Large differences in composition were found between xylem sapcollected from Actinidia chinensis (Chinese gooseberry or kiwifruit) as bleeding sap and sap collected by vacuum extraction.A comparison of saps collected by the two methods has littlemeaning, however, unless the position on the plant from whichsap was collected and the prior treatment of the plant are specified.Furthermore the composition of bleeding sap changes rapidlywith time, probably because of marked gradients in concentrationof individual solutes in the xylem sap from the base to thetop of the plant. Contamination of vacuum-extracted sap by cellularcontents was shown to be insignificant. Sap collected as bleeding sap and by vacuum extraction are ofsomewhat different origins. It would be difficult to predictthe composition of bleeding sap simply from a knowledge of vacuum-extractedsap: it may be similarly unwise to predict the composition ofthe transpiration stream from that of vacuum extracted sap. Seasonal variation was followed in the content of total a-aminoacids, arginine, calcium, total carbohydrate, ß-galactosidaseactivity, magnesium, nitrate, phosphatase activity, phosphateand sulphate in vacuum-extracted xylem sap from Actinidia chinensisvar. hispida, the Chinese gooseberry or kiwifruit. There wasa marked increase in the concentration of most sap componentsjust prior to leaf emergence followed by a rapid decrease afterthe leaves had expanded. Experiments with excised extensionshoots showed that much of this spring-time increase in concentrationsof sap components was due to mobilization of nutrients withinthe shoot itself. Sap from the trunk and the older brancheschanged less in composition than did sap from the younger partsof the plant. The amplitude and direction of trends in concentrationof sap from the different parts of the plant varied with nutrientand with time. Analysis of vacuum-extracted xylem sap collectedduring periods of rapid transpiration from early summer onwardsgives a reliable indication of the composition of the transpirationstream. Bleeding sap of Actinidia chinensis and Betula populifolia andguttation fluidof Avena sativa were analysed for sugars, amino-acids,auxin, and certain enzymes. A wide range of amino-acids wasfound in all three. Auxin was not detected in the bleeding sap,but was present in Avena guttation fluid (5.1 µg IAA equivalent/l).‘IAA oxidase’, acid phosphatase, ribonuclease, deoxyribonuclease,and protease were detected in the bleeding sap and guttationfluid. The possibility that some of the substances found insap and guttation fluid are products of autolysing, differentiatingxylem cells in the roots is discussed.  相似文献

Chemical Composition of Bleeding Xylem Sap from Kiwifruit Vines   总被引:5,自引:0,他引:5  
A study of the chemical composition and charge balance was madeof bleeding xylem sap collected from excised one-year-old extensionshoots of healthy, Mn-deficient, Mn-toxic and Zn-deficient kiwifruitvines (Actinidia deliciosa (A. Chev.) C. F. Liang et A. R. Ferguson)immediately prior to leafburst. The exudates were analysed formacronutrient cations and anions, trace elements, amino acids,organic acids and sugars. Major charged species measured wereCa (13.3 mM), K (8.9 mM), Mg (5.6 mM), malate (12.5 mM) andphosphate (5.8 mM). Glutamine (12 mM) was the predominant Ncarrier identified, accounting for 58 per cent of the totalN followed by NO2-N (4.5 per cent), NH4+-N (3.5 per cent)and arginine-N (2.9 per cent). Approximately 22 per cent ofthe N was in a hydrolysable proteinaceous fraction comprisingmainly glutamine and glutamate. Eighteen free proteinaceousamino acids were idetified in sap, the most abundant being glutamine,glutamic acid, valine, isoleucine and phenylalanine. Computersimulation of the chemical composition predicted that in additionto hydrated cations, ion pairs formed between inorganic components(SO42–, HPO42–, H2PO4) and cations (Ca2+,Mg2+, Mn2+), plus metal-organic ligand complexes (Ca Malate,Zn Malate, FeCit, CuHis, CuGln) are important species involvedin translocation. The solubility product of hydroxyapatite wasexceeded in all exudates although in vitro precipitation wasnot observed. To achieve electroneutrality with the componentsmeasured, however, formation of precipitate precursors (hydroxyapatitenuclei) had to be assumed. Irregularities in Mn nutrition (butnot Zn) were clearly indicated by the elemental compositionof exudate suggesting the use of sap analysis as a possiblepre-season indicator of nutritional status for this species. Antitranspirant Activity in Xylem Sap of Maize Plants   总被引:18,自引:1,他引:17  
Xylem sap from unwatered maize plants was collected and testedfor antitranspirant activity. Two assays were used. These werea transpiration assay with detached wheat leaves and a stomatalbio-assay involving the direct microscopic observation of epidermisof Commelina communis. The reduction in transpiration of detached wheat leaves promotedby xylem sap could be duplicated almost exactly by the applicationof solutions of ABA of equivalent concentration to that foundin the xylem sap. Removal of virtually all the ABA from thexylem sap, using an immunoaffinity column, removed virtuallyall the antitranspirant activity in both assays. These results are discussed in the context of other resultswhich suggest the presence of as-yet unidentified inhibitorsin the xylem sap of unwatered plants. We suggest that with maize plants at least, stomatal responsesto soil drying can be entirely explained by enhanced concentrationof ABA in the xylem stream. Removal of fruits and treatment with SADH (succinic acid 2,2-dimethylhydrazide) were used to change the balance between sources and sinks for photosynthates in Malus domestica‘Golden Delicious’. Sorbitol and sugar content were measured in bleeding sap, in xylem sap prepared by suction, and in 80% methanol extracts of the roots. Concentration as well as total amount of sorbitol in bleeding sap sampled in July and September were lower in fruiting than in defruited trees, and so was the total amount of sorbitol in xylem sap from the trunk. SADH treatment tended to reduce the sorbitol content. Sorbitol in root extracts, expressed as percentage of methanol (80%) insoluble root dry matter, was highest in fruiting trees; but root dry matter was here less than half of that in defruited trees. Sorbitol content in xylem sap as well as sorbitol + sugar percentage of roots showed a distinct maximum in late winter followed by a heavy decrease during spring. It is suggested that sorbitol in xylem sap during the growth season represents a return transport from the roots, and that the level of sorbitol in this return transport reflects, to a certain degree, the ratio between leaf area and assimilate demand by the tree.  相似文献

Vascular Development and Sap Flow in Apple Pedicels   总被引:4,自引:2,他引:2  
Xylem and phloem tissues of the pedicel of apple fruit increasein cross-sectional area throughout development. The increasein phloem is similar in the two cultivars examined (Cox's OrangePippin and Royal Gala) and reflects a steadily increasing phloemsap flow to the fruit. The increase in xylem tissue is due toa proliferation of non-conducting, structural, components sinceclose examination reveals no increase in the number of vesselelements from just after flowering onwards. The greater number,and the larger diameter, of the vessels in Cox's explains theinitially higher xylem conductance found in this cultivar. In vitro measurements of xylem exudation reveal a decline duringthe growing season in the xylem conductance of both cultivarsand an increasing proportion of fruit (particularly in Cox's)in which the xylem comes to be totally non-conducting. Thisobservation is in line with previously reported measurementsof xylem sap flow in vivo. Barley plants (Hordeum vulgare L. cv. Mazurka) were grown inaerated solution cultures with 2 mM or 8 mM inorganic nitrogensupplied as nitrate alone, ammonium alone or 1:1 nitrate+ammonium.Activities of the principal inorganic nitrogen assimilatoryenzymes and nitrogen transport were measured. Activities ofnitrate and nitrite reductases, glutamine synthetase and glutamatesynthase were greater in leaves than in roots but glutamatedehydrogenase was most active in roots. Only nitrate and nitritereductases changed notably (4–10 times) in response tothe different nitrogen treatments. Nitrate reductase appearedto be rate-limiting for nitrate assimilation to glutamate inroots and also in leaves, where its total in vitro activitywas closely related to nitrate flux in the xylem sap and wasslightly in excess of that needed to reduce the transportednitrate. Xylem nitrate concentration was 13 times greater thanthat in the nutrient solution. Ammonium nitrogen was assimilatedalmost completely in the roots and the small amount releasedinto the xylem sap was similar for the nitrate and the ammoniumtreatments. The presence of ammonium in the nutrient decreasedboth export of nitrate to the xylem and its accumulation inleaves and roots. Nitrate was stored in stem bases and was releasedto the xylem and thence to the leaves during nitrogen starvation.In these experiments, ammonium was assimilated principally inthe roots and nitrate in the leaves. Any advantage of this divisionof function may depend partly on total conversion of inorganicnitrogen to amino acids when nitrate and ammonium are givenin optimal concentrations. Summary Abscisic acid (ABA) has been identified by gas chromatography in xylem sap of the woody species apple, peach, willow, sugar maple, Tecomaria capensis and Actinidia chinensis. Arabidopsis halleri is a Cd hyperaccumulator; however, the mechanismsinvolved in the root to shoot translocation of Cd are not wellunderstood. In this study, we characterized Cd transfer fromthe root medium to xylem in this species. Arabidopsis halleriaccumulated 1,500 mg kg–1 Cd in the shoot without growthinhibition. A time-course experiment showed that the releaseof Cd into the xylem was very rapid; by 2 h exposure to Cd,Cd concentration in the xylem sap was 5-fold higher than thatin the external solution. The concentration of Cd in the xylemsap increased linearly with increasing Cd concentration in theexternal solution. Cd transfer to the xylem was completely inhibitedby the metabolic inhibitor carbonyl cyanide 3-chlorophenylhydrazone(CCCP). Cd concentration in the xylem sap was decreased by increasingthe concentration of external Zn, but enhanced by Fe deficiencytreatment. Analysis with 113Cd-nuclear magnetic resonance (NMR)showed that the chemical shift of 113Cd in the xylem sap wasthe same as that of Cd(NO3)2. Metal speciation with Geochem-PCalso showed that Cd occurred mainly in the free ionic form inthe xylem sap. These results suggest that Cd transfer from theroot medium to the xylem in A. halleri is an energy-dependentprocess that is partly shared with Zn and/or Fe transport. The composition of xylem sap and exudate from stem incisionsof Nicotiana glauca Grah. was compared in detail. Exudationfrom stem incisions occurred over a 5 min period in certainplants, enabling collection of 5–30 µl of sap. Therate of exudation showed an exponential decline. Exudate hada high dry matter content (170–196 mg ml–1) andhigh sugar (sucrose) levels. Xylem sap had a low pH (5.8) andexudate a pH of 7.9. Glutamine dominated the amino compoundsin xylem sap and exudate, and K+ was the major cation. Totalamino compounds in stem exudate reached 10.8 mg ml–1 whereasxylem sap contained much lower levels (0.28 mg ml–1).All mineral elements and amino compounds with the exceptionof calcium were more concentrated in stem exudate than in xylemsap. Sucrose was labelled heavily in stem exudate following pulsingof an adjacent leaf with 14CO2. A concentration gradient ofsugar (2.1 bar m–1) was recorded for stems. Levels ofsucrose, amino compounds and K+ ions in stem exudate showeda diurnal periodicity. Each commodity reached maximum concentrationat or near noon and minimum concentration about dawn. The evidencesuggests that exudate from stem incisions of N. glauca is arepresentative sample of solutes translocated in the phloem. The effect of two different copper conditions (deficiency andexcess) on the amino acid composition in B. carinata xylem sapwas analysed. When the Cu in the nutrient solution was increasedfrom 0.12 to 2.5 or 5 µM, the concentrations of histidine,threonine, glutamine, proline, methionine, and glycine weremuch increased in the xylem sap. When Cu was made deficientin the nutrient solution by decreasing its concentration from0.12 µM to 0 µM, nicotianamine, glutamine, and threoninewere significantly increased in the xylem sap. Aqueous solutionscontaining different Cu–amino acid complexes (simulatedsaps) responded in a specific way to the changes in pH, providinga signature that was used to evaluate, by comparison with thereal xylem sap, the importance of each amino acid in the xylemtransport of Cu. For a single amino acid, the free solutionCu2+ concentration versus pH titration curves for histidineand proline were the most similar to that for xylem under Cuexcess. Under Cu deficiency, this Cu concentration versus pHtitration curve appeared to be very similar to that for nicotianamine.It is concluded that increased Cu concentrations induced theselective synthesis of certain amino acids in the sap, of whichhistidine and proline are the most important. Under Cu deficiency,the concentration of nicotianamine was induced the most. Thefact that nicotianamine is induced under Cu starvation and notunder Cu excess, is in contrast to similar studies indicatingspecies-specific reactions. However, the induction of nicotianamineunder Cu starvation is in line with recent molecular data ofthe role of nicotianamine in intracellular Cu delivery. Seedlings (180-d-old) of Casuarina cunninghamianaM L., C. equisetifoliaMiq. and C. glauca Sieber inoculated with each of two differentsources of Frankia, were analysed for translocated nitrogenouscompounds in xylem sap. Analyses were also made on sap fromnodulated and non-nodulated plants of C. glauca grown with orwithout a range of levels of combined nitrogen. Xylem exudateswere collected from stems, roots, and individual nodules ofnodulated plants and from stems and roots of non-nodulated plants.While the proportional composition of solutes varied, the samerange of amino compounds was found in xylem sap from the threedifferent symbioses. In C. glauca asparagine was the major aminoacid in the root sap followed by proline, while in symbioticC. cunninghamiana arginine accounted for more than 25% of theamino compounds. Citrulline was the major translocated productfound in the stem exudate of symbiotic C. equisetifolia. Increasingconcentrations of ammonium nitrate in the nutrient solutionresulted in increasing levels of free ammonia and glutaminein xylem sap from stems of nodulated and non-nodulated C. glauca,but there was relatively little change in the prominent solutes,e.g. citrulline, proline, and arginine. The composition of nitrogenoussolutes in stem or root exudates of C. glauca was similar tothat of exudate collected from individual nodules and on thisbasis it was not possible to distinguish specific products ofcurrent N2 fixation in xylem. The main differences in N solutecomposition between the symbioses were apparently due to hostplant effects rather than nodulation or the levels of combinedN. Also, the data indicate that the use of the proportion ofN in sap as citrulline (or indeed any other organic N solute)could not be used as an index of nitrogen fixation.  相似文献

Kouchi, H. and Higuchi, T. 1988. Carbon flow from nodulatedroots to the shoots of soybean {Glycine max L. Merr.) plants:An estimation of the contribution of current photosynthate toureides in the xylem stream.–J. exp. Bot. 39: 1015–1023. Well-nodulated, water-cultured soybean plants were allowed toassimilate 13CO2 at a constant specific activity for 10 h andthe 13C-labelling of total carbon and ureides in xylem sap wasinvestigated. Labelled carbon appeared very rapidly in the xylem stream. Percentageof labelled carbon (relative specific activity, RSA) in xylemsap was 18% at 2 h after the start of 13CO2 assimilation andreached 53% at the end of the 10 h assimilation. The amountof labelled carbon exported from nodulated roots to the shootsvia the xylem during the 10 h labelling period accounted for33% of total labelled carbon imported into the nodulated roots.Ureides (allantoin and allantoic acid) in xylem sap were stronglydependent on currently assimilated carbon. The RSA of ureidesin xylem sap had reached 83% at the end of the assimilationperiod. Labelled carbon in ureides accounted for 51% of totallabelled carbon returned from nodulated roots to the shootsvia the xylem during the 10 h assimilation period. A treatmentwith 20 mol m–3 nitrate in the culture medium for 2 ddecreased the ureide concentration in the xylem sap slightly,but greatly decreased the RSA of ureides. By comparing the data with the results of analysis of the xylemsap of nodule-detached plants, it was concluded that the majorityof labelled carbon exported to the xylem stream from noduleswas in ureide form. A considerable amount of carbon was alsoreturned from roots to shoots via the xylem stream but it wasmore dependent on (non-labelled) carbon reserved in the roottissues. Plants of Helianthus annuus were grown in soil in pots suchthat approximately 30% of the root system protruded throughthe base of the pot. After 7 d further growth in aerated nutrientsolution, the attached, protruding roots were air-dried for10–15 min and thereafter surrounded with moist still air,in the dark, for 49 h, whilst the soil was kept at field capacity.The roots of the control plants remained in the nutrient solutionthroughout the experiment. This treatment rapidly reduced the water content of protrudingroots from 20.5 to 17.8 g g–1 dry mass (DM), which remainedless than that of the control roots for the rest of the experiment.This treatment also reduced root turgor and water potential.The abscisic acid (ABA) concentrations in the protruding roots,xylem sap and leaves of the treated plants increased significantly,compared to values recorded for control plants. In treated roots, the ABA concentration was significantly increased4 h after treatment, with a maximum of 4.4+0.1 nmol g–1(DM) after 25 h. The ABA concentration in the xylem sap of thetreated plants was significantly greater than in the controls25 h, 30 h, and 49 h after the partial drying of the roots,with a maximum concentration of approximately 970 pmol ABA cm-3at 49 h. Initially, the ABA concentration in the leaves was0.45 nmol g–1 (DM) which increased significantly to 1.1±0.1 nmol g–1 at 25 h, to 1.7±0.3 nmol g–1at 49 h. Leaf conductance was significantly less in plants with air-driedroots than in the controls 8 h after the start of the treatmentand thereafter. The water relations of the leaves of the treatedplants did not differ from those of the control plants. These results confirm previous reports that ABA is rapidly generatedin partially-dried and attached root systems and demonstratesa concomitant large increase in the ABA content of the xylemsap. It is suggested that partial dehydration of some of theroots of Helianthus annuus, increases ABA concentration in thetranspiration stream and decreases leaf conductance in the absenceof changes in leaf water status. As these responses were initiatedin free-growing roots the stimulus is independent of any increasesin soil shear strength that are associated with soil drying. The principal forms of amino nitrogen transported in xylem were studied in nodulated and non-nodulated peanut (Arachis hypogaea L.). In symbiotic plants, asparagine and the nonprotein amino acid, 4-methyleneglutamine, were identified as the major components of xylem exudate collected from root systems decapitated below the lowest nodule or above the nodulated zone. Sap bleeding from detached nodules carried 80% of its nitrogen as asparagine and less than 1% as 4-methyleneglutamine. Pulse-feeding nodulated roots with 15N2 gas showed asparagine to be the principal nitrogen product exported from N2-fixing nodules. Maintaining root systems in an N2-deficient (argon:oxygen, 80:20, v/v) atmosphere for 3 days greatly depleted asparagine levels in nodules. 4-Methyleneglutamine represented 73% of the total amino nitrogen in the xylem sap of non-nodulated plants grown on nitrogen-free nutrients, but relative levels of this compound decreased and asparagine increased when nitrate was supplied. The presence of 4-methyleneglutamine in xylem exudate did not appear to be associated with either N2 fixation or nitrate assimilation, and an origin from cotyledon nitrogen was suggested from study of changes in amount of the compound in tissue amino acid pools and in root bleeding xylem sap following germination. Changes in xylem sap composition were studied in nodulated plants receiving a range of levels of 15N-nitrate, and a 15N dilution technique was used to determine the proportions of accumulated plant nitrogen derived from N2 or fed nitrate. The abundance of asparagine in xylem sap and the ratio of asparagine:nitrate fell, while the ratio of nitrate:total amino acid rose as plants derived less of their organic nitrogen from N2. Assays based on xylem sap composition are suggested as a means of determining the relative extents to which N2 and nitrate are being used in peanuts.  相似文献

Collection of Xylem Sap at Flow Rate Similar to in vivo Transpiration Flux   总被引:3,自引:0,他引:3  
We have explored a method to collect xylem sap using a Scholanderpressure chamber for potted plants. Intact root system in potswhich fitted the pressure chamber was pressurised at a pneumaticpressure numerically equal to the absolute value of shoot waterpotential. The rate of xylem flow obtained from the stem stumpunder such pressure was found similar to the rate of transpirationbefore detopping. The rate of pressurised flow from detop-pedroots was linearly related to the pressure applied in both well-wateredand soil-dried plants. The osmotic concentration of the xylemsap was negatively related to the rate of volume flow, suggestingthe necessity to collect xylem sap at in vivo flow rate if originalsolute concentration is to be evaluated. The concentration ofABA in the xylem sap, however, did not show such a relationshipwith water flux. Both well-watered and soil-dried plants showedthe concentration of ABA in xylem sap largely stable with arange of volume flow rate, indicating a linear relationshipbetween the rate of ABA delivery through xylem and that of volumeflow. We also compared the concentrations of ABA in xylem sapsequentially collected from pressurised roots with that fromdetached shoots of the same plants. The concentration of ABAin the initial saps from shoots showed to be similar to thatfrom roots. However, a decrease in the concentration of ABAin the xylem sap collected from detached leaf or twig was observedwhen more volume of sap was collected, which might also be dependenton the plant species and the volume of xylem vessels concerned. Broccoli (Brassica oleracea var. italica cv Premium Crop) plantswere germinated in soil, transferred to vermiculite three weekslater and grown in the glasshouse, then either supplied continuouslywith boron levels ranging from 0.0 (deficient) to 12.5 (toxic)mg l–1 of nutrient solution or transferred from 2.5 to0.0 mg B l–1 at the initiation of inflorescence development.At commercial maturity the concentrations of various inorganicand organic solutes in phloem exudates and xylem saps, as wellas plant characteristics and elemental composition of the variousplant parts, were determined. Under deficient B levels leaf midrib and stem corkiness wereevident, together with signs of stem pith breakdown, symptomswhich resemble the initiation of the hollow stem disorder. Thexylem sap B concentration declined by about 50 % when B wasnot supplied or was removed after a period of adequate supply;the phloem concentration was unaffected. Also, the decreasingB concentration gradients from mature transpiring tissues toyoung developing sinks disappeared. Therefore, it is concludedthat when B is deficient, it is retranslocated from source leavesin the phloem stream supplying the developing leaves and inflorescence.The data also suggested that at toxic levels B undergoes extensivelateral transfer, probably from xylem to xylem, thereby enhancingthe B concentration of developing sinks. The B regime influenced dry-matter partitioning, retranslocationof some elements, and the synthesis and distribution of aminoacids and sugars, reflecting the general nature of B involvementin plant processes. Sycamore seedlings were grown with their root systems dividedequally between two containers. Water was withheld from onecontainer while the other container was kept well-watered. Effectsof soil drying on stomatal behaviour, shoot water status, andabscisic acid (ABA) concentration in roots, xylem sap and leaveswere evaluated. At 3 d, root ABA in the drying container increased significantly,while the root ABA in the unstressed container of the same plantsdid not differ from that of the control. The increase in rootABA was associated with the increase in xylem sap ABA and withthe decrease in stomatal conductance without any significantperturbation in shoot water status. At 7 d, despite the continuous increase in root ABA concentration,xylem sap ABA showed a marked decline when soil water contentwas depleted below 013 g g–1. This reduction in xylemsap ABA coincided with a partial recovery of stomatal conductance.The results indicate that xylem sap ABA is a function of rootABA as well as the flow rate of water from roots to shoots,and that this ABA can be a sensitive indicator to the shootof the effect of soil drying. This paper concerns nutrient concentrations in xylem sap collectedfrom above and below M.8, M.9, M.26 and M.7 dwarfing interstocks.The sap flowing from above an interstock had lower concentrationsof nutrients than that from the rootstock levels and these differencesincreased with the dwarfing effect of the interstock. There were indications that nutrient concentrations were enhancedbelow interstocks but were reduced in the sap flowing from theinterstocks into the scions. Generally these reductions weregreater the more dwarfing the effect of the interstock. Analyses of sap from above, mid-way along and below interstocksindicated that the changes in content were produced in, or closeto, the graft union between scion and interstock.  相似文献
