A series of experiments were conducted to characterize the water stress-induced changes in the activities of RuBP carboxylase (RuBPCO) and sucrose phosphate synthase (SPS), photosystem 2 activity, and contents of chlorophylls, carotenoids, starch, sucrose, amino acids, free proline, proteins and nucleic acids in mulberry (Morus alba L. cv. K-2) leaves. Water stress progressively reduced the activities of RuBPCO and SPS in the leaf extracts, the chlorophyll content, and PS2 activity in isolated chloroplasts. Plants exposed to drought showed lower content of starch and sucrose but higher total sugar content than control plants. While the soluble protein content decreased under water stress, the amino acid content increased. Proline accumulation (2.5-fold) was noticed in stressed leaves. A reduction in the contents of DNA and RNA was observed. Reduced nitrogen content was associated with the reduction in nitrate reductase activity. SDS-PAGE protein profile showed few additional proteins (78 and 92 kDa) in the water stressed plants compared to control plants.  相似文献

The control of photosynthetic starch/sucrose formation in leaves of soybean (Glycine max L. Merr.) cultivars was studied in relation to stage of plant development, photosynthetic photoperiod, and nitrogen source. At each sampling, leaf tissue was analyzed for starch content, activities of sucrose-metabolizing enzymes, and labeling of starch and sucrose (by 14CO2 assimilation) in isolated cells. In three of the four varieties tested, nodulated plants had lower leaf starch levels and higher activities of sucrose phosphate synthetase (SPS), and isolated mesophyll cells incorporated more carbon (percentage of total 14CO2 fixed) into sucrose and less into starch as compared to nonnodulated (nitrate-dependent) plants. The variation among cultivars and nitrogen treatments observed in the activity of SPS in leaf extracts was positively correlated with labeling of sucrose in isolated cells (r = 0.81) and negatively correlated with whole leaf starch content (r = −0.66). The results suggested that increased demand for assimilates by nodulated roots may be accommodated by greater partitioning of carbon into sucrose in the mesophyll cells. We have also confirmed the earlier report (Chatterton, Silvius 1979 Plant Physiol 64: 749-753) that photoperiod affects partitioning of fixed carbon into starch. Within two days of transfer of nodulated soybean Ransom plants from a 14-hour to a 7-hour photoperiod, leaf starch accumulation rates doubled, and this effect was associated with increased labeling of starch and decreased labeling of sucrose in isolated cells. Concurrently, activities of SPS, sucrose synthase, and uridine diphosphatase in leaves were decreased.  相似文献

The grapevine (Vitis vinifera L. cv. Riesling) plants subjected to water deficit were studied for changes in relative water content (RWC), leaf dry mass, contents of chlorophyll (Chl), total leaf proteins, free amino acids, and proline, and activities of ribulose-1,5-bisphosphate carboxylase (RuBPC), nitrate reductase (NR), and protease. In water-stressed plants RWC, leaf dry matter, Chl content, net photosynthetic rate (P N), and RuBPC and NR activities were significantly decreased. The total leaf protein content also declined with increase in the accumulation of free amino acids. Concurrently, the protease activity in the tissues was also increased. A significant two-fold increase in proline content was recorded.  相似文献

Changes of leaf pigments, ribulose-1,5-bisphosphate carboxylase (RuBPC) and photosynthetic efficiency were examined in grapevine (Vitis vinifera L.) plants grown under ambient irradiation (maximum daily PAR = 1500 micromol m(-2) s(-1)) for 7 days to low night temperature (LNT) of 5 degrees C (daily from 18:00 to 06:00). The contents of chlorophyll (Chl) and carotenoids (Car) per fresh mass were lower in LNT leaves than in control leaves. The contents of alpha + beta carotene and lutein-5,6-epoxide remained unaffected, but the de-epoxidation state involving the components of xanthophyll cycle increased. RuBPC activity and soluble proteins were also significantly reduced in LNT leaves. In isolated thylakoids, a marked inhibition of whole chain (PS I + PS II) and PS II activity were observed in LNT leaves. Smaller inhibition of PS I activity was observed in LNT leaves. The artificial exogenous electron donors, MnCl2, DPC and NH2OH did not restored the loss of PS II activity in LNT leaves. The same results were obtained when F(v)/F(m) was evaluated by Chl fluorescence measurements. The marked loss of PS II activity in LNT leaves was due to the marked loss of D1 protein which was determined by immunological studies.  相似文献

The expression of a sucrose-phosphate synthase (SPS) gene from maize (Zea mays, a monocotyledon) in tomato (Lycopersicon esculentum, a dicotyledon) resulted in marked increases in extractable SPS activity in the light and the dark. Diurnal modulation of the native tomato SPS activity was found. However, when the maize enzyme was present the tomato leaf cells were unable to regulate its activation state. No detrimental effects were observed and total dry matter production was unchanged. However, carbon allocation within the plants was modified such that in shoots it increased, whereas in roots it decreased. There was, therefore, a change in the shoot:root dry weight ratio favoring the shoot. This was positively correlated with increased SPS activity in leaves. SPS was a major determinant of the amount of starch in leaves as well as sucrose. There was a strong positive correlation between the ratio of sucrose to starch and SPS activity in leaves. Therefore, SPS activity is a major determinant of the partitioning of photosynthetically fixed carbon in the leaf and in the whole plant. The photosynthetic rate in air was not significantly increased as a result of elevated leaf SPS activity. However, the light- and CO2-saturated rate of photosynthesis was increased by about 20% in leaves expressing high SPS. In addition, the temporary enhancement of the photosynthetic rate following brief exposures to low light was increased in the high SPS plants relative to controls. We conclude that the level of SPS in the leaves plays a pivotal role in carbon partitioning. Furthermore, high SPS levels have the potential to boost photosynthetic rates under favorable conditions.  相似文献

The effects of short-term exposure to chilling temperature (10 °C) on sucrose synthesis in leaves of the cold-tolerant sugarcane cultivars Saccharum sinense R. cv. Yomitanzan and Saccharum sp. cv. NiF4, and the cold-sensitive cultivar S. officinarum L. cv. Badila were studied. Plants were grown at day/night temperatures of 30/25 °C, and then shifted to a constant day/night temperature of 10 °C. After 52-h exposure to the chilling temperature, sucrose content in the leaves of NiF4 and Yomitanzan showed a 2.5- to 3.5-fold increase relative to that of the control plants that had been left on day/night temperatures of 30/25 °C. No such increase was observed in Badila leaves. Similarly, starch content in the leaves of NiF4 and Yomitanzan was maintained high, but starch was depleted in Badila leaves after the 52-h exposure. During the chilling temperature, sucrose phosphate synthase (SPS; E.C.2.4.1.14) activity was relatively stable in the leaves of NiF4 and Yomitanzan, whereas in Badila leaves SPS activity significantly decreased. There was no significant change in cytosolic fructose-1,6-bisphosphatase activity for the three cultivars at the chilling temperature. This supports the hypothesis that: (1) on exposure to chilling temperature, sucrose content in sugarcane leaves is determined by the photosynthetic rate in the leaves, and is not related to SPS activity; (2) SPS activity in sugarcane leaves at chilling temperature is to be determined by sugar concentration in the leaves.  相似文献

Rice (Oryza sativa L. cv. IR-30) was grown season-long in outdoor, controlled-environment chambers at 33 Pa CO2 with day/night/paddy-water temperatures of 28/21/25 °C, and at 66 Pa CO2 with five different day/night/paddy-water temperature regimes (25/18/21, 28/21/25, 31/24/28, 34/27/31 and 37/30/34 °C). Sucrose phosphate synthase (SPS) activities in leaf extracts at 21, 48 and 81 days after planting (DAP) were assayed under saturating and selective (limiting) conditions. Diel SPS activity data indicated that rice SPS was light regulated; with up to 2.2-fold higher rates during the day. Throughout the growth season, leaf SPS activities were up-regulated in the CO2-enriched plants, averaging 20 and 12% higher than in ambient-CO2 grown plants in selective and saturating assays, respectively. Similarly, SPS activities increased 2.4% for each 1 °C rise in growth temperature from 25 to 34 °C, but de creased 11.5% at 37 °C. Leaf sucrose content was higher, and mirrored SPS activity better, than starch, although starch was more responsive to CO2 treatment. Leaf sucrose and starch contents were significantly higher throughout the season in plants at elevated CO2, but the N content averaged 6.5% lower. Increasing growth temperatures from 25 to 37 °C caused a linear decrease (62%) in leaf starch content, but not in sucrose. Consequently, the starch:sucrose ratio declined with growth temperature. The data are consistent with the hypothesis that the up-regulation of leaf SPS may be an acclimation response of rice to optimize the utilization and export of organic-C with the increased rates of inorganic-C fixation in elevated CO2 or temperature growth regimes.  相似文献

The functional activities of the photosynthetic apparatus of two grapevine genotypes (Vitis vinifera L. cvs. Müller-Thurgau and Lagrein) were investigated after low night temperature (LNT) treatment for 7 d. LNT caused important reductions of the net photosynthetic rate (PN) of Lagrein plants due to non-stomatal components. These non-stomatal effects were not evident in Müller-Thurgau. At LNT treatment, the contents of photosynthetic pigments decreased significantly in Lagrein, but in Müller-Thurgau the contents of chlorophyll (Chl) remained unchanged whereas the contents of carotenoids (Car) increased. An increase and decrease of Chl a/b was shown in Mü ller-Thurgau and Lagrein stressed plants, respectively. RuBPC activity and content of soluble proteins were also significantly reduced in Lagrein. Under LNT treatment, photosystem (PS) 2 was markedly more inhibited in Lagrein than in Müller-Thurgau. The decrease in PS 2 activity in Lagrein was mostly due to the marked loss of D1, 47, 43, 33, 28-25, 23 and 17 kDa proteins determined by immunological and SDS-PAGE studies.  相似文献

Experiments were conducted in controlled growth chambers to evaluate how increase in CO2 concentration affected sucrose metabolizing enzymes, especially sucrose phosphate synthase (SPS; EC 2.4.1.14) and sucrose synthase (SS; EC 2.4.1.13), as well as carbon metabolism and partitioning in a tropical epiphytic orchid species (Oncidium goldiana). Response of ribulose‐1,5‐bisphosphate carboxylase/oxygenase (Rubisco; EC 4.1.1.39) to elevated CO2 was determined along with dry mass production, photosynthesis rate, chlorophyll content, total nitrogen and total soluble protein content. After 60 days of growth, there was a 80% and 150% increase in dry mass production in plants grown at 750 and 1 100 μl l?1 CO2, respectively, compared with those grown at ambient CO2 (about 370 μl l?1). A similar increase in photosynthesis rate was detected throughout the growth period when measured under growth CO2 conditions. Concomitantly, there was a decline in leaf Rubisco activity in plants in elevated CO2 after 10 days of growth. Over the growth period, leaf SPS and SS activities were up‐regulated by an average of 20% and 40% for plants grown at 750 and 1100 μl l?1 CO2, respectively. Leaf sucrose content and starch content were significantly higher throughout the growth period in plants grown at elevated CO2 than those at ambient CO2. The partitioning of photosynthetically fixed carbon between sucrose and starch appeared to be unaffected by the 750 μl l?1 CO2 treatment, but it was favored into starch under the 1 100 μl l?1 CO2 condition. The activities of SPS and SS in leaf extracts were closely associated with photosynthetic rates and with partitioning of carbon between starch and sucrose in leaves. The data are consistent with the hypothesis that the up‐regulation of leaf SPS and SS might be an acclimation response to optimize the utilization and export of organic carbon with the increased rate of inorganic‐carbon fixation in elevated CO2 conditions.  相似文献

Three clones of tobacco transformed with the T-DNA of Agrobacterium rhizogenes Ri plasmid A4b cultivated in vitro on a solid agar medium neither showed pronounced morphological diversity nor significantly differed in chlorophyll (Chl) contents from control plants. However, the transformation caused a 27 to 83 % decay in leaf oxygen evolution and in both ribulose-1,5-bisphosphate carboxylase (RuBPC) and phosphoenolpyruvate carboxylase (PEPC) activities. Therefore, the transformation brought about a reduction of active PEPC as well as activable RuBPC amounts in plant tissues. Individual clones did not mutually differ. In tobacco transformed with A. rhizogenes strain TR101 and grown in soil only the mean leaf area tended to reduce. Chl contents, Chl a/b ratio, oxygen evolution, and activities of both RuBPC and PEPC were insignificantly affected by the transformation.  相似文献

We generated transgenic tobacco plants with high levels of fructose-1,6-bisphosphatase expressing cyanobacterialfructose-1,6-/sedoheptulose-1,7-bisphosphatase in the cytosol. At ambient CO2 levels (360 ppm), growth, photosynthetic activity, and fresh weight were unchanged but the sucrose/hexose/starch ratio was slightly altered in the transgenic plants compared with wild-type plants. At elevated CO2 levels (1200 ppm), lateral shoot, leaf number, and fresh weight were significantly increased in the transgenic plants. Photosynthetic activity was also increased. Hexose accumulated in the upper leaves in the wild-type plants, while sucrose and starch accumulated in the lower leaves and lateral shoots in the transgenic plants. These findings suggest that cytosolic fructose-1,6-bisphosphatase contributes to the efficient conversion of hexose into sucrose, and that the change in carbon partitioning affects photosynthetic capacity and morphogenesis at elevated CO2 levels.  相似文献

Experiments were conducted with vegetative soybean plants (Glycine max [L.] Merr., `Ransom') to determine whether the activities in leaf extracts of key enzymes in sucrose metabolism changed during the daily light/dark cycle. The activity of sucrose-phosphate synthase (SPS) exhibited a distinct diurnal rhythm, whereas the activities of UDP-glucose pyrophosphorylase, cytoplasmic fructose-1,6-bisphosphatase, and sucrose synthase did not. The changes in extractable SPS activity were not related directly to photosynthetic rates or light/dark changes. Hence, it was postulated that the oscillations were under the control of an endogenous clock. During the light period, the activity of SPS was similar to the estimated rate of sucrose formation. In the dark, however, SPS activity declined sharply and then increased even though degradation of starch was linear. The activity of SPS always exceeded the estimated maximum rate of sucrose formation in the dark. Transfer of plants into light during the normal dark period (when SPS activity was low) resulted in increased partitioning of photosynthate into starch compared to partitioning observed during the normal light period. These results were consistent with the hypothesis that SPS activity in situ was a factor regulating the rate of sucrose synthesis and partitioning of fixed carbon between starch and sucrose in the light.  相似文献

以河套蜜瓜为试材,在果实迅速膨大期通过去果处理改变库源关系,研究源叶净光合速率,蔗糖、还原糖和淀粉含量及其代谢相关酶活性的昼夜变化规律。结果表明:(1)源叶的净光合速率为单峰曲线,无明显的"光合午休"现象,去果处理对其无影响。(2)源叶中蔗糖和还原糖含量的昼夜变化为单峰曲线,蔗糖磷酸合成酶和蔗糖合成酶合成方向活性的昼夜变化为双峰曲线,蔗糖合成酶分解方向、酸性转化酶和中性转化酶活性的昼夜变化无明显规律,改变库源关系对这些指标均无显著影响;蔗糖含量升高受蔗糖磷酸合成酶和蔗糖合成酶合成方向正调控,而蔗糖含量降低则受多种酶的共同调节。(3)源叶中淀粉含量和腺苷二磷酸葡萄糖焦磷酸化酶活性的昼夜变化为单峰曲线,去果处理可以显著提高淀粉含量和腺苷二磷酸葡萄糖焦磷酸化酶活性,淀粉含量升高受腺苷二磷酸葡萄糖焦磷酸化酶正调控。  相似文献

光质可影响植物光合特性、形态以及生理过程。本试验研究了不同光质(白光W、红光R、红蓝混合光RB、蓝光B)对桑树植株生长、形态和光合作用的影响。结果表明:与白光对照相比,红光、蓝光和红蓝混合光处理下植株的生长、干物质积累受到抑制;红光处理下植株的株高、叶面积显著高于白光、红蓝混合光、蓝光处理;而白光、红蓝混合光、蓝光处理下植株的LMA、叶绿素a/b比值、可溶性蛋白含量、蔗糖、淀粉含量和叶片总N含量显著高于红光处理;红蓝混合光处理下植株的Pn、Gs、ΦPSⅡ与白光处理相近,红光、蓝光处理下植株的Pn、ΦPSⅡ低于白光、红蓝混合光处理,同时红光、红蓝混合光、蓝光处理下植株的抗氧化酶活性高于白光处理,而MDA含量低于白光处理;红光处理下植株的叶片厚度、栅栏组织和海绵组织厚度显著小于白光处理。因此,一定比例的红蓝混合光可以使桑树植株的生长、光合特性、生理特征和叶片解剖结构与白光下生长植株相近,并减少单质红光、单质蓝光对植株生长发育的不利影响。  相似文献

There is evidence suggesting that in plants changes in the photosynthetic source/sink balance are an important factor that regulates leaf photosynthetic rate through affects on the leaf carbohydrate status. However, to resolve the regulatory mechanism of leaf photosynthetic rate associated with photosynthetic source/sink balance, information, particularly on mutual relationships of experimental data that are linked with a variety of photosynthetic source/sink balances, seems to be still limited. Thus, a variety of manipulations altering the plant source/sink ratio were carried out with soybean plants, and the mutual relationships of various characteristics such as leaf photosynthetic rate, carbohydrate content and the source/sink ratio were analyzed in manipulated and non-manipulated control plants. The manipulations were removal of one-half or all pods, removal of one-third or two-third leaves, and shading of one-third or one-half leaves with soybean plants grown for 8 weeks under 10 h light (24 degrees C) and 14 h darkness (17 degrees C). It was shown that there were significant negative correlations between source/sink ratio (dry weight ratio of attached leaves to other all organs) and leaf photosynthetic rate; source/sink ratio and activation ratio (percentage of initial activity to total activity) of Rubisco in leaf extract; leaf carbohydrate (sucrose or starch) content and photosynthetic rate; carbohydrate (sucrose or starch) content and activation ratio of Rubisco; amount of protein-bound ribulose-1,5-bisphosphate (RuBP) in leaf extract and leaf photosynthetic rate; and the amount of protein-bound RuBP and activation ratio of Rubisco. In addition, there were significant positive correlations between source/sink ratio and leaf carbohydrate (sucrose or starch) content; source/sink ratio and the amount of protein-bound RuBP; carbohydrate (sucrose or starch) content and amount of protein-bound RuBP and the activation ratio of Rubisco and leaf photosynthetic rate. The plant water content, leaf chlorophyll and Rubisco contents were not affected significantly by the manipulations. There is a previous report in Arabidopsis thaliana that the amount of protein-bound RuBP in leaf extract correlates negatively with the activation ratio of Rubisco in the leaf extract. Therefore, the results obtained from the manipulation experiments indicate that there is a regulatory mechanism for the leaf photosynthetic rate that correlates negatively with leaf carbohydrate (sucrose and starch) status and positively with the activation state of Rubisco under a variety of photosynthetic source/sink balances.  相似文献

The effect of iron deficiency on photosynthetic pigments, ribulose-1,5-bisphosphate carboxylase (RuBPC), and photosystem activities were investigated in field grown grapevine (Vitis vinifera L. cv. Pinot noir) leaves. The contents of chlorophyll (Chl) (a+b) and carotenoids per unit fresh mass showed a progressive decrease upon increase in iron deficiency. Similar results were also observed in content of total soluble proteins and RuBPC activity. The marked loss of large (55 kDa) and small (15 kDa) subunits of RuBPC was also observed in severely chlorotic leaves. However, when various photosynthetic electron transport activities were analysed in isolated thylakoids, a major decrease in the rate of whole chain (H2O methyl viologen) electron transport was observed in iron deficient leaves. Such reduction was mainly due to the loss of photosystem 2 (PS2) activity. The same results were obtained when Fv/Fm was evaluated by Chl fluorescence measurements in leaves. Smaller inhibition of photosystem 1 (PS1) activity was also observed in both mild and severely chlorotic leaves. The artificial electron donors, diphenyl carbazide and NH2OH, markedly restored the loss of PS2 activity in severely chlorotic leaves. The marked loss of PS2 activity was evidently due to the loss of 33, 23, 28-25, and 17 kDa polypeptides in iron deficient leaves.  相似文献

Maize(Zea mays L.) plants were grown in a greenhouse with differentlevels of nitrate-N (2 to 20 millimolar). Nitrogen nutritionhad dramatic effects on plant growth and photosynthetic characteristicsof mature leaves. Increasing nitrogen resulted in greater biomassproduction, shoot/root ratios, and rates of leaf expansion duringthe day. The elongating zone of high-N plants had higher activities(per gram fresh weight) of sucrose synthase and neutral invertasethan low-N plants, suggesting that increased leaf growth wasrelated to a greater biochemical capacity for sucrose metabolism. Mature leaves of high-N plants had higher rates of photosynthesisand assimilate export (sucrose formation), and partitioned morecarbon into sucrose relative to starch. Increased photosyntheticrates (leaf area basis) were associated with higher levels ofribulose-l,5-bisphosphate carboxylase, phosphoenolpyruvate carboxylaseand pyruvate, phosphate dikinase (determined immunochemically).In addition, N-nutrition affected the functional organizationof chlorophyll in the leaves. Large increases in the numberof PS I reaction centers were observed which fully accountedfor increases in leaf chlorophyll content with increasing nitratesupply. Collectively, the results suggest that increased growth of maizeplants at high light and optimal nitrogen nutrition is relatedto greater capacity for photosynthesis and translocation inmature leaves, and possibly increased capacity for sucrose metabolismin expanding leaves. The high sucrose phosphate synthase (SPS) capacity and the low soluble acid invertase activity of mature leaves of the first flush of leaves remained stable during second flush development. Conversely, fluctuations of sucrose synthase (SS) activity were in parallel with the sucrose requirement of the second flush. Sucrose synthase activity (synthesis direction) in first flush leaves could increase in 'response' to sink demand constituted by the second flush growth. Only the ptotosynthates provided by flush mature leaves were translocated for a current flush, while the starch content of these leaves remained stable. After their emergence, second flush leaves showed an increase in SPS and SS (Synthetic direction) activities. The high sucrose synthesis in second flush leaves was used for leaf expansion. When young leaves were 30% fully expanded (stage II20), SPS activity showed little change whereas SS activity declined rapidly toward and after full leaf expansion. The starch accumulation in the young leaves occured simultaneously with their expansion. Developing leaves showed a high level of acid invertase activity until maximum leaf expansion (stage II1). In first and second flush leaves, changes in acid invertase activity correlated positively with changes in reducing sugar concentrations. Alkaline invertase and sucrose synthase (cleavage direction) activities showed similar changes with low values when compared with those of acid invertase activity, especially in second flush leaves. The present results suggest that soluble acid invertase was the primary enzyme responsible for sucrose catabolism in the expanding common oak leaf.  相似文献

Changes in various components of photosynthetic apparatus during the 4 d dark incubation at 25°C of detached control and ultraviolet-B (UV-B) treatedVigna unguiculata L. leaves were examined. The photosynthetic apparatus was more degraded in younger control seedlings and for a longer time UV-B treated seedlings than in the older or for a shorter time UV-B treated seedlings. This was shown by determining the losses in chlorophyll (Chl) and protein contents, variable fluorescence yield, photosystem (PS) 2, PS1 and ribulose-1,5-bisphosphate carboxylase (RuBPC) activities, and photosynthetic14CO2 fixation.
