We studied brainstem auditory evoked potentials (BAEPs) in 8 fetal sheep in utero, ranging in gestational age from 105 to 142 days gestation (normal term 147 days). We could not elicit BAEPs prior to 117 days of gestation. After this age rapid maturation was seen, with three discernible peaks observed prior to 120 days and five peaks after 120 days. A significant (P less than 0.05) gestational age related fall in peak latencies and interpeak latencies was observed. The rate of stimulus presentation that could be tolerated without significant changes in wavepeak latency or amplitude also increased with advancing gestational age. In older fetuses (greater than 125 days), where a differentiated electrocorticogram (ECOG) was observed, differences were seen in latency and amplitude of several of the BAEP wavepeaks dependent upon the state. In high voltage ECOG states the amplitudes of wave IV and V were significantly (P less than 0.05) greater than in the low voltage ECOG state and the latencies of wave I, II and V were significantly (P less than 0.05) longer in low as compared to high voltage ECOG state. The BAEP, being obtainable over very short periods of time, appears to provide a useful indice of neural maturation for the sheep fetus in utero.  相似文献

We studied the effects of inhibitors of prostaglandin synthesis on fetal breathing movements on 17 occasions in 11 lambs (gestational age 125-141 days). We gave 12 h infusions of sodium mechlofenamate (8.6-22.2 mg.kg-1) in 13 studies and indomethacin (21.8-38.8 mg.kg-1) in four studies. Results were similar with both agents and did not correlate with drug dosage. There were no changes in fetal arterial blood pressure, pH or blood gas tensions. We assessed fetal breathing movements by measurements of tracheal pressure for a control period of 224 h prior to and 208 h during the infusion of inhibitors of prostaglandin synthesis; their administration caused a marked stimulation of fetal breathing movements judged from the following four variables: (1) incidence of fetal breathing movements increased from 38.4 to 69.2% of the time (P < 0.001); (2) average amplitude of change in tracheal pressure during fetal breathing movements increased from 4.1 to 6.0 torr (P < 0.01); (3) maximal amplitude of change in tracheal pressure during fetal breathing movements increased from 8.8 to 13.4 torr (P < 0.01); and (4) the duration of the longest continuous episode of fetal breathing movements increased from 37 to 229 min (P < 0.05). Two fetuses had electrocorticogram (ECoG) recordings. In control periods, fetal breathing movements occurred only during low voltage, high frequency ECoG activity; however, during infusions of inhibitors of prostaglandin synthesis, fetal breathing movements occurred also during high voltage, low frequency ECoG activity. We conclude that inhibitors of prostaglandin synthesis stimulate fetal breathing movement in fetal sheep. These results suggest that a component of the prostaglandin system is a factor which inhibits breathing movements during fetal life.  相似文献

Extra-dural or cerebroventricular intracranial pressure was measured in 7 unanaesthetized fetal sheep (123-137 days gestation). Basal intracranial pressure was 6.7 +/- 1.7 mmHg, but there were many transient increases of pressure in association with spontaneous changes of amniotic pressure, fetal intrathoracic pressure, and particularly when the fetal nuchal muscles were active. These spontaneous increases of intracranial pressure were often associated with cessation of breathing movements and change of the electrocorticogram from low to high voltage activity. To test whether increased intracranial pressure influenced breathing movements and electrocortical activity, intracranial pressure was raised either by occluding the superior vena cava for 1 min with an implanted extravascular cuff, or by extra-dural injection of 0.3-1.0 ml of 0.9% NaCl. Increasing the intracranial pressure 5-15 mmHg by either method during low voltage electrocortical activity caused cessation of breathing movements, electro-ocular activity, and change of the electrocorticogram from low to high voltage in a significant proportion of trials. We propose that natural fluctuations of intracranial pressure caused by compression of the fetal body or skull, by body movements or by uterine activity, may cause changes in electrocortical activity and breathing movements.  相似文献

The time related hemodynamic responses to forskolin-elicited increases in cAMP were studied in the near-term fetus. Catheters and electrodes were inserted into 6 fetal sheep to measure arterial, venous and thoracic pressures, electrocorticogram, and electrocardiogram. At gestational day 134, experiments were performed to determine the effect of forskolin infusion (400 micrograms/ml at 1.03 ml/min for 5 min) on fetal blood pressure, coronary and cerebral blood flow and resistance. Blood flow measurements were made using 15 microns microspheres labelled with radioactive isotopes during the control period and at 0, 5, 10, 15, and 45 min after forskolin infusion. Forskolin infusion was always initiated during a high-voltage electrocortical epoch and was given twice in each animal. In each case, forskolin caused electrocortical activity to change from high-voltage state to an intermediate voltage state. Blood pressure fell significantly by the end of the infusion period and returned to control levels 10 min later. Fetal heart rate and coronary blood flow were immediately elevated by forskolin (P less than 0.01) whereas cerebral blood flow did not increase until 5 min later (P less than 0.01). Cerebral blood flow was still elevated (P less than 0.05) 45 min after the end of forskolin infusion, whereas coronary blood flow had returned to control levels. Both cerebral and coronary vascular resistance fell significantly in response to forskolin infusion (P less than 0.01). This effect lasted at least 15 min and had returned to control levels 45 min after forskolin had been terminated.  相似文献

为了探讨将胎儿自主神经发育及其功能状况作为判断胎儿宫内发育迟缓(IUGR)的可行性,采用酒精灌胃加限食法建立IUGR动物模型,利用自回归(AR)模型法对IUGR胎鼠心率变异性(HRV)进行功率谱分析。结果表明,无论是低频功率、高频功率还是总功率,实验组均明显高于对照组,但标化后的高频功率实验组则明显低于对照组,标化后的低频功率实验组仍明显高于对照组。提示,利用FHRV谱分析评价胎儿自主神经发育及其功能状况,进而对IUGR进行产前诊断的方法是可行的。  相似文献

The objective of this study was to examine the effect of subchronic administration of an aqueous extract of smokeless tobacco (ST) on the development of the CD-1 mouse fetus. Mice were administered ST for approximately 5 weeks: for 2 weeks prior to breeding, during breeding, and during gestational days 0 to 17. Thus the initial peak nicotine levels occurred prior to breeding and not during the critical periods of gestation. Two ST dosages were administered by gastric intubation three times daily: ST/D-1, equivalent to a dose of 12 mg nicotine/kg of body weight, and ST/D-2, equivalent to 20 mg nicotine/kg body weight. Maternal plasma nicotine levels were determined 30 min after the second intubation during the pretreatment and gestational phases of treatment. At these ST dosages, the weight gain of ST-treated dams was not significantly affected in comparison to treated controls. The mean maternal plasma nicotine level for the low-dosage group was 363 ng/ml, and 481 ng/ml for the high-dosage group, with maternal lethality observed at 9.6% and 28.2%, respectively. No significant differences were seen between control and ST/D-1 maternal and/or fetal values, except for placental weights which were heaviest in the ST group (P less than 0.05). Several differences were noted between the ST/D-2 group and controls: fetal weights were reduced by 5.4% (P less than 0.05); decreased ossification was seen in femur measurements and in nine of ten characteristics measured (P less than 0.05); the frequency of resorptions (7.6%) was almost doubled (controls 4.2%); and the frequency of deaths and malformations was not affected. Under these experimental conditions, the low dose produced a negligible effect on the CD-1 mouse fetus and the dam. The high dose demonstrated growth retardation (P less than 0.05), increased embryotoxicity, and a significant decrease in ossification (P less than 0.05).  相似文献

Somatosensory evoked potentials were recorded in utero from 13 chronically instrumented fetal lambs (97 to 148 days of gestation) following electrical stimulation of the upper lip or upper limb. Several clear and reproducible peaks were observed. Following upper lip stimulation, peaks were seen with mean peak latencies of 9, 13.2, 17.8, 21.3, 33.8 and 206 ms at a gestational age of 125 days. Similar peaks, but of slightly later mean latencies, were seen following limb stimulation. These peaks demonstrated significant gestational age related falls in peak latencies (P less than 0.05). Several of the mid to late latency peaks, notably those occurring at 21.3, 33.8 and 206 ms, demonstrated changes (P less than 0.05) in both latency (longer in low voltage) and amplitude (reduced in low voltage) dependent on electrocorticographic state. Rate of stimulus presentation also had a significant effect on both amplitude and latency of several peaks (P less than 0.05) with this effect lessening with advancing gestational age. Evoked potentials can thus be successfully obtained from chronically instrumented fetal lambs and provide a useful indice for studies of neural maturation.  相似文献

Fetuses of 12 near-term sheep were prepared for microsphere determination of cerebral blood flow. Experiments were performed 5 days postsurgery. The regional blood flows were measured in successive high (HV), low (LV) and high voltage electrocorticographic states. Comparisons were made between the observations made in the LV and averaged flanking HV cycles. Total cerebral blood flow was 95 +/- 8, 119 +/- 11 and 100 +/- 9 ml/min/100 g in HV, LV and HV, respectively. Low voltage electrocortical activity increased average cerebral blood flow by 22% (P less than 0.01). Significant changes were seen in all regions except the occipital cortex. The maximum change was observed in the thalamus in which the flows were 152 +/- 23, 243 +/- 35 and 138 +/- 20 ml/min/per 100 g tissue, respectively. The increase was 68% (P less than 0.001). The percent changes seen in the cerebrum are as follows: Frontal grey + 18%, frontal white + 22%, parietal white + 22%, temporal + 18%. A + 17% change was seen in the cord (P less than 0.03). It is concluded that in low voltage electrocortical activity all of the brain, except the occipital region, shows an increase in cerebral blood flow. This is probably secondary to a variance in cerebral activity. This preparation may be useful in localizing function in the fetal brain.  相似文献

Local cerebral blood flow in four near term fetal sheep was evaluated continuously before and after natural alternations in fetal behavioral state. Measurements were made in fetuses several days following an aseptic surgery to place electrodes for behavioral state recordings as well as heated and reference thermojunctions in cortical and subcortical tissue. These thermojunctions were used to qualitatively assess local cerebral blood flow. The time of transition between rapid eye movement sleep (REMS) and non-rapid eye movement sleep (NREMS) was based on visual inspection of strip chart recordings of electrocortical, electroocular, and neck electromyographic activity and application of published criteria for their assessment. To confirm that transition had occurred, the amplitude of the spectrum of the electrocorticogram in one-third octave bands centered around 1 Hz and 20 Hz was measured before and after the transition point. Mean cerebral blood flow rose significantly by 24 s (P less than 0.05) after the transition from NREMS to REMS and fell by 12 s after the transition from REMS to NREMS (P less than 0.05).  相似文献

Local blood flow was measured with radioactive microspheres in 9 near-term ewes 2 min into successive high and low voltage electrocortical activity states. In an additional 8 animals the umbilical blood flow was measured using an electromagnetic flow-probe on the common umbilical vein. The microsphere data indicated that the blood flow during low and high voltage electrocortical activity was 185 +/- 22 ml/min per kg of fetus (SEM) and 165 +/- 22 ml/min per kg of fetus (P less than 0.01) respectively. Using the electromagnetic flowprobe the average flow during low and high voltage electrocortical activity was 203 +/- 14 ml/min per kg of fetus and 196 +/- 13 ml/min per kg of fetus (P less than 0.05) respectively. We observed that the decrease in the umbilical blood flow preceded the change from low to high voltage electrocortical activity by approximately 1 min. In that time the flow is significantly lower than it was during the preceding measurements taken during the low voltage electrocortical activity periods. This depression was still significantly lower at 3 min into the high voltage electrocortical activity whereas at 5 min into the high voltage state it was elevated to near average values. We conclude that the umbilical blood flow, on the average, is lower in high voltage states than it is in low voltage states and that this change precedes the switch from low to high voltage electrocortical activity.  相似文献

Adult 15 female Japanese monkeys showing regular menstrual cycles were subjected to the daily blood sampling for the measurement of estradiol (E2), progesterone (P) and biological LH in the mating season. Monkeys were maintained under controlled conditions in a standardized environment. Of the 35 cycles observed, 18 (51.4%) were estimated as anovulatory cycles and 17 (48.6%) were ovulatory cycles. The anovulatory cycles were classified into three types according to the peak level of E2 (Type I: E2 less than 50 pg/ml 3 cycles, Type II: E2 less than 170 pg/ml 7 cycles, Type III: E2 greater than 170 pg/ml 8 cycles). The ovulatory cycles were classified into two Types according to the peak level of P (Type IV: P less than 5.0 ng/ml 5 cycles, Tyep V: P greater than 5.0 ng/ml 12 cycles). The menstrual cycle was 27.5 +/- 7.8 days. The differences between mid cycle LH surge and P level in Type IV and in Type V were statistically significant. It was revealed that female Japanese monkeys kept under controlled condition in the mating season showed high incidence of various types of anovulatory cycles and that the ovulatory cycles with low P elevation in the mid luteal phase showed low LH and P secretions on the mid cycle date.  相似文献

How nonhuman species perceive the world is a biological question of fundamental importance, and has major significance for establishing the validity and possible limitations of animal models of human sensory function and perception. Studies in comparative hearing have revealed that almost all animals, including monkeys, are worse than humans at discriminating tone frequencies. Less is known, however, about comparative differences in discriminating more spectrally complex sounds. We compared the capacity of macaques and humans to discriminate complex sound patterns by measuring spectral-contrast sensitivity using stimuli having sine-modulated power spectra, analogous to sine-wave gratings used in visual studies. We found that the auditory system of the macaque is far less sensitive than the human system over the sine-profile frequency range tested (0.5-2.0 cycles/octave). These results indicate that rhesus macaques hear at least some spectrally complex sounds with less fidelity than do humans, and demonstrate large differences in primate species' abilities to process low-resolution spectral patterns. These results cannot be accounted for by traditional, narrowband peripheral filter models of spectral analysis, but instead, imply the involvement of a central, frequency integration process that may differ significantly across species.  相似文献

The role of cortisol as a factor controlling the deposition of glycogen in the pig fetus was examined by infusing either a low dose (1 mg/day) or a high dose (3 mg/day) of cortisol into chronically-catheterized hypophysectomized fetal pigs for five days beginning on day 100-104 of gestation. After infusion, liver glycogen was significantly higher (P less than 0.05) and lung glycogen significantly lower (P less than 0.05) than in uninfused hypophysectomized litter mates although concentrations were significantly different from intact litter mates (P less than 0.05). Although skeletal and cardiac muscle content increased after infusion this difference was not significant. Changes in tissue glycogen content were similar for both the low and high rates of infusion. These observations indicate that exogenous cortisol alone is able to stimulate liver glycogen deposition and reverse the effect of hypophysectomy. Although other factors may be necessary for maximal response this suggests that cortisol is an important stimulant for liver glycogen deposition in the fetal pig. The effect of cortisol on muscle glycogen was equivocal suggesting that other hormones may play a more important role in this tissue.  相似文献

Data collected over a 10-yr period on 1,778 service intervals in 1,111 Arsi (Bos indicus), cows at the Ethiopian Ministry of Agriculture ranch were analyzed. Conception rate to first service was 48%, cows calving from June to February being more likely to conceive at first oestrus. Overall interval between services in animals returning for insemination averaged 53 +/- 40 (SD), range 6-262 d. There were (6%) "short" cycles of less than 17 d, mostly during the first (69%) and second (30%) intervals (P less than 0.01). A quarter of service periods (24%) were within the normal length of 18-24 d, 52% were its multiples while 18% were abnormal and probably indicative of embryonic mortality. First oestrus cycle length was affected by year (P less than 0.01) and season of calving (P less than 0.05). Second oestrus cycle length was influenced by season of calving and first oestrus onset (P less than 0.05). It is concluded that the low frequency of normal cycles is to a large extent due to heats which remained undetected by visual observation in Arsi cows under artificial insemination programmes.  相似文献

The experimental objective was to evaluate how continuous infusion of oxytocin during the anticipated period of luteolysis in cattle would influence secretion of progesterone, oestradiol and 13,14-dihydro-15-keto-prostaglandin F-2 alpha (PGFM). In Exp. I, 6 non-lactating Holstein cows were infused with saline or oxytocin (20 IU/h, i.v.) from Day 13 to Day 20 of an oestrous cycle in a cross-over experimental design (Day 0 = oestrus). During saline cycles, concentrations of progesterone decreased from 11.0 +/- 2.0 ng/ml on Day 14 to 2.0 +/- 1.3 ng/ml on Day 23; however, during oxytocin cycles, luteolysis was delayed and progesterone secretion remained near 11 ng/ml until after Day 22 (P less than 0.05). Interoestrous interval was 1.6 days longer in oxytocin than in saline cycles (P = 0.07). Baseline PGFM and amplitude and frequency of PGFM peaks in blood samples collected hourly on Day 18 did not differ between saline and oxytocin cycles. In Exp. II, 7 non-lactating Holstein cows were infused with saline or oxytocin from Day 13 to Day 25 after oestrus in a cross-over experimental design. Secretion of progesterone decreased from 6.8 +/- 0.7 ng/ml on Day 16 to less than 2 ng/ml on Day 22 of saline cycles; however, during oxytocin cycles, luteolysis did not occur until after Day 25 (P less than 0.05). Interoestrous interval was 5.9 days longer for oxytocin than for saline cycles (P less than 0.05). In blood samples taken every 2 h from Day 17 to Day 23, PGFM peak amplitude was higher (P less than 0.05) in saline (142.1 +/- 25.1 pg/ml) than in oxytocin cycles (109.8 +/- 15.2 pg/ml). Nevertheless, pulsatile secretion of PGFM was detected during 6 of 7 oxytocin cycles. In both experiments, the anticipated rise in serum oestradiol concentrations before oestrus, around Days 18-20, was observed during saline cycles, but during oxytocin cycles, concentrations of oestradiol remained at basal levels until after oxytocin infusion was discontinued. We concluded that continuous infusion of oxytocin caused extended oestrous cycles, prolonged the secretion of progesterone, and reduced the amplitude of PGFM pulses. Moreover, when oxytocin was infused, pulsatile secretion of PGFM was not abolished, but oestrogen secretion did not increase until oxytocin infusion stopped.  相似文献

The effects of pharmacological doses of glucagon (0.5 micrograms/kg/min) were studied in 8 chronically-catheterised fetal sheep. These doses of glucagon raised fetal blood glucose, and caused a small fall in fetal arterial PO2 (P less than 0.05). Arterial PCO2 rose (P less than 0.05) and pH fell (P less than 0.05) while plasma osmolality increased (P less than 0.01). There were no effects of glucagon on fetal renal function but in the fetus, like the adult, i.v. glucagon caused increases in heart rate (P less than 0.01) which were not associated with changes in arterial pressure.  相似文献

We examined expiratory muscle fatigue during expiratory resistive loading in 11 normal subjects. Subjects breathed against expiratory resistances at their own breathing frequency and tidal volume until exhaustion or for 60 min. Respiratory muscle strength was assessed from both the maximum static expiratory and inspiratory mouth pressures (PEmax and PImax). At the lowest resistance, PEmax and PImax measured after completion of the expiratory loaded breathing were not different from control values. With higher resistance, both PEmax and PImax were decreased (P less than 0.05), and the decrease lasted for greater than or equal to 60 min. The electromyogram high-to-low frequency power ratio for the rectus abdominis muscle decreased progressively during loading (P less than 0.01), but the integrated EMG activity did not change during recovery. Transdiaphragmatic pressure during loading was increased 3.6-fold compared with control (P less than 0.05). These findings suggest that expiratory resistive loaded breathing induces muscle fatigue in both expiratory and inspiratory muscles. Fatigue of the expiratory muscles can be attributed directly to the high work load and that of the inspiratory muscles may be related to increased work due to shortened inspiratory time.  相似文献

Adenosine is known to inhibit nonshivering thermogenesis in adult brown fat. These experiments were undertaken to test whether fetal adenosine, normally present in high concentrations, suppresses lipolysis in utero and then falls after birth, permitting thermogenesis to begin. To test this hypothesis, we measured fetal plasma adenosine concentration [ADO] using high-performance liquid chromatography in 11 fetal sheep at 135-140 days gestation during simulated birth. During an initial control period, fetal [ADO] averaged 1.9 +/- 0.3 microM, about four times maternal [ADO] (0.4 +/- 0.1 microM, P less than 0.001). The fetus was then cooled by circulating cold water through a plastic coil encircled about the fetal torso. One hour later, when fetal core temperature had decreased 2.3 degrees C, fetal [ADO] averaged 2.8 +/- 0.5 microM, a 50% increase (P less than 0.05), while thermogenesis remained inactive. Next the fetal lungs were ventilated with O2 to raise arterial Po2 to greater than or equal to 150 Torr. Fetal [ADO] decreased only slightly, and thermogenic responses were modest. Finally, the umbilical cord was occluded. Fetal [ADO] decreased rapidly and 60 min later averaged 1.1 +/- 0.2 microM, 40% below initial control (P less than 0.05) and 57% below the previous period (P less than 0.001). As [ADO] fell, strong thermogenic responses became apparent, as indicated by seven- to eightfold increases in plasma glycerol (P less than 0.001) and a doubling in fetal O2 consumption (P less than 0.001). To investigate the hypothesis that the rate of fatigue development is not influenced by the absolute duration of contraction (train duration) and relaxation (off-phase of duty cycle) at constant duty cycle, strips of the diaphragm from 36 male adult rats (mean +/- SD wt 152 +/- 21 g) were stimulated directly for periods of 180, 250, and 320 ms at a constant duty cycle of 50%. The frequency of stimulation was adjusted to produce 40% of maximal tetanic tension at supramaximal voltages. After 30 min of stimulation, analysis of twitch characteristics between control and experimental groups indicated a prolongation of contraction time of 9% (P less than 0.05), an increase in relaxation time of 75% (P less than 0.05), and a decrease in twitch tension by 78% (P less than 0.05). Similarly, reductions (P less than 0.05) in isometric force output at high stimulation frequency (100 Hz) of 58% and at low frequency (20 Hz) of 67% were also noted. These changes were accompanied by an approximately 60% reduction in the maximal velocity of shortening. No difference was observed for any of the mechanical measures between experimental conditions. After 30-min stimulation, decreases of between 43 and 46% were noted for ATP (P less than 0.05) and increases of between three- and fourfold noted for IMP (P less than 0.05). No changes were found for either ADP or AMP. Total adenine nucleotide concentrations declined (P less than 0.05) an average of 24%. As with the mechanical data, no differences were found between the different stimulation conditions. It is concluded that for the conditions studied, fatigue mechanisms become manifest early in the stimulation period and are only minimally altered by the duration of specific contractions provided the relaxation period is of equal duration.  相似文献

The present study was designed to investigate the effects of clothing skin pressures exerted by two different types of brassieres (a conventional higher skin-pressured brassiere and a newly devised low skin-pressured brassiere) on the autonomic nervous system (ANS) activity. Six healthy young women (22.8 +/- 1.4 yrs.) with regular menstrual cycles participated in this study. The ANS activities were assessed by means of heart rate variability power spectral analysis. The skin pressures exerted by the brassieres were measured with an air-pack type contact surface pressure sensor at five different points. The total amount of clothing pressure, and the pressures at the center and the side regions of the brassieres were significantly greater in the high than in the low skin-pressured brassiere (Total 9816.1 +/- 269.0 vs. 6436.8 +/- 252.4 Pa, P < 0.01; Center 2212.1 +/- 336.3 vs. 353.8 +/- 85.8 Pa, P < 0.01; Side 2556.8 +/- 316.1 vs. 1747.2 +/- 199.2 Pa, P < 0.05). Concerning the ANS activity, the Total power, and the very low frequency (VLF) and the high frequency (HF) components were significantly decreased in the high skin-pressured brassiere than those in the low skin-pressured brassiere (Total 531.6 +/- 57.3 vs. 770.5 +/- 54.2 ms2, P < 0.01; VLF 60.7 +/- 14.6 vs. 179.2 +/- 38.1 ms2, P < 0.05; HF 209.5 +/- 33.2 vs. 283.2 +/- 61.5 ms2, P < 0.01). Our data indicate that the higher clothing pressures exerted by a conventional brassiere have a significant negative impact on the ANS activity, which is predominantly attributable to the significant decrease in the parasympathetic as well as the thermoregulatory sympathetic nerve activities. Since the ANS activity plays an important role in modulating the internal environment in the human body, excess clothing pressures caused by constricting types of foundation garments on the body would consequently undermine women's health.  相似文献
