We propose a joint model for cognitive decline and risk of dementia to describe the pre-diagnosis phase of dementia. We aim to estimate the time when the cognitive evolution of subjects in the pre-dementia phase becomes distinguishable from normal evolution and to study whether the shape of cognitive decline depends on educational level. The model combines a piecewise polynomial mixed model with a random change point for the evolution of the cognitive test and a log-normal model depending on the random change point for the time to dementia. Parameters are estimated by maximum likelihood using a Newton-Raphson-like algorithm. The expected cognitive evolution given age to dementia is then derived and the marginal distribution of dementia is estimated to check the log-normal assumption. 相似文献

When repeated measures of an exposure variable are obtained on individuals, it can be of epidemiologic interest to relate the slope of this variable over time to a subsequent response. Subject-specific estimates of this slope are measured with error, as are corresponding estimates of the level of exposure, i.e., the intercept of a linear regression over time. Because the intercept is often correlated with the slope and may also be associated with the outcome, each error-prone covariate (intercept and slope) is a potential confounder, thereby tending to accentuate potential biases due to measurement error. Under a familiar mixed linear model for the exposure measurements, we present closed-form estimators for the true parameters of interest in the case of a continuous outcome with complete and equally timed follow-up for all subjects. Generalizations to handle incomplete follow-up, other types of outcome variables, and additional fixed covariates are illustrated via maximum likelihood. We provide examples using data from the Multicenter AIDS Cohort Study. In these examples, substantial adjustments are made to uncorrected parameter estimates corresponding to the health-related effects of exposure variable slopes over time. We illustrate the potential impact of such adjustments on the interpretation of an epidemiologic analysis. 相似文献

The development of model-based methods for incomplete data has been a seminal contribution to statistical practice. Under the assumption of ignorable missingness, one estimates the joint distribution of the complete data for thetainTheta from the incomplete or observed data y(obs). Many interesting models involve one-to-one transformations of theta. For example, with y(i) approximately N(mu, Sigma) for i= 1, ... , n and theta= (mu, Sigma), an ordinary least squares (OLS) regression model is a one-to-one transformation of theta. Inferences based on such a transformation are equivalent to inferences based on OLS using data multiply imputed from f(y(mis) | y(obs), theta) for missing y(mis). Thus, identification of theta from y(obs) is equivalent to identification of the regression model. In this article, we consider a model for two-level data with continuous outcomes where the observations within each cluster are dependent. The parameters of the hierarchical linear model (HLM) of interest, however, lie in a subspace of Theta in general. This identification of the joint distribution overidentifies the HLM. We show how to characterize the joint distribution so that its parameters are a one-to-one transformation of the parameters of the HLM. This leads to efficient estimation of the HLM from incomplete data using either the transformation method or the method of multiple imputation. The approach allows outcomes and covariates to be missing at either of the two levels, and the HLM of interest can involve the regression of any subset of variables on a disjoint subset of variables conceived as covariates. 相似文献

Nonlinear mixed effects models have become important tools for growth and yield modeling in forestry. To date, applications have concentrated on modeling single growth variables such as tree height or bole volume. Here, we propose multivariate multilevel nonlinear mixed effects models for describing several plot-level timber quantity characteristics simultaneously. We describe how such models can be used to produce future predictions of timber volume (yield). The class of models and methods of estimation and prediction are developed and then illustrated on data from a University of Georgia study of the effects of various site preparation methods on the growth of slash pine (Pinus elliottii Engelm.). 相似文献

We present a new modification of nonlinear regression models for repeated measures data with heteroscedastic error structures by combining the transform-both-sides and weighting model from Caroll and Ruppert (1988) with the nonlinear random effects model from Lindstrom and Bates (1990). The proposed parameter estimators are a combination of pseudo maximum likelihood estimators for the transform-both-sides and weighting model and maximum likelihood (ML) or restricted maximum likelihood (REML) estimators for linear mixed effects models. We extend the proportional hazards model to a two-level model with a random intercept term and random coefficients. The parameters in the multilevel model are estimated by a combination of EM and Newton-Raphson algorithms. Even for samples of 50 groups, this method produces estimators of the fixed effects coefficients that are approximately unbiased and normally distributed. Two different methods, observed information and profile likelihood information, will be used to estimate the standard errors. This work is motivated by the goal of understanding the determinants of contraceptive use among Nepalese women in the Chitwan Valley Family Study (Axinn, Barber, and Ghimire, 1997). We utilize a two-level hazard model to examine how education and access to education for children covary with the initiation of permanent contraceptive use. 相似文献

A method for modeling survival data with multilevel clustering is described. The Cox partial likelihood is incorporated into the generalized linear mixed model (GLMM) methodology. Parameter estimation is achieved by maximizing a log likelihood analogous to the likelihood associated with the best linear unbiased prediction (BLUP) at the initial step of estimation and is extended to obtain residual maximum likelihood (REML) estimators of the variance component. Estimating equations for a three-level hierarchical survival model are developed in detail, and such a model is applied to analyze a set of chronic granulomatous disease (CGD) data on recurrent infections as an illustration with both hospital and patient effects being considered as random. Only the latter gives a significant contribution. A simulation study is carried out to evaluate the performance of the REML estimators. Further extension of the estimation procedure to models with an arbitrary number of levels is also discussed. 相似文献

In a 1992 Technometrics paper, Lambert (1992, 34, 1-14) described zero-inflated Poisson (ZIP) regression, a class of models for count data with excess zeros. In a ZIP model, a count response variable is assumed to be distributed as a mixture of a Poisson(lambda) distribution and a distribution with point mass of one at zero, with mixing probability p. Both p and lambda are allowed to depend on covariates through canonical link generalized linear models. In this paper, we adapt Lambert's methodology to an upper bounded count situation, thereby obtaining a zero-inflated binomial (ZIB) model. In addition, we add to the flexibility of these fixed effects models by incorporating random effects so that, e.g., the within-subject correlation and between-subject heterogeneity typical of repeated measures data can be accommodated. We motivate, develop, and illustrate the methods described here with an example from horticulture, where both upper bounded count (binomial-type) and unbounded count (Poisson-type) data with excess zeros were collected in a repeated measures designed experiment. 相似文献

This paper extends the multilevel survival model by allowing the existence of cured fraction in the model. Random effects induced by the multilevel clustering structure are specified in the linear predictors in both hazard function and cured probability parts. Adopting the generalized linear mixed model (GLMM) approach to formulate the problem, parameter estimation is achieved by maximizing a best linear unbiased prediction (BLUP) type log‐likelihood at the initial step of estimation, and is then extended to obtain residual maximum likelihood (REML) estimators of the variance component. The proposed multilevel mixture cure model is applied to analyze the (i) child survival study data with multilevel clustering and (ii) chronic granulomatous disease (CGD) data on recurrent infections as illustrations. In many settings, such as interlaboratory testing, small area estimation in sample surveys, and heritability studies, investigators are interested in estimating covariance components for multivariate measurements. However, the presence of outliers can seriously distort estimates obtained using standard procedures such as maximum likelihood. We propose a procedure based on M-estimation for robustly estimating multivariate covariance components in the presence of outliers; the procedure applies to balanced and unbalanced data. We present an algorithm for computing the robust estimates and examine the performance of the estimator through a simulation study. The estimator is used to find covariance components and identify outliers in a study of variability of egg length and breadth measurements of American coots. 相似文献

Analysis of longitudinal data with excessive zeros has gained increasing attention in recent years; however, current approaches to the analysis of longitudinal data with excessive zeros have primarily focused on balanced data. Dropouts are common in longitudinal studies; therefore, the analysis of the resulting unbalanced data is complicated by the missing mechanism. Our study is motivated by the analysis of longitudinal skin cancer count data presented by Greenberg, Baron, Stukel, Stevens, Mandel, Spencer, Elias, Lowe, Nierenberg, Bayrd, Vance, Freeman, Clendenning, Kwan, and the Skin Cancer Prevention Study Group[New England Journal of Medicine 323 , 789–795]. The data consist of a large number of zero responses (83% of the observations) as well as a substantial amount of dropout (about 52% of the observations). To account for both excessive zeros and dropout patterns, we propose a pattern‐mixture zero‐inflated model with compound Poisson random effects for the unbalanced longitudinal skin cancer data. We also incorporate an autoregressive of order 1 correlation structure in the model to capture longitudinal correlation of the count responses. A quasi‐likelihood approach has been developed in the estimation of our model. We illustrated the method with analysis of the longitudinal skin cancer data. 相似文献

Multiple outcomes are often used to properly characterize an effect of interest. This paper proposes a latent variable model for the situation where repeated measures over time are obtained on each outcome. These outcomes are assumed to measure an underlying quantity of main interest from different perspectives. We relate the observed outcomes using regression models to a latent variable, which is then modeled as a function of covariates by a separate regression model. Random effects are used to model the correlation due to repeated measures of the observed outcomes and the latent variable. An EM algorithm is developed to obtain maximum likelihood estimates of model parameters. Unit-specific predictions of the latent variables are also calculated. This method is illustrated using data from a national panel study on changes in methadone treatment practices. 相似文献

Summary . We study quantile regression (QR) for longitudinal measurements with nonignorable intermittent missing data and dropout. Compared to conventional mean regression, quantile regression can characterize the entire conditional distribution of the outcome variable, and is more robust to outliers and misspecification of the error distribution. We account for the within-subject correlation by introducing a ℓ2 penalty in the usual QR check function to shrink the subject-specific intercepts and slopes toward the common population values. The informative missing data are assumed to be related to the longitudinal outcome process through the shared latent random effects. We assess the performance of the proposed method using simulation studies, and illustrate it with data from a pediatric AIDS clinical trial. 相似文献

We examine the hypothesis of an increase of humus disintegration in central European forests by modeling data from the case study “Zierenberg”, which is carried out in Germany. We analyse a spatio‐temporal regression model, which was constructed after an exploratory data analysis. The data are unbalanced repeated measurements collected at several sites and soil depths between 1989 and 1995 with a huge amount of missing observations. Spatial dependencies are considered by coupling linear models for the distinct depths using autoregressive terms and adding random effects. Under the assumption of normality, we provide formulae for maximum likelihood estimation as well as test statistics. As a result of the data analysis, we find that some chemical substances might be influential in the process of humus disintegration. 相似文献

We present a method for estimating the parameters in random effects models for survival data when covariates are subject to missingness. Our method is more general than the usual frailty model as it accommodates a wide range of distributions for the random effects, which are included as an offset in the linear predictor in a manner analogous to that used in generalized linear mixed models. We propose using a Monte Carlo EM algorithm along with the Gibbs sampler to obtain parameter estimates. This method is useful in reducing the bias that may be incurred using complete-case methods in this setting. The methodology is applied to data from Eastern Cooperative Oncology Group melanoma clinical trials in which observations were believed to be clustered and several tumor characteristics were not always observed. 相似文献
