The mechanism regulating the divergent healing processes following tooth replantation is unclear. This study clarifies the relationship between the healing pattern, the time taken for tooth replantation, and the influence of occlusal force. We investigated the pulpal healing process after tooth replantation by immunohistochemistry for 5-bromo-2′-deoxyuridine and nestin and by histochemistry for tartrate-resistant acid phosphatase. The upper right first molar of 3-week-old mice was extracted and repositioned in the original socket immediately or 30 min to 6 h after the operation. We divided the animals into a non-occluded group in which the lower right first molar was extracted and an occluded group without extraction of the counterpart tooth. In control teeth (upper left first molar), the periphery of the coronal dental pulp showed intense nestin-positive reaction. Tooth replantation weakened the nestin-positive reaction in the pulp tissue. On postoperative days 5–7, tubular dentin formation commenced next to preexisting dentin in which nestin-positive odontoblast-like cells were arranged in successful cases. In other cases, bone-like tissue formation occurred in the pulp chamber until day 14. The ratio of tertiary dentin formation was significantly higher in the non-occluded group. The intentionally prolonged time for the completion of tooth replantation induced bone-like tissue formation, expanded inflammatory reaction, or fibrous tissue formation in pulp tissue. Thus, the lack of a proper oxygenated medium is probably decisive for the survival of odontoblast-lineage cells, and occlusal force during and/or after operation worsens the fate of these cells. Dental pulp is assumed to possess the capacity to elaborate both bone and dentin matrix under the pathological conditions following tooth injury. This study was undertaken to clarify the mechanism inducing bone formation in the dental pulp by investigating the pulpal healing process, after tooth replantation, by micro-computed tomography (μ-CT), immunocytochemistry for heat-shock protein (HSP)-25 and cathepsin K (CK), and histochemistry for both alkaline phosphatase (ALP) and tartrate-resistant acid phosphatase (TRAP). Under deep anesthesia, the upper right first molar of 4-week-old Wistar rats was extracted and immediately repositioned in the original socket. In control teeth at this age, the periphery of the coronal dental pulp showed intense ALP-positive and HSP-25-positive reactions, whereas there were no TRAP-positive or CK-positive cells. Tooth replantation weakened or terminated ALP-positive and HSP-25-positive reactions in the pulp tissue at the initial stages. At 3–7 days after operation, the ALP-positive region recovered from the root apex to the coronal pulp followed by HSP-25-positive reactions in successful cases showing tertiary dentin formation. In other cases, TRAP-positive and CK-positive cells appeared in the pulp tissue of the replanted tooth at postoperative days 5–10 and remained associated with the bone tissue after 12–60 days. Immunoelectron microscopy clearly demonstrated that CK-positive osteoclast-lineage cells made contact with mesenchymal cells with prominent nucleoli and well-developed cell organelles. Recently, we demonstrated that a pulse of BrdU given to prenatal animals reveals the existence of slow-cycling long-term label-retaining cells (LRCs), putative adult stem or progenitor cells, which reside in the dental pulp. This study aims to clarify responses of LRCs to allogenic tooth transplantation into mouse maxilla using prenatal BrdU-labeling, in situ hybridization for osteopontin and periostin, and immunohistochemistry for BrdU, nestin, and osteopontin. The upper-right first molars were allografted in the original socket between BrdU-labeled and non-labeled mice or between GFP transgenic and wild-type mice. Tooth transplantation caused degeneration of the odontoblast layer, resulting in the disappearance of nestin-positive reactions in the dental pulp. On postoperative days 5–7, tertiary dentin formation commenced next to the preexisting dentin where nestin-positive odontoblast-like cells were arranged in the successful cases. In BrdU-labeled transplanted teeth, dense LRCs were maintained in the center of the dental pulp beneath the odontoblast-like cells including LRCs, whereas LRCs disappeared in the area surrounding the bone-like tissue. In contrast, LRCs were not recognized in the pulp chamber of non-labeled transplants through the experimental period. Tooth transplantation using GFP mice demonstrated that the donor cells constituted the dental pulp of the transplant except for endothelial cells and some migrated cells, and the periodontal tissue was replaced by host-derived cells except for epithelial cell rests of Malassez. These results suggest that the maintenance of BrdU label-retaining dental pulp cells play a role in the regeneration of odontoblast-like cells in the process of pulpal healing following tooth transplantation.  相似文献

Tooth wear scores (ratios of exposed dentin to total crown area) were calculated from dental casts of Australian Aboriginal subjects of known age from three populations. Linear regression equations relating attrition scores to age were derived. The slope of the regression line reflects the rate of tooth wear, and the intercept is related to the timing of first exposure of dentin. Differences in morphology between anterior and posterior teeth are reflected in a linear relationship between attrition scores and age for anterior teeth but a logarithmic relationship for posterior teeth. Correlations between age and attrition range from less than 0.40 for third molars (where differences in the eruption and occlusion of the teeth resulted in different patterns of wear) to greater than 0.80 for the premolars and first molars. Because of the generally high correlations between age and attrition, it is possible to estimate age from the extent of tooth wear with confidence limits of the order of +/- 10 years.  相似文献

Orthodontic tooth movements are described as the differences between initial and final tooth positions. A computer based method for determination of tooth movements for different treatment methods was developed. A total of 20 casts of the upper jaw of patients treated with tooth positioners or fixed appliances were used as a basis for this study. Tooth movement was analysed on casts before (C i) and after treatment (C f). The casts were digitized either with a COMT or 3D laser scanning systems. After digitization, the models were superimposed in the palate by using a surface–surface matching algorithm. Tooth surfaces of the orthodontically moved teeth were segmented and determination of tooth movement was accomplished by matching the moved teeth from C i to C f. The resulting transformations delivered three dimensional information on translations and rotations. An accuracy of 0.2?mm in translations and 1 degree in rotations could be demonstrated, showing the different efficiency of treatment schemes.  相似文献

目的:探索采用大鼠牙胚细胞构建可注射组织工程牙齿的可行性及在体内肾被膜移植的成牙能力研究。方法:采用出生后4d SD仔鼠磨牙牙胚细胞,培养传代后将牙胚细胞与牛真皮基质蛋白溶液混合构建组织工程牙胚,利用注射器植入SD大鼠肾被膜下,2wk后取材,HE染色观察移植物中的组织成熟情况,Masson’s三色法观察组织工程牙胚的成牙能力。结果:组织工程牙胚移植后2wk可在肾被膜下形成乳白色矿化组织,HE染色上皮细胞和间充质细胞进行重组,形成典型的牙髓牙本质复合体样组织和釉质样结构,Masson’s三色法可见绿色矿化基质形成并包绕牙乳头样组织。结论:牙胚细胞仍保留了牙齿发育的位置信息,采用牛真皮基质蛋白溶液作为组织工程支架有助于牙胚细胞相互作用并重新排列,构建的组织工程牙胚具有进一步成熟、矿化并形成牙齿的潜能。  相似文献

Tooth size changes among Nubian archaeological populations dating from the Mesolithic through the Christian era, a period of approximately 12,000 years, are analyzed. Standard length and breadth dimensions of all permanent teeth from several cultural horizons are combined to form three large samples: Mesolithic, 10000-70000 B.C.; Agriculturalist, 3300-1100 B.C. (A-group, C-group, Pharaonic); and Intensive Agriculturalist, A.D. 0-1400 (Meroitic, X-Group, Christian). Such information not only fills a void in the knowledge of Nubian skeletal biology, but also provides a much needed African reference point for the comparison of tooth size data among anatomically modern Homo sapiens from various regions of the world. Changes in mean tooth size and associated t-tests reveal strong and significant reduction in dental size between the Mesolithic and Agriculturalist samples, followed by a continued although diminished trend of reduction for only the molar teeth between the two Agriculturalist groups. These patterns are best observed by examining tooth breadths, which are considered as the most reliable indicator of tooth size. Previously published odontometrics of the Nubian Mesolithic are briefly compared to the findings of this diachronic analysis of Nubian dental change.  相似文献

Tooth dislocation (tilting) was recorded in 1,200 skulls from 34 museum collections. The findings of dislocation by tooth type, tooth wear, and abscess location are presented. A model for dislocation based upon the progressive loss of tooth support provides a rational explanation for the phenomenon. Physiological continuous tooth eruption was considered to account for a component of the progressive loss of tooth attachment. The process of attrition, pulp perforation, and dental abscess cavity formation resulted in further, more severe loss of tooth support. Heavy functional forces, in association with greatly reduced bone support, tilted the crown lingually and root buccally. When the tooth had tilted to such an extent that the root apices protruded from the bone and, presumably (in life) through the gingival/mucosal tissues, the infected root canals were effectively isolated from the internal environment. The tooth continued to function. The more typical consequence of severe attrition and dental abscess formation was tooth loss; it also isolated an infected tooth from living tissue, but without the benefit of retaining function.  相似文献

Tooth emergence data from a mixed-longitudinal sample of 58 chimpanzees of known age were analyzed using probit and survival techniques to produce median emergence ages, ranges of variability, and emergence sequences for primary and permanent teeth. Between-group comparisons were made to test for statistically significant differences in emergence ages. No such differences were found between right and left sides, or between maxilla and mandible, for any primary or permanent teeth. Male-female comparisons did demonstrate significant emergence-age differences for some teeth, although they were not always bilaterally symmetrical. More complete data are required to further clarify the nature of sex differences in tooth emergence in chimpanzees. Regression models for age prediction from the number of emerged teeth were generated and indicate that males achieve a given number of emerged teeth at a significantly later age than females. However, when fewer than five teeth have emerged, males are predicted to be younger than females. The sizable root mean square error values for these models suggest that this method of age prediction has limited usefulness owing to the amount of variability in timing of tooth emergence in chimpanzees. The implications of these data for studies on tooth emergence in early hominids are addressed.  相似文献

摘要 目的:建立延期再植牙大鼠模型并观察牙根不同部位预后特点,比较不同组织学分析方法的合理性,为相关临床、基础研究提供参考。方法:6周龄雄性SD大鼠10只,右侧上颌切牙脱位后延期再植,术后30 d 处死,分离上颌骨;利用micro CT从不同截面分析上颌切牙及牙周组织生理结构。切片后通过HE染色、MASSON染色、TRAP染色观察比较牙根不同部位愈合情况、破骨细胞分布情况。结果:大鼠切牙唇侧有釉质覆盖,腭侧牙骨质覆盖,且再植牙不同部位愈合情况、破骨细胞分布有差异,其牙根唇侧未见明显吸收,腭侧根中1/3相比根尖1/3和根颈1/3吸收范围更广、深度更深,破骨细胞分布数量更多,差异具有统计学意义(P<0.001),提出了腭侧中1/3作为牙根吸收组织学分析区域的合理性。结论:本实验比较了再植牙大鼠模型牙根不同部位的愈合情况,提出了一种较为合理的组织学分析方法,能客观反映再植牙愈合方式,为进一步研究再植牙愈合机理提供了较好的研究模型。  相似文献

牙齿磨耗是一个多因素、多阶段、复杂渐进的过程。其特征是牙齿硬组织逐渐消失,牙齿磨耗程度可以一定程度反映饮食结构和生活方式。国外很多学者探讨了年龄、性别、饮食结构、断奶、社会分工、社会经济地位等因素对儿童牙齿磨耗的影响,及乳齿恒齿磨耗差异、牙列磨耗模式特点、牙齿磨耗与错𬌗畸形的关系等,而国内儿童牙齿磨耗研究相对薄弱。本文通过梳理和归纳相关文献,对儿童牙齿磨耗研究作简要概述,并对国内相关研究作简要回顾及展望。  相似文献

Tooth development and replacement in fetal and adult viviparous caecilians (Amphibia: Gymnophiona) are described and analyzed according to current theories of tooth succession. The fetal dentition differs from that of the adult in morphology, position, and function. Teeth are used by fetuses to scrape the oviducal epithelium, thus stimulating the secretion of a nutrient substance. Fetal dentitions vary in morphology and position in different species. The ontogeny of teeth of several species is described and the patterns of addition of loci and of replacement are analyzed. Loci are added both posteriorly along the jaw and between existing loci as the jaw grows prior to ossification; subsequently addition is restricted to the posterior part of the jaw. Tooth replacement is alternate. The several rows and patches of teeth are the result of retention of replacement series on the dentigerous elements. Tooth development and replacement in a series of juveniles and adults of different sizes in a single species are also considered. Post-fetal patterns of development and replacement are similar to those seen in larvae and adults of oviparous species. Variation in numbers of teeth and proportions of teeth at particular stages occurs ontogenetically and among individuals of the same size, though proportions occur in a similar pattern throughout the series. The general pattern of tooth replacement in fetuses and adults can be explained by either Edmund's Zahnreihen theory or by Osborn's Tooth Family theory, but replacement in fetal tooth patches and the fetal-adult dentitional transition are explained by neither.  相似文献

Our recent study suggested that allogenic tooth transplantation may affect the maintenance of dental pulp stem/progenitor cells. This study aims to elucidate the influence of allograft on the maintenance of dental pulp stem/progenitor cells following tooth replantation and allo- or auto-genic tooth transplantation in mice using BrdU chasing, immunohistochemistry for BrdU, nestin and Ki67, in situ hybridization for Dspp, transmission electron microscopy and TUNEL assay. Following extraction of the maxillary first molar in BrdU-labeled animals, the tooth was immediately repositioned in the original socket, or the roots were resected and immediately allo- or auto-grafted into the sublingual region in non-labeled or the same animals. In the control group, two types of BrdU label-retaining cells (LRCs) were distributed throughout the dental pulp: those with dense or those with granular reaction for BrdU. In the replants and autogenic transplants, dense LRCs remained in the center of dental pulp associating with the perivascular environment throughout the experimental period and possessed a proliferative capacity and maintained the differentiation capacity into the odontoblast-like cells or fibroblasts. In contrast, LRCs disappeared in the center of the pulp tissue by postoperative week 4 in the allografts. The disappearance of LRCs was attributed to the extensive apoptosis occurring significantly in LRCs except for the newly-differentiated odontoblast-like cells even in cases without immunological rejection. The results suggest that the host and recipient interaction in the allografts disturbs the maintenance of dense LRCs, presumably stem/progenitor cells, resulting in the disappearance of these cell types.  相似文献

以军都山墓地为代表的"玉皇庙文化"系中国北方青铜时代的一支具有鲜明地方特色的少数部族文化。本文对军都山墓地出土人骨牙齿标本作牙齿磨耗状况分析,试图为其饮食结构和社会经济形态的探讨提供线索。结果显示:军都山古代人群中,相同年龄组的男女两性牙齿磨耗差异不大;前部牙齿磨耗轻于后部臼齿,第一臼齿磨耗最重;臼齿磨耗样式大多呈现"正常平匀"式,臼齿磨耗角度大多较小,两者均随年龄而变化但没有显著的性别差异。经与其他样本组的对比认为军都山古代人群牙齿磨耗的特点可能与其社会经济农牧兼营的性质有关,推测动物性食物(肉食)可能在军都山古代人群主要饮食结构中占据重要比例,并包含有一定的农业经济成分。不同游牧人群牙齿磨耗程度差异较大,只有结合了磨耗形态及其他信息才有可能更切实地反映其饮食状况。  相似文献

Recent studies have demonstrated that human dental pulp contains adult stem cells. A pulse of the thymidine analog BrdU given to young animals at the optimal time could clarify where slow-cycling long-term label-retaining cells (LRCs), putative adult stem cells, reside in the pulp tissue. This study focuses on the mapping of LRCs in growing teeth and their regenerative capacity after tooth injuries. Two to seven peritoneal injections of BrdU into pregnant Wistar rats revealed slow-cycling long-term dense LRCs in the mature tissues of born animals. Numerous dense LRCs were postnatally decreased in number and reached a plateau at 4 weeks after birth when they mainly resided in the center of the dental pulp, associating with blood vessels. Mature dental pulp cells were stained with Hoechst 33342 and sorted into (<0.76%) side population cells using FACS, which included dense LRCs. Some dense LRCs co-expressed mesenchymal stem cell markers such as STRO-1 or CD146. Tooth injuries caused degeneration of the odontoblast layer, and newly differentiated odontoblast-like cells contained LRCs. Thus, dense LRCs in mature pulp tissues were supposed to be dental pulp stem cells possessing regenerative capacity for forming newly differentiated odontoblast-like cells. The present study proposes the new hypothesis that both granular and dense LRCs are equipped in the dental pulp and that the dense LRCs with proliferative capacity play crucial roles in the pulpal healing process following exogenous stimuli in cooperation with the granular LRCs.  相似文献

Tooth replacement poses many questions about development, pattern formation, tooth attachment mechanisms, functional morphology and the evolution of vertebrate dentitions. Although most vertebrate species have polyphyodont dentitions, detailed knowledge of tooth structure and replacement is poor for most groups, particularly actinopterygians. We examined the oral dentition of the bluefish, Pomatomus saltatrix, a pelagic and coastal marine predator, using a sample of 50 individuals. The oral teeth are located on the dentary and premaxillary bones, and we scored each tooth locus in the dentary and premaxillary bones using a four-part functional classification: absent (A), incoming (I), functional (F=fully ankylosed) or eroding (E). The homodont oral teeth of Pomatomus are sharp, deeply socketed and firmly ankylosed to the bone of attachment. Replacement is intraosseus and occurs in alternate tooth loci with long waves of replacement passing from rear to front. The much higher percentage of functional as opposed to eroding teeth suggests that replacement rates are low but that individual teeth are quickly lost once erosion begins. Tooth number increases ontogenetically, ranging from 15–31 dentary teeth and 15–39 premaxillary teeth in the sample studied. Teeth increase in size with every replacement cycle. Remodeling of the attachment bone occurs continuously to accommodate growth. New tooth germs originate from a discontinuous dental lamina and migrate from the lingual (dentary) or labial (premaxillary) epithelium through pores in the bone of attachment into the resorption spaces beneath the existing teeth. Pomatomus shares unique aspects of tooth replacement with barracudas and other scombroids and this supports the interpretation that Pomatomus is more closely related to scombroids than to carangoids.  相似文献

Tooth loss and tooth retention in adult South African Bantu-speaking Negroes were studied from data derived from 500 dry mandibles. The tribe, sex, and stated age of the specimens were available. Mandibles were equally distributed over the third to seventh decade of life. The presence of dental units was recorded and analysed within and between decade age groups. The mandibles carried a total of 5,459 dental units. Between the third and fourth decades of life and between the sixth and seventh decades, the reduction of the frequency of dental units was mild and statistically not significant. It was, however, severe and significant between the fourth and fifth, and the fifth and sixth decades. In the third-decade group, most teeth were found in the anterior region with a tendency to diminish in number posteriorly. After the third decade there was a definite persistence of first premolars and canines with significant tendency toward loss of teeth anterior and posterior to them. Only the incisors and third molars showed significant reduction in number from the third to the fourth decade. The reduction of all teeth was statistically significant later, until the sixth decade of life. Although there was loss of teeth from the sixth to the seventh decade, it was not significant for any of the homologous dental units. There is a possibility that some dietary differences existed between the older subjects (50–70 years old) and the younger ones (20–49 years old) which may have had some influence on the results.  相似文献

Apoptosis is a key phenomenon in the regulation of the life span of odontoblasts, which are responsible for dentin matrix production of the teeth. The mechanism controlling odontoblasts loss in developing, normal, and injured human teeth is largely unknown. A possible correlation between apoptosis and dental pulp volume reduction was examined. Histomorphometric analysis was performed on intact 10 to 14 year-old premolars to follow dentin deposition and evaluate the total number of odontoblasts. Apoptosis in growing healthy teeth as well as in mature irritated human teeth was determined using a modified TUNEL technique and an anti-caspase-3 antibody. In intact growing teeth, the sequential rearrangement of odontoblasts into a multi-layer structure during tooth crown formation was correlated with an apoptotic wave that leads to the massive elimination of odontoblasts. These data suggest that apoptosis, coincident with dentin deposition changes, plays a role in tooth maturation and homeostasis. Massive apoptotic events were observed after dentin irritation. In carious and injured teeth, apoptosis was detected in cells surrounding the lesion sites, as well as in mono-nucleated cells nearby the injury. These results indicate that apoptosis is a part of the mechanism that regulate human dental pulp chamber remodeling during tooth development and pathology.  相似文献
