目的:探讨脉冲电流经皮刺激肝区对运动性疲劳大鼠肝脏线粒体呼吸链酶复合物(I~IV)活性的影响。方法:选取健康8周龄Wistar雄性大鼠80只为实验对象,将实验动物随机分为安静对照组(A组)、疲劳对照组(B组)、疲劳前刺激组(C组)、疲劳后刺激组(D组),造模成功后分别在1、3、5周末分批处死。采用分光光度法测定1、3、5周各组动物的肝脏线粒体呼吸链酶复合物(I~IV)活性,大鼠肝脏线粒体蛋白定量用Bradfood蛋白定量法测定。结果:第3、5周末D组和B组、C组大鼠游泳力竭时间差异比较具有统计学意义(P0.05)。A组、C组及D组大鼠肝脏线粒体呼吸链酶复合物(I~IV)活性水平在第3、5周末均高于B组(P0.05)。D组大鼠肝脏线粒体呼吸链酶复合物(I~IV)活性水平在第3、5周末均高于C组(P0.05)。结论:脉冲电流经皮刺激运动性疲劳大鼠肝区可以提高其氧化呼吸链酶复合物(I~IV)的活性水平。  相似文献

为探讨合欢花对慢性应激大鼠生长和脑单胺类神经递质的影响,采用15只大鼠,设置了对照组、应激组和合欢花组3组实验。应激组和合欢花组均接受7天的应激刺激,之后合欢花组再灌胃合欢花10天。实验结束后,取3组大鼠的脑组织,用高效液相色谱法测定高香草酸(HVA)、去甲肾上腺素(NE)、多巴胺(DA)和5-羟色胺(5-HT)的含量。结果表明,应激组大鼠日增重显著低于对照组(P=0.011);而合欢花组大鼠的日增重极显著高于应激组(P=0.002)。应激组大鼠海马、纹状体和前额叶中的HVA含量与对照组相比,虽有升高的趋势,但无显著差异;两组间的NE、DA和5-HT也无显著差异。合欢花组大鼠海马中的HVA、DA含量明显高于应激组,而前额叶中的多巴胺和5-羟色胺,以及纹状体中的5-羟色胺均明显低于应激组。这表明合欢花对慢性应激引起的大鼠生长受抑有缓解作用,对其脑内单胺类神经递质有调节作用。  相似文献

为探讨贯叶连翘对慢性应激大鼠生长和脑单胺类神经递质的影响,用15只大鼠设置对照组、应激组和贯叶连翘组3组实验。应激组和贯叶连翘组均进行7天的应激刺激后,贯叶连翘组灌胃贯叶连翘10d。实验结束后,取3组大鼠的脑组织,用高效液相色谱法测定高香草酸(HVA)、去甲肾上腺素(NE)、多巴胺(DA)和5-羟色胺(5-HT)的含量。结果表明,应激组大鼠日增重明显低于对照组;而贯叶连翘组大鼠的日增重明显高于应激组。应激组大鼠海马、纹状体和前额叶中的HVA、NE、DA和5-HT与对照组间均无显著差异。贯叶连翘组大鼠纹状体中的DA含量明显高于应激组;而前额叶中的DA则明显低于应激组。因此,贯叶连翘对慢性应激引起的大鼠生长受抑有缓解作用,对其脑内单胺类神经递质有部分调节作用。  相似文献

目的建立大鼠运动疲劳模型,观察运动疲劳对大鼠各项生理、生化指标的影响。方法20只大鼠随机分为正常对照组和运动疲劳模型组,运动疲劳模型组大鼠每日按照方案进行锻炼。实验结束后大鼠检测相关指标:血清MDA含量和红细胞SOD活性,肝脏与骨骼肌MDA含量、SOD活性,骨骼肌线粒体游离钙离子含量,骨骼肌线粒体膜电位,下丘脑神经递质。电镜观察骨骼肌线粒体细微结构。结果运动疲劳模型组大鼠造模2周以后其血清、肝和骨骼肌MDA含量均有显著升高,红细胞和骨骼肌SOD活性均有显著降低,骨骼肌线粒体膜电位显著性降低,骨骼肌线粒体游离Ca2+含量有显著性降低,下丘脑GABA、5-HT含量有显著升高,下丘脑DA、ACh含量有显著性下降,电镜观察显示骨骼肌超微结构改变并以线粒体改变较为明显。结论大鼠跑台运动2周可造成运动疲劳模型,并造成大鼠骨骼肌线粒体损伤。  相似文献

目的:观察一次性力竭运动过程及恢复期大鼠纹状体细胞外液中多巴胺(DA)和5-羟色胺(5-HT)及其代谢物浓度的动态变化规律。方法:采用活体微透析结合毛细管电泳-激光诱导技术,连续观察清醒大鼠在一次性力竭运动过程及恢复期纹状体细胞外液中酪氨酸(Tyr)、5-HT、5-羟吲哚乙酸(5-HIAA)、色氨酸(Trp)和DA浓度的动态变化。结果:大鼠纹状体细胞外液中Trp、5-HT、5-HIAA水平运动初期均未见显著变化(P>0.05),运动后期、力竭及恢复期均显著高于安静水平(P<0.05,P<0.01);DA、Tyr水平在运动后期、力竭及恢复期显著高于安静水平(P<0.05,P<0.01);DA/5-HT运动初期显著升高(P<0.05,P<0.01),运动后期出现下降趋势,力竭前15 min降至最低点,而恢复期略有回升,但运动后期、力竭及恢复期与安静状态相比均无显著差异。结论:力竭运动过程中大鼠纹状体细胞外液中DA和5-HT的动态变化具有阶段性特征,运动疲劳过程中状体内DA和5-HT两种神经递质的代谢水平均显著增强,而其中以5-HT的作用占优。  相似文献

观察了支链氨基酸(BCAA)对大鼠运动能力和血清游离氨基酸代谢的影响。实验用21只雄性wistar大鼠,随机分为3组:正常组、游泳对照组和游泳补充BCAA组。2个运动组每天游泳训练1h,10天后游泳6h,观察补充BCAA对大鼠游泳运动能力和血清游离氨基酸水平的影响。实验结果表明,补充BCAA可明显提高大鼠游泳存活率,抑制血清中必需氨基酸、非必需氨基酸和总氨基酸水平升高,游泳运动后大鼠的血清中乳酸和LDH的升高幅度有所降低,抑制骨骼肌LDH活力的下降。说明补充BCAA可明显提高大鼠的运动能力,减少运动造成的蛋白质分解  相似文献

目的:观察低氧性肺动脉高压大鼠肺内5-HT1B受体的分布和表达变化,探讨低氧性肺动脉高压的形成机制.方法:40只健康雄性SD大鼠随机分为正常组(control)、低氧3周组(2w)、低氧4周组(4w)和低氧5周组(5w).除正常组外,其余3组大鼠分别在低氧环境中饲养3周、4周和5周.测定各组大鼠的平均肺动脉压力(mPAP)、右心室收缩压(RVSP)和右心室肥厚度[RV/(LV+S)%].应用免疫组织化学法观察大鼠肺组织中5-HT1B受体的分布和表达,Western blot法测定大鼠肺组织中5-HT1B受体的蛋白含量.结果:和正常组相比,低氧3周组大鼠的mPAP、RVSP和右心室肥厚度均显著升高(P均<0.05),并且随着低氧时间的延长而持续升高(P均<0.05).免疫组织化学结果显示:5-HT1B受体主要分布在正常大鼠肺动脉的内膜层,而平滑肌层中仅有少量表达:和正常组相比,低氧3周组大鼠肺动脉平滑肌层中5-HT1B受体的表达显著增多;随着低氧时间的延长,大鼠肺动脉平滑肌层中5-HT1B受体表达持续增多.Western blot结果表明,大鼠肺组织中5-HT1B受体的蛋白含量变化和免疫组织化学结果相一致.结论:低氧性肺动脉高压大鼠肺动脉中5-HT1B受体呈过度表达,这可能是低氧性肺动脉高压形成的分子机制之一.  相似文献

目的:探讨不同睡眠剥夺时间对大鼠认知功能的影响以及对下丘脑内单胺类神经递质去甲肾上腺素、多巴胺、五羟吲哚乙酸、五羟色胺的含量的影响。方法:32只健康雄性wistar大鼠随机分为4组,即96 h、120 h、144 h睡眠剥夺,正常对照组。利用睡眠剥夺箱建立大鼠SD模型,避暗穿梭法测试大鼠认知功能,高效液相电化学检测法测定下丘脑内单胺类神经递质含量。结果:大鼠避暗穿梭实验,与对照组比较,96 h、120 h组大鼠潜伏期显著缩短(P0.05);与对照组比较,各组大鼠下丘脑内NA含量均有下降(P0.05);与对照组比较,各组大鼠下丘脑内DA含量均显著下降,(P0.01),96 h、120 h、144 h组间比较,表现出含量逐渐减少的趋势;与对照组比较,各组5-HIAA含量均有上升,且120 h组明显高于其他各组(P0.05),其他组无显著性差异(P0.05);与对照组比较,各组5-HT含量均有升高,120 h、144 h组显著升高(P0.01),96 h组无显著性(P0.05)。结论:睡眠剥夺可以使大鼠中枢NA、DA含量下降,5-HIAA、5-HT含量升高,且随着睡眠剥夺时间的延长,变化更为明显,这可能是睡眠剥夺损害认知功能的原因之一。  相似文献

目的:探讨复方中药对运动大鼠中枢神经递质含量的影响,进一步认识中药提高运动能力和促进运动性疲劳恢复的作用机理。方法:选8周龄大鼠64只,随机分成服药组和对照组,服药组灌服中药煎剂8周。然后,每组再分成4个亚组分别于不同状态下断头处死,测其中枢递质含量。结果:服药组大鼠力竭运动时间极显著长于对照组(P〈0.01);安静时,除谷氨酸(GLU)含量服药组极显著高于对照组(P〈0.01)外、其余各指标无组间显著性差异;定量负荷后,服药组5-羟色胺(5-HT)、5-羟吲哚乙酸(5-HIAA)、弘氨基丁酸(GABA)、多巴胺(DA)含量和5-HT/5-HIAA显著低于对照组,GLU、GLU/GABA和DA/5-HT明显高于对照组;力竭即刻,服药组5-HT、GABA含量和5-HT/5-mAA显著低于对照组(P〈0.05),GLU含量、DA/5-HT和GLU/GABA显著高于对照组(P〈0.05);恢复12h。服药组5-HT含量和5-HT/5-HIAA极显著低于对照组(P〈0.01),GLU、DA、GABA含量和DA/5-HT明显高于对照组(P〈0.05)。结论:在大鼠运动至力竭性的过程中,复方中药制剂有明显抑制5-HT、5-HIAA、DA、GABA生成和促进GLU中枢递质合成的作用,其综合效应表现为兴奋性递质相对显著增多,使中枢神经兴奋性增强、明显延长大鼠运动时间和促进中枢疲劳的恢复。  相似文献

通过对6种藓类植物,即褶叶青藓(Brachythecium salebrosum(Web.et Mohr.)B.S.G.)、湿地匐灯藓(Plagiomnium acutum(Lindb.)Kop.)、侧枝匐灯藓(Plagiomnium maximoviczii(Lindb.)Kop.)、大凤尾藓(Fissidensnobilis Griff.)、大羽藓(Thuidium cymbifolium(Doz.et Molk.)B.S.G.)和大灰藓(Hypnum plumaeforme Wils.)嫩茎和老茎的石蜡切片和显微观察发现,同一藓类植株的嫩茎和老茎,茎结构稳定,不同种藓类植物茎横切面具有不同特征.植物体茎横切面形状、表层细胞的层数、细胞大小和细胞壁厚薄、皮层细胞大小和形状、中轴的有无以及比例等特征可以作为藓类植物的分科分类依据之一.  相似文献

The levels of endogenous phytohormones and respiratory rate in nine sorts of flowers such as Cymbidium faberi Rolfe, Nopalxochia ackermannii Kunth and others were investigated both at full bloom and senescence and meanwhile the effect of exogenous phytohormones on prolonging the blossoms and promoting ethylene production were tested. There is a high content of endogenous ethylene in all the long-lived flowere, about 3–16 folds higer than the short-lived ones. There is a high level of ABA at full blooming flowers of short-lived flowers, in which there is no or only some cytokinins in it, but the ratio of CTK (6BA+zeatin)/ABA is smaller(l.7). The endogenous ABA reached a much higher level at senescence in all nine sorts of flowers, so it is reasonable to consider that it is ABA which plays an important role of regulation in controlling flower's senescence. There is a much higher level of GA3 and zeatin in the long-lived flowers which is not demonstrated in the shortlived ones. The respiratory rate is one of the factors controtling the longevity of flowers, but it does not play a decided role. Application of 6BA and zeatin prolongs distinctly orchid’s longevity, however exogenous IAA through the promotive action on ethylene production, evidently extends the longevity of the flowers of the Nopalxochia ackermannii Kunth.  相似文献

Summary Embryogenic tissues of sugi (Cryptomeria japonica) were induced on a modified Campbell and Durzan (CD) medium containing 1 μM 2,4-dichlorophenoxyacetic acid (2,4-D) and 600 mg l−1 glutamine, and subcultured in the medium of the same composition for over 1 yr. This resulted in a mixed culture of embryogenic and non-embryogenic cells. When embryogenic cells were isolated and cultured independently, their capacity to form embryogenic aggregates was lost. Thus, the non-embryogenic cells present within a mixed culture system were essential to the formation of embryogenic aggregates. When embryogenic tissues were isolated and cultured independently on a high glutamine-containing (2400 mg l−1) medium, dry weights and endogenous levels of glutamine increased, and the tissue could generate a large number of embryogenic aggregates. Amino acid analysis of embryogenic and non-embryogenic cells from the maintenance culture indicated a higher level of glutamine was present in the latter. The high endogenous level of glutamine in the non-embryogenic portion of mixed cell masses may be the supplier of glutamine for maintaining the embryogenic property of the tissues.  相似文献

The review deals with study of enzymologic properties of a novel highly specific acetylcholinesterase substrate, N-(β-acetoxyethyl) piperidinium iodomethylate (“piperidylcholine”), and its 30 derivatives that were tested as effectors of cholinesterases of mammals and various species of Pacific squids. It was proven for the first time that responsible for specificity of action was structure of cyclic ammonium grouping of the alcohol part of molecule of the ester substrate. Analysis of specificity is performed based on enzymatic hydrolysis parameters—activity of catalytic center of cholinesterases and bimolecular constant of the reaction rate that are determined at optimal and low substrate concentrations. Among the specially synthesized group of thioester compounds there is revealed one more highly specific acetylcholinesterase substrate—N-(β-acetoxyethyl) piperidinium.  相似文献

Seed germination of an aurea mutant of tomato ( Lycopersicon esculentum Mill.) is promoted by continuous irradiation with red, far-red or long-wavelength far-red (758 nm) light as well as by cyclic irradiations (5 min red or 5 min far-red/25 min darkness). Far-red light applied immediately after each red does not change the germination behaviour. Seed germination of the isogenic wild-type, cv. UC-105, is promoted by continuous and cyclic red light while it is inhibited by continuous and cyclic far-red light and by continious 758 nm irradiation. Far-red irradiation reverses almost completely the promoting effect of red light. The promoting effect (in the aurea mutant) and the inhibitory effect (in the wild-type) of continuous far-red light do not show photon fluence rate dependency above 20 nmol m−2 s−1. It is concluded that phytochrome controls tomato seed germination throgh low energy responses in both the wild type and the au mutant. The promoting effect of continuous and cyclic far-red light in the au mutant can be attributed to a greater sensitivity to Pfr.  相似文献

Fluid collected from hatching eggs of Haemonchus contortus contained a lipase which hydrolysed 2-naphthyl laurate (about 0·7 μmol naphthol freed /h/106 eggs). The fluid also hydrolysed l-leucinamide (about 2·3 μmol leucine freed/h/106 eggs). The fluid when added to normal or heated eggs caused ‘hatching’. ‘Hatching’ also occurred in exsheathing fluid from infective juveniles and in a preparation of pancreatic lipase containing leucine aminopeptidase. A purified mammalian leucine aminopeptidase in combination with several different lipases did not attack egg shells.The ‘spontaneous’ hatching of eggs of H. contortus was strongly inhibited by 1,10-phenanthroline, 10?3M, and this inhibition was reversed by Zn2+. However, the inhibition of ‘hatching’ of eggs in externally applied hatching fluid, or the hydrolysis of leucinamide in hatching fluid was generally less marked.  相似文献
