Callinan A. P. L. 1979. The ecology of the free-living stages of Trichostrongylus vitrinus. International Journal for Parasitology9: 133–136. The ecology of the free-living stages of Trichostrongylus vitrinus was studied in western Victoria in 1975–1976. Where development of eggs (E1–E2) did occur, preinfective larvae (L1–L2) were recovered within 0–8 days, infective larvae (L3) in faeces within 10–39 days and on L3 herbage and soil within 4–26 days. The mean development time to L3 on herbage and soil was 12.5 ± 4.8 days and the mean development time to maximum yield of these was 37.9 ± 9.7 days. The mean percentage of L3 on herbage and soil actually on herbage was 70.8 ± 6.5 %. L3 persisted in faeces for up to 106 days and on herbage and soil for up to 208 days. Yields of L3 on herbage and soil varied from 0 to 5.05 % of the number of eggs deposited. Yields were highest from those eggs deposited in autumn-winter. No L3 were observed to survive over summer.  相似文献

In Nippostrongylus brasiliensis infection in rats there is evidence that antibodies ‘damage’ the worms rendering them susceptible to other components of the host immune response. Some of the experiments from which this evidence was obtained were repeated in Trichostrongylus colubri-formis infections in guinea-pigs and sheep.Lesions similar to those described in ‘damaged’ N. brasiliensis were present in T. colubriformis, but unlike damaged N. brasiliensis, damaged T. colubriformis were not rapidly expelled from the intestine of either normal or adoptively immunized hosts. Other differences between the two infections included the failure of serum from T. colubriformis-immune donors either to regularly transfer immunity against infection to non-immune recipients or to endow additional immunity on recipients of lymphoid cells from immune donors.It is suggested that the results might reflect differences between the manner in which guinea-pigs and rats achieve immune expulsion of gastro-intestinal nematode parasites.  相似文献

At a temperature of 24°C, freshly hatched second stage larvae (L2) of Rotylenchulus reniformis take 2 weeks to complete moulting and become either adult males or immature females. Measurements of different nematode stages were made using a computer-interfaced digitizer (CID). This proved to be more rapid and reproducible than conventional techniques.Moulting and development were followed in individual living specimens viewed under Nomarski optics and oil immersion. The mean loss in body volume of these specimens from L2 to immature female is about 17% compared with about 19% for the mean loss in body volume from L2 to male. The mean volumes for a total of 70 randomly selected living specimens measured are 95·6 pi for L2s, 72·9 pl for immature females and 74·2 pl for males. Mean losses in body volume during moulting calculated from these randomly selected stages were slightly higher than those observed for individuals, being about 24% for the immature female and about 22% for the male.  相似文献

Larval stages of Oesophagostomum radiatum grown in vitro and adults grown in vivo were incubated in complex media or in a simple salt solution containing radioactive glucose. Glucose disappearance and end product accumulation of third-stage larvae in a simple salt solution indicated that they excreted CO2 and acetic, propionic, and lactic acids. Larvae in third molt, fourth stage, and adults all excreted CO2, acetic, propionic, and lactic acids at twice the rate of third-stage larvae plus an additional product, methylbutyric acid. Carbon dioxide arose primarily from the 3 or 4 carbons of glucose. An anaerobic atmosphere (95% N2:5% CO2) had no apparent effect on metabolism. When incubation was done in complex media, isobutyric and 3-methylbutyric acids were seen as major excretion products (10 and 24%, respectively). However, these acids were quantitatively minor when incubations took place in simple salts-glucose medium (1 and 0–3%, respectively).  相似文献

An improved technique for measuring the water content of nematodes is described using an electronic interferometer. Changes in phase of a laser beam passing through a known pathlength of the nematode have been used to measure the refractive index and hence the water content and relative volume of the animal. Third-stage larvae of Nippostrongylus brasiliensis and Haemonchus contortus which possess an excretory ampulla, differed from second-stage larvae lacking this ampulla in requiring a greater fall in the osmolarity of artificial tap water before there was a significant increase in their water content. Increases in the pulsation frequency of the ampulla also occurred in less hypotonic solutions than those required to increase the water content of the third-stage larvae. The ampulla pulsation frequency of third-stage larvae of N. brasiliensis increased after locomotor activity in hypotonic tap water and locomotory wave frequency of third-stage larvae of N. brasiliensis was independent of the extent of hypotonicity for a range of solutions that reduced wave propogation by its second-stage larva. The results suggest that the ampulla is an adaptation to hypotonic conditions favouring a volume homeostasis that is required for optimal locomotor activity of the third-stage infective larvae of these nematodes.  相似文献

Survival of Nippostrongylus brasiliensis larvae after freezing over liquid nitrogen. International Journal for Parasitology4: 173–176. Third stage larvae of N. brasiliensis were frozen over liquid nitrogen and after storage for 7 days were thawed rapidly and inoculated subcutaneously into rats. When ensheathed larvae were used, none survived freezing as judged by motility and infectivity trials. Separate vials of exsheathed larvae survived freezing in proportions ranging from 10 to 64 per cent. Female worms, derived from frozen exsheathed larvae, had a normal complement of eggs in the uterus and both male and female worms had a normal histological appearance. Exsheathed larvae frozen in the presence of 10 per cent dimethylsulphoxide had the same survival rate as those frozen without the addition of cryoprotectant. The addition of 10 per cent glycerol adversely effected the survival of frozen exsheathed larvae.  相似文献

Bioassays were conducted to determine the susceptibility of egg masses of Mamestra brassicae and Spodoptera littoralis to different spore doses of Paecilomyces fumoso-roseus and Nomuraea rileyi at 20° and 25°C. P. fumoso-roseus was highly virulent against eggs, whereas N. rileyi provoked only a deferred mortality of larvae hatched from treated eggs. Nevertheless, larval mortality of S. littoralis caused by N. rileyi at 25°C was more effective after first-instar larval contamination than after egg mass treatment. The duration of the egg stage could explain differences of susceptibility between the two noctuids at 25°C. Scanning electron microscopical observations suggested two ways of contamination of newly hatched larvae. First, fungal germinations on the chorion surface suggested that newly hatched larvae might be infected by penetration of the egg integument before hatching. Second, conidia on the egg cuticle could be an entomopathogenic inoculum for newly emerging larvae which fed upon chorions. Results showed that pathogenicity of Hyphomycetes to noctuid eggs might be a promising area of investigation for biological control.  相似文献

Dobson C. & Owen M. E. 1978. Effect of host sex on passive immunity in mice infected with Nematospiroides dubius. International Journal for Parasitology8: 359–364. Female C3H but not Quackenbush (Q) mice harboured fewer Nematospiroides dubius than male C3H and Q mice. Both strains lost worms 21 days after infection. C3H and Q mice became progressively immune to infection following 4 sequential doses of N. dubius larvae and showed a sex resistance to infection. Hypothymic nu/nu CBA Balb/c mice did not show these effects on N. dubius infection. The reciprocal transfer of male and female immune mesenteric lymph node cells (IMLNC) to syngeneic male and female recipients showed that the female environment enhanced the protective qualities of both male and female IMLNC but the male environment suppressed these effects. Gonadectomized male and female recipients of male and female IMLNC had levels of infection similar to the entire female recipients. Serum from immune female donor mice protected both male and female recipients better than immune serum from male donors, but female mice in each treatment group were better protected than male mice. Immune serum transferred greater levels of protection then IMLNC to recipient mice against N. dubius infections. These data are consistent with the conclusion that the male environment suppresses lymphocyte activity and the production of protective antibodies and additionally may depress the effectiveness of sensitized lymphocytes and antibodies in ejecting N. dubius. On the other hand the female environment does not appear to adversely affect the mobilization of the protective immune response and may enhance immune effector mechanisms in protecting mice against N. dubius infections.  相似文献

Callinan A. P. L. and Arundel J. H. 1982. Population dynamics of the parasitic stages of Ostertagia spp. in sheep. International Journal for Parasitology12: 531–535. The development and survival of continuing infections of Ostertagia spp. in weaner sheep were studied in order to develop a general model of the parasitic stages of the life cycle of these sheep nematodes. After 10 days, 13.8% of infective larvae (L3) given at the rate of 1 dose of 1000 L3 twice per week (group 1) and 20.8% of L3 given at the rate of 10,000 L3 twice per week (group 2) were recovered in the first of the serial nematode counts. In the final counts at 137 days, 7.7 and 0.7% were recovered in these groups. The build up and maintenance of nematode populations was regulated and related to the level of infection. A model in which the death rate of the parasitic stages was a function of the time of exposure to infection and rate of infection was used to describe the serial total nematode counts. During the experiment there was no noticeable trend in numbers of fourth stage larvae (L4) in nematode counts, the size of adult nematodes, nematode egg counts (EPG) and egg output per female nematode (EPF). After 112 days, liveweight gains were significantly reduced in group 2 only, but increases in wool lengths were significantly reduced in both groups.  相似文献

Cultivation of fourth stage Brugia pahangi and B. malayi larvae from infective larvae (stage 3) were obtained in culture medium RPMI 1640 supplemented with 10% human AB serum and an LCC-MK2 rhesus monkey kidney continuous cell line feeder layer. This culture system kept larvae alive in excess of 7 weeks, and served as a source for collection of the worms' secretory, excretory, and moulting antigens.  相似文献

High (H) and low (L) immune responder mice (Sitepu & Dobson, 1982) were immunized with 5, 10, 20, 40 or 80 Nematospiroides dubius larvae, drenched 3 weeks later with anthelmintic then challenged with 100 larvae. Size of the inoculum of larvae correlated positively with the numbers of N. dubius eggs passed in the faeces of all these mice after the immunizing dose, and the size of the inoculum of immunizing larvae was negatively correlated with faecal epg, worm numbers and lengths of worms recovered from H but not L mice after challenge infection with 100 larvae. Increasing the number of N. dubius larvae in the immunizing inoculum enhanced the immunization of H mice, whereas the L mice were progressively immunosuppressed by increasing numbers of larvae.  相似文献

Brachymeria lasus and Pachycrepoideus vindemiae failed to develop in vitro on sterol-free artificial media, and dietary acetate and squalene failed to maintain and/or support growth. The sterols, cholesterol, cholestanol, β-sitosterol, 7-dehydrocholesterol, and cholesterol linoleate were all utilized and maintained larvae of both species. Larval survival and development rate were greatest with cholesterol followed by cholestanol, β-sitosterol and 7-dehydrocholesterol. Although cholesterol linoleate maintained larvae little growth occurred and mortality was high. Cholestanol followed by β-sitosterol and 7-dehydrocholesterol displayed partial cholesterol sparing activity. Cholesterol linoleate had little effect on larval growth when fed with suboptimal levels of cholesterol or cholestanol. Both species contained 5 to 10% of the total body lipids as free sterol with traces of sterol ester. The major free sterol appears to be cholesterol.  相似文献

International Journal for Parasitology15: 353–359. Similar proportions of the inoculum of Nematospiroides dubius larvae reached sexual maturity by 14 days after administration of 50–400 larvae but more adult worms had been expelled by day 63 after infection from those mice infected with 50 vs 400 larvae. There was a significant correlation between time and worm expulsion for all inoculum size groups except for mice given 400 larvae.In mice reinfected with 100 larvae, after termination of primary infections derived from 10 through 400 larvae, more worms from the challenging dose were recovered from mice given greater compared with those given smaller numbers of larvae at primary infection. The N. dubius population size after challenge infection was correlated positively both with number of larvae administered as the primary infection and with the resultant population size during that infection. The serum anti-N. dubius antibody titres after reinfection were higher in mice given 400 compared with those given fewer larvae at primary infection, and the fecundity and female to male sex ratio of the N. dubius populations decreased in proportion to these antibody titres.Protective immunity against challenge N. dubius infection, in mice which had been drenched free of adult worms established from 400 larvae for 5 down to 1 weeks before reinfection, increased from 45% (1 week) to 80% (5 weeks). There was a negative correlation between the population size of N. dubius during challenge infection and the duration between anthelmintic treatment and challenge infection.  相似文献

Dobson C. and Cayzer C. J. R. 1982. Immunosuppressive activity in serum from mice infected with Nematospiroides dubius following passive serum transfer. International Journal for Parasitology12: 561–566. Anti-N. dubius antibody titres increased with time after primary and secondary infections with 100 larvae in mice and 4 days after anthelmintic termination of a 28 day infection. Mice injected with serum from infected mice harboured fewer, smaller worms with reduced fecundities compared with control mice; the difference was greater with serum from donors given two compared with those given one infection.Mice injected with serum from donors infected for 14 days had fewer N. dubius than recipients of serum from mice infected for 28 days. Serum taken from mice 4 days after termination of a primary infection of 28 days duration was more protective when passively transferred to mice than serum from mice infected for 28 days. Serum from mice infected with 50 N. dubius larvae was more protective than serum, with a higher anti-N. dubius antibody titre, from, mice infected with 400 larvae. These observations are discussed in relation to immunosuppressive activities in the donated serum and associated with N. dubius.  相似文献

A polydnavirus, Cotesia plutellae bracovirus (CpBV), possesses a segmented and dispersed genome that is located on chromosome(s) of its symbiotic endoparasitic wasp, C. plutellae. When the host wasp parasitizes larvae of the diamondback moth, Plutella xylostella, at least 27 viral genome segments are delivered to the parasitized host along with the wasp egg. The parasitized P. xylostella exhibits significant immunosuppression and a prolonged larval development. Parasitized larvae take about 2 days longer than nonparasitized larvae to develop until the wandering stage of the final larval instar, and die after egress of the full grown wasp larvae. Developmental analysis using juvenile hormone and ecdysteroid analogs suggests that altering endocrine signals could induce the retardation of larval developmental rate in P. xylostella. In this study we used a transient expression technique to micro-inject individual CpBV genome segments, and tested their ability to induce delayed larval development of P. xylostella. We demonstrated that a CpBV segment was able to express its own encoded genes when it was injected into nonparasitized larvae, in which the expression patterns of the segment genes were similar to those in the larvae parasitized by C. plutellae. Twenty three CpBV genome segments were individually cloned and injected into the second instar larvae of P. xylostella and their effects assessed by measuring the time taken for host development to the cocooning stage. Three CpBV genome segments markedly interfered with the host larval development. When the putative genes of these segments were analyzed, it was found that they did not share any common genes. Among these segments able to delay host development, segment S27 was predicted to encode seven protein tyrosine phosphatases (CpBV-PTPs), some of which were mutated by insertional inactivation with transposons, while other encoded gene expressions were unaffected. The mutant segments were unable to induce prolonged larval development of P. xylostella. These results suggest that CpBV can induce prolonged larval development of P. xylostella, and that at least some CpBV-PTPs may contribute to the parasitic role probably by altering titers of developmental hormones.  相似文献

The immune response of mice to the nematode Trichinella spiral's was markedly altered when the infection was superimposed upon an existing infection with Nematospiroides dubius. The expulsion of a primary infection of T. spiralis was delayed in such mice, and the worms persisted for at least 4 weeks longer than they did in control mice. The degree to which expulsion was suppressed was related to the number of N. dubius present. It would appear that both adult and larval stages of N. dubius can exert a suppressive effect, since the expulsion of T. spiralis was affected within days of a super-imposed (i.e., larval) N. dubius infection. When adult N. dubius were removed from mice 4 days before infection with T. spiralis, the mice expelled the latter parasite within the normal time, indicating that recovery from the suppressive effects of concurrent infection occurred rapidly. Concurrent infection with N. dubius appeared to affect both the afferent and efferent arms of the immune response to T. spiralis, since sensitization by, and memory of, prior infection were impaired and the expression of acquired immunity was inferior to that of controls.  相似文献

The Angoumois grain moth, Sitotroga cerealella, grew well in an artificial pellet containing about 11 per cent whole wheat germ as its only crude adjuvant. A study of the insect's lipid requirements showed that while dietary wheat germ oil slightly enhanced the larval growth rate it was not required to complete metamorphosis. Normal development without dietary methyl linoleate further confirmed the insect's capacity to mature into an aerial adult without ingesting polyunsaturated fatty acids. A study of the insect's sterol requirements revealed that while a dietary sterol was essential, all free sterols (6) and sterol esters (3) tested permitted equivalent growth and development. Results also showed that larvae did not utilize the sterol relatives, squalene, lanosterol, and cholestane. It was concluded that Sitotroga had a non-specific sterol requirement. Factors in the lipid-free portion of wheat germ, which probably include vitamins, minerals, and amino acids, proved to be essential for normal growth and development, but their exact nature has yet to be determined. The significance of the results in relation to other similar investigations on grain- and plant-feeding insects is discussed.  相似文献

Early stages (neonate to 7- or 8-day-old larvae) of Spodoptera exigua and Pseudaletia unipuncta were exposed to the entomogenous nematode, Steinernema feltiae, at concentrations of 0, 10, 25, 60, 100, or 200 nematodes per larva. Larvae of both species were susceptible to nematode infections. However, neonate larvae of S. exigua were significantly less susceptible to nematode infection than 3- or 8-day-old larvae at or above 50 nematodes per larva. Mortalities of neonate larvae exposed to 50 or more nematodes ranged from 68 to 74% while mortalities of 3- and 8-day-old larvae ranged from 91 to 100%. The results with P. unipuncta showed similar trends as described for S. exigua, albeit at a lower mortality level and usually with no statistical differences. Mortalities of neonate larvae exposed to 50 or more nematodes ranged from 34 to 44% while mortalities of 7-day-old larvae ranged from 32 to 91%.  相似文献

The potentials of Haplophyllum tuberculatum and Plectranthus cylindraceus oils to control Meloidogyne javanica were investigated in vitro and in a greenhouse. A mixture of Haplophyllum and Plectranthus oils (1:1) was highly toxic to M. javanica in vitro, as it killed all nematode juveniles and inhibited hatching of eggs at 12.5 μg/ml concentration after 24 h exposure time, as did carbofuran at the same concentration. In the green-house, tomatoes grown in soil treated with a combination (1:1) of the two oils developed fewer root galls than those grown in soil treated with higher doses of either oil. The oil mixture, at 2.5 and 5.0 μg/ml of soil, was not phytotoxic to tomato plants as evident from the appearance and height of plants after 12 weeks exposure time, compared to treatment over the same period at lower effective doses. The nematicidal activity of the combined essential oils was suggested by the presence of C10 dienes, C10 trienes and C10 phenol.  相似文献
