The excessive concentrations of toxic heavy metals in mine tailings and their very low N content make soil reclamation strategies by phytostabilization difficult. Our objective was to test if the symbiotic association between the legume Anthyllis vulneraria subsp. carpatica and the bacteria Mesorhizobium metallidurans originating from highly polluted mine tailings is able to increase N concentration in soils with contrasting Zn, Pb and Cd contents. Plants of A. vulneraria subsp. carpatica from a mine site and of a non-metallicolous subsp. praeopera from non-polluted soil were inoculated with a metallicolous or a non-metallicolous compatible Mesorhizobium spp. and grown on low and high heavy metal-contaminated soils. In contaminated soil, many nodules were observed when the metallicolous A. vulneraria was inoculated with its rhizobium species M. metallidurans, whereas the non-metallicolous A. vulneraria died after a few weeks regardless of the rhizobium inoculant. Eighty percent of the total nitrogen was derived from biological nitrogen fixation through the association between metallicolous A. vulneraria and the rhizobium grown on metal-enriched soil. The ability of the metallicolous A. vulneraria to develop a high nitrogen fixing potential opens new possibilities for promoting a low-maintenance plant cover and for stabilizing the vegetation in high heavy metal-contaminated soils.  相似文献

Background and aims

The association of the legume Anthyllis vulneraria and the grass Festuca arvernensis, was found to be very efficient for the phytostabilisation of highly multi-metal contaminated mine tailings. Our objective was to quantify the contribution of Anthyllis inoculated with its symbiotic bacteria Mesorhizobium metallidurans to the soil N pool and to test whether a starter nitrogen fertilization may improve symbiotic nitrogen fixation and the growth of Festuca.

Methods

Plants of Festuca and of Anthyllis inoculated with M. metallidurans were grown separately during eight months in pots filled with mine contaminated soil. Estimation of the N fluxes was realized using 15?N isotopic methods.

Results

Starter N fertilization (28 kg N ha?1) improved symbiotic N2 fixation and the growth of both species. Belowground N balance (N rhizodeposition – soil N uptake) of the non-fertilized Anthyllis at maturity was negative (?30.6 kg N ha?1). However, the amount of N derived from fixation, including above- and belowground parts, was 78.6 kg N ha?1, demonstrating the ability of this symbiotic association to improve soil N content after senescence.

Conclusions

i) soil N enrichment by the N2-fixing symbiotic association occurs after plant senescence, when decaying leaves and shoots are incorporated into the soil; ii) application of a starter fertilization is an efficient solution to improve phytostabilisation of highly contaminated sites.  相似文献

Aims

Phytomanagement of metal-polluted soils requires information on plant responses to metal availability in soil, but the predictability of metal accumulation in plant shoots and/or roots may be limited by metal toxicity and inherent shortfalls of the bioavailability assays.

Methods

We measured the uptake of Cd and Zn in a Salix smithiana clone grown in a pot experiment on soils with different characteristics and metal availabilities, determined by conventional soil single extractions (0.05 M Na2-EDTA and 1 M NH4NO3), soil solution obtained by centrifugation, and diffusive gradients in thin films (DGT). The Cd and Zn phytoavailability after a 2-year phytoextraction by willow was assessed by metal accumulation in the straw of the following barley culture.

Results

The phytoextraction efficiency was largest on a moderately polluted acid soil. Biomass and shoot Zn concentrations of S. smithiana were better predicted by DGT-measured Zn concentrations in soil solution (C DGT) than by Zn concentrations in the soil solution and extractable soil fractions. The weaker correlation for Cd in shoots may be related to relative Cd enrichment in the plant tissues. The metal accumulation in barley straw was unaffected or increased after a 2-year phytoextraction.

Conclusions

The shoot Zn and Cd removal of the tested Salix clone can be predicted by C DGT concentrations and is highest on either calcareous or moderately polluted acid soils. Single extraction with NH4NO3 and the C DGT value of Cd were not able to predict shoot Cd removal on the tested soils. Only shoot removal of Zn was predicted fairly well by the C DGT value.  相似文献

At present, no efficient technique is available for cleaning up soils which are highly polluted by heavy metals. Limiting the movement of pollutants out of the contaminated area by creating a dense and persistent plant cover appears to be the more reasonable approach. In this context, phytostabilization is a technique that uses metallicolous plants to revegetate highly polluted soils. This paper presents the results of an experiment performed in situ using metallicolous ecotypes of four plant species native to the Mediterranean French region, and grown in different combinations at a polluted site over two years. The soils were highly polluted with zinc, cadmium and lead. The aim was to find the best species mixture in terms of cover, biomass and duration. The four species used were the biennial legume Anthyllis vulneraria, two perennial grasses, Festuca arvernensis and Koeleria vallesiana, and the perennial forb Armeria arenaria. Mixtures which included A. vulneraria, and especially when in combination with F. arvernensis, showed the highest values of cover and biomass. After flowering, the biennial individuals of A. vulneraria disappeared but subsequent germination and survival of seedlings occurred abundantly under the two grasses. Mixtures with A. arenaria showed the lowest values of cover and biomass. Soil nitrogen increased in the plots with A. vulneraria as well as the concentration of essential nutrients (N P K) in the aerial parts of the two grasses. In contrast, the concentration of metals (Zn Pb Cd) decreased in the aboveground biomass of the latter in the same plots. These results show that reciprocal facilitation effects can act in heavy metal polluted environments, and that phytostabilization efforts in the Mediterranean region can be improved by using mixtures including local metallicolous legume and grass species.  相似文献

The paper presents results of morphometric analysis of Anthyllis vulneraria L. in Poland based on 828 herbarium specimens. This study investigates five taxa which have been recognized in Poland: Anthyllis vulneraria L. ssp. vulneraria, Anthyllis vulneraria ssp. polyphylla (DC.) Nyman, A. vulneraria ssp. maritima (Schweigg.) Corb., A. vulneraria ssp. alpestris Asch. et Graebn. and A. vulneraria ssp. carpatica (Pant.) Nyman as well as three intermediate taxa of presumably hybrid origin. Multivariate statistical analyses of 12 quantitative and 10 qualitative characteristics revealed conspicuous variation within A. vulneraria. Analysis of variance (ANOVA), principal coordinate analysis (PCoA), correspondence analysis (CA) and discriminant analysis (DA) proved the taxonomic usefulness of quantitative characteristics such as number of rosette leaves, calyx length, inflorescence length, the ratio of leaf length/width, bract length as well as qualitative characteristics such as stem hairiness, calyx colour, calyx indumentum, distances between stem leaves and form of rosette leaves. The issues concerning intermediate morphotypes occurring in Poland and their status are discussed. Finally, a key for determination of taxa within A. vulneraria in Poland and distribution maps based on the material examined are provided.  相似文献

Aim

Effects of elevated CO2 on N relations are well studied, but effects on other nutrients, especially micronutrients, are not. We investigated effects of elevated CO2 on response to variation in boron (B) availability in three unrelated species: seed geranium (Pelargonium x hortorum), barley (Hordeum vulgare), and water fern (Azolla caroliniana).

Methods

Plants were grown at two levels of CO2 (370, 700?ppm) and low, medium, and high B. Treatment effects were measured on biomass, net photosynthesis (Pn) and related variables, tissue nutrient concentrations, and B transporter protein BOR1.

Results

In geranium, there were interactive effects (P?<?0.05) of B and CO2 on leaf, stem, and total plant mass, root:shoot ratio, leaf [B], B uptake rate, root [Zn], and Pn. Elevated CO2 stimulated growth at 45?μM B, but decreased it at 450?μM B and did not affect it at 4.5?μM B. Pn was stimulated by elevated CO2 only at 45?μM B and chlorophyll was enhanced only at 450?μM B. Soluble sugars increased with high CO2 only at 4.5 and 45?μM B. High CO2 decreased leaf [B] and B uptake rate, especially at 450?μM B. Though CO2 and B individually affected the concentration of several other nutrients, B x CO2 interactions were evident only for Zn in roots, wherein [Zn] decreased under elevated CO2. Interactive effects of B and CO2 on growth were confirmed in (1) barley grown at 0, 30, or 1,000?μM B, wherein growth at high CO2 was stimulated more at 30?μM B, and (2) Azolla grown at 0, 10, and 1,000?μM B, wherein growth at high CO2 was stimulated at 0 and 10?μM B.

Conclusion

Thus, low and high B both may limit growth stimulation under elevated vs. current [CO2], and B deficiency and toxicity, already common, may increase in the future.  相似文献

Background and aims

Adequate zinc (Zn) in maize (Zea mays L.) is required for obtaining Zn-enriched grain and optimum yield. This study investigated the impact of varying Zn fertilizer placements on Zn accumulation in maize plant.

Methods

Two pot experiments with same design were conducted to investigate the effect of soil Zn heterogeneity by mixing ZnSO4·7H2O (10 mg Zn kg?1 soil on an average) in 10–15, 0–15, 25–30, 0–30, 30–60 and 0–60 cm soil layers on maize root growth and shoot Zn content at flowering stage in experiment-1, and assessing effects on grain Zn accumulation at mature stage in experiment-2.

Results

In experiment-1, Zn placements created a large variation in soil DTPA-Zn concentration (0.3–29.0 mg kg?1), which induced a systemic and positive response of root growth within soil layers of 0–30 cm; and shoot Zn content was increased by 102 %–305 % depending on Zn placements. Supply capacity of Zn in soil, defined as sum of product of soil DTPA-Zn concentration and root surface area at different soil layers, was most related to shoot Zn content (r?=?0.82, P?<?0.001) via direct and indirect effects according to path analysis. In experiment-2, Zn placements increased grain Zn concentration by up to 51 %, but significantly reduced the grain Zn harvest index from 50 % by control to about 30 % in average.

Conclusion

Matching the distribution of soil applied Zn with root by Zn placement was helpful to maximize shoot Zn content and grain Zn concentration in maize.  相似文献

Aim

This study presents a micrometre-scale map of the elemental distribution within roots and surrounding sediment of Halimione portulacoides of a contaminated salt marsh in the Tagus estuary.

Methods

Microprobe particle induced X-ray emission analysis was performed in sediment slices containing roots with tubular rhizoconcretions attached to host sediments.

Results

Strong concentration gradients were found particularly in the inner part of rhizoconcretions adjacent to the root wall. Local enrichment was observed in sediment interstices with Fe precipitates and other associated elements. A maximum of 55 % of Fe was measured near the concretion–root interface, with a decrease to <5 % in the host sediment. Maximum concentrations of P (3 %), As (1,200 μg g?1) and Zn (3,000 μg g?1) were registered in concretions, one order of magnitude above the values of the host sediment. The elemental concentration profiles across roots showed that the epidermis was an efficient selective barrier to the entrance of elements. Fe and As were retained in the epidermis. The highest Cu and Zn concentrations were also observed in the epidermis. However, the concentrations of Mn, Cu and Zn increased in the inner root.

Conclusions

As and Fe were mostly retained in the concretion, whereas P, Mn, Cu and Zn were mobilised by the root.  相似文献

The P SAG12 -ipt gene was transferred to miniature rose, as the first woody species, resulting in increased ethylene resistance due to specific up-regulation of the ipt gene under senescence promoting conditions.

Abstract

Transgenic plants of Rosa hybrida ‘Linda’ were obtained via transformation with Agrobacterium tumefaciens strain harboring the binary vector pSG529(+) containing the P SAG12 -ipt construct. A. tumefaciens strains AGL1, GV3850 and LBA4404 (containing P35S-INTGUS gene) were used for transformation of embryogenic callus, but transgenic shoots were obtained only when AGL1 was applied. The highest transformation frequency was 10 % and it was achieved when half MS medium was used for the dilution of overnight culture of Agrobacterium. Southern blot confirmed integration of 1–6 copies of the nptII gene into the rose genome in the tested lines. Four transgenic lines were obtained which were morphologically true-to-type and indistinguishable from Wt shoots while they were in in vitro cultures. Adventitious root induction was more difficult in transgenic shoots compared to the Wt shoots, however, one of the transgenic lines (line 6) was rooted and subsequently analyzed phenotypically. The ipt expression levels were determined in this line after exposure to exogenous ethylene (3.5 μl l?1) and/or darkness. Darkness resulted in twofold up-regulation of ipt expression, whereas darkness combined with ethylene caused eightfold up-regulation in line 6 compared to Wt plants. The transgenic line had significantly higher content of chlorophyll at the end of the treatment period compared to Wt plants.  相似文献

Background and aims

Acacia cyclops is an invasive species within Mediterranean ecosystems, characteristically low in soil nutrients. Thus associations with nitrogen-fixing bacteria (NFB) and arbuscular mycorrhiza (AM) may provide an advantage to these legumes. This study investigated the role of AM and NFB in the growth and nutritional physiology of A. cyclops.

Methods

Seedlings were inoculated with?naturally occurring?NFB, Glomus mosseae or both, and grown under glasshouse conditions for 5 months. Plants were cultivated in sand and supplied with a 20 % strength nutrient solution.?Xylem sap nutrients, photosynthetic rates, biomass and chemical compositions, were recorded.

Results

The dual inoculation decreased the colonization of both symbionts, compared to a single symbiosis with either symbiont. Despite low colonization levels, the dual symbiosis increased host biomass and relative growth rates. This was associated with increased photosynthetic rates and enhanced nutrition. Additionally, dual symbiotic plants had enhanced N and P acquisition and utilization rates. Xylem sap analysis showed higher levels of NH 4 + being exported from the roots to the shoots in the dual symbiotic plants compared with other treatments.

Conclusions

These findings suggest the dual symbiosis is an important factor in the growth and development of A. cyclops under nutrient limiting conditions.  相似文献

Genetic locus for tetralocular ovary ( tet - o ) in Brassica rapa was identified and it was shown that the number of locules and width of silique are associated.

Abstract

Brassica rapa is a highly polymorphic species containing many vegetables and oleiferous types. An interesting group of oleiferous types is the yellow sarson group (subspecies trilocularis) grown mostly in eastern India. This group contains lines that have bilocular ovaries, a defining trait of Brassicaceae, but also lines that have tetralocular ovaries. Yellow sarson lines commonly have high silique width which is further enhanced in the tetralocular types. We mapped the locus influencing tetralocular ovary in B. rapa using three mapping populations (F2, F6 and F7) derived from a cross between Chiifu (subspecies pekinensis, having bilocular ovary) and Tetralocular (having tetralocular ovary). QTL mapping of silique width was undertaken using the three mapping populations and a F2 population derived from a cross between Chiifu and YSPB-24 (a bilocular line belonging to yellow sarson group). Qualitative mapping of the trait governing locule number (tet-o) in B. rapa mapped the locus to linkage group A4. QTL mapping for silique width detected a major QTL on LG A4, co-mapping with the tet-o locus in bilocular/tetralocular cross. This QTL was not detected in the bilocular/bilocular cross. Saturation mapping of the tet-o region with SNP markers identified Bra034340, a homologue of CLAVATA3 of Arabidopsis thaliana, as the candidate gene for locule number. A C → T transition at position 176 of the coding sequence of Bra034340 revealed co-segregation with the tetralocular phenotype. The study of silique related traits is of interest both for understanding evolution under artificial selection and for breeding of cultivated Brassica species.  相似文献

Aims and background

We previously compared metallicolous (M) and non-metallicolous (NM) populations of Noccaea (=Thlaspi) caerulescens, Silene vulgaris, and Matthiola flavida for their abilities to tolerate and (hyper)-accumulate lead (Pb) in hydroponics. In the present study we aimed 1) to check the hyperaccumulation and tolerance abilities of these populations in controlled experiments using metalliferous soils, 2) to test the M. flavida M population for Zn and Cd hypertolerance in hydroponics.

Methods

Plants were grown in hydroponics and fertilized metalliferous substrates, collected from a Zn/Pb smelter sinter deposit near Plombières, Belgium (low pH, low Ca), and a tailing of the Irankouh Zn/Pb mine, Iran (high pH, high Ca). Metal tolerance was assessed from root growth inhibition in hydroponics, or mortality, stunting or chlorosis in the experiments with soil.

Results

Metallicolous M. flavida did not show hypertolerance or hyperaccumulation of Cd or Zn in hydroponics. Only one of the N. caerulescens M populations and the native S. vulgaris M population were able to grow in Plombières soil, whereas the others stopped growing or died within 40?days. All the populations survived and maintained growth for 40?days in Irankouh soil. When grown in Irankouh soil, the M population of M. flavida hyperaccumulated Pb. N. caerulescens hyperaccumulated Zn from Plombières soil, but not from Irankouh soil.

Conclusions

The M. flavida M population is non-Pb-hypertolerant. It hyperaccumulates Pb from Irankouh soil, but not from Pb-amended nutrient solution. N. caerulescens does not hyperaccumulate Zn from the calcareous Irankouh soil.  相似文献

Background and aims

The aim was to investigate whether different Cd salts in the nutrient solution of the Cd/Zn hyperaccumulator Noccaea (Thlaspi) praecox alter leaf Cd distribution and Cd ligand environment, and plant fitness.

Methods

Plants were grown for 8 weeks with 100/300 μM CdCl2 or CdSO4. Leaf biomass, and total chlorophyll, anthocyanin, Cd, Cl, S and P concentrations were monitored. Cd localisation and ligand environment in leaves were analysed using quantitative synchrotron-based micro-X-ray fluorescence imaging, and Cd K-edge X-ray absorption fine structure and Cd L3-edge micro-X-ray absorption near-edge structure measurements.

Results

Cd uptake and plant fitness were comparable for CdCl2 and CdSO4 treatments, and depended on applied Cd concentration. In all treatments, Cd preferentially accumulated with high concentrations of Cl in vacuoles of large vacuolarised epidermal cells, bound mainly to oxygen-based (O)-ligands. In the mesophyll of CdCl2? treated plants, Cd was preferentially sequestered in vacuoles, while for CdSO4, Cd accumulated preferentially in the apoplast. In the symplast, O-ligands increased with increasing Cd concentrations; in the apoplast, sulphur-based (S)-ligands prevailed.

Conclusions

Cd partitioning between leaf mesophyll apoplast and symplast and the Cd ligand environment in N. praecox depend on the Cd salt type and concentration added to the nutrient solution.  相似文献

Background and aims

Selenium is an essential micro-nutrient for animals, humans and microorganisms; it mainly enters food chains through plants. This study proposes to explore effect of inorganic Se forms on its uptake and accumulation in Zea mays.

Methods

Zea mays was grown in a controlled-atmosphere chamber for 2 weeks in a hydroponic solution of low-concentration selenium (10 μg/L (i.e.0.12 μM) or 50 μg/L (i.e. 0.63 μM) of Se). For each concentration, four treatments were defined: control (without selenium), selenite alone, selenate alone and selenite and selenate mixed.

Results

At low concentrations, selenium did not affect the biomass production of Zea mays. However, for both concentrations, Se accumulation following a selenite-only treatment was always higher than with selenate-only. Moreover, in the selenate-only treatment, Se mainly accumulated in shoots whereas in the selenite-only treatment, Se was stocked more in the roots. Interactions between selenate and selenite were observed only at the higher concentration (0.63 μM of selenium in the nutrient solution).

Conclusions

Se form and concentration in the nutrient solution strongly influenced the absorption, allocation and metabolism of Se in Zea mays. Selenate seems to inhibit selenite absorption by the roots.  相似文献

Aims

This study examined the effect of elevated CO2 on plant growth, root morphology and Cd accumulation in S. alfredii, and assessed the possibility of using elevated CO2 as fertilizer to enhance phytoremediation efficiency of Cd-contaminated soil by S. alfredii.

Methods

Both soil pot culture and hydroponic experiments were carried out to characterize plant biomass, root morphological parameters, and cadmium uptake in S. alfredii grown under ambient (350 μL L?1) or elevated (800 μL L?1) CO2.

Results

Elevated CO2 prompted the growth of S. alfredii, shoot and root biomass were increased by 24.6–36.7% and 35.0–52.1%, respectively, as compared with plants grown in ambient CO2. After 10 days growth in medium containing 50 μM Cd under elevated CO2, the development of lateral roots and root hairs were stimulated, additionally, root length, surface area, root volume and tip number were increased significantly, especially for the finest diameter roots. The total Cd uptake per pot was significantly greater under elevated CO2 than under ambient CO2. After 60 d growth, Cd phytoextraction efficiency was increased significantly in the elevated CO2 treatment.

Conclusions

Results suggested that the use of elevated CO2 may be a useful way to improve phytoremediation efficiency of Cd-contaminated soil by S. alfredii.  相似文献

Background and aims

Interactions between Cd and Zn occur in soils and plants but are inconsistent. This study examined how Cd/Zn interactions influence the growth of Carpobrotus rossii (Haw.) and the accumulation of Cd and Zn in plants.

Methods

Plants were grown in nutrient solutions containing 5–100 μM Zn and 0, 5 or 15 μM Cd. Plant growth and tissue concentrations were measured, and the speciation of Zn within the plant tissues determined using synchrotron-based X-ray absorption spectroscopy.

Results

There was an additive negative interaction between Cd and Zn on root growth. Only the highest level of Zn (100 μM) decreased Cd concentrations in root and shoot tissues (by 40–64%), whilst 100 μM Zn enhanced Cd translocation at 5 μM Cd but decreased it at 15 μM Cd. In contrast, both 5 and 15 μM Cd decreased Zn concentrations in root and shoot tissues but increased Zn translocation by 30–90%. This interaction was not associated with changes in Zn speciation within the plants, with most Zn associated with oxalate (48–87%).

Conclusions

The presence of Zn and Cd resulted in an additive negative effect on root growth, but an antagonistic pattern in their accumulation in shoots of C. rossii.

The critical level for SO 2 susceptibility of Populus × canescens is approximately 1.2 μL L ?1 SO 2 . Both sulfite oxidation and sulfite reduction and assimilation contribute to SO 2 detoxification.

Abstract

In the present study, uptake, susceptibility and metabolism of SO2 were analyzed in the deciduous tree species poplar (Populus × canescens). A particular focus was on the significance of sulfite oxidase (SO) for sulfite detoxification, as SO has been characterized as a safety valve for SO2 detoxification in herbaceous plants. For this purpose, poplar plants were exposed to different levels of SO2 (0.65, 0.8, 1.0, 1.2 μL L?1) and were characterized by visible injuries and at the physiological level. Gas exchange parameters (stomatal conductance for water vapor, CO2 assimilation, SO2 uptake) of the shoots were compared with metabolite levels (sulfate, thiols) and enzyme activities [SO, adenosine 5′-phosphosulfate reductase (APR)] in expanding leaves (80–90 % expanded). The critical dosage of SO2 that confers injury to the leaves was 1.2 μL L?1 SO2. The observed increase in sulfur containing compounds (sulfate and thiols) in the expanding leaves strongly correlated with total SO2 uptake of the plant shoot, whereas SO2 uptake rate was strongly correlated with stomatal conductance for water vapor. Furthermore, exposure to high concentration of SO2 revealed channeling of sulfite through assimilatory sulfate reduction that contributes in addition to SO-mediated sulfite oxidation to sulfite detoxification in expanding leaves of this woody plant species.  相似文献

Efficient Agrobacterium -mediated genetic transformation for investigation of genetic and molecular mechanisms involved in inflorescence architectures in Cornus species.

Abstract

Cornus canadensis is a subshrub species in Cornus, Cornaceae. It has recently become a favored non-model plant species to study genes involved in development and evolution of inflorescence architectures in Cornaceae. Here, we report an effective protocol of plant regeneration and genetic transformation of C. canadensis. We use young inflorescence buds as explants to efficiently induce calli and multiple adventitious shoots on an optimized induction medium consisting of basal MS medium supplemented with 1 mg/l of 6-benzylaminopurine and 0.1 mg/l of 1-naphthaleneacetic acid. On the same medium, primary adventitious shoots can produce a large number of secondary adventitious shoots. Using leaves of 8-week-old secondary shoots as explants, GFP as a reporter gene controlled by 35S promoter and hygromycin B as the selection antibiotic, a standard procedure including pre-culture of explants, infection, co-cultivation, resting and selection has been developed to transform C. canadensis via Agrobacterium strain EHA105-mediated transformation. Under a strict selection condition using 14 mg/l hygromycin B, approximately 5 % explants infected by Agrobacterium produce resistant calli, from which clusters of adventitious shoots are induced. On an optimized rooting medium consisting of basal MS medium supplemented with 0.1 mg/l of indole-3-butyric acid and 7 mg/l hygromycin B, most of the resistant shoots develop adventitious roots to form complete transgenic plantlets, which can grow normally in soil. RT-PCR analysis demonstrates the expression of GFP transgene. Green fluorescence emitted by GFP is observed in transgenic calli, roots and cells of transgenic leaves under both stereo fluorescence microscope and confocal microscope. The success of genetic transformation provides an appropriate platform to investigate the molecular mechanisms by which the various inflorescence forms are developed in Cornus plants.  相似文献

Background and aims

Whether root Zn uptake during grain filling or remobilization from pre-anthesis Zn stores contributes more to grain Zn in wheat is subject to an on-going debate. This study investigated the effects of N nutrition and post-anthesis Zn availability on the relative importance of these sources.

Methods

Durum wheat plants were grown in nutrient solution containing adequate Zn (0.5?μM) and three different N levels (0.5; 1.5; 4.5?mM). One third of the plants were harvested when they reached anthesis. One half of the remaining plants were grown to maturity with adequate Zn, whereas the Zn supply to the other half was discontinued at anthesis. Roots, straw and grains were harvested separately and analyzed for Zn and N.

Results

Depending on the N supply, Zn remobilization from pre-anthesis sources provided almost all of grain Zn when the Zn supply was withheld at anthesis; otherwise up to 100?% of grain Zn could be accounted for by Zn taken up post-anthesis. By promoting tillering and grain yield and extending the grain filling, higher N supply favored the contribution of Zn uptake to grain Zn accumulation.

Conclusion

Remobilization is critical for grain Zn accumulation when Zn availability is restricted during grain filling. However, where root uptake can continue, concurrent Zn uptake during grain development, favored by higher N supply, overshadows net remobilization.  相似文献
