Summary Cell lines from selected lepidopteran species were established for the overall purpose of use in baculovirus production. A total of 36 new cell lines from 10 lepidopteran species were generated, including cell lines from a pyralid, the European corn borer,Ostrinia nubilalis, a plutellid, the diamondback moth,Plutella xylostella, as well as eight noctuids: the black cutworm,Agrotis ipsilon, the celery looper,Anagrapha falcifera, the velvetbean caterpillar,Anticarsia gemmatalis, the corn earworm,Helicoverpa zea, the tobacco budworm,Heliothis virescens, the beet armyworm,Spodoptera exigua, the fall armyworm,Spodoptera frugiperda, and the cabbage looper,Trichoplusia ni. Tissues used for cell line establishment included fat bodies, ovaries, testes, or whole embryos/larvae/pupae. All the cell lines were subcultured numerous times, characterized by isoenzyme analysis and/or deoxyribonucleic acid amplification fingerprinting using polymerase chain reaction, and stored in liquid nitrogen. Many of the cell lines were adapted to grow in serum-free medium, with cell lines fromA. ipsilon andH. virescens being adapted to suspension culture, using shaker flasks. The potential use for these cell lines in baculovirus production is discussed. All programs and services of the U.S. Department of Agriculture are offered on a nondiscriminatory basis without regard to race, color, national origin, religion sex, age, marital status, or handicap.  相似文献

The influence of dietary allelochemical on ecdysone 20-monooxygenase activity was studied in the fall armyworm, Spodoptera frugiperda (J.E. Smith). Feeding the indoles (indole-3-carbinol, indole-3-acetonitrile), flavonoids (flavone, β-naphthoflavone), monoterpenes (menthol, menthone, peppermint oil), and a coumarin (xanthotoxin) to the larvae stimulated midgut microsomal ecdysone 20-monooxygenase activity from 28 to 200% as compared with the controls. β-Naphthoflavone was the most potent inducer among those tested. Phenobarbital, a well-known cytochrome P450 inducer, also caused a 2-fold increase in the microsomal ecdysone 20-monooxygenase activity. Ecdysone 20-monooxygenase activity was 2.7-fold higher in the microsomal fraction than in the mitochondrial fraction isolated from larval midguts. Microsomal ecdysone 20-monooxygenase activity was highest in the fat body, followed by the midgut and Malpighian tubules. Tissue localization and enzyme inducibility were different between ecdysone 20-monooxygenase and xenobiotic-metabolizing cytochrome P450 monooxygenases, including aldrin epoxidase, biphenyl hydroxylase, methoxyresorufin O-demethylase, 7-ethoxycoumarin O-deethylase, p-chloro-N-methylaniline N-demethylase, and phorate sulfoxidase in fall armyworm larvae. © 1995 Wiley-Liss, Inc.  相似文献

The symptomatology and histopathology of a nuclear polyhedrosis virus isolated from a larva of the alfalfa looper, Autographa californica, was studied by examining 13 tissues in the original and following alternate hosts: cabbage looper, Trichoplusia ni; beet armyworm, Spodoptera exigua; saltmarsh caterpillar, Estigmene acrea; corn carworm, Heliothis zea; cotton leafperforator, Bucculatrix thurberiella; and diamondback moth, Plutella xylostella. In all hosts, the hypodermal, tracheal matrix, and fat body cells were infected. Other tissues infected in some hosts included the Malpighian tubules, muscle, hemocytes, ganglia, midgut, hindgut, juvenile tissue (imaginal buds), and testes. No major changes in tissue tropisms were observed. The external symptoms were typical of nuclear polyhedrosis in all species except the corn earworm; in this host, development of the disease and death were delayed.  相似文献

Canola (Brassica napus L.) cultivars Oscar and Westar, engineered with a Bacillus thuringiensis (Bt) cryIA(c) gene, were evaluated for resistance to lepidopterous pests, diamondback moth, Plutella xylostella L. (Plutellidae) and corn earworm, Helicoverpa zea (Boddie) (Noctuidae) in greenhouse and field conditions. In greenhouse preference assays conducted at vegetative and flowering plant stages, transgenic plants recorded very low levels of damage. A 100% diamondback moth mortality and 90% corn earworm mortality were obtained on transgenic plants in greenhouse antibiosis assays. The surviving corn earworm larvae on transgenic plants had reduced head capsule width and body weight. Mortality of diamondback moth and corn earworm were 100% and 95%, respectively, at different growth stages (seedling, vegetative, bolting, and flowering) on the transgenic plants in greenhouse tests. In field tests conducted during 1995–1997, plots were artificially infested with neonates of diamondback moth or corn earworm or left for natural infestation. Transgenic plants in all the treatments were highly resistant to diamondback moth and corn earworm larvae and had very low levels of defoliation. Plots infested with diamondback moth larvae had greater damage in both seasons as compared with corn earworm infested plots and plots under natural infestation. After exposure to defoliators, transgenic plants usually had higher final plant stand and produced more pods and seeds than non-transgenic plants. Diamondback moth injury caused the most pronounced difference in plant stand and pod and seed number between transgenic and non-transgenic plants. Our results suggest that transgenic canola could be used for effective management of diamondback moth and corn earworm on canola.  相似文献

Forty-three isolates of the entomopathogenic fungus Beauveria bassiana were screened for virulence against second-instar larvae of diamondback moth (Plutella xylostella) (DBM), European corn borer (Ostrinia nubilalis) (ECB), corn earworm (Helicoverpa zea) (CEW), and fall armyworm (Spodoptera frugiperda) (FAW); 30 of these isolates were tested against beet armyworm (Spodoptera exigua) (BAW). Highly virulent isolates were also tested against black cutworm (Agrotis ipsilon) (BCW), and the most virulent isolate was also assayed against imported cabbage worm (Pieris rapae) (ICW) and cabbage looper (Trichoplusia ni) (CL). All lepidopteran species tested were susceptible to B. bassiana. Corn earworm and beet armyworm were most susceptible to fungal infection, and fall armyworm was least susceptible. Limited testing suggested low susceptibility of black cutworm and cabbage looper. B. bassiana isolate 1200 exhibited virulence against all pest species greater than or equal to commercial strain GHA of B. bassiana currently registered in the USA as BotaniGard®. In assays in which larvae were topically sprayed and maintained on the treated substrate for 24 h at 100% relative humidity, 6-day (25 °C) median lethal concentrations (LC50s) of this isolate against CEW, BAW, DBM, FAW, ICW, ECB, CL, and BCW were 4, 5, 7, 11, 12, 98, 125, and 273 conidia/mm2, respectively. The respective LC50s of commercial strain GHA against these pest species were 9, 67, 97, 1213, 29, 1668, 541, and 3504 conidia/mm2. Use of LC50 versus median lethal concentration ratios (comparing LC50s of each isolate to a “standard” strain) generated similar rankings of isolate virulence. Results from parametric ANOVAs of log LC50 values followed by Tukey HSD multiple comparisons tests and those from Kruskal-Wallis nonparametric analyses followed by sequential Bonferroni tests for means comparisons were nearly identical.  相似文献

A transgenic line of the soybean 'Jack', Glycine max (L.) Merrill, expressing a synthetic cry1Ac gene from Bacillus thuringiensis variety kurstaki (Jack-Bt), was evaluated for resistance to four lepidopteran pests in the field. Jack-Bt and genotypes serving as susceptible and resistant controls were planted in field cages and artificially infested with larvae of corn earworm, Helicoverpa zea (Boddie), and velvetbean caterpillar, Anticarsia gemmatalis (Hübner), in 1996, 1997, and 1998, and also with soybean looper, Pseudoplusia includens (Walker), in 1996. Susceptible controls included Jack (1996-1998), 'Cobb' (1996), and Jack-HPH (1996). GatIR 81-296 was used as the resistant control in all 3 yr. Compared with untransformed Jack, Jack-Bt showed three to five times less defoliation from corn earworm and eight to nine times less damage from velvetbean caterpillar. Defoliation of GatIR 81-296 was intermediate between that of Jack and Jack-Bt for corn earworm, and similar to that of Jack for velveltbean caterpillar. Jack-Bt exhibited significant, but lower resistance to soybean looper. Jack-Bt also showed four times greater resistance than Jack to natural infestations of lesser cornstalk borer, Elasmopalpus lignosellus (Zeller), in conventional field plots at two locations in 1998. Data from these experiments suggest that expression of this cry1Ac construct in soybean should provide adequate levels of resistance to several lepidopteran pests under field conditions.  相似文献

β-Glucosidase in larvae of the fall armyworm, Spodoptera frugiperda (J. E. Smith), was studied using helicin as the substrate. Enzyme activity was found in various tissues, with the midgut exhibiting the highest activity. Among the midgut subcellular fractions, the soluble fraction was the most active. The apparent Km value for the enzyme was 0.63 mM. The enzyme was fairly stable for 8 weeks when stored at −10°C. Activity was mainly located in the 60% fraction when the midgut soluble fraction was fractionated with ammonium sulfate. The system also hydrolyzed a variety of glucosides including numerous toxic plant allelochemicals. Substrate specificity of β-glucosidase was different in the fall armyworm, velvetbean caterpillar, cabbage looper and corn earworm, suggesting a qualitative difference in the enzyme among these species. In general, d-amygdalin, helicin, d(+)-cellobiose, p-nitrophenyl β-d-glucoside and 4-methylumbelliferyl β-d-glucoside were the preferred substrates, whereas sinigrin, phloridzin, α-solanine, tomatine and linamarin were poor substrates for the enzyme in these insects. Toxicity tests reveal that, in most instances, the glucosides were less toxic than their corresponding aglycones to the fall armyworm, indicating that β-glucosidase is an activation enzyme.  相似文献

Somatic embryos of jack, a Glycine max (L.) Merrill cultivar, were transformed using microprojectile bombardment with a synthetic Bacillus thuringiensis insecticidal crystal protein gene (Bt cryIAc) driven by the 35S promoter and linked to the HPH gene. Approximately 10 g of tissue was bombarded, and three transgenic lines were selected on hygromycin-containing media and converted into plants. The recovered lines contained the HPH gene, but the Bt gene was lost in one line. The plasmid was rearranged in the second line, and the third line had two copies, one of which was rear-ranged. The CryIAc protein accumulated up to 46 ng mg-1 extractable protein. In detached-leaf bioassays, plants with an intact copy of the Bt gene, and to a lesser extent those with the rearranged copy, were protected from damage from corn earworm (Helicoverpa zea), soybean looper (Pseudoplusia includens), tobacco budworm (Heliothis virescens), and velvetbean caterpillar (Anticarsia gemmatalis). Corn earworm produced less than 3% defoliation on transgenic plants, compared with 20% on the lepidopteran-resistant breeding line GatIR81-296, and more than 40% on susceptible cultivars. Unlike previous reports of soybean transformation using this technique, all plants were fertile. To our knowledge, this is the first report of a soybean transgenic for a highly expressed insecticidal gene.  相似文献

Zygotic hypocotyls of canola (Brassica napus L.) cv Oscar, cv Westar, and the breeding line UGA188-20B were transformed with a truncated synthetic Bacillus thuringiensis insecticidal crystal protein gene (Bt cryIAc) under the control of the cauliflower mosaic virus 35S promoter using Agrobacterium tumefaciens-mediated transformation. Fifty-seven independently transformed lines were produced, containing 1 to 12 copies of the transgenes. A range of cry expressors was produced from 0 to 0.4% Cry as a percentage of total extractable protein. The Brassica specialists, the diamondback month (Plutella xylostella L.) and the cabbage looper (Trichoplusia ni Hubner), were completely controlled by low-, medium-, and high-expressing lines. Whereas control of the generalist lepidopteran, the corn earworm (Helicoverpa zea Boddie), was nearly complete, the other generalist caterpillar tested, the beet armyworm (Spodoptera exigua Hubner), showed a dose response that had a negative association between defoliation and cry expression. These plants were produced as models for an ecological research assessment of the risk involved in the field release of naturalized transgenic plants harboring a gene (Bt) that confers higher relative fitness under herbivore-feeding pressure.  相似文献

Plant resistance is a useful component of integrated pest management for several insects that are economically damaging to maize, Zea mays L. In this study, 15 experimental lines of maize derived from a backcross breeding program were evaluated for resistance to corn earworm, Helicoverpa zea (Boddie); fall armyworm, Spodoptera frugiperda (J. E. Smith); southwestern corn borer, Diatraea grandiosella Dyar; and sugarcane borer, Diatraea saccharalis (F.). Experimental line 100-R-3 was resistant in the field to leaf feeding by fall armyworm and line 116-B-10 was resistant in the field to leaf feeding by fall armyworm and leaf and stalk feeding by southwestern corn borer. When corn earworm larvae were fed field harvested silks from experimental line 81-9-B in the laboratory, their pupal weights were significantly lower than the pupal weights of larvae that were fed silks from the resistant control, Zapalote Chico. Maysin levels lower than those commonly associated with corn earworm resistance were present in the resistant experimental line, 107-8-7, indicating a new basis confers resistance to corn earworm in this line. These resistant experimental lines will provide plant breeders with new sources of resistance to lepidopterous insects for the development of improved maize breeding populations.  相似文献

When caterpillars feed on maize (Zea maize L.) lines with native resistance to several Lepidopteran pests, a defensive cysteine protease, Mir1-CP, rapidly accumulates at the wound site. Mir1-CP has been shown to inhibit caterpillar growth in vivo by attacking and permeabilizing the insect's peritrophic matrix (PM), a structure that surrounds the food bolus, assists in digestion and protects the midgut from microbes and toxins. PM permeabilization weakens the caterpillar defenses by facilitating the movement of other insecticidal proteins in the diet to the midgut microvilli and thereby enhancing their toxicity. To directly determine the toxicity of Mir1-CP, the purified recombinant enzyme was directly tested against four economically significant Lepidopteran pests in bioassays. Mir1-CP LC(50) values were 1.8, 3.6, 0.6, and 8.0 ppm for corn earworm, tobacco budworm, fall armyworm and southwestern corn borer, respectively. These values were the same order of magnitude as those determined for the Bacillus thuringiensis toxin Bt-CryIIA. In addition to being directly toxic to the larvae, 60 ppb Mir1-CP synergized sublethal concentrations of Bt-CryIIA in all four species. Permeabilization of the PM by Mir1-CP probably provides ready access to Bt-binding sites on the midgut microvilli and increases its activity. Consequently, Mir1-CP could be used for controlling caterpillar pests in maize using non-transgenic approaches and potentially could be used in other crops either singly or in combination with Bt-toxins.  相似文献

Lettuce quality and yield can be reduced by feeding of several lepidopterous pests, particularly cabbage looper, Trichoplusia ni (Hübner), and beet armyworm, Spodoptera exigua (Hübner) (Lepidoptera: Noctuidae). Host plant resistance to these insects is an environmentally sound adjunct to conventional chemical control. In this study we compared the survival, development, and feeding behavior of cabbage looper and beet armyworm on two romaine lettuce cultivars, resistant 'Valmaine' and susceptible 'Tall Guzmaine'. Larval mortality of both species was significantly higher on resistant Valmaine than on susceptible Tall Guzmaine. The average weight per larva after feeding for 1 wk on Tall Guzmaine plants was 6 times (beet armyworm) and 2 times (cabbage looper) greater than that of larvae feeding on Valmaine plants. Significant reduction in larval growth on Valmaine compared with that on Tall Guzmaine resulted in a 5.9- (beet armyworm) and 2.6-d (cabbage looper) increase in larval duration and almost a 1-d increase in pupal duration. Average pupal and adult weights and successful pupation of cabbage looper and beet armyworm were reduced on Valmaine compared with Tall Guzmaine. The sex ratio of progeny did not deviate from 1:1 when larvae were reared on either Valmaine or Tall Guzmaine. The fecundity of cabbage looper and beet armyworm adults that developed from larvae reared on Valmaine was about one-third that of adults from Tall Guzmaine, but adult longevity did not significantly differ on the two lettuce cultivars. The two insect species showed different feeding preferences for leaves of different age groups on Valmaine and Tall Guzmaine. Cabbage loopers cut narrow trenches on the leaf before actual feeding to block the flow of latex to the intended site of feeding. In contrast, beet armyworms did not trench. The different feeding behavior of the two species on Valmaine may explain the superior performance of cabbage looper compared with beet armyworm.  相似文献

亚麻荠是一种很少有害虫危害的油料作物。用室内生测和Y 型嗅觉仪研究了亚麻荠对小菜蛾Plutella xylostella幼虫取食和成虫行 为反应的影响。以甘蓝作对照,用亚麻荠叶片喂养的小菜蛾初孵幼虫3天后校正死亡率为79 .2%,显示了较强的致死作用;喂养小菜蛾3龄幼虫至化蛹,其存活率、化蛹率、蛹重及成 虫寿命都显著降低,表明亚麻荠对小菜蛾幼虫的生长发育有不利影响。在幼虫的取食选择实 验中,有甘蓝叶供选择时,小菜蛾幼虫不取食亚麻荠;在无可选择的情况下,小菜蛾幼虫也 取 食亚麻荠叶片,但取食量很小,与取食甘蓝叶的量相比,差异极显著。行为反应测试表明, 小菜蛾成虫对甘蓝和亚麻荠植株的挥发物都有明显的趋性反应,与对照(净化空气)相比, 差异极显著,而在甘蓝和亚麻荠之间无选择性。说明小菜蛾成虫对亚麻荠植株的挥发物具有 较强的定向反应。  相似文献

A nuclear polyhedrosis virus isolated from the alfalfa looper, Autographa californica, was found to infect several species of caterpillars including the cabbage looper, Trichoplusia ni; the beet armyworm, Spodoptera exigua; and the saltmarsh caterpillar, Estigmene acrea. Studies were therefore conducted to determine the quantitative effects of passage through the alternate hosts, S. exigua and E. acrea, on the infectivity of this virus to newly hatched first-instar cabbage looper larvae. When 11 preparations of polyhedra obtained from a like number of primary passages through the original or alternate hosts were assayed and the mortality at 7-, 10-, and 14-day intervals were subjected to probit analysis, the LD50s for the three intervals differed but those for the preparations at any given interval did not. Therefore, any of the three hosts could be used to propagate the virus, and whichever proves the easiest to rear and provides the highest yields of polyhedra can be selected.  相似文献

Evaluation of combinations of flower odor compounds in the field revealed several compounds that were attractive or co-attractive with phenylacetaldehyde (PAA) to pest noctuid and pyralid moths. A number of moth species responded positively to the key floral odorant PAA. The floral odorants cis -jasmone, linalool, benzyl acetate, limonene, β-myrcene, methyl salicylate, and methyl 2-methoxybenzoate all increased captures of some moths when added to traps with PAA, but responses varied among the moth species that were trapped. For example, soybean looper moths, Pseudoplusia includens (Walker) (Lepidoptera: Noctuidae), most strongly responded to PAA + β-myrcene, but benzyl acetate, cis -jasmone, and limonene also increased captures of these moths when these compounds were used in traps along with PAA. Velvetbean caterpillar moths, Anticarsia gemmatalis Hübner (Lepidoptera: Noctuidae), responded most strongly to PAA + linalool, but β-myrcene, cis -jasmone, and limonene also increased captures of these moths in traps over numbers trapped with PAA. Positive responses to floral compound blends were also noted for golden looper [ Argyrogramma verruca (F.)], grass looper [ Mocis disseverans (Walker)], tobacco budworm [ Heliothis virescens (F.)], southern armyworm [ Spodoptera eridania (Stoll) (all Lepidoptera: Noctuidae)], and melonworm [ Diaphania hyalinata (L.) (Lepidoptera: Pyralidae)]. Overall, PAA + β-myrcene appeared to be the strongest floral compound combination for pest species trapped, compared to PAA or other compound blends.  相似文献

Nine pharmaceutical inhibitors of eicosanoid biosynthesis (e.g., bromophenacyl bromide, clotrimazole, diclofenamic acid, esculetin, flufenamic acid, indomethacin, nimesulide, sulindac, tolfenamic acid) that increased the susceptibility of the gypsy moth, Lymantria dispar (L.), to the nucleopolyhedrovirus LdMNPV were tested against the beet armyworm Spodoptera exigua (Hübner), the corn earworm Helicoverpa zea (Boddie) and the fall armyworm Spodoptera frugiperda (J.E. Smith) and their respective NPVs to determine whether these compounds also alter the susceptibility of these insects. The susceptibility of the beet armyworm was increased by six inhibitors (bromophenacyl bromide, clotrimazole, diclofenic acid, esculetin, flufenamic acid, nimesulide). The susceptibility of the fall armyworm was increased by seven inhibitors, (bromophenacyl bromide, diclofenamic acid, esculetin, indomethacin, nimesulide, sulindac, tolfenamic acid), whereas the susceptibility of the corn earworm was increased by only one inhibitor (sulindac). The influence of the cyclooxygenase inhibitor, indomethacin was expressed in a concentration-related manner in beet armyworms. We infer from these findings that eicosanoids, including prostaglandins and lipoxygenase products, act in insect anti-viral defenses.  相似文献

S-ethyldipropylthiocarbamate (EPTC) applied as a soil treatment or over-the-top spray on cabbage plants (Brassica oleracea L.) caused the leaves to turn ‘glossy’ for as long as 30 days. EPTC-induced glossy plants were damaged significantly less than untreated plants by diamondback moth,Plutella xylostella (L.), imported cabbage worm,Pieris rapae (L.), and cabbage looper,Trichoplusia ni (Hbn.). Reductions in damage were equivalent to those obtained from treatment with permethrin. When used in combination with permethrin, EPTC provided additive control of damage by these pests. Our calculations show EPTC-induced resistance to be cost-effective. This use of EPTC has several limitations, however. Younger plants (<9 leaves) were killed or injured by the herbicide. The growth of older plants was not affected, but plants did not become glossy for ca. 10 days after they were treated with EPTC. The crop must be protected with insecticides until the plants are mature enough to treat with EPTC, and until treated plants become glossy. In addition, since the glossy trait is only effective against first instar larvae, populations of later instars on glossy plants must be reduced with an application of insecticide. Finally, EPTC formulations are water-soluble and can be washed away from the plants by heavy rains and irrigation, which may make this use of EPTC impractical in some situations. Where its use is practical, and the indicated precautions are taken, EPTC-induced resistance could reduce dependence on chemical insecticides and reduce selection for insecticide resistance in diamondback moth.  相似文献

To identify novel bioinsecticidal agents, a bacterial strain, Serratia sp. EML-SE1, was isolated from a dead larva of the lepidopteran diamondback moth (Plutella xylostella) collected from a cabbage field in Korea. In this study, the insecticidal activity of liquid cultures in Luria-Bertani broth (LBB) and nutrient broth (NB) of a bacterial strain, Serratia sp. EML-SE1 against thirty 3rd and 4th instar larvae of the diamondback moth was investigated on a Chinese cabbage leaf housed in a round plastic cage (Ø 10×6 cm). 72 h after spraying the cabbage leaf with LBB and NB cultures containing the bacterial strain, the mortalities of the larvae were determined to be 91.7% and 88.3%, respectively. In addition, the insecticidal activity on potted cabbage containing 14 leaves in a growth cage (165×83×124 cm) was found to be similar to that of the plastic cage experiment. The results of this study provided valuable information on the insecticidal activity of the liquid culture of a Serratia species against the diamondback moth.  相似文献

In the mid-fifth instar larvae of the cabbage looper moth, Trichoplusia ni, the subcellular distribution of total superoxide dismutase was as follows: 3.05 units (70.0%), 0.97 units (22.3%), and 0.33 units (7.6%) mg-1 protein in the mitochondrial, cytosolic and nuclear fractions, respectively. No superoxide dismutase activity was detected in the microsomal fraction. Catalase activity was unusually high and as follows: 283.4 units (47.3%), 150.1 units (25.1%). 142.3 units (23.8%), and 22.9 units (3.8%) mg-1 protein in the mitochondrial, cytosolic, microsomal (containing peroxisomes), and nuclear fractions. No glutathione peroxidase activity was found, but appreciable glutathione reductase activity was detected with broad subcellular distribution as follows: 3.86 units (36.1%), 3.68 units (34.0%). 2.46 units (23.0%). and 0.70 units (6.5%) mg-1 protein in the nuclear, mitochondrial, and cytosolic fractions, respectively. The unusually wide intracellular distribution of catalase in this phytophagous insect is apparently an evolutionary adaptation to the absence of glutathione peroxidase; hence, lack of a glutathione peroxidase-glutathione reductase role in alleviating stress from lipid peroxidation. Catalase working sequentially to superoxide dismutase, may nearly completely prevent the formation of the lipid peroxidizing OH radical from all intracellular compartments by the destruction of H2O2 which together with O-2 is a precursor of OH.  相似文献
