Urban lakes belong to various groups, for example from “young” to “old” with regard to development, and from “natural” to “anthropogenic” with respect to transformations due to human activity. The majority of these lakes are eutrophic and polytrophic, but special attention should be paid to mesotrophic ones, with relatively unchanged vegetation, with species of the class Charetea, e.g. Lake Redykajny (43.3% of the phytolittoral) or Lake Tyrsko (44.0% of the phytolittoral).  相似文献

To determine the renal effects of cadmium (Cd) in older animals, we administered subcutaneously a single dose of cadmium, 3.0 mg/kg/BW, to Syrian hamsters aged 16 wk (“young”) and 60 wk (“old”). Marked morphologic changes in the kidney and renal dysfunction were observed, especially in the older animals. The concentration of MDA in the renal cortex was significantly increased only in young hamsters treated with cadmium. Concentrations of glutathione (GSH) in the renal cortex were increased in the old hamsters on d 6. Increased levels of renal MDA after cadmium treatment may induce the production of GSH in the kidney thus preventing renal damage. Aging can increase the susceptibility to the renal effects of cadmium.  相似文献

Summary The action of hydralazine on collagen prolyl hydroxylation was studied in a cell culture system using WI-38 fibroblasts. The prolyl hydroxylation level was determined by a method involving the digestion of collagen by bacterial collagenase and the examination of specific peptides. The presence of low concentrations of hydralazine (0.2 mM) in both “young” and “old” fibroblast cultures strongly inhibited collagen prolyl hydroxylation. The degree of inhibition was greater in serum-deficient cultures. No significant improvement in the degree of hydroxylation was observed by increasing either ascorbate or iron levels in the hydralazine-containing cultures in which hydroxylation was inhibited. Some of the reported side effects of hydralazine seen in patients might be related to its inhibitory effects on mixed function oxidative (MFO) hydroxylation systems. While the ascorbate dependence of the prolyl hydroxylase system of WI-38 decreased with the “age” of the culture, hydralazine inhibition of hydroxylation was dramatic with cultures of all “ages”. Understanding the effect of protection measures on recovery of endangered populations is crucial for assessing the efficiency of management plans. Following the ban of DDT, PCB and other detrimental chemicals in the 1970s, the German white-tailed eagle population recovered rapidly. Using nest monitoring data, dead recovery data and population dynamics models, we examined both short and long-term viabilities of the population. Between 1991 and 2005 the proportion of breeding pairs with an active nest among territorial pairs was α = 0.88 ± 0.007. Annual breeding success was γ = 0.62 ± 0.014. Mean fledged brood size was b = 1.50 ± 0.010 fledged/successful breeding pair. The analysis of dead recovery data of 1,273 individuals ringed as nestling between 1991 and 2006 allowed survival rates to be estimated. We needed to distinguish between “young” (5 years old) and “old” (older than 5 years) adults. Furthermore the annual survival rate was higher for juvenile and immature birds (0.921 ± 0.024) than for adults (s “young” = 0.760 ± 0.097 and s “old” = 0.875 ± 0.079), a pattern presumably explained by territorial fights. Finally, an age structured model was used to examine the joint effect of population regulation, environmental and demographic stochasticities. All results indicated that only pessimistic scenarios could lead to substantial extinction probabilities within 100 years. We conclude that management measures were successful in removing the main extinction factors of the population and that future conservation efforts need to focus on the remaining ultimate extinction causes.  相似文献

目的:观察肠淋巴液引流对失血性休克大鼠红细胞流变性指标以及血液黏度的作用。方法:Wistar雄性大鼠均分为假休克组、休克组(复制失血性休克模型)、引流组(复制失血性休克模型,自低血压1 h引流休克肠淋巴液)。在低血压3 h或相应时间,经腹主动脉取血,检测红细胞参数、红细胞电泳、红细胞沉降率(ESR)以及血液黏度,计算红细胞聚集指数、红细胞变形指数。结果:与假休克组比较,休克组红细胞数量、红细胞比积(HCT)、血红蛋白(Hb)、平均红细胞血红蛋白浓度(MCHC)、红细胞电泳率与迁移率、红细胞变形指数、全血黏度、全血低切与高切相对黏度和还原黏度显著降低,休克组平均红细胞体积、红细胞电泳时间、ESR、血沉方程K值与校正K值、红细胞聚集性指数、血浆黏度显著升高;引流组MCHC、红细胞电泳率与迁移率、全血黏度、全血低切与高切还原黏度均显著降低,引流组红细胞体积分布宽度(RDW-SD)显著增加。同时,引流组HCT、RDW-SD、红细胞变形指数、全血黏度、全血低切与高切相对黏度显著高于休克组;ESR、血沉方程K值与校正K值、红细胞聚集性指数、血浆黏度显著低于休克组。结论:休克肠淋巴液引流可改善失血性休克大鼠红细胞流变行为,从而改善血液流变性。  相似文献

肠系膜淋巴管结扎对急性失血大鼠红细胞流变性的影响   总被引:1,自引:0,他引:1  
目的:观察结扎肠系膜淋巴管(MLD)对急性失血大鼠红细胞流变性的影响。方法:20只Wistar雄性大鼠随机均分为失血组与结扎组。所有大鼠经右侧颈总动脉匀速放血(失血量为全血量的1/4),结扎组失血后结扎MLD,失血组仅在MLD下穿线。记录24h存活情况。24h后,将存活大鼠经左侧颈总动脉迅速放血,测定实验前后的红细胞沉降率(ESR)、红细胞电泳、红细胞压积(Hct),计算红细胞聚集与变形指数。结果:急性失血后24h,结扎组大鼠存活情况(9只)略好于失血组(6只)。急性失血后24h,与实验前相比,失血组与结扎组的ESR、血沉方程K值、校正K值、红细胞电泳时间均显著升高或延长,红细胞变形性降低,失血组的红细胞聚集指数显著升高、红细胞电泳长度与迁移率均显著降低;结扎组的ESR、血沉方程K值、校正K值、红细胞聚集指数、电泳时间较失血组显著降低,红细胞电泳长度与迁移率、变形性较失血组显著升高。结论:急性失血导致大鼠红细胞聚集性升高、红细胞电泳能力及变形性降低,结扎MLD可改善急性失血导致的红细胞流变性异常。  相似文献

The proteomic analysis has shown that the red cell membrane contains several kinases and phosphatases. Therefore the aim of this study was to investigate the role of protein kinases of human red cell membrane in deformability and aggregation alterations. The exposure of red blood cells (RBCs) to some chemical compounds has led to a change in the RBC microrheological properties. When forskolin (10 μM), an adenylyl cyclase (AC) and a protein kinase A (PKA) stimulator were added to RBC suspension, the RBC deformability (RBCD) was increased by 20% (p<0.05). Somewhat more significant deformability rise appeared after RBC incubation with dB-AMP (by 26%; p<0.01). The red cell aggregation (RBCA) was significantly decreased under these conditions (p<0.01). Markedly less changes of deformability were found after RBC incubation with protein kinase stimulator C (PKC)—phorbol 12-myristate 13-acetate (PMA). This drug reduced the red cell aggregation only slightly. The red cell tyrosine phosphotase activity was changed by N-vanadat and a significant RBCD rise and RBCA lowering were obtained. The similar effect was found when the cells were incubated with cisplatin as a tyrosine protein kinase (TPK) activator. It is important to note that a selective TPK inhibitor—lavendustin eliminated the above mentioned effects.  相似文献

Microrheologic properties (aggregation and deformability) were examined in young (upper fraction) and old (bottom fraction) erythrocytes separated by density gradient centrifugation at 30000 g. A significant difference in microrheologic properties of the young and old erythrocytes was revieled. Erythrocyte aggregation was studied in the plasma and serum. Aggregation of the young and old cells in plasma was higher than in serum, which points to the importance of the presence of fibrinogen in the aggregation medium. The study of aggregation kinetics showed that the absence of fibrinogen in the medium produced an insignificant effect on the rate of aggregate formation, but it dramatically affected the degree of aggregation (12 and 88% difference between aggregate number in the plasma and serum, respectively). In young erythrocytes, the difference of the degree of aggregation and kinetics of aggregate formation in the plasma and serum was not so pronounced (32 and 43%, respectively). Analysis of correlations between aggregation and deformation of the young and old erythrocytes revealed more pronounced interrelations of these parameters in the plasma compared to serum, indicating an important role of fibrinogen for the manifestation of microrheologic properties of erythrocytes. These interrelations were most pronounced during aggregate formation, which supported the bridging theory of aggregation.  相似文献

Gramicidin S (GS) is a cyclo-decapeptide antibiotic with wide Gram+ and Gram− antimicrobial spectrum. However, its therapeutic application is very limited due to hemolytic activity of GS. The presence of cholesterol defines one of the most significant differences between eukaryotic plasma membranes and bacterial inner membranes. To find out the cholesterol effect on the GS hemolytic efficiency we compared GS-induced hemolysis of erythrocytes extracted from the blood of healthy donors against donors with atherosclerosis, “naturally” enriched with cholesterol. Our results show that increased cholesterol levels significantly attenuates yet does not abolishes the GS hemolytic activity. High levels of cholesterol content in erythrocyte membranes results in a decrease in the membrane fluidity and deformability leading to a decrease in the rate of GS interaction with membranes. The results obtained confirm that hydrophobic as well as electrostatic interactions must be involved in the binding of GS to cell membranes. Lipid peroxidation occurring within atherosclerotic erythrocytes leads to considerable decrease in the degree of GS-induced erythrocyte hemolysis in vitro. These results can be applied to the rational design of GS analogs with increased antibacterial efficiency but reduced hemolytic activity.  相似文献

In experiments of Wistar male rats, changes are studied of erythrocyte hematological, biochemical (activitities of transport ATPases), and rheological properties (capability for aggregation and deformability) 7 days after bloodletting of 12-15 % of the total blood mass. It has been shown that alongside with an elevation of erythrocyte volume and of the number of immature cells - reticulocytes, there was a statistically significant increase of Na,K-ATPase and Ca-ATPase activities in the whole erythrocytes and their membrane preparations - ghosts, the increment of activity in the case of Na,K-ATPase being essentially higher in the whole cells. This indicates the appearance of an enzyme activator inside the erythrocytes. There are also revealed a decrease of firmness of erythrocyte aggregates, a deceleration of spontaneous aggregation, and an increase of index of erythrocyte deformability. The conclusion is made that changes of erythrocyte rheological properties are interconnected with changes of the Na,K-ATPase activity and are directed to optimization of blood circulation in large vessels and capillary network.  相似文献

The relationship between hemorheology, erythrocyte ATP and 2,3-diphosphoglycerate (2,3-DPG) concentrations, and von Willebrand factor antigen was studied in athletes and peripheral arterial disease patients. Lower blood viscosity, mainly due to a higher erythrocyte deformability, was found in athletes compared to control subjects. Higher 2,3-DPG/Ht levels in athletes were correlated with blood viscosity, erythrocyte deformability, the rigidity index, and erythrocyte suspension viscosity at low shear stress. It is suggested that these relationships might be determined by the predominance of immature erythrocytes in the blood circulation of the athletes. In the group of patients, a decrease in ATP/Ht was related to increased erythrocyte aggregation and a higher erythrocyte suspension viscosity. Moreover, the concentration of von Willebrand factor was positively correlated with the erythrocyte aggregation index, erythrocyte suspension viscosity, and plasma viscosity. The results show that alterations in erythrocyte and plasma rheology may be involved in the modification of the functional state of the vascular endothelium and the development of atherosclerosis.  相似文献

The ability of red blood cells (RBCs) for the reversible change of their shape under passing through capillaries in microcirculation mainly depends on membrane elasticity of these cells. Phosphorylation of some membrane proteins can result in the changes of microrheological red blood cell properties. Here we show a significant increase in RBC deformability (RBCD) after incubation with isoproterenol (10−6 M). Red blood cell aggregation (RBCA) decreased under these conditions only slightly. When forskolin (10 μM), an adenylyl cyclase (AC) stimulator, was added to the RBC suspension, RBCD increased significantly (p < 0.05). Some more changes of deformability were found after incubation of RBC with stable penetrating analog of cyclic adenosine phosphate (cAMP), dibutyryl-cAMP, (dB-cAMP, 50 μM) and after phosphodiesterase (PDE) activity inhibition with Vinpocetine, Rolipram, or IBMX. It was found that Gs-proteins inhibitor Clonidine and specific Gi-protein stimulator Mastaparan 7 increased both RBCD and RBCA. On the whole, the data clearly show that the RBC aggregation and deformation changes are related with activation of the different intracellular signaling pathways. We suppose that RBCD increase was mainly associated with activation of the adenylyl-cyclase-cAMP system.  相似文献

Den sharing by conspecific spiny lobsters (aggregation) is modulated by chemical attraction but may confer several, not necessarily mutually exclusive, antipredator byproduct benefits: a “guide effect”, which only benefits the individual attracted to a sheltered conspecific; a “dilution effect”, which reduces per-capita risk of predation simply through aggregation; or active “group defense”. Each potential benefit has a different set of predictors (relationships between aggregation and conspecific or predator densities), but conflicting results could suggest the simultaneous operation of more than one benefit. These predictions were tested for coexisting Panulirus guttatus (a reef-obligate) and Panulirus argus (a temporary reef-dweller) using data collected during 11 surveys on fixed sites over a coral reef in Mexico. P. guttatus greatly outnumbered P. argus, but P. argus showed a greater tendency to aggregate. All three benefits of den sharing operated for the more social P. argus, with “group defense” being of the most benefit for larger individuals, and the “guide” and “dilution” effects for smaller individuals recently immigrating into the reef habitat and sharing dens with larger conspecifics. P. guttatus did not display “group defense” and its aggregations appeared to be modulated by the interplay between attraction and aggressive behaviors. This species relied more on solitary crypticity, especially at larger sizes, but appeared to benefit from a “guide effect” at high conspecific densities. In experimental tanks, each species tended to aggregate when tested separately, but when tested jointly, aggregation among P. guttatus was significantly reduced. The experimental results reflect the differential patterns of aggregation between the fore-reef, where P. guttatus dominated, and the back-reef, where coexistence of both species was greater.  相似文献

The mechanical properties of vertebrate erythrocytes depend on their cytoskeletal protein networks. Membrane skeleton proteins spectrin and protein 4.1 (4.1R) cross-link with actin to maintain membrane stability under mechanical stress. Phosphorylation of 4.1R alters the affinity of 4.1R for spectrin–actin binding and this modulates the mechanical properties of human erythrocytes. In this study, phorbol 12-myristate-13-acetate (PMA)-induced phosphorylation of 4.1R was tested, erythrocyte deformability was determined and the erythrocyte elastic modulus was detected in human, chick, frog and fish. Furthermore, amino acid sequences of the functionally important domains of 4.1R were analyzed. Results showed that PMA-induced phosphorylation of 4.1R decreased erythrocyte deformability and this property was stable after 1 h. The values of Young’s modulus alteration gradually decreased from human to fish (0.388±0.035 kPa, 0.219±0.022 kPa, 0.191±0.036 kPa and 0.141±0.007 kPa). Ser-312 and Ser-331 are located within the consensus sequence recognized by protein kinase C (PKC); however, Ser-331 in zebrafish was replaced by Ala-331. The sequence of the 8 aa motif from vertebrate 4.1R showed only one amino acid mutation in frog and numerous substitutions in fish. Analyses of Young’s modulus suggested that the interaction between 4.1R with the spectrin–actin binding domain may have a special relationship with the development of erythrocyte deformability. In addition, amino acid mutations in 4.1R further supported this relationship. Thus, we hypothesize that alteration of membrane skeleton protein binding affinity may play a potential role in the development of erythrocyte deformability, and alteration of Young’s modulus values may provide a method for determining the deformability development of vertebrate erythrocytes.  相似文献

Properties of erythrocyte surface were investigated for patients with ischemic heart disease in the course of treatment with the UV-irradiated blood autotransfusion (UVIBA). Application of methods of light-scattering, photometry and cytochemistry revealed rapid and significant changes in deformability and aggregation properties of the erythrocytes immediately following each UVIBA procedure, which was accompanied by considerable blood viscosity decrease.  相似文献

The normal deformability of erythrocytes plays an important role in ensuring blood mobility, erythrocyte longevity, and microcirculation, which is the ability of erythrocytes to change shapes in response to external forces. However, the effects of curcumin extracts on the deformability of erythrocytes have not yet been evaluated. Accordingly, in this study, we explored the effects of pre-treatment with curcumin extract on erythrocyte deformation and erythrocyte band 3 (SLC4A1; EB3) expression. We also evaluated the associations between EB3 expression and erythrocyte deformability induced by hydrogen peroxide. Blood samples were divided into the control group, pre-treatment group (treated with curcumin extract or vitamin C), and negative control group, and oxidant stress parameters, antioxidant status, erythrocyte deformability and elasticity, and EB3 modifications were evaluated using immunoblotting and immunofluorescence staining. Hydrogen peroxide significantly increased oxidative stress parameters, modulus elasticity values and clustered EB3 levels and induced conjugation of membrane proteins to form high-molecular-weight complexes (p < 0.05). Erythrocyte deformability and elasticity were significantly decreased in the treated groups compared with those in the control group. Overall, our findings suggested that pre-treatment with curcumin extracts increased antioxidant status, reduced EB3 cross-linking, and improved erythrocyte deformability, to an even better extent than vitamin C. These results provide important insights into the effects of treatment with curcumin extracts on erythrocyte damage and suggest that curcumin may have applications in antioxidant therapy.  相似文献

The objective of this study was to assess the effects of nickel chloride on human and rainbow trout erythrocytes in vitro. The cells were incubated with 0, 0.5 and 1 mM nickel chloride for 1 h at pH 7.40 and 25°C, then K+ efflux, SO42− uptake and GSH and GSSG concentrations were measured. In both kind of cells, “high concentration” nickel treatment increased KCl efflux with respect to the control. The SO42− uptake was not significantly different at “low nickel concentration” but was lower in erythrocytes treated with 1 mM nickel chloride; the rate constant of SO42− uptake decreased by 35% in human erythrocytes and by 44% in fish erythrocytes. Nickel chloride also acts on cellular metabolism and in particular on erythrocyte glutathione peroxidase with consequent increase in oxidative stress; the data show a significant decrease in intracellular GSH in both human (25%) and fish erythrocytes (18%) after treatment with nickel chloride, with concomitantly high GSSG concentrations and lower GSH/GSSG ratios.  相似文献

Seasonal variations in the gonad development and sex ratio of copepodite stage V (CV) and adults were examined from February to November in order to understand the reproductive cycle and the life history of Calanus glacialis in the White Sea. Gonad maturation, sexual differentiation and moulting to adults take place during the 2nd year of development. Energy accumulation takes place in the spring and summer of the 2nd year. The following autumn/winter is the major period of CV maturation, which occurs independent of food supply. Maturation of males precedes that of females by 2–3 months. The maximum proportions of CV and adult males are found in the population in October and November. The onset of female maturation is observed in February and March, ca. 2 months prior to the spring phytoplankton bloom. Reproduction takes place between April and June. Its termination in the second half of June coincides with the warming of the surface water layer where egg laying takes place. Variations in the gonad morphology throughout the year suggest long life spans and iteroparity of females of C. glacialis in the White Sea. Many of them survive for several months after reproduction and are able to overwinter again. Therefore, females with different life histories co-occur in the population in winter: “young” females recently moulted from the overwintering CVs, and “old” females which have spawned at least once in their life, after which they return to overwintering conditions. In contrast, males have shorter life spans of 3–4 months resulting in a sex ratio skewed toward females at all seasons. Accepted: 27 April 1999  相似文献
