Yearling brown trout, Salmo trutta , were exposed to low mineral content water (nominal concentrations of 20μmol 1−1 magnesium, 7.7 μmol 1−1 potassium, 44 μmol 1−1 sodium) over a pH range of 4.0–5.2 with ambient calcium concentrations of 2.5–60 μmol 1−1. All fish died at pH 4.0 and 4.2 irrespective of ambient calcium concentration and also at pH 4.4 with only 2–3 μmol 1 −1 calcium (that is calcium-free water except for that leached from the diet or excreted by the fish). Good growth rates were obtained over the remaining treatments which extended down to pH 4.4 with as little as 7 μmol 1−1 calcium. When starved, weight loss was inversely correlated with pH. Effects on plasma chloride, percentage dry weight and calcium, potassium sodium, and phosphorus contents of skin, muscle and bone tissue were also investigated. These demonstrated pH effects on mineral metabolism in starved fish, but no effects were detected in fed fish.  相似文献

Abstract Ethylene (ethene) was found to inhibit methane formation in slurries from sewage sludge and sediment samples taken from freshwater and marine sources. Methane formation from sediment contents was inhibited by 50% at 0.07% ethylene concentration in the gas phase (approx. 5 μmol · 1−1 in the aqueous phase) and by 94% at ≥0.05% ethylene in the gas phase (≥36 μ mol · 1−1 in the aqueous phase). Sulphate reduction was not impaired. Methane formation from added acetate, hydrogen or methanol was inhibited by ≥98%, from lactate by about 90%. The inhibition was reversible, and methanogenic activity recuperated completely after ethylene removal. Cyclopentadiene and cycloheptatriene led to strong inhibition; benzene, toluene, isoprene, and 1-hexine to moderate inhibition of methanogenesis; several unsaturated linear hydrocarbons were without effect. Pure cultures of Methanospirillum hungatei, Methanothrix soehngenii , and Methanosarcina barkeri were all inhibited by 50% at 0.05–0.1% ethylene concentration in the gas phase (3.6–7.2 μmol · 1−1 in the aqueous phase). Pure cultures of Acetobacterium woodii, Halobacterium halobium and Sulfolobus acidocaldarius were not significantly inhibited by either ethylene or acetylene. Ethylene is recommended as a selective inhibitor of methanogenesis for physiological and enrichment experiments with sediment and sludge samples.  相似文献

The effects of water hardness (9 and 220 mgl−1 as CaCO3) upon zinc exchange in brown trout exposed to 0.77 μmol Zn 1−1 have been investigated using artificial soft water (<49.9 μmol Ca l-1, <40.1 μmol Mg 1−1) and mains hard water (1671.7 μmol Ca 1−1, 493.6 μmol Mg 1−1) of known composition. Both hard and soft water-adapted fish exhibited a bimodal pattern of net zinc influx. Net zinc influxes during both fast and slow uptake phases were significantly greater ( P <0.001) in soft (82.9 and 6.2 μmol Zn 100 g−1 h−1) than in hard water (46.3 and 2.4 μmol Zn 100 g h−1). Zinc efflux (- 0.2 μmol Zn 100 g−1 h−1) was enhanced only in hard water during the slow net influx phase.
Brown trout exposed to zinc in hard water and placed in metal-free media exhibited a greater net efflux (- 25.6 μmol Zn 100 g−1 h−1) of the metal than did fish in soft water (-4.2 μmol Zn 100 g−1 h−1) treated in the same manner. Tissue 65Zn activities reflected both the differences in uptake and excretion rates of the metal between hard and soft water fish. During zinc exposure (0.77 μmol Zn 1−1) high water hardness reduced tissue burdens of the metal by reducing net branchial influx, and enhancing efflux of the metal in hard water fish.  相似文献

Dimethylsulfoxide reduction by marine sulfate-reducing bacteria   总被引:2,自引:0,他引:2  
Abstract Dimethylsulfoxide (DMSO) reduction occurred in five out of nine strains of sulfate-reducing bacteria from marine or saline environments, but not in three freshwater isolates. DMSO reduction supported growth in all positive strains. In Desulfovibrio desulfuricans strain PA2805, DMSO reduction occurred simultaneously with sulfate reduction and was not effectively inhibited by molybdate, a specific inhibitor of sulfate reduction. The growth yield per mol lactate was 26% higher with DMSO than with sulfate as electron acceptor. In extracts of cells of strain PA2805 grown on sulfate, a low level of DMSO-reducing activity was present (0.013 μmol (mg protein) min); higher levels were found in cells grown on DMSO (0.56 μmol (mg protein) min). In anoxic marine environments DMSO reduction by sulfate-reducing bacteria may lead to enhanced dimethylsulfide emission rates.  相似文献

Plasma membrane vesicles were isolated from the roots of 7-day-old rice plants ( Oryza sativa L. cv. Bahía) by utilizing an aqueous polymer two-phase system with 6.2%:6.2% (w/w) Dextran T500 and polyethylene glycol 3350 (PEG) at pH 7.6. Plasmalemma vesicles of high purity were obtained as indicated by the vanadate-sensitive K+, Mg2+-ATPase activity that was 18 times higher in the upper (PEG-rich) phase than in the lower (Dextran-rich) phase and by specific staining with sodium silicotungstate. Two peaks of ATPase activity were found. One showed a pH optimum at 6.0 in the presence of 150 m M KCl and 3 m M ATP with apparent Km (ATP) and Vmax of 0.75 m M and 79 μmol (mg protein)−1 h−1, respectively. With 50 m M KCl and 7 m M ATP a pH optimum of 6.5, an apparent Km (ATP) of 6.3 m M and Vmax of 159 μmol (mg protein)−1 h−1 were determined. Both activities were specific for ATP, unspecific for monovalent cations, sensitive to sodium vanadate and Ca2+ but insensitive to azide and nitrate.  相似文献

Abstract The anoxygenic phototrophic purple sulfur bacterium Thiocapsa roseopersicina was grown in illuminated continuous cultures with thiosulfate as growth limiting substrate. Aeration resulted in completely colorless cells growing chemotrophically, whereafter the conditions were changed to a 23 h oxic/1 h anoxic regime. After 11 volume changes at a dilution rate of 0.031 h−1 (35% of μmax) a time dependent equilibrium was established. During the 23 h oxic periods bacteriochlorophyll a synthesis (BChl a ) was not observed, whereas during the 1 h anoxic periods synthesis was maximal (i.e. 1.1 μg (mg protein)−1 h−1). As a result the BChl a concentration gradually increased from zero to an average value over 24 h of 1.9 μg (mg protein)−1. Concomitantly, the protein concentration increased from 13.9 mg 1−1 during continuous oxic conditions to 28.8 mg 1−1. For comparison, the protein concentration during fully phototrophic growth at an identical thiosulfate concentration in the inflowing medium was 53.7 mg 1−1. The specific respiration rate was 8 μmol O2 (mg protein)−1 h−1 during full chemotrophic growth and gradually decreased to 3.5 μmol O2 (mg protein)−1 h−1 after 11 volume changes at the regime employed. These data show that T. rosepersicina is able to simultaneously utilize light and aerobic respiration of thiosulfate as sources of energy. The ecological relevance of the data is discussed.  相似文献

Sperm structure and motility of the freshwater teleost Cottus gobio   总被引:1,自引:0,他引:1  
When motility of spermatozoa of Cottos gobio was initiated with distilled water, the motility rate decreased to 0% within 1 min, and significant signs of osmotic alterations were observed at the end of the motility period. By contrast, in 50 mmol 1−1 NaCl solution, the motility rate persisted for 120–140 min. In both distilled water and in 50 mmol 1−1 NaCl solution, the main swimming type of spermatozoa was linear motion during the whole motility period. The initial swimming velocity (50.0 ± 2.1 μm s−1) measured 10 s after motility initiation was similar in both distilled water and in 50 mmol 1−1 NaCl solution. In distilled water, the velocity decreased to <20 μm s−1 (locally motile) during the first minute of the motility phase. In 50 mmol 1−1 NaCl solutions, it remained at a constant level during the first 60 min of the motility period, but then started to decrease to <20 μm s−1 after 120 min. When 5 mmol 1−1 potassium cyanide, antimycin or atractyloside was added to the 50 mmol 1−1 NaCl solution, the motility period was reduced to ≤2min. Ten millimoles per litre 2-deoxy-D-glucose, malonate or a mixture of 5 mmol 1−1 atractyloside and 5 mmol 1−1 carnithine did not effect the duration of the motility period. This indicates that sperm energy metabolism depends mainly on respiration rate and fatty acid metabolism.  相似文献

Even in the presence of glucose the growth of Marchantia polymorpha L. (cell line HYH-2F) requires light, and growth is more sensitive to 10−6 M 3-(3, 4-dichlorophenyl)-1, 1-dimethylurea than to 10−4 Antimycin A. The inability of the cells to grow in the dark is due to the low level of respiration. The respiration rate under light increased to four times the dark value. The values of the compensation ratio (the photosyntehtic rate/the respiration rate) for the oxygen exchange were below 1.0 daring the growth period, although oxygen evolution was found. At the early exponential phase, oxygen evolution was 0.373 μmol (mg cell dry weight)−1 h−1 [61.7 μmol (mg chlorophyll)−1 h−1]. M. polymorpha cells are unable to grow anaerobically in the light without a supply of carbon dioxide. When 1% carbon dioxide in nitrogen is supplied, photochemically produced oxygen and energy are sufficient for sustained growth although at significantly reduced yields in both cell dry weight and chlorophyll. Photosyntehtic CO2 assimilation rate was 0.13 μmol (mg cell dry weight)−1 h−1[11.3 μmol (mg chlorophyll)−1 h−1]. At least one-third of the carbon atoms in cellular constituents seem to be derived from atmospheric carbon dioxide, which indicates that M. polymorpha cells grow photomixotrophicaily.  相似文献

Iron inefficiency in the maize ( Zea mays L.) mutant ysl is caused by a defect in the uptake system for Fe-phytosiderophores. To characterize this defect further, the uptake kinetics of Fe-phytosiderophores in ysl was compared to the Fe-efficient maize cultivar Alice. Short-term uptake of 59Fe-labeled Fe-deoxymugineic acid (Fe-DMA) was measured over a concentration range of 0.03 to 300 μM. Iron uptake in Fe-deficient plants followed Michaelis-Menten kinetics up to about 30 μM and was linear at higher concentrations, indicating two kinetically distinct components in the uptake of Fe-phytosiderophores. The saturable component had similar Km (∼ 10 μM) in both genotypes. In contrast. Vmax was 5.5 μmol Fe-DMA g−1 dry weight [30 min]−1 in Alice, but only 0.6 μmol Fe-DMA g−1 dry weight [30 min]−1 in ysl. Uptake experiments with double-labeled 59Fe-[14C]DMA suggest that in both cultivars Fe-DMA was taken up by the roots as the intact chelate. The results indicate the existence of a high-affinity and a low-affinity uptake system mediating Fe-phytosiderophore transport across the root plasma membrane in maize. Apparently, the mutation responsible for Fe inefficiency in ysl affected high-affected uptake and led to a decrease in activity and/or number of Fe-phytosiderophore transporters.  相似文献

Aromatic L-amino acid decarboxylase activity was measured in brain, heart, intestine, kidney, liver, muscle, pyloric caeca, spleen and stomach of skipjack, using L-3,4–dihydroxyphenylalanine as the substrate. Aromatic L-amino acid decarboxylase activity was found to be present in all of the organs studied. The highest activity was found in the intestine (1774 μmol min −1 g−1 wet wt of tissue). The liver showed the lowest activity (48.7 umol min −1 g −1).  相似文献

Abstract: The cultivation of the hyperthermophile Pyrococcus furiosus , the thermoacidophile Sulfolobus shibatae and the halophile Marinococcus M52 in dialysis membrane reactors resulted in cell yields of 2.6 g 1−1, 114 g 1−1 and 132 g 1−1 (cell dry weight), respectively. In the case of P. furiosus neither hydrogen (up to 160 μmol 1−1) nor the metabolic products were found to be responsible for growth cessation at a cultivation temperature of 90°C. The low cell yield at an agitation speed of above 1800 rpm demonstrates the sensitivity of P. furiosus to hydrodynamic stress. The oxygen transfer rate into culture medium at extreme temperatures was shown to be equal or even greater than that under mesophilic condition.  相似文献

Anaerobic production of succinate, a common feature in animals able to sustain anoxia, has seldom been reported in plants. By the use of 1H-nuclear magnetic resonance spectroscopy we show here that succinate is produced by rice seedlings (Oryza sativa L. cv. Arborio) subjected to anoxic conditions. Starting from levels below I μmol (g fresh weight)−1 in air, after 48 h of anoxia the levels of alanine, succinate and lactate had increased to 23.8, 5.2 and 1.0 μmol (g fresh weight) −1, respectively, in shoot tissues. Succinate was accumulated in shoots, notably in the coleoptiles, but not in roots of the rice seedlings, suggesting its involvement in rice coleoptile elongation under anoxia. Other possible functions of succinate production in rice seedling, an organism highly tolerant to anoxia, are discussed.  相似文献

Seedlings of two tree species from the Atlantic lowlands of Costa Rica, Ochroma la-gopus Swartz, a fast-growing pioneer species, and Pentaclethra macroloba (Willd.) Kuntze, a slower-growing climax species, were grown under enriched atmospheric CO2 in controlled environment chambers. Carbon dioxide concentrations were maintained at 350 and 675 μl 1−1 under photosynthetic photon flux densities of 500 μol m−2 s−1 and temperatures of 26°C day and 20°C night. Total biomass of both species increased significantly in the elevated CO2 treatment; the increase in biomass was greatest for the pioneer species, O. lagopus . Both species had greater leaf areas and specific leaf weights with increased atmospheric CO2. However, the ratio of non-pho-tosynthetic tissue to leaf area also increased in both species leading to decreased leaf area ratios. Plants of both species grown at 675 μl 1−1 CO2 had lower chlorophyll contents and photosynthesis on a leaf area basis than those grown at 350 μl 1−1. Reductions in net photosynthesis occurred despite increased internal CO2 concentrations in the CO2-enriched treatment. Stomatal conductances of both species decreased with CO2-enrichment resulting in significant increases in water use efficiency.  相似文献

SUMMARY. The concentrations of trace metals in filtered and unfiltered lake water were measured using anodic stripping voltammetry, before and after digestion by ultra-violet irradiation, and by atomic absorption spectroscopy with electrothermal atomization. Total soluble components were estimated to be: zinc, 2.1 μg 1−1; cadmium, <0.05 μg l−1; lead, <(0.1 μg l−1 and copper, 0.3 μg 1−1. Atomic absorption spectrophotometric results and u.v.-digested, anodic stripping voltammetric results were in good agreement. All measurable zinc was electrochemically labile whereas copper above the detection limit of 0.09 μg l−1 was electrochemically inert.  相似文献

The neuropeptide vasoactive intestinal peptide (VIP) is anti-inflammatory and protective in the immune and nervous systems, respectively. This study demonstrated in corneal endothelial (CE) cells injured by severe oxidative stress (1.4 mM H2O2) in bovine corneal organ cultures that VIP pre-treatment (0, 10−10, 10−8, and 10−6 M; 15 min), in a VIP concentration-dependent manner, switched the inflammation-causing necrosis to inflammation-neutral apoptosis (showing annexin V-binding, chromatin condensation, and DNA fragmentation) and upheld ATP levels in a VIP antagonist (SN)VIPhyb-sensitive manner, while up-regulated mRNA levels of the anti-apoptotic Bcl-2 and the differentiation marker N-cadherin in a kinase A inhibitor-sensitive manner. As a result, VIP, in a concentration-dependent and VIP antagonist-sensitive manners, promoted long-term CE cell survival. ATP levels, a determining factor in the choice of apoptosis versus necrosis, measured after VIP pre-treatment and 0.5 min post-H2O2 were 39.6 ± 3.3, 50.8 ± 6.2, 60.1 ± 4.8, and 53.6 ± 5.3 pmoles/μg protein (mean ± SEM), respectively ( p  < 0.05, anova ). VIP treatment alone concentration-dependently increased levels of N-cadherin ( Koh et al. 2008 ), the phosphorylated cAMP-responsive-element binding protein and Bcl-2, while10−8 M VIP, in a VIP antagonist (SN)VIPhyb-sensitive manner, increased ATP level by 38% ( p  < 0.02) and decreased glycogen level by 32% ( p  < 0.02). VPAC1 (not VPAC2) receptor was expressed in CE cells. Thus, CE cell VIP/VPAC1 signaling is both anti-inflammatory and protective in the corneal endothelium.  相似文献

Abstract. Seedlings of Phaseolus vulgaris were exposed to solutions containing Cd2+ in the range 0 to 1 molm−3. Ethylene formation started following 3 h of exposure to 10−2, 10−1 and 1 mol m−3 Cd2+, peaked at 18 h and returned to a relatively low rate after 24 h. Cadmium-induced ethylene formation depended on the formation of 1-aminocyclopropane-1-carboxylic acid (ACC). Aminoethoxyvinylglycine (AVG, 0.1 mol m−3) inhibited ACC accumulation and ethylene production during exposure to 0.2 mol m−3 Cd2+.
Activity of soluble and ionically-bound peroxidase increased after 18 h of exposure to Cd2+ concentrations above 10−3 mol m−3 due to an increase in activity of cathodic isoperoxidases. Stimulation of soluble and ionically-bound peroxidase by 0.2 mol m−3 Cd2+ was reduced in the presence of 0.1 mol m−3 AVG.
Accumulation of soluble and insoluble ('ligninlike') phenolics was found in plants exposed to Cd2+ (10−2 mol m−3 or above) in the presence or absence of AVG. Deposition of insoluble (autofluorescing) material occurred in cell walls around vessels and was associated with reduced expansion and water content of leaves.  相似文献

Both reduced illumination and increased turbidity caused a significant reduction in reaction distance of Gobiusculus flavescens . The longest reaction distance, 18.9 cm for larger prey (Calanus finmarchicus) , occurred at a light level of 80 μmol m −2 s −1 compared to 12.9 cm for a smaller prey (Acartia clausi) at 8 μmol m−2 s−1. Above a light saturation level of 10 μmol m−2 s−1, additional light had little influence on reaction distance. In the turbidity experiments, the longest reaction distances were measured at turbidity levels of 10–20 JTU. Prey size influenced reaction distance at all tested light levels. Search time was influenced by prey size only at low illumination. With increasing turbidity, reaction distance to a group of prey was longer than to one prey.  相似文献

Citrus ( Citrus sinensis L. Osbeck) leaf explants completely abscise within 48 h when exposed to saturating amounts of ethylene at 25°C. When 2,5-norbornadiene was added, 2000 μl 1−1 reduced abscission of explants also exposed to 2 μl 1−1 of ethylene to the level of the control, and 8000 μl 1−1 reduced abscission in explants exposed to 10 μl 1−1 of ethylene to the level of the control, but abscission was complete when 1 000 μl 1−1 of ethylene was used in the presence of 8 000 μl 1−1 of 2,5-norbornadiene. When explants were exposed to 2 μl 1−1 of ethylene, 2000 μl 1−1 of 2,5-norbornadiene prevented abscission if applied up to 10 h after exposure to ethylene. After 18 h, applied 2,5-norbornadiene had little effect on abscission at 48 h. A Lineweaver-Burk plot gave a 1/2 maximum value of 0.12 μl 1−1 for ethylene on abscission, 2,5-Norbornadiene gave competitive kinetics with respect to ethylene with a K1 value of approximately 120 μl 1−1 of 2,5-norbornadiene. The presence of 2,5norbornadiene stimulated ethylene production, which progressively increased as the 2,5-norbornadiene concentration was increased from 250 to 8 000 μl 1−1 2,5-Norbornadiene also suppressed the induction of cellulase and polygalacturonase by ethylene. Together, 2,5-norbornadiene and 2,4-dichlorophenoxyacetic acid were more effective than either alone in reducing abscission. 2,5-Norbornadiene also was effective in preventing the reduction of indole-3-acetic acid transport induced by ethylene.  相似文献

A fathead minnow life-cycle exposure to various zinc concentrations demonstrated that the most sensitive indicators of zinc toxicity were egg adhesiveness and fragility, which were significantly affected at 145 μg Zn 1−1 and above, but were not affected at 78 μg Zn 1−1 and below. These effects occurred shortly after the eggs were spawned (during water hardening) and therefore were not related to effects on the parental fish. Hatchability and survival of larvae were significantly reduced, and deformities at hatching were significantly increased at 295 ug Zn 1−1 and above. Acclimated and unacclimated groups of larvae exposed to identical zinc concentrations for 8 weeks after hatch showed only slight differences in sensitivity.  相似文献

Abstract— The content of ATP was measured in neurons from dorsal root ganglia and in cortical and spinal astrocytes with the luciferase method. No ATP could be demonstrated in the cortical cells but in the dorsal root neurons and spinal astrocytes an average amount of 7·9 and 11·9 × 10−15 mol per cell respectively was found. An increase in the external potassium concentration had no effect on the content of ATP in neurons (7·1 × 10−15 mol/cell) but led to a significant ( P < 0·005) decline to 6·4 × 10−15 mol per cell in the astrocytes.  相似文献
