ileal absorptive cells by using a membrane-bound tracer (horseradish peroxidase-labelled cancanavalin-A: Con-A HRP). The ileal absorptive cells were exposed in vitro to Con-A HRP for 10 min at 4° C, incubated for different times in Con-A free medium at 37° C, and prepared for electron microscopy. After 1 min incubation at 37° C, invaginations of the apical cell membrane, including coated pits, and endocytic vesicles were labelled with HRP-reaction product, whereas the AT and large endocytic vacuoles were negative. After 2.5 min, almost all the large endocytic vacuoles were labelled with reaction product, which was seen in their vacuolar lumen and along the luminal surface of their limiting membrane. A few AT with reaction product were seen in the apical cytoplasm; they were in frequent connection with the reaction-positive large endocytic vacuoles. With increasing incubation time, the number of the labelled AT increased. Thus, after 15 min at 37° C, the apical cytoplasm was fully occupied by the reaction-positive AT. The ends of these AT were often continuous with small spherical coated vesicles. No reaction product was detected in the Golgi complex at any time after incubation. These observations indicate that the AT located in the apical cytoplasm probably originate by budding off from the large endocytic vacuoles, rather than being involved in the process of endocytosis.  相似文献

Large (up to 100 m long), loosely coiled, free-living spirochetes with variable diameters (from 0.4 to 3 m in the same cell) were seen at least 40 times between August 1990 and January 1993. These spirochetes were observed in mud water and enrichment media from highly specific habitats in intertidal evaporite flats at three disjunct localities, one in Spain and two in Mexico. All three are sites of commerical saltworks. Associated with Microcoleus chthonoplastes, the large spirochetes from Spain display phototaxis and a composite organization. Shorter and smaller-diameter spirochetes are seen inside both healthy and spent periplasm of larger ones. Small spirochetes attached to large ones have been observed live. From two to twelve spirochete protoplasmic cylinders were seen inside a single common outer membrane. A distinctive granulated cytoplasm in which the granules are of similar diameter (20–32 nm) to that of the flagella (26 nm) was present. Granule diameters were measured in thin section and in negatively-stained whole-mount preparations. Based on their ultrastructure, large size, variable diameter, number of flagella (3 to 6), and phototactic behavior these unique spirochetes are formally named Spirosymplokos deltaeiberi. Under anoxic (or low oxygen) conditions they formed blooms in mixed culture in media selective for spirochetes. Cellobiose was the major carbon source in 80% seawater, the antibiotic rifampicin was added, mat from the original field site was present and tubes were incubated in the light at from 18–31 °C. Within 1–2 weeks populations of the large spirochete developed at 25 °C but they could not be transferred to fresh medium.  相似文献

Mycelial growth and fruit-body production of an ectomycorrhizal Boletus sp. were examined in pure culture. Mycelia of the strain Bo1 grew well on a medium consisting of sawdust and barley grains. Mature fruit bodies bearing basidiospores were produced after incubation at 22°C for 90 days in the dark, followed by incubation at 26°C for 30–46 days under conditions of high humidity and illumination. The addition of porous stone as a casing on the medium increased fruit-body yield. Deposited spores germinated well on an agar medium and formed mycelial colonies, thus completing the life cycle of Bo1 without a host plant and under axenic conditions. The ability of Bo1 to form ectomycorrhizas was confirmed by axenic resynthesis of mycorrhizas on Quercus serrata. Cultured fruit bodies of Bo1 resembled Gyroporus castaneus and Boletus subcinnamomeus, but its taxonomic position was not elucidated at the species level.  相似文献

The effects of air humidity and temperature on the ascospore discharge of Graphostroma platystoma were experimentally investigated. The ascospores were not discharged from the stromata in air at 100% relative humidity (RH). However, they were discharged from the wetted stromata at 3°, 10°, and 24°C under 100% RH or nearly so. The amount of the discharged ascospore was large at 24°C, medium at 10°C, and small at 3°C. The ascospores in the rainwater that washed down the stromata were counted after rainfall in the field. The discharge was observed from September to the following May.  相似文献

Pectinase and sclerotium production by strains ofAspergillus flavus were determined with a pectinase culture plate assay and a Cz 3% NaNO3 medium plate assay. In theA. flavus population, 51% of isolates produced sclerotia, the toxigenic strains showing a tendency to have smaller sclerotia. Strains producing both abundant small sclerotia and a large quantity of aflatoxin were not found. There was no linear correlation between the amount of aflatoxin produced and the number of sclerotia. Levels of pectinase produced by the toxigenic strains were higher than that of the non-toxigenic strains, and this character was more obvious in the sclerotium-producing strains than in the non-sclerotium-prodcing strains. In theA. flavus population from Zhejiang in which the toxigenic strain rate was low, toxigenic strains may require higher levels of pectinase to compete with the non-toxigenic strains when infecting foodstuffs.  相似文献

A new species of sporulating saccharolytic anaerobe, designated as Clostridium quinii sp. nov., is described. A gram-positive strain BS1, was isolated from the granular metanogenic sludge (UASB) from a waste-water treatment plant at a sugar refinery. The strain exhibits a series of morphological stages, developing from a spore to a small rod to a motile rod (peritrichous flagella) in the exponential growth phase, and then swelling to form cigar-shaped cells, exhibiting tumbling movements, in the late exponential growth phase before finally becoming large nonmotile ovoid cells in the stationary phase. Swelling occurs as a result of glucose being taken up and stored as a glycogen-like substance. The main fermentation products when growing on glucose is H2, CO2, formate, acetate and ethanol as well as small amounts of butyrate during exponential growth. Lactate is formed during the stationary phase, when glucose is abundant. Optimal conditions for growth is 40–45°C and pH of around 7.4. The type strain BS1 contains 28.0% mol G+C.  相似文献

The microanatomy of immature 'white', 'slightly pigmented' and mature, 1-month-old 'black' sclerotia of Sclerotinia trifoliorum , S. sclerotiorum , and S. minor were studied by scanning electron microscopy (SEM). A surface mycelial network was present over sclerotia at maturity. Also dried exudate on the superficial, sclerotial cells at maturity was observed. At this stage of morphogenesis an outer layer of the wall of medullary hyphae was synthesized. Two zones (i.e., rind and medulla) of hyphal tissue in sections of mature sclerotia were distinguished. The wall of rind cells was thick and one-layered, whereas the wall of medullary hyphae was thick and bi-layered.
No lacunac (intercellular spaces) in sclerotial rind were found but the sclerotial medulla appeared to be lacunate in all three species. At the SEM level the structural organization of sclerotia of S. trifoliorum was identical to that one of sclerotia of S. sclerotiorum and S. minor. Thus, in the conducted investigation of the sclerotial stromata, a unique, structural characteristic of taxonomic importance to distinguish S. trifoliorum from the other Sclerotinia species was not found. Observations on the sclerotial morphogenesis in S. trifoliorum and the related species agree with and supplement the light and transmission electron microscope studies of other researchers.  相似文献
