贵州烟草根围AM真菌多样性的初步研究   总被引:1,自引:0,他引:1  
从贵州省内烟区不同土壤生态环境下采集烟草根际土样,湿筛离心法分离丛枝菌根(AM)真菌孢子,鉴定出烟草AM真菌4属20种,其中球囊霉属9种,无梗囊霉属7种,巨孢囊霉属3种,盾巨孢囊霉属1种。从土壤样品DNA中扩增AM真菌特异性片段并采用DGGE技术对AM真菌多样性进行分析。测序结果显示烟草根际土壤中菌根真菌主要菌群为球囊霉属,与湿筛离心法的鉴定结果一致。为进一步研究贵州地区AM真菌多样性以及开发应用提供了依据。  相似文献

Soil basidiomycetes play key roles in forest nutrient and carbon cycling processes, yet the diversity and structure of below ground basidiomycete communities remain poorly understood. Prescribed burning is a commonly used forest management practice and there is evidence that single fire events can have an impact on soil fungal communities but little is known about the effects of repeated prescribed burning. We have used internal transcribed spacer (ITS) terminal restriction fragment length polymorphism (T-RFLP) analysis to investigate the impacts of repeated prescribed burning every two or four years over a period of 30 years on soil basidiomycete communities in an Australian wet sclerophyll forest. Detrended correspondence analysis of ITS T-RFLP profiles separated basidiomycete communities in unburned control plots from those in burned plots, with those burned every two years being the most different from controls. Burning had no effect on basidiomycete species richness, thus these differences appear to be due to changes in community structure. Basidiomycete communities in the unburned control plots were vertically stratified in the upper 20 cm of soil, but no evidence was found for stratification in the burned plots, suggesting that repeated prescribed burning results in more uniform basidiomycete communities. Overall, the results demonstrate that repeated prescribed burning alters soil basidiomycete communities, with the effect being greater with more frequent burning.  相似文献

Aim: The aim of this study was to quantitatively and qualitatively assess the effect of sample storage on the metabolically active microbial community found in sputum samples from patients with cystic fibrosis (CF). Methods: Sputum samples were collected and split in two equal aliquots one of which was immersed in RNAlater and refrigerated immediately, the second stored at room temperature for 24 h and RNAlater was subsequently added. mRNA was extracted, and RT‐PCR‐DGGE and qPCR analysis of the bacterial and fungal communities was carried out. Results: Significant differences in the bacterial communities between the two protocols were observed but there were no significant difference seen in the fungal community analyses. Analysis by qPCR demonstrated that room temperature storage gave statistically significant increases in eubacteria and Pseudomonas spp. and a statistically significant decrease in those of Haemophilus influenzae. Conclusions: The analysis of metabolically active microbial communities from CF sputum using molecular techniques indicated that samples should be stored at 4°C upon addition of RNAlater to obtain an accurate depiction of the CF lung microbiota. Also, storing respiratory samples at room temperature may cause an over representation of Pseudomonas aeruginosa and mask the presence of other clinically significant organisms.  相似文献

为了解长期定位施肥对石灰性紫色水稻土丛枝菌根真菌(AMF)物种多样性的影响,从稻麦轮作下连续26年定位施农家肥(M)、氮肥+农家肥(NM)、氮磷肥+农家肥(NPM)、氮磷钾肥+农家肥(NPKM)、不施肥(CK)、氮肥(N)、氮磷肥(NP)、氮磷钾肥(NPK)等不同施肥处理小区采集根区土样,分离鉴定AMF、测定多样性特征。共鉴定出17种AMF,其中球囊霉属Glomus 12种,占已分离总数的70.5%,为优势属;无梗囊霉属Acaulospora 3种,占17.6%;原囊霉属Archaeospora和内养囊霉属Entrophospora各1种,各占5.9%。无论是石灰性紫色土种植水稻还是小麦,AMF侵染率最高的施肥处理均为无肥处理(CK),施肥特别是磷肥,降低AMF侵染率、孢子密度、相对多度和种丰度。变性梯度凝胶电泳(denaturing gradient gel electrophoresis,DGGE)分析结果表明,不施肥对照(CK)的AMF多样性最丰富,多样性指数最高,水稻和小麦种植下的多样性指数分别为2.68和3.02;与无机肥处理相比,农家肥配施无机肥提高了AMF多样性指数。农家肥配施无机肥更有利于AMF发育。  相似文献

湖泊沉积物中真菌的类群及其作用所知甚少。以长江中上游代表性湖泊洪湖为研究对象,从湖心截取了一个72cm长的沉积柱,通过rDNA内转录间隔区(ITS)的变性梯度凝胶电泳(DGGE),对洪湖沉积物中真菌群落结构的垂直变化进行了分析。结果显示,DGGE条带以14-18cm层次为过渡,在2-14cm和14-72cm之间存在一定差异。Shannon-Weaver index(H′)在上下层之间有波动。聚类分析上层2-18cm和中下层18-72cm分别以不同的小类聚在一起。表明14-18cm以上和以下的类群存在差异,  相似文献

Aims: To understand the effects of lactic acid bacteria (LAB) inoculation on fermentation products, aerobic stability and microbial communities of silage. Methods and Results: Wilted Italian ryegrass was stored in laboratory silos with and without inoculation of Lactobacillus rhamnosus and Lactobacillus buchneri. The silos were opened after 14, 56 and 120 days and then subjected to aerobic deterioration for 7 days. Intensive alcoholic fermentation was found in untreated silage; the sum of ethanol and 2,3‐butanediol content at day 14 was about 7 times higher than that of lactic and volatile fatty acids. Alcoholic fermentation was suppressed by L. rhamnosus and L. buchneri inoculation and lactic acid and acetic acid became the dominant fermentation products, respectively. Silages were deteriorated in untreated and L. rhamnosus‐inoculated silages, whereas no spoilage was found in L. buchneri‐inoculated silage. Enterobacteria such as Erwinia persicina, Pantoea agglomerans and Rahnella aquatilis were detected in untreated silage, whereas some of these bacteria disappeared or became faint with L. rhamnosus treatment. When silage was deteriorated, Lactobacillus brevis and Bacillus pumilus were observed in untreated and L. rhamnosus‐inoculated communities, respectively. The inoculated LAB species was detectable in addition to untreated bacterial communities. Saccharomyces cerevisiae and Pichia anomala were the main fungi in untreated and L. rhamnosus‐inoculated silages; however, P. anomala was not visibly seen in L. buchneri‐inoculated silage either at silo opening or after exposure to air. Conclusion: Inoculation with L. rhamnosus can suppress alcoholic fermentation of wilted grass silage with elimination of enterobacteria at the beginning of fermentation. Addition of L. buchneri may improve aerobic stability, with distinct inhibitory effect observed on P. anomala after silo opening. Significance and Impact of the Study: Bacterial and fungal community analyses help us to understand how inoculated LAB can function to improve the fermentation and aerobic stability of silage.  相似文献

实验选用了我国3种不同土壤研究土壤硝化活性、硝化细菌数量,并使用变性梯度凝胶电泳(DGGE)的方法研究了不同土壤中氨氧化细菌(AOB)区系变化。通过28d的土壤培养实验研究发现,潮土具有最强的硝化势,几乎100%的铵态氮转化为硝态氮;而红壤中的硝化势最弱,只有4.9%的铵态氮转化为硝态氮。对这3种土壤硝化细菌进行计数发现,3种土壤氨氧化菌数量差异显著,而3种土壤亚硝酸氧化菌(NOB)处于一个数量级。采用氨氧化菌功能基因amoA(氨单加氧酶ammoniamonooxygenase)特异PCR结合DGGE的方法对土壤氨氧化菌区系进行分析。红壤有4个氨氧化菌种属,与潮土和黄泥土没有共同的氨氧化菌种属。4个种属中两个是与潮土和黄泥土亲源性比较远的,特有的氨氧化菌种属,这两个种属与已知的Nitrosospira属的cluster3bz97838和Nitrosospira属的cluster3aAF353263亲源性比较近。潮土有5个氨氧化菌种属,潮土与黄泥土有两个共同的氨氧化菌种属,这两个种属中的一个是潮土和黄泥土特有的,与其他氨氧化菌种属亲源性比较远的氨氧化菌种属,这个种属与已知的Nitrosospira属的cluster3bZ97849亲源性比较近。黄泥土有4个氨氧化菌种属,除了与潮土共有的一个种属是两种土壤特有的氨氧化菌种属外,黄泥土还有一个与其他氨氧化菌种属亲源性比较远的,黄泥土特有的种属,与Nitrosospira属的cluster3aAF353263亲源性很近。3种土壤中分离到的硝化细菌表现出不同的硝化能力。实验结果表明,以amoA基因为目标的PCR-DGGE是比以16SrDNA为目标的PCR-DGGE更有效的研究氨氧化菌种群的方法;3种土壤的氨氧化菌种群差异显著,尤其是红壤的氨氧化菌种群与另外两种土壤差异明显,这种差异可能与红壤的低pH条件对氨氧化菌种群的长期选择有关;3种土壤中的硝化活性与土壤中的硝化细菌数量没有显著相关,可能由于3种土壤差异显著的土壤环境对硝化活性的影响造成。因此在对不同土壤硝化细菌进行研究时不仅需要对硝化细菌数量进行研究,还需要研究不同土壤中硝化细菌的种属及不同土壤环境条件对硝化细菌硝化活性的影响。  相似文献

【目的】为探讨耕作和施加有机肥、化肥对黑土表层(0-30cm)、中层(100-130cm)及深层(200-230cm)细菌群落结构的影响,【方法】应用DGGE技术对相应土层中细菌群落结构进行了解析。【结果】结果表明,与对照相比,耕作和施加有机肥、化肥对表层黑土细菌群落结构影响较大,二者差异度为4%;而对中层和深层细菌群落结构影响较小,二者差异度为2%。对于细菌群落结构的垂向变化,中层和深层中细菌群落结构的相似性远远高于同表层的相似性。【结论】可见,耕作和施加有机肥、化肥仅对黑土表层土壤(0-30cm)具有一定的影响,而对100cm以下土壤细菌群落影响较小,细菌群落随土壤深度不同的垂向变化要远高于土壤管理措施造成的影响。  相似文献

The community of arbuscular mycorrhizal fungi (AMF) colonizing the roots of Festuca pratensis and Achillea millefolium was characterized in a Swedish pasture at different times, along a gradient of fertilization. The small subunit ribosomal RNA gene was subjected to PCR and denaturing gradient gel electrophoresis (DGGE), sequencing and phylogenetic analysis. The sequences found in this study clustered in 10 discrete sequence groups, seven belonging to Glomus, two to Scutellospora and one to Diversispora. A negative correlation was observed between soil mineral nitrogen and the number of AMF sequence groups in the roots. The frequency of occurrence of AMF in roots decreased dramatically between June and September. No plant-host specificity could be detected.  相似文献

Aims:  Some fungi present on the surface of grapes may have a negative effect on the quality of wine. The aim of this study was to evaluate PCR-denaturing gradient gel electrophoresis (PCR-DGGE), for the establishment of fungal community profiles from grapes, in order to monitor fungi potentially involved in wine defects.
Methods and Results:  A fragment of the β-tubulin gene was amplified from filamentous fungi and yeasts described from grapes and analysed using two different denaturing gradient gels to constitute a reference database. The use of β-tubulin sequences instead of ITS rDNA in PCR-DGGE showed a progress in the discrimination of these fungal species but comigration problems were still observed. The technique was then applied on grape samples. The profiles counted up to 10 bands of which half corresponded to species which were not recorded in the reference database.
Conclusion:  PCR-DGGE represents a useful tool to compare environmental samples for the study of the dynamics of fungal communities, but comigrations represent a limit in its use to describe the species present.
Significance and Impact of the Study:  A better knowledge of the fungal diversity on grapes, particularly species responsible for wine defect, is necessary to develop accurate molecular detection tools.  相似文献

以泸州老窖1、50、100和400年窖泥为研究对象,采用变性梯度凝胶电泳(DGGE)研究浓香型白酒窖泥放线菌的群落结构及其多样性。DGGE图谱显示,除1年样品外,其余窖底泥多样性指数(H)均低于同窖龄窖壁泥,但均匀度指数(EH)较高。不同窖池相同部位窖泥的群落结构变化趋势为:随窖龄的延长,窖壁泥H值逐渐上升,为1.74—2.28;窖底泥下降,为1.73—2.07。EH值均为波动下降,分别在0.986—0.991和0.971—0.994之间。窖底泥相似性系数(SC)逐渐上升,为0.46—0.82;窖壁泥为0.31—0.62。DGGE条带测序结果显示,窖泥放线菌归于Olsenella、Atopobium、Streptomyces和Corynebacterium 4个属。Olsenella和Atopobium属为共有的优势属,且在窖壁泥中的优势度(di)均随窖龄延长而降低,在窖底泥中升高。实验结果表明,浓香型白酒窖泥蕴藏着丰富的放线菌资源,群落结构和多样性存在差异,菌群演替呈现一定规律性。  相似文献

Refuse-derived fuels (RDF) pellets manufactured in Japan have been reported to contain a relatively high number of viable bacterial cells, and these bacteria generated a large amount of hydrogen gas during fermentation under wet conditions. In this study, we compared hydrogen gas generation from RDF pellets manufactured in Japan and in Germany and found that a large amount of hydrogen gas was generated from the Japanese RDF pellets but not from the German ones. This difference can be explained by the absence and presence of a biodegradation process before molding of raw garbage into RDF pellets. That is, the German process includes a biodegradation (or biological drying) process with forced aeration for a week, and this appears to reduce BOD in the garbage. Denaturing gradient gel electrophoresis analysis of 16S rRNA gene followed by DNA sequencing indicated that microbiotas of the RDF pellets manufactured in Japan and in Germany were very different.  相似文献

When adenosine (Ia) was allowed to react with 2-methylthio-4H-l,3,2-benzodioxaphosphorin-2-oxide (MTBO) (II), a new phosphorylating agent, in the presence of cyclohexylamine, adenosine 5′-S-methyl phosphorothiolate (IVa) and adenosine 2′: 3′-cyclic phosphate (Va) were obtained.

Ribonucleoside 5′-S-methyl phosphorothiolates (IV) were selectively synthesized directly from borate complexes of ribonucleosides (VI) and MTBO in the presence of cyclohexylamine, followed by removal of metaboric acid by co-distillation with methanol, in 68~86% yields.

Under anhydrous conditions, the phosphorothiolates (IV) were cyclized to give ribonucleoside 3′: 5′-cyclic phosphates (X) by iodine oxidation. Five 3′: 5′-cyclic nucleotides including cyclic AMP were synthesized by this procedure in 38~64% yields after purification.  相似文献

To study the structure of microbial communities in the biological hydrogen production reactor and determine the ecological function of hydrogen producing bacteria, anaerobic sludge was obtained from the continuous stirred tank reactor (CSTR) in different periods of time, and the diversity and dynamics of microbial communities were investigated by denaturing gradient gel electrophoresis (DGGE). The results of DGGE demonstrated that an obvious shift of microbial population happened from the beginning of star-up to the 28th day, and the ethanol type fermentation was established. After 28 days the structure of microbial community became stable, and the climax community was formed. Comparative analysis of 16S rDNA sequences from reamplifying and sequencing the prominent bands indicated that the dominant population belonged to low G+C Gram-positive bacteria (Clostridium sp. andEthanologenbacterium sp.), β-proteobacteria (Acidovorax sp.), γ-proteobacteria (Kluyvera sp.), Bacteroides (uncultured bacterium SJA-168), and Spirochaetes (uncultured eubacterium E1-K13), respectively. The hydrogen production rate increased obviously with the increase ofEthanologenbacterium sp.,Clostridium sp. and uncultured Spirochaetes after 21 days, meanwhile the succession of ethanol type fermentation was formed. Throughout the succession the microbial diversity increased however it decreased after 21 days. Some types ofClostridium sp.Acidovorax sp.,Kluyvera sp., and Bacteroides were dominant populations during all periods of time. These special populations were essential for the construction of climax community. Hydrogen production efficiency was dependent on both hydrogen producing bacteria and other populations. It implied that the cometabolism of microbial community played a great role of biohydrogen production in the reactors.  相似文献

为了研究生物造粒流化床污水处理反应器颗粒污泥中微生物群落结构及其动态变化,分别从生物造粒流化床10、60、110cm处取颗粒污泥,通过细胞裂解直接提取颗粒污泥细菌基因组DNA。以细菌和古细菌16S rRNA基因通用引物530F/1490R,对活性污泥中提取的细菌基因组DNA进行PCR扩增,长约1kb的PCR扩增产物纯化后经变性梯度凝胶电泳(DGGE)分离,获得微生物群落的DNA特征指纹图谱。结果显示,生物造粒流化床反应器颗粒污泥中的微生物群落非常丰富,在10cm处微生物的种属达到23种,60cm处为21种,110cm处为20种;生物造粒流化床不同高度都有一些各自的特有种属和共有种属,反应器不同高度的微生物群落演替不明显,微生物群落相似性为83.1%,群落结构较为稳定。  相似文献

Exotic invasive plants can show strong plant–soil feedback responses, but little is known about time scales for significant changes in soil microbial communities to occur after invasion. Previous work has suggested that plant invasions can modify arbuscular mycorrhizal (AM) fungal community structure. However, there is a lack of understanding about how long it takes for these changes to develop. To test this we investigated temporal changes in AM fungal communities colonising the invasive plant Vincetoxicum rossicum (Apocynaceae). We hypothesised that AM fungal community structure would change in a particular direction during the invasion process. We collected soil from two sites with a long history of invasion by this plant, with each site having paired invaded and uninvaded plots. Soil from these plots was used in a glasshouse experiment to characterise AM fungal community structure in the roots of V. rossicum at different times throughout a simulated growing season. AM fungal community structure differed between invaded and uninvaded plots. However, contrasting with our hypothesis, AM fungal communities colonising V. rossicum growing in soil from uninvaded plots did not change towards those in plants growing in previously invaded soil. Our data suggest that changes to AM fungal communities in the presence of V. rossicum require longer than the first growing season after establishment to develop.  相似文献

对筛选到的1组稻秆腐解复合菌系RSS-4,以腐解产物的pH值、酶活及稻秆失重率为指标,研究了该复合菌系的性质稳定性和功能稳定性;并用变性梯度凝胶电泳法(DGGE)分析了该复合菌系的菌种组成稳定性。结果表明,多代继代培养过程中各代复合菌系腐解产物的pH值变化趋势基本一致,pH值从起始的6.72左右迅速升至最高点7.75,然后逐渐下降并稳定在7.20;多代继代培养后复合菌系各代之间的酶活动态变化及其对稻秆腐解能力的差异均很小。在连续继代培养的过程中,DGGE图谱揭示第15~23代复合菌系之间的菌种组成没有明显差异。  相似文献

Ectomycorrhizal fungal communities have been characterized in a number of ways. Here we compare colonized root-tip and mycelia views of an ectomycorrhizal fungal community. Ectomycorrhizal fungi, both as mycelia and colonized root tips, were identified in soil samples taken from a pine plantation. We determined that for some ectomycorrhizal fungal species multiple root tips from a single soil sample were not independent. Therefore in the comparison of root-tip and mycelia views, we considered species to be present or absent from each soil sample irrespective of the number of root tips colonized by the species. We observed 39 ectomycorrhizal fungal species in total, but 12 were observed exclusively as mycelia and 11 exclusively colonizing root tips. The relative frequencies of 10 species occurring as both mycelia and root tips were not independent of the method of observation. Our results suggest that ectomycorrhizal fungal species differ in their spatial distributions on root tips, and that root-tip and mycelia views of the community are different.  相似文献
