Levels of DNA single-strand break were assayed in brain cells from rats acutely exposed to low-intensity 2450 MHz microwaves using an alkaline microgel electrophoresis method. Immediately after 2 h of exposure to pulsed (2 μs width, 500 pulses/s) microwaves, no significant effect was observed, whereas a dose rate-dependent [0.6 and 1.2 W/kg whole body specific absorption rate (SAR)] increase in DNA single-strand breaks was found in brain cells of rats at 4 h postexposure. Furthermore, in rats exposed for 2 h to continuous-wave 2450 MHz microwaves (SAR 1.2 W/kg), increases in brain cell DNA single-strand breaks were observed immediately as well as at 4 h postexposure. © 1995 Wiley-Liss, Inc.  相似文献

Helix aspersa neurons were irradiated with continuous-wave (CW) and noise-amplitude-modulated microwaves (carrier frequency 2450 MHz, 20% AM, 2 Hz-20 kHz) in a specially designed waveguide exposure system. Continuous-wave microwave irradiations were conducted at 8 degrees, 21 degrees, and 28 degrees C, while noise-modulated irradiation was performed at 21 degrees C. The results showed that exposure of snail neurons to CW microwaves for 60 min at 12.9 W/kg inhibited spontaneous activity and reduced input resistance at 8 degrees and 21 degrees C but not at 28 degrees C. The relative decrease in resistance at 21 degrees C was half that at 8 degrees C. Exposure of neurons to noise-modulated microwaves at 6.8 and 14.4 W/kg predominately caused excitatory responses characterized by augmented membrane resistance and the appearance of greater activity. The effect differed qualitatively from the inhibition observed with continuous, unmodulated microwave irradiation.  相似文献

Microwave‐induced corneal endothelial damage was reported to have a low threshold (2.6 W/kg), and vasoactive ophthalmologic medications lowered the threshold by a factor of 10–0.26 W/kg. In an attempt to confirm these observations, four adult male Rhesus monkeys (Macaca mulatta) under propofol anesthesia were exposed to pulsed microwaves in the far field of a 2.8 GHz signal (1.43 ± 0.06 µs pulse width, 34 Hz pulse repetition frequency, 13.0 mW/cm2 spatial and temporal average, and 464 W/cm2 spatial and temporal peak (291 W/cm2 square wave equivalent) power densities). Corneal‐specific absorption rate was 5.07 W/kg (0.39 W/kg/mW/cm2). The exposure resulted in a 1.0–1.2 °C increase in eyelid temperature. In Experiment I, exposures were 4 h/day, 3 days/week for 3 weeks (nine exposures and 36 h total). In Experiment II, these subjects were pretreated with 0.5% Timolol maleate and 0.005% Xalatan® followed by 3 or 7 4‐h pulsed microwave exposures. Under ketamine–xylazine anesthesia, a non‐contact specular microscope was used to obtain corneal endothelium images, corneal endothelial cell density, and pachymetry at the center and four peripheral areas of the cornea. Ophthalmologic measurements were done before and 7, 30, 90, and 180 days after exposures. Pulsed microwave exposure did not cause alterations in corneal endothelial cell density and corneal thickness with or without ophthalmologic drugs. Therefore, previously reported changes in the cornea exposed to pulsed microwaves were not confirmed at exposure levels that are more than an order of magnitude higher. Escherichia coli pol A+ and pol A? strains were exposed to 8.8-GHz microwaves pulsed at 1,000 Hz (1-μs pulse width) and an SAR of 40 W/kg, which increased the temperature of the cell culture by 7 °C. Our goal was to investigate effects of long-term exposure to pulsed microwave radiation. The major emphasis was to expose a large sample of experimental animals throughout their lifetimes and to monitor them for effects on general health and longevity. An exposure facility was developed that enabled 200 rats to be maintained under specific-pathogen-free (SPF) conditions while housed individually in circularly-polarized waveguides. The exposure facility consisted of two rooms, each containing 50 active waveguides and 50 waveguides for sham (control) exposures. The experimental rats were exposed to 2,450-MHz pulsed microwaves at 800 pps with a 10-microseconds pulse width. The pulsed microwaves were square-wave modulated at 8-Hz. Whole body calorimetry, thermographic analysis, and power-meter analysis indicated that microwaves delivered at 0.144 W to each exposure waveguide resulted in an average specific absorption rate (SAR) that ranged from 0.4 W/kg for a 200-g rat to 0.15 W/kg for an 800-g rat. Two hundred male, Sprague-Dawley rats were assigned in equal numbers to radiation-exposure and sham-exposure conditions. Exposure began at 8 weeks of age and continued daily, 21.5 h/day, for 25 months. Animals were bled at regular intervals and blood samples were analyzed for serum chemistries, hematological values, protein electrophoretic patterns, thyroxine, and plasma corticosterone levels. In addition to daily measures of body mass, food and water consumption by all animals, O2 consumption and CO2 production were periodically measured in a sub-sample (N = 18) of each group. Activity was assessed in an open-field apparatus at regular intervals throughout the study. After 13 months, 10 rats from each group were euthanatized to test for immunological competence and to permit whole-body analysis, as well as gross and histopathological examinations. At the end of 25 months, the survivors (11 sham-exposed and 12 radiation-exposed rats) were euthanatized for similar analyses. The other 157 animals were examined histopathologically when they died spontaneously or were terminated in extremis.  相似文献

Electrical characteristics of motor cortical neurons were studied in acute experiments on immobilized cats. Values of the input resistances varied from units to tens of megohms (mean 11.11±3.93 MΩ). The threshold current is a hyperbolic function of input resistance of the corresponding neurons and negative correlation was found between the axonal conduction velocity and input resistance. The time constant (τ0) of the membrane was 7.1±3.46 msec. A time constant τ1, of 1.65±0.36 msec, could also be distinguished in some neurons. Electrotonic lengths of dendrites of the cortical neurons were calculated by the use of Rall's model: mean 3.66±0.94 (in units of length constant).  相似文献

Na+, K+, and Ca2+ concentrations in the blood serum and submaxillary salivary gland (SSG) were investigated in adult, male rats exposed to 2880-MHz microwaves modulated with 1.5-μs pulses at a pulse repetition rate of 1000 Hz or in a hyperthermal environment. Rats were exposed, one at a time, for 30 min to microwaves producing a specific absorption rate (SAR) of: 4.2, 6.3,6.8,8.4, 10.8, or 12.6 W/kg, or were sham exposed under similar environmental conditions. In a second series, one group of rats was exposed singly for 15, 30, or 60 min to microwaves producing an SAR of 9.5 W/kg and other rats were exposed for similar periods at 40 °C; and 10 rats were sham exposed. Flame photometric analysis indicated that the thresholds of microwave radiation required to induce a change in Na+, K+, and Ca2+ concentrations in the salivary glands are 6.8, 6.8, and 6.3 W/kg, respectively. The directions of Na+, K+, and Ca2+ ion shifts in exposed rats' salivary glands are similar, whether affected by microwaves or hyperthermia. Greater changes in Na+ and K+ concentrations in SSG of rats exposed to microwaves for 15 and 30 min were found than in those exposed at 40 °C. On the other hand, exposure to hyperthermia at 40 °C or to microwaves for 1 h caused Na+ concentration to be increased by 68.7 and 59.5% and K+ concentration to be decreased by 29.6 and 21.7%, respectively.  相似文献

In the course of electrophysiological experiments, two types of the neurons of the edible snail Helix lucorum were detected, which responded by different way to application of antibodies to the neuron-specific calcium-binding S 100 protein (AS1000). Under effect of AS100, frequency of the action potential (AP) generation in the spontaneously active V1, V3, V17, and RPa6 cells decreased, whereas in V4 and V6 cells increased. On addition of quinine solution the AP generation frequency of these neurons decreased more than twice, while the AP duration (t S) rose 6 times. The combined action of AS100 and quinine did not change statistically significantly the AP generation frequency, membrane potential (MP) and AP generation threshold (APt), as compared with the effect of AS100 in saline. The value of the AP duration (t S) increased 1.6 times, which was less pronounced as compared with the quinine action in saline. This means that AS100 prevents an increase of the AP duration after the quinine application (block of the Ca-depended K-channels). The main AS100 effect at the level of the ionic currents is shown to consist in a decrease of the maximal value of the input current, on average, by 20%, while of the output current, on average, by 12%.  相似文献

The behavior of cultured myotubes from chick embryos exposed to microwaves has been experimentally analyzed. Recordings of acetylcholine-induced currents have been obtained via patch-clamp techniques using both cell-attached (single-channel current recording) and whole-cell (total current recording) configurations. During the exposure to low-power microwaves the frequency of the ACh-activated single channel openings decreased, while the ACh-induced total current showed a faster falling phase. Channel open time and conductance were not affected by microwave irradiation. It is concluded that the exposure to microwaves increases the rate of desensitization and decreases the channel opening probability. The nonthermal origin and the molecular interaction mechanisms governing these electromagnetic-induced effects are discussed.  相似文献

The electrophysiological responses of neurons in the canine area postrema (AP) to ionophoretic application of neuropeptides and transmitters were studied and correlated with the presence or absence of an emetic response on systemic administration. Of 17 common neuropeptides 11 were emetic when applied systemically at doses of 0.03-0.35 mg/kg. The emesis was dose dependent and was no longer observed in animals with chronic ablation of the AP. The responses of 122 AP single units were recorded. Neurons were silent at rest, and most were excited by glutamate, apomorphine, and dopamine. Excitatory responses to each of eight emetic peptides were recorded in 22-65% of cells studied; no responses were found to two peptides that were not emetic. The response to glutamate was always a brief, high-frequency discharge; the responses to all 13 other excitatory substances were of long latency, low frequency, and long duration. With high ionophoretic current or multiple applications, units would frequently become spontaneously active for many minutes or longer. The similarity of response of so many substances on small neurons suggests a common ionic or metabolic mechanism underlying the response. The direct correlation between the occurrence of emesis on systemic administration and the presence of excitatory receptors on AP neurons provides strong support for the proposed role of the AP as the chemoreceptor trigger zone for emesis.  相似文献

Microwave evoked body movements were studied in mice. A resonant cavity was used to provide head and neck exposure of the mouse to pulsed and gated continuous wave (CW) 1.25 GHz microwaves. No difference in response to pulsed and gated CW stimuli of equal average power was found. The incidence of the microwave evoked body movements increased proportionally with specific absorption (dose) when the whole-body average specific absorption rate was at a constant level (7300 W/kg). Under a constant average specific absorption rate, the response incidence reached a plateau at 0.9 kJ/kg. For doses higher than 0.9 kJ/kg, response incidence was proportional to the specific absorption rate and reached a plateau at 900 W/kg. Body movements could be evoked by a single microwave pulse. The lowest whole-body specific absorption (SA) tested was 0.18 kJ/kg, and the corresponding brain SA was 0.29 kJ/kg. Bulk heating potentials of these SAs were less than 0.1 °C. For doses higher than 0.9 kJ/kg, the response incidence was also proportional to subcutaneous temperature increment and subcutaneous heating rate. The extrapolated absolute thresholds (0% incidence) were 1.21 °C temperature increment and 0.24 °C/s heating rate. Due to high subcutaneous heating rates, these microwaves must be perceived by the mouse as an intense thermal sensation but not a pain sensation because the temperature increment was well below the threshold for thermal pain. Results of the present study should be considered in promulgation of personnel protection guideline against high peak power but low average power microwaves. © 1994 Wiley-Liss, Inc.  相似文献

Chopper neurons in the cochlear nucleus are characterized by intrinsic oscillations with short average interspike intervals (ISIs) and relative level independence of their response (Pfeiffer, Exp Brain Res 1:220–235, 1966; Blackburn and Sachs, J Neurophysiol 62:1303–1329, 1989), properties which are unattained by models of single chopper neurons (e.g., Rothman and Manis, J Neurophysiol 89:3070–3082, 2003a). In order to achieve short ISIs, we optimized the time constants of Rothman and Manis single neuron model with genetic algorithms. Some parameters in the optimization, such as the temperature and the capacity of the cell, turned out to be crucial for the required acceleration of their response. In order to achieve the relative level independence, we have simulated an interconnected network consisting of Rothman and Manis neurons. The results indicate that by stabilization of intrinsic oscillations, it is possible to simulate the physiologically observed level independence of ISIs. As previously reviewed and demonstrated (Bahmer and Langner, Biol Cybern 95:371–379, 2006a), chopper neurons show a preference for ISIs which are multiples of 0.4 ms. It was also demonstrated that the network consisting of two optimized Rothman and Manis neurons which activate each other with synaptic delays of 0.4 ms shows a preference for ISIs of 0.8 ms. Oscillations with various multiples of 0.4 ms as ISIs may be derived from neurons in a more complex network that is activated by simultaneous input of an onset neuron and several auditory nerve fibers.  相似文献

Female CD-1 mice immunized against the bacterium Streptococcus pneumoniae type III were exposed to 9-GHz pulsed microwaves (pulse repetition rate 970-1,000, pulse width 1.0 microseconds, peak power 1 W/cm2) at an average incident power density of 1 mW/cm2 (calculated SAR congruent to 0.47 W/kg) for 2 h per day for 5 days. Circulating antibody titers for the microwave-exposed animals were not significantly different from those of the sham-irradiated animals, and there were no differences in any of the hematological parameters analyzed, indicating that 9-GHz pulsed microwaves at 1 mW/cm2 do not alter the immune response of mice immunized against S pneumoniae.  相似文献

In experiments on mice, we studied changes in the level of analgesia induced by irradiation of the antipain acupuncture point (AP) E36 by low-intensity microwaves under conditions of modification of the serotonin level in the brain; this level was modified by injection of 300 mg/kg DL-p-chlorophenylalanine (pCPA). The duration of the nociceptive behavioral reaction (licking the limb) caused by injection of the formalin solution into the foot dorsal surface increased 24, 48, and 72 h after pCPA injection by 99.9, 84.4, and 114.4%, as compared with those in animals subjected to microwave irradiation of the AP E36 with no preliminary pCPA injection. It is concluded that the brain serotonergic system is actively involved in the analgesia effects induced by irradiation of the AP by low-intensity microwaves.  相似文献

Action potentials (APs) and impulse responses in the soma and axon of the rapidly and slowly adapting (SA) abdominal stretch receptor neurons of the crayfish (Astacus leptodactylus) were recorded with single microelectrode current-clamp technique. Impulse frequency response to constant current injection was almost constant in the SA neuron while the response decayed completely in the rapidly adapting (RA) neuron. Mean impulse frequency responses to current stimulations were similar in the receptor neuron pairs. In the RA neuron additional current steps evoked additional impulses while a sudden drop in the current amplitude caused adaptation. Impulse duration was dependent on the rate of rise when current ramps were used. Adaptation was facilitated when calculated receptor current was used. Exposing the neuron to 3 mmol/l TEA or scorpion venom resulted in partly elongated impulse responses. SA neuron could continuously convert the current input into impulse frequency irrespective of previous stimulation conditions. Exposing the SA neuron to 3 mmol/l TEA or 1 mmol/l Lidocaine reduced impulse duration to large current stimulations. The SA neuron fired spontaneously if it was exposed to 5-10 mmol/l Lidocaine or 10(-2) mg/ml Leiurus quinquestriatus venom. The action potential (AP) amplitudes in the RA soma, RA axon, SA soma, and SA axon were significantly different between components of all pairs. Duration of the AP in the axon of the RA neuron was significantly shorter than those in the RA soma, SA soma, and SA axon. Diameter of the RA axon was larger than that of the SA axon. Non-adapting impulse responses were promptly observed only in the SA axons. The results indicate that the RA neuron is a sort of rate receptor transducing the rapid length changes in the receptor muscle while the SA neuron is capable of transducing the maintained length changes in the receptor muscle. The differences in firing properties mainly originate from the differences in the active and passive properties of the receptor neurons.  相似文献

Although spike-frequency adaptation is a commonly observed property of neurons, its functional implications are still poorly understood. In this work, using a leaky integrate-and-fire neural model that includes a Ca2+-activated K+ current (I AHP), we develop a quantitative theory of adaptation temporal dynamics and compare our results with recent in vivo intracellular recordings from pyramidal cells in the cat visual cortex. Experimentally testable relations between the degree and the time constant of spike-frequency adaptation are predicted. We also contrast the I AHP model with an alternative adaptation model based on a dynamical firing threshold. Possible roles of adaptation in temporal computation are explored, as a a time-delayed neuronal self-inhibition mechanism. Our results include the following: (1) given the same firing rate, the variability of interspike intervals (ISIs) is either reduced or enhanced by adaptation, depending on whether the I AHP dynamics is fast or slow compared with the mean ISI in the output spike train; (2) when the inputs are Poisson-distributed (uncorrelated), adaptation generates temporal anticorrelation between ISIs, we suggest that measurement of this negative correlation provides a probe to assess the strength of I AHP in vivo; (3) the forward masking effect produced by the slow dynamics of I AHP is nonlinear and effective at selecting the strongest input among competing sources of input signals.  相似文献

Hypersalivation is an important mechanism for heat dissipation by animals without sweat glands. The water content and conductivity (at 20 kHz) in sub-maxillary salivary gland (SSG) and in other tissues were investigated in adult male rats exposed to microwaves (2880 MHz, 1.5 μs pulses at 1000 Hz) or to conventional heat at 40 °C. Eighty rats in one series were exposed, one at a time, for 30 min to microwaves producing a specific absorption rate (SAR) of 4.2,6.3,6.8,8.4,10.8 or 12.6 W/kg. Fifty rats were sham-exposed under similar environmental conditions. In the second series, ten rats were sham-exposed, 33 rats were exposed, one at time, for 15, 30 or 60 min to microwaves at a SAR of 9.5 W/kg, and 32 rats were exposed for similar periods to conventional heat at 40 °C. In rats of the first series colonic temperatures were elevated significantly at a SAR of 4.2 W/kg, while SSG water content and conductivity increased significantly at SAR values of 6.3 W/kg and higher. In the second series of experiments increases in colonic temperature and SSG water content were greater after 15 and 30 min of microwave exposure than after exposure to heat. Also, SSG conductivity was significantly depressed by heat and significantly increased by microwaves after exposure for 15 or 30 min. The results support the hypothesis that water content and conductivity of SSG of rats can be used as a sensitive specific test of a microwave induced thermal response.  相似文献

We have previously shown that late-developing avian nucleus magnocellularis (NM) neurons (embryonic [E] days 19–21) fire action potentials (APs) that resembles a band-pass filter in response to sinusoidal current injections of varying frequencies. NM neurons located in the mid- to high-frequency regions of the nucleus fire preferentially at 75 Hz, but only fire a single onset AP to frequency inputs greater than 200 Hz. Surprisingly, NM neurons do not fire APs to sinusoidal inputs less than 20 Hz regardless of the strength of the current injection. In the present study we evaluated intrinsic mechanisms that prevent AP generation to low frequency inputs. We constructed a computational model to simulate the frequency-firing patterns of NM neurons based on experimental data at both room and near physiologic temperatures. The results from our model confirm that the interaction among low- and high-voltage activated potassium channels (KLVA and KHVA, respectively) and voltage dependent sodium channels (NaV) give rise to the frequency-firing patterns observed in vitro. In particular, we evaluated the regulatory role of KLVA during low frequency sinusoidal stimulation. The model shows that, in response to low frequency stimuli, activation of large KLVA current counterbalances the slow-depolarizing current injection, likely permitting NaV closed-state inactivation and preventing the generation of APs. When the KLVA current density was reduced, the model neuron fired multiple APs per sinusoidal cycle, indicating that KLVA channels regulate low frequency AP firing of NM neurons. This intrinsic property of NM neurons may assist in optimizing response to different rates of synaptic inputs.  相似文献

Previous studies in our laboratory have established that pulsed microwaves at 2.45 GHz and 10 mW/cm2 are associated with production of corneal endothelial lesions and with disruption of the blood-aqueous barrier in the non-human primate eye. In the study reported here we examined ocular damage in monkeys (M. mulatta and M. fascicularis) following topical treatment with one of two ophthalmic drugs (timolol maleate and pilocarpine) that preceded exposure to pulsed microwaves. Anesthetized monkeys were sham exposed or exposed to pulsed, 2.45 GHz microwaves (10 microseconds, 100 pps) at average power densities of 0.2, 1, 5, 10, or 15 mW/cm2 4 h a day for 3 consecutive days (respective SARs were 0.052, 0.26, 1.3, 2.6, and 3.9 W/kg). Immediately before microwave exposure, one or both eyes were treated topically with one drop of 0.5% timolol maleate or of 2% pilocarpine. Following administration of a drug, we observed a significant reduction in the power-density threshold (from 10 to 1 mW/cm2) for induction of corneal endothelial lesions and for increased vascular permeability of the iris. Diagnostic procedures (in vivo specular microscopy and fluorescein iris angiography) were performed following each exposure protocol. In addition, increased vascular permeability was confirmed with horseradish peroxidase tracer techniques. Although we did not measure intraocular temperatures in experimental animals, the results suggest that a mechanism other than significant heating of the eye is involved. Our data indicate that pulsed microwaves at an average SAR of 0.26 W/kg, if administered after pretreatment with ophthalmic drugs, can produce significant ocular effects in the anesthetized primate.  相似文献
