Carrot and leek seed was osmotically primed in polyethylene glycol solution (273 g/kg water and 342 g/kg water respectively) for 10, 14 or 17 days before accelerated ageing for 0, 24, 48, 72 or 96 h. Priming reduced the germination time compared with non-primed seed. Accelerated ageing increased germination and emergence times and decreased percentage germination and emergence to a greater extent for the primed seeds than for non-primed seeds in both species. Primed and dried but non-aged seed from both species was stored at 10°C for 12 months. There was no loss of viability and improvements in germination time due to priming were maintained throughout the storage period for all the priming treatments in leek, and for the 10 and 14 day priming treatments in carrot. Carrot seed primed for 17 days lost some viability after 12 months storage compared with non-stored seed.  相似文献

Bean seed storage ability is of major interest for seed firms and depends, at least partially, on the effect of desiccation microclimate and desiccation rate on the mother plant; however, their effects are contradictory and not quantified. Therefore, our aims were (a) to predict seed desiccation rate from microclimatic variables at the pod level and (b) to measure the effects of seed desiccation rate and temperature on seed storage ability. For 4 years, beans (Phaseolus vulgaris) were sown at different dates in the field and once in a greenhouse. Pods having seeds at the beginning of desiccation stage were selected according to their colour and then dried in the field or under controlled conditions. In the field, pods were dried under both natural conditions and conditions modified either by defoliation or by water spraying. Under controlled conditions, pods were dried under a wide range of temperature and relative humidity combinations. Seed water content, temperature and relative humidity were measured throughout the desiccation phase. Storage ability was measured by a standard germination test after either 11 or 15 days of controlled deterioration. Seed desiccation rate was highly correlated to air vapour pressure deficit, which measures the combined effects of temperature and relative humidity on drying. With a logit transformation, deterioration test results showed a linear decrease in storage ability as seed desiccation rate increased for either controlled or field‐dried lots. However, for controlled lots desiccated at temperatures lower than 24°C, the lower the temperature, the lower the storage ability at a given desiccation rate. Moreover, the relationship between the results of the two deterioration periods differed between controlled and field‐dried lots. Finally, the high correlation between seed storage ability, desiccation rate and vapour pressure deficit in the field has given new insights for building a crop decision‐oriented model for optimising harvesting.  相似文献

Onion seeds were osmotically primed in polyethylene glycol solution (342 g/kg water) either for 14 days before accelerated ageing at 40°C. 18% m.c. for 0, 24, 48, 72 or 96 h, or for 10, 14 or 17 days after ageing. Priming improved the rate of germination compared with non-primed seed. Priming before ageing delayed the loss of viability due to ageing, but priming after ageing had no effect on viability. Primed and dried onion seed was stored for 18 months at 10°C, 9% m.c. with no effect on viability; improvements in germination rate due to priming were maintained over the storage period. Conductivity measurements of seed leachates were not a consistently reliable indicator of germination performance.  相似文献

Methyl methanesulphonate (MMS), applied on barley seeds, inhibits the seed respiration and thein vitro activity of cytochrome-oxidase and catalase, whereas doubles the activity of total dehydrogenases and has no influence on the activity of peroxidase. Storage of MMS-treated seeds for 10 days at 30% seed water content, thus under conditions favouring the recovery from the MMS-induced toxic and genetic effects, resulted in an enhancement of the cytochrome oxidase and catalase activity, which reached or exceeded the activity of the control. The activity of peroxidase, although increasing during storage, did not attain the value of the control and the activity of total dehydrogenases remained two times higher as compared to the control.  相似文献

Germinated seeds of five vegetable crops were stored at 1 °C in aerated water or humid air for periods of up to 15 days and then fluid drilled in the field. Viability of germinated seed was maintained in carrot and parsnip for a minimum of 15 days, salad onion for 9 days, lettuce for 6 days and cabbage for 3 days. When salad onion, lettuce and cabbage seeds were stored for longer than these times there was a reduction in the number of seedlings emerging. In species where storing seeds for up to 15 days caused a reduction in emergence an adjustment to the seed rate related to the length of storage can be made to achieve the desired population and still retain the advantages of fluid drilling pre-germinated seeds. This is possible because with one exception (an increase in the variation of salad onion stem diameters) the performance of plants from all stored germinated seeds after emergence was similar to plants from freshly germinated seeds.  相似文献

Germination of Archontophoenix alexandrae seeds and embryos were studied under gradient water content treatments throughout the seed development phases of maturation in 2005 to investigate seed desiccation tolerance and storage characteristics. During the maturation process, seed water content decreased gradually from55 DAF (days after flowering) to 70 DAF, and seeds reached the maximum dry-weight at 90 DAF. Seed germinability appeared after 60 DAF. Seeds germinated with a temperature range from15℃- 40℃ under alternating photoperiod (14 h light, 10 h dark, 12μmol m- 2s - 1 ), while the best germination percentage was obtained between 30℃- 35℃. A maximum germination capacity reached at 70 DAF. However, seed germination was greatly inhibited by light. Desiccation tolerance of seeds and embryos increasedgradually from 55 DAF to 90 DAF and reached the maximum at 90 DAF with a semilethal water content of 0.18 g/g ( seed) and 0.3 g/g ( embryo) respectively. Rapid dehydration maintained higher seed germination percentage than thatof slow dehydration when drying to the same water content. Seeds with without water content treatments failed to germinate after 1 month storage under - 18℃, whereas appropriate desiccation treatment prolonged seed longevity under 4℃, 10℃ and 15℃ storage temperatures. It revealed obviously the recalcitrant characteristics of Archontophoenix alexandrae seeds torage behaviour which are tolerant toward neither deep desiccation nor low temperatures.  相似文献

Alfalfa (Medicago sativa L. cv. Saranac) seed were soaked for 20 minutes in water, acetone, or methanol containing 10 or 50 mg/ml of oxamyl (Vydate L) or coated with a 2% aqueous cellulose solution containing the same amounts of oxamyl. Seed were analyzed for oxamyl by HPLC immediately after treatment and after 9 and 26 months of storage. Oxamyl content of alfalfa seed did not decline after 26 months of storage. The effects of seed treatment on growth of alfalfa and nematode control were examined using soils infested with Pratylenchus penetrans and Meloidogyne hapla. Germination was not affected by any of the seed treatments. Twenty-one days after sowing, the total growth of alfalfa seedlings grown from seed treated with 50 mg/ml of oxamyl in P. penetrans-infested soils had increased by 62% over controls. Nodulation per pot increased by as much as 267%, and the densities of P. penetrans per gram of root were reduced by as much as 73% compared to control plants. In M. hapla-infested soils, increases in plant growth (32%) and nodulation (71%) also occurred with oxamyl-treated seeds. Root gall reduction (86%) was also substantial due to oxamyl seed treatment.  相似文献

Background and Aims

Most priming studies have been conducted on commercial seed lots of unspecified uniformity and maturity, and subsequent seed longevity has been reported to both increase and decrease. Here a seed lot of Digitalis purpurea L. with relatively uniform maturity and known history was used to analyse the effects of priming on seed longevity in air-dry storage.

Methods

Seeds collected close to natural dispersal and dried at 15 % relative humidity (RH), 15 °C, were placed into experimental storage (60 % RH, 45 °C) for 14 or 28 d, primed for 48 h at 0, −1, −2, −5, −10 or −15 MPa, re-equilibrated (47 % RH, 20 °C) and then returned to storage. Further seed samples were primed for 2 or 48 h at −1 MPa and either dried at 15 % RH, 15 °C or immediately re-equilibrated for experimental storage. Finally, some seeds were given up to three cycles of experimental storage and priming (48 h at −1 MPa).

Key Results

Priming at −1 MPa had a variable effect on subsequent survival during experimental storage. The shortest lived seeds in the control population showed slightly increased life spans; the longer lived seeds showed reduced life spans. In contrast, seeds first stored for 14 or 28 d before priming had substantially increased life spans. The increase tended to be greatest in the shortest lived fraction of the seed population. Both the period of rehydration and the subsequent drying conditions had significant effects on longevity. Interrupting air-dry storage with additional cycles of priming also increased longevity.

Conclusions

The extent of prior deterioration and the post-priming desiccation environment affect the benefits of priming to the subsequent survival of mature seeds. Rehydration–dehydration treatments may have potential as an adjunct or alternative to the regeneration of seed accessions maintained in gene banks for plant biodiversity conservation or plant breeding.Key words: Digitalis purpurea, priming, re-drying, seed longevity, seed storage, ageing  相似文献

Changes in seed quality in pepper (Capsicum annuum L.) were monitored during seed development and maturation in two seasons. Seed quality was assessed by a number of different tests, but principally by determining seed storage longevity in laboratory tests and seedling growth in glasshouse tests. Mass maturity (defined as the end of the seed-filling phase) occurred 49–53 days after anthesis (DAA) in 1989 (varying among fruit layers) and 53 DAA in 1990 when seed moisture contents were 51–53%. The onset of both germinability and desiccation tolerance occurred either just before or at mass maturity. Maximum potential longevity (assessed by the value of the seed lot constant Ki) was achieved 63–65 DAA, i.e. not until 10–12 days after mass maturity (DAMM), in both years. Seedling dry weights in the glasshouse growth tests were maximal later still - for seeds harvested 17–21 DAMM in 1989 and 17 DAMM in 1990; the effects on seedling weight arose from differences in times from sowing to emergence (P < 0.005) among different seed harvests, with no significant differences in subsequent relative growth rates (P > 0.25). Seed priming reduced mean germination times for seeds harvested at all stages of development, but had little effect on germination capacity and potential longevity, and did not affect the pattern of changes in potential longevity during seed development and maturation. The results contradict the hypothesis that seed quality is maximal at the end of the seed-filling phase and that viability and vigour begin to decline immediately thereafter.  相似文献

The germination requirements and the basis of the optimal water content before and after cryopreservation were studied for ten endangered Brazilian species of Bromeliaceae. Constant and alternating temperature regimes were used to determine the best conditions for seed germination. The relationship between seed water content and relative humidity was evaluated using water sorption isotherms at 15 °C. Seeds were cryostored at four water contents (3, 5, 7 and 9%) and three storage periods (0, 180 and 365 days), and loss in viability and vigour were estimated. Fresh seeds of all species showed maximum germination in < 30 days at temperatures between 20 and 30 °C, indicating the absence of a physical/morphological dormancy. A sigmoidal relationship between seed water content and relative humidity was observed with no apparent differences in sorption characteristics among the species. The optimum water content for cryopreservation of most of these species was c. 7%. Ultra‐drying (3% seed water content) had a detrimental effect on seed viability and vigour. Our experiments suggested orthodox storage behaviour for all species of Bromeliaceae examined as they are able to survive desiccation and freezing. This study has shown the feasibility of ex situ conservation in seed cryobanks of endangered bromeliads from the Brazilian Atlantic Forest to support future reintroduction of these species in nature. © 2014 The Linnean Society of London, Botanical Journal of the Linnean Society, 2014, 176 , 567–576.  相似文献

The role of auxins in the morphogenesis of immature seeds of Medicago truncatula was studied, focusing on the transition from the embryo cell division phase to seed maturation. We analyzed seed development in vitro, by flow cytometry, and through the determination of the kinetics of seed fresh weight and size. Thus, seeds were harvested at 8, 10 and 12 days after pollination and cultured in vitro on a medium either without auxin or supplemented with indole‐3‐butyric acid (IBA) or naphthalene acetic acid (NAA) at 1 mg l?1. All parameters studied were determined every 2 days from the start of in vitro culture. The results showed that both auxins increased the weight and size of seeds with NAA having a stronger effect than IBA. We further demonstrated that the auxin treatments modulate the transition between mitotic cycles and endocycles in M. truncatula developing seed by favoring sustained cell divisions while simultaneously prolonging endoreduplication, which is known to be the cytogenetical imprint of the transition from the cell division phase to the storage protein accumulation phase during seed development.  相似文献

Effects of various chemical and physical factors on the germination of several seed lots of reed canary-grass (Phalaris arundinacea L.) have been studied. Germination at the optimum constant temperatures of 24 to 27°C was significantly stimulated by the following treatments: moist chilling in light, red light given during the first 3 days of imbibition, three 2-h periods at 12°C given during the second day of imbibition, ethylene, increased oxygen tension and soaking in aerated water for 4 days. Dry storage at 20–30°C had no effect on the germination ability of the seeds. No significant quantities of germination inhibitors were found either in water or methanol extracts of seed dispersal units. By comparing three cultivars with various degrees of seed dormancy, respiration measurements showed that there was a significant positive correlation between oxygen uptake prior to visible germination and germination capacity. Similarly, germination-stimulating treatment significantly enhanced oxygen uptake prior to visible germination.  相似文献

When a combination of hydrogen peroxide and hypochlorite was used to surface sterilize rice seeds, a 10(2)- to 10(4)-fold decrease in CFU was observed during the first 15 h after inoculation of the rice rhizosphere organism Burkholderia vietnamiensis TVV75. This artifact could not be eliminated simply by rinsing the seeds, even thoroughly, with sterile distilled water. When growth resumed, a significant increase in the frequency of rifampin- and nalidixic acid-resistant mutants in the population was observed compared to the control without seeds. This phenomenon was a specific effect of hypochlorite; it was not observed with hydrogen peroxide alone. It was also not observed when the effect of hypochlorite was counteracted by sodium thiosulfate. We hypothesized that the hypochlorite used for disinfection reacted with the rice seed surface, forming a chlorine cover which was not removed by rinsing and generated mutagenic chloramines. We studied a set of rifampin- and nalidixic acid-resistant mutants obtained after seed surface sterilization. The corresponding rpoB and gyrA genes were amplified and sequenced to characterize the induced mutations. The mutations in five of seven nalidixic acid-resistant mutants and all of the rifampin-resistant mutants studied were found to correspond to single amino acid substitutions. Hypochlorite surface sterilization can thus be a source of artifacts when the initial bacterial colonization of a plant is studied.  相似文献

研究了多毛坡垒(Hopea mollissima)种子在自然条件下的寿命及成熟度、温度、光照、土壤水分条件对多毛坡垒种子萌发的影响,结果表明:多毛坡垒种子适宜萌发的温度范围是20~30℃,最适宜温度为25℃;光照对多毛坡垒种子的萌发没有显著性影响,但在周期性光照条件下以25~30℃萌发结果最好,全黑暗条件下以20~25℃萌发结果最好;种子适宜萌发的土壤含水量为20%~60%,最适含水量为50%~60%;种子成熟后在树上长时间宿存明显降低了种子的发芽率和活力;在室内自然摊放条件下,10天后发芽率减半,16天后完全丧失发芽能力。研究认为,该物种对热带湿性季节性雨林原生境有很强的依赖性,目前多毛坡牟的濒危状态主要是由于滥砍乱伐和森林破坏造成的,由于其种子具有顽拗性种子的一些特点,日前该物种宜采取活体保仔的方法,以就地保护为主,活植物迁地保护为辅。  相似文献

Abstract Uniola paniculata (sea oats) rhizomes uprooted by hurricanes and deposited as wrack could be salvaged and replanted in dune restoration. To test this unexplored technique, percent tiller emergence was observed for 4 years from U. paniculata rhizomes replanted after submersion in seawater; air exposure of 0, 1, 3, 5, 7, and 11 days; freshwater rinse; and reburial in pots (watered) or on the beach (with or without supplemental water). In addition, U. paniculata rhizomes uprooted by Hurricane Georges were experimentally planted, and effects of soil salinity and moisture on emergence were tested in the greenhouse. Tiller emergence declined with increasing length of air exposure and decreasing size of rhizome. Tiller survival was enhanced by rainfall, rinsing with salt or fresh water during exposure and immediately after planting or supplemental beach watering. Although emergence was not reduced by soil salinity of 1,800 μS/cm, emergence was reduced by soil salinity of 5,800 μS/cm. Across the 4 years of the study tiller emergence from treated rhizomes varied considerably. U. paniculata rhizomes lost bud viability after 3–5 days of beach exposure, unless fresh water from rainfall, wet burlap storage, or watering was applied within 3 days. Bud viability was extended through 11 days when supplied with water. Thus, reburial within 3–11 days after a storm is a viable restoration technique.  相似文献

The effects of time of seed maturation and dry seed storage and of light and temperature requirements during seed incubation on final germination percentage and germination rate were assessed for the invasive shrub Prosopis juliflora (Sw.) D.C., grown under desert environmental conditions of the United Arab Emirates (UAE). Seeds were collected from Fujira on the northern coast of the UAE at different times during the growing seasons (autumn, winter and spring) and were germinated immediately and after 8 months of dry storage under room temperature (20±3 °C). Seeds were germinated at three temperatures (15, 25 and 40 °C) in both continuous light and darkness. The results showed significant effects for time of seed collection, seed storage, light and temperature of seed incubation and many of their interactions on both germination percentage and rate. Fresh seeds matured during autumn and winter germinated significantly greater at 40 °C and in light than at lower temperatures and in dark. Storage significantly increased germination percentage and rate; the increase was greater for seeds matured during winter than for seeds matured during spring. This indicates that dormancy breakage was greater in seeds of winter than seeds of spring. The need for high temperature to achieve greater germination was significantly reduced after seed storage, especially for seeds matured in autumn and winter.  相似文献

Early and late varieties of the short day plant, Abelmoschusesculentus (L.) Moench. (= Hibiscus esculentus L.), were subjectedto various numbers of short days (SD; 10 h natural daylight)before transfer to 16 h long days. Short day treatments werestarted at the time of cotyledon release following seed germination,and their effects on the induction and development of flowersand fruits were recorded. While early okra required 6 SD cyclesfor the induction of flowering in all plants, late okra required14. However, both varieties needed more than 20 SD cycles forflower opening; flower buds produced by plants given fewer SDcycles abscinded with little or no development. In both varieties,the number of flower buds produced on each plant increased asthe number of short day cycles was increased from 5 to 20; flowerbuds were initiated at more axillary bud sites on the main stem.Three fruits were formed on each plant of both varieties subjectedto more than 30 SD cycles but, while the last formed fruit abscindedwithout development, the other two developed and became filledwith seeds. However, the greatest yield in fruit dry weightwas from plants kept under SD throughout and also in late okraplants given 55 SD. In general, the total number of seeds perplant increased as a larger number of SD cycles were imposed.  相似文献

In water soaked barley seeds the respiration rate (QO2) at 25 °C is about 100 times higher at 30% than at 20% water content. The treatment of seeds with mutagenic methyl methanesulphonate led to the depression of the respiration rate in dependence on the applied mutagenic dose. Seed storage at 30% water content resulted in a recovery from the inhibition of seed respiration caused by methyl methanesulphonate. In contrast, at 20% seed water content the respiration rate either slightly decreased or remained unchanged in the course of storage. The results are discussed in relation to the previously described changes in the yield of induced mutagenic effects in barley seeds treated with monofunctional alkylating agents and stored at 30% and 20% seed water content.  相似文献

Survival and biocontrol activity of Clonostachys rosea (isolate IK726) conidia during storage on barley seeds were investigated. The initial density of colony forming conidia on seed was 4 &#50 10 3 to 9 &#50 10 4 colony forming units (cfu)/seed. After 5 months storage at 4&#176;C, the density decreased by less than one order of magnitude and the biocontrol efficacy against seedling blight caused by seed-borne Bipolaris sorokiniana was maintained at a significantly high level ( > 80% disease reduction) for > 5 months. Conidial survival on seeds stored at 20&#176;C declined more rapidly than at 4&#176;C, and biocontrol efficacy was significantly reduced after 3-5 months. However, conidia produced on solid media over 20 days survived better than conidia produced in liquid culture and conidia from solid media produced over 12 days. In contrast, when seeds treated with conidia were packed with silica gel and stored at 20&#176;C, the cfu density decreased by less than one order of magnitude after 5 months and the biocontrol efficacy was still high after 6 months. A dose-response curve revealed that 103 cfu/seed were needed for 80% control of seedling blight. Similar control was obtained in storage experiments when approximately 103 cfu/seed were recovered from seed, indicating that conidia which survived also retained a high ability to control disease.  相似文献
