1. 3,3'-Diaminobenzidine was shown to serve as an electron donor to photosystem 1 in the presence of 3-(3,4-dichlorophenyl)-1,1-dimethylurea. In Tris-treated chloroplasts diaminobenzidine serves as an electron donor to photosystem 1 and to photosystem 2; the latter is sensitive to 3-(3,4-dichlorophenyl)-1,1-dimethylurea. 2. Addition of diaminobenzidine to Tris-treated chloroplasts causes an increase in fluorescence yield. 3. Diaminobenzidine-dependent electron transport mediated by photosystem 2 is coupled to synthesis of ATP even in the absence of an electron acceptor. This phosphorylation which is presumably supported by cyclic electron flow, is sensitive to 3-(3,4-dichlorophenyl)-1,1-dimethylurea. 4. Diaminobenzidine-dependent ATP formation, in Tris-treated chloroplasts exhibits the red-drop phenomenon. 5. The diaminobenzidine-induced cyclic photophosphorylation (mediated by photosystem 2) is resistant to a large extent to KCN-treatment which is known to inhibit reactions catalyzed by photosystem 1. On the other hand ATP formation supported by electron transport from diaminobenzidine to methyl viologen [in the presence of 3-(3,4-dichlorophenyl)-1,1-dimethylurea] is largely inhibited by KCN-treatment. This observation suggests that there are two coupling sites of ATP formation, one catalyzed by diaminobenzidine as a donor to photosystem 1 (in the presence of 3-(3,4-dichlorophenyl)-1,1-dimethylurea), and the other supported by diaminobenzidine which acts both as a donor to photosystem 2 (in Tris-treated chloroplasts) and as an acceptor (in its oxidized form) from a carrier located between the two photosystems.  相似文献

The mutant strain U of Euglena gracilis, different from the wild type strain Z, has lost the ability to form chloroplasts during growth in the dark.Chloroplastic DNA could not be detected by CsCl density analysis in the dark-grown strain U. Chloroplast nucleoids fluoro-stained by DAPI were found in the light-grown cells, but not in the dark-grown U. Target number analysed by UV irradiation on the chloroplast formation ability decreased rapidly during cell-growth in darkness. Using a novel, pulsed micro-second time-resolved photoacoustic (PA) instrument, we measured thermal dissipation and energy storage (ES) in the intact cells of wild type (WT) Chlamydomonas reinhardtii, and mutants lacking either PSI or PSII reaction centers (RCs). On this time scale, the kinetic contributions of the thermal expansion component due to heat dissipation of absorbed energy and the negative volume change due to electrostriction induced by charge separation in each of the photosystems could be readily distinguished. Kinetic analysis revealed that PSI and PSII RCs exhibit strikingly different PA signals where PSI is characterized by a strong electrostriction signal and a weak thermal expansion component while PSII has a small electrostriction component and large thermal expansion. The calculated ES efficiencies at ~10?μs were estimated to be 80?±?5 and 50?±?13% for PSII-deficient mutants and PSI-deficient mutants, respectively, and 67?±?2% for WT. The overall ES efficiency was positively correlated with the ratio of PSI to PSI?+?PSII. Our results suggest that the shallow excitonic trap in PSII limits the efficiency of ES as a result of an evolutionary frozen metabolic framework of two photosystems in all oxygenic photoautotrophs.  相似文献

The photosynthetic energy storage yield of uncoupled thylakoid membranes was monitored by photoacoustic spectroscopy at various measuring beam intensities. The energy storage rate as evaluated by the half-saturation measuring beam intensity (i50) was inhibited by 3-(3,4-dichlorophenyl)-1,1 dimethylurea, by heat inactivation or by artificial electron acceptors specific for photosystem I or photosystem II; and was activated by electron donors to photosystem I. The reactions involving both photosystems were all characterized by a similar maximal energy storage yield of 16±2 percent. The photochemical apparatus organization in Synechococcus 6301 (Cyanophyceae) was investigated under various experimental conditions. Wild type (WT) Synechococcus produced phycobilisomes (PBSs) containing normal levels of phycocyanin (Phc) and allophycocyanin (Aphc). The ratio of reaction centers(RC) RCII/RCI of 0.4 was the same in WT and the mutant strain AN112, whereas RCH/PBS was 1.9:1 in WT and 1:1 in AN112. Excitation of WT cells with broad-band 620 nm light, which is absorbed primarily by Phc and Aphc and to a much lesser extent by chlorophyll (Chl), sensitized the RC of photosystem (PS) II at about 15 times the rate it sensitized RCI. This implies that PBSs are associated exclusively with PSII complexes and that PBS excitation is not transferred to PSI. The AN112 mutant of Synechococcus produced smaller PBSs consisting of the Aphc-containing core and of only six Phc-containing hexamers, respectively. It lacked about 65% of the Phccontaining rod substractures. Under our experimental conditions, the effective absorption cross section of the mutant PBS was only about half that of the WT. In agreement, the rate of RCII excitation by 620 nm light was also about half of that measured in the WT. Thus, the rate of light absorption by PSII depends directly on PBS size and composition. The low rate of RCI excitation with 620 nm light was the same in WT and AN112 cells, apparently independent of the PBS effective absorption cross section. We propose a strict structural-functional association between PBS and PSII complex. Excessive light conditions repressed the levels of mRNAs accumulation of multiple Lhc genes encoding light-harvesting chlorophyll-a/b (LHC) proteins of photosystem (PS)II in the unicellular green alga, Chlamydomonas reinhardtii. The light intensity required for the repression tended to decrease with lowering temperature or CO(2) concentration. The responses of six LhcII genes encoding the major LHC (LHCII) proteins and two genes (Lhcb4 and Lhcb5) encoding the minor LHC proteins of PSII (CP29 and CP26) were similar. The results indicate that the expression of these Lhc genes is coordinately repressed when the energy input through the antenna systems exceeds the requirement for CO(2) assimilation. The Lhc mRNA level repressed under high-light conditions was partially recovered by adding the electron transport inhibitor 3-(3,4-dichlorophenyl)-1,1-dimethylurea, suggesting that redox signaling via photosynthetic electron carriers is involved in the gene regulation. However, the mRNA level was still considerably lower under high-light than under low-light conditions even in the presence of 3-(3,4-dichlorophenyl)-1,1-dimethylurea. Repression of the Lhc genes by high light was prominent even in the mutants deficient in the reaction center(s) of PSII or both PSI and PSII. The results indicate that two alternative processes are involved in the repression of Lhc genes under high-light conditions, one of which is independent of the photosynthetic reaction centers and electron transport events.  相似文献

Filamentous cyanobacteria are the main primary producers in biological desert sand crusts. The cells are exposed to extreme environmental conditions including temperature, light, and diurnal desiccation/rehydration cycles. We have studied the kinetics of activation of photosynthesis during rehydration of the cyanobacteria, primarily Microcoleus sp., within crust samples collected in the Negev desert, Israel. We also investigated their susceptibility to photoinhibition. Activation of the photosynthetic apparatus, measured by fluorescence kinetics, thermoluminescence, and low temperature fluorescence emission spectra, did not require de novo protein synthesis. Over 50% of the photosystem II (PSII) activity, assembled phycobilisomes, and photosystem I (PSI) antennae were detected within less than 5 min of rehydration. Energy transfer to PSII and PSI by the respective antennae was fully established within 10 to 20 min of rehydration. The activation of a fraction of PSII population (about 20%-30%) was light and temperature-dependent but did not require electron flow to plastoquinone [was not inhibited by 3-(3,4-dichlorophenyl)-1,1-dimethylurea]. The cyanobacteria within the crusts are remarkably resistant to photoinhibition even in the absence of protein synthesis. The rate of PSII repair increased with light intensity and with time of exposure. Consequently, the extent of photoinhibition in high-light-exposed crusts reached a constant, relatively low, level. This is in contrast to model organisms such as Synechocystis sp. strain PCC 6803 where PSII activity declined continuously over the entire exposure to high illumination. Ability of the crust's organisms to rapidly activate photosynthesis upon rehydration and withstand photoinhibition under high light intensity may partly explain their ability to survive in this ecosystem.  相似文献

Stationary delayed fluorescence (DF) of chlorophyll in isolated membrane preparations from thermophilic cyanobacterium Synechococcus elongatus was investigated as a function of temperature. Two peaks at different temperatures were observed. The low-temperature peak (54-60 degrees C) coincided with the main maximum of the thermally-induced delayed fluorescence of chlorophyll in intact cells and PSII-particles with active oxygen-evolving system. The high-temperature peak (78 degrees C) coincided with the minor band of delayed light emitted by intact cells. It was also observed in the delayed fluorescence emission from a PSI-enriched fraction preparation. The intensities of the DF peaks were dependent on the presence of inhibitors, donors and acceptors that cause specific effects on electron transport of the two photosystems. The low-temperature and high-temperature peaks were related to PSII and PSI, respectively. The manifestation of delayed fluorescence from PSI and PSII at different temperatures seems to be a specific property of thermophilic cyanobacteria. The reason for this may be a high thermal stability of the photosystems and the lack of the PSII antenna complex in isolated membranes. Consequently, the relative yield of delayed fluorescence from PSI markedly increases. Thermally-induced fluorescence seen in membranes of cyanobacteria showed a high sensitivity to structural and functional membrane alterations induced by pH changes, different electron transport stabilizing agents or different concentrations of MgCl2.  相似文献

Eosin, a known generator of singlet oxygen, applied to leaf discs of Pisum sativum L. sensitized the inhibition of photosynthesis. Analysis of partial photosynthetic electron-transport reactions and of the kinetics of variable chlorophyll fluorescence located the damage at photosystem II. This injury required the presence of oxygen and was also caused by the irradiation of eosin-treated leaf tissue with green light. Photosynthetic electron flow changed considerably during desiccation and re-hydration of the intertidal macroalgae Porphyra haitanensis. Activities of both photosystem (PSI) and photosystem (PSII) increased significantly at moderate desiccation levels. Whereas PSII activity was abolished at an absolute water content (AWC) <24 %, PSI remained active with progressive decreases in AWC to values as low as 16 %. This result suggested that cyclic electron flow around PSI was still active after inactivation of linear electron flow following severe desiccation. Moreover, the PSI activity was restored more rapidly than that of PSII upon re-hydration. Pretreatment of the blades with 3-(3′,4′-dichlorophenyl)-1,1-dimethylurea (DCMU) suppressed PSII activity following desiccation to an AWC of ~16 % AWC. Cyclic electron flow around PSI decreased markedly in blades pretreated with DCMU than in blades without pretreatment of DCMU during re-hydration in seawater containing DCMU. All results suggested that the activity of PSII under desiccation conditions plays an important role in the operation of cyclic electron flow during desiccation and its recovery during re-hydration. Therefore, we proposed the PSII activity during desiccation could eventually lead to the accumulation of NADPH, which could serve as electron donor for P700+ and promote its recovery during re-hydration, thereby favoring the operation of cyclic electron flow.  相似文献

Photosynthetic activities were analyzed in Chlamydomonas reinhardtii mitochondrial mutants affected in different complexes (I, III, IV, I + III, and I + IV) of the respiratory chain. Oxygen evolution curves showed a positive relationship between the apparent yield of photosynthetic linear electron transport and the number of active proton-pumping sites in mitochondria. Although no significant alterations of the quantitative relationships between major photosynthetic complexes were found in the mutants, 77 K fluorescence spectra showed a preferential excitation of photosystem I (PSI) compared with wild type, which was indicative of a shift toward state 2. This effect was correlated with high levels of phosphorylation of light-harvesting complex II polypeptides, indicating the preferential association of light-harvesting complex II with PSI. The transition to state 1 occurred in untreated wild-type cells exposed to PSI light or in 3-(3,4-dichlorophenyl)-1,1-dimethylureatreated cells exposed to white light. In mutants of the cytochrome pathway and in double mutants, this transition was only observed in white light in the presence of 3-(3,4-dichlorophenyl)-1,1-dimethylurea. This suggests higher rates of nonphotochemical plastoquinone reduction through the chlororespiratory pathway, which was confirmed by measurements of the complementary area above the fluorescence induction curve in dark-adapted cells. Photo-acoustic measurements of energy storage by PSI showed a stimulation of PSI-driven cyclic electron flow in the most affected mutants. The present results demonstrate that in C. reinhardtii mutants, permanent defects in the mitochondrial electron transport chain stabilize state 2, which favors cyclic over linear electron transport in the chloroplast.  相似文献

The present study shows that in the presence of 600 nm light, sulfide acts as a specific inhibitor of photosynthetic electron transport between water and Photosystem II in the cyanobacteria Aphanothece halophytica and Synechococcus 6311 as well as in tobacco chloroplasts. In the presence of 600 nm light sulfied affects the fast fluorescence transients as does a low concentration (10 mM) of hydroxylamine; the fluorescence yield decreases in the presence of either chemical and can be restored by the addition of 3-(3,4-dichlorophenyl)-1,1-dimethylurea. In chloroplasts, however, NH2OH, an electron donor at high concentrations (40 mM), relieves the sulfide effect. In the dark, sulfide affects the cyanobacterial fluorescence transients through decrease of oxygen tension. The fluorescence yield increases in a similar pattern to that observed under nitrogen flushing. Upon omission of sulfide in A. halophytica, the characteristic aerobic fluorescence transients return, consistent with the ease of alternation between oxygenic and sulfide-dependent anoxygenic photosynthesis in many cyanobacteria.  相似文献

Sargassum fusiforme, a species of brown seaweed with economic importance, inhabits lower intertidal zones where algae are often exposed to various stresses. In this study, changes in the photosynthetic performance of S. fusiforme under saline stress were investigated. The PSII performance in S. fusiforme significantly improved, when the thalli were exposed to 0% salinity, and remained high with prolonging treatment time. In contrast, the PSII activity declined considerably under salinities of 4.5 and 6%. The PSI activity did not change remarkably under saline stress, thus demonstrating higher tolerance to saline stress than PSII. In addition, the PSI activity could be also restored after saline treatments, when PSII was inhibited by 3-(3,4-dichlorophenyl)-1,1-dimethylurea. It might be as a result of changes in the NAD(P)H content in the thalli under saline stress. Our results suggested that PSI was much more tolerant to different saline stress than PSII in S. fusiforme. We demonstrated that S. fusiforme was much more tolerant to hyposaline than to hypersaline stress.  相似文献

The fluorescence induction curve of photoinhibited thylakoids measured in the presence of 3-(3,4-dichlorophenyl)-1,1-dimethyl urea was modeled using an extension of the model of Lavergne and Trissl (Biophys. J. 68:2474-2492), which takes into account the reversible exciton trapping by photosystem II (PSII) reaction centers and exciton exchange between PSII units. The model of Trissl and Lavergne was modified by assuming that PSII consists of photosynthetically active and photoinhibited (inactive in oxygen evolution) units and that the inactive PSII units can efficiently dissipate energy even if they still retain the capacity for the charge separation reaction. Comparison of theoretical and experimental fluorescence induction curves of thylakoids, which had been subjected to strong light in the presence of the uncoupler nigericin, suggests connectivity between the photoinhibited and active PSII units. The model predicts that photoinhibition lowers the yield of radical pair formation in the remaining active PSII centers. However, the kinetics of PSII inactivation in nigericin-treated thylakoids upon exposure to photoinhibitory light ranging from 185 to 2650 micromol photons m-2 s-1 was strictly exponential. This may suggest that photoinhibition occurs independently of the primary electron transfer reactions of PSII or that increased production of harmful substances by photoinhibited PSII units compensates for the protection afforded by the quenching of excitation energy in photoinhibited centers.  相似文献

Initiation of thylakoid membrane assembly was examined in degreened cells of Chlamydomonas reinhardtii y1 cells depleted of thylakoid membranes and photosynthetic activity by growth in the dark for 3 to 4 d. Photoreductive activities of photosystem II (PSII) and photosystem I (PSI) increased with no apparent lag when degreened cells were exposed to light at 38[deg]C. However, fluorescence transients induced by actinic light, which reflect the functional state of PSII, changed only slightly during the first 2 h of greening. When these cells were treated with 3-(3,4-dichlorophenyl)-1,1-dimethyl urea (DCMU) or saturating light, fluorescence increased commensurate with the cellular content of chlorophyll. In similar experiments with greening cells of C. reinhardtii CC-2341 (ac-u-g-2.3), a PSI-minus strain, fluorescence increased with chlorophyll without treatment with DCMU. These data suggested that fluorescence of initial PSII centers in greening y1 cells was quenched by activity of PSI. Continuous monitoring of fluorescence in the presence or absence of DCMU showed that assembly of quenched PSII centers occurred within seconds after exposure of y1 cells to light. These results are consistent with initial assembly of PSI and PSII within localized domains, where their proximity allows efficient energy coupling.  相似文献

Photosystem (PS) II activity of a sulfoquinovosyl diacylglycerol (SQDG)-deficient mutant (hf-2) of Chlamydomonas was partially decreased compared with that of wild-type. The susceptibility to 3-(3,4-dichlorophenyl)-1,1-dimethylurea (DCMU) was also modified in the mutant. Photometric measurements in the isolated thylakoid membranes of hf-2 revealed that the lowered activity in the mutant was derived from a decrease in the efficiency of the electron donation from water to tyrosine Z, not from the efficiency of the electron transport from Q(A) to Q(B). This result was confirmed by the decay kinetics of chlorophyll fluorescence determined in vivo. We conclude that SQDG contributes to maintaining the conformation of PSII complexes, particularly that of D1 polypeptides, which are necessary for maximum activities in Chlamydomonas.  相似文献

Induction of chlorophyll a fluorescence and photosynthesis as affected by temperature were measured in cucumber leaf discs. Abrupt changes of the maximal variable fluorescence, Fv(p), and photosynthesis were observed around 9° and 21°C when the temperature was decreased from 30° to 0°C. The temperature-dependent maximal fluorescence of DCMU-treated leaf discs showed a single change around 21°C. Photosystem I-driven cyclic electron transport was measured in intact cells of Synechococcus sp PCC 7942 grown under different light intensities using photoacoustic and spectroscopic methods. The light-saturated capacity for PS I cyclic electron transport increased relative to chlorophyll concentration, PS I concentration, and linear electron transport capacity as growth light intensity was raised. In cells grown under moderate to high light intensity, PS I cyclic electron transport was nearly insensitive to methyl viologen, indicating that the cyclic electron supply to PS I derived almost exclusively from a thylakoid dehydrogenase. In cells grown under low light intensity, PS I cyclic electron transport was partially inhibited by methyl viologen, indicating that part of the cyclic electron supply to PS I derived directly from ferredoxin. 2-Amino-1,1,3-tricyanopropene inhibits photosynthetic O2 evolution, but, unlike 3-(3,4-dichlorophenyl)-1,1-dimethylurea, has little effect on the steady-state fluorescence of chlorophyll. In chloroplasts prepared from spinach leaves and inhibited by 2-amino-1,1,3-tricyanopropene, a 3-(3,4-dichlorophenyl)-1,1-dimethylurea-sensitive photoreduction of ferricyanide may be restored by addition of semicarbazide. It is concluded that 2-amino-1,1,3-tricyanopropene acts at a point close to the photo-oxidation of water.  相似文献
