Background and Aims

To date, current research involving pollen viability has been evaluated in a relatively low number of orchid species. In the present study, we focused on five related Mediterranean orchid genera (Anacamptis, Orchis, Dactylorhiza, Ophrys and Serapias) that are characterized by different types of deceptive pollination.

Methods

The in vitro germination ability of increasingly aged pollinaria of eight food-, seven sexually and two shelter-deceptive species was evaluated. Pollination experiments on two food-, one sexually and one shelter-deceptive species were also performed and the percentage of embryonate seeds derived from the increasingly aged pollinaria was checked.

Key Results

All of the examined species showed long-term viabilities (=50 % pollen tube growth) that ranged from 8 to 35 d. Species with the same deceptive pollination strategies exhibited the same pollen viability trends. Interestingly, pollen viabilities of species groups with different deception types have shown significant differences, with sexually and shelter- deceptive species exhibiting a shorter life span than food-deceptive species.

Conclusions

This study confirms the prolonged germination and fertilization capacities of orchid pollinaria, and to our knowledge is the first report demonstrating a clear relationship between pollen viability and pollination system. It is proposed that this relationship is attributed to the different types of reproductive barriers, pre- or post-zygotic, that characterixe Ophrys and Serapias and the food-deceptive species, respectively.  相似文献

Paspalum L. is a large and complex genus, enclosing more than 300 species, whose boundaries and infrageneric classification are still being studied. Recent phylogenetic analyses suggest that Paspalum inaequivalve Raddi and Paspalum microstachyum J. Presl, from the Inaequivalvia informal group, should be excluded from Paspalum. Focused on the unclear taxonomic position of P. inaequivalve and P. microstachyum, their leaf anatomy was studied, and some atypical features related to C4 photosynthesis were found. This atypical Kranz syndrome is the aim of this research. Transverse leaf blade sections from fresh and herbarium material of P. inaequivalve were studied by light, fluorescence, and transmission microscopy. Additionally, sheaths and culms of P. inaequivalve and leaf blades of P. microstachyum were observed by light microscopy. δ13C isotope discrimination was determinated for P. inaequivalve. We compared our results with available anatomical data from related taxa. As well as typical mesophyll cells (PCA) and mestome sheath cells (PCR), a third type of cells, here called ‘globose parenchymatous cells’, was observed in leaf blades of P. inaequivalve and P. microstachyum. These cells are placed externally to the mestome sheaths of the first and second vascular bundles, they have thin walls, with no developed suberine lamella, few chloroplasts with 1–2 starch grains, thylakoids not organized in grana, and a large central vacuole. The globose parenchymatous cells represent a novel trait in P. inaequivalve and P. microstachyum, further supporting the close relation between both species and their exclusion from the genus Paspalum. This atypical Kranz syndrome has not been described in Paspalum before, but the globose parenchymatous cells here described resemble the distinct cells considered as remnants of the outer parenchymatous sheath described for Anthaenantiopsis, some sections of Panicum L., and Chaetium Nees, providing possible taxonomic significance.  相似文献

The metabolites produced by the secretory canals of the root cortex from four Smallanthus species belonging to the yacon group were identified as ent-kaurane-type diterpenes. The dichloromethane root cortex extracts of the four species were treated with diazomethane and analyzed comparatively by GC–MS using a simple and rapid procedure which is very sensitive and reproducible permitting detection of minor components. In all cases, ent-16-kauren-19-oic acid (kaurenoic acid) methyl ester was the main component, differences being observed only in the minor components. The minor components identified were grandiflorenic acid methyl ester, ent-16-kauren-19-al, 16α,17-epoxy-15α-angeloyloxy-kauran-19-oic acid methyl ester and several O-acyl derivatives at C-15 or C-18 of kaurenoic acid. One of the minor components, 18-isobutyroyloxy-ent-kaur-16-en-19-oic acid is a new kaurenoic acid derivative. Grandiflorenic acid and 15-α-angeloyloxy-16,17-α-epoxy-ent-16-kauren-19-oic acid were present only in Smallanthus sonchifolius and Smallanthus siegesbeckius which showed very similar GC traces. The different GC profile of RC diterpenes from Smallanthus connatus and Smallanthus macroscyphus supports the view that they are different taxa. Some chemotaxonomic aspects of the genus Smallanthus and the subtribe Milleriinae are briefly discussed.  相似文献

The morphological and anatomical nature of perennating storage organs of the predominantly Australasian orchid tribe Diurideae (Orchidoideae: Orchidaceae) as well as anatomical concepts in tribes Orchideae and Diseae of Orchidoideae have been problematic for 150 yr, reflected in conflicting or vague terminology and questions about polystely and even monophyly of Orchidoideae. From a representative survey of underground organs of 145 species in 37 ingroup genera (Diurideae) and two outgroup genera (Spiranthes and Disa), the so-called “root-stem tuberoids” are here interpreted as root tubers (except for the stem tubers of Rhizanthella) borne on either droppers or stolonoid roots. All root tubers examined are bounded by a 1–4 layered velamen and exodermis, whereas droppers and stolonoid roots may have velamen-exodermis or a simple epidermis depending on the taxon and often bear multiseriate or uniseriate trichomes associated with mycorrhiza (as do roots of some taxa). The “polystely” reported in tubers of many Orchideae also occurs in tubers of many Diurideae but represents only dissection of the siphonostele into 2–13 traces. Cladistic analyses of data show extraordinarily high levels of homoplasy in characters related to root, dropper/stolonoid root, and tuber, so that these characters alone are of limited usefulness.  相似文献

The Heliohebe group of Veronica (sect. Hebe) consists of five species occurring in the South Island of New Zealand. These species and a hybrid were analysed for their flavonoids. Five flavone glycosides were isolated and identified by NMR spectroscopy and three additional glycosides were detected by LC–UV–MS. Luteolin 7-O-, 3′-O- and 4′-O-glucosides and apigenin 7-O-glucoside were present in all six taxa investigated, 6-hydroxyluteolin glycosides were found in five and a luteolin caffeoylglycoside in four taxa, while a hypolaetin 7-O-glycoside was detected only in Veronica pentasepala. The 3′-O- and 4′-O-glucosides of luteolin are also common in other species of Veronica sect. Hebe (restricted to the Southern Hemisphere), but are rare in Northern Hemisphere species of Veronica and thus act as good chemotaxonomic markers for the section. The relatively simple flavonoid profiles found in the Heliohebe group are plesiomorphic and consistent with the group's status as sister to the Hebe clade. Based on the detected flavonoids, two groups could be distinguished within the Heliohebe clade: (1) Veronica hulkeana, Veronica lavaudiana and Veronica raoulii, characterised by luteolin caffeoylglycoside, and (2) V. pentasepala and Veronica scrupea, where this compound is replaced by a 6-hydroxyluteolin dihexoside.  相似文献

Thielaviopsis basicola is a hemibiotrophic root pathogen causing black root rot in a wide range of economically important crops. Our initial attempts to transform T. basicola using standard Agrobacterium tumefaciens–mediated transformation (ATMT) protocols were unsuccessful. Successful transformation required the addition of V8 juice (to induce germination of T. basicola chlamydospores) and higher concentrations of acetosyringone in the co-cultivation medium, and of chlamydospores/endoconidia, A. tumefaciens cells during co-cultivation. With these modifications, two T. basicola strains were successfully transformed with the green (egfp) or red (AsRed) fluorescent protein genes. Chlamydospores/endoconidia transformed with the egfp gene exhibited strong green fluorescence, but their fluorescence became weaker as the germ tubes emerged. Transformants harbouring the AsRed gene displayed strong red fluorescence in both chlamydospores/endoconidia and germ tubes. Fluorescent microscopic observations of an AsRed-labelled strain colonizing roots of transgenic Nicotiana benthamiana plants, which express the actin filaments labelled with EGFP, at 24 hours post inoculation showed varying levels of fungal germination and penetration. At this stage, the infection appeared to be biotrophic with the EGFP-labelled host actin filaments not being visibly degraded, even in host root cells in close contact with the hyphae. This is the first report of ATMT of T. basicola, and the use of an AsRed-labelled strain to directly observe the root infection process.  相似文献

The geographical distribution and analysis of the essential oils of species from three sections of Hypericum L. (Guttiferae/Clusiaceae/Hypericaceae) from Portugal are presented. Hypericum perfoliatum (section Drosocarpium) grows wild in the centre and south of Portugal; Hypericum humifusum and Hypericum linarifolium are both from section Oligostema, the former occurring throughout the country, while the second is distributed mainly in the north and centre; Hypericum pulchrum (section Taeniocarpium) is confined to the littoral north of Portugal. The essential oils were obtained by distillation–extraction, hydrodistillation and distillation in a modified Marcusson apparatus from the dried aerial parts of the different populations and were analysed by GC and GC–MS. Monoterpene hydrocarbons constituted the main fraction in all oils (43–69%, 53–85%, 28–45% and 48–65% for H. perfoliatum, H. humifusum, H. linarifolium and H. pulchrum, respectively). Sesquiterpene hydrocarbons (2–13%, 6–18%, 21–27% and 16–18%, respectively) and a third fraction of non-terpenic compounds (20–29%, 3–16%, 2–14% and 5–11%, respectively) from the four species attained relatively high amounts in all oils. Within each species, no major differences were detected in the essential oil composition, despite the fact that different locations, phenological phases and extraction methodologies were used. Notwithstanding the dominance of α-pinene in all four species' oils, cluster and principal components analysis on the identified components showed that the range of α-pinene, β-pinene and n-nonane supported a separation of the four species. The essential oil composition of the four species showed some qualitative resemblances, which correlate well with the taxonomical classification based on morphological characters.  相似文献

Thymus is taxonomically a very complex genus with a high frequency of hybridisation and introgression among sympatric species. The variation in accumulation of leaf-surface flavonoids was investigated in 71 wild populations of Thymus from different putative hybrid swarm areas in Andalucia, Spain. Twenty-two flavones, five flavanones, two dihydroflavonols, a flavonol and two unknowns were detected by HPLC–DAD combined with LC–APCI-MS analysis. The majority of compounds were flavones with a luteolin-type substitution of the B-ring, in contrast to previous reports on Macedonian taxa, which predominantly accumulate flavones with apigenin-type substitution of the B-ring. Anatomical and morphometric studies, supported by cluster analysis, identified pure Thymus hyemalis and Thymus baeticus populations, and a large number of putative hybrids. Flavonoid variation was closely related to morphological variation in all populations and is suspected to be a result of genetic polymorphism. Principal component analysis identified the presence of species-specific and geographically linked chemotypes and putative hybrids with mixed morphological and chemical characteristics. Qualitative and quantitative flavonoid accumulation appears to be genetically regulated, while external factors play a secondary role. Flavonoid profiles can thus provide diagnostic markers for the taxonomy of Thymus and are also useful in detecting hybridising taxa.  相似文献

Starch grains are a ubiquitous component of plants that have been used in tandem with phytoliths, pollen, and macrofossils to reconstruct past floral diversity. This tool has yet to be fully explored for aquatic plants, specifically seagrasses, which lack phytoliths and are rarely preserved as macrofossils or pollen. If starch grains in seagrasses are morphologically distinct, this method has the potential to improve seagrass identification in the fossil record in such cases where its starch is preserved (e.g. scratches and occlusal surfaces of tooth enamel from seagrass consumers). The goals of this study were twofold: (1) to determine if starch is present in seagrass material and (2) to assess how starch grain morphology differs between different seagrasses.This study focused on four abundant and ecologically distinct seagrasses from the Caribbean: Halodule wrightii, Ruppia maritima, Syringodium filiforme, and Thalassia testudinum. Starch grains were observed in all species except S. filiforme. Grains from H. wrightii are typically observed in side-on orientation, are sub-round to angular, and are fairly small (3-19 μm, end-on). Grains of R. maritima are small spherical grains (4-8 μm) that have a centric hilum and a straight extinction cross with a median angle between the arms of 90°. Grains from T. testudinum are large (9-31 μm, end-on), conical in side-on and round/sub-round in end-on orientation, have a slightly eccentric hilum with an obvious particle, and prominent lamellae.Visual assessment and comparative statistics demonstrate that the morphology of starch grains from T. testudinum, R. maritima, and H. wrightii are significantly different. With more extensive research, there is potential for the positive identification of starch grains from an unknown seagrass. The ability to identify seagrass from starch grains could facilitate the identification of seagrasses in the fossil record and supply information on seagrass evolution and distribution, climate effects on seagrass distribution, and the diets of seagrass consumers.  相似文献

The potential allelopathic effects of 14 stilbenoids and five flavonoids, isolated from leaves of Carex distachya Desf., were evaluated on the seed germination and seedling growth of three coexisting Mediterranean species (Dactylis hispanica, Petrorhagia velutina, and Phleum subulatum). The structures of the metabolites have been elucidated on the basis of their spectroscopic features (1D and 2D NMR experiments and EI–MS and ESI–MS data). The bioassays showed species-specific effects of the metabolites from C. distachya, specially on the plant growth (root and shoot elongation) which resulted significantly stimulated or inhibited at 10−4 M concentration. The effects on root elongation is generally greater than the shoot growth at all the tested concentrations (10−4–10−8 M). Cluster of biological data showed interesting relationships between the chemical structures of the compounds and their biological effects.  相似文献

Two fossil taxa Tubulifloridites antipodica and T. viteauensis recovered from the Eocene Knysna Lignite of South Africa were examined with scanning (SEM) and transmission electron microscopy (TEM). The details of their sculpturing and wall structure are similar to the same species of fossil dispersed pollen taxa recovered from southwestern Africa and South America. Fifteen species of the woody South African taxa, Brachylaena (9 species) and Tarchonanthus (6 species) were investigated with SEM and TEM. All of the taxa are tricolporate, spherical to slightly prolate, microechinate to echinate and have a bilayered columellate infrastructure, except B.ilicifolia, which has a single columellate infrastructural level with the “granularization” of the outer portion of the infrastructural layer or the inner layer of the tectum. There is a similar distribution of plesiomorphic and derived pollen characters in a number of aster subfamilies and tribes suggesting a similar evolutionary progression of pollen, and pollen wall character evolution was occurring synchronously in a variety of aster subfamilies during the middle Tertiary and that these unique pollen features may be important to the evolution and diversification of the Asteraceae.  相似文献

Chemical signals can be the basis of interspecific recognition and speciation in many animals. The Columbretes Islands wall lizard, Podarcis atrata is very close genetically to the mainland Iberian wall lizard Podarcis hispanica. However, a previous study suggested that chemosensory interspecific recognition would avoid reproductive interactions and hybridization between these two species. These results suggested that chemicals used in intraspecific communication might differ in composition and/or proportions between these two species. In this paper, we used gas chromatography–mass spectrometry (GC–MS) to characterize the chemical composition of the lipophilic fraction from femoral gland secretions of male P. atrata and P. hispanica. The analysis showed that chemicals found in femoral secretions varied in composition and proportions between species and between populations. Seven steroids and two unidentified waxy esters, were exclusive of P. atrata lizards from the islands. In contrast, nine steroids and other six compounds were only found in mainland P. hispanica. There were also differences in proportions of shared compounds between species. Moreover, all these differences were higher between P. atrata and P. hispanica than between any population of P. hispanica. Chemical differences might be consequence of genetic differences, but they could also be explained by adaptation to different habitats with different climatic conditions or diet resources. Compounds that are specific of each species, or differences in the pattern of compounds, could explain species recognition. Therefore, these results of chemical composition and previous studies of chemosensory recognition reinforce the fact that the genetic differences between P. hispanica and P. atrata may result in an effective reproductive isolation between these two taxa.  相似文献

Seventeen fructose-6-phosphate phosphoketolase-positive bacterial strains were isolated from the digestive tract of wild pigs (Sus scrofa). Most of them were identified as Bifidobacterium boum according to sequences of 16S rRNA gene. Two strains isolated from the small intestine content had unusual morphology of cells in comparison with bifidobacteria. Cells growing in liquid anaerobic media were regular shaped rods arranged mostly in pairs. These isolates showed relatively low 16S rRNA gene sequence similarities (maximum identity of 94%) to members of the family Bifidobacteriaceae. Nevertheless, phylogenetic analyses of 16S rRNA, hsp60 and xfp gene sequences revealed that these strains are more related to recently described Neoscardovia, Aeriscardovia and other scardovial genera, than to Bifidobacterium species. Partial gene sequences of other phylogenetic markers showed low (65.8–89.5%) similarities to genome sequences of bifidobacteria and Gardnerella vaginalis. The major fatty acids detected in cells of the representative strain DPTE4T were C16:0, C18:1, C14:0. The peptidoglycan type of the DPTE4T strain was A3β l-Orn(l-Lys)-l-Ser(l-Ala)-l-Ala2. Polar lipid analysis revealed two phosphoglycolipids and phospholipids, a glycolipid and diphosphatidylglycerol. The results of phylogenetic, genotypic and phenotypic analyses support the proposal of a novel taxa, Pseudoscardovia suis gen. nov., sp. nov. (type strain = DPTE4T = DSM 24744T = CCM 7942T).  相似文献

Root-to-shoot cadmium (Cd) translocation in Solanum torvum is lower than that of the eggplant Solanum melongena; therefore, grafting S. melongena onto S. torvum rootstock can effectively reduce the Cd concentration in eggplant fruits. We hypothesized that Cd transport in S. torvum roots is restricted in the path between the epidermis and xylem vessel; hence, we investigated the Cd distribution in the roots at the micron-scale. Elemental maps of Cd, Zn and Fe accumulation in S. melongena and S. torvum root sections were obtained by synchrotron micro X-ray fluorescence spectrometry. The Cd was localized in both the stele and the epidermis of the S. melongena root cross sections regardless of the distance from the root apex. In S. torvum root sections taken at 30 and 40 mm above the root apex, a higher abundance of Cd was found within the cells of the endodermis and pericycle. The results suggested that the symplastic uptake and xylem loading of Cd in S. torvum roots were restricted, and thereby, the Cd that was unable to be loaded into the xylem accumulated in the endodermis and in the pericycle. Because symplastic uptake differs only slightly between the two species, the difference in xylem loading would explain the comparatively lower Cd concentration in S. torvum shoots.  相似文献

The fine structure of the green stem of Dianthus caryophyllus, the leaf petiole and the flower pedicel of Zantedeschia aethiopica were studied using light and scanning electron microscopy. It was revealed that these non-foliar plant parts of both species possess epidermis with numerous stomata. Stomatal density of D. caryophyllus stem was found to be relatively high (79 vs 100 per mm2 found on leaf surface). Z. aethiopica petioles and pedicels also possess numerous stomata (17 per mm2), yet stomatal density was found to be about half of that of leaves. Anatomical differences observed between petioles and pedicels were only minor. Stems of D. caryophyllus as well as petioles and pedicels of Z. aethiopica have a chlorenchyma-type tissue whose fine structure is quite similar to the leaf palisade chlorenchyma. Yet, the palisade of Z. aethiopica petioles and pedicels shows a peculiar arrangement: palisade cells are arranged with their long axis parallel to the longitudinal organ axis. Palisade tissue found in the aforementioned non-foliar plant organs in both species shows strong red chlorophyll auto-fluorescence under epi-fluorescence optics, consists of cells with abundant chloroplasts, possesses high percentage of intercellular spaces (13 and 20%, respectively) and its cells expose considerable part of their surface to the intercellular air. The fine structure of this stem palisade tissue along with the abundance of functional stomata found on the epidermis may support efficient photosynthesis.  相似文献

In this paper we analyze through a polyphasic approach several Bradyrhizobium strains isolated in Spain and Morocco from root nodules of Retama sphaerocarpa and Retama monosperma. All the strains have identical 16S rRNA genes and their closest relative species is Bradyrhizobium lablabi CCBAU 23086T, with 99.41% identity with respect to the strain Ro19T. Despite the closeness of the 16S rRNA genes, the housekeeping genes recA, atpD and glnII were divergent in Ro19T and B. lablabi CCBAU 23086T, with identity values of 95.71%, 93.75% and 93.11%, respectively. These differences were congruent with DNA–DNA hybridization analysis that revealed an average of 35% relatedness between the novel species and B. lablabi CCBAU 23086T. Also, differential phenotypic characteristics of the new species were found with respect to the already described species of Bradyrhizobium. Based on the genotypic and phenotypic data obtained in this study, we propose to classify the group of strains isolated from R. sphaerocarpa and R. monosperma as a novel species named Bradyrhizobium retamae sp. nov. (type strain Ro19T = LMG 27393T = CECT 8261T). The analysis of symbiotic genes revealed that some of these strains constitute a new symbiovar within genus Bradyrhizobium for which we propose the name “retamae”, that mainly contains nodulating strains isolated from Retama species in different continents.  相似文献
