Geoactive soil fungi were investigated for phosphatase‐mediated uranium precipitation during growth on an organic phosphorus source. Aspergillus niger and Paecilomyces javanicus were grown on modified Czapek–Dox medium amended with glycerol 2‐phosphate (G2P) as sole P source and uranium nitrate. Both organisms showed reduced growth on uranium‐containing media but were able to extensively precipitate uranium and phosphorus‐containing minerals on hyphal surfaces, and these were identified by X‐ray powder diffraction as uranyl phosphate species, including potassium uranyl phosphate hydrate (KPUO6.3H2O), meta‐ankoleite [(K1.7Ba0.2)(UO2)2(PO4)2.6H2O], uranyl phosphate hydrate [(UO2)3(PO4)2.4H2O], meta‐ankoleite (K(UO2)(PO4).3H2O), uramphite (NH4UO2PO4.3H2O) and chernikovite [(H3O)2(UO2)2(PO4)2.6H2O]. Some minerals with a morphology similar to bacterial hydrogen uranyl phosphate were detected on A. niger biomass. Geochemical modelling confirmed the complexity of uranium speciation, and the presence of meta‐ankoleite, uramphite and uranyl phosphate hydrate between pH 3 and 8 closely matched the experimental data, with potassium as the dominant cation. We have therefore demonstrated that fungi can precipitate U‐containing phosphate biominerals when grown with an organic source of P, with the hyphal matrix serving to localize the resultant uranium minerals. The findings throw further light on potential fungal roles in U and P biogeochemistry as well as the application of these mechanisms for element recovery or bioremediation.  相似文献

Five new open-framework compounds of gallium have been synthesized by hydrothermal methods and their structures determined by single crystal X-ray diffraction studies. The compounds, [C8N4H26][Ga6F4(PO4)6], I, [C5N3H11][Ga3F2(PO4)3]·H2O, II, [C6N3H19][Ga4(C2O4)(PO4)4(H2PO4)]·2H2O, III, [Ga2F3(HPO4)(PO4)]·2H3O, IV, and [C3N2H5]2[Ga4(H2O)3(HPO3)7], V, possess three-dimensional structures. All the compounds are formed by the connectivity between the Ga polyhedra and phosphite/phosphate units. The observation of SBU-6 (I and II) and spiro-5 (IV) secondary building units (SBUs) are noteworthy. The flexibility of the formation of gallium phosphate frameworks has been established by the isolation of two related structures (I and II) from the same SBU units but different organic amines. Some of the present structures have close resemblance to the gallium phosphate phases known earlier. The compounds have been characterized by CHN analysis, powder XRD, IR, and TGA.  相似文献

The thermophilic, gram-positive bacterium Thermoterrabacterium ferrireducens coupled organotrophic growth to the reduction of sparingly soluble U(VI) phosphate. X-ray powder diffraction and X-ray absorption spectroscopy analysis identified the electron acceptor in a defined medium as U(VI) phosphate [uramphite; (NH4)(UO2)(PO4) . 3H2O], while the U(IV)-containing precipitate formed during bacterial growth was identified as ningyoite [CaU(PO4)2 . H2O]. This is the first report of microbial reduction of a largely insoluble U(VI) compound.  相似文献

Two new zinc complexes, Zn(HL1)2 (1) and [Zn2(H2L2)(OAc)2]2 (2) [H2L1 = Schiff base derived from o-vanillin and (R)-(+)-2-amino-3-phenyl-1-propanol, H3L2 = Schiff base derived from o-vanillin and 2-amino-2-ethyl-1,3-propanediol], have been synthesized and characterized by single crystal X-ray diffraction, elemental analyses, TG analyses, solid fluorescence, IR, UV-Vis and circular dichroism spectra. The structural analysis shows that complex 1 has a right-handed double helical chain along the crystallographic b axis. A homochiral 3D supramolecular architecture has been further constructed by intermolecular C-H··· π, O-H···O and C-H···O interactions. Complex 2 includes two crystallographically independent binuclear zinc molecules. The two binuclear zinc molecules are isostructural. The 2-D sheet supramolecular structure was formed by intermolecular hydrogen bonding interaction. The fluorescence of ligands and complexes in DMF at room temperature are studied. The interactions of two complexes with calf thymus DNA (CT-DNA) are investigated using UV-Vis, CD and fluorescence spectroscopy. The results show that complex 1 exhibits higher interaction with CT-DNA than complex 2. In addition, in vitro cytotoxicity of the complexes towards four kinds of cancerous cell lines (A549, HeLa, HL-60 and K562) were assayed by the MTT method. Investigations on the structures indicated that the chirality and nuclearity of zinc complexes play an important role on cytotoxic activity.  相似文献

Metal localisation was investigated in the lichenised ascomycete Trapelia involuta growing on a range of uraniferous minerals including metazeunerite [Cu(UO2)2(AsO4)2·8H2O], metatorbernite [Cu(UO2)2(PO4)2·8H2O], autunite [Ca(UO2)2(PO4)2·10H2O] and uranium-enriched iron oxide and hydroxide minerals at the abandoned South Terras mine site, Cornwall, UK. Apothecia from samples collected from waste dumps at the mine have an unusually dark colour that decolorized with NaOCl, an observation which together with Fourier Transform Infrared Spectroscopy of apothecial extracts, suggested the presence of melanin-like pigments. X-ray element mapping and probe traverses across the lichen-rock interface identify the highest U, Fe, and Cu concentrations in the outer parts of melanised apothecia. Accumulation of mineral particulates and complexing with lichen acids are not considered responsible for this since element ratios in the traverses do not correspond with those of likely mineral phases and lichen metabolites are localised in different tissues. Metal biosorption by melanin-like pigments are likely to be responsible for the observed metal fixation. No detectable U or Cu was observed in control samples although Fe showed a similar localisation in some specimens. The high concentrations of mucopolysaccharides and P recorded inside apothecia (within asci containing reproductive spores and hypothecium) suggests that the formation of melanised tissues may help protect vital reproductive tissues from the toxic effects of U and other metals, since the uranyl ion complexes strongly with phosphate species.  相似文献

Pseudomonas syringae pv. glycinea PG4180 produces the polyketide phytotoxin coronatine. The effects of environmental, nutritional, and host factors on growth and coronatine production by PG4180 were examined by varying the components of a defined basal medium which contained the following nutrients per liter: glucose (10 g), NH4Cl (1 g), MgSO4 · 7H2O (0.2 g), KH2PO4 (4.1 g), K2HPO4 · 3H2O (3.6 g), and FeCl3 (2 μM). Bacterial growth was recorded as dry weight, and coronatine production was measured by high-performance liquid chromatography. Both growth and the quantity of coronatine synthesized were significantly affected by carbon source, nutrient levels (glucose, NH4Cl, phosphate, Mg, and SO4), amino acid supplements, and the presence of complex carbon and nitrogen sources. The yield of coronatine generally declined when conditions were varied from those in the basal medium. Coronatine production and growth were not affected when the pH was adjusted from 6.5 to 7.8. Increases in the osmolarity of the basal medium significantly decreased coronatine production without affecting growth. The addition of plant extracts, plant-derived secondary metabolites, or zinc did not affect growth or coronatine production, while the addition of millimolar levels of KNO3 or micromolar levels of FeCl3 significantly enhanced coronatine production. The yield of coronatine was maximized after a 7-day incubation at 18°C and 280 rpm. The results of the present study were used to formulate a medium which allowed for enhanced coronatine production in nearly all strains of P. syringae tested. A rapid method for extracting coronatine from small volumes of culture supernatant was also developed.  相似文献

The kinetics for the reduction of sulfate alone and for concurrent uranium [U(VI)] and sulfate reduction, by mixed and pure cultures of sulfate-reducing bacteria (SRB) at 21 ± 3°C were studied. The mixed culture contained the SRB Desulfovibrio vulgaris along with a Clostridium sp. determined via 16S ribosomal DNA analysis. The pure culture was Desulfovibrio desulfuricans (ATCC 7757). A zero-order model best fit the data for the reduction of sulfate from 0.1 to 10 mM. A lag time occurred below cell concentrations of 0.1 mg (dry weight) of cells/ml. For the mixed culture, average values for the maximum specific reaction rate, Vmax, ranged from 2.4 ± 0.2 μmol of sulfate/mg (dry weight) of SRB · h−1) at 0.25 mM sulfate to 5.0 ± 1.1 μmol of sulfate/mg (dry weight) of SRB · h−1 at 10 mM sulfate (average cell concentration, 0.52 mg [dry weight]/ml). For the pure culture, Vmax was 1.6 ± 0.2 μmol of sulfate/mg (dry weight) of SRB · h−1 at 1 mM sulfate (0.29 mg [dry weight] of cells/ml). When both electron acceptors were present, sulfate reduction remained zero order for both cultures, while uranium reduction was first order, with rate constants of 0.071 ± 0.003 mg (dry weight) of cells/ml · min−1 for the mixed culture and 0.137 ± 0.016 mg (dry weight) of cells/ml · min−1 (U0 = 1 mM) for the D. desulfuricans culture. Both cultures exhibited a faster rate of uranium reduction in the presence of sulfate and no lag time until the onset of U reduction in contrast to U alone. This kinetics information can be used to design an SRB-dominated biotreatment scheme for the removal of U(VI) from an aqueous source.  相似文献

Enterolith fragments from two tapir species and horses were subjected to x-ray diffraction analysis. Tapir enteroliths were formed as layers of mineral deposited around a foreign nidus. The structure was similar to that of equine enteroliths except that tapir enteroliths lacked a central region of radially symmetrical coarse crystals. The enteroliths from tapirs were composed primarily of vivianite [Fe3(PO4)2 · 8H2O] and newberyite [MgH(PO4) · 3H2O], instead of the struvite [Mg(NH4)(PO4) · 6H2O] of enteroliths from horses. The reason for this difference is not known. Based on the chemistry of these mineral precipitates and information from other species, it was concluded that dietary manipulation to maximize carbohydrate fermentation and minimize protein fermentation in the large intestine may help prevent enterolithiasis in tapirs. The uranium(IV) complexes [U(EDTA)(H2O)2], [U(HOEDTA)]+, and [U(DTPA)]? are well-formed in the pH fange 2–3 ([DTPA]5- = diethylenetriaminepentaacetate; [HOEDTA]3-  N-(2-hydroxyethyl)ethylenediaminetriacetate). Of these, only [U(DTPA)]- is extracted from an aqueous phase at pH 2 by the perchlorate salt of the primary amine, Primene JM-T. As the aqueous phase pH was raised, extraction occurred in all three cases and hydrolysed species may be extracted from EDTA and HOEDTA solutions but [U(DTPA)]? resists hydrolysis. The addition of sulphate had a marked effect on the extraction of U(IV) from EDTA and HOEDTA through the formation of [U(EDTA)(SO4)(H2O)]2- and [U(HOEDTA)(SO4)(H2O)n]?. The equilibrium constant, log β1, for: [(U(EDTA)(H2O)2] 2 [SO4]2? ? [U(EDTA)(SO4)(H2O)]2- 2 H2O was found to be 2.43 ± 0.04 (I = 1 mol dm?3, NaClO4; pH 2.0; 20 °C) from spectrophotometric data.With tri-n-octylphosphine oxide (TOPO) electronic spectroscopy showed that the same U(IV) complex was extracted at pH 2 for Cs2UCl6, U(IV)/ HOEDTA, and U(IV)/DTPA and the aminepoly- carboxylates were aqueous phase masking agents but with [U(EDTA)(H2O)2] oxidation gave a uranyl(VI) organic phase species.Uranium(IV) is strongly extracted from aqueous solutions of HOEDTA at pH 2 or 3 by bis(2-ethyl- hexyl)phosphoric acid (HBEHP) but less so from EDTA and DTPA. Since U(IV) is completely extracted from Cs2UCl6 it could be that the amine- polycarboxylates were aqueous phase masking agents although spectral evidence did not support this.  相似文献

A field isolate of Desulfovibrio desulfuricans was grown in defined medium in a two-stage continuous culture apparatus with different concentrations of phosphate in the feed medium. The first state (V1) was operated as a conventional chemostat (D = 0.045 h−1) that was limited in energy source (lactate) or phosphate. The second stage (V2) received effluent from V1 but no additional nutrients, and contained a healthy population of transiently starved or resting cells. An increase in the concentration of phosphate in the medium fed to V1 resulted in increased corrosion rates of carbon steel in both V1 and V2. Despite the more rapid corrosion observed in growing cultures relative to that in resting cultures, corrosion products that were isolated under strictly anaerobic conditions from the two culture modes had similar bulk compositions which varied with the phosphate content of the medium. Crystalline mackinawite (Fe9S8), vivianite [Fe3(PO4)2 · 8H2O], and goethite [FeO(OH)] were detected in amounts which varied with the culture conditions. Chemical analyses indicated that the S in the corrosion product was almost exclusively in the form of sulfides, while the P was present both as phosphate and as unidentified components, possibly reduced P species. Some differential localization of S and P was observed in intact corrosion products. Cells from lactate-limited, but not from phosphate-limited, cultures contained intracellular granules that were enriched in P and Fe. The results are discussed in terms of several proposed mechanisms of microbiologically influenced corrosion.  相似文献

Early studies with Geobacter sulfurreducens suggested that outer-surface c-type cytochromes might play a role in U(VI) reduction, but it has recently been suggested that there is substantial U(VI) reduction at the surface of the electrically conductive pili known as microbial nanowires. This phenomenon was further investigated. A strain of G. sulfurreducens, known as Aro-5, which produces pili with substantially reduced conductivity reduced U(VI) nearly as well as the wild type, as did a strain in which the gene for PilA, the structural pilin protein, was deleted. In order to reduce rates of U(VI) reduction to levels less than 20% of the wild-type rates, it was necessary to delete the genes for the five most abundant outer surface c-type cytochromes of G. sulfurreducens. X-ray absorption near-edge structure spectroscopy demonstrated that whereas 83% ± 10% of the uranium associated with wild-type cells correspond to U(IV) after 4 h of incubation, with the quintuple mutant, 89% ± 10% of uranium was U(VI). Transmission electron microscopy and X-ray energy dispersion spectroscopy revealed that wild-type cells did not precipitate uranium along pili as previously reported, but U(IV) was precipitated at the outer cell surface. These findings are consistent with those of previous studies, which have suggested that G. sulfurreducens requires outer-surface c-type cytochromes but not pili for the reduction of soluble extracellular electron acceptors.  相似文献

Lettuce (Lactuca sativa L. cv Minetto) seeds were primed in aerated solutions of 1% K3PO4 or water at 15°C in the dark for various periods of time to determine the manner by which seed priming bypasses thermodormancy. Seeds which were not primed did not germinate at 35°C, whereas those which were primed for 20 h in 1% K3PO4 or distilled H2O had up to 86% germination. The rate of water uptake and respiration during priming were similar regardless of soak solution. Cell elongation occurred in both water and 1% K3PO4, 4 to 6 h prior to cell division. Both processes commenced sooner in water than K3PO4. Radicle protrusion (germination) occurred in the priming solution at 21 h in water and 27 h in 1% K3PO4.

Respiration, radicle protrusion and cell division consistently occurred sooner in primed (redried) seeds compared to nonprimed seeds when they were imbibed at 25°C. Cell division and elongation commenced after 10 h imbibition in primed (redried) seeds imbibed at 35°C. Neither process occurred in nonprimed seeds. Respiratory rates were higher in both primed and nonprimed seeds imbibed at 35°C compared to those imbibed at 25°C, although radicle protrusion did not occur in nonprimed seeds which were imbibed at 35°C. It is apparent that cell elongation and division are inhibited during high temperature imbibition in nonprimed lettuce seeds. Seed priming appears to lead to the irreversible initiation of cell elongation, thus overcoming thermodormancy.

The mixed-metal trinuclear cluster cations [H3Ru2(C6Me6)2Os(C6H6)(O)]+ (1), [H3Ru2(1,2,4,5-C6H2Me4)2Os(p-MeC6H4iPr)(O)]+ (2) and [H3Ru2(1,2,4,5-C6H2Me4)2Os(C6H6)(O)]+ (3) have been synthesised from the corresponding dinuclear precursors [H3Ru2(arene)2]+ and the corresponding mononuclear complexes [Os(arene)(H2O)3]2+, isolated and characterised as the tetrafluoroborate and hexafluorophosphate salts. The cations 1, 2 and 3 are heteronuclear analogues of the cluster cation [H3Ru3(C6H6)(C6Me6)2(O)]+ that possesses a homonuclear metallic core. The single-crystal X-ray structure analyses of [1][BF4], [2][PF6] and [3][PF6] reveal an equiangular metal triangle despite the presence of an osmium atom in the metallic core.  相似文献

Kinetic parameters and the role of cytochrome c3 in sulfate, Fe(III), and U(VI) reduction were investigated in Desulfovibrio vulgaris Hildenborough. While sulfate reduction followed Michaelis-Menten kinetics (Km = 220 μM), loss of Fe(III) and U(VI) was first-order at all concentrations tested. Initial reduction rates of all electron acceptors were similar for cells grown with H2 and sulfate, while cultures grown using lactate and sulfate had similar rates of metal loss but lower sulfate reduction activities. The similarities in metal, but not sulfate, reduction with H2 and lactate suggest divergent pathways. Respiration assays and reduced minus oxidized spectra were carried out to determine c-type cytochrome involvement in electron acceptor reduction. c-type cytochrome oxidation was immediate with Fe(III) and U(VI) in the presence of H2, lactate, or pyruvate. Sulfidogenesis occurred with all three electron donors and effectively oxidized the c-type cytochrome in lactate- or pyruvate-reduced, but not H2-reduced cells. Correspondingly, electron acceptor competition assays with lactate or pyruvate as electron donors showed that Fe(III) inhibited U(VI) reduction, and U(VI) inhibited sulfate loss. However, sulfate reduction was slowed but not halted when H2 was the electron donor in the presence of Fe(III) or U(VI). U(VI) loss was still impeded by Fe(III) when H2 was used. Hence, we propose a modified pathway for the reduction of sulfate, Fe(III), and U(VI) which helps explain why these bacteria cannot grow using these metals. We further propose that cytochrome c3 is an electron carrier involved in lactate and pyruvate oxidation and is the reductase for alternate electron acceptors with higher redox potentials than sulfate.  相似文献

Summary Dihydrogen phosphate ion activity (H2PO4) and phosphate potential were measured in the saturation extracts of 14 representative Red Mediterranean soils (Alfisols) of Greece, estimating the ionic strength from a) measured ionic concentrations b) actual ionic concentrations and c) electrical conductivity measurements. The values of (H2PO4) and phosphate potential thus obtained were correlated with P-uptake by rye-grass in a pot experiment. The correlation coefficients did not differ significantly (p≤0.001) from one another and from those of (H2PO4) and phosphate potential measured in 0.01M CaCl2 extracts.  相似文献

A new three-dimensional open framework structure of mixed valence ethylenediamine-vanadium phosphate [C2H10N2][(HVIVO3)(HVVO2)(PO4)] (1), has been synthesized under mild hydrothermal conditions and characterized by elemental analyses, IR, fluorescent spectrum, TG-DTA and single crystal X-ray diffraction. Compound 1 exhibits a novel three-dimensional (3D) vanadium phosphate anion framework composed of vanadium, phosphate, and oxygen atoms through covalent bonds, with the diprotonated ethylenediamine [NH3CH2CH2NH3]2+ cations residing in the channels along c-axis. The organic diprotonated ethylenediamine cations interact with the O atoms in the inorganic network through hydrogen bonds. The electrochemical behavior of 1 has also been studied in detail by cyclic voltammograms, which is very important for practical applications in electrode modification. Furthermore, the strong photoluminescence property of compound 1 is also measured at room temperature.  相似文献

Sulforaphane (SFN) is an isothiocyanate found in cruciferous vegetables with anti-inflammatory, anti-oxidant and anti-cancer activities. However, the antioxidant and anticancer mechanism of sulforaphane is not well understood. In the present research, we reported binding modes, binding constants and stability of SFN–DNA and -RNA complexes by Fourier transform infrared (FTIR) and UV–Visible spectroscopic methods. Spectroscopic evidence showed DNA intercalation with some degree of groove binding. SFN binds minor and major grooves of DNA and backbone phosphate (PO2), while RNA binding is through G, U, A bases with some degree of SFN–phosphate (PO2) interaction. Overall binding constants were estimated to be K(SFN–DNA)=3.01 (± 0.035)×104 M-1 and K(SFN–RNA)= 6.63 (±0.042)×103 M-1. At high SFN concentration (SFN/RNA = 1/1), DNA conformation changed from B to A occurred, while RNA remained in A-family structure.  相似文献

A synthetic study in the Cd-Se-O-Cl system led to formation of the new oxochloride compound Cd4(SeO3)2OCl2 via solid state reactions. The compound crystallizes in the orthorhombic space group Fmmm with cell parameters a = 7.3610(3) Å, b = 15.4936(2) Å, c = 17.5603(3) Å, Z = 8, S = 0.969, F(000) = 2800, R = 0.0185, Rw = 0.0384. Single crystal X-ray data were collected at 293 K. The crystal structure can be considered as layered and the building units are distorted [Cd(1)O6] octahedra, distorted [Cd(2)O8] cubes, irregular [Cd(3)O4Cl2] polyhedra and SeO3E trigonal pyramids. There are two crystallographically unique Cl atoms that both are half occupied. Thermogravimetric studies show that the compound starts to decompose at 500°C. The crystal structure of the new compound is closely related to the previously described compound Cd4(SeO3)2Cl4(H2O).  相似文献

Reaction of Cu(ClO4)2·6H2O, SRaaiNR′ (1-alkyl-2-[(o-thioalkyl)phenylazo]imidazole) and NH4SCN (1:1:2 mol ratio) affords distorted square pyramidal, [CuII(SRaaiNR′)(SCN)2] (3) compound while identical reaction with [Cu(MeCN)4](ClO4) yields -SCN- bridged coordination polymer, [CuI(SRaaiNR′)(SCN)]n (4). These two redox states [CuII and CuI] are interconvertible; reduction of [CuII(SRaaiNR′)(SCN)2] by ascorbic acid yields [CuI(SRaaiNR′)(SCN)]n while the oxidation of [CuI(SRaaiNR′)(SCN)]n by H2O2 in presence of excess NH4SCN affords [CuII(SRaaiNR′)(SCN)2]. They are structurally confirmed by single crystal X-ray diffraction study. Cyclic voltammogram of the complexes show Cu(II)/Cu(I) redox couple at ∼0.4 V and azo reductions at negative to SCE. UV light irradiation in MeCN solution of [CuI(SRaaiNR′)(SCN)]n (4) show trans-to-cis isomerisation of coordinated azoimidazole. The reverse transformation, cis-to-trans, is very slow with visible light irradiation while the process is thermally accessible. Quantum yields (?t→c) of trans-to-cis isomerisation are calculated and free ligands show higher ? than their Cu(I) complexes. The activation energy (Ea) of cis-to-trans isomerisation is calculated by controlled temperature experiment. Copper(II) complexes, 3, do not show photochromism. DFT and TDDFT calculation of representative complexes have been used to determine the composition and energy of molecular levels and results have been used to explain the solution spectra, photochromism and redox properties of the complexes.  相似文献

Single cell green algae (microalgae) are rapidly emerging as a platform for the production of sustainable fuels. Solar-driven H2 production from H2O theoretically provides the highest-efficiency route to fuel production in microalgae. This is because the H2-producing hydrogenase (HYDA) is directly coupled to the photosynthetic electron transport chain, thereby eliminating downstream energetic losses associated with the synthesis of carbohydrate and oils (feedstocks for methane, ethanol and oil-based fuels). Here we report the simultaneous knock-down of three light-harvesting complex proteins (LHCMB1, 2 and 3) in the high H2-producing Chlamydomonas reinhardtii mutant Stm6Glc4 using an RNAi triple knock-down strategy. The resultant Stm6Glc4L01 mutant exhibited a light green phenotype, reduced expression of LHCBM1 (20.6% ±0.27%), LHCBM2 (81.2% ±0.037%) and LHCBM3 (41.4% ±0.05%) compared to 100% control levels, and improved light to H2 (180%) and biomass (165%) conversion efficiencies. The improved H2 production efficiency was achieved at increased solar flux densities (450 instead of ∼100 µE m−2 s−1) and high cell densities which are best suited for microalgae production as light is ideally the limiting factor. Our data suggests that the overall improved photon-to-H2 conversion efficiency is due to: 1) reduced loss of absorbed energy by non-photochemical quenching (fluorescence and heat losses) near the photobioreactor surface; 2) improved light distribution in the reactor; 3) reduced photoinhibition; 4) early onset of HYDA expression and 5) reduction of O2-induced inhibition of HYDA. The Stm6Glc4L01 phenotype therefore provides important insights for the development of high-efficiency photobiological H2 production systems.  相似文献
