Several studies have focused on understanding the biochemistry and morphology of the fat body of the hematophagous mosquito Aedes aegypti (L.) (Diptera: Culicidae). In contrast, few studies, if any, have focused on morphological characters of the fat body in other mosquitoes, especially non-hematophagous taxa such as the culicid Toxorhynchites. Larvae of Toxorhynchites prey upon the larvae of other mosquito species and are used in vector mosquito control. We investigated aspects of the fat body trophocytes, including the morphometric analyses of the lipid droplets, protein granules and nuclei, during Toxorhynchites theobaldi (Dyar & Knab) post-embryonic development. Following the body weight increase from larval stage L2 to L4, the size of lipid droplets within the trophocytes also increase, and are likely the result of lipogenesis. Lipid droplets decrease in size during L4 to the female pupal stage and increase once again during the period from newly-emerged to mature adult females. Protein granules are observed for the first time in female pupae, and their appearance might be related to protein storage during metamorphosis. The size of the nucleus of trophocytes also increases during larval development, followed by a decrease during metamorphosis and an additional increase as adult female ages. In conclusion, the morphology of the fat body of T. theobaldi changes according to the developmental stage. Our study provides for the first time important insights into T. theobaldi fat body development and contributes to understand this species biology.  相似文献

The ultrastructural study on the fat body of gynes (virgin queens) of the basal ant species Cyphomyrmex rimosus and Mycetarotes parallelus and the derived Acromyrmex disciger and Atta laevigata queens showed vesicular rough endoplasmic reticulum, Golgi complex, and mitochondria in trophocytes, suggesting the involvement of these cells in protein synthesis, in addition to digestive vacuoles associated with the digestion of endocytosed compounds or rejected cell organelles. Oenocytes, another cell type present in the fat body of these species exhibit mitochondria, digestive vacuoles, and vesicles, indicating a mobilization of compounds by these cells. In A. laevigata, oenocytes also exhibited large storage sites of glycogen, in addition to a well-developed vesicular rough endoplasmic reticulum, suggesting an intensive participation of these cells in protein synthesis. The ultrastructural cytochemistry study also revealed electrodense granules of basic proteins present throughout the cytoplasm of trophocytes. The same was observed in oenocytes, although with smaller amounts of proteins. In the cytoplasm of trophocytes and oenocytes were also found droplets or electrodense granules of lipids. In oenocytes of A. disciger and in trophocytes of A. laevigata, lipids were observed in mitochondria, suggesting that this organelle might be a site of synthesis of these compounds. The chemical analysis of lipids revealed that in gynes, the main compounds present in fat body cells were saturated fatty acids, while in queens, saturated as well as unsaturated fatty acids were found. In conclusion, the present study showed that the fat body cells of gynes and queens, in general, maintained the same compounds and original features through the evolution process of the Attini tribe.  相似文献

20 species of blood-sucking mosquitoes were recorded from the northern taiga of Ural (the northern part of the Sverdlovsk region): 15 species of the genus Aedes, 2 species of Culex, 2 species of Culiseta and 1 species of Anopheles. Larvae of Aedes occur in masse in small temporary water bodies. In summer after rains larvae of Culiseta and Culex as well as some species of Aedes appear. In May overhibernated Culiseta alaskaensis, C. bergrothi and An. maculipennis begin the flight of mosquitoes. The peak of the number and specific variety of attacking mosquitoes was recorded in July; in August their activity started to decrease. Aedes communis and A. punctor are mass species, A. excrucians, A. hexodontus and C. bergrothi are rather abundant, the other species are small in number or rare.  相似文献

The ultrastructural cytochemistry study and the chemical analysis carried out to examine the fat body of workers of the basal Attini Cyphomyrmex rimosus and Mycetarotes parallelus and the derived Acromyrmex disciger and Atta laevigata revealed very electrondense protein granules of varied sizes and shapes distributed throughout the cytoplasm of parietal and perivisceral trophocytes and oenocytes. Lipids were present in the cytoplasm of trophocytes as very electrondense granules. In M. parallelus, A. disciger, and A. laevigata, lipids were present as droplets. In parietal as well as perivisceral oenocytes, lipid granules were very electrondense. In A. laevigata, lipids were present as droplets. The chemical analysis using gas chromatography showed that the main lipid compounds present in trophocytes and oenocytes were saturated (myristic, palmitic, and stearic) and unsaturated fatty acids (oleic and linoleic). Also, in basal species, lipid compounds consisted of mainly saturated fatty acids, while in derived species saturated as well as unsaturated fatty acids were observed.  相似文献

Involvement of esterase-mediated hydrolysis as a mechanism of pyrethroidresistance in three species of mosquitoes,viz., Aedes aegypti, Culex quinquefasciatus andAnopheles Stephensi was investigated by microplate assay of Β-esterases in individual larva and adult female and male mosquitoes. Assuming an absorbance value of 0.4 and above at 555 nm as the threshold level of elevated esterase activity which confers resistance, frequency distributions of such individual test mosquitoes were constructed in resistant and susceptible populations. The results indicate the involvement of ester hydrolysis of Pyrethroids as a predominant mechanism of pyrethroid-resistance in the larvae ofCulex quinquefasciatus but not inAedes aegypti. However, a marginal role of esterases is indicated in the larvae ofAnopheles stephensi.  相似文献

A cysteine proprotease has been identified in developing embryos of the cockroach Blattella germanica and found to be a maternally encoded gene product that is transferred endocytically to the oocyte. The present study aims at establishing how this maternally derived proprotease is synthesized, packaged, and secreted during vitellogenesis. To this end, proprotease was localized immunocytochemically in the fat body of postmating females and its localization compared with that of vitellogenin over the same developmental periods. Fat bodies in cockroaches are comprised of two different cell types: trophocytes and bacteriocytes. Data show that proprotease and vitellogenin come to colocalize in compound granules of the fat body trophocytes. While synthesis of vitellogenin can be traced back to granules resulting from the coalescence of Golgi-derived vesicles in the trophocyte cytoplasm, proprotease appears to be localized predominantly on the cytolysosomes of both trophocytes and bacteriocytes. When probed with an anti-proprotease antiserum, bacteria are also positively labeled, regardless of whether they are segregated inside the cytolysosomes or free in the bacteriocyte cytoplasm. Since vitellogenin and proprotease colocalize within the same cell organelle, it is assumed that Golgi-derived vesicles, which contain vitellogenin, may fuse with cytolysosomes bearing proprotease to yield compound secretory granules. To account for the present observations, the origin and role of proprotease are discussed in relation to the turnover of bacteria in the fat body and to the requirements of endosymbiosis.  相似文献

Adult mosquito surveillance was conducted during 2005 at 22 US military installations and two training sites located in six provinces in the Republic of Korea. Adult mosquitoes were collected in New Jersey light traps from 1 May to 15 October to determine threshold levels to initiate pesticide applications based on trap indices. A total of 27 363 adult mosquitoes (22 368 [81.7%] females and 4995 [18.3%] males) comprising 15 species (including five members of the Anopheles sinensis group) belonging to seven genera were collected. The most common species collected were members of the Anopheles sinensis Wiedemann group (50.2%), followed by Culex tritaeniorhynchus Giles (30.6%), Aedes vexans nipponii (Theobald) (11.8%), and Culex pipiens Coquillett (6.3%) females. Trap indices varied widely for species over their range, due in part to geographical distribution and degree of association with urban or rural communities.  相似文献

A library of monoclonal antibodies (mAB) directed against yolk polypeptides of the mosquito Aedes aegypti was utilized to visualize the secretory pathway of these polypeptides in the fat body and their accumulative pathway in developing oocytes. Single and double immunolabelling using mABs and colloidal gold of different sizes confirmed biochemical observation that 200 +/- 5 and 65 +/- 3 kDa polypeptides represent subunits of the yolk protein. This immunocytochemical analysis showed that, in trophocytes of the fat body, both the subunits of the yolk protein were routed simultaneously through the Golgi complex into secretory granules and were subsequently secreted. The yolk protein subunits were also directed together through all the steps of the accumulative pathway in the oocyte. Double immunogold labelling revealed that the subunits were present together during their binding to the oocyte membrane, transportation into and accumulation in the transitional yolk body, and, finally, crystallization in the mature yolk body. Electron microscopical immunocytochemistry also confirmed immunofluorescent data and showed that mABs directed against different steps in the biosynthetic processing of the yolk protein in the fat body, as well as in its accumulative pathway in oocytes.  相似文献

Amblyospora species and other aquatic Microsporidia were isolated from mosquitoes, black flies, and copepods and the small subunit ribosomal RNA gene was sequenced. Comparative phylogenetic analysis showed a correspondence between the mosquito host genera and their Amblyspora parasite species. There is a clade of Amblyospora species that infect the Culex host group and a clade of Amblyospora that infect the Aedes/Ochlerotatus group of mosquitoes. Parathelohania species, which infect Anopheles mosquitoes, may be the sister group to the Amblyospora in the same way that the Anopheles mosquitoes are thought to be the sister group to the Culex and Aedes mosquitoes. In addition, by sequence analysis of small subunit rDNA from spores, we identified the alternate copepod host for four species of Amblyospora. Amblyospora species are specific for their primary (mosquito) host and each of these mosquito species serves as host for only one Amblyospora species. On the other hand, a single species of copepod can serve as an intermediate host to several Amblyospora species and some Amblyospora species may be found in more than one copepod host. Intrapredatorus barri, a species within a monotypic genus with Amblyospora-like characteristics, falls well within the Amblyospora clade. The genera Edhazardia and Culicospora, which do not have functional meiospores and do not require an intermediate host, but which do have a lanceolate spore type which is ultrastructurally very similar to the Amblyospora spore type found in the copepod, cluster among the Amblyospora species. In the future, the genus Amblyospora may be redefined to include species without obligate intermediate hosts. Hazardia, Berwaldia, Larssonia, Trichotuzetia, and Gurleya are members of a sister group to the Amblyospora clades infecting mosquitoes, and may be representatives of a large group of aquatic parasites.  相似文献

ITmD37E, a unique class II transposable element (TE) with an ancient origin, appears to have been involved in multiple horizontal transfers in mosquitoes as ITmD37E sequences from 10 mosquito species of five genera share high nucleotide (nt) identities. For example, ITmD37E sequences from Aedes aegypti and Anopheles gambiae, which have an estimated common ancestor of 145-200 million years ago, display 92% nt identity. The comparison of ITmD37E and host mosquito phylogenies shows a lack of congruence. The wide distribution of conserved ITmD37Es in mosquitoes and the presence of intact copies suggest that this element may have been recently active.  相似文献

The used tire trade has facilitated the introduction, spread, and establishment of the Asian tiger mosquito, Aedes albopictus, and other mosquito species in several countries of America, Africa, Oceania, and Europe. A strategy for detecting these imported mosquito vectors was developed in Spain during 2003-2004 by EVITAR (multidisciplinary network for the study of viruses transmitted by arthropods and rodents). A survey in 45 locations found no invasive species. Eight autochthonous species of mosquitoes were detected in used tires, including Culex pipiens, Cx. hortensis, Cx. modestus, Anopheles atroparvus, An. claviger, Culiseta longiareolata, Cs. annulata, and Aedes caspius. Dominant species were Cx. pipiens and Cs. longiareolata. Aedes caspius was found in only once, near its natural breeding habitat. Considering the recent discovery of an established population of Ae. albopictus in Catalonia, the increasing commerce of used tires in Spain for recycling, storage, and recapping might greatly contribute to the rapid spread of this species across the Iberian Peninsula.  相似文献

Surveillance of adult mosquitoes was conducted at 29 US military installations and training sites in six provinces in the Republic of Korea during 2003. Adult mosquitoes were collected in New Jersey light traps and dry ice‐baited New Jersey light traps from 1 May to 15 October. Mosquito surveillance was conducted to determine threshold levels to initiate pesticide applications and identify malaria infection rates at selected army installations and training sites. A total of 42 024 adult mosquitoes (32 594 females [77.6%] and 9430 males [22. 4%]) comprising 14 species and Anopheles sinensis s.l. (a complex of five species), representing seven genera, were collected. The most common species were members of the Anopheles sinensis Wiedemann complex (54.9%), followed by Aedes vexans nipponii (Theobald) (19.0%), Culex pipiens Coquillett (14.3%) and Culex tritaeniorhynchus Giles (10.6%). BackgroundAn important function of the fat body in adult female mosquitoes is the conversion of blood meal derived amino acids (AA) into massive amounts of yolk protein precursors. A highly efficient transport mechanism for AAs across the plasma membrane of the fat body trophocytes is essential in order to deliver building blocks for the rapid synthesis of large amounts of these proteins. This mechanism consists in part of AA transporter proteins from the solute carrier family. These transporters have dual function; they function as transporters and participate in the nutrient signal transduction pathway that is activated in the fat body after a blood meal. In this study we focused on the solute carrier 7 family (SLC7), a family of AA transporters present in all metazoans that includes members with strong substrate specificity for cationic AAs.Methodology/principal findingsWe identified 11 putative SLC7 transporters in the genome sequence of Aedes aegypti. Phylogenetic analysis puts five of these in the cationic AA transporter subfamily (CAT) and six in the heterodimeric AA transporter (HAT) subfamily. All 11 A. aegypti SLC7 genes are expressed in adult females. Expression profiles are dynamic after a blood meal. We knocked down six fat body-expressed SLC7 transporters using RNAi and found that these ‘knockdowns’ reduced AA-induced TOR signaling. We also determined the effect these knockdowns had on the number of eggs deposited following a blood meal.Conclusions/significanceOur analysis stresses the importance of SLC7 transporters in TOR signaling pathway and mosquito reproduction.  相似文献

Endosymbiotic Wolbachia bacteria are potent modulators of pathogen infection and transmission in multiple naturally and artificially infected insect species, including important vectors of human pathogens. Anopheles mosquitoes are naturally uninfected with Wolbachia, and stable artificial infections have not yet succeeded in this genus. Recent techniques have enabled establishment of somatic Wolbachia infections in Anopheles. Here, we characterize somatic infections of two diverse Wolbachia strains (wMelPop and wAlbB) in Anopheles gambiae, the major vector of human malaria. After infection, wMelPop disseminates widely in the mosquito, infecting the fat body, head, sensory organs and other tissues but is notably absent from the midgut and ovaries. Wolbachia initially induces the mosquito immune system, coincident with initial clearing of the infection, but then suppresses expression of immune genes, coincident with Wolbachia replication in the mosquito. Both wMelPop and wAlbB significantly inhibit Plasmodium falciparum oocyst levels in the mosquito midgut. Although not virulent in non-bloodfed mosquitoes, wMelPop exhibits a novel phenotype and is extremely virulent for approximately 12-24 hours post-bloodmeal, after which surviving mosquitoes exhibit similar mortality trajectories to control mosquitoes. The data suggest that if stable transinfections act in a similar manner to somatic infections, Wolbachia could potentially be used as part of a strategy to control the Anopheles mosquitoes that transmit malaria.  相似文献

Abstract

Metabolic demands such as growth and reproduction of insects relate directly to the fat body. This organ is responsible for the biosynthesis and storage of nutrients, and participates in the neutralization and detoxification of xenobiotics. Studies show a relationship between its ability to accumulate nutrients and activities, such as diapause and reproduction in the beetle Anthonomus grandis, an important pest of cotton plants. However, there are no detailed studies on the structure and histochemistry of the fat body in this insect. We describe the ultrastructure and histochemistry of the fat body in A. grandis and discuss some of its characteristic features. The fat body is composed of a single cell type, the trophocyte, which is a voluminous cell with a spherical or bilobed nucleus, and cytoplasm containing several lipid droplets, glycogen granules (electron-dense), and protein granules. These trophocytes have few areas of contact with each other and form lobes, enclosed by connective tissue. A high metabolic activity of the fat body is deduced by the intense histochemical staining for protein and polysaccharide compounds, demonstrating that specific processes, such as the synthesis and secretion of vitellogenin, could be a target for more specific studies on methods to control this insect.  相似文献

Experiments were conducted on the artificial infection of larvae of 8 species of bloodsucking mosquitoes (Aedes diantaeus, A. punctor, A. cinereus, A. flavescens, A. aegypti, Culex pipiens, Anopheles messeae, Anopheles atroparvus) with spores of Amblyospora opacita var. sibirica, Parathelohania costatus sp. n., P. obesa, P. anophelis. A programme of surveillance was initiated in 1992, in the French Riviera, to detect a possible introduction of Aedes albopictus from Italy and to prevent nuisances caused by mosquitoes in the touristic localities of the C?te d'Azur. In five years, numerous mosquito breeding places have been located. Nine species have been collected: Anopheles claviger, An. plumbeus, Aedes geniculatus, Ae. vittatus, Culex hortensis, Cx. impudicus, Cx. pipiens, Culiseta fumipennis, Cs. longiareolata but no Ae. albopictus was found. Nuisances were mainly due to hypogean populations of Cx. pipiens. Breeding places in urban sites have been controlled or suppressed. The discovery of an important larval population of An. plumbeus in an urban area might further stress the importance of this species already suspected to transmit indigenous malaria in cities.  相似文献

Cardinal differences in the spatial organization of polytene chromosomes in the nuclei of salivary gland cells (malphigian tubules) and nurse ovarian cells in malarial mosquitoes of Anopheles maculipennis complex are found. The chromosomes have typical chromocentric organization in salivary gland cells and malphigian tubules. In contrast to these cells, the trophocytes have no single chromocentre and some of their chromosomes are rigidly attached to the nucleus envelope with the centromeric parts or with the particular loci. The loci-specificity of the X-chromosome intercalary parts' attachment to the nucleus envelope is found in Anopheles messeae.  相似文献
