The common and routine procedure for the quantification of chlorophyll a (chl a) in aquatic studies has a series of steps. Here, we sought to find optimal conditions for phytoplankton cell harvesting, chl a extraction, and chl a measurement and calculation, to find an effective, cost-saving, safe, and environment-friendly procedure for determining phytoplankton chl a concentration. We replaced the traditional GF/C filters with inorganic polymer flocculants (IPFs) and clay for phytoplankton harvesting, and then various solvents (acetone, ethanol, DMF, and DMSO), IPFs (PAC, PFS, and PAFS) and clay were tested for their suitability for chl a extraction, with or without homogenization at different temperatures for different extraction durations. About 0.3–1.0 g l?1 of PAC or PFSA combined with 1.0–2.5 g l?1 clay were found to provide optimal conditions in terms of yield and cost for phytoplankton cell harvesting from water samples. Based on our results, we recommend flocculation and centrifugation instead of glass-fiber membrane filtration for harvesting phytoplankton cells from environmental water samples, 95% ethanol for chl a extraction without homogenization and heating, and spectrophotometry to determine chl a concentration.  相似文献

The phytoplankton community structure of a hypertrophic lake was quantitatively determined with the aid of flow cytometry. The flow cytometry signals were calibrated to obtain cell‐specific information, such as the chl a content and the biovolume per cell. The reliability of this method was tested with laboratory cultures. The results of the phytoplankton structure in a hypertrophic lake with respect to chl distribution in the different algal groups obtained by flow cytometry were compared with the results from HPLC pigment fingerprinting. Both methods yield the percentage contribution of the different algal groups to total chl a. The chl a specific absorption coefficient of the phytoplankton (a*Phy) was determined via visible (VIS) spectroscopy of samples taken from a hypertrophic lake (Auensee) in 2003. The results indicated that a*Phy of the total cell suspension is dependent on the phytoplankton structure as well as on environmental factors. The linear relationship between a*Phy at 675 nm and the product of the chl a content per cell and the biovolume offered the possibility to normalize phytoplankton absorption spectra to acquire the taxon‐specific a*Phy. The estimated a*Phy (675 nm) values were used to normalize single cell absorption spectra at this wavelength to obtain the a*Phy between 400 and 750 nm for representatives of the major algal groups. Our measurements show that the absorption coefficient for the whole phytoplankton community varies within the season. Finally, we used the a*Phy and the chl a distribution to calculate the light absorption of each algal group in the hypertrophic lake.  相似文献

We studied chlorophyll a (chl. a), biovolume and species composition of benthic algae and phytoplankton in the eutrophic lower River Spree in 1996. The chl. a concentration was estimated as 3.5 (2.7–4.5) µg/cm2 for epipsammon, 9.4 (7.4–11.9) µg/cm2 for epipelon and 6.7 (5.7–7.8) µg/cm2 for the epilithon (median and 95% C. L.). The mean total biomass of benthic algae was significantly higher (6.0 µg chl. a/cm2) than the areal chl. a content of the pelagic zone (1.6 µg chl. a/cm2). Although certain phytoplankton taxa were abundant in the periphyton, benthic taxa generally dominated the assemblages. Seasonal dynamics of benthic algae were probably controlled by light and nitrate supply (sand), discharge fluctuations (sand, mud) and invertebrate grazing (stones). The quantitative composition of the chloroplast pigments of phytoplankton sampled weekly at one station in the Trondheimsfjord was studied by circular paper chromatography throughout 18 months. The concentrations of total chlorophyll a (T-chl a obtained by the trichromatic method) as well as of chromatographically purified chlorophyll a (chl a) followed the variations in phytoplankton concentration. Two spring blooms and a weak autumn flowering of phytoplankton were clearly reflected in the pigment contents found, namely 14–16 mg T-chl a/m3 for the spring maxima, corresponding to nearly 300 mg T-chl a/m2 for the euphotic zone; and 3–4 mg/m3 or 32 mg/m2 for the autumn peak. The concentrations between blooms amounted to ≈ 1 mg T-chl a/m3, while concentrations down to 0.03 mg/m3 were found for winter samples.The content of T-chl a was high in diatom cells prior to a bloom (20–40 × 10?9 mg/cell). During rapid growth (a more or less exponential phase) the cell content of chloroplast pigments decreased (to 5–10 × 10?9 mg). No degradation product of chlorophylls could be detected during this phase and the percentage of chl a (of T-chl a) was high (70–80 %). At the peak of the bloom, and especially when the nitrate content in the surrounding water had been exhausted, low values for T-chl a were found (0.3–0.5 × 109?mg/cell). As soon as the cell counts started to fall, or even before the decline could be clearly detected, the percentage of chl a dropped (to 40-20 %) and derived chlorophylls (not phaeophytin a) were present in the samples. Model studies with cultured algae showed a similar behaviour.It is concluded that the proportion of chl a to T-chl a and the occurrence of chlorophyll derivatives in phytoplankton samples can give valuable information on the stage of development of the algal populations involved.  相似文献

In the last 40 years, the shallow steppe lake, Neusiedler See, was ice covered between 0 and 97 days. The North Atlantic Oscillation (NAO) as well as the Mediterranean Oscillation affected the lake and its conditions during winter. Both climate indices correlated negatively with the duration of ice cover and the timing of ice-out. Average winter phytoplankton biomass increased from less than 0.2 (0.05–0.84) mg FM l?1 in the late 1960s/beginning of 1970s to 3.1 (1.72–5.61) mg FM l?1 in the years 2001–2004. The increase in annual winter biomass of phytoplankton was associated with a significant shift in the composition of the algal assemblage. In the winter 1997/1998, diatoms contributed between 40 and 80% to the phytoplankton biomass while in 2006/2007 cyanoprokaryotes contributed 46%. Mean chlorophyll-a concentrations during winter were significantly correlated with those of total phosphorus (Ptot). Together with cold-water species (rotifer Rhinoglena fertöensis), perennial, eurythermal ones (copepod Arctodiaptomus spinosus) contributed to the zooplankton community. High zooplankton numbers were encountered when rotifers, particularly when densities of Rhinoglena fertöensis were high (r 2 = 0.928). Zooplankton abundance and biomass varied from year to year but correlated positively with Chl-a (biomass ? r 2 = 0.69; numbers ? r 2 = 0.536). Winter zooplankton populations were primarily influenced by winter conditions, but in early winter also by survival of autumn populations, i.e., the more adults of Arctodiaptomus spinosus survived into winter, the higher was the zooplankton biomass in early winter. Phyto- and zooplankton dynamics in shallow lakes of the temperate region seem to critically depend on the biomass in autumn and on winter conditions, specifically on ice conditions and thus are related to climate signals such as the NAO.  相似文献

In spring 2005, monthly sampling was carried out at a sublittoral site near Tautra Island. Microphytobenthic identification, abundance (ABU), and biomass (BIOM), were performed by microscopic analyses. Bacillariophyceae accounted for 67% of the total ABU, and phytoflagellates constituted 30%. The diatom floristic list consisted of 38 genera and 94 species. Intact light‐harvesting pigments chl a, chl c, and fucoxanthin and their derivatives were identified and quantified by HPLC. Photoprotective carotenoids were also observed (only as diadinoxanthin; no diatoxanthin was detected). Average fucoxanthin content was 4.57 ± 0.45 μg fucoxanthin · g sediment dry mass?1, while the mean chl a concentration was 2.48 ± 0.15 μg · g?1 dry mass. Both the high fucoxanthin:chl a ratio (considering nondegraded forms) and low amounts of photoprotective carotenoids indicated that the benthic microalgal community was adapted to low light. Microphytobenthic primary production was estimated in situ (MPPs, from 0.15 to 1.28 mg C · m?2 · h?1) and in the laboratory (MPPp, from 6.79 to 34.70 mg C · m?2 · h?1 under light saturation) as 14C assimilation; in April it was additionally estimated from O2‐microelectrode studies (MPPO2) along with the community respiration. MPPO2 and the community respiration equaled 22.9 ± 7.0 and 7.4 ± 1.8 mg C · m?2 · h?1, respectively. A doubling of BIOM from April to June in parallel with a decreasing photosynthetic activity per unit chl a led us to suggest that the microphytobenthic community was sustained by heterotrophic metabolism during this period.  相似文献

This study investigates the dynamics of phytoplankton communities and nitrogen uptake in the Indian sector of the Southern Ocean during spring and summer. The study area is oligotrophic (Chl a stocks <50 mg m−2); nevertheless, a large spatial variation of phytoplankton biomass and community structure was observed. During both seasons the phytoplankton community in the seasonal ice zone showed higher biomasses and was mainly composed of large diatom cells. However, in the permanently open ocean zone the community had low biomass and was chiefly composed of nano- and picoflagellates. In the polar front zone, although biomass was higher, the community structure was similar to the open ocean zone. The results suggest that the variation in phytoplankton community structure on a larger scale resonates with gradients in water column stability and nutrient distribution. However, significant changes in biomass and nutrient stocks but little change in community structure were observed. Absolute nitrogen uptake rates were generally low, but their seasonal variations were highly significant. During spring the communities displayed high specific nitrate uptake (mean rate = 0.0048 h−1), and diatoms (in the seasonal ice zone) as well as nano- and picoflagellates (in the permanently open ocean zone and polar front zone) were mainly based on new production (mean ƒ-ratio = 0.69). The transition to summer was accompanied by a significant reduction in nitrate uptake rate (0.0048 h−1 → 0.0011 h−1) and a shift from predominantly new to regenerated production (ƒ-ratio 0.69 → 0.39). Ammonium played a major role in the seasonal dynamics of phytoplankton nutrition. The results emphasize that, despite a large contrast in community structure, the seasonal dynamics of the nitrogen uptake regime and phytoplankton community structure in all three subsystems were similar. Additionally, this study supports our previous conclusion that the seasonal shift in nitrogen uptake regime can occur with, as well as without, marked changes in community structure. Ecosystem functioning is simultaneously affected by changes in community composition and environmental change such as increasing atmospheric carbon dioxide (CO2) and subsequent ocean acidification. However, it largely remains uncertain how the effects of these factors compare to each other. Addressing this question, we experimentally tested the hypothesis that initial community composition and elevated CO2 are equally important to the regulation of phytoplankton biomass. We full‐factorially exposed three compositionally different marine phytoplankton communities to two different CO2 levels and examined the effects and relative importance (ω2) of the two factors and their interaction on phytoplankton biomass at bloom peak. The results showed that initial community composition had a significantly greater impact than elevated CO2 on phytoplankton biomass, which varied largely among communities. We suggest that the different initial ratios between cyanobacteria, diatoms, and dinoflagellates might be the key for the varying competitive and thus functional outcome among communities. Furthermore, the results showed that depending on initial community composition elevated CO2 selected for larger sized diatoms, which led to increased total phytoplankton biomass. This study highlights the relevance of initial community composition, which strongly drives the functional outcome, when assessing impacts of climate change on ecosystem functioning. In particular, the increase in phytoplankton biomass driven by the gain of larger sized diatoms in response to elevated CO2 potentially has strong implications for nutrient cycling and carbon export in future oceans.  相似文献

The potential interactions between the bloom-forming dinoflagellates and other phytoplankton during the algal bloom cycle are interesting, while the causes for the phytoplankton community changes were not fully understood. We hypothesized that phytoplankton community structure and photosynthetic activities of total phytoplankton have their special characteristics in different phases of the algal blooms. To test this hypothesis, a survey covering the process of a Prorocentrum donghaiense bloom in coastal waters between Dongtou and Nanji Islands was carried out from 9 to 20 May 2016, and the changes in the phytoplankton community and photosynthetic activities of total phytoplankton were determined. Surface seawater was sampled for microscopic analysis of phytoplankton composition and pulse amplitude modulated (PAM) chlorophyll fluorescence analysis of photosynthetic activities of the total phytoplankton species. A total of 25, 31, and 19 phytoplankton species were identified in its growth (9–12 May), maintenance (13–18 May) and dissipation phases (19–20 May), respectively. Diatoms were dominant in terms of species number while dinoflagellates were predominant at cell abundance. Dinoflagellates were the major dominant species during three phases of the bloom based on the dominance (Y) value, whereas the dominant species extended to dinoflagellates and diatoms including P. donghaiense, Coscinodiscus argus, Gonyaulax spinifera, Cyclotella sp. and Scrippsiella trochoidea in the dissipation phase. In the maintenance phase, the average cell abundances of the total phytoplankton and P. donghaiense were consistent with the chlorophyll a (Chla) concentration in the seawater; for the diversity indices of total phytoplankton species, Simpson index (C) was the highest while Shannon index (H′) and Pielou evenness index (J′) were the lowest. Furthermore, photosynthetic activities of the total phytoplankton species represented by the effective quantum yield (Fq'/Fm') and the maximum relative electron transport rate (rETRmax) in the maintenance phase were higher than those in the growth and dissipation phases. The results indicated that the characteristics of phytoplankton community structure and photosynthetic activities could be regarded as criteria in predicting the phases of algal blooms.  相似文献

A study was done of the relationship between hydrographic variables and the composition, abundance, community structure and biomass spectrums of coastal phytoplankton at scales greater than 100 km on the Yucatan Peninsula (SE Gulf of Mexico). This was done during the season of greatest environmental instability in the region, the northwind season (late fall to winter). Samples were collected at stations in the west (Campeche), north (Yucatan), and east (Quintana Roo) zones of the Peninsula. Measurements were taken of temperature, salinity, dissolved oxygen, dissolved inorganic nutrients (ammonia, nitrite, nitrate and phosphate) and chlorophyll a, and samples were taken for phytoplankton analysis. The hydrographic results showed the Campeche zone as having the lowest salinity (<35 psu) values, as well as the highest inorganic nutrient and chlorophyll a values, all of which are related to continental water contributions. The Yucatan zone had the lowest temperatures and the lowest inorganic nutrient values, indicating influence from the Yucatan Current and the Gulf of Mexico. A total of 159 phytoplankton species were identified, dominated by diatoms (>80%) and dinoflagellates. Phytoplankton exhibited greater concentration, richness, equitability and diversity in Campeche, while the lowest community structure values were had in the Quintana Roo zone. The ordination analysis demonstrated that the dominant genera were the diatoms Chaetoceros, Pseudonitzschia and Thalassionema. The biomass spectrums exhibited the lowest slope in environments of higher heterogeneity, with Campeche being the most disturbed and heterogeneous zone and Quintana Roo that with the least heterogeneity.  相似文献

A legacy of pre-regulation coal mining in many areas has been acid mine drainage (metal-rich, low pH water). Today, numerous remediation strategies may be implemented, but there is little data on efficacy in restoring biological condition. Two alkaline doser projects in Ohio were assessed using diatom assemblages, and biofilm extracellular enzyme activities (EEA). In one stream, water quality steadily increased downstream of the doser; pH increased from 3.8 to 7.2 and Fe decreased from 107 to 0.42 mg l?1. Likewise, the periphyton biomass (chl a 7.15–12.77 mg m?2) increased and periphyton index scores (4–27) improved. As well, EEA data showed greater activity for phosphorus, nitrogen, and one carbon acquisition enzyme. For the other stream, the conductivity remained high (>720 μS cm?1) and pH and alkalinity decreased downstream. Biological data, including EEA, varied along the stream length with higher numbers in the middle reach, such as chlorophyll a (0.56 to 87.75 to 2.77 mg m?2), and index scores (7 to 29 to 11). The first remediation site showed positive results in chemistry, biological community and measures of ecosystem function. The second stream was highly variable in these parameters suggesting further AMD inputs are hampering recovery.  相似文献

A high resolution study of chlorophyll a and primary production distribution was carried out in the Atlantic sector of the Southern Ocean during the austral summer of 1990–91. Primary production (14C assimilation) and photosynthetic capacity levels at frontal systems were among the highest recorded during the cruise (2.8–6.3 mgC·m–3·h–1, and 1.3–4.7mgC·mgChl a –1·h–1, respectively). Blooms at ocean fronts were strongly dominated by specific size classes and species. This suggests that the increase in biomass was probably the result of an enhancement of in situ production by selected components of the phytoplankton assemblage, rather than accumulation of cells through hydrographic forces. This hypothesis is supported by the high variability of photosynthetic capacities at adjacent stations along the transects. Blooms (ca 2.7–3.5 mg Chl a·m–3) were found at three oceanic fronts (the Subtropical, Subantarctic and Antarctic Polar Fronts) during the early summer. These were equivalent to, or denser than, blooms in the Marginal Ice Zone and at the Continental Water Boundary. Seasonal effects on phytoplankton community structure were very marked. In early summer (December), netphyto-plankton (>20 m) was consistently the major component of the frontal blooms, with the chain-forming diatoms Chaetoceros spp. and Nitzschia spp. dominating at the Subantarctic and Antarctic Polar Fronts, respectively. During late summer (February), nanophytoplankton (1–20 m) usually dominated algal communities at the main frontal areas. Only at the Antarctic Polar Front did netphytoplankton dominate, with the diatom component consisting almost exclusively of Corethron criophilum. An early to late summer shift of maximum phytoplankton biomass from north to south of the Antarctic Polar Front was observed. Spatial covariance between silicate levels and water-column stability appeared to be the main factor controlling phytoplankton production at the Antarctic Polar Front. Low silicate concentrations may have limited diatom growth at the northern edge of the front, while a deep mixed layer depth reduced production at the southern edge of the front.  相似文献

The biomass and primary production of phytoplankton in Lake Awasa, Ethiopia was measured over a 14 month period, November 1983 to March 1985. The lake had a mean phytoplankton biomass of 34 mg chl a m–3 (n = 14). The seasonal variation in phytoplankton biomass of the euphotic zone (mg chl a m–2 h–1) was muted with a CV (standard deviation/mean) of 31%. The vertical distribution of photosynthetic activity was of a typical pattern for phytoplankton with light inhibition on all but overcast days. The maximum specific rates of photosynthesis or photosynthetic capacity (Ømax) for the lake approached 19 mg O2 (mg chl a)–1 h–1, with high values during periods of low phytoplankton biomass. Areal rates of photosynthesis ranged between 0.30 to 0.73 g O2 m–2 h–1 and 3.3 to 7.8 g O2 m–2 d–1. The efficiency of utilisation of PhAR incident on the lake surface varied from 2.4 to 4.1 mmol E–1 with the highest efficiency observed corresponding to the lowest surface radiation. Calculated on a caloric basis, the efficiency ranged between 1.7 and 2.9%. The temporal pattern of primary production by phytoplankton showed limited variability (CV = 21 %).  相似文献

Weekly sampling was carried out in Saanich Inlet, British Columbia throughout the winter of 1975–1976. The surface water column was characterized by exposure to low solar radiation energy (<150 g cal·cm?2 · day?1), slight stratification with occasional vertical mixing, and abundant algal nutrients. Phytoplankton were mostly distributed above 5 m in the water column, with a fairly low biomass averaging <1 μgchla·1?1. Dominant phytoplankton organisms were nanoflagellates occasionally accompanied by dinoflagellates as the second dominant. Centric diatoms, which were dominant in the blooms, were always present but less than a few percentage of the total phytoplankton biomass. Daily photosynthetic productivity was exclusively limited by available radiant energy. Low solar radiation and occasional mixing of the surface zone prohibited the centric diatoms from becoming dominant.  相似文献

Water extracts of a no. 2 fuel oil, a Nigerian crude oil, and used crankcase oil were examined for their effects on algal communities in experiments lasting several weeks conducted under near-natural conditions. No. 2 fuel oil extracts depressed algal biomass (chlorophyll a) and resulted in blue-green algal (cyanobacterial) dominance and decreased diatom occurrence. Changes in concentrations of chlorophyll c, which was specific for diatoms in this work, and phycocyanin, which was specific for blue-green algae, confirmed the observations. Used crankcase oil extracts also depressed biomass, but Nigerian crude extracts did not, and both these extracts had less effect on community composition than did no. 2 fuel oil extracts. Photosynthetic 14C incorporation was both stimulated and depressed by exposure to extracts with hydrocarbon concentrations 0.038 to 0.124 mg/liter. Short-term exposure to higher concentrations (1.17 to 15.30 mg of hydrocarbons per liter) of no. 2 fuel oil extracts depressed photosynthetic 14C incorporation by Vaucheria-dominated communities in all tests but one. Toxicity was greater from extracts prepared in the light than from extracts prepared in the dark.  相似文献

It is widely recognized that the mesoscale eddies play an important part in the biogeochemical cycle in ocean ecosystem, especially in the oligotrophic tropical zones. So here a heterogeneous cyclonic eddy in its flourishing stage was detected using remote sensing and in situ biogeochemical observation in the western South China Sea (SCS) in early September, 2007. The high-performance liquid chromatography method was used to identify the photosynthetic pigments. And the CHEMical TAXonomy (CHEMTAX) was applied to calculate the contribution of nine phytoplankton groups to the total chlorophyll a (TChl a) biomass. The deep chlorophyll a maximum layer (DCML) was raised to form a dome structure in the eddy center while there was no distinct enhancement for TChl a biomass. The integrated TChl a concentration in the upper 100 m water column was also constant from the eddy center to the surrounding water outside the eddy. However the TChl a biomass in the surface layer (at 5 m) in the eddy center was promoted 2.6-fold compared to the biomass outside the eddy (p < 0.001). Thus, the slight enhancement of TChl a biomass of euphotic zone integration within the eddy was mainly from the phytoplankton in the upper mixed zone rather than the DCML. The phytoplankton community was primarily contributed by diatoms, prasinophytes, and Synechococcus at the DCML within the eddy, while less was contributed by haptophytes_8 and Prochlorococcus. The TChl a biomass for most of the phytoplankton groups increased at the surface layer in the eddy center under the effect of nutrient pumping. The doming isopycnal within the eddy supplied nutrients gently into the upper mixing layer, and there was remarkable enhancement in phytoplankton biomass at the surface layer with 10.5% TChl a biomass of water column in eddy center and 3.7% at reference stations. So the slight increasing in the water column integrated phytoplankton biomass might be attributed to the stimulated phytoplankton biomass at the surface layer.  相似文献

We compared nitrate concentrations, phytoplankton biomass, and phytoplankton community structure in lakes fed by glacier melt and snowmelt (GSF lakes) and by snowmelt only (SF lakes) within North Cascades National Park (NOCA) in Washington State, USA. In the U.S. Rocky Mountains, glacier melting has greatly increased nitrate concentrations in GSF lakes (52–236 µg NO3–N L?1) relative to SF lakes (1–14 µg NO3–N L?1) and thereby stimulated phytoplankton changes in GSF lakes. Considering NOCA contains approximately one-third of the glaciers in the continental U.S., and many mountain lakes that receive glacier meltwater inputs, we hypothesized that NOCA GSF lakes would have greater nitrate concentrations, greater phytoplankton biomass, and greater abundance of nitrogen-sensitive diatom species than NOCA SF lakes. However, at NOCA nitrate concentrations were much lower and differences between lake types were small compared to the Rockies. At NOCA, nitrate concentrations averaged 13 and 5 µg NO3–N L?1 in GSF and SF lakes, respectively, and a nitrate difference was not detectable in several individual years. There also was no difference in phytoplankton biomass or abundance of nitrogen-sensitive diatoms between lake types at NOCA. In contrast to the Rockies, there also was not a significant positive relationship between watershed percent glacier area and lake nitrate at NOCA. Results demonstrate that biogeochemical responses to global change in Western U.S. mountain lake watersheds may vary regionally. Regional differences may be affected by differing nitrogen deposition, climate, geology, or microbial processes within glacier environments, and merit further investigation.  相似文献

The interplay among microorganisms profoundly impacts biogeochemical cycles in the ocean. Culture-based work has illustrated the diversity of diatom–prokaryote interactions, but the question of whether these associations can affect the spatial distribution of microbial communities is open. Here, we investigated the relationship between assemblages of diatoms and of heterotrophic prokaryotes in surface waters of the Indian sector of the Southern Ocean in early spring. The community composition of diatoms and that of total and active prokaryotes were different among the major ocean zones investigated. We found significant relationships between compositional changes of diatoms and of prokaryotes. In contrast, spatial changes in the prokaryotic community composition were not related to geographic distance and to environmental parameters when the effect of diatoms was accounted for. Diatoms explained 30% of the variance in both the total and the active prokaryotic community composition in early spring in the Southern Ocean. Using co-occurrence analyses, we identified a large number of highly significant correlations between abundant diatom species and prokaryotic taxa. Our results show that key diatom species of the Southern Ocean are each associated with a distinct prokaryotic community, suggesting that diatom assemblages contribute to shaping the habitat type for heterotrophic prokaryotes.  相似文献

Diel variations of cellular optical properties were examined for cultures of the haptophyte Imantonia rotunda N. Reynolds and the diatom Thalassiosira pseudonana (Hust.) Hasle et Heimdal grown under a 14:10 light:dark (L:D) cycle and transferred from 100 μmol photons · m?2 · s?1 to higher irradiances of 250 and 500 μmol photons · m?2 · s?1. Cell volume and abundance, phytoplankton absorption coefficients, flow‐cytometric light scattering and chl fluorescence, and pigment composition were measured every 2 h over a 24 h period. Results showed that cell division was more synchronous for I. rotunda than for T. pseudonana. Several variables exhibited diel variability with an amplitude >100%, notably mean cell volume for the haptophyte and photoprotective carotenoids for both species, while optical properties such as flow‐cytometric scattering and chl a–specific phytoplankton absorption generally showed <50% diel variability. Increased irradiance induced changes in pigments (both species) and mean cell volume (for the diatom) and amplified diel variability for most variables. This increase in amplitude is larger for pigments (factor of 2 or more, notably for cellular photoprotective carotenoid content in I. rotunda and for photosynthetic pigments in T. pseudonana) than for optical properties (a factor of 1.5 for chl a–specific absorption, at 440 nm, in I. rotunda and a factor of 2 for the absorption cross‐section and the chl a–specific scattering in T. pseudonana). Consequently, diel changes in optical properties and pigmentation associated with the L:D cycle and amplified by concurrent changes in irradiance likely contribute significantly to the variability in optical properties observed in biooptical field studies.  相似文献
