以清洁级ICR雄性小白鼠为实验动物,研究不同剂量乙酸铜对小白鼠的生殖毒性。采用小白鼠精子畸形实验及小白鼠骨髓嗜多染红细胞(以下简称PCE)微核实验等方法。分别对成年小白鼠腹腔每天注射0.25~16.00mg/kg8个剂量的乙酸铜,染毒7天。结果表明:乙酸铜对小白鼠的体重增长及睾丸重量具有一定的抑制作用,其中组VI、组VII的最明显。不同剂量的乙酸铜均使雄性小白鼠精子密度(P<0.001)、精子活力明显降低,具有明显的剂量效应。各实验组精子畸形率、PCE微核率均明显高于对照组(P<0.001)(P<0.05或P<0.001),且均随乙酸铜剂量的增加而明显升高。结果表明实验剂量的乙酸铜对ICR雄性小白鼠具有明显的生殖毒性效应。 相似文献

目的探明孕马血清促性腺激素(PMSG)调控哺乳动物生殖生理机制。方法用4 IU PMSG分别处理出生第5天、第10天、第15天和第21天幼鼠,观察其对卵巢和子宫发育的影响。结果(1)PMSG处理后,出生第15天幼鼠的卵巢指数(33.08%)与对照组(27.16%)间差异显著(P〈0.05);(2)PMSG处理后,第5天、10天和第15天幼鼠的初级卵泡和有腔卵泡相对数量与对照相比无差异,而21 d处理组次级卵泡相对数量显著增加;(3)虽然出生后10 d前的幼鼠子宫对PMSG的反应较弱,但是,PMSG对出生后15-21 d幼鼠子宫重量指数、子宫肌层和子宫内膜层厚度发育具有明显的促进作用,而对子宫腺体数量增加无影响。结论PMSG对幼鼠卵巢的发育无明显影响;PMSG对出生后第15天前后的子宫发挥作用,而且其作用随着出生日龄的增加而增强。 相似文献

Exercise increases both the consumption of oxygen and the production of reactive species in biological tissues, and this is counterbalanced by antioxidant adaptations to regular physical training. When the intensity of exercise fluctuates between mild and moderate, it improves the status of reduction–oxidation balance in the brain and induces neuroplasticity. However, intense exercise can oxidize the brain and impair neurological function. The effect of the frequency of exercise, which is an important factor in physical training, is still unknown. The effect of periodic exercise on biomarkers of oxidative stress in the hippocampus of mice was evaluated in this study. Mice were made to run on a treadmill for 8 weeks, two, three, or five times per week, and their hippocampi and quadriceps femoris muscles were then dissected. Biomarkers of oxidative damage were negatively correlated with the frequency of exercise and mitochondrial muscular activity, while the sulfhydryl contents were positively correlated with exercise frequency. A logistic analysis revealed a dose-dependent effect of exercise on these biomarkers. In summary, these results suggested that manipulating the frequency of physical exercise could induce antioxidant-related adaptations in the hippocampi of adult mice. 相似文献

The Arabidopsis thaliana genome contains 20 CNGCs, which are proposed to encode cyclic nucleotide gated, non-selective, Ca2+-permeable ion channels. CNGC7 and CNGC8 are the two most similar with 74% protein sequence identity, and both genes are preferentially expressed in pollen. Two independent loss-of-function T-DNA insertions were identified for both genes and used to generate plant lines in which only one of the two alleles was segregating (e.g., cngc7-1+/−/cngc8-2−/− and cngc7-3−/−/cngc8-1+/−). While normal pollen transmission was observed for single gene mutations, pollen harboring mutations in both cngc7 and 8 were found to be male sterile (transmission efficiency reduced by more than 3000-fold). Pollen grains harboring T-DNA disruptions of both cngc7 and 8 displayed a high frequency of bursting when germinated in vitro. The male sterile defect could be rescued through pollen expression of a CNGC7 or 8 transgene including a CNGC7 with an N-terminal GFP-tag. However, rescue efficiencies were reduced ∼10-fold when the CNGC7 or 8 included an F to W substitution (F589W and F624W, respectively) at the junction between the putative cyclic nucleotide binding-site and the calmodulin binding-site, identifying this junction as important for proper functioning of a plant CNGC. Using confocal microscopy, GFP-CNGC7 was found to preferentially localize to the plasma membrane at the flanks of the growing tip. Together these results indicate that CNGC7 and 8 are at least partially redundant and provide an essential function at the initiation of pollen tube tip growth. 相似文献

Establishment and maintenance of the correct epigenetic code is essential for a plethora of physiological pathways and disturbed epigenetic patterns can provoke severe consequences, e.g. tumour formation. In recent years, epigenetic drugs altering the epigenome of tumours actively have been developed for anti-cancer therapies. However, such drugs could potentially also affect other physiological pathways and systems in which intact epigenetic patterns are essential. Amongst those, male fertility is one of the most prominent. Consequently, we addressed possible direct effects of two epigenetic drugs, decitabine and vorinostat, on both, the male germ line and fertility. In addition, we checked for putative transgenerational epigenetic effects on the germ line of subsequent generations (F1–F3). Parental adult male C57Bl/6 mice were treated with either decitabine or vorinostat and analysed as well as three subsequent untreated generations derived from these males. Treatment directly affected several reproductive parameters as testis (decitabine & vorinostat) and epididymis weight, size of accessory sex glands (vorinostat), the height of the seminiferous epithelium and sperm concentration and morphology (decitabine). Furthermore, after decitabine administration, DNA methylation of a number of loci was altered in sperm. However, when analysing fertility of treated mice (fertilisation, litter size and sex ratio), no major effect of the selected epigenetic drugs on male fertility was detected. In subsequent generations (F1–F3 generations) only subtle changes on reproductive organs, sperm parameters and DNA methylation but no overall effect on fertility was observed. Consequently, in mice, decitabine and vorinostat neither affected male fertility per se nor caused marked transgenerational effects. We therefore suggest that both drugs do not induce major adverse effects—in terms of male fertility and transgenerational epigenetic inheritance—when used in anti-cancer-therapies. 相似文献

为了研究镉对小鼠生殖力和生精细胞的作用,本文对氯化镉处理后的雄性小鼠进行了交配实验,以观察其对怀孕率、每窝产仔数及子代性比的影响。测定比较了注射镉后,小鼠成熟精子的总LDH酶和LDH-X酶的活性;还用双向电泳方法分析了成熟精子的蛋白质变化。结果表明,处理组在怀孕率、每窝产仔数及子代性比方面无统计学意义的差异。成熟精子的总LDH活性经镉处理后未发现明显变化,但镉能显著地抑制与精子运动的能量有关的LDH-X酶的活性。双向电泳图谱表示,镉处理后,精子中含量较少的三组蛋白质或消失不见,或发生明显变化。 相似文献

Intravenous vaccination of mice with oil-treated mycobacterial cell walls resulted in a marked macrophage accumulation in the lungs and spleens of vaccinated animals. Injection of oil emulsion alone or of cell walls alone failed to elicit the macrophage response. Although a correlation existed between the magnitude of the macrophage response and the degree of immunity against aerosol challenge with H(37)Rv organisms, the findings presented here do not rule out the possibility that qualitative differences may be present in the macrophages of animals vaccinated against tuberculosis. The ability of oil-treated cell walls to elicit an immune response appeared to be a function of the physical association of cell wall fragments and the surface of oil droplets. 相似文献

Correlated Responses in Male Reproductive Traits in Mice Selected for Litter Size and Body Weight

下载免费PDF全文 Correlated responses in male reproductive traits were determined at 4, 6 and 8 weeks of age in lines of mice selected for large litter size (L+), large 6-week body weight (W+), large litter size and small body weight (L+W-) and small litter size and large body weight (L-W+), and in an unselected control (K). Concentration of serum testosterone and weights of testes, seminal vesicles, epididymides and adrenal glands increased with age. Line differences in testosterone concentration were not detected. L+ and W+ males exhibited positive correlated responses in testes, epididymides and seminal vescile weights. Testis weight adjusted for body weight was significantly larger for L+ than controls and approached significance for W+. Realized genetic correlation betestis weight and litter size was 0.60 ± 0.04, and the realized partial genetic correlation holding body weight constant was 0.42. Therefore, pleiotropic loci, acting via the hypothalamic-pituitary axis, affect testis weight and litter size independently of body weight. Additionally, genes influencing overall growth have a pleiotropic effect on testis weight and litter size in mice; the realized genetic correlations of body weight with testis weight and with litter size were 0.60 ± 0.03 and 0.52 ± 0.10. Testis weight increased in both L+W- and L-W+ males. The positive correlated response in L+W- may have resulted from changes in frequency of genes controlling reproductive processes; whereas, in L-W+ it could have been the result of changes in the frequency of genes associated with body weight. 相似文献

In this study, employing energy dispersive x-ray analysis facilities on electron microscope, presence of different elements in human male reproductive organs-??testis, epididymis, caput, corpus and cauda, prostate gland, seminal vesicle, Cowper??s gland and vas deferens??seminal plasma and spermatozoa pellet was studied. Several elements were observed. Gold was one among them that was present in seminal plasma and spermatozoa. It was also present in epididymis caput. There are possible hazardous health effects of exposure to radiofrequency electromagnetic radiations emitted from mobile phone on the human reproductive pattern. It is more effective while keeping mobile phones in pocket or near testicular organs. Present review examines the possible concern on radio frequency radiation interaction and biological effects such as enzyme induction, and toxicological effects, including genotoxicity and carcinogenicity, testicular cancer, and reproductive outcomes. Testicular infertility or testicular cancer due to mobile phone or microwave radiations suggests an increased level of reactive oxygen species (ROS). Though generation of ROS in testis has been responsible for possible toxic effects on physiology of reproduction, the reviews of last few decades have well established that these radiations are very harmful and cause mutagenic changes in reproductive pattern and leads to infertility. The debate will be focused on bio-interaction mechanism between mobile phone and testicular cancer due to ROS formation. This causes the biological damage and leads to several changes like decreased sperm count, enzymatic and hormonal changes, DNA damage, and apoptosis formation. A tobacco clone TPD1 (Tobacco Pollen Development), characterized by an extended flowering period, was obtained using a serial agrobacterial transformation of tobacco (Nicotiana tabacum L., cv. Samsun) by constructing a DNA carrying the kanamycin resistance gene inserted into the binary pBin19 vector. However, the characteristics of the vegetative growth of these plants were similar to those of other tobacco clones and the wild type. In the insertion mutant, pollen was 1.5 times smaller than in the wild type and germinated on the stigmata of neither its own clone nor the wild-type plants. Cytochemical investigation of pollen of the TPD1-mutant did not reveal any activity of respiratory enzymes, succinate dehydrogenase and peroxidase, indicating that the pollen was nonviable. Unlike the wild type, theTPD1 mutant exhibited disturbed trophic interactions within the anther tissues, particularly suppressed starch hydrolysis in the anther wall tissues at the stage of rapidly growing microspores. We conclude that the insertion of T-DNA into the TPD1 gene produced structural and metabolic changes during the development of anther tissues in the mutant clone, resulting in pollen sterility. 相似文献

下载免费PDF全文 This review describes the various viruses identified in the semen and reproductive tracts of mammals (including humans), their distribution in tissues and fluids, their possible cell targets, and the functional consequences of their infectivity on the reproductive and endocrine systems. The consequences of these viral infections on the reproductive tract and semen can be extremely serious in terms of organ integrity, development of pathological and cancerous processes, and transmission of diseases. Furthermore, of essential importance is the fact that viral infection of the testicular cells may result not only in changes in testicular function, a serious risk for the fertility and general health of the individual (such as a fall in testosteronemia leading to cachexia), but also in the possible transmission of virus-induced mutations to subsequent generations. In addition to providing an exhaustive account of the data available in these domains, this review focuses attention on the fact that the interface between endocrinology and virology has so far been poorly explored, particularly when major health, social and economical problems are posed. Our conclusions highlight the research strategies that need to be developed. Progress in all these domains is essential for the development of new treatment strategies to eradicate viruses and to correct the virus-induced dysfunction of the endocrine system. 相似文献

LEAFY gene is the positive regulator of the MADS-box genes in flower primordia. The number of MADS-box genes presumably increased
by gene duplications before the divergence of ferns and seed plants. Most MADS-box genes in ferns are expressed similarly
in both vegetative and reproductive organs, while in gymnosperms, some MADS-box genes are specifically expressed in reproductive
organs. This suggests that (1) the increase in the number of MADS-box genes and (2) the subsequent recruitment of some MADS-box
genes as homeotic selector genes were important for the evolution of complex reproductive organs. The phylogenetic tree including
both angiosperm and gymnosperm MADS-box genes indicates the loss of the A-function genes in the gymnosperm lineage, which
is presumably related to the absence of perianths in extant gymnosperms. Comparison of expression patterns of orthologous
MADS-box genes in angiosperms, Gnetales, and conifers supports the sister relationship of Gnetales and conifers over that
of Gnetales and angiosperms predicted by phylogenetic trees based on amino acid and nucleotide sequences.

The case of a 40-year-old sighted woman with free-running sleep-wake and melatonin rhythms is presented. The subject was studied for 102 days. During the pre-treatment period, both the sleep-wake and melatonin rhythms had a period of 25.1 hr, similar to the average period of humans living in temporal isolation. Treatment consisted of bright artifical light exposure (2500 lx Vita-Lite) for 2 hr each day upon awakening. Clock time of light exposure was held constant for 6 days and then slowly advanced until the subject was arising at her desired time of day. The subject continued the light treatment at home and was able to live on a 24-hr day for the 30-day follow-up study. While other factors may be operating in this situation, it is possible that the light treatment caused the stabilization of the free-running rhythms, advancement to a normal phase and entrainment to the 24-hr day. We suspect that the tendency to free-run was related to sleep onsets that were abnormally delayed relative to the circadian phase response curve for light. By scheduling a 2-hr pulse of bright light each morning, this tendency to delay would be counteracted by light-induced advances, resulting in normal entrainment. 相似文献

Epilepsy is one of the prevalent and major neurological disorders, and approximately one-third of the individuals with epilepsy experience seizures that do not respond well to available medications. We investigated whether oxysophocarpine (OSC) had anticonvulsant and neuroprotective property in the pilocarpine (PILO)-treated mice. Thirty minutes prior to the PILO injection, the mice were administrated with OSC (20, 40, and 80 mg/kg) once. Seizures and electroencephalography (EEG) were observed, and then the mice were killed for Nissl and Fluoro-jade B (FJB) staining. The oxidative stress was measured at 24 h after convulsion. Western blot analysis was used to examine the expressions of the Bax, Bcl-2, and Caspase-3. In this study, we found that pretreatment with OSC (40, 80 mg/kg) significantly delayed the onset of the first convulsion and status epilepticus (SE) and reduced the incidence of SE and mortality. Analysis of EEG recordings revealed that OSC (40, 80 mg/kg) significantly reduced epileptiform discharges. Furthermore, Nissl and FJB staining showed that OSC (40, 80 mg/kg) attenuated the neuronal cell loss and degeneration in hippocampus. In addition, OSC (40, 80 mg/kg) attenuated the changes in the levels of Malondialdehyde (MDA) and strengthened glutathione peroxidase and catalase activity in the hippocampus. Western blot analysis showed that OSC (40, 80 mg/kg) significantly decreased the expressions of Bax, Caspase-3 and increased the expression of Bcl-2. Collectively, the findings of this study indicated that OSC exerted anticonvulsant and neuroprotective effects on PILO-treated mice. The beneficial effects should encourage further studies to investigate OSC as an adjuvant in epilepsy, both to prevent seizures and to protect neurons in brain. 相似文献
