The carbon metabolism in cell walls of Chlorella ellipsoideawas studied by following 14C incorporation into cell wall constituentsin photosynthesizing, synchronously growing cells. The rateof incorporation was higher at an early growth phase of thecell cycle. The 14C was incorporated into both the major cellwall constituents, hemicellulose and ‘rigid wall’,and the radioactivity in the latter was distributed into itstwo components, glucosamine and amino acids. In pulse-chaseexperiments, the 14C fixed photosynthetically in the precedingcell cycle was rapidly transferred into the cell wall constituentsat the early growth phase of the ongoing cell cycle, and thereafterwas gradually released from the cell walls, although the totalamount of 14C in the cells remained constant. It was concludedthat the cell wall constituents are turned over during the growthphase of the algal cell cycle, and that the cell wall metabolismin the ongoing cell cycle is closely connected with the carbonmetabolism in the preceding cell cycle. Rates of CO2 and HCC3 fixation in cells of various Chlorellaspecies in suspension were compared from the amounts of 14Cfixed during the 5 s after the injection of a solution containingonly 14CO2 or H14CO3. Results indicated that irrespectiveof the CO2 concentration during growth, Chlorella vulgaris 11h and C. miniata mainly utilized CO2, whereas C. vulgaris C-3,C. sp. K. and C. ellipsoidea took up HCO3 in additionto CO2. Cells of C. pyrenoidosa that had been grown with 1.5%CO2 (high-CO2 cells) mainly utilized CO2, whereas those grownwith air (low-CO2 cells) utilized HCO3 in addition toCO2. Cells that utilized HCO3 had carbonic anhydrase(CA) on their surfaces. The effects of Diamox and CA on the rates of CO2 and HCO3fixation are in accord with the inference that HCO3 wasutilized after conversion to CO2 via the CA located on the cellsurface. CA was found in both the soluble and insoluble fractions;the CA on the cell surface was insoluble. Independent of the modes of utilization, the apparent Km (NaHCO3)for photosynthesis was much lower in low-CO2 cells than in high-CO2ones. The kinetics of 14C-2-acetate assimilation by Chlorella pyrenoidosain the light were examined. Under aerobic conditions the primaryproduct of acetate assimilation was succinic acid which, afterten seconds, contained over 60 per cent of the 14C incorporatedby the cells. The percentage of the total 14C in succinate fellwith time, while that in citrate and glutamate increased. After1800 sec over 60 per cent of 14C was present in two compounds,glutamic acid and an unknown compound (X). Glucose-6-phosphate,fructose-6-phosphate, phosphoglyceric acid and phosphoenolpyruvicacid became labelled after 60 sec but together never containedmore than one per cent of the total 14C incorporated. Underanaerobic conditions succinate was still the primary productof acetate assimilation, and the absence of carbon dioxide resultedin a decrease in 14C incorporation into compound X. The patternof acetate assimilation in acetate grown and acetate adaptedChlorella was very similar to that in photo-autotrophicallygrown Chlorella. In the presence of 10–6M DCMU, succinicacid was the primary product of acetate assimilation, but therewas an early Incorporation of 14C into glutamate, aspartate,and malate. 4 x10–3M MFA did not effect the early incorporationof 14C into succinic acid, but resulted in accumulation of 14Cin citrate and a decreased amount in glutamate and in compound X.  相似文献

Photosynthesis under conditions known to favour glycollate excretionby algae did not result in glycollate excretion in a strainof Chlorella pyrenoidosa unless an inhibitor of glycollate oxidase,-hydroxypyridin-2yl-methane sulphonate (-HPMS), was present.This inhibitor increased the total amount of glycollate presentin the supernatant from the cells during photosynthetic carbondioxide fixation and gave accumulation of 14C in glycollateduring 14CO2 fixation under conditions favouring glycollatesynthesis. At pH 8.3 -HPMS did not stimulate photosynthetic14CO2 fixation in C. pyrenoidosa as occurs with some algae.Photoassimilation of acetate was inhibited by -HPMS, and thiswas shown to result from acetyl-CoA synthetase inhibition by-HPMS.  相似文献

Glycollate Formation during the Photorespiration of Acetate by Chlorella   总被引:1,自引:0,他引:1  
WhenChlorella pyrenoidosa photoassimilates 3H-14C-acetate theglycollic acid formed shows a high 3H/14C ratio, the only othercompounds showing similar ratios being glycerate and serine.The 3H/14C ratio of glycollate was unaffected by the TCA cycleinhibitors MFA, diethylmalonate and arsenite showing that 3Hin glycollate does not result from the oxidation of acetatevia the TCA cycle, the resulting NADP3H2 or NAD3H2 being usedfor the reduction of the glycollate precursor. Although DCMUdecreased the 3H/14C ratio, complete inhibition of glycollatelabelling was not observed with 10–6 M DCMU, at whichconcentration complete inhibition of the Hill reaction is achieved.Although the 3H/14C ratio was unaltered, total dpm of both 14Cand 3H in glycollate were increased by INH. The 3H/14C ratiosof glycerate and serine were decreased by INH, as were the totaldpm of 3H and 14C incorporated into these compounds. Thus, INHinhibits the further metabolism of glycollate to glycerate andserine. The effect of INH on incorporation of 14C-I-acetateinto various cell fractions was investigated. The incorporationof 14C into polysaccharide and lipid was decreased, while theincorporation of 14C into the water-soluble fraction of cellsand therelease of 14CO2 were little affected. Although glycollicacid was an early product of acetate photoassimilation in Chlorellapyrenoidosa, glycollate excretion does not take place undera wide range of environmental conditions shown to favour glycollateexcretion by other algae. However, small amounts of labelledglycollate were detected in the supernatant from the cells duringthe photoassimilation of 3H-14C-acetate, but this glycollatedid not show the high 3H/14C ratio of glycollate present withinthe cell. The failure of Chlorella pyrenoidosa to excrete appreciableamounts of glycollate when photoassimilating acetate or carbondioxide was considered to result from the presence of glycollateoxidase (EC 1.1.3.1 [EC] ) which allowed the further metabolism ofglycollate. Besides glycollate oxidase, glyoxylate reductasewas also demonstrated in Chlorella pyrenoidosa so that glycollatecould function in hydrogen transfer during the photoassimilationof acetate.  相似文献

Ammonia Induces Starch Degradation in Chlorella Cells   总被引:3,自引:0,他引:3  
When ammonia was added to cells of Chlorella which had fixed14CO2 photo synthetically, 14C which had been incorporated intostarch was greatly decreased. A similar effect was observedwhen potassium nitrate and sodium nitrite were added. The ammonia-induceddecrease in 14C-starch was observed in all species of Chlorellatested. With cells of C. vulgaris 11h, most of the radioactivityin starch was recovered in sucrose, indicating that ammoniainduces the conversion of starch into sucrose. The percent of14C recovered in sucrose differed from species to species andpractically no recovery in sucrose was observed in C. pyrenoidosa.In most species tested, the enhancing effects of blue lightand ammonia on O2 uptake as well as the ammonia effect on starchdegradation were greater in cells which had been starved inphosphate medium in the dark than in non-starved cells. In contrast,the enhancing effect of ammonia on dark CO2 fixation was muchgreater in non-starved cells. C. pyrenoidosa was unique in thatblue light did not show any effect on its O2 uptake. Green cells of Chlorella protothecoides when incubated in amedium containing acetate but no nitrogen source, have beenshown to be bleached as strongly as in glucose-induced bleaching.Using U-14C-acetate as tracer, the acetate metabolism of algalcells during the process of acetate-induced bleaching was investigated.Changes in algal cell activities for respiration and assimilationof added 14C-acetate were followed during bleaching processesin "acetate-adapted" and "non-adapted" green cells. As in glucose-inducedbleaching of algal cells, algal cell activity for incorporating14C into lipids showed the most characteristic change, suggestingthat lipogenesis is causally related to the occurrence of bleachingin algal cells. 1) With Chlorella ellipsoidea cells, in the presence of 5x10–6M DSPD, or in its absence, the amounts of 14CO2 incorporatedin P-esters, serine-plus-glycine and alanine were larger underred light than under blue light, whereas blue light specificallyincreased 14CO2-incorporation in aspartate, glutamate, malateand fumarate (blue light effect). The amount of total 14C fixedunder blue or red light was greatly decreased by the additionof DSPD. When the concentration of DSPD was raised to 5x10–4M, practically no radioactivity was found, under blue or redlight, in aspartate, glutamate and fumarate. Radioactivity inalanine was greatly increased. Effects of higher concentrationof DSPD are explained as due to the inhibition of PEP carboxylaseactivity in Chlorella cells. 2) The percentage incorporation of 14C into aspartate and theother compounds mentioned above, under near infra-red illuminationwas significantly smaller than that under blue light and wasalmost equal to that under red light. Photomixotrophic cell suspension culture was established fromthe leaf derived callus cells of Gisekia pharnaceoides L., aC4 dicotyledonous weed. The late log phase cells possessed shade-typecharacters such as low chlorophyll a/b ratio, less pronouncedO2 evolution and CO2 fixation, saturation of photosyntheticCO2 fixation at low intensity. The chloroplasts from these cellscontained granal stacking with high degree of a very few granawhich are characterized by their wide and high degree of stackings. The predominant labelling of 3-phosphoglyceric acid and sugarphosphates (40% of the total 14C incorporated) during 5 s exposureto 14CO2 in light and subsequent decrease in percentage of 14Cin these compounds with increase in exposure time indicatedthe operation of the C3 pathway in these cells. The simultaneoussynthesis of malate (23% of the total 14C incorporated) is relatedto the much pronounced glycolytic and tricarboxylic acid cycleactivities in these cells. The initial proliferation of callimainly from the zones of vascular supplies in the leaf, highstarch content of the cells, presence of large starch grainsin all the chloroplasts, activities of Calvin cycle enzymes,heavy labelling of C3 type intermediates and less labellingof aspartate as early photosynthates and rapid accumulationof radioactivity into starch during 14CO2 assimilation indicatedthat most of the cells in photomixotrophic culture were derivedfrom bundle sheath cells or the leaf cells of Gisekia changetheir function under culture conditions. 1Present address: Tropical Botanic Garden and Research Institute,Navaranga Road, Trivandrum 695 011, India. The development of taurine uptake into the unicellular greenalga Chlorella fusca 211-8b was characterized as a specificresponse to either nitrate or sulphate limitation. Taurine transportunder nitrogen starvation was stimulated by low pH and showeda biphasic kinetics with Km-values of 1.1 x 10–3 mol dm–3and 1.0 x 10–2 mol dm–3. Uptake was substantiallyinhibited by all - and ß-amino acids tested, whereassulphonate analogues failed to diminish taurine accumulation.Thus, uptake seemed to be mediated by a ‘general aminoacid permease’, unable to discriminate between carboxyland sulphonyl groups. However, Chlorella fusca could not catabolizethis unusual ß-amino acid and mobilize the amino-boundnitrogen for growth. Only a small group of -amino acids supportedthe growth of Chlorella fusca as an efficient nitrogen source. In a Chlorella culture grown asynchronously under autotrophicconditions, two biosynthetic enzymes of putrescine—ornithinedecarboxylase (ODC) and arginine decarboxylase (ADC)—weredetected. Both enzymes require pyridoxal phosphate and dithiothreitolfor their activity but differ in their optimal pH, the ionicstrength of their buffer, temperature of inactivation, and Km.In addition, L-canaline was found to inhibit the activity ofODC but not that of ADC. During the logarithmic phase of growth,ODC activity increased sharply, then decreased before the onsetof the stationary phase. The starch content as well as the rate of photosynthetic starchformation in Chlorella ellipsoidea was studied throughout thecell cycle. The starch level in Chlorella cells rose markedlyduring the growing phase in the light, but it started to decreaseafter about 14 to 16 hr regardless of illumination. The rateof starch synthesis, measured by the level of 14C-incorporationinto starch, increased rapidly in the growing phase until 10hr, and decreased promptly thereafter, even in the light. Glucose, either uniformly labelled with14C, or specificallylabelled in the I, 2, or 6 position, was added to C. vulgaris.Radio-active carbon dioxide was produced initially ten timesfaster from glucose-I-14C than from glucose-6-14C. This differencewas found with carbohydrate-starved cultures, exponentiallygrowing cultures, and cultures assimilating ammonia or nitraterapidly. A similar difference was also found with C. pyrenoidosaand Ankistrodesmus. 37 per cent. of the 14C added as glucose-1-14Cto exponentially growing cells was recovered as carbon dioxidebut generally the recovery was less than this. During induction of nitrate reductase in Chlorella vulgaris,synthesis of the precursor, demolybdo cytochrome c reductase,exceeds the synthesis of active enzyme. Evidence is also presentedwhich shows that the purification procedure of Funkhouser etal. The distribution of radioactivity in Chlorella during dark 14CO2fixation was investigated either (a) in normal cells with andwithout added ammonium chloride, or (b) in nitrogen-starvedcells supplied with intermediates of the Krebs-Henseleit ureacycle. In the control experiments almost all the activity was presentin compounds of or associated with, the tricarboxylic acid cycle. The amino-acids citrulline and arginine became radioactive onlyin the presence of ammonia or ornithine where initially theycomprised 40–60 per cent. of the total activity, reactionsof the Krebs–Henseleit urea cycle being implicated intheir formation. No evidence could be found for a complete ureacycle. Unidentified compounds deriving their radioactivity fromthe C4 carbon of citrulline and/or arginine were detected andformed up to 40 per cent. of the total 14CO2 incorporated after25 min.  相似文献

Analysis of products formed in Chlorella vulgaris 11 h cellsduring photosynthesis in air containing 3,000 ppm 14CO2 at varioustemperatures revealed that the level of 14C-starch was maximumaround 20–24?C and decreased with further rise in temperatureuntil 40?C, while 14C-sucrose greatly increased at temperaturesabove about 28?C. Elevating the temperature from 20 to 38?Cduring photosynthetic 14CO2 fixation resulted in a remarkabledecrease in 14C in starch and a concomitant increase in 14Cin sucrose. This conversion of starch to sucrose when shiftingthe temperature from 20 to 38?C proceeded even in the dark.Hydrolysis of sucrose by rß-fructosidase showed that,irrespective of the experimental conditions, the radioactivitiesin sucrose were equally distributed between glucose and fructose.The enhancement of starch degradation with temperature risewas more remarkable than that of the activity of ribulose bisphosphatecarboxylase from the same cells. When Chlorella cells whichhad been preloaded with 14C-starch after photosynthesis for30 min at 20?C were incubated in the dark for an additional30 min at 20?C, 14C-starch was degraded by only about 4%. However,the values after 30-min dark incubation at 28, 32, 36 and 40?Cwere increased by about 10, 19, 36 and 50%, respectively. Duringthe temperature-dependent conversion of starch to sucrose, nosignificant amount of radioactivity accumulated in free glucoseand maltose. To study the wavelength-effect on photosynthetic carbon metabolism,14C-bicarbon-ate was added to Chlorella vulgaris 1 lh suspensionunder monochromatic blue (456 nm) and red (660 nm) light. Thelight intensities were so adjusted that the rates of 14CO2 fixationunder blue and red light were practically equal. Analysis of14C-fixation products revealed that the rates of 14CO2 incorporationinto sucrose and starch were greater under red light than underblue light, while blue light specifically enhanced 14CO2 incorporationinto alanine, aspartate, glutamate, glutamine, malate, citrate,lipid fraction and alcohol-water insoluble non-carbohydratefraction. Pretreatment of the algal cells in phosphate mediumin the dark, which was essential for the blue light enhancementof PEP carboxylase activity, was not necessary to induce theabove wavelength effects. Superimposition of monochromatic bluelight at low intensity (450 erg.cm–2.sec–1) on thered light at saturating intensity caused a significant decreasein the rate of 14CO2 incorporation into sucrose and increasein incorporation into alanine, lipid-fraction, aspartate andother related compounds, indicating that the path of carbonin photosynthesis is regulated by short wavelengdi light ofvery low intensity. Carbon dioxide enhanced chloroplast regeneration in glucose-bleachedcells of Chlorella protothecoides in the presence of CMU inthe light. Both the formation of chlorophyll and the synthesesof RNA and protein were considerably enhanced. The CO2 metabolism of algal cells during greening was investigatedusing 14C-bicarbonate as the tracer. Changes in photosynthetic carbon metabolism during the glucosebleaching of Chlorella protothecoides cells were investigatedusing NaH14CO3 as tracer. Several hours after incubating thegreen algal cells in the glucose medium in the dark, the ratesof 14C-incorporation into glucose polymers and sucrose decreasedand the incorporation into the lipid fraction (fatty acids)greatly increased. At this stage, the rate of photosynthetic14CO2 fixation and the chlorophyll content were practicallythe same as in the starting green cells. Afterwards, the photosyntheticcapacity and chlorophyll content continued to decrease throughoutthe experimental period. The antibiotic thiolactomycin inhibits the fatty acid synthesisfrom both [1-14C]- acetate and [2-14C]malonyl-CoA of spinachleaves, developing castor bean endosperms and avocado mesocarp.On the other hand, fatty acid synthetases of Brevibacteriumammoniagenes and Corynebacterium glutamicum are much less sensitiveto this antibiotic. As has been indicated that thiolactomycininhibits fatty acid synthetase of Escherichia coli but has littleeffect on the synthetases of yeast and rat liver [Hayashi etal. (1983) Biochem. Biophys. Res.
