A new triterpene glycoside, repandoside (1) was isolated together with six known saponins (2-7) from the methanol extract of Cyclamen repandum tubers. The isolated saponins were characterized by high resolution mass spectrometry and both 1D and 2D NMR experiments. The in vitro effect of saponins on LPS-induced IL-8 and TNF-α mRNA level (by quantitative RT-PCR) and protein release (by ELISA) was evaluated in human THP-1 macrophages. We found that compounds 1 (repandoside), 2 (deglucocyclamin) and 4 (anagalloside B) at 100 μM inhibited the LPS-induced IL-8 and TNF-α expressions.  相似文献

In our preliminary screening study on the anti-inflammatory activity, eight triterpenes, one sterol, and one chalcone were isolated from the CH2Cl2-soluble extract of the stems and leaves of Rhus sylvestris Siebold and Zucc (Anacardiaceae). On the basis of their spectroscopic data, these compounds were identified as 10α-cucurbitadienol (1), glut-5-en-3-ol (2), β-amyrin acetate (3), β-amyrin (4) and lupeol (5), cycloart-24-en-3-one (6), cycloart-25-en-3,24-dione (7), 24-hydroxycycloart-25-en-3-one (8), β-sitosterol (9), and 2′-hydroxy-4,4′-dimethoxychalcone (10). All of them were isolated from this plant for the first. Furthermore, the compounds in non-cytotoxic concentrations (0–1.0 μM) were tested for their ability to block inflammatory cytokine secretion in the presence of LPS in the murine RAW264.7 macrophage cell line. Among the compounds that were tested, compounds 8 and 9 reduced the LPS-induced secretion of IL-6, as well as TNF-α, in a mouse RAW264.7 macrophage cell line. Moreover, compounds 2, 3, 7, and 10 specifically diminished only the secretion of TNF-α even in 0.01 μM concentrations. It is thus suggested that they are potential therapeutics of TNF-α-related diseases and conditions, such as transplant rejection, type II diabetes, and atherosclerosis.  相似文献

The tumor necrosis factor alpha (TNF-α) and interleukin 1 beta (IL-1β) inhibitory activities of Cajanus cajan (leaves) crude methanolic extract, its fractions and its phytochemical constituents were evaluated in lipopolysaccharide (LPS) stimulated RAW 264.7 and J774A.1 cells. Phytochemical investigation of the active ethyl acetate (CCE) and n-butanol (CCB) fractions of C. cajan L. leaves yielded 14 compounds. It was observed that both pinostrobin (9) and cajanus lactone (4) were found to be most active in inhibiting TNF-α (IC50 < 22 μM) and IL-1β (IC50 < 40 μM) whereas compounds 2, 3, 58, 10 and 14 showed moderate and mild effects (IC50 = 35.50–81.22 μM for TNF-α and 38.23–89.10 μM for IL-1β) in both the cell lines. Furthermore, at dose of 20 mg/kg, both pinostrobin (9) and cajanus lactone (4) were found to reduce LPS-induced TNF-α levels by 48.6% and 55.0% respectively and IL-1β levels by 53.1% and 41.8% respectively in Sprague Dawley (SD) rats. These findings suggest that C. cajan L. leaves can be developed as an effective herbal remedy for the treatment and prevention of inflammation or associated ailments.  相似文献

Three new phenolic glycosides 2-(3-O-β-d-glucopyranosyl-4-hydroxyphenyl) ethanol 1-O-β-d-glucopyranoside (1), 2-(4-O-β-d-fructopyranosylphenyl) ethanol 1-O-β-d-galactopyranoside (2) and 3-methoxy-4-O-β-d-allopyranosyl acetophenone (3), along with nine known compounds (4–12), were isolated from the ethanol extract of the whole plant of Aconitum tanguticum (Maxim.) Stapf. Their structures were elucidated by analysis of spectroscopic data including 1D-, 2D-NMR and HRESIMS, and the reported literature data comparison. All the compounds were evaluated for their potential anti-inflammatory effects by the inhibition of TNF-α production on LPS-stimulated RAW264.7 macrophages. Compounds 1, 3, 5 and 79 showed certain inhibition activity and their IC50 values were 38.18, 27.64, 3.25, 84.45, 12.76 and 18.44 μg/mL, respectively.  相似文献

Cycloaddition reaction of 4-chloro-2-oxo-2H-chromene-3-carbaldehydes (3a-g) and 4-chloro-2H-chromene-3-carbaldehydes (7a-h) with activated alkynes (4a-b) provided the 2-oxo-2H-chromenyl-5-oxo-2,5-dihydrofuran-3-carboxylates (5a-n) and 2H-chromenyl-5-oxo-2,5-dihydrofuran-3-carboxylates (8a-p). All the prepared compounds were screened for anti-inflammatory activity. In vitro anti-inflammatory activity data demonstrated that the compounds 5g, 5i, 5k-l and 8f are effective among the tested compounds against TNF-α (1.108 ± 0.002, 0.423 ± 0.022, 0.047 ± 0.001, 0.070 ± 0.002 and 0.142 ± 0.001 µM) in comparison with standard compound Prednisolone (0.033 ± 0.002 µM). Based on in vitro results, three compounds (5i, 5k and 8f) have been selected for in vivo experiments and these compounds are identified as better compounds with respect to anti-inflammatory activity in LPS induced mice model. Sanguisorba officinalis L. was well known as a traditional herbal medicine to treat inflammation and allergic skin diseases. The aim of this research was to indentify compounds with anti-allergic inflammatory property. Twenty-five compounds (125) were isolated from S. officinalis including two new compounds (1 and 8), and their chemical structures were identified by NMR and ESIMS analysis. Consequently, the anti-allergic inflammatory activities of these isolates were investigated by inhibiting β-hexosaminidase and IL-4 production in PMA/A23187-stimulated RBL-2H3 cells. Compounds 6, 8, 13, 1718 and 25 significantly inhibited β-hexosaminidase release and IL-4 production. Additionally, compounds 8, 17 and 25 effectively suppressed the activation of NF-κB and NF-κB p65 translocation into the nucleus. Anti-inflammatory effects of isolated compounds were evaluated in LPS-stimulated RAW264.7 macrophages, and they showed dramatic inhibition on LPS-induced overproduction of nitric oxide (NO) and TNF-α. Consistently, the protein levels of iNOS and COX-2 were remarkably decreased by the single compounds 8, 13 and 25. These results showed that compounds 8, 13 and 25 from S. officinalis may have a therapeutic potential for allergic inflammatory diseases.  相似文献

A new adduct of iriflophene and flavonoid, 1, 8, 10, 10b-tetrahydroxy-3-methoxy-5a-(3,4-dihydr- oxyphenyl)-9-(4-hydroxybenzoyl)-5a,10b-dihydro-11H-benzofuro[2,3-b]chromen-11-one (1), together with six known compounds were isolated from the EtOH extract from Sedum aizoon L. Their chemical structures were elucidated by analyses of spectral data. The chemotaxonomic analysis of the compounds was discussed, compounds 14 provide important chemotaxonomic markers for S. aizoon.  相似文献

Using various chromatographic techniques, 23 triterpene saponins (1–23) were isolated from an ethanol extract of Stauntonia hexaphylla, including two new compounds (12 and 15). Their chemical structures were established by comprehensive spectroscopic methods such as 1D- and 2D-NMR, and HR-ESI-MS, and chemical reactions. The anti-inflammatory activities of the isolated saponins were determined using the nitric oxide (NO) assay. Compound 13 exhibited the greatest inhibitory effect (IC50?=?0.59?μM). In addition to NO, compound 13 suppressed the secretion of PGE2, IL-1β, and IL-6, but not TNF-α, and inhibited the protein expression of iNOS and COX-2 in LPS-activated RAW264.7 cells. The chemical derivatives of the isolated compounds were studied using structure–activity relationships. The results suggested that compound 13 isolated from S. hexaphylla might be useful for treating inflammation. This is the first comprehensive study of saponins from the leaves of S. hexaphylla based on anti-inflammatory extract screening guidelines.  相似文献

Chemical investigation of the EtOAc extract of the medicinal plant Thymelaea lythroides yielded eight secondary metabolites, including bassiatin (1), daphenone (2), daphnelone (3), daphnoretin (4), δ-sesamin (5), wikstromol (6), trans-tiliroside (7), and rutarensin (8). The structures of the isolated compounds were determined on the basis of 1D and 2D NMR spectroscopy as well as mass spectrometry. Glandularia selloi (Verbenaceae) presents phenylethanoids, iridoids and flavone glycosides as the main constituents. Two novel chrysoeriol derivatives, selloiside A (1) and selloiside B (2) were isolated from the methanolic extract of the aerial parts. Both flavones are acylated disaccharides. Two known compounds, verbascoside (3) and 6β-hydroxy-ipolamiide (4), were isolated from the methanolic extract of the roots. The structures were elucidated using 1D, 2D NMR and MS. Acylflavones have taxonomic significance since they occur mainly is species from the order Lamiales, specifically from the family Lamiaceae, closely related to Verbenaceae. The compounds 14 and the methanolic extract were investigated for antichemotactic activity.  相似文献

Fourteen compounds were isolated from the 95% ethanol reflux extract of Asarum sieboldii Miq. var. Seoulense Nakai, including five phenanthrene derivatives (15), three isobutyl amides (68), three phenylpropanoids (911) and three lignins (1214). The structures of these compounds were identified by spectroscopic methods and by comparison with the reported spectroscopic data. Among them, compounds 6 and 11 were firstly reported from the family Aristolochiaceae, and compounds 3 and 4 were reported for the first time from the genus Asarum. Additionally, compounds 1, 2 and 8 were isolated from A. sieboldii Miq. var. Seoulense Nakai for the first time. These compounds have shown chemical relationships between A. sieboldii Miq. var. Seoulense Nakai and other species of Asarum as well as those found in the genus Aristolochia in the family Aristolochiaceae.  相似文献

The investigation of the ethanol extract of fresh aerial parts of the Patagonian shrub Nardophyllum bryoides collected in the province of Chubut, Argentina, yielded eleven terpenoids. These include: three seco-ent-halimane diterpenoids (13), two ent-halimanes (45) and six pentacyclic oleanane and ursane triterpenoids (611). Four of these compounds (2, 6, 8 and 11) are hitherto unknown, while two others (1 and 4) have been previously reported but only as synthetic products. Several of these compounds showed moderate cytotoxicity against a human pancreatic adenocarcinoma cell line while compounds 4 and 5 were active at micromolar concentrations. The main component, seco-chiliolidic acid (1), could be isolated from this extract in large amounts, turning N. bryoides into a sustainable source of this bioactive compound.  相似文献

The phytochemical study was done on the leaves of Gambeya lacourtiana. This plant has been used in traditional medicine to treat of different ailments such as uterine heamorrhage, metritis and other vaginal infections. Chromatographic fractionation and purification on the leaves crude extract afforded lupeol acetate (1), lupenone (2), lupeol (3), taraxerol (4) stigmasterol (5), erythrodiol (6), chamaedrydiol (7), methyl pheophorbide-a (8), corosolic acid (9), tormentic acid (10), epicatechin (11) and 22-dihydrospinasterol 3-O-β-D-glucopyranoside (12). The structures of compounds 112 were elucidated on the basis of 1D and 2D NMR, mass spectrometric and the spectroscopic data as well as comparison with the literature. Compounds 4, 710 and 12 were isolated for the first time from Gambeya genus. Crude extract, fractions and compounds 6–12 were evaluated for their antibacterial activity. Methyl pheophorbide-a (8) demonstrated moderate activity against Salmonella typhi CPC with MIC values of 62.5 μg/mL. The chemophenetic significance of these compounds is also discussed.  相似文献

The phytochemical study was done on the methanol extract from of the leaves of Symphonia globulifera. This plant has been used in traditional medicine to treat of different ailments such as malaria, diseases of the skin, diabetes, cough, intestinal worms, pre-hepatic jaundice and fever. Chromatographic fractionation and purification of this extract led to the isolation and characterization of twelve compounds (1–12) including pristriol (1), robustaflavone (2), polycarpol (3), 7''-O-methylrobustaflavone (4), amentoflavone (5), stigmasterol glucoside (6), epicatechin (7), apigenin (8), luteolin (9), 1,5-dihydroxyxanthone (10), β-sitosterol 3-β-D-glucopyranoside (11) and a mixture of stigmasterol and β-sitosterol (12). The structures of compounds 1–12 were elucidated on the basis of 1D and 2D NMR, mass spectrometric and the spectroscopic data as well as comparison with the literature. All these compounds were isolated for the first time from Symphonia genus. The NMR spectra and structure elucidation of compound 1 were reported for the first in the literature. The antibacterial and antioxidant activities of three of these compounds were evaluated. The chemophenetic significance of these compounds is also discussed.  相似文献

Seven polyhydroxylated steroidal glycosides (17) and two polyhydroxylated sterols (8, 9) were isolated and identified from the 75% ethanol extract of starfish Culcita novaeguineae which was collected in Xisha Islands of South China Sea. The structures of the isolated compounds were elucidated on the basis of spectroscopic data, physical and chemical evidence, MS spectrum and assisted by comparison with the reported data. To the best of our knowledge, this is the first time to report compounds 16, 8 and 9 from genus Culcita. Moreover, compounds 1, 46, 8 and 9 were isolated from family Oreasteridae for the first time. Phytochemical investigation of the stem bark of Ficus natalensis afforded eleven compounds including one ceramide (1), two anthraquinones (2, 3), four triterpenes (47), two polyols (8, 9) and two steroids (10, 11). The structures of the compounds were determined by spectroscopic analyses including IR, UV, MS, 1D- and 2D- NMR (1H, 13C, 1H–1H COSY, HMQC, HMBC and NOESY), as well as by comparison with literature data. The antibacterial activity and the cytotoxicity of the extract, fractions and some isolated compounds (3, 5, 8 and 9) were evaluated. Some fractions and sub-fractions from various column chromatography displayed moderate antibacterial activity with diameter zone of inhibition (DZI) ranging from 7 to 10 mm. None of the compounds tested had activity. In the present study, all the compounds are isolated for the first time from the species F. natalensis. Compounds 2, 47, 10 and 11 were previously reported from the genus Ficus. The chemophenetic significance of the isolated compounds is discussed.  相似文献

Anti-inflammatory assay-guided separation of extracts from the roots of Caesalpinia mimosoides Lamk. led to isolation of seven compounds: four diterpenes (14), a dimer (9), and two dibenzo[b,d]furans (10, 11) together with eleven known compounds. Their structures were elucidated by 1D- and 2D-NMR techniques as well as UV, IR, mass spectral data and comparison with literature values. The anti-inflammatory activities of all compounds were evaluated for inhibitory activities against lipopolysaccharide (LPS) induced nitric oxide (NO) production in RAW264.7 cell lines. Compounds 4, 6, 8, and 1214 were also tested for the inhibitory effect on LPS-induced tumor necrosis factor-alpha (TNF-α) release in RAW264.7 cells. The results indicated that 4 possessed potent inhibitory activity for both tests with IC50 values of 3.0 and 6.5 μM, respectively.  相似文献

Phytochemical investigation of the ethanolic extract from the leaves of Cinnamomum parthenoxylon (Jack) Meisn. led to the isolation of (3R, 4R, 3′R, 4′R)-6,6′-dimethoxy-3, 4, 3′, 4′-tetrahydro-2H, 2′H-[3, 3′]bichromenyl-4, 4′-diol (1), 4-hydroxybenzaldehyde (2), 1,2,4-trihydroxybenzene (3), kaempferol-3-O-α-l-rhamnoside (4), herbacetin (5), quercetin-3-O-α-l-rhamnoside (6), daucosterol (7), and β-sitosterol (8). The structures were established by extensive analysis of their MS and NMR spectroscopic data and comparison with literature data. In the present research, all of the isolated compounds 18 are reported for the first time in the species C. parthenoxylon. Compounds 16 were firstly isolated from genus Cinnamomum. Compounds 1, 3, 5 and 6 have not been reported from any species in Lauraceae family. The chemotaxonomic significance of the isolated compounds is discussed.  相似文献

Two new prenylflavones 5,7,3′,4′-tetrahydroxy-3-methoxy-8-geranylflavone (1) and 5,7,3′,4′-tetrahydroxy-3-methoxy-8,5′-diprenylflavone (2), as well as four known ones, uralenol (3), papyriflavonol A (4), broussoflavonol B (5) and broussochalcone A (6) were isolated and purified from an ethyl acetate-soluble extract of the barks of Broussonetia papyrifera. Their structures were determined with the spectroscopic methods including HR-EI-MS, 1D and 2D NMR. We found that compounds 26 showed potent anti-proliferation effects on ER-positive breast cancer MCF-7 cells in vitro. The IC50 values of compounds 2 and 5 were 4.41 and 4.19 μM respectively after the treatment of 72 h. We also found that compounds 2 and 5 strongly down-regulated expression concentrations of estrogen receptor-α (ER-α) and were able to inhibit tumor growth in a xenograft model of the human breast cancer line BCAP-37 in vivo. Our results demonstrated that prenylflavones from B. Papyrifera exhibit potent anti-tumor activity.  相似文献

Investigation of the 95% ethanolic extract of red yeast rice fermented with the pink mutant of the fungus Monascus pilosus BCRC PK-1 led to the isolation of one new pyridine alkaloid, namely (E)-methyl 4-(2-acetyl-4-oxotridec-1-enyl)-6-propylnicotinate (1), together with 9 known compounds (210). Their structures were elucidated on the basis of spectroscopic data, including homo- and heteronuclear correlation NMR experiments (COSY, HSQC and HMBC) and comparison with data reported in the literature. We also evaluated the inhibitory effects of some isolates on the production of tumor necrosis factor (TNF-α) induced by lipopolysaccharide (LPS). Among the isolates, compounds 1 and 2 inhibited TNF-alpha production in U937 cells in vitro, and the IC50 values are 1.52 and 2.56 μg/ml, respectively.  相似文献
