Cold tolerance and dehydration in Enchytraeidae from Svalbard   总被引:4,自引:1,他引:3  
When cooled in contact with moisture, eight species of arctic Enchytraeidae from Svalbard were killed by freezing within minutes or hours at −3 and −5 °C; an exception was Enchytraeus kincaidi which survived for up to 2 days. When the temperature approached 0 °C the enchytraeids apparently tried to escape from the moist soil. The supercooling capacity of the enchytraeids was relatively low, with mean supercooling points of −5 to −8 °C. In contrast, specimens of several species were extracted from soil cores that had been frozen in their intact state at −15 °C for up to 71 days. Compared to freezing in a moist environment, higher survival rates were obtained during cooling at freezing temperatures in dry soil. Survival was recorded in species kept at −3 °C for up to 35 days, and in some species kept at −6 °C for up to 17 days. Slow warming greatly increased survival rates at −6 °C . The results strongly suggest that arctic enchytraeids avoid freezing by dehydration at subzero temperatures. In agreement with this, weight losses of up to ca. 42% of fresh weight were recorded in Mesenchytraeus spp. and of up to 55% in Enchytraeus kincaidi at water vapour pressures above ice at −3 to −6 °C. The effect of exposure to low temperatures (5 °C) on lymphocyte proliferation, leukocyte populations, and serum complement levels was examined in the northern leopard frog, Rana pipiens. Proliferation of T lymphocytes in response to phytohemagglutinin stimulation was significantly decreased in frogs kept for 2, 3, and 5 months at 5 °C compared to that of animals kept at 22 °C. A significant increase in the average percentage of neutrophils and a decrease in the mean percentage of eosinophils was observed in the blood of frogs held for 5 months in the cold compared to animals held at 22 °C for the same length of time. Mean serum complement activity after 1 month at 5 °C was significantly reduced in comparison to animals held at 22 °C and was not detectable after 5 months in the cold. Recovery of complement levels at room temperature (22 °C) was also examined after cold exposure. Complement levels were significantly higher than controls (at 22 °C) in frogs returned to 22 °C for 7 and 14 days after 5 months in the cold. After frogs were held at 5 °C for 1 month, serum complement levels increased significantly within 2 days after returning to 22 °C and continued to rise 5 and 9 days after warming. Injections with Aeromonas hydrophila following a 5-week exposure to 5 °C failed to cause death or observable symptoms of disease in frogs that were returned to 22 °C. In the Swiss Prealps Entomobrya nivalis hibernates in an inactive state, hidden under bark flakes on spruce. For freeze avoidance it relies on thermal hysteresis proteins (THPs) and polyols (mainly ribitol, with small amounts arabitol and threitol). Polyols are present only during the inactive state, THPs additionally protect during the transition phase in spring and autumn, when animals are still active but frosts may occur. Peak values were recorded in February/March for THPs (3.5 °C hysteresis between melting and freezing point) and for polyols (26 μg mg−1 FW; hemolymph osmolality 680 mosmol l−1). E. nivalis is able to control its hemolymph osmolality independently of body water content. Mean osmolality in summer was 350– 440 mosmol l−1, in winter it was elevated to 650 mosmol l−1, due to a synthesis mainly of ribitol. Body water content varied between 1.8 and 3.3 mg H2O mg−1 DW, depending on humidity conditions. Experiments on triggering of antifreeze synthesis showed the action of temperature and photoperiod as cues, but there was also evidence for an endogenous rhythm. No clear correlation between antifreeze concentration and supercooling ability could be established, suggesting that gut content or other parameters also play an inportant role. This investigation examined the influence of soil moisture and associated parameters on the cold hardiness of the Colorado potato beetle (Leptinotarsa decemlineata Say), a temperate-zone species that overwinters in terrestrial burrows. The body mass and water content of adult beetles kept in sand at 4 °C varied over a 16-week period of diapause according to the substratum's moisture content. Changes in body water content, in turn, influenced the crystallization temperature (range −3.3 to −18.4 °C; n = 417), indicating that environmental moisture indirectly determined supercooling capacity, a measure of physiological cold hardiness. Beetles held in dry sand readily tolerated a 24-h exposure to temperatures ranging from 0° to −5 °C, but those chilled in sand containing as little as 1.7% water (dry mass) had elevated mortality. Thus, burrowing in dry soils not only promotes supercooling via its effect on water balance, but may also inhibit inoculative freezing. Mortality of beetles exposed to −5 °C for 24 h was lower in substrates composed of sand, clay and/or peat (36–52%) than in pure silica sand (78%) having an identical water content (17.0% dry mass). In addition to moisture, the texture, structure, water potential, and other physico-chemical attributes of soil may strongly influence the cold hardiness and overwintering survival of burrowing insects. We examined the effects of climate and allocation patterns on stem respiration in ponderosa pine (Pinus ponderosa) growing on identical substrate in the cool, moist Sierra Nevada mountains and the warm, dry, Great Basin Desert. These environments are representative of current climatic conditions and those predicted to accompany a doubling of atmospheric CO2, respectively, throughout the range of many western north American conifers. A previous study found that trees growing in the desert allocate proportionally more biomass to sapwood and less to leaf area than montane trees. We tested the hypothesis that respiration rates of sapwood are lower in desert trees than in montane trees due to reduced stem maintenance respiration (physiological acclimation) or reduced construction cost of stem tissue (structural acclimation). Maintenance respiration per unit sapwood volume at 15°C did not differ between populations (desert: 6.39 ± 1.14 SE μmol m−3 s−1, montane: 6.54 ± 1.13 SE μmol m−3 s−1, P = 0.71) and declined with increasing stem diameter (P = 0.001). The temperature coefficient of respiration (Q 10) varied seasonally within both environments (P = 0.05). Construction cost of stem sapwood was the same in both environments (desert: 1.46 ± 0.009 SE g glucose g−1 sapwood, montane: 1.48 ± 0.009 SE glucose g−1 sapwood, P = 0.14). Annual construction respiration calculated from construction cost, percent carbon and relative growth rate was greater in montane populations due to higher growth rates. These data provide no evidence of respiratory acclimation by desert trees. Estimated yearly stem maintenance respiration was greater in large desert trees than in large montane trees because of higher temperatures in the desert and because of increased allocation of biomass to sapwood. By analogy, these data suggest that under predicted increases in temperature and aridity, potential increases in aboveground carbon gain due to enhanced photosynthetic rates may be partially offset by increases in maintenance respiration in large trees growing in CO2-enriched atmospheres. The thermal and metabolic physiology of Chalinolobus gouldii, an Australian vespertilionid bat, was studied in the laboratory using flow-through respirometry. Chalinolobus gouldii exhibits a clear pattern of euthermic thermoregulation, typical of endotherms with respect to body temperature and rate of oxygen consumption. The basal metabolic rate of euthermic Chalinolobus gouldii is approximately 86% of that predicted for a 17.5-g mammal and falls into the range of mass-specific basal metabolic rates ascribed to vespertilionid bats. However, like most vespertilionid bats, Chalinolobus gouldii displays extreme thermolability. It is able to enter into torpor and spontaneously arouse at ambient temperatures as low as 5 °C. Torpid bats thermoconform at moderate ambient temperature, with body temperature ≈ ambient temperature, and have a low rate of oxygen consumption determined primarily by Q 10 effects. At low ambient temperature (< 10 °C), torpid C. gouldii begin to regulate their body temperature by increased metabolic heat production; they tend to maintain a higher body temperature at low ambient temperature than do many northern hemisphere hibernating bats. Use of torpor leads to significant energy savings. The evaporative water loss of euthermic bats is relatively high, which seems unusual for a bat whose range includes extremely arid areas of Australia, and is reduced during torpor. The thermal conductance of euthermic C. gouldii is less than that predicted for a mammal of its size. The thermal conductance is considerably lower for torpid bats at intermediate body temperature and ambient temperature, but increases to euthermic values for torpid bats when thermoregulating at low ambient temperature. Freeze-avoiding fire-colored beetle larvae, Dendroides canadensis, were monitored seasonally to explore the role of endogenous hemolymph ice nucleators and antifreeze proteins on the maintenance of supercooling. In preparation for overwintering, D. canadensis depressed hemolymph ice nucleator activity and increased thermal hysteresis activity [mean value circa 0. 5 °C (summer) versus circa 5 °C (midwinter)] resulting in decreased larval and hemolymph supercooling points [−7 °C (summer) versus −20 °C (midwinter)]. Results of gel filtration chromatography, flotation ultracentifugation and quantitative investigation of ice nucleator activity using hemolymph from summer and winter collected larvae strongly suggest that highly active protein and lipoprotein ice nucleators are removed in preparation for overwintering. Additions of either purified antifreeze proteins or midwinter hemolymph with high antifreeze protein activity to a mixture of protein or lipoprotein ice nucleators isolated from D. canadensis hemolymph inhibited the activity of these nucleators. This suggests that in addition to seasonal removal, inhibition of hemolymph ice nucleators by antifreeze proteins contributes to seasonal increases in hemolymph supercooling capacity. To study the physiological responses induced by immersing in cold water various areas of the upper limb, 20 subjects immersed either the index finger (T1), hand (T2) or forearm and hand (T3) for 30 min in 5°C water followed by a 15-min recovery period. Skin temperature of the index finger, skin blood flow (Qsk) measured by laser Doppler flowmetry, as well as heart rate (HR) and mean arterial blood pressure (ˉBPa) were all monitored during the test. Cutaneous vascular conductance (CVC) was calculated as Qsk / ˉBPa. Cold induced vasodilatation (CIVD) indices were calculated from index finger skin temperature and CVC time courses. The results showed that no differences in temperature, CVC or cardiovascular changes were observed between T2 and T3. During T1, CIVD appeared earlier compared to T2 and T3 [5.90 (SEM 0.32) min in T1 vs 7.95 (SEM 0.86) min in T2 and 9.26 (SEM 0.78) min in T3, P < 0.01]. The HR was unchanged in T1 whereas it increased significantly at the beginning of T2 and T3 [+13 (SEM 2) beats · min−1 in T2 and +15 (SEM 3) beats · min−1 in T3, P < 0.01] and then decreased at the end of the immersion [−12 (SEM 3) beats · min−1 in T2, and −15 (SEM 3) beats · min−1 in T3, P < 0.01]. Moreover, ˉBPaincreased at the beginning of T1 but was lower than in T2 and T3 [+9.3 (SEM 2.5) mmHg in T1, P < 0.05;  +20.6 (SEM 2.6) mmHg and 26.5 (SEM 2.8) mmHg in T2 and T3, respectively, P < 0.01]. The rewarming during recovery was faster and higher in T1 compared to T2 and T3. These results showed that general and local physiological responses observed during an upper limb cold water test differed according to the area immersed. Index finger cooling led to earlier and faster CIVD without significant cardiovascular changes, whereas hand or forearm immersion led to a delayed and slower CIVD with a bradycardia at the end of the test. This study investigated the effects on running economy (RE) of ingesting either no fluid or an electrolyte solution with or without 6% carbohydrate (counterbalanced design) during 60-min running bouts at 80% maximal oxygen consumption (O2max). Tests were undertaken in either a thermoneutral (22–23°C; 56–62% relative humidity, RH) or a hot and humid natural environment (Singapore: 25–35°C; 66–77% RH). The subjects were 15 young adult male Singaporeans [O2max = 55.5 (4.4 SD) ml kg−1 min−1]. The RE was measured at 3 m s−1 [65 (6)% O2max] before (RE1) and after each prolonged run (RE2). Fluids were administered every 2 min, at an individual rate determined from prior tests, to maintain body mass (group mean = 17.4 ml min−1). The O2 during RE2 was higher (P < 0.05) than that during the RE1 test for all treatments, with no differences between treatments (ANOVA). The mean increase in O2 from RE1 to RE2 ranged from 3.4 to 4.7 ml kg−1 min−1 across treatments. In conclusion, the deterioration in RE at 3 m s−1 (65% O2max) after 60 min of running at 80% O2max appears to occur independently of whether fluid is ingested and regardless of whether the fluid contains carbohydrates or electrolytes, in both a thermoneutral and in a hot, humid environment. Phosphorus magnetic resonance spectroscopy (31P-MRS) was used to investigate the influence of maximal aerobic power (˙VO 2max) on the recovery of human calf muscle from high-intensity exercise. The (˙VOO2max) of 21 males was measured during treadmill exercise and subjects were assigned to either a low-aerobic-power (LAP) group (n = 10) or a high-aerobic-power (HAP) group (n = 11). Mean (SE) ˙VO 2max of the groups were 46.6 (1.1) and 64.4 (1.4) ml · kg−1 · min−1, respectively. A calf ergometry work capacity test was used to assign the same relative exercise intensity to each subject for the MRS protocol. At least 48 h later, subjects performed the rest (4 min), exercise (2 min) and recovery (10 min) protocol in a 1.5 T MRS scanner. The relative concentration of phosphocreatine (PCr) was measured throughout the protocol and intracellular pH (pHi) was determined from the chemical shift between inorganic phospate (Pi) and PCr. End-exercise PCr levels were 27 (3.4) and 25 (3.5)% of resting levels for LAP and HAP respectively. Mean resting pHi was 7.07 for both groups, and following exercise it fell to 6.45 (0.04) for HAP and 6.38 (0.04) for LAP. Analysis of data using non-linear regression models showed no differences in the rate of either PCr or pHi recovery. The results suggest that ˙VO2max is a poor predictor of metabolic recovery rate from high-intensity exercise. Differences in recovery rate observed between individuals with similar ˙VO2max imply that other factors influence recovery. Accepted: 17 December 1996  相似文献

Unlike northern hemisphere conifer families, the southern family, Podocarpaceae, produces a great variety of foliage forms ranging from functionally broad-, to needle-leaved. The production of broad photosynthetic surfaces in podocarps has been linked qualitatively to low-light-environments, and we undertook to assess the validity of this assumption by measuring the light response of a morphologically diverse group of podocarps. The light response, as apparent photochemical electron transport rate (ETR), was measured by modulated fluorescence in ten species of this family and six associated species (including five Cupressaceae and one functionally needle-leaved angiosperm) all grown under identical glasshouse conditions. In all species, ETR was found to increase as light intensity increased, reaching a peak value (ETRmax) at saturating quantum flux (PPFDsat), and decreasing thereafter. ETRmax ranged from 217 μmol electrons · m−2 · s−1 at a PPFDsat of 1725 μmol photons · m−2 · s−1 in Actinostrobus acuminatus to an ETR of 60 μmol electrons · m−2 · s−1 at a PPFDsat of 745 μmol electrons · m−2 · s−1 in Podocarpus dispermis. Good correlations were observed between ETRmax and both PPFDsat and maximum assimilation rate measured by gas-exchange analysis. The effective quantum yield at light saturation remained constant in all species with an average value of 0.278 ± 0.0035 determined for all 16 species. Differences in the shapes of light response curves were related to differences in the response of non-photochemical quenching (q n), with q n saturating faster in species with low PPFDsat. Amongst the species of Podocarpaceae, the log of average shoot width was well correlated with PPFDsat, wider leaves saturating at lower light intensities. Fluxes of CO2 and N2O were measured from both natural and experimentally augmented snowpacks during the winters of 1993 and 1994 on Niwot Ridge in the Colorado Front Range. Consistent snow cover insulated the soil surface from extreme air temperatures and allowed heterotrophic activity to continue through much of the winter. In contrast, soil remained frozen at sites with inconsistent snow cover and production did not begin until snowmelt. Fluxes were measured when soil temperatures under the snow ranged from –5°C to 0°C, but there was no significant relationship between flux for either gas and temperature within this range. While early developing snowpacks resulted in warmer minimum soil temperatures allowing production to continue for most of the winter, the highest CO2 fluxes were recorded at sites which experienced a hard freeze before a consistent snowpack developed. Consequently, the seasonal flux of CO2 C from snow covered soils was related both to the severity of freeze and the duration of snow cover. Over-winter CO2 C loss ranged from 0.3 g C m−2 season−1 at sites characterized by inconsistent snow cover to 25.7 g C m−2 season−1 at sites that experienced a hard freeze followed by an extended period of snow cover. In contrast to the pattern observed with C loss, a hard freeze early in the winter did not result in greater N2ON loss. Both mean daily N2O fluxes and the total over-winter N2ON loss were related to the length of time soils were covered by a consistent snowpack. Over-winter N2ON loss ranged from less 0.23 mg N m−2 from the latest developing, short duration snowpacks to 16.90 mg N m−2 from sites with early snow cover. These data suggest that over-winter heterotrophic activity in snow-covered soil has the potential to mineralize from less than 1% to greater than 25% of the carbon fixed in ANPP, while over-winter N2O fluxes range from less than half to an order of magnitude higher than growing season fluxes. The variability in these fluxes suggests that small changes in climate which affect the timing of seasonal snow cover may have a large effect on C and N cycling in these environments. In this study we measured growth and milk intake and calculated energy intake and its allocation into metabolism and stored tissue for hooded seal (Cystophora cristata) pups. In addition, we measured mass loss, change in body composition and metabolic rate during the first days of the postweaning fast. The mean body mass of the hooded seal pups (n = 5) at the start of the experiments, when they were new-born, was 24.3 ± 1.3 kg (SD). They gained an average of 5.9 ± 1.1. kg · day−1 of which 19% was water, 76% fat and 5% protein. This corresponds to an average daily energy deposition of 179.8 ± 16.0 MJ. The pups were weaned at an average body mass of 42.5 ± 1.0 kg 3.1 days after the experiment was initiated. During the first days of the postweaning fast the pups lost an average of 1.3 ± 0.5␣kg of body mass daily, of which 56% was water, 16% fat and 28% protein. During the nursing period the average daily water influx for the pups was 124.6 ± 25.8 ml · kg−1. The average CO2 production during this period was 1.10 ± 0.20 ml · g−1 · h−1, which corresponds to a field metabolic rate of 714 ± 130 kJ ·  kg−1 · day−1, or 5.8 ± 1.1 times the predicted basal metabolic rate according to Kleiber (1975). During the postweaning fast the average daily water influx was reduced to 16.1 ± 6.6 ml · kg−1. The average CO2 production in␣this period was 0.58 ± 0.17 ml · g−1 · h−1 which corresponds to a field metabolic rate of 375 ± 108 kJ · kg−1 · day−1 or 3.2 ± 0.9 times the predicted basal metabolic rate. Average values for milk composition were 33.5% water, 58.6% fat and 6.2% protein. The pups drank an average of 10.4 ± 1.8␣kg of milk daily, which represents an energy intake of 248.9 ± 39.1 MJ · day−1. The pups were able to store 73.2 ± 7.7% of this energy as body tissue. This study compared the effects of supplementing the normal diets of six trained cyclists [maximal oxygen uptake O2max) 4.5 (0.36)l · min−1; values are mean (SD)] with additional carbohydrate (CHO) on muscle glycogen utilisation during a 1-h cycle time-trial (TT). Using a randomised crossover design, subjects consumed either their normal diet (NORM) for 3 days, which consisted of 426 (137) g · day−1 CHO [5.9 (1.4) g · kg−1 body mass (BM)], or additional CHO (SUPP) to increase their intake to 661 (76) g · day−1 [9.3 (0.7) g · kg−1 BM]. The SUPP diet elevated muscle glycogen content from 459 (83) to 565 (62) mmol · kg−1 dry weight (d.w.) (P < 0.05). However, despite the increased pre-exercise muscle glycogen stores, there was no difference in the distance cycled during the TT [40.41 (1.44) vs 40.18 (1.76) km for NORM and SUPP, respectively]. With NORM, muscle glycogen declined from 459 (83) to 175 (64) mmol · kg−1 d.w., whereas with SUPP the corresponding values were 565 (62) and 292 (113) mmol · kg−1 d.w. Accordingly, both muscle glycogen utilisation [277 (64) vs 273 (114) mmol · kg−1 d.w.] and total CHO oxidation [169 (20) vs 165 (30) g · h−1 for NORM and SUPP, respectively] were similar. Neither were there any differences in plasma glucose or lactate concentrations during the two experimental trials. Plasma glucose concentration averaged 5.5 (0.5) and 5.6 (0.6) mmol · l−1, while plasma lactate concentration averaged 4.4 (1.9) and 4.4 (2.3) mmol · l−1 for NORM and SUPP, respectively. The results of this study show that when well-trained subjects increase the CHO content of their diet for 3 days from 6 to 9 g · kg−1 BM there is only a modest increase in muscle glycogen content. Since supplementary CHO did not improve TT performance, we conclude that additional CHO provides no benefit to performance for athletes who compete in intense, continuous events lasting 1 h. Furthermore, the substantial muscle CHO reserves observed at the termination of exercise indicate that whole-muscle glycogen depletion does not determine fatigue at this exercise intensity and duration. Photoperiodic time measurement regulating larval diapause in the pitcher-plant mosquito, Wyeomyia smithii, varies in a close relationship with latitude. The critical photoperiod mediating the maintenance and termination of diapause is positively correlated with latitude (r 2 = 0.977) among six populations from southern (30–31° N), intermediate (40° N), and northern (46–49° N) latitudes in North America. The developmental response to unnaturally short and to unnaturally long photoperiods declines with increasing latitude, so that longer critical photoperiods are associated with a downward rather than a lateral shift in the photoperiodic response curve. Exotic light and dark cycles of varying period (T) with a short (10 h) photophase and a scotophase ranging from 14 (T = 24) to 62 (T = 72) h, reveal two geographic patterns: a decline in perturbability of the photoperiodic clock with increasing latitude, and no change with latitude in the 21-h period of rising and falling development with increasing T. These results show (1) that there is a rhythmic component to photoperiodic time measurement in W. smithii, (2) that the period of this rhythm is about 21 h in all populations, and (3) that more northern populations show decreasing responsiveness to photoperiod and increasing stability against perturbation by exotic period lengths (T > 24). Previous studies on W.␣smithii indicate that this single temperate species of a tropical and subtropical genus has evolved from south to north. We therefore conclude that the evolution of increasing critical photoperiod in W. smithii during its adaptive radiation into North America has more likely involved the amplitude and not the period of the underlying circadian pacemaker. Water relations dynamics during simulated sunflecks at high (36°C) and medium (27°C) temperatures and high and low vapour pressure deficits beween leaf and air (VPD) were studied on shade-grown Piper auritum H.B. & K. plants, a pioneer tree, common in gaps and clearings of tropical rain forests. The leaves of P. auritum wilt rapidly when exposed to high light. Exposure to high VPD and high light caused substantial and rapid dehydration of leaves. Dehydration could be prevented under high humidity irrespective of temperature. Water stored in leaf cells served as initial source for transpiration upon high light exposure. This effect increased with increasing VPD and temperature. The pronounced decrease in leaf water content over time in high light caused a rapid decrease in leaf water potential (Ψl) and a concomitant increase in water potential gradient (ΔΨ/Δx) between trunk and leaf, yet the high leaf elasticity (small bulk elastic modulus, ε) allowed turgor maintenance under most conditions. Under high VPD and high temperature, stomata remained open and ΔΨ/Δx frequently exceeded 0.95 MPa · m−1, the cavitation-inducing threshold (ΔΨ/Δx cav) causing high rates of acoustic emissions from stems and leaf petioles and leading to concomitant losses in hydraulic conductance per leaf area (k l). At medium temperature (high VPD), stomatal closure contained xylem embolism by keeping ΔΨ/Δx at or below this threshold. We argue that wilting substantially contributes to creating a sufficient driving force for water uptake from the soil, and reducing the VPD (through a decrease in radiation load and thus leaf temperature) to avoid excessive dehydration. Short-latency vestibular-evoked potentials to pulsed linear acceleration were characterized in the quail. Responses occurred within 8 ms following the onset of stimuli and were composed of a series of positive and negative peaks. The latencies and amplitudes of the first four peaks were quantitatively characterized. Mean latencies at 1.0 g ms−1 ranged from 1265 ± 208 μs (P1, N = 18) to 4802 ± 441 μs (N4, N = 13). Amplitudes ranged from 3.72 ± 1.51 μV (P1/N1, N = 18) to 1.49 ± 0.77 μV (P3/N3, N = 16). Latency-intensity (LI) slopes ranged from −38.7 ± 7.3 μs dB−1 (P1, N = 18) to −71.6 ± 21.9 μs dB−1 (N3, N = 15) and amplitude-intensity (AI) slopes ranged from 0.20 ± 0.08 μV dB−1 (P1/N1, N = 18) to 0.07 ± 0.04 μV dB−1 (P3/N3, N = 11). The mean response threshold across all animals was −21.83 ± 3.34 dB re: 1.0 g ms−1 (N = 18). Responses remained after cochlear extirpation showing that they could not depend critically on cochlear activity. Responses were eliminated by destruction of the vestibular end organs, thus showing that responses depended critically and specifically on the vestibular system. The results demonstrate that the responses are vestibular and the findings provide a scientific basis for using vestibular responses to evaluate vestibular function through ontogeny and senescence in the quail. The purpose of this study was to investigate the effect of a thiamin derivative, thiamin tetrahydrofurfuryl disulfide (TTFD), on oxygen uptake (˙VO2), lactate accumulation and cycling performance during exercise to exhaustion. Using a randomized, double-blind, cross-over design with a 10-day washout between trials, 14 subjects ingested either 1 g · day−1 of TTFD or a placebo (PL) for 4 days. On day 3, subjects performed a progressive exercise test to exhaustion on a cycle ergometer for the determination of ˙VO2submax, ˙VO2peak, lactate concentration ([La ]), lactate threshold (ThLa) and heart rate ( f c). On day 4, subjects performed a maximal 2000-m time trial on a cycle ergometer. A one-way analysis of variance (ANOVA) with repeated measures was used to determine significant differences between trials. There were no significant differences detected between trials for serial measures of ˙VO2submax, [La] or f c. Likewise, ˙VO2peak [PL 4.06 (0.19) TTFD 4.12 (0.19) l · min−1, P = 0.83], ThLa [PL 2.47 (0.17), TTFD 2.43 (0.16) l · min−1, P = 0.86] and 2000-m performance time [PL 204.5 (5.5), TTFD 200.9 (4.3) s, P = 0.61] were not significantly different between trials. Unidirectional flux rates of Ca2+ across gastrointestinal tissues from sheep and goats were measured in vitro by applying the Ussing-chamber technique. Except for the sheep duodenum, mucosal to serosal Ca2+ flux rates (J ms) exceeded respective flux rates in the opposite direction (J sm) in both species and in all segments of the intestinal tract. This resulted in net Ca2+ flux rates␣(J net = J ms − J sm) ranging between −2 and 9 nmol · cm−2 · h−1 in sheep and between 10 and 15 nmol cm−2 · h−1 in goats. In sheep, only J net in jejunum, and in goats, J netin duodenum and jejunum were significantly different from zero. Using sheep rumen wall epithelia, significant J net of Ca2+ of around 5 nmol · cm−2 · h−1 could be detected. Since the experiments were carried out in the absence of an electrochemical gradient, significant net Ca2+ absorption clearly indicates the presence of active mechanisms for Ca2+ transport. Dietary Ca depletion caused increased calcitriol plasma concentrations and induced significant stimulations of net Ca2+ absorption in goat rumen. J net of Ca2+ across goat rumen epithelia was significantly reduced by 1 mmol · l −1 verapamil in the mucosal buffer solution. In conclusion, there is clear evidence for the rumen as a main site for active Ca2+ absorption in small ruminants. Stimulation of active Ca2+ absorption by increased plasma calcitriol levels and inhibition by mucosal verapamil suggest mechanistic and regulatory similarities to active Ca2+ transport as described for the upper small intestines of monogastric species. The ontogeny of deep-body cold sensitivity was studied in 1 to 12 days old Pekin ducklings Anas platyrhynchos. Deep-body cold sensitivity was determined by means of thermodes implanted in the abdominal cavity. The thermodes were perfused with cold water for 15-min periods to lower the core temperature. Cooling of the body core elicited increases in metabolic rate and vasoconstrictions in the legs of all the ducklings. From the changes induced in metabolic rate and core temperature, deep-body cold sensitivity values of between −5.17 and −6.36 W · kg−1 · °C−1, were estimated. These values, which are in the range of those reported previously for adult Pekin ducks, did not change with age, and it is concluded that deep-body cold sensitivity is fully developed at hatching. Our next aim was to investigate whether the autonomic responses elicited by exposure of ducklings to cold ambient conditions could be explained by temperature changes within the body core. During cold exposure, the increase in metabolic rate was not accompanied by a concomitant decrease in core temperature. On the contrary, deep-body temperature increased slightly during the initial phase of cold exposure. The ducklings attained a metabolic rate amounting to 85–90% of their peak metabolic rate before the core temperature fell below the regulated level measured at thermoneutrality. Thus, despite the findings that Pekin ducklings have a highly-developed deep-body cold sensitivity, their metabolic cold defence under natural conditions seems to be mediated primarily by peripheral thermoreceptors.
